The involvement of cortical microtubules in the control of plant cell expansion was studied in the Arabidopsis root epidermis. In the zone of fast elongation microtubules were transverse to the root axis in all epidermal cells. However when cells entered the differentiation zone cell type-specific microtubule reorientation took place. In the trichoblasts that were then approximately 130 µm long and formed the root hair bulge, the microtubules switched to a random distribution. In the adjoining atrichoblasts microtubules adopted a slightly oblique orientation. In more proximal parts of the differentiation zone atrichoblast microtubules were found in a more oblique and finally in a longitudinal orientation. Upon exposure to ethylene or 1-aminocyclopropane-1-carboxylic acid (ACC – the precursor of ethylene) at a saturating dose, cell elongation abruptly stopped. From then on trichoblast cells reached only a length of about 35 µm, and developed root hairs. Cortical microtubules changed orientation within 10 min. In trichoblasts they adopted the typical random orientation, in atrichoblasts however, they took up a longitudinal orientation. Microtubule reorientation was complete within 60 min. The possible role of microtubules in the control of cell elongation is discussed.  相似文献

Bean ( Phaseolus vulgaris L.) cell suspensions were adapted for growth in 12 µ M dichlobenil (2,6-dichlorobenzonitrile or DCB) by a stepwise increase in the concentration of the inhibitor in each subculture. Non-tolerant suspensions (I 50  = 0.3 µ M ) gave rise to single cells or small clusters while tolerant cell suspensions (I 50  = 30 µ M ) grown in DCB formed large clusters. The cells in these clusters were surrounded by a thick and irregular cell wall with a lamellate structure and lacking a differentiated middle lamella. Analysis of habituated cell walls by Fourier transform infrared spectroscopy and cell wall fractionation revealed: (1) a reduced amount of cellulose and hemicelluloses, mainly xyloglucan (2) qualitative and quantitative differences in pectin levels, and (3) a non-crystalline and soluble β-1,4-glucan. When tolerant cells were returned to medium lacking DCB, the size of the cell clusters was reduced; the middle lamella was only partly formed, and the composition of the cell wall gradually reverted to that obtained with non-tolerant cells. However, dehabituated cells (I 50  = 12 µ M ) were 40-fold more tolerant to DCB than non-tolerant cells and were only 2.5-fold more sensitive than tolerant cells.  相似文献

Broad-leaved dock, Rumex obtusifolius L. (Polygonaceae), is the main host plant of the green dock beetle, Gastrophysa viridula Degeer (Coleoptera: Chrysomelidae). Adult beetles are able to attach and walk on leaves of this plant. Leaf surface is rather uneven, because of irregularly shaped prominent epidermal cells with a maximum height of about 9 µm. The surface is covered with a smooth epicuticular wax layer having relatively low free surface energy (FSE). The aim of this study was to measure beetle attachment force applying a 'centrifugal technique' on adult insects on the leaf surface and other substrates, in order to understand the effect of surface architecture and its physicochemical properties on insect attachment. We compared forces on an adaxial leaf surface with forces on a smooth silanized glass plate having low FSE, and on a polishing paper having slightly lower FSE and similar surface roughness (asperities' size = 9 µm). Smooth plate made of normal untreated clean glass with high FSE was used as a reference substrate. On the leaf surface and the polishing paper, attachment forces were lower compared to both glass samples. There was no significant difference between force values on leaves and the polishing paper, but on both glass surfaces they were significantly higher than on the other substrates. Hydrophobicity alone explains a decrease in attachment force of the beetle, but when combined with roughness the decrease in force is four times greater.  相似文献

In basaltic glass from the southern Mid-Atlantic-Ridge conducive environmental conditions for biogenic weathering resulted in excellent preserved microbial morphologies on glass surfaces. The distinct glass interface and open spaces between palagonite sheet and glass indicate a dissolution-reprecipitation mechanism of glass alteration potentially supported by microorganisms. On internal fracture surfaces, branching channels with widths at 20–30 μm containing longish structures with targeted dissolution of the glass by growing tips were observed. Alteration resulted in enrichment of Fe, Ti, P, and K in palagonite in amorphous mineral forms.  相似文献

