Amplification of the second internal transcribed spacer (ITS2) of ribosomal DNA was used to compare seven samples of the Tetranychus kanzawai Kishida-- T. hydrangeae Pritchard & Baker mite complex from five different countries: Australia, the Congo, Indonesia, Japan and the USA. No morphological differences were detected between these mites and their ITS2 sequences displayed strong similarity except for a small nucleotide divergence of 0.2% in specimens from Australia and Indonesia. Reciprocal crosses and backcrosses between mites assumed to be T. kanzawai and T. hydrangeae respectively showed reproductive compatibility. Fertile hybrid females were obtained in all cases, indicating conspecificity of the mites tested. It is concluded that T. hydrangeae is a synonym of T. kanzawai. The evidence suggests that T. kanzawai originated in South-east Asia and probably spread throughout the world on Hydrangea spp. cuttings.  相似文献

Sequence variation of the complete second internal transcribed spacer (ITS2, 445 bp) of nuclear ribosomal DNA and part of the mitochondrial cytochrome oxidase I gene (COI, 350 bp) was examined in Amphitetranychus viennensis (Zacher) mites (Acari:Tetranychidae) from four French and four Japanese locations. Sequence analysis consistently revealed the separation of the samples in two major groups: French mites differed from Japanese by 3.8–4.1% of the nucleotide divergence in COI sequences. These two groups also displayed distinct ITS2 consensus sequences (2.1% nucleotide divergence). A few variations, not affecting the diagnostic sites around the consensus sequence, were revealed among cloned copies of the same individual. Reciprocal crosses and backcrosses between one French and two Japanese populations disclosed strong reproductive incompatibility. However, fertile hybrid females were obtained, indicating the conspecificity of the tested mites. Despite the presence of Wolbachia in the French strain, but not in the Japanese ones, our crosses did not display the unidirectional incompatibility typically produced by this microorganism, but rather a bidirectional – although asymmetrical – incompatibility pattern. The post-zygotic incompatibilities in A. viennensis cannot be explained by the presence of Wolbachia but to some extent by mite genome divergence resulting from limited gene exchange between allopatric populations. Experiments of Wolbachia elimination by antibiotic treatment and subsequent crosses with cured strains are still needed to fully understand the reproductive incompatibility patterns in this mite species.  相似文献

The relative importance of the braconid Cotesia sesamiae, a gregarious larval parasitoid of lepidopteran stemborers, varies greatly with region in Africa; while the most common parasitoid of noctuid stemborers in eastern Africa, it is rare in western Africa. Thus, several strains of C. sesamiae from Kenya are envisaged for introduction into western Africa. The present study investigates the reproductive compatibility between four populations of C. sesamiae from West Africa and Kenya with the noctuid Sesamia calamistis as the host using reciprocal crosses as well as backcrosses of hybrid females with males of the parental populations. Searching time of the male for the female and mating period varied significantly with couple and ranged between 0.78-1.9 min and 3.4-12.8 s, respectively. Crosses that involved females from inland Kenya (KI) did not yield any female offspring. However, backcrosses of hybrid female bearing a KI male genome with a KI male yielded both female and male offspring. Thus, there was a partial reproductive incompatibility between KI and West African populations which suggested that the latter were infected with Wolbachia sp. However, this should not affect the efficacy of a population introduced from East into West Africa, as there is a high degree of sib-mating in this gregarious parasitoid species. It was concluded that the regional differences in the relative importance of C. sesamiae was due to differences in the insect and plant host range of the different populations.  相似文献

