Background

Different evolutionary hypotheses predict a correlation between the fitness of a genotype in the absence of infection and the likelihood to become infected. The cost of resistance hypothesis predicts that resistant genotypes pay a cost of being resistant and are less fit in the absence of parasites. The inbreeding-infection hypothesis predicts that the susceptible individuals are less fit due to inbreeding depression.

Methods and Results

Here we tested if a host''s natural infection status was associated with its fitness. First, we experimentally confirmed that cured but formerly infected Daphnia magna are genetically more susceptible to reinfections with Octosporea bayeri than naturally uninfected D. magna. We then collected from each of 22 populations both uninfected and infected D. magna genotypes. All were treated against parasites and kept in their asexual phase. We estimated their relative fitness in an experiment against a tester genotype and in another experiment in direct competition. Consistently, we found no difference in competitive abilities between uninfected and cured but formerly infected genotypes. This was the case both in the presence as well as in the absence of sympatric parasites during the competition trials.

Conclusions

Our data do not support the inbreeding-infection hypothesis. They also do not support a cost of resistance, however ignoring other parasite strains or parasite species. We suggest as a possible explanation for our results that resistance genes might segregate largely independently of other fitness associated genes in this system.  相似文献

1. Evolutionary increases in dispersal‐related traits are frequently documented during range expansions. Investment in flight‐related traits is energetically costly and a trade‐off with fecundity may be expected during range expansion. 2. However, in contrast to wing‐dimorphic species, this trade‐off is not general in wing‐monomorphic species. In the absence of a dispersal‐‐fecundity trade‐off, an increased investment in clutch size at the expansion front is expected possibly at a cost of reduced offspring size. 3. The study evaluated investment in female flight morphology and fecundity‐related traits (clutch size, hatchling size) and potential trade‐offs among these traits in replicated populations of the poleward range‐expanding damselfly Coenagrion scitulum. 4. Females at the expansion front had a higher relative thorax length, indicating an increased investment in flight; this can be explained by spatial sorting of dispersal ability or in situ natural selection at the expansion front. Edge females produced larger hatchlings, however, this pattern was totally driven by the population‐specific thermal larval regimes and could not be attributed to the range expansion per se. By contrast, clutch sizes did not differ between core and edge populations. There was no signal of a dispersal–fecundity trade‐off either for a trade‐off between clutch size and hatchling size. 5. These results indicate that evolution of a higher dispersal ability at the expansion front of C. scitulum does not trade off with investment in fecundity, hence a dispersal–fecundity trade‐off is unlikely to slow down range expansion of this species.  相似文献

In comparisons across Drosophila species, faster pre-adult development is phenotypically correlated with increased pre-adult competitive ability, suggesting that these two traits may also be evolutionary correlates of one another. However, correlations between traits within- and among- species can differ, and in most cases it is the within-species genetic correlations that are likely to act as constraints on adaptive evolution. Moreover, laboratory studies on Drosophila melanogaster have shown that the suite of traits that evolves in populations subjected to selection for faster development is the opposite of the traits that evolve in populations selected for increased pre-adult competitive ability. This observation led us to propose that, despite having a higher carrying capacity and a reduced minimum food requirement for completing development than controls, D. melanogaster populations subjected to selection for faster development should have lower competitive ability than controls owing to their reduced larval feeding rates and urea tolerance. Here, we describe results from pre-adult competition experiments that clearly show that the faster developing populations are substantially poorer competitors than controls when reared at high density in competition with a marked mutant strain. We briefly discuss these results in the context of different formulations of density-dependent selection theory.  相似文献

Pyrethroid resistance in mosquitoes   总被引:3,自引:0,他引:3
Repeated blood feedings throughout their life span have made mosquitoes ideal transmitters of a wide variety of disease agents. Vector control is a very important part of the current global strategy for the control of mosquito-associated diseases and insecticide application is the most important component in this effort. Pyrethroids, which account for 25% of the world insecticide market, are currently the most widely used insecticides for the indoor control of mosquitoes and are the only chemical recommended for the treatment of mosquito nets, the main tool for preventing malaria in Africa. However, mosquito-borne diseases are now resurgent, largely because of insecticide resistance that has developed in mosquito vectors and the anti-parasite drug resistance of parasites. This paper reviews our current knowledge of the molecular mechanisms governing metabolic detoxification and the development of target site insensitivity that leads to pyrethroid resistance in mosquitoes.  相似文献

