In the strictly aerobic, gram-negative bacterium Vitreoscilla strain C1, oxygen-limited growth conditions create a more than 50-fold increase in the expression of a homodimeric heme protein which was recognized as the first bacterial hemoglobin (Hb). The recently determined crystal structure of Vitreoscilla Hb has indicated that the heme pocket of microbial globins differs from that of eukaryotic Hbs. In an attempt to understand the diverse functions of Hb-like proteins in prokaryotes, we have cloned and characterized the gene (vgb) encoding an Hb-like protein from another strain of Vitreoscilla, V. stercoraria DW. Several silent changes were observed within the coding region of the V. stercoraria vgb gene. Apart from that, V. stercoraria Hb exhibited interesting differences between the A and E helices. Compared to its Hb counterpart from Vitreoscilla strain C1, the purified preparation of V. stercoraria Hb displays a slower autooxidation rate. The differences between Vitreoscilla Hb and V. stercoraria Hb were mapped onto the three-dimensional structure of Vitreoscilla Hb, which indicated that the four changes, namely, Ile7Val, Ile9Thr, Ile10Ser, and Leu62Val, present within the V. stercoraria Hb fall in the region where the A and E helices contact each other. Therefore, alteration in the relative orientation of the A and E helices and the corresponding conformational change in the heme binding pocket of V. stercoraria Hb can be correlated to its slower autooxidation rate. In sharp contrast to the oxygen-regulated biosynthesis of Hb in Vitreoscilla strain C1, production of Hb in V. stercoraria has been found to be low and independent of oxygen control, which is supported by the absence of a fumarate and nitrate reductase regulator box within the V. stercoraria vgb promoter region. Thus, the regulation mechanisms of the Hb-encoding gene appear to be quite different in the two closely related species of Vitreoscilla. The relatively slower autooxidation rate of V. stercoraria Hb, lack of oxygen sensitivity, and constitutive production of Hb suggest that it may have some other function(s) in the cellular physiology of V. stercoraria DW, together with facilitated oxygen transport, predicted for earlier reported Vitreoscilla Hb.  相似文献

The activation of DnaA protein by cardiolipin is inhibited by fluphenazinein vitro. We therefore examined the sensitivity of temperature-sensitivednaA mutants ofEscherichia coli to fluphenazine and other phenothiazine derivatives. Among the eightdnaA mutants tested,dnaA5, dnaA46 dnaA602, anddnaA604, mutants with mutations in the putative ATP binding site of DnaA protein, showed higher sensitivities to phenothiazine derivatives than did the wild-type strain. ThednaA508 anddnaA167 mutants, which have mutations in the N-terminal region of DnaA protein, also showed higher sensitivities to phenothiazine derivatives. On the other hand, thednaA204 anddnaA205 mutants, with lesions in the C-terminal region of the DnaA protein, showed the same sensitivity to phenothiazine derivatives as the wild-type strain. Isoelectric focusing in the ultranarrow immobilized (7.1–7.5) pH gradient (IPG) of hemoglobin and high-performances liquid chromatography (HPLC) of globin chains were used to investigate Hb polymorphism in Italian river buffalo. Six different phenotypes, each characterized by two or four different Hbs, were detected by IPG, whereas two differentIIα-globin chains were separated from two differentIα-chains by HPLC. Two α-chains (Iα1 andIIα3), and Hbs with similar mobilities (Hb1 andHb3), were associated with the AA Hb phenotype: two α-chains (Iα2 andIIα4), and Hbs with different mobilities (Hb2 andHb4), were associated with the BB phenotype: two sets of doublet Hbs were associated with the AB phenotype, thus suggesting allelic polymorphisms at the two α loci. An allele at the β locus is responsible for increasing to as many as eight the number of different Hbs, thus further complicating the notable Hb polymorphism of the river buffalo.  相似文献

