The association of FMRFamide-related peptides (FaRPs) with the spermatheca of Locusta migratoria was demonstrated using radioimmunoassay and immunohistochemical techniques. The physiological effects of various FaRPs on the neurally evoked contractions of the spermatheca were also examined. FMRFamide-like immunoreactivity (FLI) was demonstrated in processes and cell bodies situated in the VIIIth (terminal) abdominal ganglion. These included an anterior, central and posterior pair of ventral cell bodies positioned near the midline of the ganglion, in addition to two bilaterally paired dorsal cell bodies in the posterior region of the VIIIth abdominal ganglion. Two axons displaying FLI proceed down the ventral ovipositor nerve (VON) and into the receptaculum seminis nerve which innervates the anterior regions of the spermatheca. FLI was also noted in processes on the spermathecal muscle with the highest density occurring on the spermathecal sac and coil duct. FaRPs applied to the spermathecal muscle included GQERNFLRFamide, NFIRFamide, ADDRNFIRFamide, YGGFMRFamide, FMRFamide, ADVGHVFLRFamide and SchistoFLRFamide (PDVDHVFLRFamide). Dose-dependent physiological effects were only noted for FMRFamide, ADVGHVFLRFamide and SchistoFLRFamide. FMRFamide led to a dose-dependent increase in the amplitude of neurally evoked contractions with a threshold of approximately 5 x 10(-7) M. SchistoFLRFamide, and ADVGHVFLRFamide, had an inhibitory effect, decreasing the amplitude of neurally evoked spermathecal contractions.  相似文献

Release of spermatozoa from the receptaculum seminis of Schistocerca vaga was studied by means of electrical and mechanical stimulation. Electrical stimulation of the receptaculum nerve, or the ductus aperture nerve, leads to release of spermatozoa from the receptaculum seminis, provided the spermathecal innervation is intact. Mechanical stimulation of the ductus aperture in the genital chamber also leads to sperm release, provided the neural loop, ductus aperture/terminal abdominal ganglion/receptaculum seminis, has not been interrupted at any point. Ten somata in the terminal abdominal ganglion, including 6 dorsal unpaired medial (DUM) neurons, innervate the receptaculum seminis; some of these somata may be neurosecretory. Approximately 80 presumed sensory axons run from the ductus aperture to the same ganglion. On the basis of these neuroanatomical data and the results of electrical and mechanical stimulation, a schema of how the release mechanisms operate in S. vaga is proposed.  相似文献

Octopamine was identified in the spermathecal tissue of Locusta migratoria using HPLC and immunohistochemical techniques. Octopamine-like immunoreactive unpaired median neurons were identified in the VIIth and VIIIth (terminal) abdominal ganglia and octopamine-like immunoreactive axons were present in the ventral ovipositor nerve (branches from this nerve innervate the spermatheca). Stimulatory actions of octopamine on myogenic and neurogenic contractions were observed. Dose-dependent increases in the frequency of myogenic contractions and the amplitude of neurogenic contractions were elicited by the application of octopamine to the spermathecal muscle. Non-sustained basal tension increases were noted in some preparations, although these were not found to be dose-dependent. SchistoFLRFamide (PDVDHVFLRFamide) inhibited octopamine-induced contractions by a maximum of about 30%. In the presence of 3-isobutyl-1 -methylxanthine, octopamine increased cAMP levels in all regions of the spermathecal. The largest increase in cAMP content was found in the spermathecal sac, followed by the straight duct and coil duct. Phentolamine blocked octopamine-induced increases in cAMP levels and abolished the actions of octopamine on myogenic contractions.  相似文献

