Roots of Phleum pratense L. were photographed during both vertical growth and gravitropic bending, and positions of anticlinal rhizodermal cell walls were digitized on the physically upper and lower flanks of the root in the curvature plane. By using B-splines, arc lengths of these positions, i.e. distances along the root surface, values of curvature, and relative elemental rates of elongation were estimated. The whole graviresponse can be divided into phases according to growth-rate values: (i) an increase of rates on the upper side of the root and a decrease on the lower side during the first 1–11/2h after the root has been moved from the vertical to a horizontal position, (ii) a transient equality of the rates on both sides, (iii) 2–3 h after the beginning of graviresponse, the growth gradient is inverted, and (iv) finally, after about 4 h, the growth rates of both flanks are approximately equal again. Curvature begins 15–20 min after horizontal placement of the root. During the first 2 h of graviresponse, plots of curvature versus arc length show one maximum value. After 2–21/2 h, two maximum values can be observed, the apical one near the root tip always keeping the same distance from the tip, the other one drifting basipetally relative to the growing tip. By evaluating photographs of high magnification, a group of six rhizodermal cells on each side of the root was identified which are the first cells showing gravitropic bending. These cells are located at the beginning of the elongation zone, enclosing the region 480–680 m from the root tip. The aim of this work was to assess whether agroforestry species have the ability to acquire P from pools unavailable to maize. Tithonia diversifolia(Hemsley) A. Gray, Tephrosia vogelii Hook f., Zea mays and Lupinus albusL. were grown in rhizopots and pH change and depletion of inorganic and organic P pools measured in the rhizosphere. Plants were harvested at the same growth stage, after 56 days for maize and white lupin and 70 days for tithonia and tephrosia, and the rhizosphere sampled. The rhizosphere was acidified by tithonia (pH change –0.3 units to pH 4.8) and lupins (–0.2 units to 4.9), alkalinised by tephrosia (+0.4 units to pH 5.4), and remained unchanged with maize growth. Concurrent with acidification in the rhizosphere of tithonia there was a decline in resin-P (0.8 g P g–1). However, there was also a decline in NaOH extractable inorganic P (NaOH-Pi) (5.6 g P g–1 at the root surface) and organic P pools (NaOH-Po) (15.4 g P g–1 at 1.5 mm from the root), which would not be expected without specific P acquisition mechanisms. Alkalinisation of tephrosia rhizosphere was accompanied by changes in all measured pools, although the large depletion of organic P (21.6 g P g–1 at 5 mm from the root) suggests that mineralisation, as well as desorption of organic P, was stimulated. The size of changes of both pH and P pools varied with distance away from the rhizoplane. Decline of more recalcitrant P pools with the growth of the agroforestry species contrasted with the effect of maize growth, which was negligible on resin-P and NaOH-Pi, but led to an accumulation of P as NaOH-Po (14.2 g P g–1 at 5 mm from the root). Overall the depletion of recalcitrant P pools, particularly Po, suggests that the growth of tithonia and tephrosia enhance desorption and dissolution of P, while also enhancing organic P mineralisation. Both species appear to have potential for agroforestry technologies designed to enhance the availability of P to crops, at least in the short term.  相似文献

One-year-old Prunus avium L. were grown under greenhouse conditions in a Countesswells soil in all combinations of 2 pH and 2 P levels. The soil, obtained from a long-term liming and fertilizer experiment, provided pH values throughout the experiment of 3.75–3.99 (pH 1) and 4.81–5.41 (pH 2). The P treatments had 0.43% acetic acid extractable P of 31–44 g g-1 (P1) and 145–173 g g-1 (P2). The trees were harvested 92 (H1), 134 (H2), and 168 (H3) days after initiation of growth.Top (leaf+new stem) dry weight was significantly increased for pH 2 and P2 at H2 and H3. P2 also increased leaf weight (H1), the weight of the original stem-root (H2 and H3), and root length but decreased root diameter at both soil pHs (H2 and H3). Total tree uptake of N, P, K, Ca, and Mg was also increased by pH-P combinations which had significantly greater dry matter production and root length. Total Mn uptake decreased at pH2. Root nutrient inflows (uM m-1 day-1) were increased for Ca at pH2 and for P at P2. Mn inflow decreased at pH2 and at pH1 P2 although the increased root length associated with the latter treatmen resulted in increased total tree Mn uptake. In general, high nutrient inflows occurred in all trees at H1 and in severely stunted trees at pH1 P1; both had larger than average root diameters.  相似文献

