琥珀酸脱氢酶(SDH)的活性在判断心肌缺血,再灌性损伤中起重要作用。本文通过IBAS图象分析系统对缺血再灌的培养心肌细胞内的SDH定量评估,以求探讨大豆磷脂脂质体对心肌缺血再灌性损伤的保护作用。结果表明,在缺血及再灌组的心肌细胞内出现较强的SDH损伤性反应,大豆磷脂脂质体能明显减轻这种反应,提示大豆磷脂脂质体对缺血及缺血/再灌心肌具有明显的保护作用。  相似文献

目的:观察非选择性一氧化氮合酶抑制剂NG-硝基-L-精氨酸(NG-nitro-L-arginine,L-NA)对局灶性脑缺血大鼠脑线粒体的损伤作用,以探讨其改善缺血性脑损伤的作用机制。方法:将大鼠随机分为假手术组、缺血对照组、L-NA治疗组,采用线栓法阻断大鼠大脑中动脉(MCAO)复制局灶性脑缺血模型,分别于缺血后2h、6h、12h给药治疗3d,迅速断头取脑,差速离心法提取缺血侧脑组织线粒休,迅速测定线粒体膜肿胀度及线粒体活力,测定线粒体总ATP酶、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性,以及线粒休一氧化氮(NO)、丙二醛(MDA)含量:电镜观察缺血后皮层神经元超微结构的改变及L-NA对其影响。结果:在大鼠MCAO后线粒体膜肿胀度增加,线粒体活力下降,线粒体NO、MDA含量明显增加,线粒体总ATP酶、SOD、GSH-Px活性均明显下降:缺血后2h、6h、12h给予L-NA治疗3d与缺血对照组相比NO含量明显下降,缺血后12h治疗组线粒体膜肿胀度、线粒体活力、总ATP酶、SOD、GSH-Px活性均显著升高、MDA含量下降。电镜结果显示脑缺血后皮层神经元水肿,线粒体肿胀、嵴断裂、溶解、消失,且随缺血时间延长损伤加重;缺血后12h给予L-NA治疗能明显改善脑缺血引起的神经元水肿、线粒体肿胀和空泡化。结论:L-NA能明显抑制脑缺血后线粒体NO生成,在缺血早期给予L-NA对缺血性脑损伤无改善作用:缺血后期给予L-NA,能明显降低线粒体膜肿胀程度,改善线粒体能量供应,增强线粒体抗氧化作用及其活力,从而减轻脑缺血损伤。  相似文献

目的:观察胰岛素对大鼠肠缺血再灌注后小肠组织损伤的影响。方法:雄性SD大鼠40只随机分为4组,每组10只,手术对照组、单纯缺血组、再灌注组、胰岛素干预组。于30min缺血和120min再灌注后,进行组织病理学和生化检测。结果:(1)单纯缺血组肠粘膜损害较手术对照组明显升高(P<0.01),超氧化物歧化酶(SOD)活性无明显变化;(2)再灌注组SOD活性明显降低,与手术对照组和单纯缺血组相比较差异均有显著性(P<0.01);(3)胰岛素组SOD活性与再灌注组相比有明显改善(P<0.01)。结论:肠缺血可以引起肠粘膜损伤,再灌注则可加重这种损伤,胰岛素可以减轻再灌注损伤。  相似文献

目的和方法:利用整体大鼠异丙肾上腺素损伤(ISO)和离体大鼠全心停灌/再灌(I/R)两种模型,观察了心肌缺血和缺血/再灌注对心肌生物膜-线粒体膜及肌纤维膜损伤的影响。结果:ISO(5mg/kg,皮下注射)和I/R(20min/20min)可导致大鼠心脏生物膜产生严重损伤,表现为心肌线粒体脂质过氧化产物明显增加,线粒体磷脂酶A2(PLA2)激活,从而导致线粒体膜磷脂(PL)含量减少,磷脂分解产物游离脂肪酸(FFA)增加,膜脂流动性(LFU)降低,线粒体Ca^2 -ATPase及肌纤维膜Na^ ,K^ -ATPase活性降低,线粒体呼吸功能降低、呼吸链氧化磷酸化解偶联,高能磷酸化合物生成减少。结论:整体ISO和离体I/R可导致大鼠心肌线粒体、肌纤维膜结构和功能损伤。  相似文献

