本文利用高效液相色谱(HPLC)对银盾革蜱饥饿、吸血和饱血后若虫整体水平20-羟基蜕皮酮含量变化规律进行了研究.结果表示20-羟基蜕皮酮存在于若虫整个发育期,其含量变化具有两个峰.第一个峰小,出现在吸血期(吸血后5天),含量为2.34ng/只,该峰出现后,若虫快速饱血并从宿主体落下;第二个峰大,出现在蜕化期(饱血后8天),含量为16.17ng/只,此峰可能与上表皮沉积有关.从饱血后第6天至饱血后第8天,20-羟基蜕皮酮含量迅速增加,皮层溶离发生在饱血后7天左右.  相似文献

Abstract The ecdysteroids (ECs) concentration and their components in the larvae of Haemaphysalis longicornis were determined by radioimmunoassay (RIA) and high performance liquid chromatography (HPLC). ECs ware found in feeding and engorged larvae and absent in unfed larvae. During feeding and the first 3 days after engorgement, ECs were in low level (less than 14. 35pg EE /larva). On the 5th day, they began to increase sharply (29. 87pg EE /larva) and reached their peak (56. 04pg EE /larva) on the 7th day. Then they declined to low value again (19. 38pg EE /larva). HPLC analysis revealed that the ECs components of the RIA peak could be 20-hydroxyecdysone (20E) and ecdysone (E) on the ratio of 20E: E = 3. 07: 1, which meant 20E was probably the main effective hormone.  相似文献

Abstract  The ecdysteroid levels in hemolymph, ovary, synganglion and whole body of diapausing female Dermacentor niveus were detected by HPLC, and compared with the results of nondiapausing female. It is revealed that the ecdysteroid levels in hemolymph and ovary of diapausing female are similar basically to that of nondiapausing female in the first few days after engorgement. From the 10th day after engorgement, the ecdysteroid levels of diapausing female decreased and even became distinctly lower than that of nondiapausing female. The paucity of ecdysteroids in these individuals would influence the normal development of oocytes. In order to explore the effect of ecdyateroids on the diapausing female, we injected 20-hydroxyecdysone with different dosages at different time into the ticks, and found that after just complete engorgement the injection with large dosages (10000 and 1375 ng/tick) caused death of the ticks. From 10th to 20th day after engorgement the ecdysteroid levels of diapausing female are lower than that of nondiapausing one before oviposition, the injection with certain dosages 50, 70 and 100 ng/tick> of 20E can accelerate vitellogenesis and terminate reproductive diapause, but the amount of eggs produced by them is less than that produced by nondiapausing female. The termination of diapause in female of ixcdid tick by exogenous ecdysteroids is reported for the first time.  相似文献

用高效液相色谱法和放射免疫测定法测定了银盾革蜱滞育雌虫的合神经节、血淋巴、卵巢和整体中蜕皮激素的含量,并与正常发育的雌虫做比较。结果表明,滞育雌虫血淋巴和卵巢中蜕皮激素的含量在饱血后前10天与正常发育雌虫基本上相同。饱血10天以后,滞育雌虫蜕皮激素的含量下降,较正常发育雌虫少得多。蜕皮激素的缺乏影响了卵母细胞的发育。为阐明蜕皮激素对滞育雌虫的影响,在不同时间向银盾革蜱滞育雌虫体内注入不同剂量的β-蜕皮素。刚饱血者注入大剂量(1.375和10000ng/蜱)的β-蜕皮素引起雌虫死亡。饱血后20—30天,注入一定剂量(50,70,100ng/蜱)的β-蜕皮素可以促进卵黄形成,并解除了雌虫的生殖滞育,但其所产的卵量明显少于正常发育的雌虫。应用外源蜕皮激素解除硬蜱雌虫的生殖滞育现象尚属首次报道。  相似文献