Dihydropyrimidinase from Pseudomonas stutzeri ATCC 17588 was purified 100-fold and characterized. It was found that dihydrouracil, dihydrothymine and hydantoin could serve as substrates for the partially purified enzyme. The K m values for dihydrouracil, dihydrothymine and hydantoin were determined to be 19.6 M, 21.3 M and 36.4 M, respectively, while their respective V max values were 0.836 mol/min, 0.666 mol/min and 2.21 mol/min. Between pH 7.5 and 9.0, enzyme activity was shown to be maximal. The optimum temperature for enzyme activity was 45 °C. Using gel filtration, the molecular weight of the enzyme was calculated to be approximately 115000 Da. Metal ions were found to influence the level of enzyme activity. Dihydropyrimidinase activity was stimulated by magnesium ions and inhibited by either zinc or copper ions.  相似文献

Three predictions of the acid-growth theory of fusicoccin (FC) action in inducing cell elongation were reinvestigated using abraded segments of maize (Zea mays L.) coleoptiles. i) Quantitative comparison of segment elongation and medium-acidification kinetics measured in the same sample of tissue shows that these FC-induced processes are strictly correlated in time and respond coordinately to cations present in the medium. ii) Fusicoccin (1 mol l-1) induces a rapid acidification of the cell-wall solution, reaching a final level of pH 3.8–4.0. Exogenous protons are able to substitute quantitatively for FC in causing segment elongation at pH 3.8–4.0. At pH 4, FC has no additional effect on cell elongation. iii) Neutral buffers (pH 7) completely abolish the FC-mediated growth response. iv) Cycloheximide (10 mg l-1) inhibits both FC-induced and acid-buffer(pH 4)-induced elongation after a lag of 40–45 min, and FC-induced H+ excretion after a lag of 2 h. Under the same conditions, indole-3-acetic acid-induced elongation and H+ excretion are inhibited without detectable lag. The contribution of Mg deficiency to Al stress in twelve different sorghum (Sorghum bicolor (L.) Moench) genotypes was investigated in nutrient solution culture under conditions of low Mg supply (between 50 and 1000 M) at two pH values. At pH 4.2, 30 M Al strongly inhibited Mg uptake. When dry matter yield was plotted as a function of the plant Mg concentration, similar response curves were obtained in the absence and the presence of Al with three genotypes. With many other genotypes dry matter yields of the control (without Al treatment) and Al-stressed plants were remarkably different at similar internal Mg concentrations, suggesting that growth had been suppressed not by Mg deficiency but by another factor, i.e. Al-induced root damage. At pH 4.8, 30 M Al hardly induced root damage but reduced Mg uptake and Al-induced Mg deficiency could almost completely account for the growth reaction of all genotypes. Therefore, at this pH the efficiency of uptake or use of Mg in different genotypes was the basis of their respective susceptibility to Al toxicity. When specific root length surpassed a certain critical range below 80–100 m per g dry root, growth control by Al-induced Mg deficiency was nearly abolished. Concentrations of ions were measured in soil solutions from beech (Nothofagus) forests in remote areas of New Zealand and in solutions from beech (Fagus sylvatica) and Norway spruce (Picea abies) forests in North-East Bavaria, West Germany, to compare the chemistry of soil solutions which are unaffected by acid deposition (New Zealand) with those that are affected (West Germany). In New Zealand, soil solution SO4 2– concentrations ranged between <2 and 58 mol L–1, and NO3 concentrations ranged between <1 and 3 mol L–1. In West Germany, SO4 2– concentrations ranged between 80 and 700 mol L–1, and NO3 concentrations at three of six sites ranged between 39 and 3750 mol L–1, but was not detected at the remaining three sites. At all sites in New Zealand, and at sites where the soil base status was moderately high in West Germany, pH levels increased, and total Al (Alt) and inorganic monomeric Al (Ali) levels decreased rapidly with increasing soil depth. In contrast, at sites on soils of low base status in West Germany, pH levels increased only slightly, and Al levels did not decline with increasing soil depth.Under a high-elevation Norway spruce stand showing severe Mg deficiency and dieback symptoms in West Germany, soil solution Mg2+ levels ranged between 20 and 60 mol L, and were only half those under a healthy stand. Alt and Ali levels were substantially higher the healthy stand than under the unhealthy stand, indicating that Al toxicity was not the main cause of spruce decline.  相似文献

