Programmed cell death can be divided into apoptosis and autophagic cell death. We describe the biological activities of TMEM166
(transmembrane protein 166, also known as FLJ13391), which is a novel lysosome and endoplasmic reticulum-associated membrane
protein containing a putative TM domain. Overexpression of TMEM166 markedly inhibited colony formation in HeLa cells. Simultaneously,
typical morphological characteristics consistent with autophagy were observed by transmission electron microscopy, including
extensive autophagic vacuolization and enclosure of cell organelles by double-membrane structures. Further experiments confirmed
that the overexpression of TMEM166 increased the punctate distribution of MDC staining and GFP-LC3 in HeLa cells, as well
as the LC3-II/LC3-I proportion. On the other hand, TMEM166-transfected HeLa and 293T cells succumbed to cell death with hallmarks
of apoptosis including phosphatidylserine externalization, loss of mitochondrial transmembrane potential, caspase activation
and chromatin condensation. Kinetic analysis revealed that the appearance of autophagy-related biochemical parameters preceded
the nuclear changes typical of apoptosis in TMEM166-transfected HeLa cells. Suppression of TMEM166 expression by small interference
RNA inhibited starvation-induced autophagy in HeLa cells. These findings show for the first time that TMEM166 is a novel regulator

Apoptosis in the malaria protozoan,Plasmodium berghei: a possible mechanism for limiting intensity of infection in the mosquito 总被引:8,自引:0,他引:8
Death by apoptosis regulates cell numbers in metazoan tissues and it is mediated by activation of caspases and results in characteristic morphological and biochemical changes. We report here that the malaria protozoan, Plasmodium berghei, exhibits features typical of metazoan apoptotic cells including condensation of chromatin, fragmentation of the nuclear DNA and movement of phosphatidylserine from the inner to the outer lamellae of the cell membrane. In addition, proteins with caspase-like activity were identified in the cytoplasm of the ookinete suggesting that the cellular mechanism of cell death may be similar to that of multicellular eukaryotes. Our data show that more than 50% of the mosquito midgut stages of the parasite die naturally by apoptosis before gut invasion. Cell death was prevented by a caspase inhibitor, treatment resulting in a doubling of parasite intensity. All these features also occur in vitro. Cell suicide thus plays a major and hitherto unrecognised role in controlling parasite populations and could be a novel target for malaria control strategies. 相似文献

Natural products are considered a good choice in the biological control of mosquitoes because they are an effective way to eliminate larvae and prevent an increase in mosquito numbers, while simultaneously not polluting the environment or damaging health. This investigation was designed to study the potential toxicity of three species of algae, Caulerpa racemosa (Weber-van Bosse, 1909), Padina boryana (Thivy, 1966), and Turbinaria ornata (Turner J. Agardh, 1848), on the larvae of Aedes aegypti mosquito, the vector of dengue and Zika viruses. Among the studied species, Caulerpa racemosa showed the greatest effectiveness in eradicating A. aegypti larvae with an LC50 = 43.5 ppm, followed by Padina boryana with an LC50 = 51.93 ppm. Both species proved to be excellent candidates as a source of larvicidal agents and could be used commercially in mosquito control programs as eco-friendly biopesticides. The combined activity of different mixtures against mosquito larvae was expressed as the coeffective factor (C.F.). C.F. values showed that the joint activity of insect growth regulator Dudim in combination with Caulerpa racemosa and Padina boryana extracts produced degrees of potentiation effects and degrees of additive effects were obtained with Dudim in combination with Turbinaria ornata extract. 相似文献

The cDNA corresponding to an inhibitor of apoptosis (IAP) from the Egyptian armyworm, Spodoptera littoralis, was cloned by RT-PCR. Sequence analysis showed that the IAP of S. littoralis (SlIAP) contains two baculoviral IAP repeat (BIR) motifs, followed by a RING finger, an organization which is very similar to that of other lepidopteran IAPs. SlIAP mRNA was detected in ovary, testis, salivary gland, fat body, epidermis, brain and midgut of S. littoralis. During the last larval instar, prepupal and pupal stages, brain mRNA levels remained approximately constant, whereas those of midgut showed a large peak centred in the prepupal stage. Midgut morphology changed during metamorphosis from a semi-transparent, cylindrical structure in last instar larvae to a brownish globular mass in pupae. TUNEL assays, LysoTracker staining and caspase-3 immunohistochemistry, indicated that programmed cell death in midgut starts actively at the onset of pupation process, coinciding with the dramatic decrease of SlIAP mRNA levels observed at the same time. 相似文献

