Plants are known to influence belowground microbial community structure along their roots, but the impacts of plant species richness and plant functional group (FG) identity on microbial communities in the bulk soil are still not well understood. Here, we used 454‐pyrosequencing to analyse the soil microbial community composition in a long‐term biodiversity experiment at Jena, Germany. We examined responses of bacteria, fungi, archaea, and protists to plant species richness (communities varying from 1 to 60 sown species) and plant FG identity (grasses, legumes, small herbs, tall herbs) in bulk soil. We hypothesized that plant species richness and FG identity would alter microbial community composition and have a positive impact on microbial species richness. Plant species richness had a marginal positive effect on the richness of fungi, but we observed no such effect on bacteria, archaea and protists. Plant species richness also did not have a large impact on microbial community composition. Rather, abiotic soil properties partially explained the community composition of bacteria, fungi, arbuscular mycorrhizal fungi (AMF), archaea and protists. Plant FG richness did not impact microbial community composition; however, plant FG identity was more effective. Bacterial richness was highest in legume plots and lowest in small herb plots, and AMF and archaeal community composition in legume plant communities was distinct from that in communities composed of other plant FGs. We conclude that soil microbial community composition in bulk soil is influenced more by changes in plant FG composition and abiotic soil properties, than by changes in plant species richness per se.  相似文献

1. Agricultural intensification has caused dramatic biodiversity loss in many agricultural landscapes over the last century. Here, we investigated whether new types of farm ponds (made of artificial substrata) in intensive systems and natural‐substratum ponds in traditional farming systems differ in their value for aquatic biodiversity conservation. 2. We analysed the main patterns of environmental variation, compared α‐, β‐ and γ‐diversity of macroinvertebrates between ponds types and evaluated the role of submerged aquatic vegetation (SAV). Generalised additive models (GAM) were used to analyse the relationships of α‐ and β‐diversity with environmental predictors, and variation partitioning to separate the effect of environmental and spatial characteristics on the variation in macroinvertebrate assemblages. Moran’s eigenvector maps (MEMs) were used to define spatial variables. 3. A principal coordinate analysis (PCoA) detected a primary environmental gradient that separated nutrient‐rich ponds from those dominated by SAV; a secondary morphometric gradient distinguished natural‐substratum ponds, with large surface area and structural complexity, from artificial‐substratum ponds with steeper slopes. Natural‐substratum ponds had almost twice the α‐ and γ‐diversity of artificial‐substratum ponds, and diversity significantly increased when SAV was present, particularly in artificial‐substratum ponds. Total phosphorus (TP) strongly contributed to explain the patterns in diversity, while SAV was a significant predictor of assemblage composition and diversity. GAMs revealed optima of both α‐diversity at intermediate SAV covers and β‐diversity at intermediate–high TP concentrations. 4. These findings have important implications for conservation planning. Adaptation of artificial‐substratum ponds by adding natural substratum and smoothing the gradient of pond margins would improve their conservation value. Development of SAV with occasional harvests and certain cautionary measures to control nutrient levels may also improve both the agronomical and environmental function of ponds.  相似文献

Inflammatory cytokines are closely related to pigmentary changes. In this study, the effects of IFN‐γ on melanogenesis were investigated. IFN‐γ inhibits basal and α‐MSH‐induced melanogenesis in B16 melanoma cells and normal human melanocytes. MITF mRNA and protein expressions were significantly inhibited in response to IFN‐γ. IFN‐γ inhibited CREB binding to the MITF promoter but did not affect CREB phosphorylation. Instead, IFN‐γ inhibited the association of CBP and CREB through the increased association between CREB binding protein (CBP) and STAT1. These findings suggest that IFN‐γ inhibits both basal and α‐MSH‐induced melanogenesis by inhibiting MITF expression. The inhibitory action of IFN‐γ in α‐MSH‐induced melanogenesis is likely to be associated with the sequestration of CBP via the association between CBP and STAT1. These data suggest that IFN‐γ plays a role in controlling inflammation‐ or UV‐induced pigmentary changes.  相似文献

