Clostridium perfringens produces disease in sheep, goats and other animal species, most of which are generically called enterotoxemias. This micro-organism can be a normal inhabitant of the intestine of most animal species including humans, but when the intestinal environment is altered by sudden changes in diet or other factors, C. perfringens proliferates in large numbers and produces several potent toxins that are absorbed into the general circulation or act locally with usually devastating effects on the host. History, clinical signs and gross post-mortem findings are useful tools for establishing a presumptive diagnosis of enterotoxaemia by C. perfringens in sheep and goats, although no definitive diagnosis of these diseases can be made without laboratory confirmation. Because all types of C. perfringens can be normal inhabitants of the intestine of most animals, culture of this micro-organism from intestinal contents of animals has no diagnostic value unless a colony count is performed and large numbers (usually more than 10(4)-10(7)CFU/g) of C. perfringens are found. The most accepted criterion in establishing a definitive diagnosis of enterotoxaemia by C. perfringens is the detection of its toxins in intestinal contents. However, some of the major toxins of C. perfringens (i.e. epsilon toxin) can also be found, albeit in small amounts, in the small intestine of clinically normal sheep, and this poses a diagnostic challenge. In such cases the histopathology of the brain must be used as an alternative diagnostic tool, since the lesions produced by epsilon toxin in the brains of sheep and goats are unique and pathognomonic for C. perfringens type D enterotoxaemia. Ancillary tests, such as measurement of urine glucose or observation of Gram stained smears of intestinal mucosa can be used and, although they have a presumptive diagnostic value when positive, they cannot be used to rule out a diagnosis of enterotoxaemia if they are negative. In conclusion, the diagnosis of C. perfringens infections in animals is complex and it is appropriate to rely on a combination of diagnostic techniques rather than one singe test.  相似文献

While nuclear pediatrics, as well as radio-pediatrics for radiology, should be considered as a full specialty of nuclear medicine, oncology applications also deserve special consideration. The child indeed is not a miniature adult, and especially in oncology, we must take into account the specificity of various diseases, the management of pain, and the fundamental psychological involvement of the disease in the child but also in his family. Cerebrovascular diseases are significant causes of death and disability in humans. Improvements in diagnostic and therapeutic approaches strongly rely on adequate gyrencephalic, large animal models being demanded for translational research. Ovine stroke models may represent a promising approach but are currently limited by insufficient knowledge regarding the venous system of the cerebral angioarchitecture. The present study was intended to provide a comprehensive anatomical analysis of the intracranial venous system in sheep as a reliable basis for the interpretation of experimental results in such ovine models. We used corrosion casts as well as contrast-enhanced magnetic resonance venography to scrutinize blood drainage from the brain. This combined approach yielded detailed and, to some extent, novel findings. In particular, we provide evidence for chordae Willisii and lateral venous lacunae, and report on connections between the dorsal and ventral sinuses in this species. For the first time, we also describe venous confluences in the deep cerebral venous system and an ‘anterior condylar confluent’ as seen in humans. This report provides a detailed reference for the interpretation of venous diagnostic imaging findings in sheep, including an assessment of structure detectability by in vivo (imaging) versus ex vivo (corrosion cast) visualization methods. Moreover, it features a comprehensive interspecies-comparison of the venous cerebral angioarchitecture in man, rodents, canines and sheep as a relevant large animal model species, and describes possible implications for translational cerebrovascular research.  相似文献

Diploid ox nuclei contain about 14% more DNA than nuclei from sheep of the same sex. Goat nuclei have a similar DNA content to those of sheep. In view of the similar chromosome banding patterns in these species, it appears that chromosome evolution must have involved numerous minute interstitial deletions or additions of DNA. Although chromosomes which have similar banding patterns in these three species may be regarded as homologous in this respect, and can be regarded as having a common evolutionary origin, they are not homologous for the quantity of their DNA.  相似文献