Group‐living animals can gain protection against parasitic infections through social contacts with previously infected conspecifics (social immunization). Recent research suggests that such protective effects can be induced through visual or chemical cues released by infected individuals, resulting in anticipatory immune upregulation among group members. Here, we study cue‐induced social resistance in rainbow trout Oncorhynchus mykiss exposed to a trematode parasite, the eye‐fluke Diplostomum pseudospathaceum. We established groups of naïve individuals (receivers) that were paired with previously infected individuals (donors) at different ratios of donors to receivers and at different time points since donor exposure to capture varying concentrations of the anticipated cues. While the pre‐infection elevated resistance among the donors, there was no evidence of social transfer of resistance, regardless of the ratio of donors and receivers in a group or the time since the pre‐infection. The results suggest that resistance through social signalling may be system‐specific and requires further study into the generality of the phenomenon as well as the nature of the cues involved.  相似文献

Encrusting algae are well-known to be able, for long periods, to withstand shading and overgrowth by other organisms. How this is achieved remains a mystery. It had been proposed that connections with unshaded (non-overgrown) parts of the thallus may allow transfer of nutrients to the shaded part. From this model, I proposed and tested the hypothesis that shaded patches of the intertidal red alga, Hildenbrandia rubra, would survive overgrowth longer, or better, when connected to unshaded thallus than when experimentally separated from surrounding alga. Experimental treatments were shading (black or transparent 80 mm perspex discs or no cover) and scraping (scraped around the disc to remove contacts, a control for effects of scraping, no treatment). The 9 orthogonal combinations of cover and scraping were applied to 3 independent, random replicates (i.e. 27 plots) in each of four randomly chosen sites.

In all 4 sites, over 13 months, shaded H. rubra survived in greater abundance (as % cover) where in contact with surrounding thallus. In one site, there was no effect of shading unless the thallus was isolated. In two sites, shading reduced cover, but was more deleterious where the thallus was isolated. In the fourth site, there were artefacts due to a perspex cover, but still less cover of alga where it was isolated.

This encrusting alga can withstand a long period of complete shading, provided there is connection to unshaded thallus. Interpreting or predicting overgrowth interactions in terms of competitive outcomes is therefore dependent on consideration of whether the overgrown species is actually being affected. It also depends on the duration of overgrowth and, as shown here, the extent to which connectivity with unshaded thallus is effective at preventing or reducing any consequences. Observations and experiments that do not ascertain these are difficult to interpret.  相似文献

Costs of resistance are expected to contribute to the maintenance of genetic variation for resistance in natural host populations. In the present study, we experimentally test for genetic trade-offs between parasite resistance and larval competitive ability expressed under varying levels of crowding and temperature. Artificial selection for increased behavioral resistance was applied against an ectoparasitic mite (Macrocheles subbadius) in replicate lines of the fruit fly Drosophila nigrospiracula. We then measured correlated responses to selection in larval competitive ability by contrasting replicate selected and control (unselected) lines in the absence of parasitism. Experiments were conducted under variable environmental conditions: two temperatures and three levels of larval density. Our results reveal a negative genetic correlation between resistance and larval-adult survival under conditions of moderate and severe intra-specific competition. At both low and high temperature, percent emergence was significantly higher among control lines than selected lines. This divergence in larval competitive ability was magnified under high levels of competition, but only at low temperature. Hence, the interaction between selection treatment and larval density was modified by temperature. As predicted, larvae experiencing medium and high levels of competition exhibited an overall reduction in female body size compared to larvae at low levels of competition. Female flies emerging from selected lines were significantly smaller than those females from control lines, but this effect was only significant under conditions of moderate to severe competition. These results provide evidence of environment-dependent trade-offs between ectoparasite resistance and larval competitive ability, a potential mechanism maintaining genetic polymorphism for resistance.  相似文献