Aim We investigated whether faunas of lentic macroinvertebrates differed among two landscape types: (1) those that are largely covered in forests (presumed to be in a more pre‐human‐impact condition) and (2) those that are completely cleared for agricultural exploitation (massively altered). Location Five pairs of landscapes (each pair referred to as a region) – one of each landscape type – across a 30,000 km2 region of north‐central Victoria, Australia. Methods Each individual waterbody was surveyed three times (austral spring 2006, autumn 2007, and spring 2007) for invertebrates. Waterbodies were characterized by measurements of static (e.g. abutting vegetation cover) and labile (e.g. pH) variables. Data were analysed using hierarchical Bayesian models of species richness, α‐ and β‐diversities and functional feeding groups. Assemblage composition was related to landscape and in‐waterbody characteristics. Results Neither measured, nor asymptotic estimates of, species richness differed among landscape types, notwithstanding consistent differences in in‐waterbody habitat characteristics among waterbodies in the two landscape types. There were no discernible differences in patterns of α‐ and β‐diversities at landscape scales relating to landscape type. Habitat diversity of waterbodies at the landscape scale did not affect β‐diversity, although distinct waterbodies within landscapes tended to have more distinct faunas. Main conclusions The lentic macroinvertebrate faunas are relatively homogeneous over the entire region, with little differentiation between wooded and cleared landscapes. The regional fauna may be a homogenized subset of native species, possibly arising from the huge numerical predominance of lentic habitats in agricultural landscapes producing ‘spill‐over’ effects into forested landscapes. Of taxa more frequently found in one or other landscape type, trophic group diversity was greater in forested landscapes.  相似文献

The genetic variation across a species’ range is an important factor in speciation and conservation, yet searching for general patterns and underlying causes remains challenging. While the majority of comparisons between central and marginal populations have revealed a general central–marginal (C–M) decline in genetic diversity, others show no clear pattern. Similarly, most latitudinal studies (although much fewer, especially those conducted rangewide) also showed latitudinal trends in genetic variation. To date, the C–M and latitudinal patterns have often been examined independently and have rarely been considered together when accounting for the observed genetic variation across species ranges. Here, in the light of the most recent findings, I show how latitude might be responsible for some of the deviations from the general C–M trends in genetic diversity, and vice versa. In the future, integrating latitude and range geometry with climate‐induced species migration would offer important insights into conservation prioritization across species ranges.  相似文献

Community assembly is determined by a combination of historical events and contemporary processes that are difficult to disentangle, but eco‐evolutionary mechanisms may be uncovered by the joint analysis of species and genetic diversity across multiple sites. Mountain streams across Europe harbour highly diverse macroinvertebrate communities whose composition and turnover (replacement of taxa) among sites and regions remain poorly known. We studied whole‐community biodiversity within and among six mountain regions along a latitudinal transect from Morocco to Scandinavia at three levels of taxonomic hierarchy: genus, species and haplotypes. Using DNA barcoding of four insect families (>3100 individuals, 118 species) across 62 streams, we found that measures of local and regional diversity and intraregional turnover generally declined slightly towards northern latitudes. However, at all hierarchical levels we found complete (haplotype) or high (species, genus) turnover among regions (and even among sites within regions), which counters the expectations of Pleistocene postglacial northward expansion from southern refugia. Species distributions were mostly correlated with environmental conditions, suggesting a strong role of lineage‐ or species‐specific traits in determining local and latitudinal community composition, lineage diversification and phylogenetic community structure (e.g., loss of Coleoptera, but not Ephemeroptera, at northern sites). High intraspecific genetic structure within regions, even in northernmost sites, reflects species‐specific dispersal and demographic histories and indicates postglacial migration from geographically scattered refugia, rather than from only southern areas. Overall, patterns were not strongly concordant across hierarchical levels, but consistent with the overriding influence of environmental factors determining community composition at the species and genus levels.  相似文献

