The lactate dehydrogenase isoenzyme pattern has been determined in different murine lymphocytic cell populations. In each cell population, the LDH activity was predominantly found in the LDH-4 and LDH-5 fractions. The percentage LDH-5 activity was significantly higher in B cells than in T cells. The same is true for lymphocytes from the spleen versus lymph node lymphocytes. The percentage LDH-5 activity is significantly higher in peripheral T lymphocytes than in thymocytes. Enrichment of the more mature thymocytes of the thymocyte cell pool by either cortisone treatment in vivo or gradient centrifugation on bovine serum albumin (BSA) results in a decrease of LDH-1 and LDH-2 fractions. In the cortisone-treated group, the shift in the LDH pattern is accompanied by a significant increase of LDH-5 and LDH-4 fractions, whereas in the BSA group only the LDH-4 fraction increases.  相似文献

Changes in the activity of serum lactate dehydrogenase (LDH) and the serum LDH isozyme pattern in healthy young men given a high carbohydrate diet (480-636 g/day, 80% of the total energy) for 21 days were examined. Serum total LDH activity showed no significant change in four healthy young volunteers who received high carbohydrate diet for 21 days. However, the percentage of LDH-4 increased significantly (P less than 0.01) from 8.5 +/- 2.4 to 10.9 +/- 1.9% after 21 days, as did also the percentage of LDH-5 (P less than 0.01) from 5.1 +/- 1.9 to 10.7 +/- 2.9%. The percentage of M-type LDH activity increased 30% during the experiment (P less than 0.05). It is concluded from the results that the high carbohydrate intake affects the percentage of LDH-4 and LDH-5, but not the total serum LDH activity.  相似文献

Beef liver mitochondrial fraction showed LDH activity (1.76 +/- 0.25 U/g pellet). Sixty seven% of the initial mitochondrial pellet LDH activity (almost M4 isoenzyme) was released when suspended in NaCl 0.15 M. When the washed particles were sonicated in a 0.15 M NaCl medium, the solubilized LDH activity (all five isoenzymes as cytosoluble fraction) was 5-fold higher than the initial pellet activity. The different isoenzymatic composition of intramitochondrial and externally bound forms of the enzyme should be taken into account when investigating the physiological role of intramitochondrial LDH. Beef liver cytosoluble LDH (very little content of M4 isoenzyme) showed no affinity for the beef liver mitochondrial fraction but purified M4-LDH isoenzyme was able to bind to the particulate fraction from the same source. This suggests an isoenzyme specificity for the interaction. The maximum amount of cytosoluble LDH bound to the mitochondrial fraction depends on the enzyme and the particulate fraction source. Therefore, binding capacity to the mitochondrial fraction depends not only on the net charge of LDH isoenzymes, which play a predominant role in the binding, but also on individual characteristics of the LDH isoenzymes and mitochondrial fractions from different sources. This suggests that electrostatic forces are not the only ones involved in the binding process.  相似文献

Summary Lactate dehydrogenase (LDH) isozyme patterns were analysed by polyacrylamide (PAA) slab gel electrophoresis in extracts prepared from various rabbit skeletal muscles of defined fibre composition and by PAA microelectrophoresis of microdissected, histochemically typed single muscle fibres. The results obtained by electrophoresis of whole muscle extracts generally agreed with the data obtained from single fibre electrophoresis, i.e. the LDH isozyme pattern corresponded to that of the predominant fibre type. Type I Fibres from soleus and semitendinosus muscles were characterized by a unique pattern of all 5 LDH isozymes with a predominance of LDH-1, 2 and 3. The major fraction (80%) of the type II fibres from extensor digitorum longus and tibialis anterior muscles contained only LDH-5 (M4). About 20% of the type II fibres contained in addition to LDH-5 small amounts of LDH-4 and LDH-3. The fraction of fibres containing LDH-5, LDH-4, and LDH-3 was similar (ca. 20%) in the histochemically defined IIA and IIB subpopulations In view of the fact that the major fractions of rabbit IIB fibres display low and of IIA fibres high aerobic oxidative capacities (Reichmann and Pette 1982), these data indicate that the expression of the H-subunit of LDH is not correlated with the aerobic-oxidative capacity of the fibre. Lactic dehydrogenase (LDH) and alpha-hydroxybutyrate dehydrogenase (HBD) and LDH isozyme patterns were studied in serum from newborns and patients with ovarian cancer or myocardial infarction. LDH and HBD activities from newborns and patients with ovarian cancer or myocardial infarction were significantly increased, compared with those from patients with benign ovarian tumor. These increases were accompanied with a decrease of LDH-H and an increase of LDH-M in serum from newborns and patients with ovarian cancer, while an increase of LDH-H in serum from patients with myocardial infarction was dominant. However, the raised HBD activities in serum from patients with benign ovarian tumor did not affect the LDH isozyme patterns. From analysis of linear regression, a negative correlation between LDH-1 or -2 and HBD activity in serum from patients with ovarian cancer was observed while there was a positive correlation between LDH-4 and HBD activity. Similar patterns in serum from newborns were observed. On the other hand, a positive correlation between LDH-1 and HBD activity and a negative correlation between LDH-4 and HBD activity were found in serum from patients with myocardial infarction.  相似文献

