Summary Potted cuttings of a 12-year-old Picea abies tree were fumigated with ozone, 100 or 300 g O3· m–3 (50 or 150 ppb O3) being added to charcoal-filtered air during the 1985 growing season for a total of 1215 h. The wax structure of ozone-fumigated needles was no different from that of controls. Because flattened wax structures and fused wax fibrils also occurred in controls, these phenomena could not serve as bioindications for the ozone concentrations applied. A smooth layer was found beneath the soluble wax layer and covered needle surface and stomatal openings of ozone-fumigated needles to a greater extent than in controls. Wax quantity was considerably reduced by fumigation with 300 g O3 · m–3. Leaf pigments (as extracted with the wax) were less abundant in needles treated with 300 gO3; the smooth layer probably contributed to the impeded extraction of pigments.  相似文献

Summary Potted cuttings of a 12-year-old, and grafts of an 80-year-old, Norway spruce (Picea abies (L.) Karst.) were subjected to 100 or 300 g O3·m–3 for 1215 h (45 h of daylight per week) during the growing season of 1985. At the end of the fumigation the plants did not exhibit any visible signs of injury. Whereas in the fumigation with 100g O3·m–3 we did not detect any significant change in gas exchange, 300 g O3·m–3 did alter the CO2 uptake after 27 weeks, and in one clone transpiration was also altered. Stomatal reaction to a change of light suggested sluggishness, but the change was not statistically significant.  相似文献

Summary Fumigation with 100 g/m3 and 200 g/m3 ozone in closed-top fumigation chambers induced starch accumulation in chloroplasts of Norway spruce. This accumulation was probably due to a partial inhibition of the starch translocation at night. The intensity of the effect was dependent on the season and the age of the needles. The accumulation was reversed in winter. It is therefore unlikely that such an effect has much significance for plant health.  相似文献

Summary Catalase (EC 1.11.1.6) activity (both total and specific activity) of particulate fractions of needles of Norway spruce [Picea abies (L.) Karst.] was elevated approximately 2-fold following exposure of trees to 60–70 g/m3 of ozone during the growing season compared to trees receiving charcoal filtered air (about 15 g/m3 ozone). Measurements were from homogenates fractionated into particulate and soluble (supernatent) activities. In contrast, the catalase activity of the supernatant was unchanged in response to ozone treatment. Catalase activity declined as the needles aged comparing current, 1-, and 2-year needles but the ozone-induced increment remained constant. Electron microscope cytochemistry using peroxidatic coupling with 3,3-diaminobenzidine carried out in parallel, revealed catalase-containing peroxisomes both in situ and in the particulate fractions analyzed for catalase activity. The tissue volume occupied by peroxisomes in response to needle age and ozone appeared to vary approximately in proportion to the measured catalase activity. Overall cytochemical reactivity for catalase declined with needle age, but, for all years, was greater in needles of trees receiving air supplemented with ozone compared to those of trees receiving charcoal filtered air.Abbreviations DAB 3,3-diaminobenzidine tetrahydrochloride  相似文献

Summary Potted young Norway spruce trees were exposed to different concentrations of the air pollutants ozone, sulphur dioxide and nitrogen dioxide under completely controlled environmental conditions. After the treatment, the potted trees were kept outdoors. Measurements of the maximum photosynthetic capacity (A2500) were performed with current-year and 1-year-old needles during and after exposure of the trees. In trees fumigated with nitrogen oxides no damage was found at the concentrations used, and the trees' ability to fix carbon dioxide was increased. Using SO2, a rapid and marked decrease in A2500 was obtained within the first days of the experiment. This decrease did not continue further, but was reversed upon cessation of the fumigation. However, a clear dose-dependent decrease in A2500 occurred when trees were fumigated continuously with an ozone concentration of 450 nl l–1 or more. The effect of ozone was not reversible, but continued during post-culture of the trees.  相似文献

