Tapes philippinarum is a bivalve mollusc of the Pacific Ocean, successfully imported for human consumption into the northern Adriatic Sea (Europe). For better knowledge of its considerable adaptive ability in comparison with similar autochthonous species, a morpho-functional characterisation of its haemocytes was carried out with the establishment of short-term cell cultures (60 min at 25 degrees C). Various methods of cytochemical staining identified four cell types in the haemolymph: granulocytes (48.05% +/- 1.43), hyalinocytes (32.18% +/- 0.99), haemoblasts (18.97% +/- 0.63) and serous cells (0.8% +/- 0.19). The granulocytes, possessing cytoplasmic granules with differing dye affinity, included basophils, neutrophils and acidophils. Such granules stained vitally with Neutral Red, and correspond to lysosomes. Hydrolytic and oxidative enzymes were mainly detectable after stimulation in the presence of yeast cells. Both granulocytes and hyalinocytes were positive for alkaline phosphatase, non-specific esterase, peroxidase, and cytochrome C oxidase, whereas only granulocytes were positive for beta-glucuronidase, acid esterase, and arylsulphatase. Both cell types were competent phagocytes towards yeast and plasma had an opsonising effect. Moreover, the respiratory burst accompanied phagocytosis with superoxide anion production, recognisable through cytoplasmic deposits of formazan after treatment with nitro blue tetrazolium. Haemoblasts were small undifferentiated cells which, due to their morphology and positivity to the anti-CD34 antibody, show the typical features of stem cells. Serous cells, probably arising from Keber's gland and belonging to another differentiation pathway, contained non-sulphate acid mucopolysaccharides and play an important role in early defence mechanisms, taking part in the formation of clots.  相似文献

This study was conducted to characterize the production of superoxide ions by leukocytes in whole blood of the American alligator (Alligator mississippiensis). We used WST-1, a tetrazolium salt which can be reduced to a water-soluble formazan compound with high molar absorptivity at 438 nm, to probe the production of superoxide by alligator leukocytes. Incubation of alligator whole blood with WST-1 resulted in a time- and concentration-dependent increase in absorbance of the plasma at 438 nm. The reduction of WST-1 was inhibited in a concentration-dependent manner by superoxide dismutase, an enzyme that catalyzes the reduction of superoxide to peroxide, confirming that the reduction of WST-1 was due to the presence of superoxide. Treatment of whole blood with nitrotetrazolium blue (NBT) resulted in the staining of heterophils and monocytes, enforcing the idea that that the production of superoxide is due to the presence of leukocytes, and not other blood cell components. It is interesting to note that the production of superoxide by the alligator leukocytes required no external stimulation while human leukocytes must be stimulated with an immunological challenge before producing superoxide. This is the first report of the production of superoxide as an innate immune mechanism in crocodilians.  相似文献

The usual method of staining polyacrylamide gel electropherograms for superoxide dismutase activity utilizes a photochemical flux of O2- to reduce nitroblue tetrazolium. Superoxide dismutases intercept O2-, preventing formazan production and thus causing achromatic bands. In the presence of H2O2, catalases also yield achromatic bands during this staining procedure. This is due to local elevation of pO2 by the catalatic decomposition of H2O2. O2, in turn, inhibits the reduction of the tetrazolium by O2-. This phenomenon provides a new activity stain for catalase. A previously described activity stain for catalase has also been reexamined and significantly improved.  相似文献