Tyramine-like immunoreactivity was identified in neurons of the VIIIth abdominal ganglion and in axons projecting to the spermatheca of adult females of Locusta migratoria. Tyramine-like immunoreactive processes were also found throughout all regions of the spermatheca and tyramine-like immunoreactive bipolar or multipolar neurons were present on the spermathecal sac. HPLC coupled with electrochemical detection revealed more tyramine than octopamine present in spermathecal tissue. Electrical stimulation of the ventral ovipositor nerve resulted in a significant increase in calcium-dependent release of tyramine from the spermatheca. Both tyramine and octopamine increase the frequency and basal tonus of spermathecal contractions in a dose-dependent manner, with octopamine having a lower threshold. When tyramine is applied along with a half maximal octopamine dose, there is an additive effect on contractions of the spermatheca with slight synergistic effects at lower doses of tyramine. High concentrations of tyramine (10(-4)M) stimulated increases in cyclic AMP levels of the spermatheca; an effect blocked by phentolamine. Phentolamine has a higher affinity (and thus a lower IC(50) value congruent with5.6x10(-8)M) than yohimbine (IC(50) congruent with1.1x10(-4)M) in reducing tyramine-induced spermathecal contractions. Taken together, these results suggest that tyramine may be a co-transmitter with octopamine at the spermatheca, with both neuroactive chemicals acting on an octopamine receptor.  相似文献

Crustacean cardioactive peptide (CCAP)-like immunoreactivity was identified in neurons of the VIIIth abdominal ganglion and in axons in the nerves that project to the spermatheca of 3-4 week old adult female locusts. In addition, lightly stained CCAP-like immunoreactive processes were localized over the spermathecae. The amount of CCAP in the spermathecal tissue was quantified using an enzyme-linked immunosorbent assay (ELISA) performed on extracts of the whole spermatheca, and on its constituent parts, namely the sperm sac, coiled duct and straight duct. The spermatheca contains 920+/-273 fmol (mean+/-SE) of CCAP equivalents, with the majority localized in the coiled duct. There are age-related differences in the amount of CCAP present in the spermathecae with less content in spermathecae from 1 to 5 day old and greater content in spermathecae from 3 to 4 week old adults. There was also no difference in CCAP content of spermathecae in mated and virgin 3 to 4 week old adults. Reversed phase-high performance liquid chromatography (RP-HPLC) followed by ELISA further confirmed the presence of CCAP-like material in extracts of locust spermathecae. Physiological assays demonstrated that CCAP increased the basal tonus and frequency of spontaneous contractions of the spermatheca, with thresholds between 10(-10) and 10(-9)M and maxima at 10(-7)M CCAP. CCAP also increases the amplitude of neurally evoked contractions with a threshold less than 10(-11)M and a maximum of 10(-7)M CCAP. The present study suggests that CCAP acts as a neuromodulator/neurotransmitter at the spermathecal visceral tissue of female Locusta migratoria.  相似文献

The control of spermathecal contractions in Locusta migratoria via a neural loop was demonstrated using mechanical stimulation and electrophysiological recordings. Extracellular electrophysiological recordings from the receptaculum seminis nerve (N2B2), which innervates the spermathecal sac, were conducted during mechanical stimulation of the genital chamber sensory cells. Activation of the genital chamber sensory cells, using a glass probe approximating the shape and size of an egg, was found to increase the action potential frequency and initiate bursts of action potentials if a tonic frequency of action potentials was present prior to stimulation. If the motor pattern initially consisted of bursts of action potentials, then mechanical stimulation of the genital chamber sensory cells resulted in an increase in firing frequency, in most preparations, with the bursting remaining. Removal of the probe from the genital chamber always returned the motor activity to that noted prior to sensory cell stimulation. Simultaneous electrophysiological recordings from both the left and right receptaculum seminis nerves (N2B2) revealed that the bursts of action potentials were coordinated, although individual action potentials were not coupled one to one. Activation of the genital chamber sensory cells also resulted in increases in spermathecal contraction frequency, an effect which was coordinated with the changes in motor activity. It is proposed that an egg in the genital chamber activates the sensory cells resulting in increases in spermathecal contraction frequency and the subsequent release of spermatozoa onto the micropyle of the egg for fertilization.  相似文献

