The presented article highlights the process of biofuel production with a special focus on bioethanol. After a short introduction to the “problems” of biofuels – the “first generation” biofuels (in regards to their competition to feed and food production) and the “second generation” biofuels (in regards to the required more complex process technology) − the different steps in the process from natural resources towards the final product are presented and the underlying biotechnological challenges discussed: the pre-treatment of the natural resources followed by the biotechnological processes of hydrolysis and fermentation. Topics such as enzyme screening for efficient or even multi-step hydrolysis as well as microbial strain selection under process conditions and the optimization of the anaerobic fermentative conversion of the saccharides to bioethanol are discussed. Optimizing the production of bioethanol to be competitive with petrochemical fuels is the main challenge for the underlying process development.  相似文献

Reactive oxygen species – superoxide, hydrogen peroxide and hydroxyl radicals – have long been suspected of constraining bacterial growth in important microbial habitats and indeed of shaping microbial communities. Over recent decades, studies of paradigmatic organisms such as Escherichia coli, Salmonella typhimurium, Bacillus subtilis and Saccharomyces cerevisiae have pinpointed the biomolecules that oxidants can damage and the strategies by which microbes minimize their injuries. What is lacking is a good sense of the circumstances under which oxidative stress actually occurs. In this MiniReview several potential natural sources of oxidative stress are considered: endogenous ROS formation, chemical oxidation of reduced species at oxic–anoxic interfaces, H2O2 production by lactic acid bacteria, the oxidative burst of phagocytes and the redox-cycling of secreted small molecules. While all of these phenomena can be reproduced and verified in the lab, the actual quantification of stress in natural habitats remains lacking – and, therefore, we have a fundamental hole in our understanding of the role that oxidative stress actually plays in the biosphere.  相似文献

Insects are often associated with symbiotic micro‐organisms, which allow them to utilize nutritionally marginal diets. Adult fruit flies (Diptera: Tephritidae) associate with extracellular bacteria (Enterobacteriaceae) that inhabit their digestive tract. These flies obtain nutrients by foraging for plant exudates, honeydew and bird droppings scattered on leaves and fruit – a nutritional niche which offers ample amounts of carbohydrates, but low quantities of available nitrogen. We identified the bacteria resident in the gut of the olive fly (Bactrocera oleae) – a worldwide pest of olives and examined their contribution to nitrogen metabolism in the adult insect. By suppressing bacteria in the gut and monitoring female fecundity, we demonstrate that bacteria contribute essential amino acids and metabolize urea into an available nitrogen source for the fly, thus significantly elevating egg production. In an ecological context, bacteria were found to be beneficial to females subsisting on bird droppings, but not on honeydew – two natural food sources. We suggest that a main gut bacterium (Candidatus Erwinia dacicola) forms an inseparable, essential part of this fly's nutritional ecology. The evolution of this symbiosis has allowed adult flies to utilize food substrates which are low or imbalanced in assimilable nitrogen and thereby to overcome the nitrogen limitations of their natural diet.  相似文献

This article examines the selective use of plant materials in design in Melanesia. It explores – through an analysis of pandanus leaf mats in New Ireland, Papua New Guinea – how makers select fibres on the basis of their capacity to express social relations to varying temporalities before their natural decay. J.J. Gibson's theory of affordance and Donald Norman's concept of mapping are critically applied for this purpose. This approach emphasizes how social and temporal relations are condensed into objects, and refocuses anthropological attention towards materials and their affordances in the production of material culture.  相似文献

A combination of founder effects and local adaptation – the Monopolization hypothesis – has been proposed to reconcile the strong population differentiation of zooplankton dwelling in ponds and lakes and their high dispersal abilities. The role genetic drift plays in genetic differentiation of zooplankton is well documented, but the impact of natural selection has received less attention. Here, we compare differentiation in neutral genetic markers (FST) and in quantitative traits (QST) in six natural populations of the rotifer Brachionus plicatilis to assess the importance of natural selection in explaining genetic differentiation of life‐history traits. Five life‐history traits were measured in four temperature × salinity combinations in common‐garden experiments. Population differentiation for neutral genetic markers – 11 microsatellite loci – was very high (FST = 0.482). Differentiation in life‐history traits was higher in traits related to sexual reproduction than in those related to asexual reproduction. QST values for diapausing egg production (a trait related to sexual reproduction) were higher than their corresponding FST in some pairs of populations. Our results indicate the importance of divergent natural selection in these populations and suggest local adaptation to the unpredictability of B. plicatilis habitats.  相似文献

The relationship between farming management and the overexploitation of natural resources is often a theme of discussion in the environmental sciences. Moreover, farmers’ choices – driven by consumer demand – have a significant effect on the agricultural production system.The Ecological Footprint methodology as it currently implemented assumes that all cropland activities are sustained by the capacity of the ecosystem, basing both demand and capacity calculations on the exact same flow accounting. This causes some confusion in the evaluation of the ecological performance of farming because it appears that this activity has no consequences on the planet.This paper proposes a solution to this duality caused by the current methodological assumption about croplands, and investigates the influence of different farming techniques on Ecological Footprint results. Starting from the concept of an embodied footprint in production, we propose a new approach for the evaluation of farming performance. This approach permits an estimation of the impact of farming activity, linked to the farmers’ technique, and a calculation of the crop Footprint in reference to the production capacity of the natural system.Building on the central methodology of the Ecological Footprint, we provide a different evaluation system and show case study results for comparison.  相似文献