The chromosomes (2n = 2x = 24) of Norway spruce are very large since their size reflects the huge amount of genomic DNA (2C = 30 × 109 bp). However, the identification of homologous pairs is hampered by their high degree of similarity at the morphological level. Data so far presented in the literature were not sufficient to solve all the ambiguities in chromosome identification. Several genomic Norway spruce DNA clones containing highly repetitive sequences have been identified and characterised in our laboratory. Three of them were selected for fluorescent in situ hybridization (FISH) experiments because of their strong signals and suitability for chromosome identification: PATR140 hybridized at the centromeric site of three chromosome pairs; PAF1 hybridized in six subtelomeric and two centromeric sites; 1PABCD6 co-localized with the subtelomeric sites identified by PAF1. The statistical analysis of microscopic measurements of chromosomes in combination with the FISH signals of these probes allowed the unambigous construction of Norway spruce karyotype. We also compared the karyotype of Norway spruce with that of other spruce species to infer the number and kind of rearrangements that have occurred during the evolution of these species.Communicated by D.B. The vitality of fine roots in a Norway spruce stand subjected to application of ammonium sulphate (NS), wood ash (A) and nitrogen-free fertilizer (V) respectively, was investigated using i) vitality classification of fine roots based on morphological characteristics and ii) the triphenyl tetrazolium chloride (TTC) method of estimating dehydrogenase activity.Although the NS-treated areas showed a 30% increase in above-ground production in response to the NS-application, the vitality of the fine-root system in the humus layer appeared to be in a state of deterioration, as indicated by a decrease in fine-root biomass, an increased amount of dead fine (0–1 mm) and small (1–2 mm) roots, a decreased specific root length (SRL = fine root length/fine root dry weight) and an increased dehydrogenase activity. The impact of the the A and V treatments was reflected in a decrease in fine-root biomass and an increase in SRL. The results make it clear that in order to study the vitality of forest trees, both fine-root studies and studies of above-ground tree parts are necessary.  相似文献

Summary The concentrations of 23 elements have been determined by instrumental neutron activation analysis in the needles and axes of 1-year-old spruce twigs from 12 different sites. Although the twigs had been washed with toluene and tetrahydrofuran prior to analysis, it was found that the amounts of eight elements (Al, Cr, Eu, Fe, La, Sc, Th and V) present were predominantly due to aerosol particles still being retained on the surface of the axes. The results of the remaining 15 elements (Ba, Br, Ca, Cl, Co, Cs, Hg, K, Mg, Mn, Na, P, Rb, Sr and Zn) were not or only slightly influenced by this effect and are considered to represent their inherent concentrations within the axes. With most of these latter 15 elements there are highly significant and linear correlations between the concentrations in the needles and in the axis, which testify to the great similarity of needles and axis of a twig. The axis/needles ratios are constant for every element (except K), but the individual elements show considerable differentiation, as evidenced by their ratios, which have values between 0.5 and 2.8. Inter-element correlations in the axes are mainly found within two groups of elements. With Na, Cl and Br these correlations are probably due to different pollution levels of the different sites, whereas with Mn, Co, Rb and Cs they are due to the pH of the soil. The activity of 137Cs from the Chernobyl fallout represents independent confirmation of the results obtained with the stable elements. With twigs sprouted before the deposition of the fallout, 137Cs acts as a tracer for aerosol particles, whereas it forms a tracer for the stable Cs inherent to the plant with twigs sprouted after the deposition.  相似文献

