刺芹侧耳生长条件和栽培特性的研究   总被引:2,自引:0,他引:2  
对刺芹侧耳[Pleurotus eryngii(DC.ex Fr.)Quel]生长条件与栽培特性进行了分析.结果表明:刺芹侧耳菌丝生长的适宜pH值为pH 5~pH 7,最适pH值为pH 6;黑暗环境有利于菌丝生长,光照对菌丝生长具有抑制作用;菌丝生长适宜的碳源为可溶性淀粉、葡萄糖和蔗糖,适宜的氮源为蛋白胨和钼酸铵,葡萄糖和蛋白胨的适宜浓度分别为50和1 g·L-1;在马铃薯黄豆粉培养基、黄豆粉玉米粉培养基和马铃薯蛋白胨培养基中可较好地形成菌丝球.采用液体菌种栽培,菌丝的生长速度比使用玉米固体菌种快,其中用马铃薯黄豆粉液体菌种栽培,菌丝生长最快;使用马铃薯蛋白胨液体菌种在木屑棉籽壳混合培养料中栽培,可获得菌柄长菌盖大的子实体,而且产量高.  相似文献

通过对尖顶羊肚菌液体培养基质与条件的研究,明确其菌丝生长的最适pH值、最适温度、适宜光照条件、适宜葡萄糖和蛋白胨浓度、适宜培养基,以便应用于尖顶羊肚菌液体菌种的生产和工业发酵。结果表明:菌丝的最适生长温度为2 5℃;最适生长pH值为6 ;葡萄糖和蛋白胨最适浓度分别为2 0 0g/L和10g/L ;菌丝在黑暗环境下生长良好,光照对菌丝生长具有抑制作用;用胡萝卜酵母膏培养基振荡培养形成的菌丝球多,菌丝生长量大;菌丝球在不同培养基中生长,可引起培养液pH值的上升或者下降;菌丝球可利用培养基内的氨基酸,使氨基酸降解,在胡萝卜酵母膏培养基中振荡培养8d的菌液总氨基酸含量较原液减少了36 71% ,亮氨酸、异亮氨酸和甲硫氨酸含量的下降幅度最大  相似文献

大黄鱼细菌性病原哈维氏弧菌培养特性的研究   总被引:1,自引:0,他引:1  
弧菌是危害水产养殖动物的最为严重的病原之一.从患病大黄鱼肝脏分离致病的细菌哈维氏弧菌GYC1108-1为材料,对哈维氏弧菌GYC1108-1株的最佳生长条件及培养基优化进行了测定.综合考察不同的培养温度、培养时间、培养基盐度、培养基的pH值对细菌生长的影响,并采用正交实验法,对培养基配方进行优化.结果表明:不同的培养温度、培养时间、培养基盐度、培养基pH值等均会影响细菌的产量.GYC1108-1适宜生长的盐度为1%-5%、pH为5~9.5、温度为15~35℃、在培养基中添加硫酸铜、硫酸氨会明显促进生长;最佳培养条件及培养基成份为:NaCl 2%,pH8.0,温度30℃,蛋白胨1.0%、牛肉膏0.75%、CuSO4 0.3mg/L、(NH4)2SO4 0.1g/L.  相似文献

本研究为从云南腾冲热泉中分离纯化得到一株产高温蛋白酶的菌株并对其进行驯化培养,用以探究该菌株的生长条件及酶学特性,通过选择培养基筛选能够分解脱脂奶粉产蛋白酶的菌株,应用常规方法液体培养菌体,探究温度、pH、碳源、氮源对菌株生长情况的影响,并采用福林酚法测蛋白酶活性。并提取蛋白酶液对酶的最适pH、温度以及热稳定性、pH稳定性进行研究。结果发现通过含脱脂奶粉的固体培养基筛选得到一株产蛋白酶菌株A-2,经过生理生化试验和16S rDNA鉴定知该菌种属于Aneurinibacillus属。酵母粉、葡萄糖、55℃、pH值7.5分别为菌株生长的最适氮源、碳源、温度和pH。此外该菌株所产的蛋白酶最适温度为60℃,在pH值7~9具有较好的酶活性。因此,该菌株为嗜热芽孢杆菌,所产的碱性蛋白酶具有较高的耐受温度和pH稳定性,为进一步开发利用提供参考的价值。  相似文献

以查氏培养基为基础,在碳源、氮源、pH值、温度等单因子条件研究的基础上,采用正交实验对淡紫拟青霉的培养条件进行了优化研究,得到了适合其生长的发酵培养基配方。实验结果表明,以蔗糖为碳源(60g/L),硝酸铵为氮源(1.5g/L),pH5~6,培养温度为30℃时最适合该菌的生长。  相似文献

