In this study, sensitivities of 156 Sclerotinia sclerotiorum isolates collected from sunflower fields of West Azarbaijan province, Iran, were assessed to carbendazim and iprodione, and the baseline sensitivities were established for azoxystrobin and tebuconazole. Resistance to carbendazim and iprodione was observed in 53.85% and 4.49% of the isolates, respectively. The 50% effective concentration (EC50) values of azoxystrobin for the isolates ranged from 0.017 to 3.515 μg/ml with a mean of 0.330 μg/ml, and 8.97% of the strains showed low levels of resistance to the fungicide. However, in the presence of salicylhydroxamic acid, all isolates were sensitive to azoxystrobin and EC50 values ranged from 0.015 to 0.263 μg/ml with a mean of 0.086 μg/ml. All isolates were found to be sensitive to tebuconazole, and EC50 values ranged from 0.003 to 0.177 μg/ml with a mean of 0.036 μg/ml. Among the multiple-resistant isolates, the strains exhibiting resistance to both carbendazim and iprodione were detected in the highest frequency (4.49%). No correlation was observed between mycelial growth and aggressiveness with fungicide sensitivity of the isolates suggesting the absence of fitness cost associated with resistance to the studied fungicides. The results indicated that iprodione, azoxystrobin and tebuconazole could be effectively used in rotation or mixture in spray programmes to manage S. sclerotiorum in the region. The baselines established for azoxystrobin and tebuconazole would be useful in monitoring the fungal populations in the province to assess possible shifts in fungicide sensitivity of S. sclerotiorum isolates in the future.  相似文献

The effects of various fungicides on mycelial growth and spore germination of Ascochyta rabiei were determined by incorporating them into potato dextrose agar and measuring colony diameter and observing colony growth and spore germination at 20 ± 2°C. Eight fungicides prevented spore germination of the pathogen at concentrations of 0.125–2 μg/ml, three hindered mycelial growth at 2–4 μg/ml and seven failed to inhibit mycelial growth even at 128 μg/ml. The reference fungicide for the pathogen, chlorothalonil, stopped conidial germination at low rates but did not prevent mycelial growth at 128 μg/ml. Thirteen fungicides were tested against seed infections of the pathogen, and benomyl + thiram, carbendazim and carbendazim + chlorothalonil seed treatments gave more than 85% inhibition on both vacuum‐infiltrated and naturally infected seeds. Coating the seeds with polymers did not increase the effectiveness of fungicides. Three fungicides; (azoxystrobin, chlorothalonil and mancozeb), gave the highest protection in the field but protection decreased with increased inoculum pressure. Addition of humic acid to fungicide suspensions did not affect their performance.  相似文献

Pepper Phytophthora blight (PPB), caused by Phytophthora capsici, is an important disease of pepper in China. The extensive application of metalaxyl has resulted in widespread resistance to this fungicide in field. This study has evaluated the activities of several fungicides against the mycelial growth and sporangium germination of metalaxyl‐sensitive and metalaxyl‐resistant P. capsici isolates by determination of EC50 values. The results showed that the novel carboxylic acid amide (CAA) fungicide mandipropamid exhibited excellent inhibitory activity against PPB both in vitro and in vivo, with averagely EC50 values of 0.075 and 0.004 μg/ml in mycelial growth and sporangium germination, respectively, and over 88% efficacy in controlling PPB. The other three CAA fungicides also provided over 70% efficacy in controlling PPB. The mycelial growth was less sensitive to quinone outside inhibitor (QoI) fungicides azoxystrobin and trifloxystrobin than that of sporangium germination in P. capsici isolates. However, azoxystrobin and trifloxystrobin provided over 80% efficacy in controlling PPB. It was noted that propamocarb and cymoxanil did not exhibit activity against the mycelial growth or sporangium germination of P. capsici isolates in the in vitro tests, with over 70% efficacy in controlling PPB. The new fungicide mixture 62.5 g/l fluopicolide + 625 g/l propamocarb (trade name infinito, 687.5 g/l suspension concentrate (SC)) produced over 88% efficacy in controlling PPB caused by both metalaxyl‐sensitive and metalaxyl‐resistant isolates. The data of this study also proved that there was obviously no cross‐resistance between metalaxyl and the other tested fungicides. Therefore, these fungicides should be good alternatives to metalaxyl for the control of PPB and management of metalaxyl resistance.  相似文献