Mixed-species stocking can foster sound landscape management while offering economic and ecological advantages compared with mono-species stocking. Producers contemplating a mixed-species enterprise should reflect on several considerations before implementing this animal management strategy. Factors applicable to a particular producer's landscape must be considered together with goals and economic constraints before implementing mixed-species stocking. A major consideration when using mixed-species stocking is how to deal with predation losses, especially among small ruminants. An approach being adopted in some commercial operations capitalizes on using innate animal behaviors to form cohesive groups of two or more livestock species that consistently remain together under free-ranging conditions. These groups are referred to as flerds. The mixing of a flock of sheep and/or goats with a herd of cattle into a flerd has been shown to protect sheep and goats from coyote predation, as well as offering other husbandry advantages. Some of the added advantages include more efficient conversion of forage into animal protein. Creation of flerds, their maintenance and advantages are discussed.  相似文献

In a series of experiments it was demonstrated that highly increased activities of aspartate aminotransferase (= GOT), total lactate dehydrogenase (LDH), and α-hydroxybutyrate dehydrogenase (HBD) occurred in serum of sheep on herring meal feeding. The alanine aminotransferase (A1AT = GPT) level remained unchanged. The enzyme increase was apparently not related to the liver toxic agent dimethylnitrosamine (DMNA) occasionally occurring in lethal doses in meals produced from raw materials preserved with excesses- of nitrite. Histological changes of the liver or other tissues were never detected in experimental animals killed at a stage with very high serum enzyme values. Diets equivalent in digestible crude protein consisting of vegetable as well as of animal protein sources other than fish meal, did not give rise to elevated serum enzyme values. Electrophoretic separation of the LDH isoenzymes in serum of herring meal-fed sheep showed an increased percentage of the LDH1 fraction, which is predominant in liver, heart, and kidney. Determination of enzyme activities in various tissues resulted in a markedly higher concentration in livers from herring meal-fed animals than in sheep fed casein at an equal protein level. It is suggested that herring meal may have a special promoting effect on the de novo synthesis of the enzymes concerned. As a practical consequence of the experiments it must be emphasized that serum determinations of these enzymes for diagnostic purposes will give a false picture of the clinical condition of sheep fed even moderate amounts of herring meal.  相似文献

Soay sheep on the island of Hirta exhibit periodic population collapses that have been proposed to result from nonlinear interactions between weather, population density, and age structure. Here we employ a diagnostic approach to reanalyze the data from 1985 to 2004 and find that climate mainly affects the equilibrium population size, thus acting as a lateral perturbation. From this, we derive a simple energetic model for a population interacting with its food supply in the presence of variable winter weather. This model explains the strong nonlinearity in the Soay sheep population regulation function and provides a framework for evaluating climatic perturbations. We examined two integrative climatic indexes, one representing effects on forage production and the other representing the severity of winter weather. Results suggest that the latter has the main effect on Soay sheep population dynamics. Models incorporating this variable provided fairly accurate predictions of Soay sheep population fluctuations. The diagnostic approach offers an objective way to develop simple, nonstructured population models that are useful for understanding the causes of population fluctuations and predicting population changes, provided they are based on a careful consideration of the underlying biological and/or ecological mechanisms.  相似文献

Wild sheep (Ovis orientalis) as the true ancestor of domestic sheep (Ovis aries) is currently listed as vulnerable (VU) by IUCN. Effective conservation of this species requires collecting all available ecological knowledge into a single framework that could be used for management decision making. Use of Bayesian Belief Networks for such purposes have been advocated over recent decades as this approach can integrate different sources of knowledge and perform comprehensive analyses. We built a decision support tool using BBN to assist in habitat management of wild sheep populations throughout the species’ geographical range. The behaviour of the model was tested using scenario and sensitivity analysis. Habitat security, food and water suitability, and thermal cover were recognised as the main habitat variables determining wild sheep habitat suitability. Integrating the complex interactions between variables, the model can be applied for both diagnostic and predictive analyses answering “what if” and “how” questions. This approach can assist wildlife conservationists for building similar models for other poorly-studied threatened species.  相似文献