We have studied the erythrocyte enzyme phosphofructokinase (PFK) from two strains of Long-Evans rats with genetically determined differences in erythrocyte 2,3-diphosphoglycerate (DPG) levels. The DPG difference is due to two alleles at one locus. With one probable exception, the genotype at this locus is always associated with the hemoglobin (Hb) electrophoretic phenotype, due to a polymorphism at the III-globin locus. The enzyme PFK has been implicated in the DPG difference because glycolytic intermediate levels suggest that this enzyme has a higher in vivo activity in High-DPG strain rats, although the total PFK activity does not differ. We report here that partially purified erythrocyte PFK from Low-DPG strain cells is inhibited significantly more at physiological levels of DPG (P<0.01) than PFK from High-DPG strain erythrocytes. Citrate and adenosine triphosphate also inhibit the Low-DPG enzyme more than the High-DPG enzyme. Therefore, a structurally different PFK, with a greater sensitivity to inhibitors, may explain the lower DPG and ATP levels observed in Low-DPG strain animals. TheEscherichia coli genesdicF anddicB encode division inhibitors, which prevent the synthesis and activity, respectively, of the essential division protein FtsZ. A mutation at the C-terminal end of the RNA polymeraseβ subunit renders cells resistant to both inhibitors. In the mutant strain the level of theftsZ gene product is higher than in the wild type. Disruption ofrpoS, which encodes the stationary phase sigma factor σS, lowers FtsZ protein levels in the mutant, and partially restores sensitivity to the inhibitors.  相似文献

Trichogramma cordubensis n. sp. is a parasite ofHeliothis armigera Hb. eggs in cotton crops in western Andalusia. Reproduced onEphestia kuehniella Zeller eggs, it is described morphologically and compared withT. chilonis Ishii andT. evanescens Westwood as both present similarities with this species.T. cordubensis can be distinguished in the male genitalia by the shape of the dorsal expansion of the gonobase and the size, shape and relative position of the median ventral projection. Male antennal setae are longer inT. cordubensis than in the other 2 species. The aim of the eurrent study is to investigate the therapeutic and preventive effects of 1α, 25dihydroxyvitaminD3 (1,25 (OH)2 D3) andAjuga iva (AI) extraet on diabetes toxicity in rats testes. Thus diabetic rats were treated with 1α, 25dihydroxyvitaminD3 orAjuga iva extract as both therapeutie and preventive treatments on diabetes toxicity in rats testes. Our results showed that diabetes indueed a decrease in testosterone and 17β-estradiol levels in testes and plasma. Besides, a fall in testicular antioxidant capacity appeared by a deerease in both antioxidant (superoxide dismutase (SOD), eatalase (CAT) and glutathione peroxidase (GPx) activities) and non-enzymatic antioxidant (copper (Cu), magnesium (Mg) and iron (Fe) levels). All theses changes enhanced testicular toxicity (inerease in testicular aspartate amino transaminase (AST), alanine amino transaminase (ALT), laetate dehydrogenase (LDH) activities and the lipid peroxidation and triglyeeride (TG) levels). In addition, a decrease in testicular total cholesterol (TCh) level was observed in diabetic rats testes. All the ehanges lead to a decrease in the total number and mobility of epididymal spermatozoa. The administration of 1α,25dihydroxyvitaminD3 andAjuga iva extract three weeks before and after diabetes induetion interfered and prevented diabetes toxicity in the reproduetive system. 1,25 (OH)2 D3 andAjuga iva extract blunted all changes observed in diabetic rats. To sum up, the data suggested that 1,25 (OH)2 D3 andAjuga iva extract have a protective effect on alloxan-induced damage in reproductive system by enhancing the testosterone and 17β-estradiol levels, consequenty protecting from oxidative stress, cellular toxicity and maintaining the number and motility of spermatozoids.  相似文献

Staphylococcus aureus is a leading cause of life-threatening infections in the United States. It requires iron to grow, which must be actively procured from its host to successfully mount an infection. Heme-iron within hemoglobin (Hb) is the most abundant source of iron in the human body and is captured by S. aureus using two closely related receptors, IsdH and IsdB. Here we demonstrate that each receptor captures heme using two conserved near iron transporter (NEAT) domains that function synergistically. NMR studies of the 39-kDa conserved unit from IsdH (IsdHN2N3, Ala326–Asp660) reveals that it adopts an elongated dumbbell-shaped structure in which its NEAT domains are properly positioned by a helical linker domain, whose three-dimensional structure is determined here in detail. Electrospray ionization mass spectrometry and heme transfer measurements indicate that IsdHN2N3 extracts heme from Hb via an ordered process in which the receptor promotes heme release by inducing steric strain that dissociates the Hb tetramer. Other clinically significant Gram-positive pathogens capture Hb using receptors that contain multiple NEAT domains, suggesting that they use a conserved mechanism.  相似文献