Sixteen species of Jaborosa (Solanaceae), including eight endemic to Argentina, were studied karyologically. The numbers n  = 12 and/or 2n  = 24 were found in all species, the majority of the cases being new reports. Mitotic chromosomes were small- to medium-sized, the average length varying from 2.95 to 4.93 µm. All species had one to three chromosome pairs with satellites. The karyotypes, obtained for 13 species, were slightly asymmetrical: A 1 ranged from 0.228 to 0.483, A 2 ranged from 0.095 to 0.182, and Paszko's asymmetry index ranged from 0.84 to 3.47. In all species, metacentric chromosomes were the most common, followed by submetacentrics, but subtelocentrics were rare. Morphological similarities and sectional arrangements were not reflected in either a principal components analysis plot or asymmetry index plot, but the species could be singled out by their karyotype formulae and the different karyotype parameters taken. In Jaborosa , a notably diversified genus, exo-morphological evolution has taken place, together with evident chromosome rearrangements, whose disposition is different and not as clear as in related genera.  © 2008 The Linnean Society of London, Botanical Journal of the Linnean Society , 2008, 156 , 467–478.  相似文献

Abstract: The human neuroblastoma cell line SK-N-BE expresses δ-opioid receptors negatively coupled to adenylyl cyclase. Prolonged treatment (2 h) of the cells with 100 n M etorphine leads to an almost complete desensitization (8.2 ± 5.9 vs. 45.8 ± 8.7% for the control). Other receptors negatively coupled to adenylyl cyclase, namely, D2-dopaminergic, α2-adrenergic, and m2/m4-muscarinic, were identified by screening of these cells, and it was shown that prolonged treatment (2 h) with 1 µ M 2-bromo-α-ergocryptine or 1 µ M arterenol resulted in a marked desensitization of D2-dopaminergic and α2-adrenergic receptors, respectively. Cross-desensitization experiments revealed that pretreatment with etorphine desensitized with the same efficiency the δ-opioid receptor and the D2-dopaminergic receptor, and pretreatment with 2-bromo-α-ergocryptine also desensitized both receptors. In contrast, pretreatment with etorphine desensitized only partly the α2-adrenergic receptor response, whereas pretreatment with 1 µ M arterenol partly desensitized the δ-opioid receptor response. It is concluded that the δ-opioid receptor-mediated inhibitory response of adenylyl cyclase undergoes heterologous desensitization, and it is suggested that δ-opioid and D2-dopaminergic receptors are coupled to adenylyl cyclase via a Gi2 protein, whereas α2-adrenergic receptor could be coupled to the enzyme via two G proteins, Gi2 and another member of the Gi/Go family.  相似文献

