Vitamin D3 (1000 iu 100 g−1 body wt) was administered to the freshwater mud eel, Amphipnous cuchia , which was maintained either in tap water or a calcium-rich environment. Elevation of serum calcium levels was observed after the Vitamin D3 administration. This response was faster in the fishes which were maintained in a calcium rich environment after the vitamin D3 administration.  相似文献

In elderly institutionalized people, confined to bedroom and receiving no vitamin D supplementation, the frequency of vitamin D deficiency is found very high. Systematic administration of vitamin D has, therefore, been proposed to correct vitamin D deficiency. Within this context, we studied 40 elderly institutionalized subjects (mean age 80.5 + 7.2 yr) with low 25(OH)D3 concentrations (4.4 + 1.8 micrograms/l). Sixteen of them (Group I) had low serum calcium concentrations (less than 2.3 mmol/l) and 24 (Group II) had normal serum calcium concentrations (from 2.3 to 2.6 mmol/l). As hypocalcemia has been shown to regulate 1,25(OH)D3 production independent of PTH in animals and in humans, we compared their respective responses to the administration of vitamin D3. Subjects received a total dose of 15 mg (600,000 IU) of vitamin D3 divided into 3 i.m. injections at one month intervals and were explored before therapy and one and 6 months after the last dose of vitamin D3. The treatment induced a similar marked rise in 25(OH)D3 levels (from 4.1 + 1.7 to 24.4 + 8.7 micrograms/l for group I and from 5.1 + 1.8 to 27.2 + 8.0 micrograms/l for group II) in both groups but increased the 1,25(OH)2D3 concentrations only in group I (from 22.9 + 6.9 to 32.6 + 11.3 ng/l). Meanwhile serum calcium concentrations rose in group I (to low normal range i.e. 2.31 + 0.07 mmol/l) and were unaffected in group II. These results suggest that hypocalcemia is a potent stimulator of renal 1-hydroxylase in elderly people. Furthermore, a transient significant (P less than 0.01) increase in serum osteocalcin (from 10.6 + 4.1 to 14.1 + 5.9 micrograms/l) could be observed in group I which demonstrates for the first time that the osteocalcin response of osteoblasts to stimulation by 1,25(OH)2D3 is retained in very old people.  相似文献

Although the cardiovascular system is not a classical target for 1,25-dihydroxyvitamin D3, both cardiac myocytes and vascular smooth muscle cells respond to this hormone. The present study aimed to elucidate the effect of active vitamin D3 on cardiovascular functions in rats exposed to glucocorticoid excess. Adult male Wistar rats were allocated into three groups: control group, dexamethasone (Dex)-treated group receiving Dex (200 μg/kg) subcutaneously for 12 days, and vitamin D3-Dex-treated group receiving 1,25-(OH)2D3 (100 ng/kg) and Dex (200 μg/kg) subcutaneously for 12 days. Rats were subjected to measurement of systolic (SBP), diastolic (DBP), and mean arterial (MAP) blood pressures and heart rate. Rate pressure product (RPP) was calculated. Rats’ isolated hearts were perfused in Langendorff preparation and studied for basal activities (heart rate, peaked developed tension, time to peak tension, half relaxation time, and myocardial flow rate) and their responses to isoproterenol infusion. Blood samples were collected for determination of plasma level of nitrite, nitric oxide surrogate. Dex-treated group showed significant increase in SBP, DBP, MAP, and RPP, as well as cardiac hypertrophy and enhancement of basal cardiac performance evidenced by increased heart rate, rapid and increased contractility, and accelerated lusitropy, together with impaired contractile and myocardial flow rate responsiveness to beta-adrenergic activation and depressed inotropic and coronary vascular reserves. Such alterations were accompanied by low plasma nitrite. These changes were markedly improved by vitamin D3 treatment. In conclusion, vitamin D3 is an efficacious modulator of the deleterious cardiovascular responses induced by glucocorticoid excess, probably via accentuation of nitric oxide.  相似文献

Background

Varanidae is a clade of tiny (<20 mm pre-caudal length [PCL]) to giant (>600 mm PCL) lizards first appearing in the Cretaceous. True monitor lizards (Varanus) are known from diagnostic remains beginning in the early Miocene (Varanus rusingensis), although extremely fragmentary remains have been suggested as indicating earlier Varanus. The paleobiogeographic history of Varanus and timing for origin of its gigantism remain uncertain.

