Ploidy of watermelon [Citrullus lanatus (Thunb.) Matsum. and Nakai shoots and plantlets was estimated by painting the lower epidermis of intact in vitro-derived leaves with fluorescein diacetate (FDA) and observing fluorescence of guard cell chloroplasts with a microscope and UV light. Leaves from in vitro shoot-tip cultures of known diploid cultivars and tetraploid breeding lines were used to establish the mean number of chloroplasts per guard cell pair. Leaves from diploid and tetraploid shoot cultures had 9.7 and 17.8 chloroplasts per guard cell pair, respectively. This method then was used to estimate the ploidy of shoots regenerated from cotyledon explants of the diploid cultivar Minilee. Approximately 11% of the 188 regenerated shoots were classified as tetraploid during in vitro culture. Putative tetraploids were transplanted to the field and self-pollinated. About 45% of tetraploids identified in vitro produced fruit and viable seed. Chloroplast counts of R1 progeny were used to confirm their ploidy. All of the putative diploids were confirmed diploid and all putative tetraploids proved to be non-chimeric true breeding tetraploids. 盾叶薯蓣四倍体与二倍体叶表皮及气孔器显微结构的比较   总被引:5,自引:1,他引:4  
对盾叶薯蓣四倍体和二倍体阴叶和阳叶表皮及气孔器的显微形态结构进行了比较。结果显示:叶片表皮细胞及气孔器的形态结构因染色体倍性及光照的不同而存在明显差异。表皮细胞有规则多边形与不规则细胞壁内褶2种类型;气孔器为典型的双子叶植物无规则型气孔器,仅分布在下表皮。四倍体表皮细胞密度、气孔器密度及气孔指数平均值分别为476.28±6.87个.mm-2、78.22±3.1个.mm-2、14.11,较二倍体的分别小4.60%、17.95%和11.98%。前者气孔器长、宽及保卫细胞宽度平均值分别为32.78±2.09μm、26.07±1.55μm、9.63±1.14μm,较后者的分别大10.73%、3.90%和18.01%,差异极显著。前者阴叶的较其阳叶的分别大5.77%、6.00%、8.72%;后者的则分别大1.72%、1.74%、2.41%。前者叶片保卫细胞中的叶绿体数目为23.93±3.19个,较后者的多74.00%。表皮细胞及气孔器密度、气孔指数,气孔器大小,保卫细胞叶绿体数目、尤其是保卫细胞宽度,可作为倍性鉴定的参考依据。四倍体高产有其良好的叶片结构基础。  相似文献

This study was carried out to estimate the broad-sense heritability for seed yield and some yield components of safflower (Carthamus tinctorius L.) cultivars. The experimental design was a randomized complete block design with three replications in the 2004 growing season in the Middle Black Sea Region conditions of Turkey. Three safflower cultivars (5-154, Din?er and Yenice) were grown at five locations (Bafra, Ladik, Suluova, Gümü?hacik?y and Osmancik). The heritability for seed yield, plant height, first branch height, number of branch, head diameter, number of seed per head, 1000-seed weight and oil content were estimates as 35%, 93%, 99%, 45%, 21%, 69%, 81% and 59%, respectively. It was found that first branch height was the least affected trait over environments and followed plant height, thousand seed weight and number of seed per head. On the other hand, head diameter, seed yield, number of branch and oil content were the most affected traits versus environmental conditions. The first branch height, plant height and 1000-seed weight could be used to succeed in selection in early generation.  相似文献

Greengram or mungbean (Vigna radiata (L.) Wilczek), which originated in the Indian sub-continent, is an important pulse crop in India. Genetic diversity and patterns of variation among 111 accessions representing the better agronomic types from the entire collection of the National Bureau of Plant Genetic Resources were studied. Wide variation in morphological and agronomic traits viz. number of pods per plant, yield per plant, 100-seed weight, fruit setting capacity, flowering period, maturity, number of pod bearing peduncles, plant height, number of primary branches, length of branch, nodulation and leafiness was observed. Grain yield was found to be significantly correlated with 100-seed weight and pod length. The data were subjected to cluster and principal components analysis. The accessions were grouped into six discrete and well-defined clusters. The study demonstrated the patterns of variation at the population level. The multivariate analysis was useful in identifying a group of accessions with yield enhancing traits within a highly diverse group of accessions and their potential value in greengram improvement is suggested.  相似文献