Pollen grains of six species of Gnetum , G. parvifolium , G. hainanense , G. luofuense , G. pendulum , G. cleistostachyum and G. montanum , collected from China were examined using light, scanning and transmission electron microscopy. Pollen grains of Gnetum are subspheroidal or irregular-apolar, inaperturate, 11.21–22.44 µm in long axis and 9.34–20.47 µm in short axis. The exine surface is covered with spinules, 0.50(0.30–0.71) µm long spaced on average 1.12(0.81–1.46) µm apart. The exine is about 0.55 µm thick and comprises ectexine and endexine. The ectexine includes a thin tectum and an infratectal granular layer. The tectum protrudes outwards, forming the spinules. The endexine is composed of discontinuous lamellae, with lacunae between lamellae. The pollen grains of Gnetum are compared with those of Ephedra and Welwitschia , and also those of the ANITA Group of angiosperms, including Amborellaceae, Nymphaeales, Illiciales, Trimeniaceae and Austrobaileyaceae. The exine ultrastructures of Gnetum , Ephedra and Welwitschia are quite similar, consisting of tectum, granular layer and lamellated endexine. The exine ultrastructure of Gnetum is also similar to that of Nymphaea colorata (Nymphaeaceae) in the transitional region between the proximal and distal poles, but differs from that of Amborellaceae, Illicium religiosum (Illiciaceae), Schisandra (Schisandraceae), Trimeniaceae and Austrobaileyaceae. Emerging diseases are increasing in incidence; therefore, understanding how pathogens are introduced into new regions and cause epidemics is of importance for the development of strategies that may hinder their spread. We used molecular data to study how a vector-borne banana virus, Banana bunchy top virus (BBTV), spread in Hawaii after it was first detected in 1989. Our analyses suggest that BBTV was introduced once into Hawaii, on the island of Oahu. All other islands were infected with isolates originating from Oahu, suggesting that movement of contaminated plant material was the main driving factor responsible for interisland spread of BBTV. The rate of mutation inferred by the phylogenetic analysis (1.4 × 10−4 bp/year) was similar to that obtained in an experimental evolution study under greenhouse conditions (3.9 × 10−4 bp/year). We used these values to estimate the number of infections occurring under field conditions per year. Our results suggest that strict and enforced regulations limiting the movement of banana plant material among Hawaiian islands could have reduced interisland spread of this pathogen.  相似文献

The ~2.63 Ga Carawine Dolomite, Hamersley Basin, Western Australia, preserves tube structures of probable microbial origin that formed in a low energy environment. The tubes are 0.4–1.8 cm in diameter and at least 10–16 cm long in outcrop. The tubes are defined by dark, 45-µm-thick dolomicritic walls, whereas the tube fill and host rock are composed of 30 µm, cloudy dolomite crystals and rare 170- to 425-µm-wide, dark well-sorted clasts. Closely spaced, rarely discontinuous laminae coat the insides of tubes; less closely spaced, peaked, discontinuous laminae coat the outsides of tubes. The laminae on the outsides of tubes are often intercalated with mammilate structures. The presence of probable microbial coatings on both the insides and the outsides of the tube walls requires that the tubes formed above the sediment–water interface. These tube structures probably formed during gas-charged fluid escape, similar to tubes observed in ancient and modern hydrocarbon seeps and cylindrical water transfer structures in sandstones. The laminae that coat the tubes have very similar geometries to modern biofilms that form in both turbulent and laminar flow, and their geometries probably reflect flow conditions during the fluid escape. The identification of these structures suggests that the preserved interaction between fluid escape and microbial growth in carbonates may be more common than previously thought.  相似文献

The mouse oviduct is covered by dense tracts of ciliated cells interspersed at random with occasional non-ciliated cells. Correlation between scanning electron microscopy and thin section images indicates that in the isolated fimbria most cilia are short (5 µm) and inactive, resting at the end of a uterad-directed effective stroke. These cilia terminate in a 9S−2 tip, the microtubules ending in an electron-dense plaque underneath the cell membrane. At the tip of the cilium a crown of fine extracellular hairs is attached to the ciliary membrane. In the ampulla and isthmus the ciliated cells decrease progressively in number and appear to lie in crypts.  相似文献

A new rhodymeniacean species, Chamaebotrys prolifera , is described from a shallow water habitat in Puerto Rico, representing the first occurrence of the genus in the Atlantic Ocean. Plants, to 5 cm across, are decumbent and comprised of compressed vesicles that are originally proliferously branched at the perimeter. Older vesicles become branched from their dorsal surfaces as well. Branches are septate at their origin and become irregularly shaped with age. Anastomoses between adjacent vesicles is common. Individual vesicles measure to 15 mm in broadest dimension. Vesicle walls consist of two layers of medullary cells and two layers of cortical cells. Tetrasporangia, which occur in diffuse nemathecia, are spherical, to 30 µm in diameter and are cut off terminally from an inner cortical cell. Cystocarps are hemispherical measuring to 800 µm in diameter and 350 µm in height. Spermatangia are apparently cut off randomly from outer cortical cells across the thallus surface. Molecular evidence confirms placement of Chamaebotrys within the Rhodymeniaceae.  相似文献

