This study describes the persistence of an embryonal structure through to sub-adulthood in the ostrich and emu. Mandibles from sub-adult ostrich and emu were subjected to special staining, light microscopy and dissected to reveal and describe Meckel's cartilage. Meckel's cartilage, composed of hyaline cartilage, was present within the neurovascular canal of both species. The persistence through to sub-adulthood of Meckel's cartilage in the ostrich and emu is a feature not previously reported in any other avian species. The proximal end of Meckel's cartilage was ossified in the region of the articular bone and the distal end was ossified in some specimens. Although this structure may ossify at a much later stage in life, the function in young and sub-adult birds may be to dampen shockwaves along the intramandibular nerve that result from the action of pecking. In the ostrich, the M. pseudotemporalis superficialis tendon inserted onto the supra-angular bone and Meckel's cartilage. In the emu, a small portion of the tendon was attached to the supra-angular bone and the main part to Meckel's cartilage. The persistence of Meckel's cartilage in adult lepidosaurs, crocodilians and ratites represents an unusual shared trait between the extant members of the above groups.  相似文献

The claw closer muscle of two estuarine crab species, Cyrtograpsus angulatus and Neohelice granulata (Grapsoidea, Varunidae): histochemical fibre type composition. —Acta Zoologica (Stockholm) 00 : 1–7. This study permitted the characterization of four types of muscle fibres in the claw closer muscles of Cyrtograpsus angulatus and Neohelice granulata. Succinic dehydrogenase (SDH) for mitochondria, periodic acid Schiff (PAS) for glycogen, Sudan Black B for lipids and myosin‐adenosine triphosphatase (m‐ATPase) preincubated at alkaline and acid pHs were used for that purpose. The mean fibre diameters, the relative areas and frequencies of each muscle fibre type were calculated. Types I and IV would be considered ‘extreme’ groups with type I fibres large, weak and acid/alkaline‐labile m‐ATPase, weak SDH, PAS and Sudan, and type IV fibres small, very strong and acid/alkaline‐resistant m‐ATPase, strong SDH and PAS, and moderate Sudan. Types II and III would belong to a predominant ‘intermediate’ group. Type IV fibres were scarce in C. angulatus but represented 25% of the total fibre population in N. granulata. In C. angulatus, the relative area occupied by type I fibres was bigger than its relative proportion, whereas in N. granulata, types I and II had similar patterns. Concluding, variations in fibre type composition in the claw closer muscles of C. angulatus and N. granulata would be linked to different habitats and feeding behaviours.  相似文献

The objective of the paper is to evaluate the effect of acellular nerve allografts (ANA) seeded with Schwann cells to promote nerve regeneration after bridging the sciatic nerve defects of rats and to discuss its acting mechanisms. Schwann cells were isolated from neonatal Wistar rats. In vitro Schwann cells were microinjected into acellular nerve allografts and co-cultured. Twenty-four Wistar rats weighing 180–220 g were randomly divided into three groups with eight rats in each group: ANA seeded with Schwann cells (ANA + SCs), ANA group and autografts group. All the grafts were, respectively, served for bridging a 10-mm long surgically created sciatic nerve gap. Examinations of regeneration nerve were performed after 12 weeks by transmission electron microscope (TEM), scanning electron microscope (SEM), and electrophysiological methods, and then analyzed statistically. The results obtained indicated that in vitro Schwann cells displayed the feature of bipolar morphology with oval nuclei. Compared with ANA group, the conduction velocity of ANA + SCs group and autograft group was faster after 12 weeks, latent period was shorter, and wave amplitude was higher (P < 0.05). The difference between ANA + SCs group and autograft group is not significant (P > 0.05). Regeneration nerve myelinated fiber number, myelin sheath thickness, and myelinated fibers/total nerves (%) in both ANA + SCs group and autograft group are higher than that in ANA group; the difference is significant (P < 0.05). The difference between the former two is not significant (P > 0.05). In conclusion, ANA seeded with SCs could improve nerve regeneration and functional recovery after bridging the sciatic nerve gap of rats, which offers a novel approach for the repair peripheral nerve defect.  相似文献