Methodology/Principal Findings

A new Varanus from the Mytilini Formation (Turolian, Miocene) of Samos, Greece is described. The holotype consists of a partial skull roof, right side of a braincase, partial posterior mandible, fragment of clavicle, and parts of six vertebrae. A cladistic analysis including 83 taxa coded for 5733 molecular and 489 morphological characters (71 previously unincluded) demonstrates that the new fossil is a nested member of an otherwise exclusively East Asian Varanus clade. The new species is the earliest-known giant (>600 mm PCL) terrestrial lizard. Importantly, this species co-existed with a diverse continental mammalian fauna.

Conclusions/Significance

The new monitor is larger (longer) than 99% of known fossil and living lizards. Varanus includes, by far, the largest limbed squamates today. The only extant non-snake squamates that approach monitors in maximum size are the glass-snake Pseudopus and the worm-lizard Amphisbaena. Mosasauroids were larger, but exclusively marine, and occurred only during the Late Cretaceous. Large, extant, non-Varanus, lizards are limbless and/or largely isolated from mammalian competitors. By contrast, our new Varanus achieved gigantism in a continental environment populated by diverse eutherian mammal competitors.  相似文献

Synopsis Acidification of the lakes of the La Cloche Mountains, Ontario, has reduced population densities of the yellow perch. Yellow perch of age groups 1 to 3 responded with increased rates of growth. Yellow perch of age groups 4 to 9 responded with reduced rates of growth. On the basis of these findings it is suggested that the growth rate of yellow perch may serve as a valuable indicator of environmental stress. Plasma 25-(OH)D3 concentrations following an intra-portal injection of 100 micrograms Kg-1 of D3 or 100 micrograms Kg-1 of 25-(OH)D3 was studied in D depleted rats fed ethanol diet and pair-fed controls. When challenged with D3, the rats under ethanol feeding were unable to increase their plasma 25(OH)D3 concentrations above those observed in controls. Plasma 25(OH)D3 concentrations following 25(OH)D3 administration were however lowered by the ethanol treatment 3 and 96 hr after 25(OH)D3 administration (p less than 0.05). These results suggest that animals chronically exposed to ethanol have an unaltered plasma 25(OH)D3 response following a pharmacological dose of D3 while the drug treatment contributes to an accelerated plasma 25(OH)D3 disappearance following 25(OH)D3.The former observations also suggest that D3 does not seem to be a high affinity substrate for the ethanol-induced cytochrome P-450.  相似文献

Extraction, lipid-reduction, and chromatographic methods suitable for the resolution and subsequent quantitation of vitamin D2, vitamin D3, 25-hydroxyvitamin D2, and 25-hydroxy-vitamin D3 from human milk are described. This procedure utilizes a methanol:methylene chloride extraction, precipitation of unwanted lipids with cold methanol and ether, backwash with alkaline buffer, silica Sep-Pak preparative chromatography, normal- and reverse-phase high-performance liquid chromatography with final quantitation of the antirachitic sterols by competitive protein binding assay. The described assay was used to determine these antirachitic sterols in milk from women receiving various supplements of vitamin D or undergoing ultraviolet phototherapy.  相似文献

Synopsis From 1979 to 1981 we followed the movement, diet, and growth of yellow perch,Perca flavescens, for their first 70 days after hatching in Lake Itasca, Minnesota. Perch spawned inshore during early spring; hatching occurred 10–20 days after spawning. Newly hatched perch were 5.6–6.2 mm total length (TL). Soon after hatching the larvae moved into the limnetic zone where they began feeding. This movement is probably a mechanism to escape intense predation in the littoral zone. Normally the first food of perch was immature copepods, but within a week they incorporated all common zooplankters into their diet. When the perch reached 25 mm TL (about day 40) they returned to the littoral zone, where they ate larger and more abundant prey than was present in the limnetic habitat. There is no correlation between growth rates and zooplankton abundances, which suggests that food quantity is not a limiting factor in the early life history of perch in Lake Itasca.  相似文献