Some microbial public goods can provide both individual and community‐wide benefits, and are open to exploitation by non‐producing species. One such example is the production of metal‐detoxifying siderophores. Here, we investigate whether conflicting selection pressures on siderophore production by heavy metals – a detoxifying effect of siderophores, and exploitation of this detoxifying effect – result in a net increase or decrease. We show that the proportion of siderophore‐producing taxa increases along a natural heavy metal gradient. A causal link between metal contamination and siderophore production was subsequently demonstrated in a microcosm experiment in compost, in which we observed changes in community composition towards taxa that produce relatively more siderophores following copper contamination. We confirmed the selective benefit of siderophores by showing that taxa producing large amounts of siderophore suffered less growth inhibition in toxic copper. Our results suggest that ecological selection will favour siderophore‐mediated decontamination, with important consequences for potential remediation strategies.  相似文献

We used a multistrain approach to study the intra‐ and interspecific variability of the growth rates of three Pseudo‐nitzschia species – P. australis, P. fraudulenta, and P. pungens – and of their domoic acid (DA) production. We carried out mating and batch experiments to investigate the respective effects of strain age and cell size, and thus the influence of their life cycle on the physiology of these species. The cell size – life cycle relationship was characteristic of each species. The influence of age and cell size on the intraspecific variability of growth rates suggests that these characteristics should be considered cautiously for the strains used in physiological studies on Pseudo‐nitzschia species. The results from all three species do not support the hypothesis of a decrease in DA production with time since isolation from natural populations. In P. australis, the cellular DA content was rather a function of cell size. More particularly, cells at the gametangia stage of their life cycle contained up to six times more DA than smaller or larger cells incapable of sexual reproduction. These findings reveal a link between P. australis life cycle and cell toxicity. This suggest that life cycle dynamics in Pseudo‐nitzschia natural populations may influence bloom toxicity.  相似文献

The leaves of Catharanthus roseus (L.) G. Don produce the first natural drugs used in cancer therapy – the dimeric terpenoid indole alkaloids vinblastine and vincristine. The study of C. roseus further revealed two other terpenoid indole alkaloids with important pharmacological activity: ajmalicine, used as an antihypertensive, and serpentine, used as sedative. The biosynthetic pathway of the medicinal alkaloids has been investigated in much detail and a number of steps are now well characterized at the enzyme and gene level and, recently, several regulatory genes have also been isolated and characterized. Since early studies of the biosynthesis of vinblastine, during the 1970s and 1980s, the dimerization reaction has attracted much attention due to its possible regulatory importance and potential application for the semi synthetic production of the dimeric alkaloids. After initial, inconclusive work suggesting the involvement of peroxidase-like enzymes, the search for the dimerization enzyme in leaf tissue detected a single dimerization activity credited to the single class III plant peroxidase present in the leaves of the plant – the basic isoenzyme CRPRX1. The enzyme was purified to homogeneity, the respective cDNA and genomic sequences were characterized, and a channeling mechanism was proposed for the peroxidase-mediated-vacuolar synthesis of the first dimeric alkaloid intermediate, α-3′,4′-anhydrovinblastine. On the other hand, the oxidation of ajmalicine into serpentine has been attributed to basic peroxidase isoenzymes localized in the vacuole of C. roseus cells. An overview of the work implying class III plant peroxidases in the biosynthesis of terpenoid indole alkaloids in C. roseus is presented here. Most bioassessment programs in Brazil face difficulties when scaling up from small spatial scales because larger scales usually encompass great environmental variability. Covariance of anthropogenic pressures with natural environmental gradients can be a confounding factor in the evaluation of biologic responses to anthropogenic pressures. The objective of this study was to develop a multimetric index (MMI) with macroinvertebrates for two stream types and two ecoregions in the Atlantic Forest biome in Rio de Janeiro state, Brazil. We hypothesized that by using two approaches – (1) testing and adjusting metrics to landscape parameters, and (2) selecting metrics using a cluster analysis to avoid metrics redundancy – the final MMI would perform better than the traditional approach (unadjusted metrics, one metric representing each category). Four MMIs were thus developed: MMI-1 – adjusted MMI with metrics selected after cluster analysis); MMI-2 – adjusted MMI with one metric from each category; MMI-3 – unadjusted MMI with metrics selected after cluster analysis; MMI-4 – unadjusted MMI with one metric from each category. We used three decision criteria to assess MMI’s performance: precision, responsiveness and sensitivity. In addition, we tested the MMI’s by using an independent set of sites to validate the results. Although all MMIs performed well in the three criteria, adjusting metrics to natural variation increased MMI response and sensitivity to impairment. In addition, the selected MMI-2 was able to classify sites of two stream types and two ecoregions. The use of cluster analysis, however, did not avoid high redundancy between metrics of different branches. The MMI-4 had the poorest performance among all tested MMIs and it was not able to distinguish adequately reference and impaired sites from both ecoregions. We present some considerations on the use of metrics and on the development of MMI’s in Brazil and elsewhere.  相似文献

