The present study investigated the extracellular biosynthesis of antimicrobial silver nanoparticles by Escherichia coli AUCAS 112 and Aspergillus niger AUCAS 237 derived from coastal mangrove sediment of southeast India. Both microbial species were able to produce silver nanoparticles, as confirmed by X-ray diffraction spectrum. The nanoparticles synthesized were mostly spherical, ranging in size from 5 to 20 nm for E. coli and from 5 to 35 nm for A. niger, as evident by transmission electron microscopy. Fourier transform spectroscopy revealed prominent peaks corresponding to amides I and II, indicating the presence of a protein for stabilizing the nanoparticles. Electrophoretic analysis revealed the presence of a prominent protein band with a molecular mass of 45 kDa for E. coli and 70 kDa for A. niger. The silver nanoparticles inhibited certain clinical pathogens, with antibacterial activity being more distinct than antifungal activity. The antimicrobial activity of E. coli was more pronounced than that of A. niger and was enhanced with the addition of polyvinyl alcohol as a stabilizing agent. This work highlighted the possibility of using microbes of coastal origin for synthesis of antimicrobial silver nanoparticles.  相似文献

There are alarming reports of growing microbial resistance to all classes of antimicrobial agents used against different infections. Also the existing classes of anticancer drugs used against different tumours warrant the urgent search for more effective alternative agents for treatment. Broad-spectrum bioactivities of silver nanoparticles indicate their potential to solve many microbial resistance problems up to a certain extent. The antibacterial, antifungal, antiviral, antiprotozoal, acaricidal, larvicidal, lousicidal and anticancer activities of silver nanoparticles have recently attracted the attention of scientists all over the world. The aim of the present review is to discuss broad-spectrum multifunctional activities of silver nanoparticles and stress their therapeutic potential as smart nanomedicine. Much emphasis has been dedicated to the antimicrobial and anticancer potential of silver nanoparticles showing their promising characteristics for treatment, prophylaxis and control of infections, as well as for diagnosis and treatment of different cancer types.  相似文献

This review gives a comprehensive overview of the widespread use and toxicity of silver compounds in many biological applications. Moreover, the bacterial silver resistance mechanisms and their spread in the environment are discussed. This study shows that it is important to understand in detail how silver and silver nanoparticles exert their toxicity and to understand how bacteria acquire silver resistance. Silver ions have shown to possess strong antimicrobial properties but cause no immediate and serious risk for human health, which led to an extensive use of silver-based products in many applications. However, the risk of silver nanoparticles is not yet clarified and their widespread use could increase silver release in the environment, which can have negative impacts on ecosystems. Moreover, it is shown that silver resistance determinants are widely spread among environmental and clinically relevant bacteria. These resistance determinants are often located on mobile genetic elements, facilitating their spread. Therefore, detailed knowledge of the silver toxicity and resistance mechanisms can improve its applications and lead to a better understanding of the impact on human health and ecosystems.  相似文献

It is of interest to document the cytotoxicity and anti microbial analysis of silver and graphene oxide nanoparticles. The plant extracts from Andrographis paniculata and Ocimum sanctum Linn were used as reducing agent. The nanoparticles were characterized using UV-visible spectroscopy, FT-IR, XRD and TEM. The antimicrobial activity was completed for oral pathogens. Brine Shrimp Lethality assay was conducted for cytotoxicity. Thus, we show that silver and graphene oxide bio based nanoparticles have antimicrobial activity with minimum cytotoxic effects.  相似文献