The sensitivity of Venturia inaequalis to trifloxystrobin and difenoconazole was studied in Uruguay. Populations of V. inaequalis were collected from apple orchards with different histories of trifloxystrobin use. Sensitivity of populations to trifloxystrobin was analysed using a method for testing spore germination published by FRAC, using a discriminatory concentration of 2.0 mg a.i./l. Resistance to trifloxystrobin was widespread in the region of commercial apple production with resistance detected in all orchards examined, the incidences ranging from 3% to 95%. The highest frequencies were found in orchards with the most intensive use of Quinone outside inhibitors (QoI) fungicides. Sensitivities of isolates of V. inaequalis to difenoconazole were assessed at five concentrations using a mycelial growth assay on isolates (33 isolates per orchard) from one non‐commercial (baseline orchard) and two commercial orchards having differing histories of difenoconazole use. Populations in both commercial orchards exhibited reduced sensitivities to difenoconazole compared to the baseline orchard. Resistance factor (RF) values of 6.6 and 11.74 were measured in the orchards with moderate (up to 4 sprays per season) and intensive use (more than 5 sprays per season) of difenoconazole, respectively. A single‐assessment concentration (SAC) was identified for assessing difenoconazole sensitivity of V. inaequalis isolates by fitting linear regressions between log10 EC50 and relative growth (RG) of the isolates at each fungicide concentration testing, and comparing the goodness‐of‐fit of the regression lines. Comparable results were obtained based on EC50 values and RG values at a SAC of 0.05 mg of active ingredient per litre (a.i. per l). Populations from both commercial orchards differed from the baseline population, in that isolates with RG ≥70 were present at substantial levels in the former but absent from the latter.  相似文献

Grey mould, caused by the fungus Botrytis cinerea Pers ex Fr., is a very destructive and important disease worldwide. Fluazinam is a phenylpyridinamine fungicide with broad‐spectrum activities. The baseline sensitivity of B. cinerea to fluazinam is yet to be established in Henan Province, China. In this study, a total of 117 field isolates of B. cinerea were collected from 49 commercial greenhouses in different locations of Henan Province, in 2016, and the sensitivities of these isolates to fluazinam were determined based on mycelial growth. The effective concentration for 50% (EC50) values ranged from 0.0038 to 0.0441 μg/ml, and the mean EC50 value was 0.0201 ± 0.0081 μg/ml for mycelial growth. The frequency distribution range presented a unimodal curve. To define the cross‐resistance relationships, the linear correlation coefficients of the EC50 values between fluazinam and carbendazim, procymidone, pyrimethanil or boscalid were analysed. The results showed that no correlation was observed between fluazinam and the other tested fungicides. These results provide important information to growers for the prevention and control of grey mould.  相似文献

Sensitivity to azoxystrobin and kresoxim‐methyl of 80 single‐spore isolates of Magnaporthe oryzae was determined. The EC50 values for azoxystrobin and kresoxim‐methyl in inhibiting mycelial growth of the 80 M. oryzae isolates were 0.006–0.056 and 0.024–0.287 µg mL?1, respectively. The EC50 values for azoxystrobin and kresoxim‐methyl in inhibiting conidial germination of the M. oryzae populations were 0.004–0.051 and 0.012–0.105 µg mL?1, respectively. There was significant difference in sensitivity to azoxystrobin or kresoxim‐methyl between the tested isolates representing differential sensitivity to carbendazim (MBC) and kitazin P (IBP); however, there was no correlation between this difference in sensitivity to azoxystrobin or kresoxim‐methyl and sensitivity to MBC or IBP, indicating that there was no cross‐resistance between azoxystrobin or kresoxim‐methyl and MBC or IBP. In the protective and curative experiments, kresoxim‐methyl exhibited higher protective and curative activity than azoxystrobin when applied at 150 and 250 µg mL?1 accordingly, while azoxystrobin exhibited stronger inhibitory activity against M. oryzae isolates than that of kresoxim‐methyl in the in vitro test. The results of field experiments also suggested that both azoxystrobin and kresoxim‐methyl at 187.5 g.a.i. ha?1 gave over 73% control efficacy in both sites, exhibiting excellent activity against rice blast. Taken together, azoxystrobin and kresoxim‐methyl could be a good substitute for MBC or IBP for controlling rice blast in China, but should be carefully used as they were both at‐risk.  相似文献