The female reproductive system of the humpbacked fly Megaselia scalaris Loew (Diptera : Phoridae) was examined in whole mount preparations and serial sections. The system includes 2 ovaries, paired lateral oviducts, a common oviduct, and a genital chamber, opening externally through a gonopore, anteriad and ventrad to the anus. The ducts of the 2 accessory glands open independently into the dorsal region of the genital chamber. The terminal duct of a 2-armed spermatheca joins the right posterior and ventral wall of the genital chamber, immediately inside the gonopore. Passing dorally, the spermathecal duct lies immediately ventral to the duct of the right accessory gland. A short distance posteriad, it divides into two branches, each supplying an arm of the spermatheca. The genital chamber extends both anteriorly and posteriorly from its junction with the common oviduct, creating anterior and posterior compartments. In the right lateral wall of the genital chamber, a distinctive loop-shaped thickening (plate) resembles a darkened thread when it is observed through the integument. Features likely to have taxonomic utility include the posterior and ventral location of the terminal portion of the spermathecal duct; and the asymmetrically arranged, loop-shaped plate.  相似文献

The musculature of the mushroom-shaped accessory gland receives innervation from trunks 5C1 of the phallic nerves, which arise from the posterior part of the terminal abdominal ganglion of the male cockroach Periplaneta americana. Anterograde cobalt filling through trunks 5C1 with the subsequent precipitating procedure has shown the fine innervation of the accessory gland. By retrograde cobalt filling through the same trunks, different types of cells have been mapped in the terminal abdominal ganglion. About 25 dorsal unpaired median (DUM) neurons have been identified among them. About 36 octopamine-like immunoreactive DUM neurons with large somata have been characterized in whole-mount preparations of the terminal abdominal ganglion. The combination of the cobalt-filling technique with immunohistochemical mapping of cells suggests an octopaminergic innervation of the musculature of the accessory gland by DUM neurons.  相似文献

【目的】明确斑衣蜡蝉Lycorma delicatula雌成虫生殖系统整体形态及超微结构特征,为蜡蝉总科昆虫分类及系统发育探讨提供更多形态学证据。【方法】采用光学显微镜与透射电子显微镜,观察斑衣蜡蝉雌成虫生殖系统整体形态和各主要器官的超微结构。【结果】斑衣蜡蝉雌成虫生殖系统主要包括1对卵巢、1个中输卵管、1个交配囊、1个交配囊管、1个前阴道、1个后阴道、1个受精囊、1个受精管和2根受精囊附腺。卵巢为端滋式,由14根卵巢小管组成,卵室由固有膜、滤泡细胞和卵细胞组成,卵巢小管中的滋养细胞清晰可见;中输卵管位于前阴道基部,由中输卵管腔、上皮细胞、肌肉鞘和基膜组成;交配囊膨大呈圆球状,囊壁由上皮细胞、肌肉层和基膜组成;交配囊管呈圆柱状,连接交配囊和后阴道,由肌肉鞘、上皮细胞层和管腔组成;前、后阴道超微结构相似,主要由肌肉鞘、基膜、上皮细胞和管腔组成,但后阴道上皮细胞细胞核周围存在分泌颗粒,且管腔内有大量微绒毛,而前阴道壁内包含有大量囊泡结构;受精管从中输卵管末端延伸至受精囊,由基膜、厚层肌肉鞘和管腔组成;受精囊为受精管近末端略膨大的囊状结构,由肌肉鞘、基膜、上皮细胞和囊腔构成;雌性受精囊附腺着生于受精囊末端,为均匀的螺旋管状,主要由肌肉层、上皮细胞层和附腺中心管腔组成。【结论】斑衣蜡蝉雌性生殖系统与已报道的蜡蝉总科其他类群的雌性生殖系统结构相似,但卵巢小管数目有差异;蝉亚目中不同总科雌成虫雌性附腺与受精囊附腺的形态特征存在明显区别;斑衣蜡蝉雌性生殖系统超微结构与叶蝉总科和沫蝉总科昆虫也存在部分差异。这些差异是否可以作为头喙亚目高级阶元的划分依据仍有待于进一步研究。  相似文献

