Tween 80 (0.1%, v/v) added to Thermomonospora curvata growing in minimal medium caused a transient lowering of the dry cell mass, decreased the optimal growth temperature of the thermophile from 62 to 54°C, and increased extracellular esterase activity. Cells grown in the presence of Tween 80 had decreased concentrations of branched chain fatty acids and increased concentrations of oleic acid. The detergent removed surface protuberances from mycelia and increased the liberation of enzymes active against crystalline cellulose, but did not stimulate liberation of enzymes active against carboxymethylcellulose, starch or pectin.  相似文献

Enzymatic conversion of 7-dehydrocholesterol to cholesterol by liver microsomes was increased by addition of Tween 80. This increase was proportional to Tween 80 concentration, and reached its maximum of 250% of baseline activity after addition of 300 μg/ml of Tween 80. This enhancement was comparable to that achieved by addition of 7.4 mg/ml of cytosol protein. No additive effect was observed with Tween 80 combined with cytosol protein. These data suggest that Tween 80 can substitute for sterol carrier protein 2 in the conversion of 7-dehydrocholesterol to cholesterol.  相似文献

The effects of the biosurfactant monorhamnolipid (monoRL) and the chemical surfactant Tween 80 on the degradation of phenol by Candida tropicalis CICC 1463 were studied. Both surfactants impeded the decay in cell concentrations at the beginning of the fermentation and enhanced the cell growth thereafter. They also increased the degradation efficiencies of 500 mg/L phenol from 86.9% in control to above 99.0% for all test concentrations within 30 h. The monoRL could also be degraded by the C. tropicalis. These results indicate that the surfactants could diminish the toxicity of phenol to the yeast, increase cell growth and improve phenol removal. The monoRL is better than Tween 80 because of biodegradability.  相似文献

Effect of Tween 80 on lipid vesicle permeability   总被引:1,自引:0,他引:1  
As part of an examination of procedures for protein/lipid reconstitution we incubated solubilized cytochrome oxidase at different temperatures with preformed vesicles composed of saturated lipids. We observed that the 22Na permeability of these vesicles was increased following such an incubation step, with the maximal effect occurring following incubations at temperatures in the vicinity of the lipid gel to liquid-crystal phase change. However, these permeability changes could be completely duplicated by incubating similar vesicles with concentrations of Tween 80 used to solubilize the extracted oxidase. Thus detergent molecules used to solubilize extracted membrane proteins can induce significant changes in the reconstituting lipid environment and the possibility of such changes must be taken into account when interpreting the properties of the reconstituted lipid/protein system under study.  相似文献

Tween 80 and proteose peptone effect on cellulase production with Trichoderma spec. was studied. Sugar cane pith was used in the medium as carbon source. Effect of Tween 80 on alkaloid-producing cultures ofClaviceps paspali   总被引:1,自引:0,他引:1  
Addition of Tween 80 to submerged cultures ofClaviceps paspali (Stevens and Hall) growing in a simple defined medium increased biomass formation and caused a temporary change in alkaloid synthesis intensity. The Tween-supplemented culture reached maximal alkaloid yields four days earlier than the control. The shift of alkaloid production was associated with a shift of organic acids and amino acids in the cell-pool. Thus the maximal formation of alkaloids was characterized by a decrease in the amount of citric acid, by the disappearance of succinic, fumaric and oxaloacetic acids and by increased accumulation of methionine, cysteine, alanine and histidine. The slow alkaloid synthesis was accompanied by a relatively high content of citric and succinic acids in the cell-pool and by the absence of methionine. The data are consistent with the hypothesis that ergot alkaloids participate in the regulation of the metabolism of cultures.  相似文献

1. Both Tween 80 and sodium fluoride significantly enhanced total extracellular glucosyltransferase activities of Streptococcus mutans. 2. Water-insoluble and water-soluble glucan formation were uniformly increased by Tween 80, whereas fluoride stimulated only water-soluble glucan formation. 3. Elevated glucan formation was due to an increase in enzymes secreted from bacterial cells. 4. Fatty acid composition and phospholipid content in bacterial membrane were changed by Tween 80, although sodium fluoride scarcely showed these changes. 5. Rapidly growing mycobacteria (14 species, 67 strains) showed a highly significant correlation between their susceptibility to smegmatocin (a Tween-hydrolysing esterase of Mycobacterium smegmatis exhibiting antimicrobial activity in the presence of Tween 80) and sensitivity to oleic acid (a hydrolysis product of Tween 80). Polyoxyethylenesorbitan ether, the other product of Tween 80 hydrolysis, also showed some toxic effect on smegmatocin-sensitive mycobacteria. From Mycobacterium diernhoferi ATCC 19340, a representative smegmatocin-sensitive strain, mutant strains with smegmatocin-resistant phenotypes were isolated. These mutants were more resistant to oleic acid than the parent strain, although their oleic acid resistance was somewhat lower than their smegmatocin resistance.  相似文献