Developmental success on 47 species of plant and reproductive compatibility were studied for four strains of the European red mite,Panonychus ulmi (Koch), collected from dwarf bamboo, elm and apple trees. The host range was variable in those strains and there was no plant species on which all four strains were able to reach maturity.Intra-populational crosses in the dwarf-bamboo strain and the two apple strains of mites, as well as crosses between the two apple strains, gave both female and male progeny. Inter-populational crosses between the dwarf-bamboo and apple strains produced only male progeny, as in the case where females of each strain were kept virgin, indicating that these two were reproductively incompatible and had an arrhenotokous reproductive system. When females of the elm strain were crossed with males of the elm, dwarf-bamboo or apple strain, no males were produced, as in the case for virgin females of the elm strain which had a thelytokous reproductive system. It is thus concluded that the strains derived from different host species were reproductively incompatible with one another.  相似文献

Recent studies have shown that certain host populations of the cereal rust mite Abacarus hystrix are highly specialized in their host use and it is likely that reproductive isolation exists between them. Here I verified this expectation by testing for reproductive barriers between ryegrass and quackgrass populations of A. hystrix. I performed reciprocal crosses between individuals from both populations and observed results of crosses. Leaves of the three grass species, ryegrass, quackgrass and wheat, were used as mating arenas. I used two criteria to determine reproductive barriers between strains: the proportion of female progeny and viability of progeny. If studied populations of this haplodiploid species are reproductively isolated a male-biased sex ratio and/or hybrid progeny of reduced viability would be expected. I found that in the presence of quackgrass and ryegrass pre-zygotic barriers between studied populations exists. On wheat I observed asymmetry in reproductive barriers. Between females from quackgrass and males from ryegrass a pre-zygotic barrier existed (only males obtained). However, the opposite reciprocal cross (females from ryegrass and males from quackgrass) produced progeny of both sexes. A male-biased sex ratio and low adult emergence suggests that post-zygotic mechanisms acted here. Low viability of progeny obtained from crosses in which females from ryegrass were engaged suggests that the origin of the female nymph acted as a predictor of hybrid inviability. The pattern of sterility observed in the cereal rust mite indicates that in the presence of three hosts (ryegrass, quackgrass and wheat) pre- and post-zygotic reproductive barriers between quackgrass and ryegrass populations of this mite exist. In addition to host fidelity (which acts as pre-zygotic barrier) there are post-zygotic barriers to gene flow.  相似文献

Collectively, populations of Acanthocyclops vernalis, a species complex of freshwater copepods, are remarkably similar as to morphology and DNA content, despite variability in chromosome number. Reproductive isolation had been reported among some populations, but with each new investigation the species boundaries and factors that may influence them appeared less clear. To clarify the pattern of biological species within this group of populations, we adopted a comprehensive approach and examined patterns of reproductive isolation in populations for which morphology, chromosome number, DNA content, and 18S rDNA sequences are known. In this study we established nine isofemale lines from four sites in Wisconsin and performed 266 crosses. Crosses within and among these lines were used to relate the degree of reproductive isolation to chromosome differences and to construct a model to explain the origin and maintenance of chromosome number variability. Different gametic and somatic chromosome numbers were observed among specimens within some isofemale lines. In a few cases, gametes with different haploid numbers were produced by a single female. Matings within isofemale lines always produced at least some reproductively successful replicate crosses (produced viable, fertile offspring). Crosses between lines from the same site showed reduced success relative to within-line crosses. Crosses between populations from distant sites showed limited genetic compatibility, producing viable, fertile F1 offspring but infertile F2 adults. One cross between lines with different chromosome numbers (one with 2n = 8 and one with 2n = 10) produced fertile viable offspring, which reproduced for at least 60 generations. These hybrids had either eight or nine chromosomes in the third generation of inbreeding, and eight chromosomes after 20 generations. These hybrids also had reduced nuclear DNA contents at the third generation, a level that persisted through the 20th generation. Successful backcrosses between some hybrids and their parental lines further demonstrated the potential for genetic compatibility among forms with different chromosome numbers. We propose a model in which alterations due to Robertsonian fusions, translocations, and/or loss of chromosomal fragments generate heritable variation, only some of which leads to reproductive isolation. Hence, some of the criteria traditionally used to recognize species boundaries in animals (morphology, DNA content, chromosome number) may not apply to this species complex.  相似文献