Mountains are among the most powerful natural gradients for testing ecological and evolutionary responses of biota to environmental influences because differences in climate and plant structure occur over short spatial scales. We describe the spatiotemporal distribution patterns and drives of fruit‐feeding butterfly diversity in the mountainous region of Serra do Cipó, Minas Gerais, Brazil. Seven elevations from 822 to 1,388 m a.s.l. were selected for evaluating the effects of abiotic factors and vegetation characteristics on butterfly diversity. A total of 44 fruit‐feeding butterfly species were recorded in a two‐year study. Species richness (local and regional) of fruit‐feeding butterflies decreased with increasing elevation. The interaction between temperature or humidity and precipitation influenced the abundance and β‐diversity of butterflies in the elevation gradient, whereas β‐diversity decreased with increasing plant richness. Butterfly richness (local and regional) and β‐diversity varied with the sampling period, with fewer species in July (2012 and 2013), the dry period, as expected for Neotropical insects. β‐Diversity in space and time was due to species replacement (turnover), indicating that butterfly composition differs throughout the mountain and over time. In summary, climate and plant richness largely influence butterfly diversity in the elevational gradient. Climatic changes in conjunction with increasing anthropic impacts on mountainous regions of southeast Brazil will likely influence the community of mountaintop butterflies in the Espinhaço Mountain Range. The adaptation of nine species of mites that infest stored products for starch utilization was tested by (1) enzymatic analysis using feces and whole mite extracts, (2) biotests, and (3) inhibition experiments. Acarus siro, Aleuroglyphus ovatus, and Tyroborus lini were associated with the starch‐type substrates and maltose, with higher enzymatic activities observed in whole mite extracts. Lepidoglyphus destructor was associated with the same substrates but had higher activities in feces. Dermatophagoides farinae, Chortoglyphus arcuatus, and Caloglyphus redickorzevi were associated with sucrose. Tyrophagus putrescentiae and Carpoglyphus lactis had low or intermediate enzymatic activity on the tested substrates. Biotests on starch additive diets showed accelerated growth of species associated with the starch‐type substrates. The inhibitor acarbose suppressed starch hydrolysis and growth of the mites. We suggest that the species with higher starch hydrolytic activity in feces were more tolerant to acarbose, and α‐amylase and α‐glucosidase of synanthropic mites are suitable targets for inhibitor‐based strategies of mite control. © 2009 Wiley Periodicals, Inc.  相似文献

The biological underpinnings linking stress to Alzheimer's disease (AD) risk are poorly understood. We investigated how corticotrophin releasing factor (CRF), a critical stress response mediator, influences amyloid‐β (Aβ) production. In cells, CRF treatment increases Aβ production and triggers CRF receptor 1 (CRFR1) and γ‐secretase internalization. Co‐immunoprecipitation studies establish that γ‐secretase associates with CRFR1; this is mediated by β‐arrestin binding motifs. Additionally, CRFR1 and γ‐secretase co‐localize in lipid raft fractions, with increased γ‐secretase accumulation upon CRF treatment. CRF treatment also increases γ‐secretase activity in vitro, revealing a second, receptor‐independent mechanism of action. CRF is the first endogenous neuropeptide that can be shown to directly modulate γ‐secretase activity. Unexpectedly, CRFR1 antagonists also increased Aβ. These data collectively link CRF to increased Aβ through γ‐secretase and provide mechanistic insight into how stress may increase AD risk. They also suggest that direct targeting of CRF might be necessary to effectively modulate this pathway for therapeutic benefit in AD, as CRFR1 antagonists increase Aβ and in some cases preferentially increase Aβ42 via complex effects on γ‐secretase.  相似文献

Objectives

Previous research has established population variation in anti‐inflammatory immunological biomarkers in human milk. This immunity is potentially ecology‐dependent and may alter the life history trade‐off between growth and maintenance in infants. The current study has two aims: (1) to assess the ecological differences in milk immunity in two populations, one from the urban U.S. and one from rural Kenya; and (2) to test the hypothesis that milk immunity can affect infant growth indicators.

Materials and Methods

Kenyan Ariaal (n = 233) and U.S. (n = 75) breastfeeding mother‐infant pairs participated in a cross‐sectional study at two separate field sites. Laboratory analysis was performed on milk for the anti‐inflammatory biomarkers TGF‐β2, sTNF‐αRI, sTNF‐αRII, and IL‐1ra using ELISA. Multiple imputation was used to extrapolate data below the limit of detection before multivariate analysis.

