Cyclic 3',5'-AMP phosphodiesterase of Neurospora crassa   总被引:13,自引:0,他引:13  
Cyclic 3′,5′-AMP (cAMP) phosphodiesterase activity can be demonstrated in extracts of Neurosporacrassa. The activity is particulate, has a pH optimum of 7.4, and consists of two forms that have different cAMP binding constants. Methylxanthines, inorganic phosphate, and EDTA are inhibitors of the diesterase as are ATP, ADP, and 8-bromo-cAMP. The enzymatic activity is stimulated by histamine and imidazole. These properties suggest that the Neurospora enzyme is more closely related to the mammalian than to bacterial cAMP phosphodiesterases.  相似文献

An enzyme system in the post mitochondrial fraction of Neurosporacrassa when supplemented with appropriate cofactors formed carnitine from ε-N-trimethyllysine. These findings together with previous studies of ε-N-lysine methylation in this fungi, illustrate that carnitine synthesis in Neurospora differs markedly in certain features from mammalian systems in that the entire synthesis is carried out employing free intermediates and cytosolic enzymes.  相似文献

Cell surface glycoproteins of Neurospora crassa conidia have been shown to bind amino acids and to be genetically associated with the previously defined amino acid transport systems of that organism. L-arginine does not readily permeate a film of Neurospora conidial lipids. Addition of glycoprotein extracts from Neurospora to the lipid film enhances permeation of arginine at an initial rate 1000 times the rate of permeation through lipid alone. The initial rate of passage exceeds the rate of unhindered passage (no lipid film) through the same cross sectional area by 10 fold.  相似文献

Neurosporacrassa was surveyed for cyclic AMP-dependent protein kinase activity. Two peaks (I and II) of protein kinase activity were demonstrated by DEAE-cellulose chromatography of wild type Neurospora extracts. Peak I was stimulated by cyclic AMP, eluted below 60 mM NaCl and had high activity using histone H2B as substrate. Peak II eluted at 200–250 mM NaCl; its activity was not cyclic AMP stimulated and was highest with dephosphorylated casein as a substrate. Cyclic AMP binding to a protein associated with the protein kinase is specifically inhibited by certain cyclic AMP analogs.  相似文献

The arom pentafunctional enzyme conjugate of Neurospora crassa was exposed to trypsin, chymotrypsin, or a protease preparation from Neurospora in the presence and absence of the first substrate, 3-deoxy-D-arabino-heptulosonate 7-phosphate. It was found that the first substrate coordinately protects all five activities from proteolytic inactivation, which indicates a conformational change induced by this compound. In addition, the data presented are consistent with the “domain” theory of conjugate structure. It is also argued that coordinate protection may be of physiological significance.  相似文献

Irradiating the mycelium of Neurospora crassa with blue light causes the photoreduction of a b-type cytochrome. The action spectrum for the photoreduction of the cytochrome b is similar to action spectra for the photoactivation of carotene synthesis and photoinhibition of the circadian rhythm of conidiation in Neurospora.  相似文献

The gel released from isolated rat liver nuclei in response to heparin treatment has been found to stain with methylene blue, azure A, and methyl green when the dyes were added to the salt-sucrose nuclear isolation medium.Azure A and methylene blue caused rapid nuclear shrinkage to as little as 14 the original nuclear volume. Subsequent treatment with heparin caused the nuclei to fade rapidly and swell to approximately 54 of the original volume. With methylene blue stained nuclei heparin caused the extrusion of deeply stained, slightly birefringent rods through apertures on the nuclear surface. Methyl green also caused nuclear shrinkage, but to a lesser degree.Studies with the Feulgen reaction demonstrated structural damage in isolated rat liver nuclei as a result of heparin action. The viscous material released by heparin was shown to be Feulgen positive by resort to hydrolysis without prior fixation, since after customary fixatives the presence of a Feulgen positive reaction outside the nucleus could not be clearly demonstrated. The possibility is suggested that the Feulgen reaction following the customary fixatives depends in part on the manner in which the DNA is bound.The nuclei of leucocytes with visually intact cell membranes included in the nuclear preparations failed to show structural damage due to heparin and it is suggested that either the cell membrane provides some protection against heparin action or that damaged cells are more susceptible to this action.Observations made provide additional basis for the conclusion that heparin replaces DNA in the nucleo-histone of the nucleus, resulting in the structural damage observed, and releasing DNA in the form of a soluble viscous protein containing complex.  相似文献