【目的】揭示绿盲蝽Apolygus lucorum腹神经节的组成结构。【方法】采用免疫组织化学染色方法,利用突触蛋白抗体对绿盲蝽成虫的腹神经节进行免疫标记,激光共聚焦扫描显微镜扫描照相获得原始数据,用图像分析软件进行标记,构建三维结构模型。【结果】绿盲蝽成虫腹神经节位于腹神经索的末端,与其前方的后胸神经节和中胸神经节紧密融合,形成后部神经节。与脑和胸神经节类似,腹神经节由周围的细胞体和内部的神经髓构成。腹神经节的神经纤维束主要包括位于腹侧的两条纵向神经连索和向两侧发出的9束神经纤维。9束神经纤维连接着9个神经原节,即富含突触联系的神经髓。这些神经原节紧密融合,无明显的边界,最后两节形成膨大的末端腹神经节。两侧的神经原节由横向的神经连锁连接起来。腹神经节外周的细胞体数量较多,排列紧密,大小一致,仅在前端背侧中间和后端腹侧中间位置分别有2个和5个体积较大的细胞体。【结论】本研究结果明确了绿盲蝽腹神经节的结构,为进一步研究昆虫的行为调控及神经系统发育和演化奠定一定的形态学基础。  相似文献

The autonomic innervation of the ovary of the dab was studied histologically and physiologically. The ovary receives a branch of nerve bundles that emerge into the abdominal cavity at the postero-ventral end of the kidney and can be traced back to the sympathetic chain in the vicinity of the 5th vertebra. Almost all the nerve fibers are AChE-positive, and some of them also emit adrenergic fluorescence. Electrical stimulation of the ovarian nerves caused ovarian contractions, and administration of ACh elicited contractions of the ovary preparations, supporting the hypothesis that the ovary is innervated by excitatory cholinergic fibers. In the ovarian nerve bundles, many AChE-positive and non-fluorescent ganglion cells are scattered. Ultrastructural studies suggest that nerve endings situated on the ovarian smooth muscle and on ganglion cells are cholinergic. These results also suggest that the cells are the post-ganglionic neurons of the cholinergic innervation and the axons of the cells reach to the muscle cells. On the other hand, the adrenergic fluoresecent fibers possibly participate in the inhibitory innervation, since the presence of inhibitory beta-adrenoceptors were demonstrated by pharmacological studies.  相似文献

The female genital system of the oonopid Silhouettella loricatula is astonishingly complex. The genital opening is situated medially and leads into an oval receptaculum that is heavily sclerotized except for the ventral half of the posterior wall that appears chitinized only. A large striking sclerite lying in the posterior wall of the uterus externus is attached anteriorly to the receptaculum and continues dorsally into a globular appendix that bears a furrow. The uterus externus shows a peculiar modification in its anterior wall: a paddle-like sclerite with a nail-like posterior process. This sclerite lies opposite to the furrow proceeding in the globular appendix and may serve females to lock the uterus externus by muscle contractions. Massive muscles connect the sclerite with the anterior scutum of the opisthosoma and with two other sclerites that are attached to the receptaculum and serve as attachments for further muscles. Gland cells extend around a pore field of the receptaculum. They produce secretion that encloses spermatozoa in a discrete package (secretory sac) inside the receptaculum. In this way, the mixing of sperm from different males and thus sperm competition may be severely limited or completely prevented. During a copulation in the laboratory the ejection of a secretory sac that most probably contained spermatozoa was observed, indicating sperm dumping in S. loricatula. The ejection of the secretory sac may be caused by female muscle contractions or by male pedipalp movements. The majority of the investigated females have microorganisms in the receptacula that could represent symbionts or infectious agents. The microorganisms can be identified partly as bacteria. They are enclosed in secretion and are always found in the same position inside the receptaculum.  相似文献

