Summary The application of the overfeeding technique (interruption of the competition during larval development) to the study of larval competition in two-strain cultures of Drosophila melanogaster demonstrates the following points: (1) viability is a function of competition time; (2) viability becomes more frequency-dependent as competition time increases; (3) the dynamics of the inner subpopulation (adults that have passed all their development in a crowded condition) and outer subpopulation (adults coming from larvae recovered by interruption of competition) vary with time as regards frequency-dependence; and (4) the wild type strain Oregon is the active agent in competition with the strain cardinal.  相似文献

Summary The surface structure of the compound eyes of 6 Drosophila species and 12 eye mutants of D. melanogaster were compared by scanning electron microscopy. D. melanogaster, D. simulans, D. hydei, D. funebris and D. virilis displayed hexagonal facets and differed only slightly in the distribution of bristles. D. lebanonensis displayed tetragonal facets.No obvious differences in surface structure of the eyes of colour mutants of D. melanogaster were found. Mutants with structural modifications of the eyes revealed irregular patterns of bristles, variations in bristle number and variations in facet shape, size and organization. The mutant spapol does not display clear-cut delineated facets.  相似文献

Survival (viability) of newly eclosed adults of D. simulans st and D. melanogaster Or R-C, which had excess quantities of dead yeast available throughout life, was measured daily for 7 days in an experiment where adult density (6 levels), species frequency (6 levels) and 32P tissue content (2 levels) were varied factorially. A separate experiment comparing viability in different types of experimental unit also was done. Similar experiments were done for D. simulans st competing against D. melanogaster y w. The entire data were subjected to a least-squares (unbalanced, missing-plot) analysis of variance. D. melanogaster Or R-C had a higher average viability than D. melanogaster y w or D. simulans st, which were equal. The competing strain of D. melanogaster influenced D. simulans st viability — viability being higher when y w was the competitor. Viability decreased over the seven day period but at different rates for the three strains. Increasing density reduced viability for all three strains, but species frequency effects, although significant, were generally not consistent. Females had higher viability than males in both D. melanogaster strains, but the reverse was true in D. simulans st. 32P lowered viability and experimental unit type altered viability. Numerous interactions were significant.Adult density was shown to have a delayed effect on viability — the delay before the appearance of the effect (an increase in death rate) being decreased as density rose. The term variably delayed density dependent has been adopted to describe the fitness component, adult viability. Some high density populations showed a readjustment (a decrease) in their death rate as a reaction to the effect of reduced density caused by high early mortality.  相似文献

Mating in the dark between individuals carrying different eye color mutants extracted from the same natural population is shown to be random. (Previous experiments had shown that darker-eye males mate more successfully under alternating light-dark conditions). Extraction of pigments and quantitative analysis by absorption curves show that all mutants have less pigmentation than wild-type individuals; unexpectedly, darker-eye mutants do not resemble the wild-type level of pigmentation any more than light-eye single mutants. The results suggest that (1) eye pigmentation is not neccessarily related to visual ability; (2) some kind of pleiotropic effect affecting the central nervous system may be involved in the sexual behaviour of male mutants.  相似文献

Uptakes of guanine into Malpighian tubules of wild-type Drosophila and the eye color mutants white (w), brown (bw), and pink-peach (p p) have been compared. Tubules for each of these mutants are unable to concentrate guanine intracellularly. The transport of xanthine and riboflavin is also deficient in w tubules. The transport of guanosine, adenine, hypoxanthine, and guanosine monophosphate is similar in wild-type and white Malpighian tubules. These data and other information about these mutants make it likely that these pteridine-deficient eye color mutants do not produce pigments because of the inability to transport a pteridine precursor. This view supports the hypothesis that mutants which lack both pteridine and ommochromes do so because precursors to both classes of pigments share a common transport system.This work was supported by Grant GM22366 from NIH.  相似文献

Kynurenine-H 3 transport and conversion to 3-hydroxykynurenine were studied in organ culture using the Malpighian tubules and developing eyes from wild type and the eye color mutants w, st, 1td, ca, and cn of Drosophila melanogaster. Malpighian tubules from wild type have the ability to concentrate kynurenine and convert it to 3-hydroxykynurenine. The tubules from w, st, 1td, and ca are deficient in the ability to transport kynurenine, as are the eyes of the mutants w, st, and 1td. This defect in kynurenine transport provides a physiological explanation for the phenotypic properties of the mutants. The relationship of these measurements to previous observations on these eye color mutants is discussed and the transport defect hypothesis is consistently supported. We have concluded that several of the eye color mutants in Drosophila are transport mutants.  相似文献