The muscle fibers of the pectoralis (M. pectoralis pars thoracicus) of a male and a female ostrich (Struthio camelus) and a male and a female emu (Dromaius novaehollandiae) were studied histochemically for succinate dehydrogenase and myofibrillar adenosine triphosphatase. Slow-tonic (ST), fast-twitch oxidative-glycolytic (FOG) and fast-twitch glycolytic (FG) fibers were approximately equal in number and distribution in the emu pectoralis examined. In the ostriches, both predominantly FG and approximately equal areas, were present. ST fibers were significantly (P ≤ 0.05) larger than the similarly (P ≥ 0.05) sized FG and FOG fibers in the female ostrich and emus. In the male ostrich ST fibers were smaller (P ≤ 0.05) than FG fibers, neither of which were significantly (P ≥ 0.05) different from FOG fibers. The ratites have the greatest percentage and widest distribution of ST fibers found in any avian pectoralis studied to date. This could represent the ancestoral avian pectoralis, neoteny or an effect of flightlessness. ST fibers are used in the maintenance of posture, which is probably the main role of the pectoralis in the emu. The predominantly FG areas of the ostrich are indicative of an additional function, namely, behavioural display. Sexual dimorphism in the ostrich pectoralis is strongly suggested.  相似文献

A comparative study of the quantitative data of the frog extraocular muscles and the cranial nerves that innervate them was performed. Oculorotatory muscles contain muscle fibres of at least 4 types which are arranged in heterogeneous layers. The zonal arrangement of the muscles does not occur on the cross-sections in the vicinity of muscle insertions. In these regions only two muscle fibre types are present and the total number of fibres is smaller by 70% than in the central region of the muscle. Most numerous are muscle fibres in the rectus inferior muscle, while the smallest number of fibres is found in rectus interior muscle. Three distinct types of nerve fibres are distinguished according to the following criteria: occurrence and thickness of myelin sheath, fibre diameter and ratio "g". The fibres with thin myelin sheaths indicate small diameters (1-5--6- mum) and their ratio "g" equals 0-82 +/- 0-08. They constitute about 30% of the myelinated fibres in the nerve supply of the oculorotatory muscles and about 14% in the supply of the retractor bulbi muscle. Both the value of the ratio "g" and a greater number of these fibres in the nerve supply of the muscles that contain slow contracting muscle fibres indicate that they are rather slow conducting nerve fibres. The range of the diameters of the fibres with thick myelin sheaths is greater (3-5--13-5 mum) and their "g" equals 0-66 +/- 0-06. These fibres constitute about 70% of the myelinated ones in the nerve supply of the oculorotatory muscles and 86% in the supply of the retractor bulbi muscles. The value of the ratio "g" in these fibres indicates that they are fast contracting ones. The smallest diameters are found in the myelinated fibres (0-5--1-7 mum). These fibres occur frequently in all the examined nerves; they constitute 36--47% of the total number of all the nerve fibres. The frog extraocular muscles are characterized by an abundal nerve supply which is reflected in the low innervation ratio (1:4--1:5). On the distal cross-section of nerves the number of nerve fibres is greater than on the proximal ones. Ganglionic neurons occur sporadically around the nerves; in the nerve III synaptic contacts between two neurons were observed.  相似文献