探讨川芎嗪联用L-精氨酸对心肌缺血/再灌注损伤(MI/RI)时心肌细胞线粒体功能的影响。方法:选用日本大耳白兔50只,随机分为正常对照组(A组)、心肌缺血/再灌注组(B组)、心肌缺血/再灌注+川芎嗪治疗组(C组)、心肌缺血/再灌注+L-精氨酸治疗组(D组)和心肌缺血/再灌注+川芎嗪+L-精氨酸治疗组(E组)。观察心肌线粒体呼吸功能、Ca^2+浓度([Ca^2+]m)、丙二醛浓度(MDA)、超氧化物歧化酶活性(SOD)和心肌组织三磷酸腺苷(ATP)、能荷(EC)的变化。结果:C、D、E组与B组比较,线粒体呼吸控制率(RCR)、Ⅲ态呼吸速率(ST3)、SOD明显升高,Ⅳ态呼吸速率(ST4)、[Ca^2+]m、MDA显著降低,心肌组织ATP、EC均明显增高;且与A组比较,E组上述指标均无明显差异。结论:川芎嗪联用L-精氨酸可通过降低氧自由基水平和减轻钙超载,而改善缺血/再灌注损伤心肌的线粒体功能。  相似文献

目的:探讨硫化氢(H2S)对急性心肌缺血大鼠线粒体功能的影响,并探讨其改善急性心肌缺血损伤的作用机制。方法:通过结扎大鼠左冠状动脉前降支建立急性心肌缺血模型。雄性SD大鼠48只随机分为6组(n=8):假手术组,缺血组,缺血+硫氢化钠(NaHS)低、中、高剂量组和缺血+炔丙基甘氨酸(PPG)组。透射电镜观察心肌组织线粒体超微结构;检测血浆中H2S含量、心肌组织CSE活性;测定心肌线粒体活力、膜肿胀度及线粒体总ATP酶、谷胱甘肽过氧化物酶(GSH-PX)、超氧化物歧化酶(SOD)的活性和丙二醛(MDA)含量。结果:与假手术组比较,缺血组大鼠血浆H2S含量和心肌组织中CSE活性降低;心肌线粒体膜肿胀,线粒体活力下降;线粒体中MDA含量明显升高,ATP酶、SOD、GSH-Px活性明显降低(P〈0.01)。与缺血组比较,缺血+NaHS低、中、高剂量组大鼠血浆H2S含量和心组织中CSE活性均升高;缺血+NaHS中、高剂量组大鼠心肌线粒体MDA含量明显减少,膜肿胀度减轻;缺血+NaHS低、中、高剂量组线粒体活力有所恢复,ATP酶、SOD、GSH-Px的活性明显升高(P〈0.05或P〈0.01)。PPG可部分减弱H2S的心肌保护作用(P〈0.05或P〈0.01)。结论:H2S可增强线粒体ATP酶、SOD、GSH-Px的活性,降低线粒体脂质过氧化水平,从而起到对大鼠急性心肌缺血的保护作用。  相似文献

目的:探讨线粒体膜通透性转换孔(MPTP)抑制剂——环孢素A(CsA)对大鼠肺常温缺血/再灌注后细胞凋亡的影响。方法:健康SD大鼠30只,随机分为3组(n=10):假手术组、缺血/再灌注组(I/R组)和环孢素A干预组(CsA组)。复制在体肺缺血/再灌注损伤模型。采用原位缺口末端标记(TUNEL)法检测肺组织细胞凋亡,免疫组化技术检测肺组织细胞细胞色素C(CytC)的含量,以及分光光度计测定肺组织细胞caspase-3的活性。结果:I/R组肺组织细胞胞浆CytC的含量、caspase-3活性明显高于假手术组(P0.01),并观察到大量肺组织细胞凋亡的发生。CsA组与I/R组相比,CytC释放明显减少(P0.01),caspase-3活性减弱,细胞凋亡的发生率明显下降(P0.01)。结论:环孢素A可能通过抑制MPTP开放,减少缺血/再灌注后线粒体CytC的释放,从而减少肺组织细胞的凋亡。  相似文献