Results

There were significant differences between U.S. and Kenyan mothers on all four milk biomarkers, with Kenyan mothers having significantly higher sTNF‐αRI and sTNF‐αRII and lower TGF‐β2 and IL‐1ra than U.S. mothers. U.S. mothers with higher milk TGF‐β2 and IL‐1ra have infants that are significantly longer and heavier for their age, while Kenyan mothers with higher sTNF‐αRI have significantly longer and heavier infants for their age, and those with higher TGF‐β2 have marginally significantly longer infants.

Discussion

There were significant differences in ecological milk immunity between U.S. and Kenyan mothers. These differences potentially play a role in the growth of their infants. Further research in milk immunity should consider the possibility of shared maternal–infant life histories.

Both enantiomers of three biologically relevant paraconic acids—MB‐3, methylenolactocin, and C75—were obtained with enantioselectivities up to 99% by kinetic enzymatic resolutions. Good enantiomeric excesses were obtained for MB‐3 and methylenolactocin, using α‐chymotrypsin and aminoacylase as enantiocomplementary enzymes, while C75 was resolved with aminoacylase. They all were evaluated for their antiproliferative, antibacterial, and antifungal activities, showing weak effects and practically no difference between enantiomers in each case. We analyzed the organization of the microtubular cytoskeleton and the distribution of centrosomes at the different stages of differentiation of the ovarian follicle of the lizard Podarcis sicula by examining immunolabeled α‐ and γ‐tubulins using confocal microscopy. We observed that in the follicular epithelium the differentiation of the nurse pyriform cells is accompanied by a reorganization of the microtubules in the oocyte cortex, changing from a reticular to a radial pattern. Furthermore, these cortical microtubules extend in the cytoplasm of the connected follicle cells through intercellular bridges. Radially oriented microtubules were still more marked in the oocyte cortex during the final stages of oogenesis, when the yolk proteins were incorporated by endocytosis. The nucleation centres of the microtubules (centrosomes) were clearly detectable as γ‐tubulin immunolabeled spots in the somatic stromal cells of the germinal bed. A diffuse cytoplasmic immunolabeling together with multiple labeled foci, resembling the desegregation of the centrosomes in early oogenesis of vertebrates and invertebrates, was revealed in the prediplotenic germ cells. In the cytoplasm of growing oocytes, a diffuse labeling of the γ‐tubulin antibody was always detectable. In the growing ovarian follicles, immunolabeled spots were detected in the mono‐layered follicle cells which surrounded the early oocytes. In follicles with a polymorphic follicular epithelium, only the small follicle cells showed labeled spots. A weak and diffuse labeling was observed in the pyriform cells while in the enlarging intermediate cells the centrosomes degenerated like in the early oocytes. Our observations confirm that in P. sicula most of the oocyte growth is supported by the structural and functional integration of the developing oocyte with the pyriform nurse cells and suggest that their fusion with the oocyte results in an acquirement by these somatic cells of characteristics typical of the germ cells. The essential oils (EOs) isolated from the leaves and twigs of Juniperus excelsa M.Bieb . growing wild in Lebanon were characterized, and their antimicrobial activity and antiradical capacity were evaluated. The EOs were obtained by hydrodistillation using a Clevenger‐type apparatus and characterized by GC and GC/MS analyses. The antimicrobial activity was evaluated by determining minimal inhibitory concentrations (MICs) against a Gram‐positive and a Gram‐negative bacterium, a yeast, and a dermatophyte with the broth microdilution technique. A total of 28 constituents was identified and accounted for 90.1 and 95.6% of the twig and leaf EO composition, respectively. Both EOs were essentially composed of monoterpene hydrocarbons (46.7 and 59.6% for twig and leaf EOs, resp.) and sesquiterpenes (39.4 and 32.1%, resp.). The main components were α‐pinene, α‐cedrol, and δ‐car‐3‐ene. The J. excelsa EOs did not show any antiradical potential, but revealed interesting in vitro antimicrobial activities against Staphylococcus aureus and Trichophyton rubrum (MICs of 64 and 128 μg/ml, resp.). The three major compounds were tested separately and in combination according to their respective amounts in the oil. δ‐Car‐3‐ene was the most active component and is undoubtedly one of the constituents driving the antifungal activity of J. excelsa essential oil, even though synergies are probably involved.  相似文献