Membrane transport plays a leading role in a wide spectrum of cellular and subcellular pathways, including multidrug resistance (MDR), cellular signaling, and cell-cell communication. Pseudomonas aeruginosa is renowned for its intriguing membrane transport mechanisms, such as the interplay of membrane permeability and extrusion machinery, leading to selective accumulation of specific intracellular substances and MDR. Despite extensive studies, the mechanisms of membrane transport in living microbial cells remain incompletely understood. In this study, we directly measure real-time change of membrane permeability and pore sizes of P. aeruginosa at the nanometer scale using the intrinsic color index (surface plasmon resonance spectra) of silver (Ag) nanoparticles as the nanometer size index probes. The results show that Ag nanoparticles with sizes ranging up to 80 nm are accumulated in living microbial cells, demonstrating that these Ag nanoparticles transport through the inner and outer membrane of the cells. In addition, a greater number of larger intracellular Ag nanoparticles are observed in the cells as chloramphenicol concentration increases, suggesting that chloramphenicol increases membrane permeability and porosity. Furthermore, studies of mutants (nalB-1 and DeltaABM) show that the accumulation rate of intracellular Ag nanoparticles depends on the expression level of the extrusion pump (MexAB-OprM), suggesting that the extrusion pump plays an important role in controlling the accumulation of Ag nanoparticles in living cells. Moreover, the accumulation kinetics measured by Ag nanoparticles are similar to those measured using a small fluorescent molecule (EtBr), eliminating the possibility of steric and size effects of Ag nanoparticle probes. Susceptibility measurements also suggest that a low concentration of Ag nanoparticles (1.3 pM) does not create significant toxicity for the cells, further validating that single Ag nanoparticles (1.3 pM) can be used as biocompatible nanoprobes for the study of membrane transport kinetics in living microbial cells.  相似文献

Germanium is an inert metal with no known biological function in prokaryotic or eukaryotic organisms. Its toxicity is low compared to that of silver. Germanium is accumulated in certain bacterial strains by either energy-independent passive binding or an energy-dependent mechanism. Little is known about the molecular aspects of silver resistance, toxicity, and accumulation in bacterial strains. This is surprising because silver has been used as an antimicrobial agent in the medical field for centuries. It is likely that silver ions are excluded (resulting in decreased silver accumulation) from certain bacterial strains or immobilized intracellularly to prevent toxic effects from being exerted. These mechanisms of silver resistance have not been fully elucidated. This review examines the toxicity and accumulation of germanium and silver in selected microbial species. In addition, resistance mechanisms to these biologically nonessential metals is discussed, with more emphasis placed on silver-resistant bacteria due to the knowledge available.  相似文献

We show here that silver nanoparticles (AgNP) were intrinsically antibacterial, whereas gold nanoparticles (AuNP) were antimicrobial only when ampicillin was bound to their surfaces. Both AuNP and AgNP functionalized with ampicillin were effective broad-spectrum bactericides against Gram-negative and Gram-positive bacteria. Most importantly, when AuNP and AgNP were functionalized with ampicillin they became potent bactericidal agents with unique properties that subverted antibiotic resistance mechanisms of multiple-drug-resistant bacteria.  相似文献

Silver nanoparticles are known to have antimicrobial properties and have been used extensively in medicine, although the mechanism(s) of action have not yet been clearly established. In the present study, the findings suggest a novel mechanism for the antibacterial effect of silver nanoparticles on Escherichia coli, namely, the induction of a bacterial apoptosis-like response. We propose a possible mechanism for the bacterial apoptosis-like response that includes the following: accumulation of reactive oxygen species (ROS) (detected with H2DCFDA staining), increased intracellular calcium levels (detected with Fura-2 AM), phosphatidylserine exposure in the outer membrane (detected with Annexin V) which is the hallmarks of early apoptosis, disruption of the membrane potential [detected with DiBAC4(3)], activation of a bacterial caspase-like protein (detected by FITC-VAD-FMK staining) and DNA degradation (detected with TUNEL assay) which is the hallmarks of late apoptosis in bacterial cells treated with silver nanoparticles. We also performed RecA expression assay with western blotting and observed activation of SOS response to repair the damaged DNA. To summarize, silver nanoparticles are involved in the apoptosis-like response in E. coli and the novel mechanisms which were identified in this study, suggest that silver nanoparticles may be an effective antimicrobial agent with far lower propensity for inducing microbial resistance than antibiotics.  相似文献

