The developmental parameters and fitness of a recent invasive insect pest of cereals in western Canada, the cereal leaf beetle, Oulema melanopus (L.) (Coleoptera: Chrysomelidae), were compared on live plants and excised leaves of commercial cultivars of potential cereal hosts including wheat (winter and spring), oats, barley, corn, rye and triticale. Host preference and utilization within the fundamental host range of O. melanopus differed. The biological parameters differed significantly when the larvae were reared on excised tissues versus on live host plants. In both studies, wheat (winter and spring), oat (cv. Morgan) and barley were the most preferred hosts in terms of development, survivorship, adult weights and fecundity. Prolonged development with low fitness gains was noted on corn. Although the development on hosts such as rye and triticale was prolonged and adult fitness was low, the survivorship was high on these hosts. Hence, these crops can act as secondary hosts for the beetle in its new eco-region. Larvae fed foliage of Waldern, a local oat cultivar, had increased developmental time and lower survivorship compared with other cereal hosts. Despite differences in fitness gains among hosts, similar numbers of eggs were laid on all hosts. Hence, the oviposition choice of O. melanopus may not be driven by fitness gains alone and it may indicate adaptive strategy to hosts in newer environments.  相似文献

This work was undertaken to determine the kinds and amount of substances that would account for the previously demonstrated differential growth of Claviceps purpurea on guttation fluids from Rosen rye, Genesee wheat, and Traill barley seedlings. Chromatographic methods were used for determining amino acids and sugars, spot tests and spectrometric methods for inorganic materials, and microbiological methods for vitamins.

Total sugar content is about equal in rye and barley fluids, but lower in wheat. Glucose is the principal sugar component of the rye and barley fluids and galactose highest in wheat. Most of the amino acid in all 3 fluids is aspartic acid or asparagine. Barley fluid is far higher than the other 2 in total amino acids, with wheat the lowest. Most inorganic elements are found to be highest in barley and lowest in wheat, with the exception of iron where rye is highest and barley lowest. Barley fluid is highest in choline, p-aminobenzoic acid, thiamine, and uracil, while rye is highest in inositol and pyridoxine. Wheat is much lower than the other 2 in choline and inositol.

Silicon deposits in the epicarp hairs of the caryopses of mature,field-grown specimens of Hordeum sativum, Avena sativa, Secalecereale and Triticum aestivum were investigated using electron-probemicroanalysis. In all four cereals, silicon was most concentratedat the extreme tips of the hairs. In barley, it was the onlyelement detected; in the other three cereals potassium and calciumwere located below the tip. In wheat, chlorine was also detected. The hair bases of the different cereals displayed variationin the elements detected. Silicon and polassium were presentin all four and calcium present in all except rye. The hairbases of wheat also contained chlorine; phosphorus and zincwere located in barley. The latter alone showed variation ofelements between hair bases. Scanning electron microscopy revealed heavy striations of thehair tips of barley and oats. In rye and wheat, the tips weresmooth but there were slight surface markings below the tip. The results are discussed in relation to the possible functionsand significance of the epicarp hairs and their silicification. Green peach aphid, Myzus persicae (Sulzer), does not overwinter outdoors in Minnesota; it arrives each spring on low-level jet streams from the south. After arrival, anholocylic reproduction occurs on numerous herbaceous species, including many common weeds, before movement to potato, Solanum tuberosum L. In investigating aphid feeding behavior on barrier crops, we observed winter wheat, Triticum aestivum L., colonized by green peach aphid. The Northern Great Plains grows 94,000 ha of potatoes and 20.5 million ha of small grain cereals each year, the latter potentially providing an early emerging and widely distributed green peach aphid host to influence early season potato colonization. Life tables statistics indicated green peach aphid had its highest reproductive potential among cereals on winter wheat, with rye (Secale cereale L.) > barley (Hordeum vulgare L.) > oats (Avena sativa L.). Green peach aphid was found to colonize barley, rye, and winter wheat, but not oats. Mean generation time, net reproductive rate, doubling time, and finite rate of increase were significantly different between host plants. Electrical penetration graph technique indicated mean nonpenetration duration by green peach aphid was significantly different among plant species, and significantly longer on winter wheat than on the other cereals. Mean xylem phase duration was not significantly different among plant species but sieve element salivation was of longest duration on potato. Phloem sap ingestion (E2) was also significantly different among plant species with longest E2 duration on winter wheat. This study demonstrates that this aphid can effectively use key cereals at the vegetative stage.  相似文献