BackgroundExcessive exposure to iron can cause kidney damage, and chelating drugs such as deferoxamine and deferiprone have limited usefulness in treating iron poisoning. This study was designed to investigate the protective effects of grape seed proanthocyanidins (GSPAs) against iron overload induced nephrotoxicity in rats. The roles of GSPAs in chelating iron, antioxidant activity, renal function, pathological section, and apoptosis-related gene expression were assessed.MethodsNewly weaned male Sprague–Dawley rats aged 21 days (weight, 65 ± 5 g) were randomly divided into four groups containing 10 rats each: normal control (negative) group, iron overload (positive) group, GSPAs group, and GSPAs + iron overload (test) group. Iron dextran injections (2.5 mg⋅ kg−1) and GSPAs (25 mg⋅ kg−1) were intraperitoneally and intragastrically administered to rats daily for 7 weeks, respectively. Measurements included red blood cell (RBC) count and hemoglobin (Hb) level, serum total iron-binding capacity (TIBC), renal iron content, glutathione peroxidase (GSH-Px) activity, superoxide dismutase (SOD) activity, malondialdehyde (MDA) content, total antioxidant activity (T-AOC), creatinine (CR) and blood urea nitrogen (BUN) levels, pathological changes, and apoptotic Fas, Bax expressions in the kidney tissue. Differences among the dietary groups were determined using one-way analysis of variance with post-hoc Tukey’s test. P < 0.05 was considered statistically significant.ResultsRBC count, Hb level, renal iron content, MDA content, CR and BUN levels, and Fas, Bax expressions significantly increased in the positive group than in the negative group; contrarily, TIBC, GSH-Px activity, and T-AOC significantly decreased in the positive group than in the negative group (P < 0.05). Although not statistically significant, SOD activity was slightly reduced in the positive group than in the negative group. Inflammatory cell infiltration and fibrous tissue proliferation were observed in the kidney tissue of the rats in the positive group; in contrast, the rats exhibited better recovery when GSPAs were used instead of iron alone. Compared with the positive group, RBC counts, Hb levels, renal iron contents, the MDA content, CR and BUN levels, and Fas, Bax expressions significantly decreased, whereas the TIBC, the GSH-Px and SOD activities as well as T-AOC significantly increased in the test group rats (P < 0.05). There were no significant differences in the RBC counts, Hb levels, TIBC, renal iron contents, the SOD activity and MDA content, CR and BUN levels, and Fas expression between the GSPAs and negative groups. The GSH-Px activity and T-AOC were significantly increased whereas Bax expression was significantly decreased in the GSPAs group rats than in the negative group rats (P < 0.05). The rats in the GSPAs, test, and negative groups displayed glomeruli and tubules with a clear structure; further, the epithelial cells in the renal tubules were neatly arranged.ConclusionsGSPAs have protective effects on nephrotoxicity in rats with iron overload. Thus, further investigation of GSPAs as a new and natural phytochemo-preventive agent against iron overload is warranted.  相似文献

TheIf-1 alleles of many inbred strains, some of common parentage with either BALB/c Gif or C57BL/Lac, were determined. Of the 23 inbred lines examined, all were eitherIf-1 h orIf-1 l , except the C57BR/cdJ strain, which gave intermediate results; the latter will have to be explored more thoroughly before the existence of a third allele can be established. Survey of the 23 lines showed no correlation betweenH-2 haplotypes andIf-1 alleles. The absence of linkage betweenH-2 andIf-1 was confirmed by typing (BALB/c×C57BL)F1×C57BL backcross progeny for bothH-2 andIf-1. The fact that some high and some low producers were of sameH-2 type made it possible to study the production of interferon in radiation chimeras. Mice of a low-producer strain, C3H/Lac (If-1 l , were lethally irradiated and their hemopoietic function restored by grafting bone marrow from eitherIf-1 l orIf-1 h mice, all donors of the sameH-2 haplotype as that of the recipients (H-2 k . NDV-induced serum interferon production was measured 31 days after irradiation and restoration. The production of interferon in all mice restored with marrow fromIf-1 l donors was equal or lower than that of syngeneic chimeras (alsoIf-1 l ). In contrast, titers of interferon in all four groups restored withIf-1 h marrow were higher than those of control chimeras, three being higher than titers in unirradiatedIf-1 l controls. These data indicate that theIf-1 locus is expressed through cells derived from hemopoietic stem cells, possibly the interferon-producing cells themselves.  相似文献