The sensitivity of Alternaria solani isolates to the fungicides mancozeb and chlorothalonil was evaluated, to determine if inadequate disease management by these fungicides could be attributed to reduced sensitivity of A. solani isolates to these fungicides. The sensitivity of 60 isolates of A. solani was assessed using the inhibition of radial mycelial growth (RG) method, using fungicide concentrations of 0, 1.0, 10, 100, 500 and 1000 μg a.i ml?1 medium. EC50 was calculated for each isolate and fungicide combination. The EC50 values of different A. solani isolates to mancozeb ranged from 9.05 to 712.65 μg ml?1. EC50 values of different isolates to chlorothalonil ranged from 4.25 to 849.4 μg ml?1. The percentage of isolates with reduced sensitivity was 46.7 and 53.3% for mancozeb and chlorothalonil, respectively. Results of the in vivo tests demonstrated decline in disease control by the two fungicides with the reduced-sensitivity isolates compared to the sensitive ones.  相似文献

Two inhibitors, aviglycine and propargylglycine, were tested for their ability to suppress methionine synthesis thus inhibit conidial germination and mycelial growth of Czapek-Dox liquid medium grown Fusarium oxysporum f. sp. luffae μM. The linear inhibition range for mycelial growth was about 7.6–762.9 μM. Although aviglycine did not completely inhibit both conidial germination and mycelial growth, it showed significant inhibitory effect at 1.5 μM. The inhibition range for propargylglycine against conidial germination and mycelial growth were from 0.08 to 8841 μM and from 0.8 to 884.1 μM, respectively. Propargylglycine inhibited conidial germination and mycelial growth at a concentration of 8841 μM. The EC50 values of aviglycine were 1 μM for conidial growth and 122 μM for mycelial growth, and the EC50 values of propargylglycine were 47.7 μM for conidial growth and 55.6 μM for mycelial growth. Supplement of methionine released inhibition of aviglycine or propargylglycine to conidial germination. In addition, a mixture of aviglycine (1.5 μM) and propargylglycine (8841 μM) showed additive inhibitive effect than applied alone on 10 isolates. From these results, both aviglycine and propargylglycine exhibited inhibitory activity, and suggest that they can provide potential tools to design novel fungicide against fungal pathogens.  相似文献

Sclerotinia stem rot, caused by Sclerotinia sclerotiorum, is a devastating disease in Henan Province, of the main rapeseed production areas in China. Fluazinam belongs to the broad‐spectrum phenylpyridinamine fungicides, which have high activity in inhibiting the mycelial growth of S. sclerotiorum. In this study, 191 field isolates were obtained from different oilseed rape fields in Henan Province, before being exposed to fluazinam in 2015. The baseline sensitivity of S. sclerotiorum to fluazinam was established. The effective concentration for 50% inhibition of mycelial growth (EC50) ranged from 0.0019 to 0.0337 μg/ml, and the mean EC50 value was 0.0084 ± 0.0055 μg/ml. The range of the frequency distribution was narrow. The results of a cross‐resistance assay revealed no cross‐resistance between fluazinam and carbendazim, dimethachlone, boscalid or fludioxonil. Field efficacy tests showed that the control efficacies of fluazinam (50% WG) applied at 150, 225 and 300 g ai ha?1 were 67%, 73% and 88%, respectively. In contrast, the control efficacies of boscalid (50% WG) and carbendazim (50% WP) applied at 225 and 1,500 g ai ha?1 were 71% and 52%, respectively.  相似文献