Silver nanoparticle-embedded polyrhodanine nanofibers were synthesized by chemical oxidation polymerization. Silver ions were reduced to silver nanoparticles by oxidizing rhodanine monomer and simultaneously complexed with the rhodanine due to coordinative interactions, resulting in the formation of silver nanoparticle-embedded polyrhodanine nanofibers. The synthesized silver/polyrhodanine nanofiber was found to have the excellent antimicrobial activity against gram-negative Escherichia coli, gram-positive Staphylococcus aureus, and Candida albicans. The modified Kirby-Bauer test demonstrated that the silver/polyrhodanine nanofiber had better antimicrobial efficacy than silver sulfadiazine.  相似文献

Conditions for obtaining stable silver nanoparticles smaller than 10 nm were developed using a binary stabilizer polyvinylpyrrolidone/sodium dodecylsulphate in optimal ratio. Optical spectra, morphology and dependence of size of the nanoparticles on the amount of reducing agent were studied. Colloidal solutions of nanosilver showed a high bactericidal activity against strains of Staphylococcus aureus, Escherichia coli and Pseudomonas aeruginosa, and fungicidal activity against Candida albicans. The mechanism of action of nanosized silver on microbial cell was examined by laser scanning confocal microscope using fluorescent label. First step of antimicrobial effect on microorganisms was membrane damage and penetration of silver nanoparticles into the cell. Prolonged stability of nanoparticles and their antimicrobial activity over the past two years were showed.  相似文献

Mycoendophytes are the fungi that occur inside the plant tissues without exerting any negative impact on the host plant. They are most frequently isolated endophytes from the leaf, stem, and root tissues of various plants. Among all fungi, the mycoendophytes as biosynthesizer of noble metal nanoparticles (NPs) are less known. However, some reports showing efficient synthesis of metal nanoparticles, mainly silver nanoparticles and its remarkable antimicrobial activity against bacterial and fungal pathogens of humans and plants. The nanoparticles synthesized from mycoendophytes present stability, polydispersity, and biocompatibility. These are non-toxic to humans and environment, can be gained in an easy and cost-effective manner, have wide applicability and could be explored as promising candidates for a variety of biomedical, pharmaceutical, and agricultural applications. Mycogenic silver nanoparticles have also demonstrated cytotoxic activity against cancer cell lines and may prove to be a promising anticancer agent. The present review focuses on the biological synthesis of metal nanoparticles from mycoendophytes and their application in medicine. In addition, different mechanisms of biosynthesis and activity of nanoparticles on microbial cells, as well as toxicity of these mycogenic metal nanoparticles, have also been discussed.  相似文献

The goal of endodontic treatment is the debridement and removal of the microbial ecosystem associated with the disease process. The need for root canal disinfectants increases especially in those cases where infection is resistant to the regular treatment and the outcome of endodontic therapy is often compromised. Therefore, it is of interest to document the known effectiveness of silver nanoparticle based root canal disinfectants with other root canal disinfectants on microbial load reduction during root canal disinfection. Known data shows that the overall risk of bias for the selected studies was moderate. Silver nanoparticle based root canal disinfectants showed superior reduction of microbial counts in majority of the studies. This data is limited to vitro studies with no clinical information to validate the use of antimicrobial properties of silver nanoparticles used as root canal disinfectant.  相似文献

Polymer-silver nanocomposites modified cotton fabrics were prepared by in situ chemical oxidative polymerization using pyrrole and silver nitrate. In a redox reaction between pyrrole and silver nitrate, silver ions oxidize the pyrrole monomer and get reduced. This reduced silver as nanoparticles deposited on/into the polypyrrole/cotton matrix layer and the interaction between silver and polypyrrole was by adsorption or electrostatic interaction. The structure and composite formation on cotton fiber was investigated using SEM, FT-IR, XPS and XRD. The results showed that a strong interaction existing between silver nanoparticles with polypyrrole/cotton matrix. FT-IR studies clearly indicated that the interaction between polypyrrole (NH) and cellulose (>COH) was by hydrogen bonding. It is observed that the conductivity of the composite coated fabrics has been increased by the incorporation of silver nanoparticles. In the synthesized composites, silver content plays an important role in the conductivity and antimicrobial activity rate of the fabrics against gram positive Staphylococcus aureus and gram negative Escherichia coli bacteria.  相似文献

