Photoinhibition of Glucose Uptake in Chlorella   总被引:1,自引:0,他引:1  
In colorless mutant cells of Chlorella vulgaris (M125), endogenousrespiration in the dark was not affected by 30-min preilluminationwith white light (9,000 mW?m–2), while exogenous respirationof glucose or fructose was inhibited significantly by the sametreatment in air, but not under N2. This light effect on exogenousrespiration was accompanied by an inhibition of hexose uptake. When autotrophically grown wild-type cells of Chlorella vulgaris(211-11h) were incubated in glucose medium with DCMU, lightalso greatly inhibited glucose uptake and growth. Blue lightwas very effective, while red light had only a slight effect.This photoinhibitory effect was also observed in algal cellsthat had been grown in a glucose-containing medium in the dark. Using SDS-gel electrophoresis, a new protein peak with a molecularweight of 35–40 kDa was detected in plasma membrane-richcell wall fractions when Chlorella vulgaris (211-11h) cellswere transferred to a glucose-containing medium. This peak disappearedafter the algal cells were returned to the glucose-free medium.These findings suggest that this protein includes the hexose-carrierprotein. When Chlorella vulgaris 11h, Chlorella vulgaris C-l, Chlamydomonasreinhardtii, Chlamydomonas moewusii, Scenedesmus obliquus, orDunaliella tertiolecta were illuminated in with 0.5 mM NaHCO3,the pH of the medium increased in a few minutes from 6 to about9 or 10. The alkalization, which was accompanied by O2 evolution,was dependent on light, external dissolved inorganic carbon(DIC) as HCO-3, and algae grown or adapted to a low, air-levelCO2 in order to develop a DIC concentrating mechanism. Therewas little pH increase by algae without a DIC concentratingprocess from growth on 3% CO2 in air. Photosynthetic O2 evolutionwithout alkalization occurred using either internal DIC or externalCO2 at acidic pH. The PH increase stopped between pH 9 to 10,but the alkalization would restart upon re-acidification betweenpH 6 and 8. Alkalization was suppressed by the carbonic anhydraseinhibitors, acetazolamide, ethoxyzolamide or carbon oxysulfide.The pH increase appeared to be the consequence of the externalconversion of HCO3 into CO2 plus OH during photosynthesisby cells with a high affinity for CO2 uptake. Cells grown onhigh CO2 to suppress the DIC pump, when given low levels ofHCO3 in the light, acidified the medium from pH 10 to7. Air adapted Scenedesmus cells with a HCO3 pump, aswell as a CO2 pump, alkalized the medium very rapidly in thelight to a pH of over 10, as well as slower in the dark or inthe light with DCMU or without external DIC and O2 evolution.Alkalization of the medium during photosynthetic DIC uptakeby algae has been considered to be part of the global carboncycle for converting H2CO3 to HCO3 and for the formationof carbonate salts by calcareous algae from the alkaline conversionof bicarbonate to carbonate. The maximum rate of photosynthetic 14CO2 fixation (Vmax) aswell as the concentration of CO2 at which the rate of photosynthetic14CO2 fixation attains one-half its maximum velocity (Km) inChlorella vulgaris 11h cells was strongly dependent on the concentrationof CO2 continuously provided during the algal growth. The Vmax (µmoles 14CO2 fixed/ml pcv?min) and Km (% CO2)of the algal cells which had been grown in air containing 4%CO2 (by volume) were ca. 10 and 0.15–0.17, while thosein the cells which had been grown in ordinary air (containing0.04% CO2) were 7 and 0.05–0.06, respectively. When the concentration of CO2 in the bubbling gas was loweredfrom 4 to 0.04% during the algal growth, their photosynthetickinetics attained the respective lower steady levels after 5–10hr. On the other hand, when the photosynthetic kinetics weredetermined 24 hr after raising the concentration of CO2 from0.04 to 4%, the Vmax and Km-values were found to have alreadyattained the respective higher levels. The carbon dioxide compensation point of the unicellular greenalga, Chloretla saccharophila, was determined in aqueous mediumby a gas chromatographic method. Compensation points decreasedmarkedly from 63 cm3 m–3 at an external pH of 4.0 to 3.2cm3 m–3 at pH 8.0 and were not affected by the O2 concentrationof the medium. The calculated CO2 concentration required tosupport the half-maximum photosynthetic rate of the algal cellsranged from 6.0 mmol m–3 at an external pH of 60 to 1.5mmol m–3 at pH 8.0 