When hypothyroid rat liver nuclei labeled in vivo with [125 I]L-triiodothyronine are incubated with micrococcal nuclease, the nuclear chromatin is digested and chromatin particles are released into the medium. The nuclease-treated nuclei contain intact nucleoli and a residual chromatin fraction. When this residual chromatin is purified, it contains only a small percentage of the initial nuclear DNA but is strikingly enriched in [125 I]L-triiodothyronine. This chromatin fraction has many of the characteristics of nucleolar chromatin including a high protein to DNA ratio, an abundance of nonhistone proteins, and a relatively high RNA to DNA ratio. An association of thyroid hormone receptors with a nucleolar component implicates this organelle in the early events of thyroid hormone action.  相似文献

Nuclei prepared from confluent and mitotically arrested populations of human diploid fibroblast-like cells of different invitro ages were subjected to digestion by micrococcal nuclease and DNase I. There was no age or culture state variation in the susceptibility of DNA to micrococcal nuclease digestion. There was, however, an age related inhibition of DNA digestion by DNase I in nuclei from older confluent but not older arrested cells. It is suggested that this is the result of an age related masking by nucleosome core histones which limits the accessibility of DNA to enzymatic activities in older confluent cells.  相似文献

DNA replication in isolated nuclei from Concanavalin A-stimulated and resting bovine lymphocytes has been studied. Nuclei from S phase lymphocytes incorporate 4–7 times more (3H)dTTP than nuclei from resting cells. The DNA synthesis was dependent on ATP, Mg2+ and all four deoxynucleoside triphosphates and was linear for about 60 min. The newly synthesized DNA is nuclear and DNase-sensitive and is the product of discontinuous and semiconservative replication. After limited digestion with micrococcal nuclease the in vitro replicated DNA was found to occur in nucleosomes prior to joining of primary DNA pieces. Addition of a protein extract from replicating cells stimulated the DNA synthesizing capacity of nuclei from resting lymphocytes. A preliminary characterization of this extract is given.  相似文献

HeLa cell nuclei contain a protein which stimules the invitro activity of HeLa cell DNA polymerase β, but does not affect the activity of DNA polymerase α and γ. The protein, which binds to both single- and double-stranded DNA, does not possess nuclease activity and is heat stable, surviving 100 degrees C for 10 min. The molecular weight of the protein is approximately 85,000 and evidence is presented that it may exert its stimulatory effect by direct interaction with β-polymerase.  相似文献

An analysis of the repeat structure of the highly repetitive sequence, component α DNA of the African green monkey, shows that the DNA contains restriction sites for EcoRI, EcoRI1, HindIII and HaeIII. All four restriction enzyme activities indicate a basic repeat length of 176 ± 4 base-pairs. In addition to primary EcoRI1 and HindIII sites, about 59% of the repeat sequences contain secondary EcoRI1 sites and about 36% of the repeat sequences contain secondary HindIII sites. The secondary sites are located less than 176 base-pairs from the primary sites and their cleavage yields several complex series of minor, intermediate segments in gels of the partial EcoRI1 or HindIII digests. Cleavage at the secondary sites yields segments shorter than the unit monomer in the limit digests. The sites for EcoRI, EcoRI1, HindIII and HaeIII have been mapped within the repeat unit.Treatment of the monkey nuclei with micrococcal nuclease at 2 °C and in the presence of 80 mm-NaCl reveals two distinct populations of nucleosomes. One population contains bulk DNA sequences, and after cleavage with micrococcal nuclease this population yields heterogeneous segments of DNA spanning 180 to 200 base-pairs in length. The other population contains component α sequences and after cleavage with micrococcal nuclease yields homogeneous segments of component α DNA that are exact multiples of the basic sequence repeat unit of 176 base-pairs. Thus, the cleavage by micrococcal nuclease of nucleosomal arrays containing component α sequences is as regular and precise as the cleavage of the purified DNA by the restriction enzymes. The resolution of the two distinct subsets of nucleosomes in the monkey nuclei is dependent upon the conditions of ionic strength and temperature employed during the nuclear isolation and the micrococcal nuclease digestion.These observations are consistent with a phase relation between the component α repeat sequences and the associated nucleosomal proteins (Musich et al., 1977b). They are also in accord with the hypothesis that the subunit structure of constitutive heterochromatin modulates or determines the repeat sequence structure and hence, the evolution of many highly repetitive mammalian DNAs (Maio et al., 1977).  相似文献

