Eight endoparasitic nematode species were recovered from 170 maize root samples in western Transvaal, Republic of South Africa. Pratylenchus zeae had the highest average population density (17,454/5 g roots), followed by P. neglectus (5,827/5 g roots), P. penetrans (5,617/5 g roots), P. brachyurus (3,060/5 g roots), Meloidogyne incognita plus M. javanica (301 juveniles/5 g roots), P. crenatus (130/5 g roots), and Rotylenchutus parvus (64/5 g roots). The 17 reasonably homogeneous farming areas (RHFA) surveyed could be ranked on the basis of the incidence of the prevalent nematode species. A positive relationship was found between the incidence of P. brachyurus and R. parvus and long-term average annual rainfall. The incidence of P. penetrans and the Meloidogyne spp. was positively related to a combination of sand percentage and long-term average annual rainfall.  相似文献

The genus Phoenix (Arecaceae) comprises 14 species distributed from Cape Verde Islands to SE Asia. It includes the economically important species Phoenix dactylifera. The paucity of differential morphological and anatomical useful characters, and interspecific hybridization, make identification of Phoenix species difficult. In this context, the development of reliable DNA markers for species and hybrid identification would be of great utility. Previous studies identified a 12 bp polymorphic chloroplast minisatellite in the trnG (GCC)-trnfM (CAU) spacer, and showed its potential for species identification in Phoenix. In this work, in order to develop an efficient DNA barcode marker for Phoenix, a longer cpDNA region (700 bp) comprising the mentioned minisatellite, and located between the psbZ and trnfM (CAU) genes, was sequenced. One hundred and thirty-six individuals, representing all Phoenix species except P. andamanensis,were analysed. The minisatellite showed 2-7 repetitions of the 12 bp motif, with 1-3 out of seven haplotypes per species. Phoenix reclinata and P. canariensis had species-specific haplotypes. Additional polymorphisms were found in the flanking regions of the minisatellite, including substitutions, indels and homopolymers. All this information allowed us to identify unambiguously eight out of the 13 species, and overall 80% of the individuals sampled. Phoenix rupicola and P. theophrasti had the same haplotype, and so had P. atlantica, P. dactylifera, and P. sylvestris (the “date palm complex” sensu Pintaud et al. 2013). For these species, additional molecular markers will be required for their unambiguous identification. The psbZ-trnfM (CAU) region therefore could be considered as a good basis for the establishment of a DNA barcoding system in Phoenix, and is potentially useful for the identification of the female parent in Phoenix hybrids.  相似文献

This study investigated freshwater fish for their current infection status with metacercariae of Clonorchis sinensis in Korea. Twenty-one species of freshwater fish (n = 677) were collected from 34 regions nationwidely from February 2007 to June 2008. They were individually examined by digestion technique. Eight species of freshwater fish from 17 different regions were recognized positive for the metacercariae of C. sinensis. The positive rates (range of metacercariae number per fish) of fish by the species were as follows: 48% (1-1,142) in Pseudorasbora parva, 60% (1-412) in Pungtungia herzi, 15.7% (1-23) in Pseudogobio esocinus, 29% (1-7) in Acheilognathus intermedia, 21% (1-4) in Odontobutis interrupta, 33% (1-6) in Zacco temmincki, 3.6% (1-4) in Zacco platypus, and 26.3% (1) in Hemibarbus labeo. The two species, P. parva and P. herzi, are able to be the index fish for estimation of C. sinensis transmission in a certain locality. Still several species of freshwater fish are briskly transmitting C. sinensis infection in many riverside areas of southern Korea.  相似文献

Freshwater fish from Vientiane Municipality and Savannakhet Province, Lao PDR were examined by the muscle compression and artificial digestion methods to know the infection status with trematode metacercariae. In the fish from Savannakhet, 2 species of metacercariae, Opisthorchis viverrini and Haplorchis taichui, were detected. O. viverrini metacercariae were found in 6 species of fish, Puntius brevis, Hampala dispar, Esomus metallicus, Mystacoleucus marginatus, Puntioplites falcifer, and Cyclocheilichthys armatus. H. taichui metacercariae were detected in 3 species of fish, P. brevis, P. falcifer, and M. marginatus. In the fish from Vientiane, 4 species of metacercariae, O. viverrini, H. taichui, Haplorchis yokogawai, and Centrocestus formosanus, were detected. Among them, O. viverrini metacercariae were found in 7 species of fish, Onychostoma elongatum, C. armatus, H. dispar, P. brevis, Cyclocheilichthys repasson, Osteochilus hasseltii, and Hypsibarbus lagleri. The metacercariae of H. taichui were detected in 6 species of fish, C. repasson, O. elongatum, C. armatus, H. dispar, Labiobarbus leptocheila, and Cirrhinus molitorella. The metacercariae of H. yokogawai were found in 9 species of fish, C. repasson, O. elongatum, C. armatus, H. dispar, Labiobarbus leptocheila, O. hasseltii. C. molitorella, Hypsibarbus wetmorei, and H. lagleri. The metacercariae of C. formosanus were detected in 4 species of fish, C. repasson, P. brevis, O. hasseltii, and C. molitorella. From these results, it is confirmed that fishborne trematode metacercariae, i.e. O. viverrini, H. taichui, H. yokogawai and C. formosanus, are prevalent in various species of freshwater fish from Savannakhet Province and Vientiane Municipality, Lao PDR.  相似文献