and these values were not affectedby O2 concentration. The Km(CO2) of nbulose-l,5-bisphosphatecarboxylase isolated from cells grown either at pH 4.0 or pH8.0 was determined to be 64 mmol m–3. These results indicatethat loss of CO2 by photorespiration does not occur in C. saccharophilacells at acid pH and the disparity between the apparent affinityfor CO2 of the intact cells and that of the carboxylase indicatesthe operation of a ‘CO2 concentrating mechanism’in this alga at acid pH. The effect of denitrification on N2 fixation was studied ina denitrifying photosynthetic bacterium, Rhodopseudomonas sphaeroidesforma sp. denitrificans. KNO3 inhibited diazotrophic growthin light, but NC3–-dependent diazotrophic growth in darknesswas found. NO3– inhibited C2H2 reduction activity in lightin cells grown with NO3–. NO3–-dependent C2H2 reductionactivity in darkness also was present in cells grown with N2plus NO3–, but not in cells grown on glutamate with NO3–.NO3– repressed the synthesis of nitrogenase in light.This repression was not overcome by the addition of methioninesulfoximine. The inhibitory and repressive effect of NO3– was not mediatedby the NO2– produced from NC3– nor by the NH4+ excretedinto the medium. But, as N2 fixation is controlled by O2 (redoxcontrol) it seems to be mediated through the denitrificationprocesses. Much of the glutamine synthetase was adenylylatedin cells grown with NO3– and its adenylylation state closelyparallelled nitrogenase activity in the cells. The permeability of the plasmalemma of Chlamydomonas reinhardtiicells was increased by treatment with poly-L-lysine or dimethylsulphoxideas indicated by 3-phosphoglyceric acid dependent O2 evolution.These treatments decreased the ability of the cells to accumulateinorganic carbon internally and hence their photosynthetic affinityfor inorganic carbon in the medium. With saturating light andinorganic carbon, the photosynthetic rate was less affectedby the poly-L-lysine and dimethylsulphoxide treatments. Thusthe poly-L-lysine and dimethylsulphoxide did not alter the activityof the chloroplasts but rather made the intracellular inorganiccarbon pool more freely exchangeable with the medium. It isconcluded that the transporting system for inorganic carbonis located at the plasmalemma. Treatment with Diamox, an inhibitor of carbonic anhydrase, didnot affect photosynthetic rate and accumulation of inorganiccarbon when CO2 was supplied but strongly inhibited both parameterswhen HCO3 was supplied. In a mutant of Chlamydomonasreinhardtii lacking a cell wall, carbonic anhydrase leaks tothe medium and uptake of inorganic carbon is much faster whenCO2 is supplied than when HCO3 is supplied. Time courses of photosynthetic 14CO2 fixation and its simulationare presented for Chlorella cells grown under low CO2 concentration(low-CO2 cells) and subsequently exposed to 0.2 mM NaH14CO3or 130 ppm 14CO2 in the presence or absence of carbonic anhydrase(CA) in the suspending medium. It was shown that Chlorella cells utilized only free CO2 whenNaHCO3 was given in the presence or absence of CA, or when CO2was bubbled in the absence of CA. However, the present simulationindicated that both CO3 and HCO3 were utilized when CO2was given in the presence of CA. Based on these results, weconcluded that 1) Chlorella cells absorb only free CO2 and 2)this gas is provided to algal cells in two ways, i.e., by directand indirect CO2 supply. Usually, the dissolved CO2 is directlyutilized by the algal cells (direct supply of CO2). However,when the concentration of dissolved CO2 is extremely low andwhen there is CA, CO2 reconverted from HCO3 is also utilizedby Chlorella cells (indirect supply of CO2). The utilizationof HCO3 indicated by the above simulation was explainedby the indirect supply of CO2. We further assumed that the indirectsupply of CO2 to ribulose 1,5-bisphosphate carboxylase occursmainly in the chloroplasts of low-CO2 cells containing highCA. Permeability coefficients (PS values) for CO2 of the plasmamembrane (PM) of the unicellular green algae Eremosphaera viridis,Dunaliella parva, and Dunaliella acidophila, and of mesophyllprotoplasts isolated from Valerianella locusta were determinedfrom 14CO2 uptake experiments using the rapid separation ofcells by the silicone oil layer centrifugation technique. Theexperimental PS values were compared with calculated numbersobtained by interpolation of Collander plots, which are basedon lipid solubility and molecular size, for D. parva cells,mesophyll protoplasts isolated from Spinacia oleracea, mesophyllcells and guard cells of Valerianella, and guard cell protoplastsisolated from Vicia faba. The conductivity of algal plasma membranes for CO2 varies between0.1 and 9 ? 