Haematophagy, the utilization of blood as food, has evolved independently among insects such as mosquitoes, bedbugs, fleas, and others. Accordingly, several distinct biological adaptations have occurred in order to facilitate the finding, ingestion and digestion of blood from vertebrate sources. Although blood meals are essential for survival and reproduction of these insects, mechanical and chemical stresses are caused by the ingestion of a sizable meal (frequently twice or more times the weight of the insect) containing large amounts of cytotoxic molecules such as haem. Here we present data showing that the stresses caused by a blood meal induce cell death in the midgut epithelium of Culex quinquefasciatus mosquitoes. The process involves apoptosis, ejection of dead cells to the midgut lumen and differentiation of basal regenerative cells to replace the lost digestive cells. The basal cell differentiation in blood-fed mosquito midguts represents an additional mechanism by which insects cope with the stresses caused by blood meals. C. quinquefasciatus adult females are unable to replace lost cells following a third or fourth blood meal, which may have a significant impact on mosquito longevity, reproduction and vectorial capacity. 相似文献

The mosquito Aedes aegypti is an increasing problem of public health, being the vector responsible for dengue and Yellow Fever in tropical and subtropical regions. The aim of this work was to determine the potential larvicidal activity of a series of meroterpenoids, compounds 1-7, previously obtained fungal secondary metabolites from Penicillium sp., against the third-instar larvae of A. aegypti. The lethal concentrations (LC(50) and LC(90)) of 1-7 were evaluated 24 h after exposure. Dehydroaustin (4) was the most active meroterpenoid in the series, with an LC(50) value of 2.9 ppm, making it an attractive natural insecticide. 相似文献

Flaviviruses, ss(+) RNA viruses, include many of mankind's most important pathogens. Their pathogenicity derives from their ability to infect many types of cells including neurons, to replicate, and eventually to kill the cells. Flaviviruses can activate tumor necrosis factor α and both intrinsic(Bax-mediated) and extrinsic pathways to apoptosis. Thus they can use many approaches for activating these pathways. Infection can lead to necrosis if viral load is extremely high or to other types of cell death if routes to apoptosis are blocked. Dengue and Japanese Encephalitis Virus can also activate autophagy. In this case the autophagy temporarily spares the infected cell, allowing a longer period of reproduction for the virus, and the autophagy further protects the cell against other stresses such as those caused by reactive oxygen species. Several of the viral proteins have been shown to induce apoptosis or autophagy on their own, independent of the presence of other viral proteins. Given the versatility of these viruses to adapt to and manipulate the metabolism, and thus to control the survival of, the infected cells, we need to understand much better how the specific viral proteins affect the pathways to apoptosis and autophagy. The Class II transposable element, piggyBac, was used to transform the yellow fever mosquito, Aedes aegypti. In two transformed lines only 15–30 of progeny inherited the transgene, with these individuals displaying mosaic expression of the EGFP marker gene. Southern analyses, gene amplification of genomic DNA, and plasmid rescue experiments provided evidence that these lines contained a high copy number of piggyBac transformation constructs and that much of this DNA consisted of both donor and helper plasmids. A detailed analysis of one line showed that the majority of piggyBac sequences were unit-length donor or helper plasmids arranged in a large tandem array that could be lost en masse in a single generation. Despite the presence of a transposase source and many intact donor elements, no conservative (cut and paste) transposition of piggyBac was observed in these lines. These results reveal one possible outcome of uncontrolled and/or unexpected recombination in this mosquito, and support the conclusion that further investigation is necessary before transposable elements such as piggyBac can be used as genetic drive mechanisms to move pathogen-resistance genes into mosquito populations. 相似文献

Population genetic studies of insect vectors can generate knowledge to improve epidemiological studies focused on the decrease of pathogen transmission. In this study, we used nine SNPs across the Aedes aegypti genome to characterize seasonal population variations of this important dengue vector. Mosquito samples were obtained by ovitraps placed over Botucatu SP from 2005 to 2010. Our data show that, regardless of the large variation in mosquito abundance (deduced from the number of eggs obtained from ovitraps), the effective population size remained stable over the years. These results suggest that Ae. aegypti is able to maintain a sufficiently large active breeding population during the dry season to keep genetic frequencies stable. These results open new perspectives on mosquito survey and control methods. 相似文献