A group of six dorsal unpaired median (DUM) neurons of the suboesophageal ganglion (SOG) of locusts was studied with neuroanatomical and electrophysiological techniques. The neurons are located posteriorly in the SOG and have axons that descend into the ganglia of the ventral nerve cord, some as far as the terminal abdominal ganglion. Within thoracic ganglia the neurons have profuse dendritic ramifications in many neuropiles, including ventral sensory neuropiles. Based on their projection patterns three different morphological types of neurons can be distinguished. These neurons receive excitatory inputs through sensory pathways that ascend from the thoracic ganglia and are activated by limb movements. They may be involved in the modulation of synaptic transmission in thoracic ganglia.  相似文献

背侧不成对中间神经元(DUM)是一类位于多种昆虫腹神经索神经节背侧的神经元,能自发产生内源性超射动作电位。在DUM神经元细胞膜表达多种受体和离子通道,且电生理特性有别于哺乳动物中枢神经元膜上同种类型的受体和离子通道。目前已证实其细胞膜上表达K+通道、电压依赖的Na+通道、Ca2+敏感的Cl-通道、Ca2+通道、氯离子通道、乙酰胆碱受体、谷氨酸受体等多种离子通道和受体。近年来因膜片钳(patch-clamp)技术进展和对受体和离子通道研究的深入,该类神经细胞已用于杀虫剂选择性神经毒性研究和杀虫剂离体筛选。  相似文献

The distribution of the ganglia and nerves of the stomatogastric nervous system and the innervation of the extrinsic and intrinsic muscles are described. Median unpaired frontal and hypocerebral ganglia and paired ingluvial ganglia are present. The anterior pharynx is innervated by branches of the frontal nerve and by the anterior and posterior pharyngeal nerves, originating from the frontal ganglion. The posterior pharyngeal nerves are linked to nerves innervating the posterior part of the pharynx which have their origin in the hypocerebral ganglion, the anterior portion of which has previously been regarded as part of the recurrent nerve. Paired esophageal nerves run the length of the esophagus and crop between the hypocerebral and and ingluvial ganglia, innervating the muscularis by serial side branches. From each ingluvial ganglion runs an ingluvial nerve which innervates the gizzard and a cecal nerve which innervates the midgut and its ceca. At the posterior end of the midgut there is a poorly developed nerve ring. Nerves running posteriorly from this nerve ring link the stomatogastric nervous system with the proctodeal innervation from the terminal abdominal ganglion. Multipolar peripheral neurons are present on the muscularis of the whole of the foregut, rather randomly distributed on the crop and gizzard but forming fairly definite groupings at some points on the pharynx. Though of varied appearance, these cells could not be divided into discrete morphological categories. Peripheral neurons on the midgut are of different and characteristic morphology, though a few cells of the same appearance as those of the foregut occur at the midgut-hindgut boundary. Nerve fibers on the gut almost invariably terminate on the fibers of the muscularis.  相似文献

