Plants of potato (Solanum tuberosum) cultivars Katahdin and Superior were inoculated with 0, 1,500, or 15,000 Pratylenchus penetrans. Transpiration, measured in the greenhouse with a porometer after 56 days of growth, was not significantly different among nematode inoculum levels or between cultivars. The rate of xylem exudation from decapitated root systems of Katahdin plants inoculated with 1,500 or 15,000 P. penetrans and Superior plants inoculated with 15,000 P. penetrans was lower than from noninoculated plants. Root weight of Katahdin and Superior was not affected by P. penetrans inoculum level. Transpiration of plants inoculated with 0, 500, 5,000 or 50,000 P. penetrans was recorded weekly from 14 to 56 days after planting. No consistent effects of nematode inoculum density on transpiration rate were observed. Root hydraulic conductivity was lower in Katahdin plants inoculated with 266 P. penetrans per plant and in Chippewa with 5,081 per plant than in noninoculated plants. Nematodes reduced leaf area of Superior, Chippewa, and Katahdin and root dry weight of Chippewa but had no effect on growth of Hudson, Onaway, or Russet Burbank plants. Assessing nematode effects on root hydraulic conductivity may provide a measure of the tolerance of potato cultivars to nematodes.  相似文献

Reproduction of Pratylenchus penetrans on the potato cultivars Hudson, Katahdin, and Superior was determined in greenhouse and field microplot experiments. Although all three cultivars were good hosts for P. penetrans, differences in reproductive rate were found. In one greenhouse experiment, Katahdin plants inoculated with 1,500 or 15,000 P. penetrans per pot had larger population densities at harvest than did Superior; however differences between these cultivars were not significant in three other greenhouse experiments. In another experiment, population densities of P. penetrans on Hudson did not differ from those on Katahdin and Superior when inoculated with 270 and 5,080 nematodes per pot after 45 days in the greenhouse. However, population densities were usually higher on Hudson and Katahdin than on Superior in field microplots at four initial population densities during two seasons. Higher population densities on Hudson were detectable 304 days after planting in one of the two microplot studies. The juvenile:female and the male:female ratios were sometimes larger on Katahdin than on Superior, but differences were inconsistent. There was no evidence of resistance in the three cultivars evaluated, but reproduction was generally highest on Hudson and lowest on Superior.  相似文献

Potato cultivars Katahdin (susceptible) and Rosa (resistant) were exposed to infective second-stage juveniles (J2) of Globodera rostochiensis for varying periods of time, after which root systems were washed and plants were placed in Hoagland''s solution to assess J2 egression and male emergence. After transfer to liquid culture, many J2 egressed from both cultivars, but significantly more egressed from the resistant Rosa than from Katahdin. Juveniles that egressed from Rosa invaded a second host, resistant or susceptible, in significantly fewer numbers than did juveniles that egressed from Katahdin. Also, significantly fewer males developed in and emerged from resistant host roots, relative to susceptible ones. These effects of resistance may be an important component of the tolerance to invasion by G. rostochiensis exhibited by Rosa.  相似文献

The fecundity of Globodera rostochiensis (R₁A) females that developed on resistant Rosa and susceptible Katahdin potato cultivars were compared. Cysts collected from each cultivar were bulked, separated into four sizes (> 500 μm, 355-500 μm, 250-355 μm, and < 250 μm), and crushed to determine fecundity as measured by viable egg content (VEC). Fewer and generally smaller cysts developed on Rosa than on Katahdin. Although cyst size significantly (P = 0.01) influenced VEC, cyst age (8 or 13 weeks) had no effect. Regardless of size, cysts produced on Rosa contained significantly fewer viable eggs than did cysts produced on Katahdin. The fecundity of progeny from cysts produced on Rosa was significantly reduced compared with that of progeny from cysts produced on Katahdin. After two generations on Katahdin, the VEC of cysts from a population originating from Rosa was significantly less than that of cysts from a population originating from Katahdin, indicating that in the presence of a pure population of G. rostochiensis R₁A, the females that develop on the resistant cultivar Rosa represent a diminished rather than a superior selected population.  相似文献