BackgroundBrucellosis is a neglected zoonosis endemic in many countries, including regions of sub-Saharan Africa. Evaluated diagnostic tools for the detection of exposure to Brucella spp. are important for disease surveillance and guiding prevention and control activities.Methods and findingsBayesian latent class analysis was used to evaluate performance of the Rose Bengal plate test (RBT) and a competitive ELISA (cELISA) in detecting Brucella spp. exposure at the individual animal-level for cattle, sheep, and goats in Tanzania. Median posterior estimates of RBT sensitivity were: 0.779 (95% Bayesian credibility interval (BCI): 0.570–0.894), 0.893 (0.636–0.989), and 0.807 (0.575–0.966), and for cELISA were: 0.623 (0.443–0.790), 0.409 (0.241–0.644), and 0.561 (0.376–0.713), for cattle, sheep, and goats, respectively. Sensitivity BCIs were wide, with the widest for cELISA in sheep. RBT and cELISA median posterior estimates of specificity were high across species models: RBT ranged between 0.989 (0.980–0.998) and 0.995 (0.985–0.999), and cELISA between 0.984 (0.974–0.995) and 0.996 (0.988–1). Each species model generated seroprevalence estimates for two livestock subpopulations, pastoralist and non-pastoralist. Pastoralist seroprevalence estimates were: 0.063 (0.045–0.090), 0.033 (0.018–0.049), and 0.051 (0.034–0.076), for cattle, sheep, and goats, respectively. Non-pastoralist seroprevalence estimates were below 0.01 for all species models. Series and parallel diagnostic approaches were evaluated. Parallel outperformed a series approach. Median posterior estimates for parallel testing were ≥0.920 (0.760–0.986) for sensitivity and ≥0.973 (0.955–0.992) for specificity, for all species models.ConclusionsOur findings indicate that Brucella spp. surveillance in Tanzania using RBT and cELISA in parallel at the animal-level would give high test performance. There is a need to evaluate strategies for implementing parallel testing at the herd- and flock-level. Our findings can assist in generating robust Brucella spp. exposure estimates for livestock in Tanzania and wider sub-Saharan Africa. The adoption of locally evaluated robust diagnostic tests in setting-specific surveillance is an important step towards brucellosis prevention and control.  相似文献

This paper evaluates the long‐term effect of an ecological network of calcareous grasslands, a habitat type that experienced dramatic habitat loss and fragmentation during the 20th century, on species richness of habitat specialist plants. Calcareous grasslands are of special conservation concern as the habitat type with the highest diversity in plant and invertebrate species in central Europe. A baseline survey in 1989 established complete vascular plant species lists for all 62 previously abandoned calcareous grassland patches in the study area and assessed the presence of 48 habitat specialist plant species. An ecological network was initiated in 1989 to reconnect these patches with existing grazed pastures (core areas) through large flock sheep herding where feasible, as sheep are thought to be the primary dispersal vectors for calcareous grassland plants. An evaluation survey in 2009 showed significant increase in species richness of habitat specialist plants in patches reconnected by sheep herding, indicating successful colonizations by habitat specialist plants, while ungrazed patches showed no significant change. Observed increase in species richness between 1989 and 2009 was related to connectivity by sheep herding and the presence of a diversity of structural elements providing microsites for establishment. Baseline species richness of the patches, which had been abandoned since at least 1960, was associated with patch area, supporting the effect of ecological drift, and with vegetation type, which suggests that delays in extinction may be related to site factors governing the strength of competition with later seral species. The implementation of this ecological network represents a long‐term ‘natural experiment’ with baseline data, manipulation, and evaluation of hypothesized effects on a clearly defined target variable. It thus provides much needed empirical evidence that species loss in fragmented calcareous grassland communities can be counteracted by restoring functional connectivity among remnant patches.  相似文献