A new species of cyst nematode, Globodera ellingtonae, is described from soil collected from a field in Oregon. Second-stage juveniles (J2) of the species are characterized by body length of 365-515 μm, stylet length of 19-22.5 μm, basal knobs rounded posteriorly and pointed anteriorly, tail 39-55 μm, hyaline tail terminus 20-32.5 μm, and tail tapering uniformly but abruptly narrowing and constricted near the posterior third of the hyaline portion, ending with a peg-like, finely rounded to pointed terminus. Cysts are spherical to sub-spherical, dark to light brown and circumfenestrate and cyst wall pattern is ridge-like with heavy punctations. Males have a stylet length of 21-25 μm and spicule length of 30-37 μm with a pointed thorn-like tip. Females have a stylet length of 20-22.5 μm, one head annule and labial disc, heavy punctations on the cuticle, and short vulval slit 7.5-8 μm long. Morphologically this new, round-cyst species differs from the related species G. pallida, G. rostochiensis, G. tabacum complex and G. mexicana by its distinctive J2 tail, and by one or another of the following: shorter mean stylet length in J2, females and males; number of refractive bodies in the hyaline tail terminus of J2; cyst morphology including Granek’s ratio; number of cuticular ridges between the anus and vulva; and in the shape and length of spicules in males. Its relationship to these closely related species are discussed. Based upon analysis of ribosomal internal transcribed spacer (ITS) sequences, G. ellingtonae n. sp. is distinct from G. pallida, G. rostochiensis, G. tabacum and G. mexicana. Bayesian and Maximum Parsimony analysis of cloned ITS rRNA gene sequences indicated three clades, with intraspecific variability as high as 2.8%. In silico analysis revealed ITS restriction fragment length polymorphisms for enzymes Bsh 1236I, Hinf I, and Rsa I that overlap patterns for other Globodera species.  相似文献

An identification key to 63 species of Pratylenchus is given. A compendium of the most diagnostic characters to be used directly in identification of species is included as a practical alternative and supplement to the key. P. tenuis, P. similis, P. impar, P. ranjani, and P. neocapitatus are recognized as valid species on the basis of study of type specimens. P. hyderabadensis Singh &Gill, 1986 is synonymized with P. dasi Fortuner, 1985. P. hexincisus Taylor &Jenkins, 1957 is confirmed as occasionally having 4 -6 lines in lateral field (instead of 6 only). Comments on the status of some species and a list of species of the genus are given.  相似文献

The ant genus Paratrechina is reexamined based on the discovery of two new species from Madagascar (P. ankarana sp. n. and P. antsingy sp. n.). Paratrechina kohli, a species known from central Africa, is transferred to Paratrechina from Prenolepis based on a new morphological interpretation of the genus and an updated morphological diagnosis of the genus is provided. This means that other than the widespread P. longicornis, whose origins remain uncertain, all Paratrechina are restricted either to the Afrotropical or Malagasy regions. It would also appear that of the five Paratrechina species now known, three are from dry forest habitats. With this reexamination of the genus, the possible origins of P. longicornis are discussed. A key to the genera of the Prenolepis genus-group is provided, as is a key to the workers of Paratrechina. In addition, we also designate a lectotype for Paratrechina kohli.  相似文献