We have studied how callus cultures from two clones of Norway spruce influence the growth of two pathogens, Ceratocystis polonica and Heterobasidion annosum, when co-cultivated in vitro. In field experiments, trees of clone 409 were susceptible to both fungi, whereas clone 589 was less affected. Callus was cultured on medium containing cytokinins (benzylaminopurine, kinetin) and with or without auxin (2,4-dichlorophenoxyacetic acid). For co-cultivation with fungus, one piece of callus was placed towards the edge of each Petri dish. One and 14 days after inoculation with callus the dishes were co-inoculated with the fungus. Both clones strongly stimulated the initial growth of both fungi. Clone 589 inhibited the growth of both fungi when the fungi were inoculated one day after the callus. When the callus was cultured on medium without auxin for 14 days before co-inoculation clone 589 strongly inhibited the growth of both fungi, whereas clone 409 inhibited H. annosum only. Studies of crystallinity of Scots pine and Norway spruce cellulose   总被引:3,自引:0,他引:3  
The variation in the mass fraction of crystalline cellulose (crystallinity of wood), the intrinsic crystallinity of cellulose, and the thickness of cellulose crystallites in early wood of Norway spruce [Picea abies (L.) Karst.], and Scots pine (Pinus sylvestris L.) grown in Finland were studied using wide angle X-ray scattering and nuclear magnetic resonance spectroscopy. The mass fraction of crystalline cellulose in wood increased slightly with the distance from the pith and was about 30±4% in mature wood of both species. The crystallinity of cellulose and the thickness of cellulose crystallites were almost constant for both species. The crystallinity of cellulose was 52±3% for both species and the average thickness of the cellulose crystallites was 32±1 Å and 31±1 Å for Norway spruce and Scots pine, respectively. The mass fraction of cellulose in wood, calculated from the crystallinity values, increased with the distance from the pith for both species.  相似文献

Summary Development of spruce shoot aphid (Cinara pilicornis Hartig) populations was monitored in natural and artificial infestations of Norway spruce (Picea abies Karst.) seedlings, exposed to air pollutants in an experimental field. The pollutants, applied both singly and in mixtures, were gaseous sulphur dioxide, NaF (30 mg l-1 F) and Ca(NO3)2 or (NH4)2SO4 in aqueous solutions (200 mg l-1 N). Aphid numbers on 10 seedlings in each treatment and two control plots were counted at 2-week intervals. At the beginning of the experiment aphid numbers did not differ between treatments. Aphid populations peaked in late June and early July. All the pollutants and their combinations significantly increased the numbers of aphids per seedling. Four apterous females were transferred to spruce seedlings which were growing in containers in the same plots. After 4–5 weeks aphid numbers were significantly higher in the fluoride treatment and in the combined treatment of fluoride, nitrogen and SO2. The pollution treatments did not have a significant effect on shoot growth. Concentrations of F and S in needles were higher in treatments involving these pollutants. There were no significant differences in the concentrations of free amino acids in shoot stems between control and fluoride treatment. However, the relatively low concentration of arginine in the F treatment at the end of the growing season might indicate disturbances in the nitrogen metabolism of spruce seedlings.  相似文献

The differentiation of a surface layer on the embryonal mass is one ofthe first markers for normal embryo development in Norway spruce. We havepreviously shown that this differentiation is closely interlinked with a switchin the expression pattern of Pa18, a putative lipidtransfer protein (LTP) gene. In transgenic embryos ofNorway spruce under- or overexpressing the Pa18 gene under the maize ubiquitin promoter, there is no switch in the expression pattern ofthe Pa18 gene and the embryos are blocked in theirdevelopment early during maturation. In this work, we describe how under- andoverexpression of Pa18 affect sequential developmentalstages during somatic embryogenesis. The differentiation of somatic embryosfromproembryogenic masses is not affected, but the morphology of early somaticembryos is changed. Both under and overexpressing somatic embryos can gothrougha maturation process, although at a much lower frequency than the controlembryos. Germination is not affected by altered Pa18expression. However, plants regenerated from under and highly overexpressingsomatic embryos cannot survive prolonged culture.  相似文献

Mycorrhizal growth rates were measured monthly, using a new method, in two neighouring plots of a natural spruce stand. One of the plots was irrigated while the other suffered from drought during the late summer and autumn months. Drought did not completely stop mycorrhizal growth. It caused a higher rate of root dormancy and a reduced elongation rate of the parent roots but an increased development of new mucorrhizal last order laterals. Thus, the branching density of the very fine root system was increased, even though fewer growing mycorrhizae were found in the non-irrigated plot during the dry period. Similar results were observed in a water-stress experiment with pot-cultures. After rewetting, elongation rate was stimulated and the number of growing mycorrhizae increased rapidly on the non-irrigated plot. Possible relationships between dry weight, distribution and branching density of growing fine root systems are presented.  相似文献