We selected six lines of mosquito Aedes aegypti for earlier or later pupation and measured the correlated responses of several life history traits: adult size, two fecundity measures and pre-adult survival. We further examined the influence of two environmental parameters – larval food availability and infection by the microsporidian parasite Edhazardia aedis– on the correlated responses. Pre-adult survival did not respond to selection for age at pupation in any environment. For all of the other traits, the environment influenced the correlated response, though the contribution of the different environmental aspects differed among traits. While the correlated response of adult size was influenced only by larval food availability, the likelihood that a female laid eggs was influenced by parasite infection, and the correlated response of the number of eggs was influenced by the interaction of the two environmental parameters. Generally, a deteriorating environment moved the correlated response from one favouring later pupation to one favouring earlier pupation. Larval food availability and parasite infection also influenced the association between the mean wing length and fecundity of the selected lines. At high food availability, there was a positive relationship between adult size and fecundity, while infected mosquitoes reared at low food availability showed the opposite trend. We discuss these results in light of the coevolutionary potential of the host–parasite interaction. 相似文献

Programmed cell death (PCD), now known as apoptosis, is accompanied by specific morphological features. In this study, fusaric acid, a fusarium mycotoxin, was used to examine cell death in saffron (Crocus sativus Linnaeus) roots, using several apoptosis assays. Our results show that moderate FA doses (50–100 μM) induce apoptotic features while high FA doses (> 200 μM) stimulate necrosis. The apoptotic-like features induced by moderate doses of FA include chromatin condensation, formation of condensed chromatin spheres which bud from the nucleus, fragmentation of nucleosomal DNA into ∼ 180 bp fragments, exposure of phosphatidyl serine to the external membrane leaflet, delivery of cytochrome c to cytosol, and generation of H2O2. These apoptotic alterations in root cells are not observed in the presence of serine protease, caspase-1 or caspase-3 inhibitors. It is proposed that production of H2O2 and release of cytochrome c into the cytosol may activate caspase-like proteases and thus establish the apoptotic pathway. As nuclei budding spheres formed in plant root cells after exposure to 50–100 μM FA doses seem to be digested inside the cytosol, we suggest labeling them as internal apoptotic bodies (IAB) that may be more informative than previously used term, apoptotic-like bodies.Electronic Supplementary Material Supplementary material is available to authorised users in the online version of this article at . 相似文献

In Puerto Rico, the first records of the transmission of Chikungunya (CHIKV) and Zika (ZIKV) viruses were confirmed in May 2014 and December 2015, respectively. Transmission of CHIKV peaked in September 2014, whereas that of ZIKV peaked in August 2016. The emergence of these mosquito‐transmitted arboviruses in the context of a lack of human population immunity allowed observations of whether the outbreaks were associated with Aedes aegypti (Diptera: Culicidae) densities and weather. Mosquito density was monitored weekly in four communities using sentinel autocidal gravid ovitraps (AGO traps) during 2016 in order to provide data to be compared with the findings of a previous study carried out during the 2014 CHIKV epidemic. Findings in two communities protected against Ae. aegypti using mass AGO trapping (three traps per house in most houses) were compared with those in two nearby communities without vector control. Mosquito pools were collected to detect viral RNA of ZIKV, CHIKV and dengue virus. In areas without vector control, mosquito densities and rates of ZIKV detection in 2016 were significantly higher, similarly to those observed for CHIKV in 2014. The density of Ae. aegypti in treated sites was less than two females/trap/week, which is similar to the putative adult female threshold for CHIKV transmission. No significant differences in mosquito density or infection rates with ZIKV and CHIKV at the same sites between years were observed. Little is known about mating, ovariole number and sperm production of Aedes aegypti (L.) in Australia, especially in relation to climate. To determine the extent of interpopulation variation and thermal dependence of reproductive traits in A. aegypti, laboratory studies are conducted using colonies originating from up to four locations in Queensland, Australia. Observations of insemination reveal that these behaviours are temperature‐dependent, although humidity levels appear to have little effect, with only small increases in insemination at higher humidities. No noteworthy variations in thermal optima (temperature ranges within which maximal performance occurs) for such behaviours are observed between colonies, with all showing high levels of insemination between 25 and 35 ° C. Both male and female maximum fecundity for A. aegypti are also found to be temperature‐dependent. Sperm counts, not hitherto obtained for Australian A. aegypti, range from approximately 3000–5000 per male, with counts increasing with increased rearing and holding temperature, despite a decreasing body size. Conversely, ovary size decreases with temperature and body size, from approximately 100 ovarioles per female at 17 °C, to 85 at 35 °C. The lack of variation in reproductive capacity between colonies of different geographical origin indicates that any locally‐acting selective pressures are not driving divergence in key reproductive traits such as insemination ability and fecundity. This may be because the source populations used are not from sufficiently diverse climates. Nonetheless, the broad thermal optima for reproductive traits in A. aegypti are suggestive of limitations on these traits not being responsible for limiting distribution and population growth in the event of projected mean temperature rises. 相似文献