Alfalfa is a host of Pratylenchus penetrans and P. neglectus, whereas crested wheatgrass is a host of P. neglectus but not of P. penetrans. In a 120-day greenhouse experiment at 24 ñ 3 C, P. neglectus inhibited the growth of ''Lahontan'' alfalfa and ''Fairway'' crested wheatgrass. There were no differences in persistence and plant growth of alfalfa and crested wheatgrass, or reproduction of P. neglectus, in single plantings of alfalfa (AO) or crested wheatgrass (CWO), or in interplanted alfalfa and crested wheatgrass (ACW) treatments. On alfalfa, P. penetrans inhibited growth and reproduced more than did P. neglectus. Inhibition of plant growth and reproduction of P. penetrans was greater on alfalfa in AO than in ACW treatments. Pratylenchus penetrans did not reproduce on crested wheatgrass, but inhibited growth of crested wheatgrass in interplanted treatments and was avirulent in single planted treatments. Results were similar in a controlled growth chamber experiment at 15, 20, 25, and 30 C. Both nematode species inhibited alfalfa growth at all temperatures, and P. penetrans was more virulent than was P. neglectus to alfalfa at all temperatures and treatments. Plant growth inhibition and reproduction of P. penetrans on alfalfa in single and interplanted treatments were similar at 15-20 C, but were greater in single than in interplanted treatments at 25-30 C. Pratylenchus penetrans was avirulent to crested wheatgrass in the single planted treatments at all temperatures, but inhibited growth of crested wheatgrass in interplanted treatments at 20-30 C. Plant growth and reproduction of P. neglectus on crested wheatgrass was similar in single and interplanted treatments at 20-30 C and 15-30 C, respectively.  相似文献

Five grape rootstocks were inoculated with 0, 100, 1,000, and 10,000 Pratylenchus vulnus. Dogridge and Saltcreek supported low average total numbers of P. vulnus, 136-705/pot, at 12 months after inoculation. Growth of both rootstocks was not affected. Harmony, Couderc 1613, and Ganzin 1 supported high average total numbers, 6-856 times the inoculum levels. Numbers in Harmony continued to increase at all levels but reduced root weight only at the 10,000 level after 12 months. Numbers in Couderc 1613 decreased by 15-30% after 12 months, and root weight was reduced at the 10,000 level. In Ganzin 1, total nematode numbers diminished after 12 months but were still at high levels; growth reduction was proportional to numbers of nematodes added. Meloidogyne incognita, M. javanica, and M. arenaria produced galls and egg masses in Harmony and Couderc 1613 only at 36 C. Galling in Ganzin 1 increased with increasing temperature. Galls in Ganzin 1 at 18 C supported mature females after 90 days. Harmony was resistant to M. incognita in single and concomitant inoculations of P. vulnus and M. incognita. At 250 days after inoculation, total numbers of P. vulnus increased above the inoculum level and the 150-day values; increase was greatest in P. vulnus added singly. Malaria, a disease affecting humans and other animals, is caused by a protist of the genus Plasmodium. At the intraerythrocytic stage, the parasite synthesizes a high amount of phospholipids through a bewildering number of pathways. In the human Plasmodium falciparum species, a plant-like pathway that relies on serine decarboxylase and phosphoethanolamine N-methyltransferase activities diverts host serine to provide additional phosphatidylcholine and phosphatidylethanolamine to the parasite. This feature of parasitic dependence toward its host was investigated in other Plasmodium species. In silico analyses led to the identification of phosphoethanolamine N-methyltransferase gene orthologs in primate and bird parasite genomes. However, the gene was not detected in the rodent P. berghei, P. yoelii, and P. chabaudi species. Biochemical experiments with labeled choline, ethanolamine, and serine showed marked differences in biosynthetic pathways when comparing rodent P. berghei and P. vinckei, and human P. falciparum species. Notably, in both rodent parasites, ethanolamine and serine were not significantly incorporated into phosphatidylcholine, indicating the absence of phosphoethanolamine N-methyltransferase activity. To our knowledge, this is the first study to highlight a crucial difference in phospholipid metabolism between Plasmodium species. The findings should facilitate efforts to develop more rational approaches to identify and evaluate new targets for antimalarial therapy.  相似文献

Damage and reproductive potentials of Pratylenchus brachyurus and P. penetrans on soybean, Glycine max, cvs. Essex, Forrest, and Lee 68, were determined in microplot tests. Cultivar Essex was generally tolerant to P. brachyurus. Yield of Forrest was suppressed linearly with increasing Pi''s in the sandy soil (r = -0.92) and loamy sand soil (r = -0.99). Low to moderate Pi''s in the sandy clay loam gave an increase in yields as compared to plants without nematodes. Yield was not affected by this nematode in muck. Lee 68 was very sensitive to P. penetrans in microplots. Yield vs. Pi was fitted by a quadratic model (r = 0.82) with yield decreasing sharply as Pi''s were increased. The reproduction of both species decreased with increases in Pi. Lee 68 was a good host for P. penetrans, whereas Essex and Forrest were fair to poor hosts for P. brachyurus.  相似文献