The distribution and chemical content of cutaneous nerves in 3- to 13-day-old emu chicks (Dromaius novaehollandiae) were examined by using double-labelling immunohistochemistry. Seven different subpopulations of cutaneous nerves were identified based on their neurochemistry. No intraepidermal nerve fibres were found. However, axons were located within the dermis and were often associated with blood vessels, pennamotor muscles and feather follicles or innervated Herbst corpuscles. Both similarities and differences exist between subpopulations of cutaneous nerves in the emu and volant birds. As in volant birds, a subpopulation of cutaneous axons innervates the superficial skin layers and contains immunoreactivity to both substance P and calcitonin gene-related peptide (CGRP). This suggests that the neuropeptide content of these presumptive free nerve endings is conserved throughout the evolution of birds. In contrast, Herbst corpuscles in the emu are innervated by axons that contain immunoreactivity for CGRP or neuropeptide Y (NPY) but that lack the calbindin D-28k immunoreactivity found in fibres innervating Herbst corpuscles of volant birds. The nerves and nerve endings in the skin of tropical cattle were studied using histological and histochemical techniques. Many nerve trunks and fibres were present in the reticular and papillary dermis in both hairy and non-hairy skin sites. In non-hairy skin locations such as the muzzle and lower lip, encapsulated endings akin to Krause and Ruffini end bulbs, which arise from myelinated nerve trunks situated lower down the dermis were observed at the upper papillary layer level. Some fibre trunks seen at this level extended upwards to terminate within dermal papillae as bulb-shaped longitudinally lamellated Pacinian-type endings, while other onion-shaped lamellated nerve structures were located either within dermal papillae or near the dermo-epidermal area. Intraepidermal free-ending nerve fibres, appearing non-myelinated were observed in areas with thick epidermis. Intraepidermal free-ending nerve fibres, appearing non-myelinated were observed in areas with thick epidermis. On hairy skin sites, however, organized nerve endings or intraepidermal nerve endings were not readily identifiable.  相似文献

Morphological features of a collection of unknown-age wild kiwi (Apteryx mantelli) embryos from early development to point of hatch are described. Using these features, we assign developmental stages to each embryo and compare the progress of development to similar-staged ostrich (Struthio camelus) and chicken (Gallus gallus) embryos. Two ageing schemes for the kiwi embryos are developed by comparing measurements of their hindlimb segments, bills and crown–rump lengths with those of ostrich and chicken embryos at various stages of development. One of the 20 kiwi embryos was of known age. Both the ostrich model and the chicken model gave identical predictions for the marker and four other embryos. Developmental timing of some features differed between all three species, most markedly in the bill, with growth in the kiwi bill being relatively faster to achieve its larger relative and absolute size at hatch.  相似文献

Summary The distribution of monoamines in the hypothalamus of the Japanese quail (Coturnix coturnix japonica) has been studied using a histochemical fluorescence technique. In the posterior hypothalamus catecholamine-containing nerve fibres are localised in the nucleus tuberis and nucleus hypothalamicus posterior medialis and are linked by fluorescent tracts running in the stratum cellulare internum. Further tracts may be traced from the nucleus tuberis around the base of the third ventricle to the sub-ependymal layer of the median eminence, where they then appear to pass through the hypothalamo-hypophysial neurosecretory tract to terminate in the palisade zone on the portal vascular bed. The innervation of the palisade layer by catecholamines is sparse. The fluorescent terminals are spread evenly throughout both the anterior and posterior divisions of the median eminence. There is no monoamine innervation of the pars nervosa. The paraventricular organ has both 5-hydroxytryptamine- and catecholamine-containing cell bodies and axons may be traced into the region of the nucleus hypothalamicus posterior medialis. In the anterior hypothalamus the neurosecretory paraventricular nucleus contains many catecholamine nerve fibres and terminals. These are linked by fibre tracts to the nucleus basalis and to the nucleus hypothalamicus posterior medialis. The supraoptic nucleus is less well innervated although a dense accumulation of fibres lies in the preoptic recess. The latter is thought to give rise to long axons which pass in association with the neurosecretory tract to end in the nucleus tuberis.Supported by a Grant (AG 24/36) from The Agricultural Research Council. Chromosomes were studied in female specimens of the ostrich, Struthio camelus L., cassowary, Casuarius casuarius (L.), emu, Dromiceius novaehollandiae (Lath.) and rhea, Rhea americana L. by means of blood and feather pulp culture techniques. Male karyotypes were also studied in the emu and rhea. The diploid chromosome number was most likely 80 in the ostrich and rhea and 82 in the emu, while the exact number could not be determined in the cassowary. Karyotypes of the 4 species were strikingly similar and apparently interchangeable with one another with slight modifications of the centromeric position in one or two pairs of macrochromosomes. No heteromorphic macrochromosomal pair was found either in female specimens or in male ones of the ratite species so far examined, except for a female rhea. This specimen was found to possess an acrocentric chromosome which was evidently a member of nos. 4–6, but considerably smaller than any other chromosome of the group. 3H-thymidine autoradiography provided no more information than the straightforward morphological analysis with regard to the differentiation of the sex-chromosomes.Contributions from the Chromosome Research Unit, Hokkaido University. Central and peripheral neuronal pathways revealed by backfilling with neurobiotin in the optic, tentacular and small labial nerves of Lymnaea stagnalis. —Acta Zoologica (Stockholm) 93 : 28–47. The TOLm complex in Lymnaea stagnalis contains nerves n. tentacularis, n. opticus and n. labialis minor. Ligatures close to where the complex enters the central nervous system (CNS) did not prevent penetration of retrograde‐transported neurobiotin into fibres of an adjacent nerve. Axonal bifurcation within the common nerve trunk or tight junctions may be involved, providing a basis for peripheral axon reflexes. Peripheral terminations of n. tentacularis, n. labialis minor and n. opticus revealed numerous cell bodies in the tentacular epithelium, some in the tentacle and lip region, and some in the retina. These cell bodies’ central projections were mapped by neurobiotin and verified by dissections of the cerebro‐cerebral commissure and cerebro‐pleural connective. Afferent fibres of the nerves form dense sensory neuropils in the ipsilateral cerebral ganglia. Direct connections between n. tentacularis and some visceral as well as parietal nerves were demonstrated by backfillings through n. pallialis dexter internus et externus, n. pallialis sinister and n. intestinalis. Labelling of n. tentacularis revealed neuronal bodies in every ganglion and stained fibres in most of the peripheral nerves. Fewer neurons were identified through n. labialis minor and n. opticus. We discuss our results in relation to different behavioural forms like defence and feeding reactions in L. stagnalis.  相似文献

