The present study was carried out to determine an optimum honeybee colony’s carrying capacity of selected valleys dominated by Ziziphus spina-christi and Acacia tortilis in the Al-Baha region, Kingdom of Saudi Arabia. The study was conducted based on the assessment of the number of colonies kept, their productivities and the existing productive bee forage resources in the target valleys with its economic implication. In the existing beekeeping practice, the average number of managed honeybee colonies introduced per square kilometer was 530 and 317 during the flowering period of Z. spina-christi and A. tortilis, respectively. Furthermore, the overall ratios of productive bee forage plants to the number of honeybee colonies introduced were 0.55 and 11.12 to Ziziphus trees and A. tortilis shrubs respectively. In the existing situation the average honey production potential of 5.21 and 0.34 kg was recorded per Ziziphus and A. tortilis plants per flowering season, respectively. The present study, revealed that the number of honeybee colonies introduced in relation to the existing bee forage potential was extremely overcrowding which is beyond the carrying capacity of bee forage resources in selected valleys and it has been observed to affect the productivities and subsequent profitability of beekeeping. The study infers that, by keeping the optimum honeybee colony’s carrying capacity of valleys (88 traditional hives/km2 or 54 Langstroth hives/km2 in Ziziphus field and 72 traditional hives/km2 or 44 Langstroth hives/km2 in A. tortilis field), profitability of beekeeping can be boosted up to 130.39% and 207.98% during Z. spina-christi and A. tortilis, flowering seasons, respectively.  相似文献

Lut desert is situated in one of the extremely arid climatic zones of Iran and is one of the hottest deserts in our plant with the extreme fluctuation of temperature over a day. The main objective of this study is to characterize the diversity of the culturable actinomycetes and preliminary evaluation of their extracts as antimicrobial components on drug resistant pathogens. Twenty-four soil samples were collected, successively diluted and inoculated into the different culture media to support the growth of most culturable bacteria including actinomycetes. Phenotypic and molecular methods were used for accurate identification of recovered isolates particularly actinomycetes at the genus and species levels. The isolates were also evaluated for their inhibitory activities against drug resistant Acinetobacter baumannii, Enterococcus faecium, Klebsiella pneumoniae and Staphylococcus aureus. A total of 56 isolates recovered from the samples. Based on phenotypic tests, 41 isolates were identified as actinomycetes, amongst them 8 isolates were active against drug resistant pathogens. Our study revealed Lut desert, as one of the hottest deserts in the world, is the habitat to diverse taxa of bacteria particularly actinomycetes which have potential novel antimicrobial components.

The aim of this study was to isolate and screen actinomycetes from solitary wasp and swallow bird mud nests for antimicrobial activity. The actinomycetes were isolated from soil of nests of solitary wasp and swallow bird, and identified on the basis of morphological characteristics and molecular biological methods. A total of 109 actinomycetal isolates were obtained from 12 soil samples (6 from each habitat) using two media. The highest number of actinomycetes were recovered on Humic acid vitamin agar media (65.13%, n = 71) as compared to actinomycetes isolation agar media (34.86%, n = 38). The antimicrobial activity of actinomycetes isolates was determined using the agar plug method. Among 109 isolates, 51 isolates (46.78%) showed antibacterial activity by agar plug assay. The morphological and molecular characteristics confirmed that the most of active isolates in both sample belonged to the genus Streptomyces, the other potential genera like Streptosporangium, Actinomadura, Saccharopolyspora, Thermoactinomycetes and Nocardia were also recovered, but in a low frequency. The isolates designated as 8(1)*, BN-6, MN 2(6), MN 2(7) and MN 9(V) showed most promising activity against various drug resistant bacterial pathogens. It seems that the promising isolates from these unusual/unexplored habitats may prove to be an important step in development of drug for treating multi-drug resistant bacterial pathogens.  相似文献

