The aerial microalga Trentepohlia aurea has beeninvestigated in relation to removal characteristics of nitrate, nitrite,ammonium and phosphate ions. When the alga was cultured in medium with veryhighconcentrations of ammonium, nitrate and phosphate ions, it showed relativelyhigh growth and removal rates. It also grew quite well with high nitriteconcentration (< 141 mg NO2-N L–1).The removal rate was 0.28 mg NO2-N L–1day–1 in the 40-day culture, when it was cultured in modifiedBold's basal medium with added 51 mg NO2-NL–1. In addition, we examined simultaneous removal of nutrientions. The biomass was 1.5 times higher in medium which N- and P-sourcesufficient than in ordinary medium. Higher removal ratios of nitrite andnitratefrom medium were shown in a 30-day culture, reaching 37% and 32%, respectively.It is concluded that T. aurea has the potential for use inthe purification of wastewater.  相似文献

Removal of inorganic nitrogen sources by cells of the aerial microalga Trentepohlia aurea grown on the surface of substrate, such as filter paper, has been investigated in a batch system. When the alga grew on the paper dampened with medium, it actively ingested inorganic nitrogenous compounds in the medium. Immobilized cells on the filter papers were called algal biofilm in this study. When the algal biofilms were soaked in modified Bold's Basal medium (using 1 g NH4Cl l–1 as a N source), the removal rate was 4.25 mg ammonium-N l–1 day–1 in 40 days. In modified medium with added 26 mg nitrite-N, the removal rate of the total inorganic N ion by the biofilms reached 5.11 mg N l–1 day–1. This removal rate of total N ion was higher than that in the medium by addition of 26 mg nitrate-N. In addition, we tried to examine simultaneous removal of ammonium, nitrate, and nitrite ions and growth inhibition of cyanobacteria in the medium by using the algal biofilms. Consequently, it was demonstrated that the algal biofilms of T. aurea could be utilized as a biofunctional material for the purification of wastewater.  相似文献

Application of a laboratory-scale photobioreactor containing a biofilter composed of the aerial microalga Trentepohlia aurea to the removal of ammonium from synthetic wastewater was assessed to determine whether the system could be applied to water purification and the treatment of eutrophic water. The removal efficiency of the photobioreactor was tested after ten biofilter sheets (total dry weight cells: 50 mg) were cycled in nitrogen-free Bold’s basal (BB) medium for 72 h (pretreatment cycle). The ammonium removal ability was significantly enhanced when the photobioreactor was operated after performing the pretreatment cycle using nitrogen-free BB medium supplemented with magnesium. Moreover, the illumination conditions during the treatment were shown to affect the nitrogen removal ability, and this ability was strongly dependent on the concentrations of organic compounds (e.g., α-ketoglutarate and pyruvate) for assimilating the nitrogen source in the T. aurea biofilter.  相似文献

The paper presents a five-phase model to describe batch culture of Spirulina maxima under limitations of light and nutrients nitrogen andsulfur. The general equation for the exponential, linear, decelerating andstationary phases of the growth curve took into account that (i) the specificgrowth rate was proportional to the local light intensity in thephotobioreactor; (ii) light attenuation was due to cell's absorption andreflection and observed the Lambert-Beer's law with a total absorptioncoefficient (T) that was the sum of absorption coefficient(a) and reflection coefficient (r); (iii) thecomposition of the alga and its absorption a changed withtime and (iv) the specific growth rate was influenced by nutrients ofnitrogen and sulfur in the culture medium according to a Monod's law.Two successive equations describing the death phase were proposed basedon the hypothesis that the death rate accelerated with the %PSC until itreached its maximum. From that point the %PSC decreased and the deathrate reduced. Our model fitted very well the cultures grown in the photobioreactor developed in our laboratory.  相似文献