Paragonimiasis is a food-borne parasitic zoonosis caused by infection with lung flukes of the genus Paragonimus. In Vietnam, research on Paragonimus and paragonimiasis has been conducted in northern and central regions of the country. Using a combination of morphological and molecular methods, 7 Paragonimus species, namely P. heterotremus, P. westermani, P. skrjabini, P. vietnamensis, P. proliferus, P. bangkokenis and P. harinasutai, have been identified in Vietnam. Of these, the first 3, P. heterotremus, P. westermani and P. skrjabini, are known to infect humans in other countries. However, in Vietnam, only P. heterotremus, found in some northern provinces, has been shown to infect humans. Even nowadays, local people in some northern provinces, such as Lai Chau and Yen Bai, are still suffering from P. heterotremus infection. In some provinces of central Vietnam, the prevalence and infection intensity of P. westermani metacercariae in freshwater crabs (the second intermediate hosts) are extremely high, but human cases have not been reported. Likewise, although P. skrjabini was found in Thanh Hoa Province, its pathogenicity to humans in Vietnam still remains uncertain. The results of molecular phylogenetic analyses of Vietnamese Paragonimus species provides new insights on the phylogeny and taxonomy of the genus Paragonimus. Comprehensive molecular epidemiological and geobiological studies on the genus in Vietnam and adjacent countries are needed to clarify the biodiversity and public health significance of the lung flukes.  相似文献

Pratylenchoides hispaniensis n. sp. is described and illustrated from a bisexual population found in a natural habitat at Santa Elena, Jaen, central Spain. Its main distinctive characters are very long esophageal gland lobe (81-117 μm; N'' = 51-71) overlapping the intestine 3 to 5 times the body width; lateral field with six incisures; stylet knobs sloping posteriorly; labial disc encircled by the irregular sectors of the first annule; tail cylindrical, extremity annulated, and frequently with a slight dorsal indentation of the hyaline portion at the end of the lateral field. Pratylcnchoides hispaniensis n. sp. appears closely related to P. megalobatus and P. nevadensis. It differs from the former primarily by its longer body length (761-998 vs. 430-621 μm), longer stylet length (20.5-24.4 vs. 18-21 μm), six incisures in the lateral field vs. four for P. megalobatus, and posteriorly sloping stylet knobs vs. rounded or anteriorly flattened knobs in P. megalobatus. It differs from P. nevadensis mainly by the shape of the stylet knobs (sloping in P. hispaniensis vs. rounded in P. nevadensis), length of esophageal lobe (81-117 vs. 34-82 μm), and position of esophageal gland nuclei (all posterior to esophago-intestinal junction in P. hispaniensis vs. at least one nucleus anterior to junction in P. nevadensis).  相似文献

Pasteuria penetrans is an endospore-forming bacterial parasite of Meloidogyne spp. This organism is among the most promising agents for the biological control of root-knot nematodes. In order to establish the phylogenetic position of this species relative to other endospore-forming bacteria, the 16S ribosomal genes from two isolates of P. penetrans, P-20, which preferentially infects M. arenaria race 1, and P-100, which preferentially infects M. incognita and M. javanica, were PCR-amplified from a purified endospore extraction. Universal primers for the 16S rRNA gene were used to amplify DNA which was cloned, and a nucleotide sequence was obtained for 92% of the gene (1,390 base pairs) encoding the 16S rDNA from each isolate. Comparison of both isolates showed identical sequences that were compared to 16S rDNA sequences of 30 other endospore-forming bacteria obtained from GenBank. Parsimony analyses indicated that P. penetrans is a species within a clade that includes Alicyclobacillus acidocaldarius, A. cycloheptanicus, Sulfobacillus sp., Bacillus tusciae, B. schlegelii, and P. ramosa. Its closest neighbor is P. ramosa, a parasite of Daphnia spp. (water fleas). This study provided a genomic basis for the relationship of species assigned to the genus Pasteuria, and for comparison of species that are parasites of different phytopathogenic nematodes.  相似文献

The reproductive potential and damage functions for Meloidogyne hapla and M. arenaria race 1 on Virginia-type peanuts (Arachis hypogaea cv. Florigiant) were determined over 2 years in microplot experiments in North Carolina. Peanut yield suppression and damage to pods as a result of galling were greatest in response to M. arenaria (P = 0.01). Damage functions for the two species were adequately described by the quadratic models: yield (g/plot) = 398 - 17.1 (log₁₀[Pi + 1]) - 17.0(log₁₀[Pi + 1])²; (R² = 0.83, P = 0.0001) for M. arenaria; and yield = 388 - 10.2(log₁₀[Pi + 1]) - 7.5(log₁₀[Pi + 1])², (R² = 0.30, P = 0.0001) for M. hapla. Both species caused galling on pods, but this was more severe in response to M. arenaria. Reproduction of M. arenaria race 1 was greater than M. hapla on peanut, which accounts in part for the more severe pod galling. Peanut was an excellent host for both M. arenaria race 1 and for M. hapla, but reproduction by M. hapla was more variable.  相似文献