Pratylenchus penetrans suppressed the tuber yields of potato cultivars ''Katahdin'', ''Kennebec'', and ''Superior'', but did not affect yields of ''Russet Burbank''. In comparison with noninfested controls, all initial nematode densities (Pi) of P. penetrans (Pi = 38, 81, 164, 211/ 100 cm³ of soil) suppressed yields of Superior; a moderate Pi (81/100 cm³ soil) suppressed yields of Kennebec; and on Katahdin, a moderate Pi enhanced yields, but higher Pi''s caused a marked loss. In general, yields were related to the tolerance of the cultivars to nematode colonization. Highest nematode densities were found in the roots of Russet Burbank; the next highest, in succeeding order, were found in roots of Kennebec, Katahdin, and Superior. Symptoms of nematode invasion were confined to losses of tuber yield and root weight.  相似文献

The influence of Pratylenchus penetrans on the incidence and severity of Verticillium wilt was examined in the potato cultivars ''Kennebec'', ''Katahdin'', and ''Abnaki''. Single-stem plants were grown in soil maintained at a temperature of 22 ± 1 C. Axenically cultured nematodes were suspended in water and introduced to the soil, at a rate of ca 5,000/25.4-cm pot, through holes made around each stem. Ten days after infestation with nematodes, conidial suspensions of Verticillium albo-atrum were introduced into the soil at a rate of ca 1,000,000/pot. Among Katahdin plants, the severity of foliar symptoms was increased in the presence of both pathogens 2 and 3 weeks after soil intestation. During the remaining 5 weeks, severity of foliar symptoms was not different between plants infected by both pathogens and those infected by Verticillium alone. Within the wilt-susceptible cultivar Kennebec and the resistant eultivar Abnaki, no effects on foliar symptom severity were observed. When plant heights, shoot weights, and tuber yields were analyzed, a Pratylenchus-Verticillium interaction was not evident within any of the cultivars tested. Nematode populations in roots and rhizosphere were suppressed in Kennebec and Katahdin plants in the presence of Verticillium.  相似文献

Brine shrimp Artemia (Crustacea, Anostraca) diverge in biometry and nutritional quality. These differences in Artemia characteristics are significant not only from strain to strain but also from one harvest to another within same strain. The main objective of this study was to compare Artemia salina cysts harvested from Sabkhet El Adhibet (southeast Tunisia) on different dates between 2002 and 2007 with special regard to their use in aquaculture, using cysts and naupliar biometrics, protein, carbohydrate, and lipid content. Fatty acid profiles as well as hatching characterisation were also evaluated. Hydrated cysts measures ranged between 258.1 and 263.7 μm, while the freshly hatched nauplii of Artemia measures ranged between 458.1 and 476.1 μm. Lipid contents of the samples ranged from 16.2 to 18.3% of the dry weight. Fatty acid profiles showed that cysts from Sabkhet El Adhibet contain a high quantity of eicosapentaenoic acid (20: 5n-3) with a percentage ranging between 7.8 and 14.3% of the dry weight. The highest hatching efficiency was obtained for decapsulated cysts collected in 2007 (139500 nauplii g−1 of cysts). Cysts treated with hydrogen peroxide had a hatching percentage of 14.49 to 42.99%. The hatching synchronization time for untreated cysts varied between 23.5 to 27.4 h.  相似文献

Hatching studies with Heterodera glycines typically have been conducted with a mixture of egg-mass and encysted eggs. Laboratory research was conducted to compare hatching of H. glycines eggs from external egg masses with that of eggs extracted from within females and cysts (encysted eggs). Egg-mass eggs were collected by soaking infected soybean roots in 0.5% sodium hypochlorite, and encysted eggs were collected from females and cysts dislodged from the same roots with a stream of water. Eggs were incubated at 25 °C in deionized water, 3.0 mM ZnSO₄solution, or one of three synthetic H. glycines hatch inhibitors, mad hatched juveniles were counted every other day for 22 days. Samples of eggs collected at the beginning and end of all experiments were analyzed to determine extent of embryo development. Egg-mass eggs hatched more rapidly than encysted eggs during the first 16 days, but not thereafter. Throughout the experiments, hatch of egg-mass eggs in deionized water was greater than that of encysted eggs. From day 8 to day 22, egg-mass eggs were less sensitive than encysted eggs to the hatch inhibitor 2-(2''-carboxyethyl)-5-[carboxy(hydroxy)methylidenyl]cyclopentanone. A greater proportion of egg-mass eggs contained vermiform juveniles than did encysted eggs at the beginning of the experiments, but not at the end. Results indicated that H. glycines egg-mass and encysted eggs have different hatching behaviors that cannot be explained entirely by differences in embryological development.  相似文献