目的:复制L-6TG大鼠肌母细胞缺血/再灌注损伤的细胞模型.方法:将培养的L-6TG大鼠肌母细胞随机分为2组:①正常对照组(C组),②缺血/再灌注组(I/R组),观测了培养上清中乳酸脱氢酶(LDH)、细胞内超氧化物歧化酶(SOD)、黄嘌呤氧化酶(XOD)、Ca2 含量的变化;采用MTT法检测线粒体的功能;在光镜下观察细胞的形态学改变.结果:与对照组相比,L-6TG大鼠肌母细胞IR 4h后培养上清中LDH、细胞内XOD、Ca2 含量明显增加,细胞内SOD及线粒体呼吸功能明显降低,细胞严重受损,明显圆缩,并有脱落现象.结论:应用模拟缺血液和再灌液可成功复制L-6TG大鼠肌母细胞缺血/再灌注损伤的细胞模型.  相似文献

目的 制备一种新型的心肌急性缺血再灌注损伤模型,以探讨一种更符合临床实际需求的实验方法.方法 将20只雌性SD(Sprague-Dawley)大鼠随机分成2组(对照组、实验组),采用结扎主动脉根部引起心肌缺血5min再灌注30 min建立心肌急性缺血再灌注模型;通过应用透射电镜观察心肌细胞超微结构的改变,同时检测心肌组织匀浆丙二醛(Maleic Dialdehyde,MDA)含量、超氧化物歧化酶(Superoxide Dismutase,SOD)活力.结果 透射电镜下超微结构显示实验组较对照组明显加重了心肌组织结构和线粒体的损害;实验组心肌组织MDA明显高于对照组(P<0.01),而SOD明显低于对照组(P<0.01).结论 本实验成功建立了方法简便、易于操作、取材范围广泛的心肌缺血再灌注损伤模型,为心肌缺血再灌注损伤研究提供了一种更为可行的模型.  相似文献

目的:探讨参麦注射液对肢体缺血/再灌注时肺脂质过氧化损伤的防护作用。方法:复制家兔缺血/再灌注(I/R)损伤模型,分别从右颈外静脉和左颈总动脉取血,代表入肺血和出肺血,观察入、出肺血及肺组织超氧化物歧化酶(SOD)、丙二醛(MDA)及参麦注射液对上述指标的影响。结果:与对照组比较,缺血再灌组松夹后4h入、出肺血及肺组织SOD活性明显降低,MDA含量增高(P<0.01);再灌前30min静脉给予参麦注射液后,SOD活性升高,而MDA含量降低(P<0.01)。相关分析显示MDA与SOD间存在明显负相关(P<0.05)。结论:缺血再灌注时伴有肺脏氧自由基代谢紊乱,参麦注射液通过清除氧自由基,对抗脂质过氧化,减轻肺损伤。  相似文献

Mitochondria isolated from 3-day-old etiolated corn shoots (Zea mays L.) can be categorized into three separate groups, each group characteristic of the cell type from which the mitochondria were isolated. Phloem sieve tubes and some adjacent parenchyma cells contain mitochondria that have few cristae and little amorphous matrix. Mitochondria from meristematic and undifferentiated cells have more cristae and matrix. Vaculate and differentiated cells have mitochondria with well-developed cristae and abundant matrix. Each mitochondrial type exhibits typical in vitro spontaneous swelling and substrate-induced contraction responses. characterized by change or lack of change in cristae size and in density of amorphous material. For the second and third types of mitochondria, swelling and contraction are characterized by a change in degree of cristae size and in matrix density. The first type undergoes few changes upon swelling or contraction. Radical changes of the inner membrane, withdrawal and infolding, are associated with cell differentiation and not with swelling and contraction of isolated corn shoot mitochondria.  相似文献