Sheep epizoochory has often been proposed as an important vector which can help to overcome the dispersal limitation of plants in fragmented landscapes. In order to evaluate the contribution of herbivores to recruitment especially of target species, the dispersal and post-dispersal fate of such seeds must be known. In a field experiment sheep with seeds of mainly target species (experimentally attached to their coats) were present at three sand plots for 24 h. Natural epizoochorous dispersal was already shown for most of the species in our study area. Seed detachment, trampling intensity and seed shadow were measured; seedling emergence and survival were recorded over an 8-month period. In addition, the effect of sheep trampling on seedling emergence and survival of two threatened species, Jurinea cyanoides and Koeleria glauca, were studied.  相似文献

Experimental infections of calves were carried out with either isolates of predominantly Ostertagia ostertagi, pure O. leptospicularis or a mixed isolate of equal numbers of both these species. The total worms established on day 21 for the mixed species from a total inoculum of 100 000 infective larvae, was 1.2 times greater than from 100 000 larvae of the O. ostertagi isolate and 3.3 times that of the pure O. leptospicularis isolate. The increased establishment in the mixed inoculum referred to both O. ostertagi and O. leptospicularis (days 17 and 21). These differences were both highly significant (P less than 0.01). The severity of the pathological changes was also greater in the mixed infections. It is suggested that these findings must be taken into account when control measures involving alternate grazing of sheep and cattle are being employed.  相似文献

Single-strand conformational polymorphism analysis and DNA sequencing were used to characterize Mhc-DRB second exon variability in the Latxa and Karrantzar breeds of sheep. The presence of more than two sequences in some animals indicates that alleles of two different loci have been amplified. Six new alleles were identified by sequencing. The allele frequency distribution of the DRB1 gene is striking, with two alleles accounting for half of the gene pool in both breeds under study. The most frequent allele in both breeds was the same (named DRB1*0702), with some specific amino acids: Tyr in position 31 and Thr in 51. A species variability analysis was also performed including the entire set of sheep DRB exon 2 sequences. Based on the patchwork patterns of different alleles, interallelic recombination appears to be playing a significant role in the generation of allelic diversity at this locus in sheep. The phylogenetic tree of all known Caprinae DRB sequences shows that certain alleles from one species are more closely related to those from other species than they are to each other. Allele DRB1*0702 merits special attention due to its high similarity to the Mufflon allele. As this is the most frequent in both breeds analyzed, one can hypothesize that in sheep, both Mufflon and Argali have had different influences depending on the sheep breed under study and that the relationship between domestic sheep and Mufflon is greater than previously thought. The data generated in this study can serve as a basis for developing a typing assay for the sheep DRB genes in the Latxa and Karrantzar populations.  相似文献

In animals, the response to decompression scales as a power of species body mass. Consequently, decompression sickness (DCS) risk in humans should be well predicted from an animal model with a body mass comparable to humans. No-stop decompression outcomes in compressed air and nitrogen-oxygen dives with sheep (n = 394 dives, 14.5% DCS) and humans (n = 463 dives, 4.5% DCS) were used with linear-exponential, probabilistic modeling to test this hypothesis. Scaling the response parameters of this model between species (without accounting for body mass), while estimating tissue-compartment kinetic parameters from combined human and sheep data, predicts combined risk better, based on log likelihood, than do separate sheep and human models, a combined model without scaling, and a kinetic-scaled model. These findings provide a practical tool for estimating DCS risk in humans from outcomes in sheep, especially in decompression profiles too risky to test with humans. This model supports the hypothesis that species of similar body mass have similar DCS risk.  相似文献