Here we report that the bacterial catabolism of 4-hydroxy-3,5-dimethylbenzoic acid 1 takes a different course inRhodococcus rhodochrousN75 andPseudomonassp. strain HH35. The former organism accumulates a degradation metabolite of the acid which we isolated and identified as 2,6-dimethylhydroquinone 2. The latter bacterial strain converts the acid and the hydroquinone into a dead-end metabolite. This novel compound was characterised unequivocally by mass spectrometry and1H and13C NMR and UV spectroscopy as 4-acetonyl-4-hydroxy-2-methylbut-2-en-1,4-olide 4, a cyclic tautomer of (3-methylmaleyl)acetone, which exists as the enol carboxylate form 3 in aqueous solution.  相似文献

The upr-I gene controlling UV sensitivity in Neurospora crassa has been transferred from a microconidial strain into a macroconidial strain. The properties of the strain with respect to UV sensitivity and photoreactivation (PR) have been modified in the macroconidial background leading to an enhancement of resistance to UV by a factor of about 2 and some suggestion of enzymatic PR. The upr-I gene confers sensitivity to nitrous acid to about the same degree as that for UV. However, inactivation of strains carrying the upr-I gene by white light in the presence of photosensitizing dyes is equivalent to that of strains exhibiting normal sensitivity to UV.  相似文献

The effect of intermittent hypoxia (IHx) on blood hemoglobinconcentration ([Hb]) and the underlying mechanisms werestudied in rats exposed to 10%O2, 1 h/day, for up to 5 wk. IHxprotocols with longer daily hypoxic exposure show persistentpolycythemia; however, it is unknown whether [Hb] increasestransiently during hypoxia in protocols without polycythemia. Hypoxiaproduced a reversible [Hb] increase after 4 days of IHx butnot in normoxic controls (NxC) or after shorter period of IHx.Splenectomy abolished the phenomenon. Plasma epinephrine andnorepinephrine levels during hypoxia were comparable in IHx and NxCgroups, but the epinephrine-induced [Hb] increase waslarger in IHx. The 1- and2-adrenoreceptor blockade(phentolamine) and 2-blockade(yohimbine) abolished the [Hb] increase of IHx rats.Conversely, 2-receptorstimulation (oxymetazoline) increased [Hb] during normoxiain IHx but not in NxC. In conclusion, this IHx protocol results inreversible [Hb] increases during hypoxia via spleniccontraction mediated by increased2-adrenoreceptor response. Thismay protect O2 supply duringhypoxia without the cardiovascular burden of polycythemia during normoxia.

The streptomycete strain ETH 7437, producing granaticin, was characterized according to present taxonomic standards and compared with the strainsStreptomyces olivaceus (Waksman) Waksman and Henrici,Streptomyces coelicolor (Müller) andStreptomyces violaceruber (Waksman & Curtis). The strain ETH 7437 formed gray aerial mycelium, straight and flexuos sporophores, even single hooks and open spirals. Normally the amount of spores in chains was over 50, the surface of spores being smooth. The strain utilizedd-glucose,d-arabinose,d-xylose andd-fructose. The strain was capable of forming antibiotic pigment stably. For the strain ETH 7437 the termStreptomyces granaticolor was proposed.  相似文献

In the luminal contents of metronidazole-treated rats, there was a dominant Bifidobacterium species. A strain has been isolated, its 16S rRNA gene has been sequenced, and the strain has been named Bifidobacterium pseudolongum strain Patronus. In this study, using an experimental model of healthy rats, the effects of metronidazole treatment and B. pseudolongum strain Patronus administration on the luminal and mucosa-associated microbiota and on gut oxidation processes were investigated. Metronidazole treatment and the daily gavage of rats with B. pseudolongum strain Patronus increased the numbers of bifidobacteria in cecal contents and in cecal mucosa-associated microbiota compared with those in control rats. Metronidazole reduced the colonic oxidative damage to proteins. This is the first evidence that B. pseudolongum strain Patronus exerts an effect on a biomarker of oxidative damage by reducing the susceptibility to oxidation of proteins in the colon and the small bowel. Antioxidant effects of metronidazole could be linked to the bifidobacterial increase but also to other bacterial modifications.  相似文献

A new autosomal codominant hemoglobin mutation alters hemoglobin M of the primitive red cell line and hemoglobin D found in definitive cells. That Hb M and Hb D are altered by the same gene mutation supports the idea that Hb M shares a polypeptide chain with Hb D. It is concluded that in the switch from primitive hemoglobins to those of the definitive type, there are at least two α chains conserved; αA of Hb E in Hb A and αD of Hb M in Hb D.  相似文献