Summary In the present study the central innervation of the guinea-pig pineal gland was investigated. The habenulae and the pineal stalk contain myelinated and non-myelinated nerve fibres with few dense-cored and electron-lucent vesicles. Some myelinated fibres leave the main nerve fibre bundles, lose their myelin-sheaths and terminate in the pineal gland. Although direct proof is lacking, the non-myelinated fibres appear to end near the site where the bulk of the myelinated fibres are located. Here a neuropil area exists where synapses between non-myelinated fibre elements are abundant. Neurosecretory fibres were also seen. The results support the concept of functional interrelationships between hypothalamus, epithalamus and the pineal gland.  相似文献

Serial and interval electron micrograph series were used to examine the rostral and anterodorsal nerves of 12.5‐day‐old amphioxus larvae and trace selected fibres to their targets in the nerve cord. The nerves contain a variety of fibre types, including axons from at least two types of epithelial sensory cells and neurites derived from dorsal (Retzius) bipolar cells located within the cord. The rostral epithelial cells form basal synapses with a population of peripheral neurites that probably derive from the dorsal bipolar cells, though other sources are possible. Varicosities containing dense‐core vesicles occur at the tip of the rostrum, indicating the presence of efferent innervation at this site. Within the cord, some peripherally derived rostral afferents terminate at the level of the anterior cerebral vesicle, others synapse directly with both motoneurones and the notochord, but those in the largest bundle target the dendrites of the large paired neurones (LPNs) located in the primary motor centre. LPN dendrites also receive synapses from sensory fibres arriving via the anterodorsal nerves, from the anterior‐most of the dorsal bipolar cells, referred to here as tectal cells, and from a single fibre derived from the frontal eye. This convergence of multiple inputs accords with other evidence that the LPNs are key intermediaries in the sensorimotor pathway that activates the larval escape response. The rostral nerves are much larger at metamorphosis, but the ventral tracts that derive from them are still comparatively small. This is because the majority of rostral fibres are diverted into a late‐developing dorsal tract that travels within the cord to the front end of the dorsolateral neuropile, where most of its fibres disperse and form synapses. The positioning of the dorsal and ventral tracts strongly suggests homology with vertebrate olfactory and terminal nerves, respectively. This, and the question of whether the amphioxus central nervous system has anything comparable to the olfactory bulb, a telencephalic structure, is discussed.  相似文献