10–6 m s–1, whereas for the plasmalemmaof cells and protoplasts isolated from leaves of higher plantsvalues between 0.3 and 11 ? 10–6 m s–1 were measured.By assuming that these measurements are representative for plantsand algae in general, it is concluded that the CO2 conductivityof algal PM is of the same order of magnitude as that of thehigher plant cell PM. Ps values of plasma membranes for CO2are lower than those for SO2, but are in the same order of magnitudeas those measured for H2O. On the basis of these results itis concluded that theoretical values of about 3000 ? 10–6m s–1 believed to be representative for higher plant cells(Nobel, 1983) and which are frequently used for computer-basedmodels of photosynthesis, lack experimental confirmation andrepresent considerable overestimations. However, with severalsystems, including higher plant cells, the conductance of thePM for CO2 was significantly higher in light than in darkness.This suggests that in light, additional mechanisms for CO2 uptakesuch as facilitated diffusion or active uptake may operate inparallel with diffusional uptake. Reducing the concentration of sucrose in the culture mediumover successive subcultures has been tested as a method forincreasing the ability of rose shoots grown in vitro (Rosa cvsIceberg and Peace) to take up CO2. Shoots maintained on ‘constant’10, 20 and 40 g I–1 sucrose showed decreased levels ofCO2 uptake at higher sucrose concentrations, although cv. Peacegrew least at 10 g l–1 and showed correspondingly lowamounts of CO2 uptake compared with 20 and 40 g l–1. Bothcultivars died when sucrose was omitted from the medium. Assucrose concentration was reduced in the medium, so CO2 uptakeof shoots initially cultured on 20 and 40 g l–1 sucrosewas found to increase, although a concentration of 10 gl –1sucrose seemed to be limiting, below which the growth and chlorophylllevels of shoots declined. Colman, B. and Rotatore, C. 1988. Uptake and accumulation ofinorganic carbon by a freshwater diatom.—J. exp Bot 39:1025–1032. The mechanism of uptake of inorganic carbon and its accumulationhas been studied in the freshwater diatom Navicula pelliculosa.No external carbonic anhydrase could be detected, although itwas detected in cell extracts. The rate of photosynthetic O2evolution, in media in the range pH 7.5–8.5, exceededthe calculated rate of CO2 supply 2- to 5-fold, indicating thatHCO3 was taken up by the cells. At an external pH of7.5, the internal pH, measured by 14C-dimethyloxazolidine-2,4-dione distribution between the cells and the medium, was pH7.6 in the light and pH 7.4 in the dark. Accumulation of inorganiccarbon was determined by the silicone oil centrifugation methodand inorganic carbon pools of 23.5 mol m–3 were found,a concentration 21.6-fold that in the external medium. The resultsindicate an active accumulation of inorganic carbon againstpH and concentration gradients in this diatom, probably by activeHCO3 uptake. The photosynthetic response to CO2 concentration, light intensityand temperature was investigated in water hyacinth plants (Eichhorniacrassipes (Mart.) Solms) grown in summer at ambient CO2 or at10000 µmol(CO2) mol–1 and in winter at 6000 µmol(CO2)mol–1 Plants grown and measured at ambient CO2 had highphotosynthetic rate (35 µmo1(CO2) m–2 s–1),high saturating photon flux density (1500–2000) µmolm–2 s–1 and low sensitivity to temperature in therange 20–40 °C. Maximum photosynthetic rate (63 µmol(CO2)m–2 s–1) was reached at an internal CO2 concentrationof 800 µmol mol–1. Plants grown at high CO2 in summerhad photosynthetic capacities at ambient CO2 which were 15%less than for plants grown at ambient CO2, but maximum photosyntheticrates were similar. Photosynthesis by plants grown at high CO2and high light intensity had typical response curves to internalCO2 concentration with saturation at high CO2, but for plantsgrown under high CO2 and low light and plants grown under lowCO2 and high light intensity photosynthetic rates decreasedsharply at internal CO2 concentrations above 1000 µmol–1. Acclimation of single plants of Lolium temulentum to changing[CO2] was studied on plants