Agriculture played a significant role in increasing the number of pathogen species and in expanding their geographic range during the last 10,000 years. We tested the hypothesis that a fungal pathogen of cereals and grasses emerged at the time of domestication of cereals in the Fertile Crescent and subsequently speciated after adaptation to its hosts. Rhynchosporium secalis, originally described from rye, causes an important disease on barley called scald, although it also infects other species of Hordeum and Agropyron. Phylogenetic analyses based on four DNA sequence loci identified three host-associated lineages that were confirmed by cross-pathogenicity tests. Bayesian analyses of divergence time suggested that the three lineages emerged between approximately 1200 to 3600 years before present (B.P.) with a 95% highest posterior density ranging from 100 to 12,000 years B.P. depending on the implemented clock models. The coalescent inference of demographic history revealed a very recent population expansion for all three pathogens. We propose that Rhynchosporium on barley, rye, and Agropyron host species represent three cryptic pathogen species that underwent independent evolution and ecological divergence by host-specialization. We postulate that the recent emergence of these pathogens followed host shifts. The subsequent population expansions followed the expansion of the cultivated host populations and accompanying expansion of the weedy Agropyron spp. found in fields of cultivated cereals. Hence, agriculture played a major role in the emergence of the scald diseases, the adaptation of the pathogens to new hosts and their worldwide dissemination.  相似文献

Newly germinated seedlings of susceptible cultivars of oats, wheat, barley and rye were inoculated with second-stage juveniles of Heterodera avenae in pots of sand. Subsequent examination showed oat root tips to be more commonly invaded, and by a greater range of nematode numbers than the other cereals. A comparison of oats and barley showed that lower nematode numbers in barley were not due to a higher emigration from barley; invasion, establishment and emigration by nematodes all being greater in oats. Second-stage juveniles were more likely to migrate prior to establishment in barley than in oats.  相似文献

We traced the liberation and biological effect of volatile substances released from the roots of cereals,i. e. barley, wheat, rye and oats, on seedlings of the same and other plant species. Experiments were carried out in a closed glass apparatus with a static or circulating atmosphere in which the CO2 and O2 were permanently absorbed and supplemented, respectively. In some experiments the air was bubbled through water or through solutions of boric acid, barium hydroxide and potassium permanganate. The roots of all four cereals tested released volatile substances with a biological activity which appeared to be non-specific with respect to plant species. The effect of volatile substances was partially decreased by bubbling through water, barium hydroxide and boric acid and was completely removed after passing through the solution of potassium permanganate. Volatile substances liberated from roots of barley inhibited elongation of roots and coleoptile, decreased SH-group content and caused excessive formation of root hairs as well as inhibition of both dry matter production and respiration of roots of rye seedlings. Ethylene was found in the atmosphere of experimental vessels.  相似文献

Deoxynivalenol (DON) is a hazardous and globally prevalent mycotoxin in cereals. It commonly accumulates in the grain of wheat, barley and other small grain cereals affected by Fusarium head blight (caused by several Fusarium species). The concept of reducing DON in naturally contaminated grain of wheat or barley using a DON-degrading bacterium is promising but has not been accomplished. In this study, we isolated a novel DON-utilising actinomycete, Marmoricola sp. strain MIM116, from wheat heads through a novel isolation procedure including an in situ plant enrichment step. Strain MIM116 had background degradation activity, and the activity was enhanced twofold by the consumption of DON. Among Tween 20, Triton X-100 and Tween 80, we selected Tween 80 as a spreading agent of strain MIM116 because it promoted DON degradation and the growth of strain MIM116 in the presence of DON. The inoculation of MIM116 cell suspension plus 0.01% Tween 80 into 1,000 harvested kernels of wheat and barley resulted in a DON decrease from approximately 3 mg kg?1 to less than 1 mg kg?1 of dry kernels, even when cells had only basal levels of DON-degrading activity. To the best of our knowledge, this is the first report that describes (1) the isolation of a DON-degrading bacterium from wheat heads, (2) the effects of surfactants on the biodegradation of DON and (3) the decrease of DON levels in naturally contaminated wheat and barley grain using a DON-degrading bacterium.  相似文献

The relative suitability of potato and crops frequently grown in rotation with potato as hosts for Pratylenchus penetrans was evaluated. Suitability of rye, wheat, corn, oat, sorgho-sudangrass, and potato were compared in pot studies based on ratios of final population : initial population density and densities of nematodes in roots at harvest. Population densities increased more on potato, oat, and corn than on rye, wheat, and sorgho-sudangrass. There were no differences among the four rye cultivars or between the two oat cultivars in host suitability. Population increases were not related to root weight or consistently to nematode densities in roots. Although rye and wheat were equally suitable hosts in pot studies, P. penetrans increased more on wheat than on rye in a field study, indicating that reproduction was reduced or mortality was increased on rye under field conditions.  相似文献