Tetraploid plants were produced by inducing chromosome doubling using colchicine in in vitro shoot tips of poplar and black locust clones. Many of the plants treated with colchicine showed modified morphological characteristics like stunted growth, thicker leaves and modified leaf morphology. The counting of chloroplast number in the epidermal guard cells of stomata was used for the rapid screening of tetraploids. The differences in mean chloroplast numbers between diploid and tetraploid plants were highly significant. For all plants tested, the tetraploid genotype had almost double the number of chloroplasts per guard cell compared to the diploid origin. Some plants were further analysed by flow cytometry to verify their ploidy status that was determined by chloroplast numbers. The results of this study demonstrated for the first time that chloroplast counting in poplar and black locust could be an effective and reliable method for pre-screening large numbers of plants for their ploidy level. The protocol might be applicable in a wide scope of breeding programs.  相似文献

Summary The potential of tuber disc culture for chromosome doubling was investigated in somaclonal populations of four dihaploid genotypes and one tetraploid cultivar of potato (Solanum tuberosum). Laser scanning confocal microscopy (LSCM) was used for rapid determination of the ploidy level based on the number of chloroplasts in stomatal guard cells of leaves. Factorial analysis of chloroplast number in 58 clones and two leaf types showed that somaclones were clearly divided in two groups. Clones with 5–7 chloroplasts per cell as observed in tuber derived diploid controls were classified as 2X (not doubled), while those with 9–14 chloroplasts resembled the tuber derived tetraploid controls and were considered 4X (doubled). A high frequency of spontaneous chromosome doubling, 42% – 50%, was detected in 3 dihaploid genotypes, whereas no doubling was observed in one of the dihaploids as well as the tetraploid cultivar Yukon Gold. Effects of leaf type on chloroplast number was also significant. The middle leaf showed significantly higher chloroplast number than the younger leaves.  相似文献

Diploid sugar beet plants demonstrate a broad variability of the number of chloroplasts in stoma guard cells, which is related to myxoploidy of cell populations in leaf apical meristems (epigenomic variability). In addition to random organelle segregation between daughter cells, this variability is affected by factors disrupting the mitotic cycle: (1) plant treatment with a mitotic poison, such as colchicine; (2) duration of the life cycle of a plant; the variability in second-year plants is greater than in first-year ones; (3) the mode of plant reproduction; the variability in inbred plants is greater than in the initial population. Treatment of germinating seeds with a diluted colchicine solution increases the number of organelles in cells in the myxoploid generation (generation C0) and the variance of the distributions in the first vegetation year. The variability in the organelle number in stoma cells correlates with that in maternal meristem cells. It is concluded that the epidermal cell monolayer (including stoma guard cells) keeps record of the epigenomic and epiplastome variability in meristem cells. The variability of the number of chloroplasts in stoma guard cells is approximated by binomials with negative powers.  相似文献

采用扫描电镜和石蜡切片法,以‘寒富’苹果二倍体及经秋水仙素加倍获得的同源四倍体植株为材料,比较两种倍性植株叶片超微结构、叶绿素含量及叶绿素荧光参数的日变化规律。结果显示:(1)与二倍体植株相比较,其同源四倍体叶片厚度、栅海比、气孔长、气孔宽、分别增加了15.1%、16.1%、70.5%、27.2%,而气孔密度显著减少了58.7%;其同源四倍体叶保卫细胞中叶绿体数和叶绿素含量分别比二倍体植株高出125.3%、37.7%。(2)‘寒富’苹果同源四倍体与其二倍体的叶绿素荧光参数PSⅡ的原初光能转化效率(Fv/Fm)、PSⅡ的潜在光化学效率(Fv/F0)和以吸收光能为基础的光合性能指数(PI)值的日变化趋势相似,但PI平均值比二倍体显著高出38.6%。研究表明,同源四倍体较二倍体叶片在形态上更大、更厚,气孔更大、密度更小,栅海比更大,表现出抗病的叶片结构;同时同源四倍体较二倍体含有更高的叶绿素含量,表现更优良的光合特性。  相似文献