Honey bee males and queens mate in mid air and can fly many kilometres on their nuptial flights. The conservation of native honey bees, such as the European black bee (Apis mellifera mellifera), therefore, requires large isolated areas to prevent hybridisation with other subspecies, such as A. m. ligustica or A. m. carnica, which may have been introduced by beekeepers. This study used DNA microsatellite markers to determine the mating range of A. m. mellifera in two adjacent semi-isolated valleys (Edale and Hope Valley) in the Peak District National Park, England, in order to assess their suitability for native honey bee conservation and as isolated mating locations. Three apiaries were set up in each valley, each containing 12 colonies headed by a virgin queen and 2 queenright drone producing hives. The virgin queens were allowed to mate naturally with drones from the hives we had set up and with drones from hives owned by local beekeepers. After mating, samples of worker larvae were taken from the 41 queens that mated successfully and genotyped at 11 DNA microsatellite loci. Paternity analyses were then carried out to determine mating distances and isolation. An average of 10.2 fathers were detected among the 16 worker progeny. After correction for non-detection and non-sampling errors, the mean effective mating frequency of the test queens was estimated to be 17.2, which is a normal figure for honey bees. Ninety percent of the matings occurred within a distance of 7.5 km, and fifty percent within 2.5 km. The maximal mating distance recorded was 15 km. Queens and drones did occasionally mate across the borders between the two valleys, showing that the dividing mountain ridge Losehill does not provide complete isolation. Nevertheless, in the most isolated part of Edale sixty percent of all matings were to drones from Edale hives. The large majority of observed mating distances fell within the range of Hope Valley, making this site a suitable location for the long term conservation of a breeding population of black bees.  相似文献

Fifty-sixGluconobacter strains and oneAcetobacter strain were isolated from honey bees and their environment in three different regions in Belgium and identified phenotypically. Polyacrylamide gel electrophoresis of the soluble cell proteins showed that two different types exist within theGluconobacter isolates: strains from type A were found in samples of the three regions, whereas strains from type B were only isolated in two of the three regions. Both types could occur in bees from the same region, from several hives of one bee keeper and from one hive. Strains from type A were almost identical with collection strainG. oxydans subsp.suboxydans NCIB 9018, whereas strains from type B consituted a new protein electrophoretic type within the genusGluconobacter. AlthoughGluconobacter is apparently associated with honey bees, it is not known whether it is important or required for the bees or any hive product.  相似文献

Thirty-eight actinomycetes were isolated from sediment collected from the Mariana Trench (10,898 m) using marine agar and media selective for actinomycetes, notably raffinose-histidine agar. The isolates were assigned to the class Actinobacteria using primers specific for members of this taxon. The phylogenetic analysis based on 16S rRNA gene sequencing showed that the isolates belonged to the genera Dermacoccus, Kocuria, Micromonospora, Streptomyces, Tsukamurella and Williamsia. All of the isolates were screened for genes encoding nonribosomal peptide and polyketide synthetases. Nonribosomal peptide synthetase sequences were detected in more than half of the isolates and polyketide synthases type I (PKS-I) were identified in five out of 38 strains. The Streptomyces isolates produced several unusual secondary metabolites, including a PKS-I associated product. In initial testing for piezotolerance, the Dermacoccus strain MT1.1 grew at elevated hydrostatic pressures.  相似文献

Feral European Honey Bee (Apis mellifera) has been identified as a potential nest competitor for Australian hollow nesting species, but few studies have investigated the impact of feral honey bee competition on Threatened species. Our study used data from Glossy Black‐cockatoo (Calyptorhynchus lathami halmaturinus) nests on Kangaroo Island, monitored and managed over an 11‐year period, and found 12% of nests became occupied by feral honey bees during that period. Our results indicate that feral honey bees were less likely to occupy nest boxes made of PVC (5%) compared with wooden nest boxes (24%) or natural hollows in Eucalyptus trees (14%). The removal of feral honey bee hives from nests is a priority for long‐term conservation of glossy black‐cockatoos on Kangaroo Island. We recommend that PVC nest boxes are chosen for future nesting habitat restoration, due to the more frequent use of wooden nest boxes by feral honey bees.  相似文献

Microbial communities, associated with almost all metazoans, can be inherited from the environment. Although the honeybee (Apis mellifera L.) gut microbiome is well documented, studies of the gut focus on just a small component of the bee microbiome. Other key areas such as the comb, propolis, honey, and stored pollen (bee bread) are poorly understood. Furthermore, little is known about the relationship between the pollinator microbiome and its environment. Here we present a study of the bee bread microbiome and its relationship with land use. We estimated bacterial community composition using both Illumina MiSeq DNA sequencing and denaturing gradient gel electrophoresis (DGGE). Illumina was used to gain a deeper understanding of precise species diversity across samples. DGGE was used on a larger number of samples where the costs of MiSeq had become prohibitive and therefore allowed us to study a greater number of bee breads across broader geographical axes. The former demonstrates bee bread comprises, on average, 13 distinct bacterial phyla; Bacteroidetes, Firmicutes, Alpha‐proteobacteria, Beta‐proteobacteria, and Gamma‐proteobacteria were the five most abundant. The most common genera were Pseudomonas, Arsenophonus, Lactobacillus, Erwinia, and Acinetobacter. DGGE data show bacterial community composition and diversity varied spatially and temporally both within and between hives. Land use data were obtained from the 2007 Countryside Survey. Certain habitats, such as improved grasslands, are associated with low diversity bee breads, meaning that these environments may be poor sources of bee‐associated bacteria. Decreased bee bread bacterial diversity may result in reduced function within hives. Although the dispersal of microbes is ubiquitous, this study has demonstrated landscape‐level effects on microbial community composition.  相似文献