Nine species ofNeochloris can be divided into three groups on the basis of comparative ultrastructure of the flagellar apparatus, the cell wall and the pyrenoid of zoospores. In Group I,N. wimmeri andN. minuta, zoospores are thin-walled, pyrenoids are penetrated by stromal channels, and the basal bodies are in the clockwise absolute orientation and connected by the distal and two proximal fibers. In Group II,N. aquatica, N. vigenis, N. terrestris, N. pyenoidosa, andN. pseudostigmatica, zoospores are naked or covered by fuzzy material, pyrenoids are covered by a continuous starch sheath or invaginated by cytoplasmic channels, basal bodies are directly opposed, the distal fiber is differentiated into a ribbed structure at the central region, a striated microtubule-associated component (SMAC) is continuous between opposite two-membered rootlets and connected to the ribbed structure, proximal ends of basal bodies are covered by partial caps, each two-membered rootlet and a basal body are connected by a striated fiber to the X-membered rootlet associated with the opposite basal body, and the basal bodies, when oriented at wide angles, are joined at their proximal ends by core extensions. In Group III,N. pseudoalveolaris andN. cohaerens, zoospores are naked, pyrenoids are traversed by parallel thylakoids, basal bodies are in the counterclockwise absolute orientation and overlapped, and each X-membered rootlet is connected to the end of the opposite basal body by a terminal cap. It is suggested that the genusChlorococcopsis gen. nov. be erected for the Group I species. Group II, which includes the type species,N. aquatica, should be preserved asNeochloris. The group appears to be closely related to the coenobial generaPediastrum, Hydrodictyon, andSorastrum, and to have affinities with the coenocytic generaSphaeroplea andAtractomorpha as well. It is also suggested that the genusParietochloris gen. nov. be erected in thePleurastrophyceae for the species of Group III.  相似文献

Based on biotesting, we carried out an estimation of the water quality in the Amursky and Nakhodka Bays (Sea of Japan) using Plagioselmis prolonga (Cryptophyta). The obtained data were compared with the data from water biotesting using Dunaliella salina (Chlorophyta). It was shown that water from the Amursky Bay produced more a negative effect on both microalgae than water from the Nakhodka Bay. It was established that sensitivity of the P. prolonga microalga exceeded that of D. salina. This was confirmed by a sharp decrease of the P. prolonga motile cell number in the studied water.  相似文献

Batch cultures of algae grown at low (0.1 %) and elevated (2.0 %) concentrations of CO2, as well as in original BBM (Bold Basal Medium) and BBM modified with phosphate, EDTA and a combination of both, were exposed to cadmium (Cd(NO3)2·4H2O, 3CdSO4·8H2O and CdCl2·H2O) for 24 h. Regardless of the salt applied, the concentration-dependent relationships of Cd toxicity were found to be biphasic, suggesting the different affinity of target sites to cadmium. Nominal values of EC50 obtained for algae grown in original BBM and at low CO2 were 18.0, 16.44 and 15.37 mg·dm−3 for cadmium nitrate, sulphate and chloride, respectively. However, it was estimated that 97 % of the free cadmium in the added salts were bound by components of original BBM such as EDTA, phosphates, chloride and sulphate. The effect of Cd-salts at concentrations corresponding to EC50 values on algae were tested in media with 10-fold reduced phosphates (BBM-P), BBM depleted of EDTA (BBM-EDTA) and of both phosphates and EDTA (BBM-P-EDTA). For algae grown at low CO2 and BBM-P, cadmium was about 25 % less toxic than those applied in original BBM. Cadmium greatly inhibited (about 85 % of the control) the growth of algae cultured in BBM-EDTA; this effect was only slightly dependent on the CO2 concentration. Deficits of both EDTA and P led to effects similar to those brought about by the absence of EDTA only. The toxicity of cadmium depends on CO2 concentration only when algae are grown in original BBM. The growth of algae under high CO2 conditions was reduced considerably less (about 80% of control) compared with low CO2 concentrations (about 50 % of control). A relationship was found between the toxicity of cadmium salts and final pH values only in variants of low-CO2 grown algae; with an increase of medium pH the toxicity decreased. The results suggest that both growth conditions and the binding ability of the medium markedly affect the toxicity of cadmium towards microalgae.  相似文献