Phagocytosis is important in the immune system of the prawn and is believed to be a defence parameter. Previous studies have demonstrated that CpG oligonucleotides enhance the activation of the prophenoloxidase activating system of the prawn through either the G-protein/protein kinase C (PKC) or the cAMP pathway. This study investigated the influence of CpG ODN on the respiratory burst used as the indicator of phagocytic activity and on the initiation of the signal pathway in haemocytes of Macrobrachium rosenbergii. When haemocytes were treated in vitro with 50 microg ml(-1) of ODN2006 for 15 min, the increase of nitroblue-tetrazolium (NBT) reduction suggested that the respiratory burst of haemocytes can be enhanced by ODN2006 stimulation. In an attempt to determine which signal transduction pathway is involved in the enhancement effect, haemocytes were separately treated with activators or inhibitors of specific signalling components. The results showed that the NBT reduction of haemocytes increased after treatment with sodium fluoride (a G-protein activator) and decreased after treatment with GDP-beta-S (a G-protein inhibitor). When ODN2006-stimulated haemocytes were treated with GDP-beta-S, the inductive effect was significantly reduced. In haemocytes treated with 8-bromo-cAMP (a PKA activator), the NBT reduction was not significantly different from the control. The addition of phosphodiesterase-inhibiting caffeine, which inhibits the degradation of cAMP, decreased the NBT reduction of ODN2006-stimulated haemocytes; however, the addition of phenol-12-myristate-13-acetate (PMA) significantly increased the NBT reduction. When PMA-stimulated haemocytes were treated with chelerythrine (a PKC inhibitor), the induced NBT reduction was significantly reduced. Furthermore, the study of ODN2006-stimulated haemocytes treated with chelerythrine showed that the enhancement effect of ODN2006 on the NBT reduction was significantly decreased. All results suggest that the enhancement of the respiratory burst of prawn haemocytes is induced by ODN2006 via a PKC-activating signalling pathway, but negatively regulated via the cAMP pathway.  相似文献

On contact with micro-organisms or any other antigenic particles which are recognised as phagocytosable, the leucocytes of vertebrates raise their oxygen consumption suddenly (respiratory burst). The consumed oxygen is then converted into reactive oxygen species (ROS). An effort has been made in the present study to estimate the production of superoxide anion, one of the ROS, by the resident as well as activated head-kidney leucocytes of Indian major carps. Activation was accomplished by immunisation with formalin killed (FK) and heat killed (HK) whole cell bacterins of Aeromonas hydrophila. In the ex vivo experiment conducted, activated leucocytes yielded a significant increase in O.D. values for nitroblue tetrazolium (NBT) reduction reaction, reflecting an increase in superoxide anion production, from day 1 post immunisation. Of the three species of Indian major carps immunised, Catla catla showed the greatest production of superoxide anion, followed by Labeo rohita and then by Cirrhinus mrigala. The enhancement of superoxide anion production of leucocytes by immunisation justifies the role of immunisation in the microbicidal defence mechanism of fish.  相似文献

The respiratory burst of rat alveolar macrophages stimulated by a variety of agents declines as a function of time of exposure to hyperoxia. Previous studies have evaluated this effect in terms of the stimulated O2⨪ production of a population of cells. The present study was designed to determine whether this decline is due to a “turning off” of the respiratory burst activity of some cells within the alveolar macrophage population or a general suppression of the activity of all cells. The phorbol myristate acetate (PMA) initiated respiratory burst of individual rat alveolar macrophages was monitored using the reaction of nitroblue tetrazolium (NBT), which results in the formation of a precipitate on active cells. The formazan staining was evaluated using black and white photographs of the cells and comparison to a scale constructed from photographed cells of four differing intensities of staining. Frequency distributions indicated that when the respiratory burst capability in the population of alveolar macrophages is impaired approximately 50% by oxygen exposure and/or culture in plastic vessels with artificial media, there is a gradual shift in NBT reduction rather than an “all or nothing” mechanism, in which the distribution would have reflected a shift from darkly stained cells to the very lessened or negligible staining observed at the end stage of oxygen toxicity.  相似文献

The total haemocyte count (THC), phenoloxidase activity, respiratory burst (release of superoxide anion), superoxide dismutase activity, and phagocytic activity and clearance efficiency to the pathogen Lactococcus garvieae were measured when freshwater giant prawns Macrobrachium rosenbergii (16.2 +/- 2.1 g) were individually injected with saline, or dopamine at 0.5, 5.0, or 50.0 pmol prawn(-1). The results show that a transient period of immunosuppression occurred between 2 and 8 h after injection of dopamine for all immune parameters except circulating haemocytes and all immune parameters returned to control values within 8-16 h after receiving dopamine. Injection of dopamine also significantly increased the mortality of M. rosenbergii challenged with the pathogen L. garvieae. These results suggest that stress-inducing dopamine suppresses the immune system, which in turn promotes the susceptibility to L. garvieae in M. rosenbergii.  相似文献