Effects of water stress on needle ultrastructure of 2-year-old Scots pine (Pinus sylvestris L.) and 5-year-old Norway spruce [Picea abies (L.) Karst.] seedlings were studied in greenhouse experiments. Drought stress was induced by leaving seedlings without watering, and waterlogging stress was produced by submerging the seedling containers in water. Needle samples for ultrastructural analyses were collected several times during the experiments, and samples for nutrient analyses at the end of the experiments. In drought stress, plasmolysis of mesophyll and transfusion parenchyma tissues, aggregation of chloroplast stroma and its separation from thylakoids and decreased size and abundance of starch grains in needles of both species were observed. The concentration of lipid bodies around the chloroplasts were detected in pine needles. Calcium and water concentrations in spruce needles were lower by the end of the experiments compared to controls. In waterlogging treatment, swelling of phloem cells in pine needles and large starch grains, slight swelling of thylakoids and increased translucency of plastoglobuli in chloroplasts of both species studied were observed. The phosphorus concentration in pine needles was higher while phosphorus, calcium and magnesium concentrations in spruce needles were lower in the waterlogging treatments compared to controls. Typical symptoms induced by drought stress, e. g. aggregation of chloroplast stroma and its separation from thylakoids, were detected, but, in waterlogging stress, ultrastructural symptoms appeared to be related to the developing nutrient imbalance of needles.  相似文献

Ring-width chronologies of Picea abies (L.) Karst. from ten sites in the Tatra Mountains, Poland, were developed to explore growth/climate responses in stands along an altitudinal gradient ranging from 839 to 1468 m a.s.l. There were positive relationships between current-year radial growth and mean monthly temperatures in March, April, June and July, but with increasing elevation, the strength of this correlation declined for March–April and increased for June–July temperatures. The mean monthly temperature in October of the previous year positively influenced radial growth of trees at all sites. Lower mean temperatures in January negatively affected growth of trees at the high-elevation sites. Trees at the low-elevation sites responded positively to a warm early spring, whereas trees at the high-elevation sites showed positive growth responses to higher summer temperatures. Growth of trees at the high-elevation sites was better explained by the temperature regime than was growth at the lower-elevation sites. Therefore, it is likely that these sites may be particularly sensitive to potential effects of temperature change.  相似文献

Summary Photosynthetic performance of Norway spruce needles [Picea abies (L.) Karst.] was measured over a 1-year period. The trees grew in an area of heavy air pollution and forest decline on a mountain ridge in the eastern Ore Mountains (Czech Republic). Photosynthetic capacity, as well as light use efficiency, decreased dramatically with time, starting in July (2 months after bud-break) to finally reach zero (respiration only) by February of the following year. Two months later all needles from upper crown parts were shed. Needles from lower crown parts, on the other hand, were undamaged. The chlorophyll and Mg content decreased transiently during the cold season, with Mg reaching deficiency thresholds during winter. However, total sulfur, as well as organic and sulfate S increased with time. The increase was higher in needles from the upper parts of the crown, which were exposed to windy air throughout the year, than in the lower parts of the crown, which were covered by grass during summer and by snow during most of the winter.  相似文献