Three Gram-negative bacterial strains were isolated from the biofilter of a recirculating marine aquaculture. They were non-pigmented rods, mesophiles, moderately halophilic, and showed chemo-organoheterotrophic growth on various sugars, fatty acids, and amino acids, with oxygen as electron acceptor; strains D9-3T and D11-58 were in addition able to denitrify. Phototrophic or fermentative growth could not be demonstrated. Phylogenetic analysis of the 16S rRNA gene sequences placed D9-3T and D11-58, and D1-19T on two distinct branches within the alpha-3 proteobacterial Rhodobacteraceae, affiliated with, but clearly separate from, the genera Rhodobacter, Rhodovulum, and Rhodobaca. Based on morphological, physiological, and 16S rRNA-based phylogenetic characteristics, the isolated strains are proposed as new species of two novel genera, Defluviimonas denitrificans gen. nov., sp. nov. (type strain D9-3T = DSM 18921T = ATCC BAA-1447T; additional strain D11-58 = DSM19039 = ATCC BAA-1448) and Pararhodobacter aggregans gen. nov., sp. nov (type strain D1-19T = DSM 18938T = ATCC BAA-1446T).  相似文献

The effects of temperature on pollutant removal of two plant species (Oenanthe javanica D.C. and Nasturtium officinale) were evaluated using simulated microcosms of the floating eco-island system (FEIS). Both the planted FEIS (P-FEIS) and the non-planted FEIS (NP-FEIS) dramatically decreased NH4–N concentration in the hypereutrophic water at low (10 °C), medium (22 °C), and high (35 °C) temperatures, and to a greater extent for the P-FEIS and at medium temperature. The NO2–N concentration was effectively decreased from 0.23 to 0.01 mg/L after 4 d treatment with the P-FEIS at all the three temperatures, but was slightly increased in the NP-FEIS at low temperature. The P-FEIS could decrease NO3–N concentration in the eutrophic water over 1–3 times depending on temperature, with greater decrease at high temperature. The remove of total P (TP) reached 78%, on average, with the FEIS treatment for 4 d at all temperatures, which was over three times greater than those with the NP-FEIS at low temperature. The removal rates of Chla, CODMn, and BOD5 by the P-FEIS from the hypereutrophic water were, on average, 70%, 85%, 83% at 22 °C and 35 °C, respectively, while over 1–2 times smaller at 10 °C. More effective removals of Chla, CODMn, and BOD5 (over 1–2 times) were noted with the P-FEIS than those with the NP-FEIS. N. officinale showed more efficiency in removing ammonium and TN at low temperature, and BOD5 at medium and high temperatures, as compared to O. javanica. Whereas O. javanica could more effectively decrease Chla at 22 °C and 35 °C and CODMn at 10 °C than N. officinale after 4 d treatment. Higher dissolved oxygen concentration and pH was found in the FEIS with N. officinale than that with O. javanica D.C. The results imply that plant eco-island system had remarkable purification ability to remove pollutants from hypereutrophic water, and mixed planting of O. javanica D.C. with N. officinale on the FEIS may enhance nutrient removal and water quality improvement of eutrophic water bodies, especially at low temperature season.  相似文献

An indigenous polychlorinated biphenyl (PCB)-degrading bacterial consortium was obtained from soils contaminated by transformer oil with a high content of PCBs. The PCB degrader strains were isolated and identified as Brevibacterium antarcticum, Pandoraea pnomenusa, and Ochrobactrum intermedium by 16S rRNA gene sequence phylogenetic analysis. The PCB-degrading ability of the consortium and of individual strains was determined by using GC/MS. The PCB-degrading capacities of the consortium were evaluated for three concentrations of transfomer oil ranging from 55 to 152 μM supplemented with 0.001% biphenyl and 0.1% of Tween 80 surfactant. PCB biodegradation by the consortium was favored in the presence of both additives and the greatest extent of biodegradation (67.5%) was obtained at a PCB concentration of 55 μM. Each bacterial species exhibited a particular pattern of degradation relating to specific PCB congeners. Isolated strains showed a moderate degradation capability towards tetra-, hepta-, and octa-chlorobiphenyls; although no effect on penta-, hexa-, and nona-chlorobiphenyls was observed. Recently, PCB degradation capacity was recognized in a Pandorea member; however, this is the first study that describes the ability of Brevibacterium and Ochrobactrum species to degrade PCBs.  相似文献