Summary The distribution of certain catecholamines and indoleamines in the ventral nerve cord and the body segments of the medical leech, Hirudo medicinalis, was studied with the fluorescence microscope technique of Falck and Hillarp, with microspectrofluorometry, and with chemical determinations of the amines. The six cells of the segmental ganglia previously shown to be chromaffin were found to contain an amine, most probably 5-hydroxytryptamine. In the two giant cells, the amine was found on the surface of coarse intracellular granules, lying mainly at the cell membrane, and at the nucleus. The two giant cells send their axons to the body muscles, which thus seem to have a 5-hydroxytryptaminergic innervation. The four smaller amine-containing cells of the segmental ganglia send their axons to the neuropil of the ganglion.The only cell type found to contain a catecholamine (probably noradrenaline) was situated in the anterior segmental nerve in the cell cluster anterior of the nephridial duct, one cell in each nerve. The axon of this cell terminates in two or more segmental ganglia; thus these neurons seem to be afferent.This work was supported by grants from the Swedish Natural Science Research Council (project no. 99-35) and the Swedish Medical Research Council (projects no. Summary Three antisera were used to study the distribution and anatomy of bovine pancreatic polypeptide (BPP)-like/FMRFamide-like immunoreactive neurones within the unfused abdominal ganglia of the migratory locust, Locusta migratoria. All the antisera used stained two or more clusters of perikarya, localized anteriorly and posteriorly near the midline within each unfused abdominal ganglion. Double labelling experiments with intracellular dye injection, or differential backfilling, combined with subsequent immunostaining were carried out to identify these neurones. Two of the antisera (antisera 1 and 2, both raised against FMRFamide) stained three groups of midline neurones, located anterior dorsal, anterior ventral and posterior dorsal within the ganglion. Neurones of the former of these two clusters projected via the anterior median nerve to a neurohaemal organ. The posterior cluster of midline cells comprised immunopositive perikarya all but one of which also projected via the anterior median nerve to innervate the neurohaemal organ. Double labelling with Lucifer yellow and antisera 1 and 2 showed that the remaining neurone was the previously identified doral unpaired median (DUM)heart1 neurone. The third antiserum (AK141), also raised against FMRFamide, stained neurones within an anterior dorsal cluster, and in a posterior cluster. Double labelling with differential Co2+/Ni2+-backfilling and the antiserum 3 (AK141) demonstrated that the large neurones of both clusters belonged to the population of bilaterally projecting neurones (BPNs), including the DUMheart1 neurone. Since the antisera cross-react with BPP and fail to label neurones when preadsorped with BPP or FMRFamide, we conclude that the labelled neurones contain polypeptides of the FMRFamide/BPP-family.  相似文献

Female Harpactea lepida possess a single genital opening leading into a diverticulum. This diverticulum shows no secretory layer. It continues posteriorly into a receptaculum which is associated with gland cells. In the two already described dysderids, Dysdera crocata and D. erythrina, the bilobed spermatheca lies anteriorly to the diverticulum. Gland cells are associated with the spermatheca and the diverticulum. In H. lepida, the sclerotized genital structures lie dorsally to the diverticulum and consist of a posterior and an anterior part. The posterior part shows a lamella extending laterally to sclerites functioning as muscle attachments. The anterior part has two roundish structures. A hollow stalk-like sclerite functioning as muscle attachment extends towards anterior. The posterior and the anterior part of the sclerotized genital structures fit together. A narrow uterine valve connecting the uterus externus with the diverticulum forms between them. It may be opened by muscles as also suggested for D. erythrina. In H. lepida, spermatozoa embedded in secretion are found in the diverticulum and the receptaculum. There is no evidence that they are stored under different conditions like in D. erythrina. Additional spermatozoa are found in the uterus externus of H. lepida which could be an indication for internal fertilization. Spermatogenesis occurs in cysts in the testes of male H. lepida. In the vasa deferentia, the ductus ejaculatorius and the palpal bulb, the spermatozoa are embedded in homogenous secretion. The palpal bulb has a distal extension bearing a crown-like structure. Summary The nervus corporis cardiaci III (NCC III) of the locust Locust migratoria was investigated with intracellular and extracellular cobalt staining techniques in order to elucidate the morphology of neurons within the suboesophageal ganglion, which send axons into this nerve. Six neurons have many features in common with the dorsal, unpaired, median (DUM) neurons of thoracic and abdominal ganglia. Three other cells have cell bodies contralateral to their axons (contralateral neuron 1–3; CN 1–3). Two of these neurons (CN2 and CN3) appear to degenerate after imaginal ecdysis. CN3 innervates pharyngeal dilator muscles via its anterior axon in the NCC III, and a neck muscle via an additional posterior axon within the intersegmental nerve between the suboesophageal and prothoracic ganglia. A large cell with a ventral posterior cell body is located close to the sagittal plane of the ganglion (ventral, posterior, median neuron; VPMN). Staining of the NCC III towards the periphery reveals that the branching pattern of this nerve is extremely variable.