Two types of filamentous microfossil are preserved within vessel elements and rays of pyritized and partly carbonized twigs from the Lower Eocene London Clay. The first type, probably a Streptomyces -like actinomycete, is slender (<1 µm) with branches and some regular septation. Wider filaments (>2 µm) are fungal hyphae; no reproductive structures are preserved. These filamentous organisms probably started growing saprophytically after the death of the twigs; the fungi created lysis tracks on cell walls. In this paper we describe carbon-rich filamentous structures observed in association with the zeolite mineral phillipsite from sub-seafloor samples drilled and collected during the Ocean Drilling Program (ODP) Leg 197 at the Emperor Seamounts. The filamentous structures are ~5 µm thick and ~100–200 µm in length. They are found attached to phillipsite surfaces in veins and entombed in vein-filling carbonates. The carbon content of the filaments ranges between ~10 wt% C and 55 wt% C. They further bind to propidium iodide (PI), which is a dye that binds to damaged cell membranes and remnants of DNA.
Carbon-rich globular microstructures, 1–2 µm in diameter, are also found associated with the phillipsite surfaces as well as within wedge-shaped cavities in phillipsite assemblages. The globules have a carbon content that range between ~5 wt% C and 55 wt% C and they bind to PI. Ordinary globular iron oxides found throughout the samples differ in that they contain no carbon and do not bind to the dye PI. The carbon-rich globules are mostly concentrated to a film-like structure that is attached to the phillipsite surfaces. This film has a carbon content that ranges between ~25 wt% C and 75 wt% C and partially binds to PI. EDS analyses show that the carbon in all structures described are not associated with calcium and therefore not bound in carbonates. The carbon content and the binding to PI may indicate that the filamentous structures could represent fossilized filamentous microorganisms, the globules could represent fossilized microbial cells and the film-like structures could represent a microbially produced biofilm.
Our results extend the knowledge of possible habitable niches for a deep biosphere in sub-seafloor environments and suggests, as phillipsite is one of the most common zeolite mineral in volcanic rocks of the oceanic crust, that it could be a common feature in the oceanic crust elsewhere.  相似文献

Aim Eleutherodactylus coqui (commonly known as the coqui) is a frog species native to Puerto Rico and non‐native in Hawaii. Despite its ecological and economic impacts, its potential range in Hawaii is unknown, making control and management efforts difficult. Here, we predicted the distribution potential of the coqui on the island of Hawaii. Location Puerto Rico and Hawaii. Methods We predicted its potential distribution in Hawaii using five biophysical variables derived from Moderate Resolution Imaging Spectroradiometer (MODIS) as predictors, presence/absence data collected from Puerto Rico and Hawaii and three classification methods – Classification Trees (CT), Random Forests (RF) and Support Vector Machines (SVM). Results Models developed separately using data from the native range and the invaded range predicted potential coqui habitats in Hawaii with high performance. Across the three classification methods, mean area under the ROC curve (AUC) was 0.75 for models trained using the native range data and 0.88 for models trained using the invaded range data. We achieved the highest AUC value of 0.90 using RF for models trained with invaded range data. Main conclusions Our results showed that the potential distribution of coquis on the island of Hawaii is much larger than its current distribution, with RF predicting up to 49% of the island as suitable coqui habitat. Predictions also show that most areas with an elevation between 0 and 2000 m are suitable coqui habitats, whereas the cool and dry high elevation areas beyond 2000 m elevation are unsuitable. Results show that MODIS‐derived biophysical variables are capable of characterizing coqui habitats in Hawaii.  相似文献