These methods utilize the reduced coenzymes DPNH or TPNH as substrate. The reaction involves a direct transfer of electrons from reduced coenzyme to the respective diaphorase which in turn transfers the electrons to tetrazolium salt, reducing it to the insoluble formazan. Competition for electrons by preferential acceptors in the respiratory chain was prevented by various inhibitors. In the presence of respiratory inhibitors the rate of tetrazolium reduction was markedly increased. The greatest reduction was observed when amytal was used. Sites of diaphorase activity appeared as deposits of blue-black metal formazan chelate measuring 0.2 to 0.3 micro in diameter. Small mitochondria contained 2 deposits, while larger ones contained up to 6. Considerable differences were observed in the rate of tetrazolium reduction and cellular localization of diaphorase activity when DPNH was used as substrate as compared to TPNH. In each instance DPNH was oxidized more rapidly by tissues than TPNH. These findings support the concept that the oxidation of coenzymes I and II is mediated through separate diaphorases.  相似文献

The reaction catalyzed by cysteamine oxygenase on cysteamine in the presence of phenazine methosulphate as cofactor like compound is inhibited by nitroblue tetrazolium, a scavenger of superoxide ions. The reaction is not inhibited by superoxide dismutase and allyl alcohol and it is not activated by superoxide ions produced in solution. Nitroblue tetrazolium is reduced by cysteamine or mercaptoethanol and phenazine methosulphate. This reaction is completely inhibited by superoxide dismutase. In the presence of cysteamine oxygenase the reduction with mercaptoethanol is greatly enhanced and it is only partially inhibited by superoxide dismutase. According to these data a reaction mechanism is proposed in which superoxide ions and thiyl radicals are produced at the active site during catalysis.  相似文献

The respiratory burst is an important feature of the immune system. The increase in cellular oxygen uptake that marks the initiation of the respiratory burst is followed by the production of reactive oxygen species (ROS) such as superoxide anion and hydrogen peroxide which plays a role in the clearance of pathogens and tissue regeneration processes. Therefore, the respiratory burst and associated ROS constitute important indicators of fish health status. This paper compares two methods for quantitation of ROS produced during the respiratory burst in common carp: the widely used, single-point measurement based on the intracellular reduction of nitroblue tetrazolium (NBT) and a real-time luminol-enhanced assay based on the detection of native chemiluminescence. Both assays allowed for detection of dose-dependent changes in magnitude of the respiratory burst response induced by β-glucans in head kidney cells of carp. However, whereas the NBT assay was shown to detect the production of only superoxide anions, the real-time luminol-enhanced assay could detect the production of both superoxide anions and hydrogen peroxide. Only the chemiluminescence assay could reliably record the production of ROS on a real-time scale at frequent and continual time intervals for time course experiments, providing more detailed information on the respiratory burst response. The real-time chemiluminescence assay was used to measure respiratory burst activity in macrophage and neutrophilic granulocyte-enriched head kidney cell fractions and total head kidney cell suspensions and proved to be a fast, reliable, automated multiwell microplate assay to quantitate fish health status modulated by β-glucans.  相似文献

For the first time, morpho-functional characterisation of haemocytes from the cockle Cerastoderma glaucum was performed to identify circulating cell types and to study their involvement in immune responses. Haemocyte mean number was 5.5 (x 10(5)) cells/mL haemolymph. Two main haemocyte types were found in haemolymph: granulocytes (85%), about 10 microm in diameter and with evident cytoplasmic granules, and hyalinocytes (15%), 8 to 14 microm in diameter, with a few or no granules. Most of the cytoplasmic granules stained in vivo with Neutral Red, indicating that they were lysosomes. On the basis of haemocyte staining properties, granulocytes and hyalinocytes were further classified as basophils and acidophils. Acidophil hyalinocytes were the largest haemocyte type (about 14 microm in diameter) and had an eccentric nucleus and a large cytoplasmic vacuole. Both granulocytes and hyalinocytes (except acidophils) were able to phagocytise yeast cells, although the basal phagocytic index was very low (about 2%). It increased significantly (up to 26%) after pre-incubation of yeast in cell-free haemolymph, suggesting that haemolymph has opsonising properties. Haemocytes also produced superoxide anion. Moreover, both granulocytes and hyalinocytes (except acidophils) were positive for some important hydrolytic and oxidative enzymes. Lysozyme-like activity was recorded in both cell-free haemolymph and haemocyte lysate, although enzyme activity in cell lysate was significantly higher. Results indicate that haemocytes from C. glaucum are effective cells in immune responses.  相似文献