grown in controlled environmentsat 20°C with an 8 h photoperiod. In the first experimentplants were grown at 135 µ;mol m–2 s–1 photosyntheticphoton flux density (PPFD) at 415µl l–1 or 550µll–1 [CO2] with some plants transferred from the lowerto the higher [CO2] at emergence of leaf 4. In the second experimentplants were grown at 135 and 500 µmol m–2 s–1PPFD at 345 and 575 µl l–1 [CO2]. High [CO2] during growth had little effect on stomatal density,total soluble proteins, chlorophyll a content, amount of Rubiscoor cytochrome f. However, increasing [CO2] during measurementincreased photosynthetic rates, particularly in high light.Plants grown in the higher [CO2] had greater leaf extension,leaf and plant growth rates in low but not in high light. Theresults are discussed in relation to the limitation of growthby sink capacity and the modifications in the plant which allowthe storage of extra assimilates at high [CO2]. Blooms of the toxic red tide phytoplankton Heterosigma akashiwo(Raphidophyceae) are responsible for substantial losses withinthe aquaculture industry. The toxicological mechanisms of H.akashiwoblooms are complex and to date, heavily debated. One putativetype of ichthyotoxin includes the production of reactive oxygenspecies (ROS) that could alter gill structure and function,resulting in asphyxiation. In this study, we investigated thepotential of H.akashiwo to produce extracellular hydrogen peroxide,and have investigated which cellular processes are responsiblefor this production. Within all experiments, H.akashiwo producedsubstantial amounts of hydrogen peroxide (up to 7.6 pmol min–1104 cells–1), resulting in extracellular concentrationsof ~0.5 µmol l–1 H2O2. Measured rates of hydrogenperoxide production were directly proportional to cell density,but at higher cell densities, accuracy of H2O2 detection wasreduced. Whereas light intensity did not alter H2O2 production,rates of production were stimulated when temperature was elevated.Hydrogen peroxide production was not only dependent on growthphase, but also was regulated by the availability of iron inthe medium. Reduction of total iron to 1 nmol l–1 enhancedthe production of H2O2 relative to iron replete conditions (10µmol l–1 iron). From this, we collectively concludethat production of extracellular H2O2 by H.akashiwo occurs througha metabolic pathway that is not directly linked to photosynthesis.  相似文献

A comparison of the activity and properties of the enzyme phosphoenolpyruvatecarboxylase (PEPC) was made for plants of Sedum telephium L.grown under low (70 µmol m–2 s–1) or high(500µmol m–2 s–1) PPFD and subjected to varyingdegrees of water stress. Under well-watered conditions onlyplants grown under high PPFD accumulated titratable acidityovernight and the extractable activity of PEPC was almost 2-foldhigher in these plants than in plants grown under low PPFD.Increasing drought stress resulted in a substantial increasein the activity of PEPC extracted both during the light anddark periods and a decrease in the sensitivity to inhibitionby malic acid. The magnitude of these changes was determinedby the severity and duration of drought and by light intensity.A comparison of the kinetic properties of PEPC from severelydroughted plants revealed that plants droughted under high PPFDhad a lower Km for PEP than plants under low PPFD. Additionof 2·0 mol m–3 malate resulted in an increase inthe Km for PEP, with plants draughted under low PPFD havinga significantly higher Km in the presence of malic acid comparedto those under high PPFD. Response to the activator glc-6-P,which lowered the Km for PEP, also varied between plants grownunder the two light regimes. Under well-watered conditions PEPCextracted from plants under high PPFD was more sensitive toactivation by glc-6-P than those under low PPFD. After the severedrought treatment, however, the Km for PEP in the presence ofglc-6-P was similar for enzyme extracted from plants grown underboth light regimes. Soluble sugars and starch were depletedovernight and were both possible sources of substrate for PEPC.With increasing drought, however, the depletion of starch relativeto soluble sugars increased under both light regimes. The propertiesof PEPC and the characteristics of carbohydrate accumulation/depletionare discussed in relation to the regulation of CAM in S. telephiumgrown under different light and watering regimes. The levels of purine and pyrimidine