I examined the intrinsic postzygotic incompatibilities between two pupfishes, Cyprinodon elegans and Cyprinodon variegatus. Laboratory hybridization experiments revealed evidence of strong postzygotic isolation. Male hybrids have very low fertility, and the survival of backcrosses into C. elegans was substantially reduced. In addition, several crosses produced female-biased sex ratios. Crosses involving C. elegans females and C. variegatus males produced only females, and in backcrosses involving hybrid females and C. elegans males, males made up approximately 25% of the offspring. All other crosses produced approximately 50% males. These sex ratios could be explained by genetic incompatibilities that occur, at least in part, on sex chromosomes. Thus, these results provide strong albeit indirect evidence that pupfish have XY chromosomal sex determination. The results of this study provide insight on the evolution of reproductive isolating mechanisms, particularly the role of Haldane's rule and the 'faster-male' theory in taxa lacking well-differentiated sex chromosomes.  相似文献

Wolbachia are cytoplasmically transmitted bacteria that infect several species of mites. In the two-spotted spider mite Tetranychus urticae Koch this symbiont can induce reproductive incompatibility. Wolbachia-induced reproductive incompatibility is observed in crosses between Wolbachia-infected (W) males and uninfected (U) females. This incompatibility is expressed in F1 broods as male-biased sex ratios, an effect called cytoplasmic incompatibility (CI). However, in the two-spotted spider mite, Wolbachia-induced reproductive incompatibility may extend to the F2: broods of virgin F1 females from U×W crosses sometimes suffer increased mortality rates. This F2 effect is called hybrid breakdown (HB). Several isofemale lines derived from mites collected from rose and cucumber plants had been previously tested for CI. Life history and reproductive parameters of the generalist predatory mite Euseius (Amblyseius) finlandicus (Oudemans) were studied in the laboratory at 25 +/- 1 degrees C, with a 16L:8D photoperiod and 60 +/- 15% RH, to investigate its response to different food sources: an eriophyid mite Aceria sp., tulip pollen Tulipa gesnerana L., and two-spotted spider mite Tetranychus urticae Koch. Total developmental time of the immature stages was the shortest on eriophyid mites, followed by pollen, and then spider mites. Fecundity was highest on pollen (43.69 eggs; 1.63 eggs/female/day), then eriophyid mites (39.73 eggs; 1.37 eggs/female/day) and lowest on spider mites (18.16 eggs; 0.80 eggs/female/day). Intrinsic rate of increase (Rm), net reproductive rate (Ro) and finite rate of increase (lambda) followed the same pattern [pollen (0.168, 27.96 and 1.183, respectively), eriophyid mites (0.153, 20.81 and 1.167), spider mites (0.110, 9.44 and 1.119)]. Mean generation time (days) was the shortest on pollen (19.90), followed by eriophyid mites (20.02), and then spider mites (20.59). Average spider mite larvae consumed by E. finlandicus during immature stages were 9.18 for males and 11.85 for females. Adult E. finlandicus females consumed an average of 166.38 spider mite protonymphs during adult stage compared to an average of 66.55 by males. The number of prey protonymphs consumed per day by females was highest in the oviposition period, lower in the pre-oviposition period and the lowest in the post-oviposition period. The eriophyid mite as a prey recorded the shortest developmental time, while pollen as food recorded the highest oviposition rate in E. finlandicus. The potential of this predator as a biocontrol agent against T. urticae is discussed.  相似文献