The transformed root culture of Polygonum tinctorium Lour. was established by infecting leaf explants with Agrobacterium rhizogenes A4. These cultures were examined for their growth and indigo content under various culture conditions. Among the four different culture media tested, SH medium showed the highest yield for root growth (28 mg dry wt/30 ml) and indigo production (152 g/dry wt). In SH medium, 30 g sucrose l–1, 2500 mg KNO3 l–1, 300 mg NH4H2PO4 l–1 were the best conditions for indigo production at pH 5.7. The production of indigo in hairy roots slightly increased with the addition of 200 mg chitosan l–1 (186 g/dry wt) and 20 U pectinase l–1 (181 g/dry wt).  相似文献

This report investigates physical changes associated with the short-term inhibition of root elongation in intact maize seedlings (Zea mays L. vs. Halamish) by exogenous auxin. Movement of root tips was assayed by video microscopy in control roots, roots grown for 45 min in 10–6 M indole3-acetic acid (IAA), or roots chilled for 3 min at 11°C. IAA and chilling treatments similarly reduced root elongation rates (from 29 ± 6 m min–1 to 6 ± 2 m min–1). Initial rates of root tip contraction induced by 300 mOsmol mannitol were used to calculate tissue contractibility values. These allowed a comparison of effects of IAA and chilling treatments on apparent rates of water transport out of the root tip tissues. Chilling treatment reduced root tip contractibility by 66%, whereas IAA had much less effect (26% reduction). Roots were also exposed to an osmotic jump treatment; the initial osmotically induced increase in elongation rate was used to determine root tip extensibility values. Both IAA and chilling treatments reduced root tip extensibilities by 57%. Inhibition of wall-yielding properties, rather than hydraulic limitations, appeared to be primarily associated with inhibition of intact root tip elongation by exogenous IAA.  相似文献

Relative elemental growth rates (REGR) and lengths of epidermal cells along the elongation zone of Lolium perenne L. leaves were determined at four developmental stages ranging from shortly after emergence of the leaf tip to shortly before cessation of leaf growth. Plants were grown at constant light and temperature. At all developmental stages the length of epidermal cells in the elongation zone of both the blade and sheath increased from 12 m at the leaf base to about 550 m at the distal end of the elongation zone, whereas the length of epidermal cells within the joint region only increased from 12 to 40 m. Throughout the developmental stages elongation was confined to the basal 20 to 30 mm of the leaf with maximum REGR occurring near the center of the elongation zone. Leaf elongation rate (LER) and the spatial distributions of REGR and epidermal cell lengths were steady to a first approximation between emergence of the leaf tip and transition from blade to sheath growth. Elongation of epidermal cells in the sheath started immediately after the onset of elongation of the most proximal blade epidermal cells. During transition from blade to sheath growth the length of the blade and sheath portion of the elongation zone decreased and increased, respectively, with the total length of the elongation zone and the spatial distribution of REGR staying near constant, with exception of the joint region which elongated little during displacement through the elongation zone. Leaf elongation rate decreased rapidly during the phase when only the sheath was growing. This was associated with decreasing REGR and only a small decrease in the length of the elongation zone. Data on the spatial distributions of growth rates and of epidermal cell lengths during blade elongation were used to derive the temporal pattern of epidermal cell elongation. These data demonstrate that the elongation rate of an epidermal cell increased for days and that cessation of epidermal cell elongation was an abrupt event with cell elongation rate declining from maximum to zero within less than 10 h.Abbreviations LER leaf elongation rate - REGR relative elemental growth rates  相似文献

Taxus chinensis suspension cells were cultured in the modified Gamborg's B5 medium. Addition of 50 mg chitosan l–1, 60 M methyl jasmonate and 30 M Ag+ resulted in the greatest paclitaxel production, at 25 mg l–1 in the cultures, being almost 40 times higher than that of the control culture, 10 times higher than that of the culture exposed to Ag+, 6 times higher than that of the culture elicited by chitosan and almost double that of the culture elicited by methyl jasmonate.  相似文献