This study used neutron diffraction to investigate a ceramide-[NP] C24/[AP] C24 /[EOS]-br C30/cholesterol/lignoceric acid (0.6: 0.3: 0.1: 0.7: 1) based stratum corneum modelling system. By adding specifically deuterated ceramides-[NP]-D3, [AP]-D3, and [EOS]-br-D3, detailed information on the lamellar and the nanostructure of the system was obtained. For the short periodicity phase a natural-like lamellar repeat distance of 5.47?±?0.02?nm was observed, similar to the [NP]/[AP] base system without the [EOS]-br. Unlike in this system the ceramides here were slightly tilted, hinting towards a slightly less natural arrangement. Due to the deuteration it was possible to observe that the long ceramide chains were overlapping in the lamellar mid-plane. This is considered to be an important feature for the natural stratum corneum. Despite the presence of a ceramide [EOS] analogue – able to form a long phase arrangement – no distinct long periodicity phase was formed, despite a slightly higher than natural ω-acyl ceramide ratio of 10?mol%. The deuterated variant of this ceramide determined that the very long ceramide was integrated into the short periodicity phase, spanning multiple layers instead. The – compared to the base system – unchanged repeat distance highlights the stability of this structure. Furthermore, the localisation of the very long ceramide in the short periodicity phase indicates the possibility of a crosslinking effect and thus a multilayer stabilizing role for the ceramide [EOS]. It can be concluded, that additionally to the mere presence of ceramide-[EOS] more complex conditions have to be met in order to form this long phase. In this paper two methods for energy analysis and environmental accounting (Gross Energy Requirement and Emergy Synthesis) are critically discussed in order to explore their ability to provide a comprehensive evaluation of the performance and environmental sustainability of human-dominated production processes. In order to allow a quantitative comparison, two cropping systems, namely 1 ha of corn production in Italy, and 1 ha of willow production in Sweden, are investigated by means of the parallel application of both methods. The case studies are carried out by performing a quantitative inventory of both natural and economic input flows to the investigated cropping systems. Such input flows are then converted into embodied energy (MJ) as well as emergy (seJ) units. Finally, performance indicators representative for each method are calculated. Results provided by the two methods and their respective theoretical features are compared and discussed in order to point out limits and potentialities of both approaches. The study shows that the two methods account for different – although complementary – categories of input flows, use different conversion factors, and answer to different questions and concerns. Gross Energy Requirement focuses on fossil fuel use and is capable to support the development of more efficient use of commercial energy. Emergy Synthesis uses broader spatial and time frames and accounts for both natural and economic resources. In so doing, it takes into consideration different forms of energy, materials, human labor and economic services on a common basis, offering larger potentiality to explore the sustainable interplay of environment and economy.  相似文献

Question: How do studies of the distribution of genetic diversity of species with different life forms contribute to the development of conservation strategies? Location: Old‐growth forests of the southeastern United States. Methods: Reviews of the plant allozyme literature are used to identify differences in genetic diversity and structure among species with different life forms, distributions and breeding systems. The general results are illustrated by case studies of four plant species characteristic of two widespread old‐growth forest communities of the southeastern United States: the Pinus palustris – Aristida stricta (Longleaf pine – wiregrass) savanna of the Coastal Plain and the Quercus – Carya – Pinus (Oak‐hickory‐pine) forest of the Piedmont. Genetic variation patterns of single‐gene and quantitative traits are also reviewed. Results: Dominant forest trees, represented by Pinus palustris(longleaf pine) and Quercus rubra (Northern red oak), maintain most of their genetic diversity within their populations whereas a higher proportion of the genetic diversity of herbaceous understorey species such as Sarracenia leucophylla and Trillium reliquum is distributed among their populations. The herbaceous species also tend to have more population‐to‐population variation in genetic diversity. Higher genetic differentiation among populations is seen for quantitative traits than for allozyme traits, indicating that interpopulation variation in quantitative traits is influenced by natural selection. Conclusion: Developing effective conservation strategies for one or a few species may not prove adequate for species with other combinations of traits. Given suitable empirical studies, it should be possible to design efficient conservation programs that maintain natural levels of genetic diversity within species of conservation interest.  相似文献

The mass production of SCP on the base of hydrocarbons opens the possibility to get a complex of further substances. It is distinguished between by-products – belonging to essential nonprotein – structures of the growing cell and the so – called coupled products accumulating intra – or extracellularly by means of regulation. An important principle of SCP-coupled synthesis of products is the use and the maintenance of proportionality between the growth rate and cell concentration of the producing cells and the accumulation of products. As examples are discussed: fermentation by yeasts and bacteria on the base of n-alkanes, methane and methanol in connection with products as fatty acids, poly-β-hydroxybutyric acid, citric and gluconic acid.  相似文献
