Expression of human hepatic genes in somatic cell hybrids   总被引:4,自引:0,他引:4  
Four diploid human cell types (lymphocytes, fibroblasts, amniotic fluid cells, and hepatocytes) were fused to mouse hepatoma cells, HH. HH synthesized and secreted several liver-specific gene products including albumin, transferrin, and alpha-fetoprotein. The resulting interspecific hybrids were compared to determine whether or not the pattern of human hepatic gene expression was similar when these various cells were fused with the mouse hepatoma line. The expression of six human hepatic genes was examined, including albumin, alpha-fetoprotein, ceruloplasmin, transferrin, alpha-1-antitrypsin, and haptoglobin. Albumin was most frequently expressed while alpha-fetoprotein was not detected in any of the hybrids studied. The patterns of expression of human serum proteins differed between the hybrid series. Hybrids derived from human fibroblasts produced primarily albumin, while those derived from lymphoblastoid cells and amniocytes had a higher frequency of clones secreting alpha-1-antitrypsin. The findings reported here suggest that the frequency of hybrid clones expressing human hepatic gene products and the array of proteins produced are influenced by the histogenetic state of the human parental cell type.  相似文献

The dephosphorylation of membrane proteins by an endogenous phosphatase has been studied both in A9 and TLX5 cells and in hybrids between them. These cells differ both in growth rate and saturation density achieved in vitro. The activity of the phosphatase seems to parallel the growth rate of these cell lines. It is considered that this phosphatase is part of an ATPase enzyme system. The enzyme from TLX5 cells is stimulated by cyclic adenosine 3′5′-monophosphate (cAMP) and inhibited by zinc and fluoride ions. Prostaglandin E1 (PGE1) has been found to have no effect on the activity of the phosphatase.  相似文献

Hybrids have been generated between mouse hepatoma cells, which actively synthesize alpha-fetoprotein (AFP), and adult hepatocytes, where AFP production is shut off. These hybrids maintain an active synthesis of mouse AFP. Using a specific radioimmunoassay, we found that rat AFP production is not activated. Southern blot analysis showed that mouse and rat AFP DNA sequences can be distinguished and that hybrid clones possessing something close to the complete chromosome sets of both parents have retained both parental AFP DNA sequences. Thus expressed and non-expressed AFP genes coexist in these hybrid cells as if their expression were dependent on a cis-acting event.  相似文献

Unidirectional loss of human chromosomes in rat-human hybrids   总被引:11,自引:0,他引:11  
Karyological analysis of 25 rat-human hybrid cell clones shows that only the human chromosomes are lost from the hybrids. Giemsa banding staining of the chromosomes of these hybrids allowed the identification of each human chromosome present in the hybrid cells.  相似文献

The immune response to the liver protein F antigen which, in the mouse, occurs in two allelic forms, is under sharp immunogenetic control in that only mice that possess the Ak molecule can respond to allo-F antigen. This response has been studied in a number of F1 hybrids between inbred strains and with recombinant inbred lines all of which express Ak, and which thus enable immune suppression effects to be detected. In the AKXL and AKXD sets, the hybrids with CBA are responders if H-2 k/H-2k, and usually nonresponders if H-2 k/H-2b or H-2 k/H-2d. Although this may be due to gene dosage effects, this cannot be the explanation for the low responsiveness of the H-2 k/H-2b relative to the H-2 k/H-2d mice found in CBA × BXD hybrids. For this, and other reasons, it seems likely that low responsiveness in any mouse possessing a responder A k allele is due to suppression, and that this is mediated by the immune suppression effects of the non-H-2 k haplotype. These H-2-mediated effects can be modified, both positively and negatively, by background genes. Thus, of the ten H-2k/H-2d members of the CBA × AKXD cross, seven are low responders and three are high responders. No other typed marker has the same strain distribution pattern at present. Major unresolved questions, therefore, concern the location and mechanism of action of the background genes and the mechanism of action of the H-2 immune suppression genes.  相似文献

Nucleolus organizer regions (NOR) are actively expressed on both sets of parental chromosomes in mouse-Chines hamster hybrid cells retaining 85–95% of each parental complement. A hamster NOR, which is active in primary fetal hamster cells, but suppressed in the established AK412 line, is reactivated in these hybrids, demonstrating control of NOR expression.  相似文献

The well known and most important function of nucleoli is ribosome biogenesis. However, the nucleolus showed delayed development and malfunction in somatic cell nuclear transfer (NT) embryos. Previous studies indicated that nearly half rRNA genes (rDNA) in somatic cells were inactive and not transcribed. We compared the rDNA methylation level, active nucleolar organizer region (NORs) numbers, nucleolar proteins (upstream binding factor (UBF), nucleophosmin (B23)) distribution, and nucleolar-related gene expression in three different donor cells and NT embryos. The results showed embryonic stem cells (ESCs) had the most active NORs and lowest rDNA methylation level (7.66 and 6.76%), whereas mouse embryonic fibroblasts (MEFs) were the opposite (4.70 and 22.57%). After the donor cells were injected into enucleated MII oocytes, cumulus cells and MEFs nuclei lost B23 and UBF signals in 20 min, whereas in ESC-NT embryos, B23 and UBF signals could still be detected at 60 min post-NT. The embryos derived from ESCs, cumulus cells, and MEFs showed the same trend in active NORs numbers (7.19 versus 6.68 versus 5.77, p < 0.05) and rDNA methylation levels (6.36 versus 9.67% versus 15.52%) at the 4-cell stage as that in donor cells. However, the MEF-NT embryos displayed low rRNA synthesis/processing potential at morula stage and had an obvious decrease in blastocyst developmental rate. The results presented clear evidences that the rDNA reprogramming efficiency in NT embryos was determined by the rDNA activity in donor cells from which they derived.  相似文献