Summary The structure of the extensible (alloscutum) and inextensible (scutum) integument of the nymph, Amblyomma variegatum was examined during the whole bloodmeal and the nymphal-adult moulting cycle. Integumental events were tentatively correlated with the ecdysteroid levels measured by radioimmunoassay. We observed that all the integumental events were realised along an anteroposterior gradient. During the 5 days corresponding to the bloodmeal, although the hormone concentration was low, a new endocuticle was deposited on both the alloscutum and scutum. Furthermore, mitoses were initiated in the capitulum. On days 1–2 after the meal, ecdysteroid titres began to increase and reached a first peak corresponding to 4.1 ng 20-hydroxyecdysone equivalents/tick on the 4th day after the ticks dropped off their host. At this time the epidermis of the capitulum was detached and the outline of the adult capitulum was already visible. Mitotic activity in the alloscutum was initiated. On day 6 post-drop, the frontal apolysis was achieved and the ecdysteroid titres declined to basal values. A second peak much higher than the first one (maximum value of 33.7 ng/tick) and identified principally as 20-hydroxyecdysone by HPLC/RIA was noted on the 13th day post-drop. During the period of increase in the ecdysteroid levels (days 9–10 post-drop), the mitotic phase ended in the alloscutum and the apolysis began. Epicuticle was deposited after day 12 postdrop. Then, while the titre fell to low values (about 1.6 ng/tick, days 16–20 post-drop), the exocuticle was deposited and the nymphal cuticle was digested. All adult structures were functional 3 days before ecdysis. In young male as in female adults the mean value of the ecdysteroid levels corresponded to about 2.5 ng/tick. Finally, hydrolysis of tick whole extracts with esterase demonstrated a low increase of RIA-positive material, demonstrating the probable presence of natural ecdysteroid fatty-acid conjugates in this species.  相似文献

Abstract The salivary gland (SG) of female Dermacentor silvarum consists of salivary ducts and lots of acini.
From the basis capituli to the end of the gland, the ducts can be divided into three parts, namely, central duct, major branch ducts and the lobular ducts, and the acini is spherical-like structures attached to various ducts. The central duct parallels with trachea. The surface of the acini is wrinkly and many small tracheae distributed on the acini. In unfed female, the length of SG is short (547.33 μm) and extends on the first day after feeding, and reaches its maximum value (1109.40 μm) on the 3rd day after feeding, while copulation has justly completed. From the 3rd day after feeding to the first three days after engorgement, no apparent change in length of the gland is observed. On the 4th day after engorgement, it shortens sharply (500.00 μm). The diameter of the acini of unfed female is also short (45.24 μm), it increases gradually during feeding and reaches its maximum (74.10 μm) on the 5th day after feeding. After engorgement, the acini becomes atrophic slowly and degenerated sharply on the 4th day.  相似文献

Summary The intact male nymph cricket, Gryllus bimaculatus DeGeer, was found to show mating-like behavior, that is, courtship-like behavior (CSLB) and copulation-like behavior (CPLB), in the 7th and 8th (last) instars. The 8th instar nymph exhibited less CSLB and CPLB than the adult but much more than the 7th instar nymph. The movement patterns of CSLB and CPLB were essentially the same as those of adults except for motor acts requiring the use of the genitalia. CSLB was short and often ceased spontaneously before it switched to CPLB. CPLB also ended earlier than in adults. The occurrence of CSLB and CPLB was almost zero the few days around ecdysis. The nymph was very sensitive to disturbance, so that he often stopped courtship for more than 30 min after stimulation. CSLB was similarly induced in the male nymph (8th instar) by pairing with a female adult, male adult, female nymph (8th) and male nymph (8th). The female nymph (8th) was observed to mount not only the male adult but also the male nymph (8th). A fixed time sexual refractoriness forming a basis of cyclical mating activity was not present after CPLB in the nymph. It appeared in association with the emergence of spermatophore protrusion behavior around day 3 after the imaginal molt. In fledglings, there were some transitions during the sexual maturation process, such as failures in hook hanging, spermatophore extrusion, and spermatophore transfer to the female. The decerebration experiments on nymphs and fresh adults agreed with behavioral observations. We examined the hemolymph ecdysteroid titer (by radioimmunoassay) and profile of the ecdysteroid receptor (EcR/USP; by [3H]ponasterone A binding, gel mobility shift assay, Western blot) in the salivary gland of the ixodid tick, Amblyomma hebraeum Koch (Acari: Ixodidae) throughout the tick feeding period and first 6 days post-engorgement. Throughout the slow phase of feeding, the hemolymph ecdysteroid titer was approximately 18 pg/microliter. The titer peaked at approximately 52 pg/microliter during the rapid phase of feeding, falling back to approximately 22 pg/microliter on the day of engorgement. Ecdysteroid titer rose again to approximately 750 pg/microliter by day 6 post-engorgement. EcR was undetectable by any of the three assays in unfed ticks. Following the onset of feeding, there appeared both specific ponasterone A binding and two major EcR bands detected by Western blot analysis. Both measurements were sustained throughout the feeding period, but declined after detachment when the salivary glands were degenerating. After ticks reached about 100 mg (by which time most females are mated), a discrete DNA-binding band was shown by gel mobility shift assay using Drosophila hsp27 EcRE as a probe. Moreover, the band intensified when hemolymph ecdysteroid titer reached its peak during the rapid phase of feeding; it declined along with decreasing EcR/USP levels, and with specific ligand binding activity following engorgement. This study suggests a role for the small hemolymph ecdysteroid peak during the rapid phase of feeding in initiating salivary gland degeneration.  相似文献