The marine microalga Isochrysisgalbana was cultivated and entrapped inalginate beads for long-term storage. Theentrapped cells were alive and maintainedtheir physiological activities after oneyear of storage in absolute darkness at4 °C without a liquid medium. Thenumber of cells in the beads increased morethan 32 times when they were subsequentlyre-cultured in an aqueous medium for fiveweeks, showing that they had remained aliveduring storage. TEM observations showedthat the entrapped cells reduced their cellcovering and pyrenoid size compared withthe normal free-living cells afterlong-term storage. The algal beads werealso applied to feed and water qualitycontrol in clam cultures' leading to amarked decrease in ammonium concentrations.Algal cells escaped from the beads provideda food source for the clams. This mightreduce the cost of clam culture compared totraditional culture methods. Therefore,immobilized I. galbana can be usedfor long-term preservation of algal stockin the laboratory and applied practicallyto clam cultures.  相似文献

The freshwater green microalga Parietochloris incisa is the richest known plant source of the polyunsaturated fatty acid (PUFA), arachidonic acid (20:4omega6, AA). While many microalgae accumulate triacylglycerols (TAG) in the stationary phase or under certain stress conditions, these TAG are generally made of saturated and monounsaturated fatty acids. In contrast, most cellular AA of P. incisa resides in TAG. Using various inhibitors, we have attempted to find out if the induction of the biosynthesis of AA and the accumulation of TAG are codependent. Salicylhydroxamic acid (SHAM) affected a growth reduction that was accompanied with an increase in the content of TAG from 3.0 to 6.2% of dry weight. The proportion of 18:1 increased sharply in all lipids while that of 18:2 and its down stream products, 18:3omega6, 20:3omega6 and AA, decreased, indicating an inhibition of the Delta12 desaturation of 18:1. Treatment with the herbicide SAN 9785 significantly reduced the proportion of TAG. However, the proportion of AA in TAG, as well as in the polar lipids, increased. These findings indicate that while there is a preference for AA as a building block of TAG, the latter can be produced using other fatty acids, when the production of AA is inhibited. On the other hand, inhibiting TAG construction did not affect the production of AA. In order to elucidate the possible role of AA in TAG we have labeled exponential cultures of P. incisa kept at 25 degrees C with [1-14C]arachidonic acid and cultivated the cultures for another 12 h at 25, 12 or 4 degrees C. At the lower temperatures, labeled AA was transferred from TAG to polar lipids, indicating that TAG of P. incisa may have a role as a depot of AA that can be incorporated into the membranes, enabling the organism to quickly respond to low temperature-induced stress.  相似文献

The cryopreservation of algae could prevent genetic drift and minimize labor costs compared to the current method of maintenance and subculturing. Clear, simple protocols for cryopreservation of marine microalga, Nannochloropsis oculata were developed and cryoprotectant choice and concentration optimized. The viability of the microalga was assessed directly after thawing, and algal concentration was measured after 2-30 days of growth. Five cryoprotectants (dimethyl sulphoxide, Me2SO; ethylene glycol, EG; glycerol, Gly; methanol, MeOH; and propylene glycol, PG) at five concentrations (10, 20, 30, 40, and 50%; v/v) were evaluated to determine the toxicity of various cryoprotectants to N. oculata. The toxicity of cryoprotectant (Me2SO, EG, MeOH, and PG) was observed only at higher concentrations of CPAs: > 20% for EG, > 30% for Me2SO and methanol, and > 40% for PG. Direct freezing of algae in liquid nitrogen resulted in a severe loss of viability and a modified cryopreservation protocol proved to be more appropriate for the preservation of N. oculata. Cryopreservation protocols developed and tested in the present study might be applied to cryopreserving other strains, or species, in this genus.  相似文献