  • 1.1. The contents of vitamin D3, 25-hydroxyvitamin D3 (25-OH-D3) and 7-dehydrocholesterol (7-DHC) in 22 kinds of fish liver samples were determined by a high-performance liquid chromatographic (HPLC) method.
  • 2.2. Vitamin D3 was detected in all fish liver samples, but its contents varied from 84 to 264,000 ng/g wet tissue. The liver of fish belonging to Carangidae and Scombridae contained large amounts of the vitamin and therefore we deduced that vitamin D3 levels in liver might have some relations with taxonomical positions of fishes.
  • 3.3. 25-OH-D3 was detected in 7 out of 22 kinds of fish liver samples, while 7-DHC was in 14 out of 22. The contents of the two sterols were generally much lower than those of vitamin D3 and there was no special relationship between the contents of the sterols and the vitamin.

Utilizing the microphthalamic mouse, (mi/mi) as a model of osteopetrosis, vitamin D3 (cholecalciferol) was administered prenatally and postnatally to study its effects on tooth development and subsequent eruption. It has previously been reported that vitamin D3 crosses the placental barrier and is absorbed into mammary gland milk. Fifteen heterozygotes (+/mi) were used as breeders. There were three study groups: A) 5.0 ng/gm cholecalciferol sulfate; B) 2.5 ng/gm cholecalciferol sulfate; and C) no therapy. Intraperitoneal injections were administered three times per week, beginning when pregnancy was evident, and continuing for 4 additional weeks during lactation. Approximately half of the 59 offspring were sacrificed at age 1 day and the other half at 4 weeks. The former group was studied for crown development, and the latter group was studied for root development and eruption. When the osteopetrotic offspring of group A were compared with osteopetrotic offspring of group C, crown development and tooth eruption were substantially more advanced. Parameters examined were maturity of the ameloblasts and odontoblasts, dentin and enamel formation, root sheath development, status of eruption, and degree of apex closure. It was concluded that cholecalciferol sulfate significantly improves tooth development and subsequent eruption in the osteopetrotic mouse. A genetic disease has had its phenotype modified by vitamin therapy during gestation.  相似文献

This paper describes a simple chromatographic technique on Sephadex LH20 for the separation of vitamin D3 sulfate from free vitamin D3 and its metabolites. This technique has been used in the study of vitamin D3 sulfate metabolism in rats. A positive relationship was observed between Secchi disc depth and dermal melanin concentration in yellow perch Perca flavescens sampled from 11 humic lakes located on the Canadian Shield in southern Quebec (Canada). Secchi disc depth explained 23% of the variations of dermal melanin concentration. Secchi disc depth and thus water transparency appear to have a positive influence on melanin production in the dermis of P. flavescens.  相似文献

Synopsis Diet, growth, energy content and mercury concentrations of yearling yellow perch,Perca favescens, were determined from June 1974 through May 1975. Benthic invertebrates predominated in the diet throughout the summer with zooplankton important in June and fish in August and September. Ash, lipid, protein and energy content of the yearling yellow perch increased through the summer reaching maximal values in August, 1974. Fish collected May 15, 1975 has reduced ash, lipid., protein and energy content with an increased moisture content. Both protein and lipid were catabolized during overwintering. Seasonal variation in organic mercury concentration was not significant (p>0.05). Inorganic mercury decreased significantly (p0.05) in September, 1974 and May, 1975 samples, reflecting changes in diet and the shorter biological half-life of inorganic mercury. The pollutant bioaccumulation model of Norstrom et al. (1976) adequately described the observed accumulation of organic mercury in young perch from June through September 1974, but either underestimated uptake or overestimated clearance for the period October 1974–May 1975.  相似文献

Vitamin D metabolites and analogs induce de novo synthesis of a specific calcium-binding protein in embryonic chick duodenum maintained in organ culture. Using calcium-binding protein biosynthesis as a specific and sensitive biochemical indicator of intrinsic biopotency, 24,24-difluoro-1α,25-dihydroxy vitamin D3 was found to be approximately four times more potent on a molar basis than the most active, naturally occurring metabolite, 1α,25-dihydroxy vitamin D3.  相似文献