红皮云杉林(Picea koraiensis Forest Alliance)是以红皮云杉为群落共优势种的森林植被类型,具有结构复杂及物种多样性高等特点。在遵从《中国植被志》研编规范的基础上,提出了红皮云杉林新的分类系统,包括5个群丛组8个群丛,分别归属于常绿针叶林、落叶与常绿针叶混交林和针叶与阔叶混交林3个植被型。群丛组分类主要基于群落层片分化,以及乔木层的共优势种和特征种的差异,并以常绿针叶树层片重要值的66%为阈值划分常绿针叶林和针叶与阔叶混交林。在群丛分类中,通过双向指示种分析筛选特征种,综合考虑群落生境和群落演替阶段等因素确定分类方案。该分类方案是《中国植被志》研编规范的一个应用示例,对植被分类工作的借鉴意义表现在4个方面。(1)作为植被分类的重要凭证,样方数据质量控制是植被分类工作的重要环节;其中,物种的准确鉴定对确保植被分类方案的合理性至关重要。(2)对于乔木层物种组成丰富、优势种不明显的群落,群落层片分化和特征种是植被类型划分的重要依据。(3)以特定物种组合为共建种所组成的多个植被类型可归属为不同的植被型。(4)森林采伐等人类活动可能对植被与环境的关系形成干扰。因此,在...  相似文献

Vector-borne diseases are of global importance to human and animal health. Empirical trials of effective methods to control vectors and their pathogens can be difficult for practical, financial and ethical reasons. Here, therefore, we use a mathematical model to predict the effectiveness of a vector-borne disease control method. As a case study, we use the tick-louping ill virus system, where sheep are treated with acaricide in an attempt to control ticks and disease in red grouse , an economically important game bird. We ran the model under different scenarios of sheep flock sizes, alternative host (deer) densities, acaricide efficacies and tick burdens. The model predicted that, with very low deer densities, using sheep as tick mops can reduce the tick population and virus prevalence. However, treatment is ineffective above a certain threshold deer density, dependent on the comparative tick burden on sheep and deer. The model also predicted that high efficacy levels of acaricide must be maintained for effective tick control. This study suggests that benignly managing one host species to protect another host species from a vector and pathogen can be effective under certain conditions. It also highlights the importance of understanding the ecological complexity of a system, in order to target control methods only under certain circumstances for maximum effectiveness.  相似文献

ABSTRACT

Photoperiodic treatments have been of practical interest in controlling seasonal reproduction in sheep, goats and horses. Melatonin is the principal mediator of the environmental photoperiodic message. To investigate the intra- and inter-subject variability of melatonin 24 h rhythm, ten female Italian Saddle horses (8–10 yrs old, mean body weight 525 ± 30 kg), ten female Sarda breed sheep (2–3 yrs old, mean body weight 40.5 ± 2.8 kg) and ten female Sarda breed goats (3–4 yrs old, mean body weight 38.9 ± 4.1 kg), housed individually in a 4 × 4 m soundproof box equipped with 50 × 100 cm opening windows, were subjected to a natural photoperiod of the vernal equinox (sunrise 06:00 h; sunset 18:00 h). Blood samples were collected from each animal, every 3 h over a 48 h period starting at 00:00 h of day 1 and ending at 00:00 h of day 3. Plasma melatonin concentrations were determined by direct radioimmunoassay (MelatoninDirect RIA, Labor Diagnostika Nord GmbH, Nordhorn, Germany). The application of single cosinor method substantiated a circadian rhythm of melatonin with a nocturnal peak in all studied species. The application of two-way ANOVA on the rhythmic parameters indicated statistically significant differences between the three species in all of the cosinor analysis-derived parameters of MESOR, amplitude, acrophase and robustness of rhythm. Analyses of intra- and inter-subject variability indicate that organization of the melatonin 24 h rhythm is characterized by great accuracy of control within and between the individuals of a breed. In conclusion, features of the 24 h rhythm of melatonin among species; however, the 24 h rhythmicity of melatonin each species showed high stability within the various subjects and within the same subject. Livestock grazing is a common management practise in semi-natural grasslands in Central Europe. Different types of livestock (horses, cattle, sheep) and grazing intensity are known to affect the richness and composition of plant species. However, knowledge of grazing-dependent effects on invertebrates is limited. We examined the influence of horse, cattle and sheep grazing on the richness, abundance and composition of land snail species in 21 calcareous nutrient-poor grassland areas in the northwestern Jura Mountains, Switzerland. Grazing by different livestock species did not affect the species richness, abundance and species composition of land snails. Furthermore, the number of open-land species and the ratio of large- to small-sized snail species or individuals did not differ among the three pasture types. However, independent of livestock species, grazing intensity negatively influenced the snail fauna. Snail species richness, abundance and number of Red list species decreased with increasing grazing intensity. Grazing intensity also affected the occurrence of individual snail species (Truncatellina cylindrica, Cecilioides acicula, Candidula unifasciata and Trichia plebeia). To preserve the snail fauna in nutrient-poor grasslands, pastures can be stocked with horses, cattle or sheep. However, both maximum stocking rate (number of livestock units per hectare) and grazing duration (number of grazing days per year) must be carefully defined for the proper management of the pastures.  相似文献