Insulin resistance is commonly observed in patients prior to the development of type 2 diabetes and may predict the onset of the disease. We tested the hypothesis that impairment in insulin stimulated glucose-disposal in insulin resistant patients would be reflected in the gene expression profile of skeletal muscle. We performed gene expression profiling on skeletal muscle of insulin resistant and insulin sensitive subjects using microarrays. Microarray analysis of 19,000 genes in skeletal muscle did not display a significant difference between insulin resistant and insulin sensitive muscle. This was confirmed with real-time PCR. Our results suggest that insulin resistance is not reflected by changes in the gene expression profile in skeletal muscle.  相似文献

We compared the activity of human recombinant alpha and gamma interferons (IFNs) on normal T lymphocytes and various T cell lines. IFN gamma, unlike IFN alpha, did not promote the antiviral state in these cells, or induce the activity of 2'-5' oligoadenylate synthetase. The lack of antiviral effect was observed using an RNA virus (VSV) and a DNA virus (HSV, type 1) as challenger viruses.  相似文献

Developing arbovirus resistance in mosquitoes   总被引:4,自引:0,他引:4
Diseases caused by arthropod-borne viruses are increasingly significant public health problems, and novel methods are needed to control pathogen transmission. The hypothesis underlying the research described here is that genetic manipulation of Aedes aegypti mosquitoes can profoundly and permanently reduce their competence to transmit dengue viruses to human hosts. Recent key findings now allow us to test the genetic control hypothesis. We have identified viral genome-derived RNA segments that can be expressed in mosquito midguts and salivary glands to ablate homologous virus replication and transmission. We have demonstrated that both transient and heritable expression of virus-derived effector RNAs in cultured mosquito cells can silence virus replication, and have characterized the mechanism of RNA-mediated resistance. We are now developing virus-resistant mosquito lines by transformation with transposable elements that express effector RNAs from mosquito-active promoters.  相似文献

1. Although spiders can colonise ecosystems by air, dispersal capabilities differ among spider species. Web‐building spiders are thought to balloon at higher rates than hunting spiders. Spider success in agricultural systems may also depend on habitat preferences. Few studies have examined the success of aerially dispersing spiders in crop systems, and information about the dispersal capabilities of spiders in putative source habitats is limited. 2. Spiders were monitored in the air and on the foliage of vineyards and adjacent oak woodland in order to compare the aerial spider faunas between these disparate habitats and to determine whether highly dispersive species contributed disproportionately to the spider community in vineyards. 3. The results show that most aerially dispersing spiders in both habitats were web‐building dwarf spiders, Erigone spp. (Linyphiidae), although hunting spiders were also well represented in the air, especially in oak woodland. Most woodland spiders in the air appeared to be residents of oak woodland and probably dispersed only short distances. 4. Conversely, only a subset of the aerial spider fauna established in vineyards in high numbers. Spiders that dominated the aerial fauna were under‐represented on vineyard foliage, whereas several hunting spiders dispersed aerially at low rates but dominated vineyard spider composition. 5. These results suggest that aerial dispersal ability may allow spiders to reach crop systems, but that establishment depends on habitat preferences and/or competitive ability.  相似文献

Voice pitch, the perceived “highness” or “lowness” of a voice, influences how humans perceive and treat each other in various ways. One example is the selection of leaders. A growing number of studies, both experimental and observational, show that individuals with lower-pitched voices are more likely to win elected office. This leads to the yet untested question of whether individuals with lower voices are actually better leaders. That is, is voice pitch a reliable signal of leadership ability? Here we address this question with an observational study of the vocal pitch and leadership ability of elected officials, and an experiment where subjects were asked to respond to persuasive political policy statements made by speakers with different pitched voices. Both studies lead to the same conclusion: voice pitch does not correlate with leadership ability.  相似文献