In Neurospora crassa two enzymes can provide glutamate: the NADPH dependent GDH and the NADH dependent GOGAT. An elevated GOGAT activity was found in Neurospora wild-type under ammonium limitation in contrast to a 4-fold lower activity on excess of am monium. Glutamate and glutamine repress this enzyme. On excess of ammonium the GDH-NADPH deficient mutant am-1 grows poorly with an elevated GOGAT activity. A GOGAT less mutant was found. It presented a lag-phase to grow on ammonium. It is concluded that N. crassa glutamate synthase provides glutamate from low am-monium concentrations. The enzyme was purified to homogeneity and shown to be composed of a single type of monomer with a molecular weight above 200,000.  相似文献

Rat liver nuclear matrix has been isolated by salt extraction and nuclease digestion of nuclei. Under the electron microscope, the matrix appears as a spongelike network joined by thinner fibrils. Biochemical analysis shows a high protein content and low amounts of nucleic acid and phospholipid. Treatment of the matrix with phospholipase C results in a release of most of the nucleic acid, and a disappearance of the fibrils, however the appearance of the matrix is largely unaffected. It seems likely that phospholipids are responsible for the hydrophobic interactions between nucleic acids and matrix fibrils. From in vitro labelling studies the released DNA is more recently synthesised than the bulk material, however the matrix bound RNA appears to label less rapidly than total nuclear RNA.  相似文献

Stimulation of transduction in Escherichia coli by ultraviolet irradiation of the transducing phage P1 requires the uvrA-uvrB nuclease but not the uvrC product or DNA polymerase I. It is hypothesized that the first step in “normal” recombination can be bypassed by any procedure generating single-stranded ends of DNA (as, for example, by uvra-uvrB nuclease activity).  相似文献

The GDH (NADPH) mutant strain am-1 of N. crassa has sizable pools of glutamine and glutamate under ammonium-limited conditions for which requires an elevated glutamine synthetase activity. Glutamine in the pres ence of 2-oxoglutarate, stimulated nicotinamide nucleotide oxidation by crude and purified extracts of the am-1 strain and led to a reductant dependent formation of two molecules of glutamate. Aminooxyacetate did not have any effect on the reaction, whereas azaserine inhibited it completely. It is concluded that in N. crassa glutamine synthetase and glutamate synthase are responsible for the assimilation of low ammonium concentrations.  相似文献

The template activity of isolated rat liver nuclei for DNA synthesis assayed with E.coli DNA polymerase was found to be dependent upon the presence of Ca2+ or Mg2+ in the incubation medium. DNA was prepared from isolated nuclei subjected to conditions which activated the template and centrifuged in an alkaline sucrose gradient. The distribution profile showed that smaller fragments were formed, suggesting enhancement of endonucleolytic activity. When isolated nuclei were incubated with NAD to induce poly(adenosine diphosphate ribose) formation and were subjected to the activation conditions, the template for DNA synthesis remained unchanged. The distribution profile in an alkaline sucrose gradient of DNA prepared from these nuclei and control nuclei was identical. The present findings suggest that the template-activating system for DNA synthesis was blocked when isolated nuclei were treated with NAD invitro.  相似文献

The dinoflagellate Gyrodiniumcohnii has a distinct nuclear membrane but apparently lacks histones associated with its chromatin. Approximately 13% of the nuclear DNA is rapidly digested by micrococcal nuclease to acid soluble fragments and not to nucleosomal sized fragments as in the typical eukaryote. Moreover in the electron microscope the chromatin of G.cohnii appears as a thin filament of 40–60 Å in width without regularly spaced nucleosomes. These observations support the view that the dinoflagellates exhibit characteristics of both prokaryotes and eukaryotes.  相似文献

Metabolic sequestration of putrescine in Neurospora crassa   总被引:3,自引:0,他引:3  
The metabolic fate of putrescine labelled invivo was investigated after administration of a trace (10?7 M) of L-[14C]ornithine to exponentially growing mycelia of Neurosporacrassa, followed by a large chase (2 mM) of L-[12C]ornithine. The specific radioactivities of putrescine and spermidine were determined during the chase period by reaction with [3H]dansyl chloride of known specific radioactivity and isolation of the dansyl-derivatives by thin-layer chromatography. Radioactivity remained in the putrescine pool for over 2 h during the chase period. This suggests that putrescine is largely sequestered (80% or more) invivo. The metabolic sequestration of polyamines may be a significant factor in the regulation of polyamine synthesis.  相似文献

Digestion of Euglena nuclei or extracted chromatin with micrococcal nuclease results in the identification of a repeating structure. The DNA repeat size, analyzed on agarose and polyacrylamide gels, is found to be 225±13 base pairs. DNase I digestion produces a serie of fragments multiples of roughly 10 bases. Eventhough pressure shearing is necessary to disrupt the though pellicule of the phytoflagellate, we confirm that, in Euglena, chromatin organization is similar to that of other eukaryotes.  相似文献