The epizootic disease caused by Apis cerana sacbrood virus (AcSBV) occurred in Eastern hive bee, A. cerana, since 2015 in Taiwan. A large-scale survey of this disease from September and December 2016 in Taiwan was performed including symptom check and molecular identification in honey bees of A. cerana hives and several A. mellifera hives, which were co-cultured with A. cerana. Based on the nucleotide sequences of partial VP1, the phylogenetic analysis with those of the known AcSBV isolates revealed that most of AcSBV isolates from Taiwan were closely relative to SBV-FZ and -JL isolates from China, whereas only one sample (N15-5-1) was in a distinct cluster, which was closely relative to SBV-LN from China too. The AcSBV prevalence was occurring in A. cerana hives in most areas of Taiwan except for those in Hualien and Pingtung Counties in Taiwan. Notably, the AcSBV prevalence rate showed the temporal increase from 47.1% to 69.6% within 4?months. In addition, 37.5% of AcSBV prevalence rate was found in A. mellifera hives. It showed that A. mellifera was also susceptible to AcSBV infection. The present results would provide the information on the epidemiology and for prospective research.  相似文献

Summary Metabolites from 942 microbial isolates were screened for insecticidal and nematicidal properties. The isolates included 302 streptomycetes, 502 novel actinomycetes including representatives of 18 genera, 28 unidentified aerobic actinomycetes, 70 fungi and 40 bacteria other than actinomycetes. When diluted 10-fold, the metabolites from 55 isolates caused nearly 100% mortality in mosquito larvae (Aedes aegypti) within 24 h. These isolates included 27 isolates ofStreptomyces, four ofActinoplanes, three isolates each ofActinomadura andStreptoverticillium, two isolates each ofMicromonospora, Bacillus andPaecilomyces, and one isolate each ofMicropolyspora, Nocardiopsis, Streptosporangium, Oerskovia, Thermomonospora, Chainia, Pseudomonas, Fusarium, Monilia andSyncephalestrum. Two fungal isolates could not be identified to the generie level. Extracts from the culture broth of 18 isolates caused 100% mortality in mosquito larvae within 15 min to 24 h at a concentration of 1000 ppm. The LC50 of partially purified products from two isolates was 1–2.5 ppm and that of the semipurified preparations from four other isolates was 50 ppm. Valinomycin was identified as an active component in the culture broth from one isolate. The taxonomic analysis of 107 actinomycete strains isolated from the bottom sediments and water of Lake Baikal showed that most of the water isolates belong to the genus Streptomyces and most of the sediment isolates belong to the genus Micromonospora. In the sediments, the number of actinomycetes increased with depth (down to 200 m). Eight Streptomyces isolates were identified to a species level.  相似文献

In a population of the social bee Melipona subnitida of about 200 hives, from Mossoró, Northeastern Brazil, 54 hives were sampled and studied electrophoretically as far as the alleles Est-3 S, Est-3 M and Est-3 F were concerned. Males are produced by eggs laid by workers (no more than 4 in the same day) and by the queen. It was estimated that 61.2% of the drones are sons of the queen and 38.8% are sons of the workers. With these values the effective number (Ne) was estimated in 395.80 which is 24% higher than in a model in which all males are sons of the queen.  相似文献

Six bee viruses, which occur in Apis mellifera, were monitored in the Czech Republic between 2006 and 2009. Samples of larvae and pupae collected from hives where American foulbrood was detected were screened for bee viruses and in the 125 samples of larvae, there was no confirmed case of a larva infected with both American foulbrood and a bee virus. Of 145 samples infected with the protozoan Nosema apis, there were 23 cases of coinfections with the BQCV virus, 18 with the DWV virus and 11 with the ABPV virus. All coinfections with three or four viruses were also statistically significant apart from the one between ABPV with CBPV and DWV. The PCA ordination diagram indicates that BQCV occurs mainly with Nosema apis and DWV mainly with ABPV.  相似文献