Both the amount and the size of alcohol dehydrogenase-like cross-reacting material was determined in 14 ethyl methanesulfonate (EMS)-induced alcohol dehydrogenase-null activity mutants. In 11 mutants cross-reacting material of the same apparent molecular weight as alcohol dehydrogenase was detected, while in 3 mutants no cross-reacting material was found. In all cases, the amount of cross-reacting material found in the mutants was lower than that in wild-type flies. High, intermediate, and low cross-reacting material-producing mutants showed similar initial rates of incorporation of labeled amino acid into alcohol dehydrogenase-like protein, presumably reflecting similar rates of synthesis. If the rate of synthesis of cross-reacting material is the same in the mutants as in the wild type, then the different levels of cross-reacting material must be due to different rates of degradation.Supported by NIH Grants GM-18254 and ES-01527 and DOE Contract EY-76-S-2965.  相似文献

The functions of sleep are still unknown, but are probably related to cellular and molecular aspects of neural function. To better understand the benefits that sleep may bring at the cellular level, recent studies have employed Drosophila melanogaster as a model system and shown that fruit flies share the fundamental features of mammalian sleep. As in mammals, sleep in Drosophila is characterized by increased arousal threshold and by changes in brain electrical activity. Fly sleep is homeostatically regulated independent of the circadian clock, is modulated by stimulants and hypnotics, and is affected by age. Also, fly sleep is associated with changes in brain gene expression similar to those observed in mammals. While Drosophila neurobiology is sufficiently complex to permit meaningful generalizations to mammals and humans, Drosophila genetics is simple enough to allow a rapid mutagenesis screening. An ongoing mutagenesis study has screened approximately 5000 mutant Drosophila lines and found that sleep amount, sleep pattern, and the homeostatic regulation of sleep are highly conserved phenotypes in flies. So far, this study has identified 10 short sleeper lines and 4 lines that show no sleep rebound after sleep deprivation. Ultimately, the characterization of these lines should help identifying crucial cellular pathways involved in the regulatory mechanisms of sleep and its functional consequences.  相似文献

In previous work analyzing variability of eye colour alleles existing in natural populations of D melanogaster, it was observed that the number of females heterozygous for some eye colour alleles was greater in a wine cellar population than in populations outside this cellar. In order to determine which mechanisms caused these eye colour alleles to be favored in the heterozygotes, the changes in the frequency of four eye colour alleles frequently seen in the cellar population (se77o, sf77m, cd77o and multichromosomal 77o) was studied in artificial populations. Two different culture media, one supplemented with 10% ethanol and the other without ethanol were used. It was found that each of the four mutants reached similar equilibrium frequencies in both media, though the safranin allele (sf77m) equilibrium frequency was significantly higher in the alcohol medium. A significant excess of heterozygotes were also observed in these populations.  相似文献

There have been several attempts to estimate the average dominance (ratio of heterozygous to homozygous effects) of spontaneous deleterious mutations in Drosophila melanogaster, but these have given inconsistent results. We investigated whether transposable element (TE) insertions have higher average dominance for egg-to-adult viability than do point mutations, a possibility suggested by the types of fitness-depressing effects that TEs are believed to have. If so, then variation in dominance estimates among strains and crosses would be expected as a consequence of variation in TE activity. As a first test, we estimated the average dominance of all mutations and of copia insertions in a set of lines that had accumulated spontaneous mutations for 33 generations. A traditional regression method gave a dominance estimate for all mutations of 0.17, whereas average dominance of copia insertions was 0.51; the difference between these two estimates approached significance (P = 0.08). As a second test, we reanalyzed Ohnishi 1974 data on dominance of spontaneous and EMS-induced mutations. Because a considerable fraction of spontaneous mutations are caused by TE insertions, whereas EMS induces mainly point mutations, we predicted that average dominance would decline with increasing EMS concentration. This pattern was observed, but again fell short of formal significance (P = 0.07). Taken together, however, the two results give modest support for the hypothesis that TE insertions have greater average dominance in their viability effects than do point mutations, possibly as a result of deleterious effects of expression of TE-encoded genes.  相似文献

We investigated the effects of genotypic frequencies on egg-to-adult viabilities in pairwise combinations of four strains of Drosophila melanogaster. The experiments involved mixture of a total of 42,000 eggs in varying proportions under controlled densities and observation of surviving adults. Viabilities were found to depend on frequencies in several genotypic combinations. In the most extreme case, the absolute viability of cn;bw females increased monotonically from 54% when common to 70% when rare. The results illustrate several statistical and methodological problems that might explain why some experiments have failed to detect frequency-dependent viabilities. These problems include heterogeneity between replications, sex differences in susceptibility to competition, and strong dependence of the experimental outcome on the choice of competitor genotypes.  相似文献