Silver has been in use since time immemorial in the form of metallic silver, silver nitrate, silver sulfadiazine for the treatment of burns, wounds and several bacterial infections. But due to the emergence of several antibiotics the use of these silver compounds has been declined remarkably. Nanotechnology is gaining tremendous impetus in the present century due to its capability of modulating metals into their nanosize, which drastically changes the chemical, physical and optical properties of metals. Metallic silver in the form of silver nanoparticles has made a remarkable comeback as a potential antimicrobial agent. The use of silver nanoparticles is also important, as several pathogenic bacteria have developed resistance against various antibiotics. 近年来,利用沼泽红假单胞菌合成银纳米粒子作为一种可靠和环境友好的方法出现。主要利用沼泽红假单胞菌的细胞滤液来还原银离子。制备的纳米粒子用紫外可见光谱(UV-vis)、X射线衍射光谱(XRD)和透射电镜(TEM)进行表征。含有银粒子溶液的UV-vis光谱显示在420 nm-460 nm处出现银纳米粒子的吸收峰。TEM图像表明所形成的银纳米粒子的粒径范围为5 nm-20 nm。纳米粒子的XRD图谱证明产物为金属银。所制备的银纳米粒子对大肠杆菌和金黄色葡萄球菌作抑菌性试验。  相似文献

Silver nanoparticles are well received in the cosmeceutical industry due to their broad spectrum of pharmacology applications. Research on the therapeutic properties exhibited by silver nanoparticles revealed that the antimicrobial and anti-inflammatory properties are the main attraction in the establishment of nanocosmeceutical products whereby their mechanisms of action are reviewed in this paper. In addition, studies on other uses of silver nanoparticles acknowledged that the particles act as antifungal agents in nail polishes and pigments in coloured beauty products such as lipsticks and eye shadows. Despite the extensive use of silver nanoparticles in the cosmetic line, there are still limited resources on the mechanism of actions and the effect of the particles on the bio-functionality of the body. The safety of silver nanoparticles could be comprehended from their skin penetration ability and toxicity to the human body in which it could be justified that both features are mainly influenced by the morphology of the particles and the method of application. This article summarizes exclusively on the synthesis of silver nanoparticles, the biomedical mechanisms and applications as well the limitations with respect to skin penetration ability and toxicity effects which will contribute significantly to the vast research on the association of nanotechnology and cosmetics.  相似文献

Comparable antimicrobial and disinfecting action of decamethoxine and silver preparations on pathogens of chronic purulent otitis media (CPOM) was studied. The clinical isolates of staphylococci proved to be most sensitive to decamethoxine whose MBcC conformed to 16.5 micrograms/ml. The antimicrobial action on Proteus spp. and Pseudomonas aeruginosa was less pronounced. The required concentrations for bactericidal action on these pathogens were 69 and 93.5 micrograms/ml, respectively. The antimicrobial activity of the silver preparations such as poviargol, collargol and protargol was low. Depending on the microbial species, the bactericidal effect of the silver preparations was 12-235 times lower than that of decamethoxin. It was also shown that decamethoxin had a high disinfecting action on CPOM pathogens. It was noted that decamethoxin had a marked ability to increase the bactericidal action of poviargol (by 2-14 times) and its disinfecting action (by 2 times) on Proteus spp., E. coli and Ps. aeruginosa.  相似文献

Much work has elucidated the pathways and mechanisms involved in the production of insect immune effector systems. However, the temporal nature of these responses with respect to different immune insults is less well understood. This study investigated the magnitude and temporal variation in phenoloxidase and antimicrobial activity in the mealworm beetle Tenebrio molitor in response to a number of different synthetic and real immune elicitors. We found that antimicrobial activity in haemolymph increased rapidly during the first 48h after a challenge and was maintained at high levels for at least 14 days. There was no difference in the magnitude of responses to live or dead Escherichia coli or Bacillus subtilis. While peptidoglylcan also elicited a long-lasting antimicrobial response, the response to LPS was short lived. There was no long-lasting upregulation of phenoloxidase activity, suggesting that this immune effector system is not involved in the management of microbial infections over a long time scale.  相似文献