The European honey bee exploits floral resources efficiently and may therefore compete with solitary wild bees. Hence, conservationists and bee keepers are debating about the consequences of beekeeping for the conservation of wild bees in nature reserves. We observed flower-visiting bees on flowers of Calluna vulgaris in sites differing in the distance to the next honey-bee hive and in sites with hives present and absent in the Lüneburger Heath, Germany. Additionally, we counted wild bee ground nests in sites that differ in their distance to the next hive and wild bee stem nests and stem-nesting bee species in sites with hives present and absent. We did not observe fewer honey bees or higher wild bee flower visits in sites with different distances to the next hive (up to 1,229 m). However, wild bees visited fewer flowers and honey bee visits increased in sites containing honey-bee hives and in sites containing honey-bee hives we found fewer stem-nesting bee species. The reproductive success, measured as number of nests, was not affected by distance to honey-bee hives or their presence but by availability and characteristics of nesting resources. Our results suggest that beekeeping in the Lüneburg Heath can affect the conservation of stem-nesting bee species richness but not the overall reproduction either of stem-nesting or of ground-nesting bees. Future experiments need control sites with larger distances than 500 m to hives. Until more information is available, conservation efforts should forgo to enhance honey bee stocking rates but enhance the availability of nesting resources.  相似文献

Isolation of bioactive actinomycetes from marine sediments using rifampicin   总被引:4,自引:0,他引:4  
Summary Bioactive actinomycetes were isolated from marine sediments using rifampicin. Plating the sediments on starch-casein agar, supplemented with rifampicin, eliminated the occurrence of contaminating microorganisms. Total counts, however, were reduced in the presence of rifampicin. Most of the isolates contained ll-2,6-diaminopimelic acid (DAP), whereas 37% contained meso-DAP. The use of increasing concentrations of rifampicin tended to yield a higher proportion of strains with cell extracts positive for meso-DAP. Streptomyces and Micromonospora represented the major genera identified. Antimicrobial activity was exhibited by 46% of the isolates, primarily against Gram-positive bacteria. Inhibition of Gram-negative bacteria was minimal, but antimycotic activity was displayed by 28% of the actinomycetes. Most of the latter activity was attributable to polyenes, particularly hexanenes. The results obtained indicate that rifampicin, added to starch-casein agar, is effective for the isolation of bioactive actinomycetes from marine sediments.  相似文献

为分析澜沧江流域北部人食用蜂蜜中毒的原因,于2013年6—9月份,对该区域蜜蜂和蜜源植物的分布情况进行了调查,观察蜜蜂采集有毒蜜源植物的行为,并进一步调查了蜜蜂巢内蜂蜜、蜂花粉的储存情况,采集了中华蜜蜂蜂蜜样品,进行蜂蜜孢粉学与营养生态位分析。该区域有大量采用传统方式进行人工饲养的中华蜜蜂(Apis cerana cerana),及少量野生中华蜜蜂、黑色小蜜蜂(Apis audreniformis)、黑色大蜜蜂(Apis laboriosa smith)群体分布;人工饲养的中华蜜蜂蜂巢内部结构与野生中华蜜蜂蜂巢相似,为自然蜂巢,内有充足的蜜粉储存,部分蜂群蜂巢内虫害严重。该区域内主要蜜源植物为荞麦(Fagopyrum esculentum Moench),其他零星辅助蜜源较多,部分地点南烛(Vaccinium bracteatum Thunb)、昆明山海棠(Tripterygium hypoglaucum(Levl.)Hutch)连片集中分布。对中华蜜蜂蜂蜜进行孢粉学和营养生态位分析,结果表明:中华蜜蜂蜂蜜标本中含有有毒蜜源植物南烛、昆明山海棠花粉,部分样品中南烛、昆明山海棠的花粉含所占比例较高;中华蜜蜂的营养生态位宽度为0.22,比其他地区中华蜜蜂生态位指数小,推测澜沧江水电枢纽的修建等人为原因已对蜜蜂种类、蜜源植物的物种组成、群落结构造成了较大影响。  相似文献