Streptococcus salivarius (ATCC 25975) produced very low or nondetectable amounts of the extracellular enzyme glucosyltransferase (GTase) when grown in a chemically defined medium. The addition of Tween 80 to this medium resulted in the production of markedly enhanced levels of the enzyme. Oleic acid, the methyl ester of oleic acid, and sucrose each could not substitute for Tween 80 in this regard. The surfactant had no direct activating effect on performed enzyme activity. Tween 80 also stimulated the production of GTase by concentrated cells suspended in defined medium during a time when no measurable growth occurred. Under these conditions, the stimulatory effect of Tween 80 was blocked by chloramphenicol. It was further found that the surfactant dramatically stimulated the differential rate of GTase synthesis. These and other data strongly suggest that Tween 80 stimulates the production of extracellular GTase by acting either directly or indirectly at the level of enzyme synthesis.  相似文献

This study investigated the effects of surfactant additives and medium pH on mycelial morphology and exopolysaccharide (EPS) production in liquid culture of a valuable medicinal fungus Cordyceps sinensis Cs-HK1. In the medium containing 20 g l−1 glucose and 6 g l−1 peptone as the sole nitrogen source, the Cs-HK1 fungal mycelia formed smooth and spherical pellets about 1.8-mm mean diameter. The mycelial pellets became less uniform at pH (4.0–5.0) lower than the optimum (pH 6.0) or turned to filamentous form at higher pH (8–9). Surfactants added to the medium inhibited pellet formation, resulting in smaller and looser pellets. Tween 80 exhibited a remarkable promoting effect on EPS production, increasing the EPS yield more than twofold at 1.5% (w/v), which was most probably attributed to the stimulation of EPS biosynthesis and release from the fungal cells by Tween 80.  相似文献

Extracellular enzymes of microorganisms play an important role in the decomposition of macromolecules in the composting process. In this study, the effects of Tween 80 and rhamnolipid on the extracellular amylase, carboxymethyl cellulose enzyme (CMCase), xylanase and protease of Penicillium simplicissimum isolated from compost were investigated during solid-state fermentation. The results showed that the enzyme activities of amylase, CMCase and xylanase were increased by Tween 80 and rhamnolipid, which, however, had a negative effect on the protease production. The stimulative effects on the three enzymes were quite different during the whole fermentation process. Tween 80 and rhamnolipid also increased the fungal biomass slightly. As a result of the enhanced enzyme activities, the organic matter were also improved to different extents by both surfactants, and the decomposition rates of hemicellulose and cellulose were increased about 8.0% and 11.6% by Tween 80 at best, respectively, as well as 5% and 5.5% by rhamnolipid.  相似文献

Effect of Tween 80 on the growth of Mycobacterium avium complex   总被引:4,自引:0,他引:4  
The effect of Tween 80 on the growth of Mycobacterium avium complex (MAC) in liquid culture condition was investigated. Observation of the colony-forming units (CFU) and the morphology of MAC with transmission and scanning electron microscopy showed that Tween 80 at 0.05% in the medium (ca. 0.5 mg/ml) had bacteriostatic action and caused cell elongation. Tween 80 at 0.5% or more in the medium (ca. 5 mg/ml) reduced the quantity of MAC glycolipids and also led to false positive or positive results in biochemical tests for mycobacterial identification using nitrate reductase, urease, or arylsulfatase. To determine whether or not surfactants could reduce the MAC permeability barrier, the minimal inhibitory concentration (MIC) of antituberculosis drugs on MAC was determined in liquid medium with or without several kinds of surfactants including Tween 80. Five surfactants including Tween 80 increased the activity of antituberculosis drugs to MAC. These findings suggest that Tween 80 acts directly on the cell wall of MAC.  相似文献

Glucan production from sucrose by Streptococcus mutans OMZ 176 was stimulated approximately threefold in the presence of 0.1% Tween 80. When OMZ 176 was grown in a medium containing glucose, the glucosyltransferase level in the medium was also increased about fivefold in the presence of 0.1% Tween 80. The glucosyltransferase level increased in proportion to the logarithm of the concentration of Tween 80 in the glucose medium. Tween 80 affected neither bacterial growth nor the activity of glucosyltransferase. The appearance of glucosyltransferase in the glucose medium was inhibited immediately by chloramphenicol and actinomycin D and, after a lag, by rifampin as well. It was observed that the fatty acid composition of the cells grown with Tween 80 was altered. These results suggest that Tween 80 stimulates glucosyltransferase synthesis either directly, or indirectly by promoting glucosyltransferase secretion.  相似文献

AIMS: The aim of this study is to clarify the effect of Tween 80 on bile tolerance of lactococci. METHODS AND RESULTS: Four out of the six strains of lactococci could grow in broth containing 0.3% bile in the presence of 1% Tween 80, but grew slightly or not at all in the absence of Tween 80. Growing with Tween 80 altered the fatty acid composition of all three strains tested, but it is not clear which fatty acid influences bile tolerance. Material that absorbed light at 260 nm leaked from the cells tested with bile, but the leakage was decreased by addition of 1% Tween 80. Coincidentally, the decrease in the cell count by exposure to bile was suppressed by addition of Tween 80. CONCLUSIONS: Tween 80 enhances bile tolerance of some strains of lactococci. SIGNIFICANCE AND IMPACT OF THE STUDY: It is clarified that Tween 80 in the broth contributes to the bile tolerance of lactococci by reducing the cellular leakage caused by bile.  相似文献