After a recall of the importance of early basic developments of in vitro established cell lines for investigations on malignant transformation, a survey of essential steps in the study of malignancy by means of somatic cell hybridization is presented. Since the early sixties, in vitro crosses of malignant versus nonmalignant parental cells have provided many experimental models in which mechanisms of expression of malignancy have been approached. Allogenic as well as xenogenic cell matings resulted in tumor-producing or nontumorigenic hybrids which have been analyzed, particularly in terms of karyology in order to determine possible chromosomal patterns linked with inheritance of malignancy and its suppression. The authors discuss the successive concepts devised for interpretation of experimental data, implicating specific genetic "normalizing" information, genetic dosage as well as, more recently, epigenetic and cytoplasmic mechanisms.  相似文献

To obtain controlled genetic variation, colon cancer was chemically induced by use of four subcutaneous injections of azoxymethane (15 mg/kg of body weight/wk) to rats of 3 inbred strains (BDIX/OrlIco, F344/NHsd, WAG/Rij). The selection was based on the availability of established colon cancer cell lines arising from these particular strains. In the first experiment, only female rats were used; in the second experiment, both sexes were studied. The goal was to select a rat strain giving the highest tumor frequency with the shortest latency period in reproducible manner. The histologic characteristics should resemble the corresponding human tumors. The size of the tumors should be at about 1 cm in diameter, as these tumor cells were intended to be used in future transplantation studies. The two experiments yielded highly reproducible results: histologic evaluation of all colon tumors in all three rat strains revealed adenomas and adenocarcinomas closely resembling their human counterpart. The BDIX strain had the highest tumor frequency (75%) in both sexes and the shortest minimal latency period (28 weeks in experiment 1; 23 weeks in experiment 2). Tumor size of about 1 cm in diameter was found most often in the BDIX strain. On the basis of results of these two experiments, the BDIX strain has been selected for future study.  相似文献

The transformed phenotype is believed to be dominant in fusions between limited lifespan cells and transformed cells, based on heterokaryon experiments and on the isolation of transformed hybrids from mass cultures of fused cells. A series of fusions has been performed between limited lifespan Lesch-Nyhan fibroblast cells and a permanent HeLa cell line with a complementary genetic marker. The growth of independently isolated hybrid clones was followed in parallel with Lesch-Nyhan cells. In fusions involving Lesch-Nyhan cells which had completed about 50% of their lifespan, all hybrids initially show fibroblastic properties. Thirty-five hybrids had a limited lifespan slightly longer than Lesch-Nyhan controls. Three other hybrid clones, and all mass cultures of hybrids, showed the appearance of transformed colonies at a rate of approx. one transformant in 2 × 105 hybrid cells. These transformed cells showed anchorage independence, low serum requirement, chromosome loss, and have been maintained in culture for 50–100 population doublings with no signs of senescence. Fusions involving enucleated HeLa cells did not show transformation. Fusions with senescent Lesch-Nyhan cells yielded hybrids which grew beyond the normal Lesch-Nyhan cell lifespan, but which again showed limited lifespan and low frequency transformation. It is concluded that limited lifespan is expressed in a dominant manner in these fusions, and that transformation or “escape from senescence” is a low frequency event requiring the presence of the HeLa nucleus.  相似文献

The generation of somatic cell hybridization-derived cell lines between highly divergent species affords the opportunity to examine the concept of 'genome dominance' in the context of genetic and epigenetic changes. While whole-scale genome dominance has been well documented in natural hybrids among closely related species, an examination of centromere position and sequence retention in 2 marsupial-eutherian hybrids has revealed a mechanism for 'centromere dominance' as a driving force in the generation of stable somatic cell hybrids following an initial period of genomic instability. While one somatic cell hybrid cell line appeared to retain marsupial centromere sequences which remained competent to recruit the centromere-specific histone variant CENP-A in a Chinese hamster background, fusion events between marsupial and mouse-derived chromosomes in another hybrid line led to a centromere sequence conversion from one species to the other. We postulate that the necessity to maintain an epigenetically defined centromere following genome hybridization may be responsible for retention of specific chromosomes and may result in rapid sequence turnover to facilitate the recruitment of CENP-A containing histones.  相似文献

Summary We produced somatic cell hybrids between HT 1080-6TG human fibrosarcoma cells and either rat white blood cells (WBC) or cells directly derived from rat spleen. Karyologic and isozyme analyses of hybrid cells indicated that they preferentially lose rat chromosomes. Hypoxanthine-aminopterine thymidine-selected hybrid clones expressing rat hypoxanthine phosphoribosyltransferase (HPRT), glucose-6-phosphate dehydrogenase (G6PD), and phosphoglycerate kinase (PGK) and containing the rat X chromosome were counterselected in a medium containing 30 g/ml of 6-thioguanine. Concordant loss of the rat X chromosome and of the expression of rat HPRT and G6PD was observed in the hybrid clones.  相似文献

Nine inbred strains of the rat (Rattus norvegicus) were screened for differences in electrophoretically detectable proteins. Interstrain variation was observed for 7 of 26 proteins. Three of these variants have not been described previously: leucine aminopeptidase (Lap-1), major urinary protein (Mup-1), and seminal vesicle protein (Svp-2). Genetic analysis revealed two autosomal alleles for each of these polymorphisms. The loci Lap-1, Mup-1, and Svp-2 are linked neither to one another nor to the previously described Svp-1 and Es-4 loci. Each of the nine strains can be identified now by a specific set of monogenic markers.  相似文献