Root diffusate from soybean plants caused greater hatching of Heterodera glycines eggs during vegetative growth of the host, but the activity declined with plant senescence. Chelation of the root diffusate with ethylenediamine tetraacetic acid (EDTA) significantly increased hatching activity for H. glycines eggs. Diffusate from leafless plants caused little hatching, whereas treatment of intact plants with the growth regulators gibberellin and kinetin had no effect on the hatching activity of root diffusate. Treating H. glycines eggs with zinc chloride and root diffusate reduced egg hatching from zinc chloride alone. Levels of zinc in the root diffusate were insufficient to induce egg hatch, based on analysis by atomic absorption spectrophotometry. The enzymatic activity of leucine aminopeptidase in H. glycines eggs was not altered by treatment with chelated or nonchelated root diffusate.  相似文献

cAMP levels in eggs of G. rostochiensis and the diameter of the nucleolus of the nucleus within the dorsal pharyngeal gland cell of the second stage juvenile have been measured as indicators of the response of the nematode to the hatching stimulus in potato root diffusate. The nucleolus increased from 2.72 ± 0.103 μm for unhatched individuals to 3.28 ± 0.14 μm and 3.88 ± 0.15 μm after soaking eggs in potato root diffusate for 3 and 4 days respectively. Juveniles expressed from unstimulated eggs in water to potato root diffusate for 4–5 days showed a similar increase in size of the nucleolus to 3.94 ±0.15 μm but those released into water for this time had smaller nucleoli of 3.20 ± 0.98 μm. The change in diameter of the nucleolus is probably related to the accumulation of secretions in this gland cell before hatching. Preliminary results with dibutyryl analogues of CAMP and cGMP showed some inhibition of hatch in 10% potato root diffusate. Theophylline had a similar effect but NaF was dissimilar in that the effect of this inhibitor was not reversible. A standard radioimmunoassay showed that significant changes in cAMP levels occurred in the unhatched juveniles within cysts after treatment with potato root diffusate for 2.5 or 8 h compared with values for cysts kept in water. This change occurs before other known responses of the juveniles to potato root diffusate and it defines the period of interest for future work on the initial action of hatching factor.  相似文献

In a comparison of four potato varieties, in-soil hatch of the golden potato cyst nematode (Globodera rostochiensis) was positively correlated to in vitro hatch in response to potato root leachate (PRL). The in-soil hatch of cysts of G. rostochiensis to two of the four varieties was significantly less than that of the control (cysts in gravel without potato plants) in the first 2 wk after plant emergence, suggesting the production of hatching inhibitors (HIs) by young potato plants. The hatching factor: hatching inhibitor ratio of PRL was positively associated with the net hatching activity of the PRL. Four zones of HI activity were resolved following gel permeation chromatography of PRL on Sephadex G-10. Hatch-inactive chemicals, which stimulated the activity of hatching factors (HFs) in PRL (hatching factor stimulants, HSs), were also isolated from PRL, hatch levels induced by individual HFs responding differently to the same HS preparation. The complex interactions between individual HFs and other hatching chemicals in PRL was illustrated when addition of the hatch-active potato glycoalkaloid α-solanine caused both inhibition and stimulation of PRL-induced hatch, depending on the α-solanine concentration.  相似文献

The effects of broadleaf tobacco, tomato, and black nightshade on juvenile hatch and reproduction of Globodera tabacum tabacum were determined in laboratory and greenhouse experiments. Root exudates from nightshade stimulated greater egg hatch than those from either ''Rutgers'' tomato or ''86-4'' tobacco. Hatch was greater at higher proportions of root exudates for all three plant species. Root exudates from plants greater than 3 weeks old stimulated more hatch than younger plants. No regression relationships existed between plant age and nematode batch. In other experiments, hatch from eggs in cysts was higher for tomato and nightshade after 10 weeks in greenhouse pots compared to tobacco and bare soil. Numbers of second-stage juveniles in eggs in cysts produced from a previous generation on the same host were highest on nightshade and less on tomato and tobacco. Cysts of variable age recovered from field soil had increased hatch in both root exudates or water compared to recently produced cysts from plants in growth chambers. Globodera t. tabacum may be subject to both host and environmentally mediated diapause.  相似文献