利用透射电镜(TEM)技术研究了泥螺卵黄发生过程中线粒体的形态结构的变化特点,结果表明,从卵黄发生早期到晚期,卵母细胞内线粒体经历了从外部形态到内部结构的一系列变化。卵黄合成初期的卵母细胞内,线粒体多,结构典型,仅部分线粒体外膜破裂,嵴 和内膜逐渐消失,卵黄发生中期,线粒体基质空泡化,嵴和内膜消失,腔内充满颗粒状物质,最后演变成卵黄颗粒,随着卵母细胞的发育,卵黄颗粒的数量和直径逐渐增加,卵黄发生后期,卵质中胞器不发达,细胞质中充满卵黄颗粒,在卵黄颗粒之间仅有少量线粒体存在,提供细胞代谢所需的能量,此外,对线粒体在卵黄形成中的功能,去向及行为变化等 进行了讨论。  相似文献

胆红素对大鼠肺脏缺血再灌注损伤保护作用的研究   总被引:3,自引:0,他引:3  
目的探讨胆红素对实验性大鼠肺缺血再灌注损伤的保护作用并探讨其发生的机制。方法60只健康Wistar大鼠随机分为3组:手术对照(C)组,缺血再灌注(IR)组,胆红素干预(B)组。每组分别于缺血第45min、再灌注30min、60min、120min4个时点,经左房放血处死大鼠,观察肺组织病理形态变化,检测血浆丙二醛(MDA)、超氧化物歧化酶(SOD)的含量,测定肺组织干/湿重(D/W)比值,TUNEL法测定肺组织中细胞凋亡指数(AI)。结果①肺组织病理变化:缺血再灌注后IR组肺组织损伤进行性加重,毛细血管充血、肺泡间隔炎性细胞浸润、肺泡腔内炎性细胞及炎性液体渗出显著,B组肺组织充血、水肿、炎性细胞浸润较IR组减轻。②血浆MDA含量:IR组在缺血再灌注后血浆MDA含量明显增加,较C组和B组同时点均显著增高(P〈0.01),而B组的MDA含量在缺血再灌注过程中的变化无显著性意义;③血浆SOD含量:经缺血再灌注后,IR和B组血浆SOD含量较C组同时点均显著下降(P〈0.05),B组减少的程度明显小于IR组(P〈0.05);④肺组织D/W比值:经缺血和再灌注后,IR组和B组的肺组织D/W比值都呈进行性下降(再灌注60、120minvs缺血45min,P〈0.01);B组下降幅度明显小于IR组(再灌注60、120min时,B组vsIR组,P〈0.05);⑤肺组织细胞AI的变化:IR组及B组缺血再灌注后均可见肺组织细胞凋亡现象,但与IR组相比,B组相同时点的肺组织细胞凋亡数显著减少(P〈0.05);结论胆红素对于实验性大鼠肺脏缺血再灌注损伤具有一定的保护作用,其作用机制与清除氧自由基、抗氧化及抗细胞凋亡有关。  相似文献

Summary Several investigators have described the ultrastructural changes that occur in the mitochondria of cells in tissue cultures after treatment with the drug ethidium bromide (E). The mitochondria swell and the cristae become greatly altered and finally disappear; in the cristae-free region of the matrix electron-dense granules can be observed. It has been assumed that intercalation of E between the base pairs of the mitochondrial DNA induces the formation of the granular inclusions. To investigate whether intercalation is really the initial step in the generation of dense granules inside the matrix, we performed a comparative incubation study of HeLa-cell mitochondria in situ using three closely related dyes (D), i.e. E, tetramethylethidium bromide (TME) and betaine B (B). They strongly differ with regard to their affinity for DNA and their ability to cross membranes. E was used as a reference dye. TME does not intercalate, but is externally bound to DNA only weakly. The neutral B is not bound at all, but can cross membranes more easily than the cation E. Moreover, in aqueous solutions at pH7.0, B is in equilibrium with its protonated cation BH. BH and E have almost equal affinities for DNA. Therefore B may quickly pass the inner mitochondrial membranes and the cristae, and should then be bound inside the matrix, thus forming a BH-DNA complex. On the assumption that intercalation is necessary for the generation of intramitochondrial electron-dense bodies, we predicted that BH/B should be more efficient than E, while TME should be relatively ineffective. In experiments using HeLa cells, these predictions were found to be inaccurate. E, TME and BH/B produced almost the same mitochondrial alterations, but at different concentrations and after different incubation periods. In contrast to our expectations TME was much more effective than E and BH/B, with the last two behaving rather similarly.Therefore, it seems unlikely that the drugs penetrate the inner mitochondrial membrane system by simple physical diffusion or that intercalation is the preliminary step for the generation of dense granules inside the matrix. Instead, we assume that hydrophobic interaction between the dye cations E, BH and TME and the cristae is the main cause of the mitochondrial changes. The favoured binding partner of the dye cations may be the divalent anion, cardiolipin: this phospholipid is an essential part of the inner membrane system but is absent in other membranes of cells. By distributing the dyes between a lipophilic phase and water, it was shown that TME is more lipophilic than E and BH; this may explain the greater effectiveness of TME. The bound dye cations disturb the organization of the cristae, which become altered and finally disappear. We assume that the electron-dense granules in the matrix are mainly composed of the dyes and former membrane materials such as phospholipids and proteins, as well as perhaps some other hydrophobic matrix materials. This would also explain why it was impossible to digest the dense granules by DNase treatment. The drugs enter the mitochondrial matrix by disordering and finally destroying the cristae.  相似文献