Cat gene expression has been investigated following PEG-mediated plasmid uptake into barley protoplasts. The uptake conditions optimised for transient expression were employed for stable transformation. Transformed protoplast-derived calli of the cvs. Dissa and Igri, were selected on medium containing G418 at 40 g ml–1 or kanamycin sulphate at 250 g ml–1. Absolute transformation frequencies of 28.9×10–5 and 21.3×10–5 were recorded for Dissa with kanamycin sulphate and G418 selection, respectively. The frequency for Igri was 11.5×10–5 with G418 selection. Four experimental predictions of the acid-growth theory of auxin (indole-3-acetic acid, IAA) action in inducing cell elongation were reinvestigated using abraded segments of maize (Zea mays L.) coleoptiles. i) Quantitative comparison of segment elongation and medium-acidification kinetics measured in the same sample of tissue reveals that these IAA-induced processes are neither correlated in time nor responding coordinately to cations present in the medium. ii) Exogenous protons are not able to substitute for IAA in causing segment elongation at the predicted pH of 4.5–5.0. Instead, external buffers induce significant segment elongation only below pH 4.5, reaching a maximal response at pH 1.75–2.5. Acid and IAA coact additively, and therefore independently, in the whole range of feasible pH values. iii) Neutral or alkaline buffers (pH 6–10) are unable to abolish the IAA-mediated growth response and have no effect on its lag-phase. iv) Fusicoccin, at a concentration producing the same H+ excretion as high concentrations of IAA, is ineffective in inducing segment elongation. Moreover, sucrose and other sugars can quantiatively substritute for IAA in inducing H+ excretion but are likewise ineffective in inducing elongation. Isolated embryos ofKarwinskia humboldtiana were cultured in vitro. The growth of embryos and development to plantlets on woody plant medium supplemented with indole-3-acetic acid 6.10-2 mol l–1, gibberellic acid (GA3) 3.10-2 mol l–1, and 6-benzylaminopurine (BA) 2 mol l–1 was obtained. Multiplication of shoots and rooting of excised shoots has been achieved. Gibberellins in the embryo-suspensor system have been considered so far only in Phaseolus coccineus. We present in this report the localization of gibberellin-like substances in the suspensors of Tropaeolum majus L. and Cytisus laburnum L. The total gibberellin activity (expressed as gibberellic-acid equivalent in the -amylase bioassay) in 2000 suspensors (106 mg fresh weight; FW) of C. laburnum and in 600 suspensors (236 mg FW) of T. majus were 50.9 g g-1 FW and 8.9 g g-1 FW respectively.Abbreviation GA gibberellin  相似文献

The rate of glycolate excretion in Euglena gracilis Z and some microalgae grown at the atmospheric level of CO2 was determined using amino-oxyacetate (AOA). The extracellular O2 concentration was kept at 240 M by bubbling the incubation medium with air. Glycolate, the main excretion product, was excreted by Euglena at 6 mol·h-1·(mg chlorophyll (Chl))-1. Excretion depended on the presence of AOA, and was saturated at 1 mM AOA. A substituted oxime formed from glyoxylate and AOA was also excreted. Bicarbonate added at 0.1 mM did not prevent the excretion of glycolate. The excretion of glycolate increased with higher O2 concentrations in the medium, and was competitively inhibited by much higher concentrations of bicarbonate. Aminooxyacetate also caused excretion of glycolate from the green algae, Chlorella pyrenoidosa, Scenedesmus obliquus and Chlamydomonas reinhardtii grown on air, at the rates of 2–7 mol·h-1·(mg Chl)-1 in the presence of 0.2–0.6 mM dissolved inorganic carbon, but the cyanobacterium, Anacystis nidulans, grown in the same way did not excrete glycolate. The basis for the outcome of competition between sulfidogens and methanogens for H2 was examined by comparing the kinetic parameters of representatives of each group separately and in co-culture. Michaelis-Menten parameters (V max and K m) for four methanogens and five sulfate-reducing bacteria were determined from H2-depletion data. Further, Monod growth parameters (max, K s, Y H2) for Desulfovibrio sp. G11 and Methanospirillum hungatei JF-1 were similarly estimated. H2 K m values for the methanogenic bacteria ranged from 2.5 M (Methanospirillum PM1) to 13 M for Methanosarcina barkeri MS; Methanospirillum hungatei JF-1 and Methanobacterium PM2 had intermediate H2 K m estimates of 5 M. Average H2 K m estimates for the five sulfidogens was 1.2 M. No consistent difference among the V max estimates for the above sulfidogens (mean=100 nmol H2 min-1 mg-1 protein) and methanogens (mean=110 nmol H2 min-1 mg-1 protein) was found. A two-term Michaelis-Menten equation accurately predicted the apparent H2 K m values and the fate of H2 by resting co-cultures of sulfate-reducers and methanogens. Half-saturation coefficients (K s) for H2-limited growth of Desulfovibrio sp. G11 (2–4 M) and Methanospirillum JF-1 (6–7 M) were comparable to H2 K m estimates obtained for these organisms. Maximum specific growth rates for Desulfovibrio sp. G11 (0.05 h-1) were similar to those of Methanospirillum JF-1 (0.05–0.06 h-1); whereas G11 had an average yield coefficient 4 x that of JF-1. Calculated max and V max/K m values for the methanogens and sulfidogens studied predict that the latter bacterial group will process more H2 whether these organisms are in a growing or resting state, when the H2 concentration is in the first-order region.  相似文献