THE HATCHING RESPONSES OF SOME ROOT EELWORMS OF THE GENUS HETERODERA   总被引:2,自引:0,他引:2  
An account is given of investigations on the hatching responses, under laboratory conditions, of nine species or varieties of Heterodera , namely, the beet, cabbage, clover, Galeopsis , carrot, hop, potato, oat and pea eelworms. In the first seven of these, marked differences occurred in hatching responses from cysts incubated in root leachings from various plants, these differences being virtually diagnostic for the eelworm species concerned. In general, good responses to leachings from host plants occurred, whereas there was little or no response in non-host leachings; but cases of response to non-hosts and of failure to respond to hosts were encountered. Promising results were obtained in the analysis, by bio-assay, of mixtures of some of these eelworm species. No appreciable response occurred from cysts of oat and pea eelworms incubated in host leachings, but there was some evidence that such a response did occur from pea eelworm cysts under field conditions. Curves for rate of hatching of seven species and for hatching in diluted leachings in five species were found to be of the form described by Fenwick for potato eelworm. Results are given of some experiments on the effect of age of plant on potency of the root leachings, and on the loss of potency during storage.  相似文献

Plant–parasite coevolution has generated much interest and studies to understand and manage diseases in agriculture. Such a reciprocal evolutionary process could lead to a pattern of local adaptation between plants and parasites. Based on the phylogeography of each partner, the present study tested the hypothesis of local adaptation between the potato cyst nematode Globodera pallida and wild potatoes in Peru. The measured fitness trait was the hatching of cysts which is induced by host root exudates. Using a cross‐hatching assay between 13 populations of G. pallida and root exudates from 12 wild potatoes, our results did not show a strong pattern of local adaptation of the parasite but the sympatric combinations induced better hatching of cysts than allopatric combinations, and there was a negative relationship between the hatching percentage and the geographical distance between nematode populations and wild potatoes. Moreover, a strong effect of the geographic origin of root exudates was found, with root exudates from south of Peru inducing better hatching than root exudates from north of Peru. These results could be useful to develop new biocontrol products or potato cultivars to limit damages caused by G. pallida.  相似文献

Aqueous solutions of 5-500 μg/ml aldicarb inhibited hatching of Heterodera schachtii. Addition of hatching agents, zinc chloride, or sugarbeet root diffusate, to the aldicarb solutions did not decrease the inhibition of hatching. When cysts were removed from the aldicarb solufions and then treated for 4 wk in sugarbeet root diffusate, larvae hatched and emerged. Treatments of newly hatched larvae of H. schachtii with 5-100 μg/ml aldicarb depressed later development of larvae on sugarbeet (Beta vulgaris). Similar treatments with aldicarb sulfoxide had less effect on larval development, and aldicarb sulfone had no effect. Numbers of treated larvae that survived and developed were inversely proportional to concentration (0.1-5.0 μg/ml) and duration (0-14 days) of aldicarb treatments. Development of H. schachtii on sugarbeet grown in aldicarb-treated soil was inversely proportional to the concentration of aldicarb in the tested range of 0.75 - 3.0 μg aldicarb/g of soil. Transfer of nematode-infected plants to soil with aldicarb retarded nematode development, whereas transfer of plants first grownin treated soil to nematode-infested soil only slightly suppressed nematode development. Development of H. schachtii was inhibited in slices of storage roots of table beet (B. vulgaris), sugarbeet and turnip, (Brassica rapa), that had grown in soil treated with aldicarb.  相似文献

Golden nematode, Globodera rostochiensis (GN) population decline under resistant potatoes was related to cyst distance from plants 23 cm apart in rows 92 cm apart. GN decline, determined by sampling an infested field planted to the resistant cultivar ''Yankee Chipper'', was 81.8% in cores 11.5 cm from plants within rows. Decline was 27.4% at 23 cm from plants between rows and 36.6% at 46 cm. Population decline of juveniles in cysts added to soil in bags was 90.3% for cysts 11.5 cm from plants within rows planted to the resistant cultivar ''Rosa''. Decline between rows was 83.5, 76.9, and 60.4% at 11.5, 23.0, and 46.0 cm from plants, respectively. Maximum decline within for rows 30.5, 46.0, 61.0, and 92.0 cm apart, respectively. Decline under fallow was 43.5%, signif- which peaked 7 weeks AE. There was no effect of soil depth on population decline at any sampling position. Decreasing row spacing resulted in 79.9, 74.2, 73.4, and 66.1% GN population decline for rows 30.5, 46.0, 61.0, and 92.0 cm apart, respectively. Decline under fallow was 43.5%, significantly less than under potatoes. Potato root weight between rows was negatively correlated with row spacing and positively correlated with GN population decline.  相似文献