Pituitary adenylate cyclase-activating polypeptide (PACAP) and helospectin are two vasoactive intestinal polypeptide (VIP)-related neuropeptides that have recently been demonstrated in the mammalian gut; the aim of this study was to reveal their occurrence and localisation in the gastrointestinal tract, swimbladder, urinary bladder and the vagal innervation of the gut of teleosts, using immunohistochemical methods on whole-mounts and sections of these tissues from the Atlantic cod, Gadus morhua and the rainbow trout, Oncorhynchus mykiss. Both PACAP-like and helospectin-like peptides were present in the gut wall of the two species. Immunoreactive nerve fibres were found in all layers but were most frequent in the myenteric plexus and along the circular muscle fibres. Immunoreactivity was also demonstrated in nerves innervating the swimbladder wall, the urinary bladder and blood vessels to the gut. Immunoreactive nerve cell bodies were found in the myenteric plexus of the gut and in the muscularis mucosae of the swimbladder. In the vagus nerve, non-immunoreactive nerve cells were surrounded by PACAP-immunoreactive fibres. Double staining revealed the coexistence of PACAP-like and helospectin-like peptides with VIP in all visualized nerve fibres and in some endocrine cells. It is concluded that PACAP-like and helospectin-like peptides coexist with VIP in nerves innervating the gut of two teleost species. The distribution suggests that both PACAP and helospectin, like VIP, are involved in the control of gut motility and secretion.  相似文献

The regeneration of the sciatic nerve fibres was studied in both normal and vitamin E-deficient rats at 30 and 60 days after crush. The vitamin E is involved in one of the most important mechanisms of protection against peroxidation of plasma membrane lipids; the plasma membrane plays certainly a role in nerve regeneration. Both the diameter and the total number of myelinated nerve fibres was calculated at different times. The number of myelinated fibres in the undenervated deficient animals was lower than that found in the undenervated normals animals. Following the nerve crush, in normal animals after two months the number of myelinated fibres exceeded the number found in undenervated normal animals, whereas in the deficient rat nerves it was significantly lower than in the corresponding controls and moreover it did not even reach the number found in the nerves of undenervated deficient rats. Finally, the caliber distribution of myelinated fibres in undenervated and denervated deficient rats shows a relative percent increase in the number of greatest axons and a decrease in smaller axons. This result confirm the vitamin E to be an important factor of the normal process of nerve regeneration.  相似文献

Some morphological data of the right and left human RLNs were evaluated with the aim of verifying possible differences in the fibre composition of the two nerves. The following parameters were evaluated in the right and left RLNs of five human cases: 1) the maximum diameter of the fibres; 2) the axon diameter and area; 3) the myelin sheath area obtained substracting the axon area from the total area of each fibre. The obtained data were plotted on histograms for each case: moreover, histograms of all fibres of both left and right nerves of all five cases were made. The results show that the values of the maximum diameter of the fibres and of the myelin sheath area are always greater in a statistically significant way in the left RLNs than in the right RLNs. On the other hand the axon diameter is nearly the same in the nerves of both sides. These data suggest that the greater calibre of the myelin sheath in the fibres of the left inferior laryngeal nerve can be responsible of the faster conduction speed in this nerve. This fact might explain the simultaneous arrival of the impulses to the laryngeal muscles of the two sides in spite to the different length of the two nerves.  相似文献