通过对白色金针菇液体菌种制备及工厂化生产中应用的研究,确定白色金针菇液体菌种培养基配方及制备条件,建立了采用液体菌种进行白色金针菇工厂化生产的技术规程.通过对茵丝干重的测定,确定了液体菌种培养基配方、培养时间、培养温度、起始pH值等技术参数.并根据本地区的资源情况,通过比较试验,确定了白色金针菇液体菌种制备工艺,工厂化生产的培养料配方及工艺流程.结果表明:采用液体菌种进行白色金针菇的工厂化生产,发茵生长周期明显缩短,污染率显著降低,为白色金针菇工厂化生产提供了技术基础.  相似文献

为研究A、C、Y、W 135群流脑多糖疫苗,针对W 135群脑膜炎球菌的生长特性,采用国产50L发酵罐,针对流脑半综合液体培养基中葡萄糖浓度;培养温度、菌种接种浓度、pH、通氧量及搅拌速度等因素对W 135群脑膜炎球菌生长的影响,选择最适的培养条件。结果表明,选择半综合培养基中补加1%的葡萄糖培养效果良好,菌种接种浓度直接影响多糖复合物产量;最适pH值6.5~7.5的范围可维持W 135群链球菌快速生长;温度控制在36.5±0.2℃可得到较高的培养产物;培养过程中加大通气量及搅拌速度对培养结果有明显改善。确立了W 135群脑膜炎球菌的最适培养条件,在确定培养条件后连续进行3批500L罐放大中试培养,达到规模化生产要求。  相似文献

目的:优化光合细菌的最佳培养条件。方法:通过绘制不同的生长曲线研究不同培养基及不同培养条件等理化因素对2株光合细菌(类球红细菌1.2174、沼泽红假单胞菌1.2180)生长的影响。结果:①1.2174最佳条件:培养基(蒸馏水配制的液体富集培养基和固体ATYP);温度(35℃);pH值(8.0和9.0);接种量(培养基/种子液5:1和4:1)。②1.2180最佳条件:培养基(海水配制的ATYP和蒸馏水配制的液体富集培养基);温度(25℃和35℃);pH值(7.0和8.0);接种量(培养基/种子液2∶1和4∶1)③两株菌的光照、氧需求程度、菌群协同和振荡条件相同。结论:通过筛选,蒸馏水配制的液体富集培养基和固体ATYP培养基、35℃、1000lx、pH值8.0、培养基/种子液4∶1、微氧、有菌群协同、振荡可作为二者的通用培养条件。  相似文献

对喇叭石蕊共生菌、藻液体培养条件进行了研究。结果表明:共生菌生长在以40g/L肌醇为碳源、2g/LL-谷氨酰胺为氮源、起始pH值为7.0的LB液体培养基中,培养温度为20℃时表现最佳。其共生藻的生长在以160g/L葡萄糖为碳源、1.75g/LNaNO3为氮源、起始pH值为5.0的BBM液体培养基中,培养温度为20℃时表现最佳。  相似文献

采用菌丝生长速率法对引起石榴枯萎病和甘薯黑斑病的甘薯长喙壳菌的形态特征,及其在不同培养基、光照、温度、pH值等基本培养条件下菌丝的生长速率等生物学特性的差异进行了比较研究。结果表明,在不同处理培养条件下,石榴枯萎病菌生长速度均快于甘薯黑斑病菌,其菌落形态存在较大差异。二者在甘薯葡萄糖培养基(SPDA)上生长速率最快。在黑暗条件下的生长速率比12h光暗交替、光照处理显著增大。二者最适生长温度皆为25℃,石榴枯萎病菌菌丝生长温度范围为10-36℃,而甘薯黑斑病菌的菌丝生长温度范围为10-33℃。二者菌丝在pH4-12范围内均能生长,最适pH值为6。石榴枯萎病菌菌丝的致死温度为50℃处理10min或52℃处理5min,甘薯黑斑病菌菌丝的致死温度为48℃处理10min或50℃处理5min。二者在完全培养液中产孢量最大,菌丝生长速率随培养基中葡萄糖浓度的减小而增大。  相似文献