The influence of 15 commercially available fungicides on the germination, growth and virulence of Metarhizium anisopliae, Beauveria bassiana, Isaria fumosorosea, and Lecanillium longisporum was evaluated. The influence of the fungicides on conidial germination was dependant on the fungicide type and dose. Most fungicides retarded conidial germination of all the fungi tested at 10× and at the recommended rate of application, however, their toxicity declined at lower concentrations. Most of the fungicides inhibited mycelial growth of B. bassiana, whereas L. longisporum growth was unaffected. Only two and eight fungicides influenced mycelial growth of I. fumosorosea and M. anisopliae, respectively. None of the fungicides influenced the virulence of B. bassiana and L. longisporum, however, tolylfluanid and azoxystrobin reduced the virulence of M. anisopliae and I. fumosorosea, respectively. These studies clearly show that certain fungicides have the potential to inhibit germination of entomopathogenic fungi in vitro but appear to have little or no effect on their virulence against target insects.  相似文献

Triazole fungicides, which are sterol demethylation inhibitors, have become the primary systemic fungicides applied to cucurbits to control gummy stem blight caused by Didymella bryoniae. Isolates of D. bryoniae from South Carolina that were never exposed to tebuconazole or exposed for several years were tested for sensitivity to tebuconazole and difenoconazole. Colony diameters, percentage germination of ascospores and conidia, and germ tube lengths were measured when isolates were grown on agar amended with 0.10–10.0 mg/l tebuconazole and 0.01–1.0 mg/l difenoconazole. All 147 isolates tested were sensitive to tebuconazole and difenoconazole with mean EC50 values of 0.41 and 0.054 mg/l, respectively. Ascospore germination was greater than conidia germination on fungicide‐amended agar. Although the length of germ tubes arising from both spore types was reduced by both fungicides, the reduction was greater for ascospore germ tubes than for conidia germ tubes. Because many watermelon growers rotate crops among fields every two years, local populations of D. bryoniae have not been exposed repeatedly to tebuconazole. In addition, growers often apply a rotation of systemic and contact fungicides. Thus, despite exposure to tebuconazole for up to nine years, isolates of D. bryoniae from South Carolina remain sensitive to triazole fungicides.  相似文献

The effects of eight commonly used fungicides: propamocarb, fenarimol, triadimefon, procimidone, azoxystrobin, carbendazim, cooper oxychloride and Trichoderma harzianum on germination, mycelial growth and virulence of Isaria fumosorosea (Wize) Brown & Smith (Ascomycota: Hypocreales) was studied. The greatest effect on germination was observed by azoxystrobin, followed by carbendazim, triadimefon and procimidone. Propamocarb, fenarimol, cooper oxychloride and T. harzianum did not affect conidial germination with germinations of 95, 93, 79 and 84%, respectively. Virulence was evaluated against early fourth instar nymphs of Trialeurodes vaporariorum (Hemiptera: Aleyrodidae). The mix containing fungal conidial suspensions plus cooper oxychloride or fenarimol presented a significant reduction in the mortality in comparison with spore suspension itself. This study suggests that the most appropriate fungicides for use in Integrated Pest Management Programs against T. vaporariorum in combination with I. fumosorosea are propamocarb and T. harzianum.  相似文献