Abstract: The effects of GABA on the kinetics of tert -[35S]butylbicyclophosphorothionate ([35S]TBPS) binding to the convulsant site of GABAA receptors were studied in membrane suspensions from the cerebral cortex of newborn (1-day-old) and adult (90-day-old) rats. TBPS dissociation was biphasic in neonates and adults, indicating that more than one interconvertible state of [35S]TBPS binding sites may be present in the cerebral cortex. In the absence of GABA, the fast ( t 1/2, 11 min) and slow ( t 1/2, 77 min) components of TBPS dissociation in newborn rats were approximately fourfold slower than in adults. The acceleration of the dissociation rates caused by 2 µ M GABA, however, was more robust in neonates than in adults (six- to ninefold vs. twofold increase, respectively). Moreover, the dissociation rates of TBPS in membranes preincubated with 2 µ M GABA (dissociation started by adding 40 µ M picrotoxin) were two- to fourfold slower than in membranes preincubated without GABA (dissociation started by adding 40 µ M picrotoxin plus 2 µ M GABA). Taken together, these results suggest that (1) the closed state of GABAA receptors is associated with a more effective steric barrier for the binding of TBPS in neonates compared with adults, (2) GABA produces a larger acceleration of the binding kinetics of TBPS in neonates than in adults, and (3) long incubations with GABA may cause receptor desensitization, which in turn slows down the dissociation rates of TBPS.  相似文献

Mineral weathering plays an important role in the global cycling of carbon and metals and there is an increasing realization that subsurface microbial activity may be a key factor controlling specific biogeochemical reactions and their rates. Methanobactin (mb) is an extracellular copper-binding compound excreted by methanotrophs to acquire copper for the regulation of methane oxidation. Bioavailable Cu regulates the expression and activity of pMMO vs. sMMO (particulate vs. soluble methane monooxygenase, respectively), key enzymes responsible for bacterial methane oxidation. In this study, we investigate the effect of mb on the dissolution of Cu-substituted borosilicate glass at low temperature and near neutral pH conditions, using batch dissolution experiments. Methanobactin promotes the dissolution of Cu-substituted glasses at rates faster than control experiments. Glasses with lower concentrations of copper (80 p.p.m.) or no copper are dissolved more rapidly by mb than those with more abundant copper (800 p.p.m.). Within the first 2 h of reaction, mb sorption onto glass surfaces limits mass transfer of Cu into solution, and at higher concentrations (100 µmol) of the ligand, inhibits dissolution rates of all glass formulations. These results suggest that both the concentration of mb in solution and the solid phase Cu concentration impact silicate weathering rates and related cycling of carbon in near-surface geological settings.  相似文献

Exotic tree plantations may serve as catalysts for native forest regeneration in agriculturally degraded landscapes. In 2001, we evaluated plant species regeneration in the understory of a 7‐year‐old experimental Eucalyptus saligna forest in Hawaii approximately 1 year after the cessation of 5 years of herbicide. These forests were organized in a 2 × 2–factorial design of planting density (1 × 1– or 3 × 3–m spacing) and fertilization (unfertilized control and regular fertilization), which resulted in varying resource availabilities. We found that understory biomass was highest under high light conditions, regardless of fertilization treatment, whereas species richness was lowest under fertilized 1 × 1–m plots. The understory was dominated by species exotic to Hawaii. The most common tree species, the noxious weed Citharexylum caudatum, was particularly successful because high light–saturated photosynthesis rates and a low light compensation point allowed for high growth and survival under both light conditions. To assess longer‐term recruitment patterns, we resurveyed a portion of this site in 2006 and also surveyed five Eucalyptus plantations in this region of Hawaii that differed in age (5–23 years), species (E. saligna, E. grandis, E. cloeziana, E. microcorys), and management (experimental, industrial, nonindustrial stewardship); all were established on previous agricultural sites within approximately 3 km of native‐dominated forest. Again, very few native species were present in any of the stands, indicating that within certain landscapes and for native species with certain life history traits, exotic plantations may be ineffective nursery ecosystems for the regeneration of native species.  相似文献