Uterine secretions were collected from 20 mature cows during estrus (day 0), metestrus (day 5), diestrus (day 10) and proestrus (day-1). Lactate dehydrogenase (LDH) and LDH isoenzymes activity were evaluated. No significant cyclic variations of LDH activity was found in the uterine secretions while the mean of the enzyme activity was higher during the estrogenic period of the cycle. The relative activity of LDH-1, LDH-2 and LDH-3 isoenzymes were higher during proestrus and estrus whereas LDH-5 activity was more important during metestrus. The LDH-3 seems to have the higher relative activity in uterine secretions of the cow.  相似文献

Lactate dehydrogenase (LDH) isoenzyme profiles in human platelets and the sera of patients with type I and II diabetes mellitus and vascular complications, as well as normal subjects were measured utilizing a recently established, modified micromethod. LDH-3 was the predominating species in platelets (37.5 +/- 3.0%), with LDH-2, 1, 4 and 5 following in decreasing order of concentration. The LDH-3/LDH-4 ratio in platelets varied from 6.2 to 1.38. Type I and type II diabetic patients with vascular complications showed a significantly higher ratio for LDH-3/LDH-4 (3.99 +/- 1.20 for DM I, 2.16 +/- 0.25 for DM II patients) than the mean ratio for normal subjects (1.14 +/- 0.08). This platelet-specific LDH isoenzyme pattern may be the result of frequent in vivo platelet-vessel wall interactions in the diabetic patients whose platelets are known to be hyperaggregable in in vitro test systems. Since non-diabetic patients patients with vascular complications also displayed a similarly elevated LDH-3/LDH-4 ratio, a wider classification is preferable, although the measurement of the LDH isoenzyme pattern will be helpful in assessing diabetic vascular complications.  相似文献

Summary Adult human testicular tissue contains up to six previously undescribed lactate dehydrogenase (LDH) isoenzymes in addition to the five LDH isoenzymes normally found and the sixth found in spermatogenic cells and spermatozoa, LDH-X. Additional LDH isoenzymes were also found in spermatozoa but not in seminal fluid or in serum. After electrophoresis one additional LDH isoenzyme of testicular tissue was localized between LDH-1 and LDH-2, two between LDH-2 and LDH-3, two between LDH-3 and LDH-4, and two between LDH-4 and LDH-5. These localizations indicate that the additional LDH isoenzymes are tetramers combining the A and B subunits of the five normal LDH isoenzymes and the C subunit of LDH-X. The additional LDH isoenzymes may be important in the metabolism of spermatogenic germ cells and spermatozoa.  相似文献