The filamentous plant pathogenic fungus Fusarium graminearum is one of the most important pathogens causing Fusarium head blight (FHB) in wheat in the Henan Province of China. Metconazole is among the demethylation inhibitor (DMI) fungicides with a higher inhibitory activity on the mycelial growth of F. graminearum. In 2016 and 2017, 119 single spore isolates of F. graminearum, prior to being exposed to metconazole, were recovered from 52 wheat fields near 11 cities in Henan Province. The inhibitory activity of metconazole on the mycelia of the Henan F. graminearum population was determined, and EC50 values were calculated. The range of EC50 values of the Henan F. graminearum population to metconazole was 0.0103 to 0.0775 μg/ml with an average EC50 value of 0.0293 ± 0.0114 μg/ml. The sensitivity frequency distribution curve presented a single peak in a narrow range. No cross-resistance was found between the DMI fungicide metconazole and the benzimidazole fungicide carbendazim or the cyanoacrylate fungicide phenamacril. Therefore, these sensitivity data could be used as the baseline of F. graminearum susceptibility to metconazole in the Henan Province and provide the basis for monitoring metconazole resistance in this area.  相似文献

An outbreak of grey leaf spot caused by Stemphylium solani was observed on tomato in Shandong Province of China in recent years and brought huge economical losses. Fludioxonil is a phenylpyrrole fungicide with strong antifungal activity against S. solani. To evaluate the risk of S. solani developing fludioxonil resistance, a total of 145 field isolates were examined for sensitivity to fludioxonil by measuring mycelial growth. The baseline sensitivity was distributed as a unimodal curve with a mean EC50 value of 0.0659 (±0.0170) µg mL?1. Five mutants with high resistance to fludioxonil (RF > 1000) were obtained by successively selecting on fludioxonil‐amended plates in the laboratory. All the resistant mutants associated with strongly reduced fitness in mycelial growth, sporulation and pathogenicity. Fludioxonil had positive cross‐resistance with procymidone and iprodione, but there was no cross‐resistance with other fungicides including boscalid, fluazinam, azoxystrobin and flusilazole. Based on the current results, resistance risk of S. solani to fludioxonil could be moderate. This is the first report of baseline sensitivity of S. solani to fludioxonil and its risk assessment. In order to delay the resistance development, it is recommended that fludioxonil can be used as one component of the mixture or fungicides with different modes of action should be alternatively used for this disease management.  相似文献

This study assessed the fenhexamid sensitivity of 143 Botrytis cinerea isolates collected from greenhouse strawberries in five regions of China between 2012 and 2013, and identified four isolates with moderate levels of resistance: two from the Xinjiang Uygur Autonomous Region and two from Hebei Province. The baseline fenhexamid sensitivity of B. cinerea exhibited a unimodal distribution with a mean EC50 value of 0.20 ± 0.10 μg/ml (SD). The EC50 values of the fenhexamid‐resistant isolates ranged from 0.05 to 0.40 μg/ml. Molecular analysis of the fenhexamid target gene erg27 revealed that the resistant isolates collected from Xinjiang (163‐6 and 163‐22) contained three mutations that led to amino acid changes (V365A, E368D and A378T) known to be associated with fenhexamid resistance, but that the isolates from Hebei lacked any mutations, indicating that an alternative mechanism could be responsible for their resistance. Most of the biological characteristics of the fenhexamid‐resistant isolates, such as mycelial growth, sclerotia production and pathogenicity, did not significantly differ from those of the sensitive ones ( .05), but it was noted that some of the resistant isolates exhibited reduced rates of sporulation and spore germination. In addition, the resistant isolates exhibited lower osmotic sensitivity than the sensitive ones. The study found no evidence of cross‐resistance with other fungicides, but that there was negative cross‐resistance with procymidone, iprodione, carbendazim and pyraclostrobin, which indicates that the inclusion of these fungicides within an integrated pest management (IPM) programme could help to minimize the risk of fenhexamid resistance developing in B. cinerea.  相似文献