Ganoderma lucidum, a white rot fungus, was exploited for its potentials to produce xylanase employing shake and solid-state culture conditions. Different culture conditions such as pH, temperature, carbon and nitrogen requirements for its growth and production of xylanase were optimized. The culture media pH 6.0-7.0 and temperatures 30 degrees-35 degrees C significantly promoted the growth as well as xylanase secretion into the media. Xylan and peptone were found to be the suitable carbon and nitrogen sources. Among the different agrowastes used, wheat bran was found to be the best substrate for the test fungus for the production of xylanase than sugarcane bagasse and rice bran in solid-state fermentation.  相似文献

Study of fungal colonial growth is a basic method to examine their behaviour in different cultivation conditions. The influence of temperature and initial pH on growth radial velocity and growth density of Botryodiplodia theobromae RC1, was studied in order to show the growth characteristics of this fungus. Both temperature and culture medium influenced growth density, but radial velocity of growth was only affected by temperatures above 40 degrees C. In addition, initial pH of culture media did not affect either parameter.  相似文献

油球藻(Graesiella sp.WBG-1)是一株适合于开放池规模化培养的产油微藻,在室外规模培养过程中常因一种病原真菌(Amoeboaphelidium sp.)的污染严重影响其生长和油脂积累,甚至导致培养失败。本文利用MTT染色法借助光学显微镜对被病原真菌感染的油球藻细胞计数并统计感染率,设置不同温度、光照强度、p H以及通气量等培养条件,研究环境因子对病原真菌感染能力的影响。结果表明:MTT染色法简单易行,可用于油球藻规模培养中病原真菌的检测;温度、光照强度、通气量和p H值均能够显著影响病原真菌的感染能力,高温、高光照强度、弱酸性环境和藻液静止等培养条件不利于病原真菌对油球藻的感染;在温度30℃、光强140μmol·m~(-2)·s~(-1)、通气量1.0 L·min~(-1)和p H 9.0±0.5的培养条件下成功建立了病原真菌感染油球藻的连续传代培养。本研究在实验室内模拟油球藻规模培养中被病原真菌感染的全过程,为深入研究该病原真菌对微藻细胞的感染行为及其感染机制提供了平台。  相似文献

大型真菌种质资源是国家种质资源库建设重要组成部分,为了充分开发利用大型真菌资源,对采自吉林省寒葱岭的一株野生菌进行分离纯化,并将获取的纯菌株作为实验材料。通过形态学和ITS序列分析,将其鉴定为薄皮干酪菌Tyromyces chioneus。本实验研究了固体培养条件下不同碳源、氮源、pH和温度对其菌丝生长的影响,并从4个单因素实验中选出3个最优水平进行正交实验。同时,在对其菌丝体最适培养条件研究结果的基础上,对其进行驯化栽培研究。结果表明,薄皮干酪菌的最适培养条件为:果糖(20 g/L)、酵母浸粉(2 g/L)、pH 4.0、温度30 ℃。正交实验结果表明,氮源对其菌丝体生长的影响最大,其次是碳源、pH和温度。驯化栽培结果表明,菌丝发菌时间为21 d;空气湿度为85%-95%,适量的散射光,降低温度至18-20 ℃刺激后原基逐步变成菇蕾,将温度、湿度分别提高至24-25 ℃、90%-98%,30 d后子实体成熟。通过本实验的研究为薄皮干酪菌的后续开发利用提供了基础实验数据。  相似文献

The effect of external factors like pH of the medium, temperature and incubation period on the growth of the ectomycorrhizal fungus, Pisolithus tinctorius was studied as a function of the total fungal biomass produced, extracellular protein content and residual pH of the medium. The effect of the three parameters on the three responses was found to be significant and interdependent. Therefore, an attempt was made to optimize the growth of the fungus under varying conditions of pH, temperature and incubation period using the Box-Behnken Design of Experiments which is a second order model involving Response Surface Methodology and a second order quadratic equation. With this design expert, the optimum conditions of the three parameters that favoured the maximum growth of the fungus in vitro were found. The experimental values corroborated with the predicted values got from the model with the correlation coefficients 0.9349 for biomass, 0.9913 for protein and 0.9959 for final pH.  相似文献

A purified culture of the fungal pathogenCurvularia lunata causing leaf blight of pearl millet was used for studies on the production of cell-wall-degrading enzymes. Czapek-Dox medium was found to be the best medium of the five different nutrient media used for the production of cellulase and growth of the fungus. Pectolytic enzymes could not be detected under different cultural conditions. Two-week incubation period, pH 6.0 and temperature 25°C were found to be the most favorable conditions for good growth and maximum production of cellulase by the fungus in present studies. The results are presented and discussed in the light of the earlier findings onC. lunata.  相似文献