Abstract  In order to explore the role of ecdysteroids in development and reproduction of ixodid ticks, we studied the quantitative variation of ecdysteroids in the hemolymph, synganglion, ovary and whole body of the female ixodid ticks, Dermacentor niveus and Haemaphysalis longicornis , before and after engorgement and oviposition by HPLC and RIA. The ecdysteroid content in eggs of these ticks was determined by HPLC. The results indicated that before engorgement the quantitative variation of ecdysteroids in the whole female body was not significant, but their levels increased rapidly after engorgement. The ecdysteroid titer in hemolymph was peaked on the 5th day after engorgement, which was one day prior to oviposition. It may be regarded as a singnal of oviposition. In the synganglion the peak of ecdysteroid level occurred also on the 5th day after engorgement. This is coincident with the secretory activity of neurosecretory cells of synganglion. From the 3rd day after engorgement until oviposition the ecdysteroid level in the ovary increased rapidly. Ecdysteroids were detected in eggs of H. longicornis too. They stem from ovary and accumulated with the process of embryonic development.  相似文献

Large peaks of ecdysone (E, 2,875 ng/g live wt) and 20-hydroxyecdysone (20-HE, 2,150 ng/g live wt) occur on days 8 and 12, respectively, of postdiapause pupal-adult metamorphosis at 20°C in the bertha armyworm, Mamestra configurata, and then decline to low levels (< 100 ng/g live wt) prior to eclosion of the moth (50% eclosion at day 31.8). These peaks of E and 20-HE can be suppressed by treating the developing pupae with a physiological dose (2,500 ng/g live wt) of 20-HE. Suppression of E and 20-HE by 20-HE treatment was dose dependent, rapid (within 24 h of treatment) and permanent. The peaks of E and 20-HE were suppressed by 20-HE treatment on days 1, 3, 5, and 7 but the 20-HE peak was not suppressed by treatment on days 9 or 11. It is proposed that the mechanism by which 20-HE suppresses the production of E and thereby its own production forms a negative feedback loop that operates during the first 0.4 units of pupal-adult development in M. configurata. The function of the transitory peaks of E and 20-HE that form this feedback loop is currently unknown. Since most adults from pupae that had their ecdysteroid levels experimentally suppressed by 20-HE treatment were morphologically normal, it seems that the peaks of E and 20-HE have little or no function in controlling morphological development in pupae of M. configurata.  相似文献

Summary

A mated Amblyomma hebraeum female will engorge on a host for about 8 days before detaching and beginning the maturation of its single egg batch which is laid during a period of about 30 days. The feeding period is characterized by an important synthesis of endocuticular material occurring before the rapid feeding phase. This latter phase, correlated with an enormous weight uptake, shows an increase of ecdysteroid levels measured in the whole animal by RIA. However, the hemolymphatic levels of ecdysteroids remain very low (12 pg 20-hydroxyecdysone equivalent (20-OH-E eq.) per μ1. Within 4 days after detachment, the salivary glands degenerate. Ecdysteroid levels in the whole animal continue to increase, reaching high values (about 500 ng 20-OH-E eq./tick) at the moment of oviposition which begins 10–14 days after dropping. During the same period, hemolymphatic ecdysteroid levels increase, rising to a peak (600 pg 20-OH-E eq./μ1) 1 day prior to the beginning of oviposition. Then, the levels decrease and stabilize around 250 pg 20-OH-E eq./μl during egg-laying. Freshly laid eggs contain large amounts of ecdysteroids (2744 pg 20-OH-E eq./mg).