We examined the effect of calmodulin (CaM) antagonists applied at the root tip on root growth, gravity-induced root curvature, and the movement of calcium across the root tip and auxin (IAA) across the elongation zone of gravistimulated roots. All of the CaM antagonists used in these studies delayed gravity-induced curvature at a concentration (1 M) that did not affect root growth. Calmodulin antagonists ( 1M) inhibited downward transport of label from 45Ca2+ across the caps of gravistimulated roots relative to the downward transport of 45Ca2+ in gravistimulated roots which were not treated with CaM antagonists. Application of CaM antagonists at the root tip ( 1M) also decreased the relative downward movement of label from 3H-IAA applied to the upper side of the elongation zone of gravistimulated roots. In general, tip application of antagonists inhibited neither the upward transport of 45Ca2+ in the root tip nor the upward movement of label from 3H-IAA in the elongation zone of gravistimulated roots. Thus, roots treated with CaM antagonists ( 1 M) become less graviresponsive and exhibit reduced or even a reversal of downward polarity of calcium transport across the root tip and IAA transport across the elongation zone. The results indicate that calmodulin-regulated events play a role in root gravitropism.  相似文献

Homogeneous phosphoribulokinase (PRK; ATP: d-ribulose-5-phosphate 1-phosphotransferase, EC 2.7.1.19) was isolated from wheat leaves with a specific activity of 15 kat mg-1 protein. The purification included ammonium sulfate cuts, isoelectric precipitation, and hydrophobic and affinity chromatography on pentylagarose and Blue Sepharose CL 6B, respectively. Gel filtration of the purified enzyme yielded a 83000 Da protein. Subunits of about 42000 Da were estimated from sodium dodecyl sulfate-polyacrylamide gels. Wheat leaf PRK was stable for at least four weeks when stored at 4°C. Saturation curves for ribulose 5-phosphate (Ru5P) and ATP followed Michaelis-Menten kinetics (K m values: K m Ru5P=50–80 M; K m ATP=70 M). The saturation curve for MgCl2 was sigmoidal (half-maximal velocity <0.5 mM). The affinity for Ru5P, ATP and Mg2+ was not affected by pH changes comparable to pH shifts in the stroma. In contrast to chloroplast fructose-bisphosphatase (Zimmermann et al. 1976, Eur. J. Biochem. 70, 361–367) the affinity for ligands remained unchanged in the dithiothreitol-activated and in the non-activated state. Forty strains ofRhizobium phaseoli, isolated from Kenyan soils, were tested for infectiveness on common bean (Phaseolus vulgaris L.). 28 strains were infective and a cultivar × Rhizobium interaction was observed. 48 strains were screened for tolerance of acidity and Al in liquid culture. Assessment of visible turbidity after 14 days indicated 34 strains tolerant of pH 4.5 but none tolerant of pH 3.5. No strain was tolerant of 50 M Al at pH 5.5. Three strains were tolerant of 20 M Al at pH 5.5 and 10 M Al at pH 4.5. Screening on a solid medium identified strains tolerant of 20 and 50 M Al at pH 5.5 and 4.5 which were sensitive to these treatments in liquid culture. Those strains tolerant to 20 M Al at pH 4.5 and 5.5 in liquid culture were correctly identified on the solid medium.  相似文献