In stomata guard cells of sugar beet, variation in the number of chloroplasts was studied in successive generations: (1) hybrid generation; (2) generation yielded by uniparental apozygotic seed reproduction; (3) generation obtained after seed treatment with a colchicine solution; (4) generation obtained after seed treatment with 5-azacytidine. As compared to hybrid generation, uniparental seed reproduction increases the average number of chloroplasts in stomata guard cells (from 13.5 to 15.0) and decreases distribution variance of this trait by a factor of 3 to 4. Colchicine increases both average number of chloroplasts in stomata guard cells (from 13.5 to 18.2) and distribution variance (about twice). 5-Azacytindine reduces the number of chloroplasts in cells (from 15.0 to 12.9) but enhances distribution variance (about 1.5 times). Variation in the number of chromosomes in stomata cells is related to myxoploidy in meristem tissue, on the one hand, and to the rate of cell division, on the other. Uniparental seed reproduction is suggested to enhance the number of organelles per cell due to high myxoploidy in cell populations, which is typical of inbred plants. Colchicine blocks spindle division and sharply increases the level of myxoploidy in cell populations and the number of organelles per cell. 5-Azacytidine hypomethylates chromosome DNA, increases the rate of cell divisions, and reduces the number of organelles per cell. The described changes in the number of chloroplasts are inherited in cell lineage ("cell hereditary memory") and successive sporophyte generations.  相似文献

Eight faba bean (Vicia faba L.) genotypes were grown at lowlands of the west-Mediterranean region of Turkey in order to estimate the broad-sense heritability for plant height, number of stems and pods per plant, seed yield, biological yield, 100-seed weight, days to flowering and maturity. The heritability for plant height, number of stems and pods per plant, seed yield, biological yield, 100-seed weight, days to flowering and maturity were estimated as 83%, 63%, 43%, 62%, 52%, 99%, 97% and 97%, respectively. It was found that seed weight was the least affected trait across changing environmental conditions and followed by days to flowering and maturity. On the other hand, number of pods per plant, biological and seed yields and number of stems per plant were the most affected traits versus environmental conditions.  相似文献

Photosynthetically-active protoplasts isolated from isogenic sets of diploid-tetraploid and tetraploid-octoploid alfalfa (Medicago sativa L.) leaves were used to investigate the consequences of polyploidization on several aspects related to photosynthesis at the cellular level. Protoplasts from the tetraploid population contained twice the amount of DNA, ribulose-1,5-bisphosphate carboxylase (RuBPCase), chlorophyll (Chl), and chloroplasts per cell compared to protoplasts from the diploid population. Although protoplasts from the octoploid population contained nearly twice the number of chloroplasts and amount of Chl per cell as tetraploid protoplasts, the amount of DNA and RuBPCase per octoploid cell was only 50% higher than in protoplasts from the tetraploid population. The rate of CO2-dependent O2 evolution in protoplasts nearly doubled with an increase in ploidy from the diploid to tetraploid level, but increased only 67% with an increase in ploidy from the tetraploid to octoploid level. Whereas leaves and protoplasts had similar increases in RuBPCase, DNA, and Chl with increase in ploidy level, it was concluded that increased cell volume rather than increased cell number per leaf is responsible for the increase in leaf size with ploidy.  相似文献

Summary A half diallel analysis involving nine cultivars showed that additive as well as non-additive gene effects were important for the inheritance of seed yield per plant, 100-seed weights, protein content and potassium per cent. For remaining traits non-additive genetic components were important. Overdominance was observed for all traits except for 100-seed weight, which expressed partial dominance. Parents PMR-T10, EC21857, EC109182, T163 and EC109189 were good general combiners for seed yield, seed weight and quality traits. In general there was a good relationship between per se performance and the gca effects of the parents for all traits. Cross combinations such as LMR8 x EC109182,LMR8 x PMR-T10,LMR8 x EC21857,PMRT10 x EC21857 and P23 x EC21857 were found promising. The seed yield was positively correlated with other quality traits. Protein had a positive correlation with methionine and phosphorus. All the values of correlation co-efficients were non-significant except for yield with potassium, 100-seed weight and protein with methionine, indicating that yield and quality attributes can be improved simultaneously by simple selection procedures.  相似文献