The protozoan parasite Toxoplasma gondii is the causative agent of the zoonosis toxoplasmosis. In sheep and goats, it is one of the most prevalent causes of infectious abortion. Also in pregnant women, a primary infection can result in miscarriage. Humans acquire the infection either by ingestion of oocysts excreted by cats, the definitive host of the parasite, or by eating raw or undercooked meat from latently infected animals (Dubey & Beattie 1988). In Sweden, toxoplasmosis is a notifiable disease, and cases of clinical disease in humans as well as animals must be reported. In both veterinary and human medicine serological assays based on detecting the humoral antibody response of the host against the parasite are used as diagnostic tools. So far, solid phase assays, such as the indirect fluorescent antibody test (IFAT) and the enzyme-linked immunosorbent assay (ELISA), have been widely used to diagnose T. gondii infection in many species including cats, pigs and sheep (Dubey & Beattie 1988). However, both IFAT and ELISA require appropriate anti-species specific immunoglobulins (Ig) that must be carefully evaluated for each species prior to use. This makes these assays complicated and time consuming. Consequently, alternative, simpler methods that do not require specific antisera would be of great value. The direct agglutination test (DA), which is based on the principle that formalin-treated organisms agglutinate in the presence of specific IgG antibodies, is such an assay (Fulton & Turk 1959). The DA-test is widely used in human medicine as a screening test for T gondii infection but it has not yet been thoroughly evaluated for use in veterinary medicine (Uggla & Buxton 1990).  相似文献

We have studied binding and membrane transfer rates of unsaturated long-chain fatty acids in sheep red cells, as previously done for human red cells, in order to elucidate the transport mechanism. Observed differences must be assigned to the different composition of the membrane in the two species. Equal surface areas of the membranes of the two species have similar binding capacities and affinities for palmitic-, linoleic-, oleic- and arachidonic acid at 37°C. The competitive bindings of linoleic- and arachidonic acid as well as the distribution of bound arachidonic acid on the two sides of the membrane are not different in the two species. However, the rate constants for membrane transfer in sheep are less than half of those measured previously for human ghosts. This finding is confirmed by the exchange efflux kinetics of ghosts containing albumin-bound fatty acid. Studies of sheep ghost membranes with oleic-, arachidonic- and linoleic acid reveal a proportionality between the membrane transfer rate constants and the number of fatty acid double bonds, as found previously for human ghost membrane, and the effect of double bonds is in harmony with a large negative activation entropy for diffusion through the membrane. The established replacement of lecithin by sphingomyelin with a low unsaturation fatty acid index in sheep membranes probably causes a lower transversal lipid phase fluidity. Double bonds diminish the flexibility of hydrocarbon chains and thus the large negative activation entropy of diffusion across the membrane. The smaller transfer rate constants of the three unsaturated fatty acids in sheep membranes support the hypothesis that the transfer is diffusion in protein defined annular lipid domains and not carrier mediated.