Life history studies were carried out with isolates of Boergesenia forbesii (from Western Australia), Valonia fastigiata (Hawaii) and V. utricularis (Canary Islands, Mediterranean Sea) cultivated under laboratory conditions. Ploidy levels of nuclei were identified by micro spectrophotometric DNA content measurements after Feulgen staining. Fundamentally, the life history in both genera is diplohaplontic. Autodiploidization phenomena have been observed favoring the diploid generation. In contrast to other genera of the Siphonocladales s. str. producing biflagellate diploid zoospores (e.g. Ernodesmis, Boodlea), Valonia develops haploid and diploid zoospores with four flagella. No diploid zoospores were observed in Boergesenia.  相似文献

The effects of three organic compounds were tested on one of the most used marine micro-algae in the aquaculture of molluscs and crustaceans, Tetraselmis suecica. Studies were made in axenic conditions with yeast extract, peptone and glucose added to the culture medium, each alone, in combinations of two or all together. Medium without any organic compound was used for the control. Cultures containing yeast extract grew best, reaching maximum cell density of 3.79 × 106 and 3.84 × 106 cells ml−1. The organic carbon source affected the biochemical composition. The components most affected were the carbohydrates, with values between 6.5 pg cell−1 in control cultures and 48.5 pg cell−1 in glucose cultures. Protein content ranged between 27.5 pg cell−1 in control cultures and 88.6 pg cell−1 in yeast + glucose + peptone cultures. The lipid content changed little. Maximum protein yields were reached in cultures with yeast + glucose and with yeast - glucose - peptone, with values of 24.6 and 28.2 mg 1−1 d−1, respectively. These values are 22 and 25 times those in control cultures. A maximum carbohydrate yield of 7.9 mg carbohydrate per litre per day was obtained in yeast + glucose + peptone cultures, 27 times that in the control cultures. The maximum lipid yield was obtained with yeast + glucose + peptone and yeast + glucose. Maximum energy values were 308 kcal 1 in yeast extract - glucose - peptone cultures and 279 kcal 1−1 in yeast extract + glucose cultures. Gross energy values in control cultures were 24.5 kcal 1−1, but peptone cultures presented the minimum energy value, 22 kcal 1−1. The yeast extract: glucose ratio in the culture medium was optimized. A ratio 2:1 produced the best yields in cells, protein, carbohydrate and gross energy.  相似文献

Use of light, transmission, and scanning electronmicroscopes revealed that the epidermal cell wall ofthe red algal agarophytes Gracilaria tikvahiaeMcLachlan and G. cornea J. Agardh consists of adecklamelle and outer and inner wall layers. The twospecies differed, with G. cornea having asignificantly thicker outer wall and a more diffusedecklamelle. After induction, the zooids of Ulvalactuca would attach to glass slides and the twospecies of Gracilaria via an adhesion pad. Within a few days, 3–5 celled germlings penetrated thedecklamelle and outer wall layer of both basiphytes. By the time the epiphyte germlings reached the 15celled stage, they had penetrated the inner walllayer. The differences in epidermal cell wallconstruction between the two basiphytes may play arole in the ability of zooids of U. lactuca toattach in nature where epiphytization of G.cornea is infrequent.  相似文献

Summary The absolute configuration of the flagellar apparatus inPyramimonas gelidicola McFadden et al. has been determined and shows identity withP. obovata, indicating that they are closely related. Comparison with the flagellar apparatus of quadriflagellate zoospores from the more advancedChlorophyceae suggest thatPyramimonas may be a primitive ancestral form. The microtubular cytoskeleton has been examined in detail and is shown to be unusual in that it does not attach to the flagellar apparatus. Cytoskeletal microtubules are nucleated individually, and this is interpreted as an adaptation to the methods of mitosis and scale deployment. In view of the primitive nature of these processes, it is proposed that this type of cytoskeletal organization may represent a less advanced condition than that of the flagellar root MTOCs (microtubule organizing centers) observed in theChlorophyceae.  相似文献