Summary A 5 X 5 diallel cross among well-adapted varieties of gram from different agroclimatic regions was studied for heterosis, combining ability and inheritance of days-to-flowering, primary branches, pods per plant, 100-seed weight and grain yield. A high degree of heterosis over mid-parent and better parent was observed for primary branches, no. of pods and grain yield, whereas very little heterosis was exhibited for days-to-flower and 100-seed weight. Crosses among lines of diverse origin generally gave higher heterosis and over-dominance than lines from the same region. Primary branch number, pod number, and grain yield exhibited positive over-dominance; days-to-flower showed negative over-dominance while 100-seed weight had no dominance. Both general and specific combining ability effects were significant for all the characters studied but g.c.a. effects appeared to be more important for days-to-flower, 100-seed weight and grain yield. Graphical analysis indicated additive effects for all the characters, with complete dominance for days-to-flower, no dominance for 100-seed weight and over-dominance for the other three characters. Dominant genes conditioned earliness, primary branch number and 100-seed weight. The role of various parents and crosses in planning a hybridization programme has been discussed.  相似文献

The heritability, the number of segregating genes and the type of gene interaction of nine agronomic traits were analysed based on F2 populations of synthetic oilseedBrassica napus produced from interspecific hybridization ofB. campestris andB. oleracea through ovary culture. The nine traits—plant height, stem width, number of branches, length of main raceme, number of pods per plant, number of seeds per pod, length of pod, seed weight per plant and 1000-seed weight—had heritabilities of 0.927, 0.215, 0.172, 0.381, 0.360, 0.972, 0.952, 0.516 and 0.987 respectively, while the mean numbers of controlling genes for these characters were 7.4, 10.4, 9.9, 12.9, 11.5, 21.7, 20.5, 19.8 and 6.4 respectively. According to estimated coefficients of skewness and kurtosis of the traits tested, no significant gene interaction was found for plant height, stem width, number of branches, length of main raceme, number of seeds per pod and 1000-seed weight. Seed yield per plant is an important target for oilseed production. In partial correlation analysis, number of pods per plant, number of seeds per pod and 1000-seed weight were positively correlated with seed yield per plant. On the other hand, length of pod was negatively correlated (r = -0.69) with seed yield per plant. Other agronomic characters had no significant correlation to seed yield per plant. In this experiment, the linear regressions of seed yield per plant and other agronomic traits were also analysed. The linear regression equation wasy = 0.074x8 + 1.819x9 + 6.72x12 -60.78 (R 2 = 0.993), wherex 8, x9 and x12 represent number of pods per plant, number of seeds per pod and 1000-seed weight respectively. The experiment also showed that erucic acid and oil contents of seeds from F2 plants were lower than those of their maternal parents. However, glucosinolate content was higher than that of the maternal plants. As for protein content, similar results were found in the F2 plants and their maternal parents. It was shown that the four quality traits, i.e. erucic acid, glucosinolate, oil content, and protein content, had heritability values of 0.614, 0.405, 0.153 and 0.680 respectively.  相似文献