Summary Abundance patterns during 6–7 years and orchid visitation were determined for 51 species of the 57 local euglossine bees. Male bees were counted at 3 chemical attractants presented in the same manner each month. Sites were separated by 75 km but included wet Atlantic forest at 500 m elevation, moist forest at 180 m near Barro Colorado Island, and cloud forest at 900 m near the Pacific ocean. 1. From 15 to 30 euglossine species of 4 genera were active in each month and site; monthly species number and general bee abundance were positively correlated. Many species had 3 annual abundance peaks (range 1–4) and were active throughout the year, but peak annual abundances rarely occurred during late wet or early dry seasons. In contrast, Eufriesea generally were present as adults only 1–2 months in a year. 2. Euglossine populations were exceptionally stable. Species at each site were more stable than any known insect population, and stability and abundance were positively associated. However, year-to-year population stability and the degree of seasonality were not correlated. Among the three sites, the more diverse (species rich) bee assemblages displayed lower stability; these were the wetter and upland sites. 3. The most abundant bees visited more orchid species. Eg. and El. each visited and average of 4 orchid species (range 0–13); Ex. and Ef. visited 0–3. Stable populations did not visit more or fewer orchid species than did unstable populations. 4. Less than 68% of species at each site visited orchid flowers; less than a few dozen of the 100–800 bees counted in a day carried orchid pollinaria. Over 20% of the euglossine species never were seen with pollinaria at any site and probably seldom visit orchids in central Panama. 5. Most bee species visited 1 or no fragrance orchids in a given habitat. Orchids tended to utilize common pollinators that seldom included more than 1 species, and they utilized stable or unstable, seasonal or aseasonal bees. However, the most stable and abundant bee, Eg. imperialis, rarely pollinated orchids; fewer than 10 of ca. 20000 bees carried pollinaria. 6. Orchids may interact primarily with discrete seasonal bee population peaks-probably the emerging adults. Although specialized orchid preferences are implicated for species that visit few or no local orchids but pollinate other species and carry pollinaria in other areas, euglossine bees do not need orchids to survive or reproduce.  相似文献

Pollination is an essential step in the seed production of canola, Brassica napus L. It is achieved with the assistance of various pollen vectors, but particularly by the honey bee, Apis mellifera L. Although the importance of pollination has been shown for the production of seed crops, the need to introduce bee hives in canola fields during flowering to increase oil seed yield has not yet been proven. With the purpose of showing this, hives of A. mellifera were grouped and placed in various canola fields in the Chaudière-Appalaches and Capitale-Nationale regions (nine fields; three blocks with three treatments; 0, 1.5, and 3 hives per hectare). A cage was used to exclude pollinators and bee visitations were observed in each field. After the harvest, yield analyses were done in relation to the bee density gradient created, by using pod set, number of seeds per plant, and weight of 1000 seeds. Results showed an improvement in seed yield of 46% in the presence of three honey bee hives per hectare, compared with the absence of hives. The introduction of honey bees contributed to production and consequently, these pollinators represented a beneficial and important pollen vector for the optimal yield of canola.  相似文献

The aim of this study was to identify novel biofilm inhibitors from actinomycetes isolated from the Arctic against Vibrio cholerae, the causative agent of cholera. The biofilm inhibitory activity of actinomycetes was assessed using biofilm assay and was confirmed using air–liquid interphase coverslip assay. The potential isolates were identified using 16S rRNA gene sequencing. Of all, three isolates showed significant biofilm inhibition against V. cholerae. The results showed that 20% of the actinomycetes culture supernatant could inhibit up to 80% of the biofilm formation. When different extracted fractions were assessed, significant biofilm inhibition activity was only seen in the diethyl ether fraction of A745. At 200 μg ml−1 of diethyl ether fraction, 60% inhibition of V. cholerae biofilm was observed. The two potential isolates were found to be Streptomyces sp. and one isolate belonged to Nocardiopsis sp. This is the first report showing a Streptomyces sp. and Nocardiopsis sp. isolated from the Arctic having a biofilm inhibitory activity against V. cholerae. The spread of drug resistant V. cholerae strains is a major clinical problem and the ineffectiveness in antibiotic treatment necessitates finding new modes of prevention and containment of the disease, cholera. The formation of biofilms during the proliferation of V. cholerae is linked to its pathogenesis. Hence, the bioactive compound from the culture supernatant of the isolates identified in this study may be a promising source for the development of a potential quorum sensing inhibitors against V. cholerae.  相似文献

Three natural populations of actinomycetes were investigated by PCR for the presence of type I 3-hydroxy-3-methylglutaryl coenzyme A reductase (HMG CoA), a gene associated with isoprenoid biosynthesis. The populations were obtained from an agricultural site (69 isolates), a coastal salt marsh (220 isolates), and a desert soil (96 isolates). A set (34) of standard actinomycete reference strains were also investigated. The target gene was only detected in 5 of the 419 actinomycetes screened, which represented 4 from the coastal salt marsh and one reference strain. The isolates that contained the gene were taxonomically diverse (4 Streptomyces spp. and 1 Nocardia sp.). These results suggest that type I HMG CoA containing pathways are rare in actinomycetes and their distribution within actinomycetes populations is not random.  相似文献