Evolutionary relationships based on ribosomal DNA (rDNA) sequence data for a previously unknown species of Globodera from Portugal, Punctodera chalcoensis from Mexico, and P. punctata from Estonia, plus previously published sequences, support the following relationships: (((Cactodera weissi, G. artemisiae, C. milleri), ((G. sp. Bouro, G. sp. Canha, G. sp. Ladoeiro), ((G. pallida, G. rostochiensis), (P. chalcoensis, P. punctata)))), Heterodera avenae). Globodera sp. from Portugal, which can be confused with potato cyst nematodes by phytosanitary services when the identification is based only on morphological characters, is clearly different based on our molecular data. In addition, the rDNA data show the Globodera sp. to be only distantly related to other European Globodera species that parasitize Asteraceae. Punctodera chalcoensis and P. punctata form a sister clade to the G. pallida + G. rostochiensis clade.  相似文献

Background and Aims

In some lupin species, phosphate deficiency induces cluster-root formation, which enhances P uptake by increasing root surface area and, more importantly, the release of root exudates which enhances P availability.

Methods

Three species of Lupinus, L. albus, L. atlanticus and L. micranthus, with inherently different relative growth rates were cultivated under hydroponics in a greenhouse at four phosphate concentrations (1, 10, 50 and 150 µm) to compare the role of internal P in regulating cluster-root formation.

Key Results

The highest growth rate was observed in L. atlanticus, followed by L. albus and L. micranthus. At 1 µm P, cluster-root formation was markedly induced in all three species. The highest P uptake and accumulation was observed in L. micranthus, followed by L. atlanticus and then L. albus. Inhibition of cluster-root formation was severe at 10 µm P in L. atlanticus, but occurred stepwise with increasing P concentration in the root medium in L. albus.

Conclusions

In L. atlanticus and L. albus cluster-root formation was suppressed by P treatments above 10 µm, indicating a P-inducible regulating system for cluster-root formation, as expected. By contrast, production of cluster roots in L. micranthus, in spite of a high internal P concentration, indicated a lower sensitivity to P status, which allowed P-toxicity symptoms to develop.  相似文献

Six new species are described, raising the number of North American Acomoptera species to seven and the genus total to ten, and nearly doubling the number of species within the putative clade containing Acomoptera, Drepanocercus, and Paratinia. These novel species forms have implications for the concept of Acomoptera that in turn, may impact our understanding of its generic relationships and the evolution and composition of Gnoristinae and Sciophilinae. The new species, Acomoptera crispa, Acomoptera digitata, Acomoptera echinosa, Acomoptera forculata, Acomoptera nelsoni,and Acomoptera vockerothi, are compared with the type species of the genus, Acomoptera plexipus (Garrett), whose diagnostic features are imaged and illustrated for the first time. The European species, Acomoptera difficilis (Dziedzicki) is also illustrated and compared. Acomoptera spinistyla (Søli) comb. n. is transferred from Drepanocercus. A key to species is provided. Future work will seek to incorporate this knowledge into a systematic phylogenetic study of relationships between these species and their sister taxa.  相似文献

Two new monosexual and one bisexual species Pratylenchus Filipjev, 1936 collected from Haryana state of India are described and illustrated. The primary distinguishing features of these species are Pratylenchus microstylus n. sp.: L = 331-458 μm, spear = 11 or 12 μm; Pratylenchus cruciferus n. sp.: L = 648-793 μm, central core of lateral fields with oblique lines, hemizonid 2-8 annules anterior to excretory pore; Pratylenchus ekrami n. sp.: spear = 11-13 μm, spermatheca oblong, post vulval uterine sac with differentiated cells, tail with 26-40 annules, males abundant. Studies on intraspecific variations of P. cruciferus, P. ekrami, and P. coffeae (Zimmermann, 1898) Goodey, 1951 revealed that spear length and value of ''V'' are the least variable characters. Body length and size of post vulval uterine sac varies to varying degrees in different species. Shape of median bulb in P. ekrami, number of incisures in P. coffeae, and tail shape in P. ekrami and P. coffeae exhibit the greatest amount of intraspecific variations. P. zeae Graham, 1936 and P. thornei Sher & Allen, 1953 are the other species collected during the present studies.  相似文献