The effect of a panel of pterins on xanthine oxidase was investigated by measuring formation of urate from xanthine as well as formazan production from nitroblue tetrazolium. The pterin derivatives, depending on their chemical structure, decreased urate as well as formazan generation: 200 μM neopterin and biopterin suppressed urate formation (90% from baseline) and formazan production (80% from baseline) as well. Their reduced forms, 7,8-dihydroneopterin and 5,6,7,8-tetrahydrobiopterin, showed a lesser but still strongly diminishing influence (40% from baseline). Another oxidized pterin namely leukopterin showed only a weak inhibitory effect. Xanthopterin, a known substrate of xanthine oxidase, had a strong effect on urate formation (80% inhibition), but a lesser effect on formazan production (30% reduction). When iron-(III)-EDTA complex was added to the reaction mixture all the effects were more pronounced. Superoxide dismutase, which removes superoxide anion by dismutation intooxygen, decreased formazan production in addition to pterin derivatives and had a small but enhancing effect on urate formation. Also the reductant N-acetylcysteine had an additive effect to pterins to diminish formazan production in a dose-dependent way. The results of our study suggest that depending on their chemical structure pterins reduce superoxide anion generation by xanthine oxidase.  相似文献

The in vitro production of the reactive oxygen metabolite superoxide (O2-) was confirmed in hemocytes from the schistosome intermediate host Biomphalaria glabrata. Active forms of the enzyme superoxide dismutase (SOD) inhibited reduction of nitroblue tetrazolium (NBT) to formazan in cells that had phagocytozed zymosan particles, whereas an inactivated form of SOD did not. Moreover, based on the prevalence of O2(-)-positive hemocytes and the relative intensity of NBT staining reactions, hemocytes from the Schistosoma mansoni-resistant 10-R2 strain of B. glabrata possessed an overall greater capacity for generating superoxide than did those from S. mansoni-susceptible M-line snails. Schistosoma mansoni excretory-secretory (E-S) products, released during in vitro transformation of miracidia to sporocysts, inhibited phagocytosis of zymosan particles and superoxide activity in hemocytes from both snail strains, but 10-R2 hemocytes maintained higher levels of phagocytosis and superoxide production than did M-line hemocytes. The dose-dependent decreases in phagocytosis observed in both snail strains in the presence of E-S products could not account fully for the concomitant decrease in superoxide levels detected, indicating that either a single E-S factor differentially affects phagocytosis and superoxide production, or that different E-S factors are involved in the specific interference of each of these hemocyte functions.  相似文献

Oxygen inhibits competitively the reduction of nitro blue tetrazolium chloride (nitro BT) by NADH and phenazine methosulfate (PMS). The oxygen-dependent inhibition is stronger in the presence of superoxide dismutase, whereas cyanide counteracts the oxygen interference. On the other hand, the oxidation of NADH mediated by PMS and dioxygen is affected only marginally by superoxide dismutase and cyanide. Therefore, it is concluded that the involvement of superoxide anions occurs at the level of nitro BT reduction via a nitro blue tetrazolinyl radical, as has been suggested by Picker and Fridovich [1984) Arch. Biochem. Biophys. 228, 155-158) and not at the level of PMS oxidation. The inhibition of the oxygen interference in the nitro BT reduction by cyanide is dependent on the cyanide concentration, whereas in nitrogen cyanide has no effect on the reduction. It is caused by competition between cyanide and oxygen to reduce or oxidize the nitro BT radical to either formazan with concomitant cyanogen production or nitro BT, respectively. For the histochemical localization and analysis of electropherograms of NAD(P)+-dependent dehydrogenase activities, the interference of oxygen can be avoided by anaerobic incubations or by the use of 5 mM nitro BT when incubating aerobically.  相似文献

A micro assay for the rapid and convenient determination of superoxide dismutase activity in limited amounts of biological material was devised and successfully employed. The combination of the formazan derivative colour formation induced by reaction of O2theta with nitroblue tetrazolium and a suitable analytical polyacrylamide gel electrophoresis system was used. It was possible to show that the reactivity of soluble superoxide dismutases in polyacrylamide gels was proportional to the enzyme concentrations employed. Bovine erythrocyte Cu, Zn-superoxide dismutase (EC 1.15.1.1) (erythrocuprein) served as a standard throughout. To measure the degree of superoxide dismutase activity, a gel-scanning apparatus was usedThe integrated scanning curve of the unstained portions of the gel proved linearly proportional to the logarithm of the superoxide dismutase activity in the range between 10(-12) and 7 X 10(-11) mol of the bovine enzyme. Although the absolute integral is dependent on the different staining conditions, the slope of the superoxide dismutase calibration curve is highly reproducible. Superoxide dismutase added to crude liver and brain homogenates could be fully detected using this assay. Thus, biological material including nucleic acids, enzymes, lipids etc. do not inhibit this reaction.  相似文献