Adult Fasciola gigantica contained 4.49 ± 0.06 % (mean ± S.D.) wet weight glycogen. Tissue homogenates contained high levels of malate dehydrogenase (MDH), NAD-linked malic enzyme (ME), Phosphoenolpyruvate carboxykinase (PEPCK) and lactate dehydrogenase (LDH). MDH, PEPCK and ME activities appeared to be localized in both cytosolic and mitoehondrial fractions, fumarase activity appeared to be predominantly mitochondrial whereas LDH and pyruvate kinase activities were cytosolic in distribution. Polyacrylamide gel electrophoresis revealed the predominance of LDH-1, LDH-2 and LDH-3 but only traces of LDH-4 and LDH-5 isoenzymes in the crude cytosolic fraction. LDH activity in the crude sample was inhibited by excess substrate (pyruvate). The mitoehondrial system showed NADH -cytochrome c oxidoreductase, succinate-cytochrome c oxidoreductase, NADH oxidase and some cytochrome c-oxygen oxidoreductase activities. Under anaerobic conditions, NADH-fumarate oxidoreductase and succinate-NAD + oxidoreductase activities of mitoehondrial preparations were stimulated in the presence of ADP and ATP respectively. Isolated mitochondria contained rhodoquinone and no ubiquinone, and isolated rhodoquinone was readily reduced by succinate in the presence of submitochondrial particles. Hydrogen peroxide was produced by submitochondrial particles in the presence or absence of KCN or in the presence of fumarate.  相似文献

1. Effect of pH on liver lactate dehydrogenase (LDH) and its isozymes was examined in the goldfish acclimated to different temperatures and some purification of the LDH was attempted. 2. The optimal pH and the Km value at 30 degrees C of the enzyme were independent of acclimation temperature. 3. the optimal pH of isozyme was more basic in the order of LDH-1, LDH-2, LDH-3, LDH-4 and LDH-5. Km values of isozymes at 30 degrees C were higher in the order of LDH-1, LDH-3 and LDH-5. 4. There was no change in the enzyme activity during thermal acclimation.  相似文献

利用SOD和LDH同工酶电泳分析,研究大豆黄酮对衰老小鼠的抗氧化作用。结果显示大豆黄酮没有改变SOD和LDH同工酶谱的特征,但对因衰老引起的小鼠脑组织LDH和SOD同工酶活性、各组分的相对活性和比活力的变化有不同程度的改善作用,即LDH同工酶中LDH-2、LDH-3的活性明显下降,LDH-1的活性下降最为明显,而LDH-4的活性有所下降,但不显著,LDH-5的活性几乎没有变化,SOD同工酶的SOD-1和SOD-2的活性有不同程度的升高。这表明大豆黄酮是通过抑制LDH同工酶H亚基的合成来降低LDH的活性,而对M亚基的合成没有影响,并且能够促进SOD同工酶SOD-1和SOD-2的合成,不影响其遗传稳定性。  相似文献

An improved method is described for the isolation of isozymes 1 and 5 of lactate dehydrogenase (LDH) from heart and skeletal muscles of foxes. The method includes salt fractionation with ammonium sulphate, chromatography on DEAE- and CM-celluloses and affinity chromatography on AMP-Sepharose. The preparations of LDH isozymes 1 and 5 turned out to be homogeneous both in 7,5% polyacrylamide gel electrophoresis and under immunodiffusion analysis. It is shown that the pH optimum for LDH-1 is 10.2-10.4 for LDH-5 it is 9.5-9.6 in the case of the direct reaction, and the pH optimum is 7.6-7.8 for LDH-1 and 7.3-7.4 for LDH-5 in the case of reverse reaction. The values of Mikhaelis constants were determined for substrates and coenzymes in direct and reverse reactions. It is found that the excess of lactate and pyruvate causes substrate inhibition of both LDH-1 and LDH-5. The activities of LDH-1 and LDH-5 showed an unexpected similar sensitivity to the inhibitory effect of high pyruvate concentrations.  相似文献