A psychrotolerant phosphate solubilizing fungus has been isolated from the rock soil of a cold desert site in Indian Himalaya. The fungus grows from 4 to 35°C (optimum 21°C), and from 2 to 13.5 pH (optimum 9) under laboratory conditions. Based on phenotypic characters and 26S rDNA analysis, the fungus is identified as Paecilomyces hepiali. In quantitative estimation that was carried out at 9, 14, and 21°C, the fungus solubilized maximum phosphate at 14°C. In view of the slow growth and persistence of the desired activity at low temperature, the estimation was carried out for a longer period, i.e., up to 6 weeks. The suboptimal conditions for growth and biomass production were found to be optimal for phosphate solubilization by the fungus. At 14 and 9°C, the solubilization touched its maximum on day 42. Decline in pH was found to be significantly correlated with the phosphate solubilization at all the temperatures, under consideration. The acid phosphatase activity was found to be more prominent than alkaline phosphatase in culture filtrate. High performance thin layer chromatography (HPTLC) analysis showed production of six organic acids, gluconic and α-keto glutaric acid being in maximum amount in the culture filtrate. The study has ecological significance in view of the nutrient cycling under low temperature environment, prevalent in Himalayan region.  相似文献

Microorganisms associated with invertebrate hosts have long been suggested to be a source for bioactive metabolites. In this study, we reported that a sponge-associated fungus, Letendraea helminthicola, produced two antifouling compounds: 3-methyl-N-(2-phenylethyl) butanamide and cyclo(D-Pro-D-Phe). To optimize the production of these antifouling compounds, we then examined the production of compounds under different culture conditions (temperature, salinity, pH, and carbon and nitrogen sources). This fungus grew well and produced more compounds at temperatures between 18 and 30°C; the fungus grew well at 75 parts per thousand (ppt) salinity but produced the highest amount of antifouling compounds at 30 and 45 ppt. The optimal initial pH value for mycelial growth was 5.5 to 6.5, whereas the production of the antifouling compounds was maximized at pH 3.5 and 4.5. Glucose and xylose (as carbon sources) increased the production of antifouling compounds. Yeast extract and peptone (as nitrogen sources) maximized the production of mycelial biomass and antifouling compounds. Our results indicate that culture conditions greatly affect the production of bioactive compounds from mycelial fungal cultures as exemplified by strain L. helminthicola and that the conditions favorable for fungal growth may not be the best conditions for bioactive compound production.  相似文献

The ability of two wood-decay basidiomycetes to produce extracellular mucilaginous material (ECMM) and its relationship with total biomass production has been investigated. Growth and ECMM production by the white-rot fungus Coriolus versicolor and the brown-rot fungus Gloeophyllum trabeum were assessed in liquid culture under different nutritional and environmental conditions. Nutritional, pH and temperature factors all influenced significantly the proportion of ECMM in the total biomass produced. When total biomass production was reduced due to unfavorable growth conditions (stress), the proportion of ECMM in the biomass was elevated. The results are discussed with regard to the possible role(s) of ECMM in the responses of these fungi to stress.  相似文献

【背景】供试菌株分别是分离自山西省宁武县管涔山的肉色香蘑Lepista irina、斑褶香蘑L. panaeolus和山西省蒲县五鹿山的紫丁香蘑L. nuda 3种野生食用菌的子实体。【目的】获得3种野生食用菌的最佳培养条件。【方法】以菌丝生长速度为指标研究不同碳源、氮源、碳氮比、pH和培养温度等各因素对菌丝生长的影响,根据Box-Benhnken中心组合试验设计原理,采用3因素3水平的响应面法确定使菌丝体达到最快生长速度的最佳培养碳源、氮源和pH。【结果】肉色香蘑在葡萄糖20.9 g/L、土豆196.47 g/L、pH 6.0、培养温度21 °C的条件下,菌丝日均生长速度达到最大,为1.13 mm/d;斑褶香蘑在甘露醇17.4 g/L、酵母膏8.1 g/L、B族维生素0.1 g/L、K2HPO4 2.5 g/L、MgSO4 2.5 g/L、pH 7.9、培养温度25 °C的条件下,菌丝日均生长速度达到最大,为0.73 mm/d;紫丁香蘑在土豆200 g/L、可溶性淀粉20.5 g/L、KNO3 2.1 g/L、K2HPO4 2.5 g/L、MgSO4 2.5 g/L、B族维生素0.1 g/L、pH 7.0、培养温度25 °C的条件下,菌丝日均生长速度达到最大,为2.38 mm/d。【结论】获得了3种香蘑属菌株的最佳培养条件,为后续优质野生食用菌的引种驯化积累了相关数据和资源。  相似文献