20-Hydroxyecdysone and ecdysone have been found to be the major free ecdysteroids in hemolymph, ovaries and eggs (verified by the HPLC-RIA technique and GC-MF of silylated HPLC fractions). Helix juice (or esterase) labile ecdysteroid conjugates do not seem to be present to any noticeable extent in hemolymph, ovaries and eggs.  相似文献

为阐明蜕皮激素在硬蜱发育和生殖中的作用,用高效液相色谱法(HPLC)和放射免疫测定法测定了银盾革蜱和长角血蜱中饱血和产卵前后雌虫整体、血淋巴、合神经节及卵巢中蜕皮激素含量的变化.此外,还用HPLC测定了长角血蜱的卵和幼虫中蜕皮激素的含量.结果表明:在饱血前雌虫整体中蜕皮激素含量的变化不大,饱血后迅速增加.血淋巴中的蜕皮激素在饱血后第5天(即产卵前1天)达到高峰,可做为产卵的信号.合神经节中蜕皮激素含量的高峰在饱血后第5天,与血淋巴中高峰出现的时间一致,这与神经分泌细胞的分泌活性变化相吻合.从饱血后第3天起,卵巢中蜕皮激素含量增加很快,直到产卵时止,在长角血蜱的卵中也测到α-蜕皮素,来源于卵巢,随胚胎发育进程其含量逐渐增加.卵产出后第4天到第12天,蜕皮激素的含量增加十几倍.  相似文献