nucleotides in suspensioncultures of Catharanthus roseus were determined 24 h after stationary-phasecells were transferred to fresh complete (‘+Pi’)or phosphate-deficient (‘–Pi’) Murashige-Skoogmedium. The levels of ATP, GTP, UTP and CTP were from approx.3 to 5-fold greater in the cells grown in ‘+Pi’medium than in the cells grown in ‘–Pi’ medium.The levels of almost all other nucleotides were slightly higherin the cells in ‘+Pi’ medium. The rates of de novoand salvage biosynthesis of purine and pyrimidine nucleotideswere estimated from the rates of incorporation of radioactivityfrom [14C]formate, [2–14C]glycine, NaH14CO3, [6–14C]orotate,[8–14C]adenine, [8–14C]adenosine, [2–14C]uraciland [2–14C]uridine. The results indicated that the activityof both the de novo and the salvage pathway was higher in thecells in ‘+Pi’ medium than in the cells in ‘–Pi’medium. The rate of degradation estimated from the rate of releaseof 14CO2 from labelled purines and pyrimidines indicated thatdegradation of uridine was significantly reduced in the cellsin ‘+Pi’ medium, but no significant difference wasfound in the degradation of adenine, adenosine and uracil. Thepossible role of Pi in the control of the biosynthesis of nucleotidesand in the degradation of uridine is discussed. Treatment of S-sufficient or S-deficient Chlorella pyrenoidosacells with NaHSO3, during an 8-h period in the light, significantlydecreased their chlorophyll and dry matter contents when thecells were incubated in the presence or absence of SO42+. Incontrols lacking HSO3, when the starting pH was 7.5,dry matter and chlorophyll contents increased slightly, whereasno significant changes in either occurred at a starting pH of3.0 when the cells and medium contained SO4. In the dark,at both pH 3.0 and 7.5, dry matter and chlorophyll contentsdecreased slightly. Bisulphite treatment in the dark causedlittle decrease of either dry matter or chlorophyll when cellsand medium contained SO42–. However, in its absence, drymatter decreased markedly, but there was little change of chlorophyllcontent in the dark. The interactions between HSO3 asa source of S and as an inhibitor of growth and chlorophyllformation are discussed in the context of the changes inducedby light and alternative sources of S. Overall, the harmfuleffects of HSO3 outweigh any role it has as a sourceof S, since its effects are ameliorated by SO42–.  相似文献

Trends in several photosynthetic parameters and their responseto changed growth light were followed for 15 d in leaves ofyoung birch saplings using a rapid-response gas exchange measuringequipment. These in vivo measurements were compared to biochemicalassays that were made from the same leaves after the gas exchangestudies. The measurements were made on leaves that were selectedprior to the study and were at that time of similar age. Forthe first 7 d the photosynthetic parameters were followed fromthe growth conditions of moderate light (200 µmol m–2s–1; referred to as controls later in the text). On day7 some of the saplings were transferred to grow either underhigh (450 µmol m–2 s–1; referred to as highlight plants) or low (75 µmol m–2 s–1; referredto as low light plants) light and the capability of the preselectedleaves for acclimation was followed for 6 d. For comparison,at the end of the experiment the measurements were made on bothcontrols and on young leaves that had developed under high andlow light. Generally the in vivo measured rate of CO2 uptake (gross photosynthesis)both at 310 ppm CO2 and 2000 ppm CO2 corresponded very wellto the biochemically determined CO2 fixation capacity in vitroafter rapid extraction (measured as the initial and total activityof Rubisco, respectively). However, if the flux of CO2 intothe chloroplasts was limited by the closure of the stomata,as was the case of the high light plants, then the in vitromeasured Rubisco activity was greater than the in vivo measuredCO2 uptake. Vmax, calculated from the mesophyll conductanceat 1% O2, exceeded the initial activity of Rubisco (assayedat saturating RuBP and CO2) constantly by 60%. The catalyticactivity of Rubisco in birch leaves was overall very low, evenwhen calculated from the total activity of Rubisco (Kcat 0.63–1.18 s–1), when compared to herbaceous C3 species. Signs of light acclimation were not observed in most of thephotosynthetic parameters and in chloroplast structure whenmature birch leaves were subjected to changes