Summary A study was made of the involvement of superoxide anions in the aerobic reduction of tetrazolium salts by NAD(P)H and phenazine methosulphate (PMS). On the basis of experiments with superoxide dismutase two mechanisms of tetrazolium reduction could be distinguished-one in which fully reduced PMS (PMSH) is the reducer and one in which superoxide anion is the reducer of tetrazolium salts. It is proposed that superoxide anion is formed after a PMSH-PMS+ dismutation reaction. The relative contributions of the two distinct pathways to tetrazolium salt reduction are controlled by the PMS redox state and the oxygen tension. The consequences of the presence of superoxide anions and scavengers of superoxide anions for quantitative dehydrogenase cytochemistry are discussed.  相似文献

Armoured ground crickets Acanthoplus discoidalis (Bradyporidae) have an arsenal of defence mechanisms in response to attack. Males but not females can stridulate when attacked, while both sexes will bite and regurgitate upon provocation. They will also autohaemorrhage. Here we have quantified these responses, examining how individuals of both sexes respond to repeated simulated predatory attack from the side (grabbing the legs with forceps) or from above (grabbing the animal by the pronotum). We found different responses depending on the method of attack. When attack was directed from the side (at the legs) the crickets can bite their attacker and males stridulate intensely. About 62% of such attacks elicited an autohaemorrhage response, where the crickets squirt 13±22 mg of acrid-smelling haemolymph 43±63 mm from seams in the connective tissue between the trochanter and coxa of each leg and from under the pronotum. By contrast, animals attacked from above could not turn and bite their attacker, and stridulation was also reduced in males. About 86% of such attacks elicited an autohaemorrhage response with 19±19 mg of haemolymph projecting 10±30 mm from the body. Autohaemorrhaging is an effective form of chemical defence against bearded dragon lizards Pogona vitticeps (Agamidae) and Aca. discoidalis haemolymph applied to Gryllus bimaculatus nymphs (which have no such chemical defence) successfully saved them from predation by striped skinks Trachylepis punctatissima (Scincidae).  相似文献

It has been reported in vitro that during the respiratory burst of phagocytic cells the superoxide anion production per cell shows a negative relation with the cell density. This process has been described as autoregulation. The aim of this work was to analyze the superoxide anion production in thioglycollate-elicited peritoneal macrophage exudates to evaluate the importance of the peritoneal cavity environment in the autoregulation process. 12-O-tetradecanoylphorbol-13-acetate (PMA) was used to stimulate the respiratory burst and superoxide anion production was measured evaluating the intracellular formazan deposits that precipitate as a result of nitro blue tetrazolium (NBT) reduction. We have demonstrated a negative correlation between superoxide anion production and cell density in the peritoneal cavity in macrophages challenged with PMA. The response of individual cells was analyzed by means of an image analyzer, measuring the amount of formazan per cell and cell-size changes during the process of activation. The results revealed that the decrease in individual cell response as a function of higher cell densities were due to a significant increase in the amount of basal reaction macrophages. Concomitantly, the number of reactive cells remained unchanged irrespective of the cell density of the population. A direct correlation between cell size and superoxide anion production was observed. This phenomenon was demonstrated in SENCAR and Balb/c strains. However, macrophages from SENCAR mice showed greater superoxide anion production than those from Balb/c.The differences between strains could be associated to the increased sensitivity to PMA tumor promotion of SENCAR mice. Based on this property, macrophages from SENCAR mice were stimulated with opsonized zymosan, a particulate stimulus that reflects the interaction macrophage-microorganism during the phagocytic process. This data will contribute to the knowledge of infection control. We conclude that variations in basal reaction cells modulates the macrophage activation response when excess macrophages are recruited to the peritoneum. This is demonstrated using different stimuli, thus suggesting that this response may be applied to a wide variety of stimuli-macrophage interactions. The differences between strains may be associated to the increased sensitivity to PMA tumor promotion of SENCAR mice.  相似文献