The aim of the present investigation was to determine the impact of relative humidity (RH) and temperature on conidial germination, nuclear position and effect of important fungicides on growth and conidial germination of Cercospora canescens. Germination of conidia was observed at RH range 92–100% at 5–35°C. Significant interaction between temperature and RH indicated that higher humidity and high temperature promoted quick germination both in the presence and absence of free moisture. Although in absence of free moisture at 92–95% RH higher temperatures 25–35°C promoted quick evaporation of moisture and no conidial germination. Number of germtube was increased significantly at the optimum temperature 25–30°C and higher humidity (98–100%). But higher temperature 25–35°C with lower RH did not support the conidial germination. This finding is very important for disease forecasting using meteorological data. The spray of Carbendazim as contact fungicide may not be useful since it is not effective against the conidia of C. canescens. Triadimefon did not inhibit the conidia germination but completely inhibited mycelium development at 50 μg/ml. Propriconazole inhibited both conidia germination and mycelial development. Therefore, Propiconazole may be taken as protective as well as curative spray. In non-systemic fungicide, Copper oxychloride gave anticipated result by inhibiting both conidial germination and mycelium development. Therefore, copper oxychloride can be used as protectant fungicides for Cercospora leaf spot caused by C. canescens.  相似文献

Using nematophagous fungi for the biological control of animal parasitic nematodes will become one of the most promising strategies in the search for alternative chemical drugs. The purpose of this study was to check the in vitro activity of four anthelmintics, four chemical fungicides and two antifungal drugs on the spore germination of nematophagous fungi: Duddingtonia flagrans (SF170), Arthrobotrys oligospora (447), Arthrobotrys superba (435) and Arthrobotrys sp. (PS011). A modified 24-well cell culture plate assay was conducted to evaluate the susceptibility of nematophagous fungi against drugs tested by calculating the effective middle concentrations (EC50) of each tested drug to inhibit the germination of fungal spores. EC50 ranged between 0·7 and 47·2 μg ml−1 for fenbendazole, thiabendazole and ivermectin, except levamisole (546·5–4057·8 μg ml−1). EC50 of tested fungicides was 0·6–2·3 μg ml−1 for carbendazim, 55·9–247·4 μg ml−1 for metalaxyl, 24·4–45·2 μg ml−1 for difenoconazole, and 555·9–1438·3 μg ml−1 for pentachloronitrobenzene (PCNB). EC50 of two antifungal drugs was 0·03–3·4 μg ml−1 for amphotericin B and 0·3–10·9 μg ml−1 for ketoconazole. The results showed that 10 tested drugs, except for levamisole and PCNB, had in vitro inhibitory effects on nematophagous fungi. The chlamydospores of Dflagrans had the highest sensitivity to nine tested drugs, except for ketoconazole.  相似文献

Succinate dehydrogenase inhibitor (SDHI) fungicides are important tools to control preharvest and postharvest brown rot of stone fruits. In this study we determined the mycelial growth rate of Monilinia fructicola isolates on various media and picked the three that allowed the fastest growth to assess the sensitivity of mycelium to SDHI fungicides boscalid, fluopyram, and penthiopyrad. Minimal medium (MM) supported mycelial growth the best and yielded lowest EC50 values for three SDHI fungicides. EC50 values corresponded with disease incidence data obtained from detached fruit assays. Penthiopyrad had significantly greater intrinsic activity in vitro compared to fluopyram at the α = 0.05 level and compared to boscalid at the α = 0.1 level. However, detached fruit assays revealed that this ‘advantage’ did not carry over in vivo. In conclusion, MM appears to be the best medium currently available to assess the sensitivity of M. fructicola mycelium in vitro.  相似文献

Botrytis cinerea and Puccinia recondita spores adhere strongly to polystyrene microtiter plates coincident with germination. We developed assays for inhibition of spore adhesion in 96-well microtiter plates by using sulforhodamine B staining to quantify the adherent spores. In both organisms, fungicides that inhibited germination strongly inhibited spore adhesion, with 50% effective concentrations (EC50s) comparable to those for inhibition of germination. In contrast, fungicides that acted after germination in B. cinerea inhibited spore adhesion to microtiter plates only at concentrations much higher than their EC50s for inhibition of mycelial growth. Similarly, in P. recondita the ergosterol biosynthesis inhibitors myclobutanil and fenbuconazole acted after germination and did not inhibit spore adhesion. The assays provide a rapid, high-throughput alternative to traditional spore germination assays and may be applicable to other fungi.  相似文献