The development of Gryllus argentinus Sauss. was studied under stable laboratory conditions: the temperature of 26°C, the air humidity of 60%, and the photoperiod of 12h light: 12 h dark. The life cycle of Gryllus argentinus includes four stages: egg, pronymph, nymph, and adult. The duration of embryonic development is 18 days. The depth of egg bedding in the peat is 9.63 ± 0.12 mm (n =145), the clutch containing 2–4 eggs. A female can lay over 1100 viable eggs during the entire oviposition period. Nymphal development includes 9 instars and lasts 97 days. The duration of nymphal instars (days) is: I—5; II—6; III—6; IV—6; V—8; VI—10; VII—13; VIII—14; IX—29. The duration of the adult life is 51 days in males and 69 days, in females. In the imaginal ontogenesis of males and females, three periods can be distinguished: pre-reproductive, reproductive, and postreproductive. Males start to emit the aggressive signal on the 6th (5–8th) day (the pre-reproductive period). They enter the reproductive period (start to emit the calling song) on the 9th (8–13th) day. Females enter the reproductive period (become capable of responding to the calling song and of copulation) on the 9th (8–10th) day. Oviposition starts on the day after the first copulation. The reproductive period lasts about 40 (15–59) days in males and 58 (21–70) days in females. The post-reproductive period starts in females at the moment of finishing the egg laying period and in males, with disappearance of reproductive behavior. 【目的】 经历了长光照和短光照条件,桃小食心虫Carposina sasakii脱果老熟幼虫分别进行非滞育和滞育发育。本研究旨在通过使用蜕皮激素类似物甲氧虫酰肼对桃小食心虫滞育进行干扰,以期达到干扰桃小食心虫滞育的目的,为开发桃小食心虫防治新方法奠定基础。【方法】 利用ELISA法测定了桃小食心虫幼虫滞育和非滞育过程中的20E滴度动态;以清水为对照,测定了不同浓度(0.05, 0.1, 1,2, 5, 7.5和10 mg/mL)甲氧虫酰肼以及0.5和1 mg/mL 20E喷沙处理对桃小食心虫结茧的影响;测定了5mg/mL甲氧虫酰肼和1 mg/mL 20E喷沙处理中桃小食心虫20E滴度,并跟踪调查了0.1和5 mg/mL甲氧虫酰肼喷沙处理对桃小食心虫滞育的干扰效果。【结果】两种光周期(15L∶9D和12L∶12D)下桃小食心虫滞育和非滞育幼虫生长过程中20E滴度没有差异,均是从幼龄到老龄逐渐降低,脱果时达到最低;但脱果时,长光照非滞育发育幼虫体内20E滴度(0.473 ng/g)显著高于短光照注定滞育幼虫体内的(0.254 ng/g)。非滞育发育幼虫脱果后进入长茧化蛹,虫体20E的滴度显著升高,在蛹期保持较高滴度区间(0.652~1.217ng/g)。滞育发育幼虫脱果后进入圆茧滞育,20E滴度缓慢上升,第4天达到第一个峰值(0.656 ng/g),随后降低,在第8天达到第2个峰值(0.790 ng/g);随着进入滞育稳定期,20E滴度逐渐降低;70 d后达到滞育解除状态,20E滴度持续保持在低滴度水平;第90天达到滞育期间最低滴度(0.424 ng/g),随后20E滴度开始上升。0.05 mg/mL甲氧虫酰肼点滴短光照桃小食心虫脱果幼虫,即可干扰部分滞育圆茧的形成,对桃小食心虫脱果幼虫的LD50为7.039 μg/头。0.05 mg/mL以上浓度甲氧虫酰肼和20E喷沙处理均可有效干扰滞育圆茧的形成,1 mg/mL相同浓度的20E比甲氧虫酰肼表现出的干扰活性更高,两者均能使注定滞育的幼虫出现结长茧、畸形茧以及无法结茧的表型;在结畸形茧和无法结茧幼虫中20E滴度显著升高,5 mg/mL甲氧虫酰肼和1 mg/mL 20E喷沙处理不能结圆茧的比例分别为70.0%和66.7%,其中结畸形茧的幼虫体内20E滴度分别显著升高16.3%和143.0%,异常不能结茧的幼虫体内20E滴度分别显著升高149.3%和278.6%。部分幼虫接触甲氧虫酰肼后虽能够结圆茧,但能够完成滞育成功羽化的比例减少,甲氧虫酰肼0.1 mg/mL喷沙处理组羽化率仅为20.8%。【结论】 桃小食心虫幼虫在滞育过程中20E保持较低滴度,在生殖发育阶段需要较高滴度。外源甲氧虫酰肼和20E均能够干扰其结圆茧滞育,虫体结茧表型与20E滴度增加幅度具有相关性,相同浓度的20E比甲氧虫酰肼表现出更高的干扰活性;甲氧虫酰肼处理减少了桃小食心虫幼虫成功完成滞育的比例。  相似文献

【目的】研究中华真地鳖Eupolyphaga sinensis(Walker)断触角再生特征及再生临界期。【方法】选取1~10龄健康的初龄若虫一条触角除基部3节外剪除,蜕皮后观察并统计触角的节数。选取生长一致健康的初龄4龄若虫,于第0~19日龄进行断触角处理,蜕皮后观察触角的再生情况和触角的节数。【结果】不同虫龄的初龄若虫断去触角蜕皮后,均能通过再生使触角的节数增加。1~10龄的若虫一条触角除基部3节外剪除,蜕皮后下一龄的触角节数分别为5、8、12、13~15、15~17、16~19、18~20、13~15、20~23、22~25、24~28节。4龄的初龄若虫一条触角分别除基部0~2、3、5、10、15、20节外剪除,蜕皮后触角的节数分别为11~12、13~15、18~21、19~25、24~27节,其中一条触角除基部20节外剪除蜕皮后基本恢复正常节数。不同日龄的4龄若虫一条触角除基部3节外剪除蜕皮后,0~8日龄的处理若虫蜕皮后触角节数为13~15节,而12日龄及以后的处理若虫蜕皮后触角节数为5~6节。4龄若虫的断触角再生临界期为9~11日龄。【结论】中华真地鳖在再生临界期内断触角后能够再生触角。  相似文献