in growth lightfor 6 d. However, the change of the growth light either to highor low light caused day-to-day fluctuations in most of the measuredphotosynthetic parameters and in the case of the high lightplants signs of photoinhibition and photodestruction were alsoobserved (decrease in the amount of chlorophyll and increasein chlorophyll a/b ratio). As a result of these fluctuationsthese plants achieved a new and lower steady-state conditionbetween the light and dark reactions, as judged from the molarratio of RuBP to Rubisco binding site. The uptake rate of carbon and nitrogen (ammonium, nitrate andurea) by the Microcystis predominating among phytoplankton wasinvestigated in the summer of 1984 in Takahamaira Bay of LakeKasumigaura. The Vmax values of Microcystis for nitrate (0.025–0.046h–1) and ammonium (0.15–0.17 h–1) were considerablyhigher than other natural phytoplankton. The ammonium, nitrateand urea uptake by Microcystis was light dependent and was notinhibited with nigh light intensity. The K1 values were farlower than the Ik values. The carbon uptake was not influencedby nitrogen enrichment. Microcystis accelerated the uptake rateby changing Vmax/K s value when nitrogen versus carbon contentin cells declined. Nitrate was scarcely existent in TakahamairiBay during the summer, when Microcystis usually used ammoniumas the nitrogen source. Inair-grown cells, O2 evolution dependent on low, but not high,HCO3 concentrations was strongly inhibited by the carbonicanhydrase inhibitor acetazolamide. Cells grown with 3% CO2 exhibitedlow rates of O2 evolution at low external inorganic C; however,after 30 min in air O2 evolution rates at low inorganic C approachedthose of air-grown cells. These results are compatible withthe view that Scenedesmus develops a ‘CO2 concentratingmechanism’ in air, with carbonic anhydrase as an importantconstituent When 3% CO2-grown cells were subjected to air-level of CO2,just a transient decline in NO3 utilization was observed,but in the presence of acetazolamide the rate of the processdecreased drastically in response to the decrease in the CO2level. In CO2-free air NO3 was taken up at high ratesbut in a deregulated manner, leading to release of NH4+. A portionof the NO3 taken up in the absence of CO2 was apparentlyassimilated Cellular nitrate reductase (NR) activity initially decreasedbut subsequently recovered after a transition from 3% CO2 toair. In the presence of acetazolamide, a persistent decreasein NR activity was observed. Cellular glutamine synthetase (GS)activity increased after transition from 3% CO2 to air, theactivity increase being unaffected by acetazolamide. NH4+ releaseto the medium in the presence of L-methionine-D, L-sulphoximine(MSO) transiently increased in 3% CO2-grown cells in responseto a transfer to air. MSO-induced NH4+ release was in fact higherin air-grown cells than in 3% CO2-grown cells. Glycollate wasinitially released after transition from 3% CO2 to air, butthere was no difference in glycollate release between MSO-treatedand untreated cells. Acclimation of fully developed leaves of Bischofia javanicaBlume to shadelight was examined. Seedlings were grown undersimulated daylight (1000 µmol m–2 s–1), thentransferred to a simulated shadelight (40 µmol m–2s–1). When a high-light leaf was transferred to low light, large negativenet photosynthetic rates (Pm) were recorded. This decrease wasrapid, but within 7 d the rate increased and became equal tothe low-light control leaf. These changes in photosynthesisdid not follow the pattern of changes in stomatal conductance(gs). Transfer to the low light resulted in a dramatic decreasein leaf weight per unit area (Lw), and most of the decreasesin Lw occurred within 3 d of transfer when the Pm of the transferredleaf was well below that of the low-light control leaf. There was a significant decrease in chlorophyll a in the transferredleaf without an appreciable change in chlorophyll b resultingin a large decrease in the chlorophyll a to chlorophyll b ratio.Leaf chlorophylls per unit area were higher in the transferredleaf than the low-light control leaf. Maximum photosyntheticrate in the transferred leaf was decreased by 40% compared tothat for the high-control leaf, but was almost at the same extenthigher than the low-light control leaf The results are discussedin the context of carbon gain capacity of its seedlings underlight-limiting forest understorey habitats.