Brazil is blast disease hot spot because severe epidemics have occurred among wheat, triticale, rye, barley and oat crops. Although the first outbreak of barley blast appeared in 1998, little information is available. Therefore, this study aimed to examine host range, mating type composition and population structure of Magnaporthe sp. from a single barley field in São Paulo, Brazil. To examine pathogenicity, 25 Magnaporthe isolates were inoculated on five, three, two and two cultivars of barley, wheat, oat and rice, respectively, and one cultivar each of rye, corn, sorghum, triticale and certain weeds (Cenchrus echinatus, Setaria geniculata, Brachiaria plantaginea and Eleusine indica). Mating type distribution of 33 isolates was investigated by molecular tools. The genotypic divergence of 41 barley and five wheat isolates was investigated by 15 random amplified polymorphic DNA primers and unweighted pair group method with arithmetic mean. The host range of the barley blast pathogen included wheat, oat, rye and triticale but not rice and weeds. Sexual reproduction appeared to not be involved in the high genotypic diversity because only a single isolate, MAT1‐2, was identified. The majority of barley isolates clustered together with wheat blast, except for four, suggesting a different origin.  相似文献

Ergot alkaloids are toxins produced by some species of fungi in the genus Claviceps, that may infect rye and triticale and, in a minor degree, other types of cereals. In this study, a new UHPLC-FLD method for the quantification of the six major ergot alkaloids as well as their corresponding epimers was developed. The sample preparation was done by a solid-liquid extraction with acetonitrile and clean-up via freeze-out. The method was fully validated and then applied to 39 samples (wheat, rye, triticale, and barley) harvested in Luxembourg in 2016. Samples were sieved (1.9?×?20 mm) prior to analysis in order to remove sclerotia, hosting the alkaloids. However, 23 samples still contained at least one ergot alkaloid >?LOQ and concentrations of the sum of the 6 ergot alkaloids ranged from 0.3 to 2530.1 μg/kg. Interestingly, the highest concentrations were measured in wheat and not in rye or triticale, suggesting that all kinds of cereals should be included in monitoring programs. The outcome of this study allowed giving a first overview of ergot alkaloid concentrations in cereals harvested in Luxembourg, and the measured concentrations were in similar ranges than in other parts of the world (e.g., Canada, France, Germany).  相似文献

Carboxypeptidase activity (hydrolysis of N-carbobenzoxy-l-phenylalanyl-l-alanine) is high in a number of temperate zone cereals, originating in Asia Minor (wheat, barley, oats, wild oats, rye, triticale) compared to other cereals originating in central America or Asia (maize, sorghum, rice). However, endopeptidase activity (hydrolysis of azocasein or hemoglobin) is relatively much higher in the latter group. Comparison of trichloroacetic acid (TCA)-soluble products derived from the hydrolysis of hemoglobin showed that carboxyterminal amino acids (histidine, arginine, and tyrosine), are released when extracts from wheat and barley endosperms are used. With extracts from corn endosperms, much more TCA-soluble ultraviolet- absorbing material is released, but very little is released as free amino acids within the first 2 hours and the expected C-terminal amino acids of hemoglobin are not detected in significant amounts. These results suggest that the method of hydrolysis of the storage proteins may be significantly different in these two classes of cereals.  相似文献

We have found extensive nucleosomal fragmentation of native DNA extracted from leaves of healthy cereal plants, as indicated by ladder patterns on agarose gels and TUNEL staining. The time of first appearance of fragmentation differed among cereals. Native DNA from the first leaf of 10-day-old plants formed a clear ladder pattern of multiples of 180 bp fragments in wheat and triticale but not in barley and oats. In one cultivar of rye a weak ladder pattern occurred but not in another. Freezing and thawing of samples before DNA extraction resulted in much more extensive DNA fragmentation in wheat but not in rye and barley, indicating that DNA-degrading enzymes are present in the cytoplasm of wheat, but not in barley and rye, at this stage. In barley, nucleosomal fragmentation was first detected in 25-day-old plants. These results indicate that programmed cell death takes place in developing leaves of young cereal plants, but that the time of onset differs among cereal species.  相似文献

A 3 yearFusarium andFusarium toxin monitoring programme was established within the food and feed control authorities of Saxony-Anhalt in 2001. The first year’s results of the analysis of deoxynivalenol in cereals and cereal products with assured origin in this federal state, showed a contamination rate of 24% for wheat and wheat products. The contamination incidence reached only 8% in rye and rye products, whereas it was 17% for barley and its products including beer. Zearalenone could be detected only in 2 of 162 analysed samples.  相似文献