Obesity is associated with inflammation and increased expression of suppressor of cytokine signaling (SOCS) proteins, which inhibit cytokine and insulin signaling. Thus, reducing SOCS expression could prevent the development of obesity-induced insulin resistance. Using SOCS-1 knockout mice, we investigated the contribution of SOCS-1 in the development of insulin resistance induced by a high-fat diet (HFD). SOCS-1 knockout mice on HFD gained 70% more weight, displayed a 2.3-fold increase in epididymal fat pads mass and increased hepatic lipid content. This was accompanied by increased mRNA expression of leptin and the macrophage marker CD68 in white adipose tissue and of SREBP1c and FAS in liver. HFD also induced hyperglycemia in SOCS-1 deficient mice with impairment of glucose and insulin tolerance tests. Thus, despite the role of SOCS proteins in obesity-related insulin resistance, SOCS-1 deficiency alone is not able to prevent insulin resistance induced by a diet rich in fat.  相似文献

Roux-en-y gastric bypass (RYGB) surgery rapidly improves glucose tolerance and reverses insulin resistance in obese patients. It has been hypothesized that this effect is mediated by the diversion of nutrients from the proximal small intestine. We utilized duodenal-jejunal bypass (DJB) as a modification of gastric bypass to determine the effect of nutrient diversion from the foregut without gastric restriction on insulin resistance in obese rats. The effects of DJB or Sham surgery on glucose homeostasis were determined in both high-fat-fed Long-Evans and Wistar rats. Body weight and food intake were measured weekly postoperatively, and body composition was monitored before and after surgery. Glucose tolerance was tested before and as early as 1 month postoperation; additionally, in Wistar rats, insulin sensitivity was determined by a hyperinsulinemic-euglycemic clamp (HIEC). DJB did not affect body weight, body composition, glucose tolerance, or insulin concentrations over the period of the study. The average glucose infusion rate (GIR) during the HIEC was 6.2 ± 1.16 mg/kg/min for Sham rats compared to 7.2 ± 1.71 mg/kg/min for DJB rats (P = 0.62), and neither endogenous glucose production (EGP; P = 0.81) nor glucose utilization (glucose disappearance (R(d)), P = 0.59) differed between DJB and Sham rats. DJB does not affect insulin resistance induced by a high-fat diet in Long-Evans and Wistar rats. These data suggest that duodenal bypass alone is an insufficient mechanism to alter insulin sensitivity independent of weight loss in obese, nondiabetic rodents.  相似文献

Drosophila melanogaster can be artificially selected for increased resistance against parasitoid wasps that attack the larvae. Lines selected for greater resistance are poorer larval competitors under conditions of resource scarcity. Here we investigated the mechanistic basis of this apparent trade-off. We found that resistant lines have approximately twice the density of haemocytes (blood cells) than that of controls. Haemocytes are involved in encapsulation, the chief cellular immune defence against parasitoids. We have previously shown that resistant lines feed more slowly than controls and hypothesize that limiting resources are being switched from trophic to defensive functions.  相似文献

The Acacia drepanolobium (also known as Vachellia drepanolobium) ant‐plant symbiosis is considered a classic case of species coexistence, in which four species of tree‐defending ants compete for nesting space in a single host tree species. Coexistence in this system has been explained by trade‐offs in the ability of the ant associates to compete with each other for occupied trees versus the ability to colonize unoccupied trees. We seek to understand the proximal reasons for how and why the ant species vary in competitive or colonizing abilities, which are largely unknown. In this study, we use RADseq‐derived SNPs to identify relatedness of workers in colonies to test the hypothesis that competitively dominant ants reach large colony sizes due to polygyny, that is, the presence of multiple egg‐laying queens in a single colony. We find that variation in polygyny is not associated with competitive ability; in fact, the most dominant species, unexpectedly, showed little evidence of polygyny. We also use these markers to investigate variation in mating behavior among the ant species and find that different species vary in the number of males fathering the offspring of each colony. Finally, we show that the nature of polygyny varies between the two commonly polygynous species, Crematogaster mimosae and Tetraponera penzigi: in C. mimosae, queens in the same colony are often related, while this is not the case for T. penzigi. These results shed light on factors influencing the evolution of species coexistence in an ant‐plant mutualism, as well as demonstrating the effectiveness of RADseq‐derived SNPs for parentage analysis.  相似文献