A greenhouse experiment was performed to evaluate the effect of Norway spruce (Picea abies (L.) Karst.) seedlings on net nutrient availability in five different growing media containing F- or H-layer and mineral soil originating from a haplic podzol in northern Sweden. The initial total amounts of eight nutrient elements (N, K, P, Ca, Mg, Mn, Fe, Zn) and exchangeable amounts of the same elements were analyzed in pots with or without spruce seedlings. In the planted pots seedling nutrient uptake was also estimated. After 26 weeks, higher net nutrient availability with seedlings was found in 25 out of the 40 (62%) growing media and nutrient element combinations. A positive seedling effect on net nutrient availability might be explained by rhizodeposition stimulating the soil microorganism activity and accelerating the weathering of minerals or by seedling roots promoting the nutrient providing processes through changes in soil chemical and physical properties. Nitrogen availability was primarily affected by what part of the forest floor the growing medium contained although the positive response to seedling presence was apparent. The positive net availability response of P, Ca, Mg, Mn, Fe and Zn to seedling presence was on the other hand relatively strong. In the case of P, K, and Zn the growing medium composition (if the F- and H-layer was pure or mixed with mineral soil) was also an important factor for the estimated net availability. Pure F-and H-layer provided greater P- and K-availability while the availability of Zn increased when mineral soil was added. The influence of growing plants ought to be considered when soil samples are used for assessing the nutrient availability.  相似文献

Summary The mycorrhizal activity of spruce in a mixed-wood forest was monitored over 1 year by measuring biochemical characters in fine roots of six canopy trees and of a regrowth stand. The concentration of adenosine 5-triphosphate (ATP), a measure of living biomass, showed two peaks per year, one at bud break and one after main shoot growth. The concentration of storage polysaccharides in mycorrhizae showed the same cycles even more pronouncedly. It is proposed that these changes reflect growth and senescence of mycorrhizae and that the timing of the cycles is controlled by translocation of assimilates from the shoot. Differences between mycorrhizae collected from canopy trees and the regrowth stand were small and not significant. Characters known to be related to fungal activity of the mycorrhizal symbiosis (concentration of trehalose, glucose uptake, respiration) also varied little among the six canopy trees. Large differences among fine-root samples from different canopy trees, however, were detected in the concentrations of ATP and storage polysaccharides, measures which seemed to be physiologically integrated within trees. If low concentrations in roots precede losses of foliage from trees, these two symptoms could be used as early indicators of growth decline in individual spruce trees.  相似文献

Summary The concentration of 20 elements (including eight nutritional elements) in spruce needles was monitored by taking samples at nine different dates in a single year. On each date the youngest and the 1-year-old needles were collected at four different sites. The needles were washed with an organic solvent to remove surface contamination before analysis by instrumental neutron activation. Although the sites showed widely different concentrations for the same element, they could be treated as a uniform population using normalized concentration values. Taking the time dependence of the concentration and the ratio of the concentrations in new and old needles as criteria, the following three groups of elements could be distinguished: 9 group I —Ca, Sr, Ba, and Mn; group II — Al, Br, Co, Fe, Hg, La, Sc, Sb, and Zn; group III — K, Rb, Cs, P, and Cl. Mg and Na did not fit into any of these groups. Within group I and III elements the strongest variations occur during the growing season (late spring and early summer) and the weakest during the dormant season; the variation is greater in the younger needles. Group II elements show an approximately linear increase throughout the year in both the younger and older needles. The concentrations of elements of groups I and II increase with time, while those of group III decrease. Na is unique insofar as its concentration is influenced by foliar uptake of road de-icing salt. The concentration of elements belonging to the same group shows strong inter-element correlations. The grouping of elements deduced here shows similarities with the chemical nature of the elements and with their transportability within the plant.  相似文献

The concentrations of Ca, Sr, Ba and Mn were determined in up to five successive needle age classes from 54 individual Norway spruce trees [Picea abies (L.) Karst] from nine different sites. The accumulation behaviour was found to be very nonuniform, going from an increase with needle age to a decrease; irregular patterns were also found. The type of accumulation is largely site specific. The increasing behaviour can in most cases be approximated by a simple arithmetic function. All four elements usually show the same accumulation pattern, the similarities being closest between Ca and Mn and least between Ca and Ba. It is postulated that the similarity between the four elements is due to their precipitation and storage as oxalates. The similarity between Ca, Sr and Ba is observed at all concentrations, that with Mn only at concentrations larger than 300 g/g. Mn at small concentrations (< 50 g/g) shows a decreasing pattern and no similarity at all with Ca, Sr and Ba, but behaves similar to mobile elements.  相似文献