The wheat curl mite (WCM) is a major pest in cereal crops around the world and the vector of at least four known pathogens capable of reducing yields in crops such as wheat, corn, barley, oats, millet and rye. Current taxonomy recognizes WCM as a single species, Aceria tosichella; however, recent genetic, physiological and ecological studies have shown that WCM is likely to be a species complex. In this study we assessed genetic variation and phylogenetic relationships among WCM from four continents and a wide range of host plants using DNA sequence data from one mitochondrial gene, one nuclear gene and a single nuclear intergenic spacer region. Phylogenetic analyses revealed 11 unique mite lineages associated with specific plant hosts including wheat and barley. Host associations were consistent across continents, often with a single haplotype dominating a host plant regardless of geographic origin. The genetic and ecological differences identified in this study support the notion that WCM is a species complex in need of major taxonomic revision. These findings have implications for control of WCM globally, particularly within the context of identifying plants that form ‘green bridge’ refuges, assessing disease transmission risk, and identifying resistance in cereal genotypes to WCM and associated pathogens.  相似文献

In the autumn of 1953 an experiment was begun to follow changes in the cereal-root eelworm population of small plots on a field in Shropshire. The plots were cropped with either oats, wheat, barley or rye, sown in the autumn and spring, and some plots had fertilizer. Each plot received the same treatment for 3 years; in the fourth year an indicator crop of spring oats was grown on all the plots.
Under rye and autumn-sown wheat the eelworm population fell to a level which permitted a good oat crop in 1957. Autumn-sown wheat, barley and rye generally produced lower eelworm populations than their spring-sown counterparts, but autumn-sown oats proved to be the most efficient host. The order of host efficiency was oats (best), barley, wheat, rye. The eelworm populations were generally higher on plots receiving fertilizer treatment. In this experiment all oat plots, and spring-sown barley plus fertilizer, produced populations which severely damaged the 1957 oat crop.  相似文献

The molecular weights ofwaxy proteins, by SDS-PAGE, and the N-terminal amino acid sequences of mature protein and of V8 protease-induced fragments were determined in diploid cereals. The homology of the primary structure was relatively high among cereals examined here, and there appeared to be a common sequence, V-F-V-G-A-E-M-A, in the vicinity of the N terminus. Based on the amino acid sequences, these cereals could be divided into two groups, including corn and rice in one and diploid wheat, fourAegilops species, rye, and barley in the other. In diploid wheat andAegilops species there were substitutions of amino acids in the primary structure. Variations of this sort suggest that the primary structure ofwaxy proteins would provide clues to the phylogenetic relations in the wheat group.  相似文献

Total globulins from embryos and endosperms of barley, wheat,rye, and oats were separated by SDS-PAGE under reducing andnon-reducing conditions. The preparations from embryos of allfour cereals contained major groups of bands with Mr's of 50-60000,which were not affected by reduction. These have been characterizedpreviously from oats and shown to correspond to subunits ofthe 7S storage globulin. Immunochemical relationships betweenthese bands (and others with Mr's between 40000 and 70000) weredemonstrated by immunodiffusion and ‘Western Blotting’using antiserum raised against the major subunits of the oat7S globulins. The 7S globulins were also prepared from hand-dissectedembryos of the four cereals using sucrose density ultracentrifugation.Their amino acid compositions were broadly similar, but differedfrom those of the 7S vicilins of legumes. Fusarium graminearum is the main causative agent of Fusarium head blight on small grain cereals and of ear rot on maize. The disease leads to dramatic yield losses and to an accumulation of mycotoxins. The most dominant F. graminearum mycotoxins are the trichothecenes, with deoxynivalenol and nivalenol being the most prevalent derivatives. To investigate the involvement of trichothecenes in the virulence of the pathogen, the gene coding for the initial enzyme of the trichothecene pathway was disrupted in three field isolates, differing in chemotype and in virulence. From each isolate three individual disruption mutants were tested for their virulence on wheat, barley and maize. Despite the different initial virulence of the three wild-type progenitor strains on wheat, all disruption mutants caused disease symptoms on the inoculated spikelet, but the symptoms did not spread into other spikelets. On barley, the trichothecene deficient mutants showed no significant difference compared to the wild-type strains: all were equally aggressive. On maize, mutants derived from the NIV-producing strain caused less disease than their wild-type progenitor strain, while mutants derived from DON-producing strains caused the same level of disease as their progenitor strains. These data demonstrate that trichothecenes influence the virulence of F. graminearum in a highly complex manner, which is strongly host as well as moderately chemotype specific.  相似文献