Phytochelatins (PCs) have been detected in a large range of plant species, but their role in heavy-metal tolerance is unclear. Various clones of Salix viminalis are differently tolerant to heavy metals, and the aim of this work was to investigate whether PCs were differently expressed in tolerant compared with sensitive clones. In a long-term study, five clones with high or low metal tolerance were cultivated 21 days without or with Cd (1 or 10 µ M ), Cu (0.3 or 7 µ M ), Ni (15 µ M ), Pb (7 µ M ) or Zn (10 or 100 µ M ). Controls were further used in a short-term study where Cd (1 µ M ) was added and samples were collected 0, 15 and 30 min, 1, 3 and 24 h after start of treatment. PCs were analysed on high performance liquid chromatography (HPLC) using two different methods: post-column derivatization using Ellmans reagent and pre-derivatization with monobromobimane. Thlaspi caerulescens treated with Cd was used as internal PC standard. No PCs could be detected in Salix with either of the two methods in any of the treatments: different clones, metals, concentrations, plant parts or treatment time. The 16 thiol peaks shown were the same in both control and treated plants. Both HPLC methods showed PC peaks when Thlaspi was used but these peaks could not be associated with any of the 16 peaks. The amino acid composition of the 16 peaks was not the expected composition of that of PCs. Thus, Salix viminalis have no detectable levels of PCs, which in turn are not involved in heavy metal tolerance in Salix .  相似文献

The seminal receptacle and the euspermatozoa and paraspermatozoa of Mysella bidentata were examined at an ultrastructural level and the results were compared with earlier findings of the same and other species of the Montacutidae. The euspermatozoon has a slender 13 µm long nucleus and a 1.1 µm long bullet-shaped acrosome. The acrosome of the paraspermatozoon is almost identical in ultrastructure to that of the euspermatozoa but is longer (1.9 µm) and more slender and is bent at an angle to the diminutive nucleus (1.1 µm long). The unpaired seminal receptacle is lined by a heavily ciliated epithelium and a non-ciliated epithelium with short and broad microvilli. Euspermatozoa only are stored in the receptacle. They are densely packed and orientated with their heads towards the non-ciliated epithelium. In this position they develop numerous extremely fine microvilli from the acrosome which apparently serve to attach them to the epithelial microvillar surface. Stored sperm may presumably remain functional for at least six months. A possible function of paraspermatozoa could be to clump sperm into sperm bags to keep them in suspension.  相似文献

Background

The tilapia family of the Cichlidae includes many fish species, which live in freshwater and saltwater environments. Several species, such as O. niloticus, O. aureus, and O. mossambicus, are excellent for aquaculture because these fish are easily reproduced and readily adapt to diverse environments. Historically, tilapia species, including O. mossambicus, S. melanotheron, and O. aureus, were introduced to Hawaii many decades ago, and the state of Hawaii uses the import permit policy to prevent O. niloticus from coming into the islands. However, hybrids produced from O. niloticus may already be present in the freshwater and marine environments of the islands. The purpose of this study was to identify tilapia species that exist in Hawaii using mitochondrial DNA analysis.

Methodology/Principal Findings

In this study, we analyzed 382 samples collected from 13 farm (captive) and wild tilapia populations in Oahu and the Hawaii Islands. Comparison of intraspecies variation between the mitochondrial DNA control region (mtDNA CR) and cytochrome c oxidase I (COI) gene from five populations indicated that mtDNA CR had higher nucleotide diversity than COI. A phylogenetic tree of all sampled tilapia was generated using mtDNA CR sequences. The neighbor-joining tree analysis identified seven distinctive tilapia species: O. aureus, O. mossambicus, O. niloticus, S. melanotheron, O. urolepies, T. redalli, and a hybrid of O. massambicus and O. niloticus. Of all the populations examined, 10 populations consisting of O. aureus, O. mossambicus, O. urolepis, and O. niloticus from the farmed sites were relatively pure, whereas three wild populations showed some degree of introgression and hybridization.

Conclusions/Significance

This DNA-based tilapia species identification is the first report that confirmed tilapia species identities in the wild and captive populations in Hawaii. The DNA sequence comparisons of mtDNA CR appear to be a valid method for tilapia species identification. The suspected tilapia hybrids that consist of O. niloticus are present in captive and wild populations in Hawaii.  相似文献