Summary

In the present study, an attempt was made to clarify the timing of the transition from proliferation stage to pupal preparation stage of Bombyx wing discs and the correlation of its timing and ecdysteroid release. An histological study was carried out during the period of ecdysteroid increase in hemolymph in the last larval instar. The number of mitoses in wing discs from the feeding stage to the spinning stage was counted. During the feeding stage, the number of mitoses was about 200/disc, with the number increasing up to about 850/disc on the day of the beginning of spinning (S0); it then decreased to about 250/disc at the S1 stage, and finally mitotic figures were absent at S2. Comparing the mitotic number with hemolymph ecdysteroid titer, ecdysteroid during the high mitotic period is close to 100ng/ml, while ecdysteroid during the low mitotic period is about 10ng/ml at day 5 and 500ng/ml at S1, respectively. Two-dimensional gel electrophoresis was carried out to identify the ecdysone-induced polypeptides or the polypeptides synthesized during the spinning stage. Five ecdysteroid-inducible polypeptides were observed during the spinning stage or after 20-hydroxyecdysone (20E) treatment. From these findings, low concentrations of ecdysteroid induce cell proliferation, whereas high concentrations inhibit cell proliferation and induce new protein synthesis.  相似文献

At 25 degrees C and under a long-day photoperiod, all 5th instar Psacothea hilaris larvae pupate at the next molt. Under a short-day photoperiod, in contrast, they undergo one or two additional larval molts and enter diapause; the 7th instar larvae enter diapause without further molt. The changes in hemolymph juvenile hormone (JH III) titers, JH esterase activity, and ecdysteroid titers in pupation-destined, pre-diapause, and diapause-destined larvae were examined. JH titers of the 5th instar pupation-destined larvae decreased continuously from 1.3 ng/ml and became virtually undetectable on day 13, when JH esterase activity peaked. Ecdysteroids exhibited a small peak on day 8, 1 day before gut purge, and a large peak on day 11, 2 days before the larvae became pre-pupae. The two ecdysteroid peaks are suggested to be associated with pupal commitment and pupation, respectively. JH titers of the 5th instar pre-diapause larvae were maintained at approximately 1.5 ng/ml for 5 days and then increased to form a peak (3.3 ng/ml) on day 11. JH esterase activity remained at a low level throughout. Ecdysteroid levels exhibited a large peak of 40 ng/ml on day 18, coincident with the larval molt to the 6th instar. JH titers of the 7th instar diapause-destined larvae peaked at 1.9 ng/ml on day 3, and a level of approximately 1.1 ng/ml was maintained even 30-60 days into the instar, when they were in diapause. Ecdysteroid titers remained approximately 0.02 ng/ml. Diapause induction in this species was suggested to be a consequence of high JH and low ecdysteroid titers.  相似文献

Summary

Vitellogenesis in ticks is known to be induced by engorgement and mating. In this paper, the synthetic pyrethroid cypermethrin (CyM) is shown to induce production of yolk protein precursor, vitellogenin (Vg), and ovarian development in unengorged mated adult female Ornithodoros moubata. The levels of Vg found in the hemolymph and ovarian development induced by CyM were dose-dependent. i.e., CyM doses of more than 0.2 and 1.0 μg/tick were needed for significant increase of Vg titer in the hemolymph and yolk deposition in oocytes, respectively. Immunological and electrophoretical analyses of Vg and Vitellin (Vn) induced by CyM were identical with those induced by engorgement. Vg titer induced by CyM in unengorged females followed approximately the same time course as that in the normal engorged females. However, Vg titer induced by CyM continued to increase after day 8 and reached a maximum (95 μg/μ1) on day 10 after treatment, while Vg titer induced by engorgement decreased again after reaching a maximum (60 μg/μ1) on day 6, correlated with yolk Vn deposition in oocytes. Ovarian development induced by even high doses (10 or 20 μg/tick) of CyM was slow compared to normal development stimulated by engorgement. 长角血蜱Haemaphysalis longicornis的交配行为包括7个时期,行为的完成依赖于性信息素的调节。生物测定表明:雄蜱的行为反应受雌蜱分泌的性信息素影响。堵塞雌蜱盾窝其行为受到抑制,点滴2,6-DCP或雌蜱盾窝腺提取物则被恢复。用气相色谱法测定了雌蜱盾窝腺中2,6-DCP的含量;吸血后1~2天含量最高(11.12 ng/只);吸血后3~5天即交配前下降交维持在一较恒定的水平;吸血后6~7天即交配后明显降低;饱血后检测不到2,6-DCP。2,6-DCP是长角血蜱性信息素的一种成分。  相似文献