The relationships between germinal bodies and mitochondria were studied in the holothurian Apostichopus japonicus and the flounder Pleuronectes asper using TEM. In the gonial cells of both species the mitochondria are arranged around germinal bodies and are in contact with the latter. A gradual disappearance of the outer membrane is found in the mitochondria that interact with the germinal substance. Later on, dispersion of the globules of the mitochondrial matrix containing mitochondrial cristae occurs. It is supposed that the substance of the mitochondrial matrix takes part in the development and functioning of the germinal plasm in both invertebrates and vertebrates.  相似文献

目的观察重组人促红细胞生成素(recombinant human erythropoietin,rhEPO)对缺血/再灌注损伤大鼠心肌细胞Mitofusin2(Mfn2)蛋白表达的影响及其抗心肌细胞凋亡的作用。方法选取成年SD大鼠35只,随机分为正常组(Normal),假手术组(Sham),缺血再灌注组(I/R),缺血再灌注EPO治疗组(I/R+EPO)。各组分别于再灌注3h和24h后,剪取心脏缺血/再灌注损伤区域,用脱氧核苷酸末端转移酶介导的缺口末端标记法(TUNEL)检测心肌细胞凋亡,免疫组化法检测Mfn2蛋白的表达。结果再灌注3h和24h后,与正常组和假手术组相比,I/R组Mfn2蛋白的表达和心肌细胞凋亡均显著增加;与I/R组相比,I/R+EPO组Mfn2蛋白的表达和心肌细胞凋亡均显著降低。结论EPO可以下调缺血再灌注损伤后心肌细胞Mfn2蛋白的表达,抑制心肌细胞的凋亡。  相似文献

目的和方法:本文用异丙肾上腺素(Iso) 造成大鼠心肌缺血动物模型,观察支链氨基酸(BCAA)对大鼠心肌缺血时线粒体结构和功能损伤的预防作用。结果:BCAA 能显著降低心肌缺血后心肌线粒体中丙二醛(MDA) 水平、维持线粒体模平均微粘度(-η) 、线粒体呼吸链中细胞色素氧化酶及心肌肌球蛋白ATPase活力。结论:给予BCAA对保护大鼠心肌线粒体的结构和功能免受缺血性损伤具有一定效果  相似文献

Pathological alterations of the fine structure of the cardiac muscle of hereditary muscular dystrophic hamsters (BIO 14.6), with peculiar changes of mitochondria, which were possible degenerative, suggest a mode of formation of intramitochondrial dense bodies. The initial stage of this process appeared in the outer compartment of a mitochondrion where the outer membrane began to vesiculate or interdigit with another outer membrane of an adjacent mitochondrion. Then this vesiculated region developed into an intramitochondrial dense area, which further invaded the central part of the mitochondrion keeping its continuity with the cristae and its vesicle substructure. Independently, some parts of the membrane of the mitochondrial cristae fused with each other and formed dark spots. They were also included in the vesiculated intramitochondrial dense area. The vesiculated outer membrane may be involved in deterioration of the calcium binding site compatible with increased calcium of the prenecrotic stage of the cardiac muscle of dystrophic hamster.  相似文献