We characterized 13 accessions of dry peas of different origins from various growing regions in Argentina, based on three replications of 20 plants cultivated in 2009 and 2010 in a greenhouse, with the objective of selecting those with favorable characteristics for use in breeding programs. Significant differences were found for length and width of stipule and pod, length of the internodes and leaflets, plant height, total number of nodes, number of nodes at the first pod, number of days to flowering and to harvest, number of pods and seeds per pod, 100-seed weight and grain diameter, demonstrating a high degree of genetic variability. Phenotypic correlation analysis demonstrated that large pods produced more seeds per pod, but the seed weight decreased. Plants with smaller number of nodes in the first pod were more productive. Estimates of genotypic correlation coefficients indicated a strong inherent association among the different traits. Clustering methods grouped the accessions into five clusters. Cluster 5 included two accessions and showed the highest values for length and width of stipules (4.9 and 4.5 cm, respectively), length of leaflets (7.43 cm) and days to flowering (122.6), while cluster 3, with one accession, and cluster 4, with two accessions, showed the highest values for number of seeds per pod (3.78 and 4.39), number of pods per plant (5.33 and 5.70), length of pods (5.54 and 5.72 cm), and width of pods (1.21 and 1.20 cm, respectively). We conclude that accessions in clusters 3 and 4 would be useful for crosses with other cultivars in pea breeding programs.  相似文献

采用双子叶植物种子数量性状的遗传模型,分析了大豆品种双列杂交F1和F2种子的粒重、粒宽、粒厚和粒长/粒宽、粒长/粒厚、粒宽/粒厚粒形性状的遗传效应。结果表明:7种粒形性状同时受制于种子直接遗传效应,而且还不同程度的受制于母体和细胞质效应。其中,百粒重、粒长、粒长/粒宽、粒长/粒厚和粒宽/粒厚的遗传以细胞质效应为主;粒宽和粒厚以母体遗传效应为主。粒重、粒长和粒长/粒宽、粒宽/粒厚的种子直接遗传率和细胞质遗传率均属中等,对其4个性状选择可以在较高世代单株和单粒选择均有效果。粒宽和粒厚母体遗传率数值较大,对其性状应以母体单株为单位早代选择,以增加粒宽和粒厚。P2和P7可作为增加百粒重、粒长/粒宽、粒长/粒厚和粒宽/粒厚的理想亲本;P1、P4和P6分别是提高粒长、粒厚和粒宽的理想亲本。  相似文献

As part of an investigation into the control of chloroplast replication the number and size of chloroplasts in mesophyll cells was examined in relation to the size of the cells. In first leaves of Triticum aestivum L. and T. monococcum L. the number of chloroplasts in fully expanded mesophyll cells is positively correlated with the plan area of the cells. The linear relationship between chloroplast number per cell and cell plan area is also consistent over a fivefold range of cell size in isogenic diploid and tetraploid T. monococcum. In T. aestivum the chloroplast number per unit cell plan area varies among cells in relation to the size of the chloroplasts. Those cells containing chloroplasts with a relatively small face area have a correspondingly higher density of chloroplasts, and consequently, the total chloroplast area per unit cell plan area is very similar in all the cells. The results indicate that the proportion of the cell surface area covered by chloroplasts is precisely regulated, and that this is achieved during cell development by growth and replication of the chloroplasts.  相似文献

采用根尖压片法对55份石斛属资源的倍性进行了鉴定,并测量它们的气孔长度、气孔密度、叶长、叶宽、叶厚、株高、茎粗、花朵直径和花瓣厚度9个形态学性状。结果显示:(1)55份供试石斛属资源材料中有:四倍体40份、占72.73%,三倍体11份、占20.00%,二倍体4份、占7.27%。(2)对石斛属倍性与9个形态学性状进行相关分析表明,气孔长度、叶长、叶宽、叶厚、茎粗、花朵直径和花瓣厚度7个性状与染色体倍性呈极显著正相关,气孔密度与染色体倍性呈极显著负相关,株高与染色体倍性相关性不显著。  相似文献

Anther culture of four cultivars of autumn cauliflower gave embryo yields of 100 embryos (per 100 anthers cultured) for one cultivar, eight embryos for a second cultivar, and almost zero for the two other cultivars. From 986 embryos which were grown on, 418 plants were regenerated. The ploidy of the regenerants was assessed by measuring guard cell lengths, pollen size, and by chromosome counts at meiosis. Only 1% of the regenerants were haploid, 41% diploid, 5% triploid and 53% tetraploid. One octoploid plant was also found. The results are discussed in relation to equivalent results for Brussels sprouts, and the use of anther culture in the breediing of cauliflowers.  相似文献