1. The C elevation of the compound action potential (CAP) was recorded with suction electrodes from dorsal roots of rats at 25 degrees C and toads (Bufo bufo) at 10 degrees C. The C fibre CAP had a conduction velocity of 0.5 +/- 0.07 SE M per sec (N = 10) and 0.25 +/- 0.04 M per sec (N = 8) in the rat and toad nerves respectively. 2. The depressant effect of applied drugs on the amplitude of the C fibres CAP was measured. Nerves from both species had similar sensitivities to GABA. EC50 5.0 microM +/- 0.5 SEM (N = 3) and 5.5 microM +/- 1.4 (N = 3) for the rat and toad respectively. Maximum depressant effects of GABA produced in rat and toad nerves were 35% +/- 5 SEM and 17% +/- 2.5 respectively. 3. In five out of ten of the rat nerves tested kainate had a clear depressant effect (maximum 36% +/- 4.3 SEM, EC50 6.8 microM +/- 0.9 SEM, N = 3) on the C fibre CAP. Kainate, at concentrations from 100 to 500 microM, had no effect on seven toad nerves. 4. Toad nerves were about 100 times less sensitive, than rat nerves, to capsaicin (ED50 values 430 microM +/- 190 SEM and 0.7 microM +/- 0.2 respectively, N = 4). 5. The similar sensitivity of nerves in both species to GABA and differing sensitivities to kainate and capsaicin suggests that amphibian C fibres specifically lack sensitivity to capsaicin and kainate.  相似文献

The splanchnic afferent signal in the myelinated fibre spectrum was analysed in cats and its relationship to viscerosomatic reflex activity was studied. In addition to Abeta and Adelta components, a further elevation was observed in the sympathetic chain neurogram. According to the conduction rate (15 plus or minus 4 m/sec), it was caused by myelinated fibres 2-3 mu in diameter. The presence of this component at thoracic dorsal root level, or in the sympathetic chain in stimulation of the postganglionic part of the nerve trunk, confirmed the afferent nature of the observed activity. The threshold stimulus for these fibres, which were termed group Adelta2, was four times higher than for Abeta fibres. Correlation of the neurogram with evoked activity in the intercostal nerves confirmed the different functional role of group Adelta2 fibres. Adelta2 fibre activity evokes the early component of the two-component viscerosomatic discharge and Adelta2 fibre activity its later component. The authors discuss the functional significance of Adelta2 fibres, which probably mediate nociceptive information from the viscera.  相似文献

The aim of this work was to gain insights into the mechanism of muscle differentiation and growth in Pagellus bogaraveo, by studying muscle fibre phenotypes identified by immunohistochemistry. At hatching, several layers of deep fast-white fibres were covered by a superficial fibre monolayer. At 5 days, slow-red fibres appeared near the lateral line nerve. At 40 days, the intermediate-pink muscle became visible, and in the slow-red and fast-white muscle layers transitions from larval myosin isoforms to the isoforms typical of adult muscle occurred. Between 70 and 100 days, small fibres with a distinct ATPase profile appeared throughout the fast-white muscle, marking the onset of “mosaic” hyperplasia. The myosin of the original superficial monolayer fibres underwent two myosin transformations, before being slowly replaced by an adult slow-red isoform. In juveniles and adults, the slow-red muscle layer could be resolved into two distinct types. The analysis of fibre phenotypes indicated that post-larval muscle growth occurred by two distinct stages of hyperplasia. This study offers a basis for further comparative and experimental studies with this economically relevant species, namely for identifying factors influencing its muscle growth dynamics and disclosing underlying mechanisms.  相似文献

The objective of this study was to determine if a recently developed human Ranvier node model, which is based on a modified version of the Hodgkin–Huxley model, could predict the excitability behaviour in human peripheral sensory nerve fibres with diameters ranging from 5.0 to 15.0 μm. The Ranvier node model was extended to include a persistent sodium current and was incorporated into a generalised single cable nerve fibre model. Parameter temperature dependence was included. All calculations were performed in Matlab. Sensory nerve fibre excitability behaviour characteristics predicted by the new nerve fibre model at different temperatures and fibre diameters compared well with measured data. Absolute refractory periods deviated from measured data, while relative refractory periods were similar to measured data. Conduction velocities showed both fibre diameter and temperature dependence and were underestimated in fibres thinner than 12.5 μm. Calculated strength–duration time constants ranged from 128.5 to 183.0 μs at 37°C over the studied nerve fibre diameter range, with chronaxie times about 30% shorter than strength–duration time constants. Chronaxie times exhibited temperature dependence, with values overestimated by a factor 5 at temperatures lower than body temperature. Possible explanations include the deviated absolute refractory period trend and inclusion of a nodal strangulation relationship. At the time of this research J. E.