A total of 65 cases of ovarian tumor admitted for surgical intervention were prospectively enrolled in this study, including 40 cases with benign ovarian tumors and 25 cases of ovarian cancer. The 25 malignancies were comprised of 22 cases of common epithelial origin and three cases with Krukenberg tumors. Serum total LDH level and its isozyme activity were measured preoperatively in each patient. The mean value (+/-SD of serum LDH in the malignant group was 876.3 (+/- 450.4) IU/1, which was significantly higher than 364.8 (+/- 87.9) IU/1 in the benign group. Twenty-two cases of ovarian malignancy (88%) and 6 cases of the benign group (15%) had elevated serum LDH levels above 450 IU/1. Analysis of the LDH isozyme pattern demonstrated that there was a significant shift to LDH-4 and LDH-5 fractions in ovarian malignancy when compared to the benign group (with the mean value of 9.5%, 14.6% vs. 6.5% and 4.6%, respectively). The elevated LDH level could yield 88% and 85% in sensitivity and specificity for detecting ovarian malignancy, while the abnormal isozyme pattern could yield 84% and 77.5%, respectively. From combined analysis of total LDH levels and isozyme patterns, the true positive detection rate and true negative detection rate could both reach 100%. In conclusion, the present findings imply that total serum LDH level and isozyme activity measurement may provide significant aids in evaluation and detection of human ovarian cancers among patients with ovarian tumors.  相似文献

The effects of four different cardiac hypertrophic stresses on cardiac lactate dehydrogenase (LDH) isozyme composition and activity were examined. Altitude-induced right ventricular hypertrophy was accompanied by an increase of 10% in the M subunit of LDH in right ventricle, left ventricle, and atria. Left ventricular hypertrophy induced by aortic stenosis also produced isozyme changes in the ventricles, but of only half the magnitude. Biventricular hypertrophy, induced by running or swimming, was accompanied by 4-5% increases in M LDH in the ventricles only. We conclude that changes in LDH activity are directly related to changes in the M subunit in all three portions of the heart. No changes in H subunit were noted under any of the stresses. It appears that the magnitude of changes in cardiac LDH isozyme composition are only marginally related to extent of hypertrophy.  相似文献

The pattern of lactate dehydrogenase isoenzyme spectrum changes on different stages of T-lymphocyte differentiation was studied An enriched population of stem cells has LDH-5, 4 and 3 isoenzymes, and much less LDH-2 activity. The isoenzyme pattern of thymic cell precursors consists of LDH-5, 4, 3 and 2. All the five LDH isoenzymes were found in cortical thymocytes. Medullary thymocytes reveal LDH-5, 4 and 3 isoenzymes. T-lymphocytes of peripheral lymphoid organs contain mainly LDH-5 and in a lesser degree LDH-4 activity.  相似文献

Summary Lactate dehydrogenase (LDH) and LDH isoenzyme activities were determined in tumor tissue, tumor cystic fluid, and serum from athymic mice with transplants of a human testicular germ cell tumor. High activity of LDH-1 was found in tumor tissue and tumor cystic fluid. By histochemical staining LDH was found in all tumor cells. Most tumor cells had a faint staining reaction while some cells dispersed throughout the tumor had a strong staining reaction. The serum LDH-1 in athymic mice with tumor transplants correlated markedly with the tumor volume. The results indicate that serum LDH-1 was derived from the testicular germ cell tumor transplants.  相似文献

The spectrum of LDH isozymes was studied using different concentration of X-100 triton for enzyme extraction from the eggs and embryos, different systems of electrophoresis and different methods of staining electrophoregrams. Similar results were obtained in all cases. The mouse eggs and embryos were shown to contain only LDH-1; the treatment of oviducal fluid with X-100 triton strengthened the activity of LDH-4 and LDH-5. The detection of the activity of LDH-5 and hybrid isozymes appears to be related to the presence of oviducal fluid in the sample under study.  相似文献

Changes in the activities of lactate dehydrogenase isoenzymes in the gut and fat body of Galleria mellonella and Barathra brassicae larvac infected by the microsporidans Nosema plodiae and Pleistophora schubergi were studied by means of dise electrophoresis. In the normal last instar G. mellonella gut and fat body three isoenzymes, LDH-1, LDH-2-3, and LDH-4, and in B. brassicae two isoenzymes, LDH-1 and LDH-2-3, were present. In the fat body of both the animals infected by N. plodiae, the isoenzyme LDH-2-3 increased in activity substantially by the fifth day of infection. The gut LDH isoenzymes were not affected by the microsporidan. The same LDH-2-3 effect could be provoked by some enzymes toxic for G. mellonella larvae such as phospholipase-C and protease preparations.  相似文献