The occurrence ofchlamydospores of Glomus fasciculatum (Gf) within cysts of the soybean cyst nematode, Heterodera glycines, and the effects of vesicular-arbuscular mycorrhizae on nematode population dynamics and soybean (Glycine max) plant growth were investigated. Chlamydospores occupied 1-24% of cysts recovered from field soil samples. Hyphae of Missouri isolate Gfl penetrated the female nematode cuticle shortly after she ruptured the root epidermis. Convoluted hyphae filled infected eggs, and sporogenesis occurred within infected eggs. G. microcarpum, G. mosseae, and two isolates of Gf were inoculated with H. glycines on plants of ''Essex'' soybeans. Each of the two Gf isolates infected about 1% of the nematode eggs in experimental pot cuhures. The Gfl isolate decreased the number of first-generation adult females 26%, compared with the nonmycorrhizal control. The total numbers of first-generation plus second-generation adult females were similar for both Gf isolates and 29-41% greater than the nonmycorrhizal control. Soybean plants with Gf and H. glycines produced more biomass than did nonmycorrhizal plants with nematodes, but only Gfl delayed leaf senescence.  相似文献

Low concentrations of ruthenium red and lanthanum chloride inhibited hatching of juveniles from eggs in cysts of Globodera rostochiensis in potato root diffusate. Pro-bit analysis for 31 cysts at seven concentrations of ruthenium red showed that 50% inhibition with 95% fiducial limits occured at 47 ± 23 μm; a similar value of 59 ± 14 μm was obtained using eggs removed from cysts. Results for 10 to 20 cysts at six concentrations of lanthanum chloride suggested a somewhat higher value for 50% inhibition of 110 ± 83 μM. In contrast hatching of eggs in cysts of Heterodera schachtii in water was unaffected by 5 ITIM lanthanum chloride and 625 μM ruthenium red, concentrations which cause over 90% inhibition of hatch in G. rostochiensis.
Two calcium ionophores synergised hatching of a 1971 population of G. rostochiensis in dilute diffusate. Optimal concentrations of 2 μM for A23187 and 10 μM for BrX537A increased the hatch from 17 ± 3–6 juveniles/cyst to 114 ± 44 juveniles/cyst and 138 ± 40 juveniles/cyst respectively. Ionophores in the absence of diffusate hatched very few eggs of this population but caused a greater hatch in a second (1975) population which gave a high hatch in water of 43 ± 10 juveniles/cyst. This was increased by A23187 to 181 ± 41 juveniles/cyst. The results with both the inhibitors and the ionophores suggest that hatching in G. rostochiensis might be a calcium-mediated process.  相似文献

Changes in the numbers of all stages of Heterodera rostochiensis in plants and soil were studied in a potato crop growing on infected land. Hatching and invasion occurred early in the season, when plants were very young and temperatures were below those considered necessary for hatching and invasion in Long Island, U.S.A. More larvae hatched from cysts nearer than away from potato roots. Males were abundant during June and July, when fertilization probably occurs. Many new cysts appeared during late June and early July. In May/June, there were many larvae in the roots of young plants but later there were few; a slight increase during August probably indicated a small second generation. The larvae for this second invasion may have come from the most recently formed cysts, which may have been stimulated to hatch either by root exudates or by substances from other soil organisms.  相似文献

The potential of several chromatographic methods for isolating hatching factors for potato cyst nematodes from potato root diffusate was investigated using a bioassay based on emergence of juveniles from cysts. Gel filtration provided an overall estimate of molecular weight of 437 Da for the hatching activity and ion exchange chromatography indicated that at least 60% of the recovered activity was anionic in nature. Material less polar than the hatching activity could be removed by passing potato root diffusate through a reversed-phase Sep-Pak C18 cartridge and the elutant showed 83.3 ± 4.4% (mean of 32 cysts) of the initial hatching activity. High performance liquid chromatography (HPLC) with a reversed phase, C18 column and gradient elution (0–80% CH3OH in water) confirmed that much of the hatching activity was polar and that it was not retained by this method of separation. A weak anion exchange resin achieved slight retention of much of the hatching activity and an ion pairing reagent lowered the polarity sufficiently to allow some retention in subsequent reversed phase HPLC on a CIS column. Both ion exchange and ion pairing HPLC suggested that hatching activity was not chromatographed as a single compound and indicated that fractions able to influence the nucleolus of the nucleus within the dorsal pharyngeal gland cell did not always show hatching activity.  相似文献