Intracellular symbiotic bacteria belonging to the Cytophaga-Flavobacterium-Bacteroides lineage have recently been described and are widely distributed in arthropod species. The newly discovered bacteria, named Cardinium sp, cause the expression of various reproductive alterations in their arthropod hosts, including cytoplasmic incompatibility (CI), induction of parthenogenesis and feminization of genetic males. We detected 16S ribosomal DNA sequences similar to those of Cardinium from seven populations of five spider mite species, suggesting a broad distribution of infection of Cardinium in spider mites. To clarify the effect of Cardinium on the reproductive traits of the infected spider mites, infected mites were crossed with uninfected mites for each population. In one of the populations, Eotetranychus suginamensis, CI was induced when infected males were crossed with uninfected females. The other six populations of four species showed no reproductive abnormalities in the F(1) generation, but the possibility of CI effects in the F(2) generation remains to be tested. One species of spider mite, Tetranychus pueraricola, harbored both Cardinium and Wolbachia, but these symbionts seemed to have no effect on the reproduction of the host, even when the host was infected independently with each symbiont.  相似文献

Three lepidopteran stemborers, Busseola fusca Fuller (Noctuidae), Sesamia calamistis Hampson (Noctuidae), and Chilo partellus (Swinhoe) (Crambidae), were evaluated for their acceptability for oviposition and suitability for development by two populations of the larval endoparasitoid Cotesia sesamiae (Cameron) (Hymenoptera: Braconidae) occurring in the highveld (>1200 m) and lowveld (<600 m) regions of Zimbabwe. Mating studies were also conducted to determine reproductive compatibility between the populations. Both C. sesamiae populations preferred the noctuids to C. partellus for oviposition, possibly reflecting differences in evolutionary history. Although B. fusca was partially suitable for development of lowveld C. sesamiae, all three hosts were suitable for development of the highveld population. Crosses between highveld and lowveld C. sesamiae were compatible, and were generally not different from the intra-population crosses in developmental time, % adult emergence and sex ratio. However, broods were much larger when highveld males were used in the mating combinations. We conclude that although there is host overlap and probably a considerable degree of outbreeding between the two C. sesamiae populations, there are still significant genetic differences between them. Within Zimbabwe, it is unlikely that the deliberate introduction of either population outside its region of occurrence will give meaningful stemborer control.  相似文献

It is widely accepted that the genetic divergence and reproductive incompat- ibility between closely related species and/or populations is often viewed as an important step toward speciation. In this study, sexual compatibility in crosses between the southern XS population and the northern TA population of the polyandrous cabbage beetle Co- laphellus bowringi was investigated by testing their mating preferences, mating latency, copulation duration, and reproductive performances of post-mating. In choice mating ex- periments, the percentages ofmatings were significantly higher in intra-population crosses than in inter-population crosses. Both isolation index (/) and index of pair sexual isolation (/PSi) indicated partial mating incompatibility or assortative mating in crosses between the two different geographical populations. In single pair mating experiments, XS females in inter-population crosses mated significantly later and copulated significantly shorter than those in intra-population crosses. However, TA females in inter-population crosses mated significantly earlier and copulated longer than those in intra-population crosses, suggesting that larger XS males may enhance heterotypic mating. The lifetime fecundity was highest in XS homotypic matings, lowest in TA homotypic matings, and intermedi- ate in heterotypic rnatings between their parents. The inter-population crosses resulted in significantly lower egg hatching rate and shorter female longevity than intra-population crosses. These results demonstrated that there exist some incompatibilities in premating, postmating-prezygotic, and postzygotic stages between the southern XS population and northern TA population of the cabbage beetle Colaphellus bowringi.  相似文献

Whereas endosymbiont-induced incompatibility is known to occur in various arthropod taxa, such as spider mites, insects and isopods, it has been rarely reported in plant-inhabiting predatory mites (Acari: Phytoseiidae). Recent cross-breeding studies with the phytoseiid mite Neoseiulus paspalivorus De Leon revealed a complete post-mating reproductive isolation between specimens collected from three geographic origins—Northeast Brazil (South America), Benin and Ghana (West Africa)—even though they are morphologically similar. We carried out a study to assess to what extent these populations exhibit genetic differences and whether endosymbionts are involved in the incompatibility. First, we used the mitochondrial cytochrome oxidase I (COI) gene to assess genetic diversity among the three populations. Second, we used a PCR-based method to check for the presence of Wolbachia and/or Cardinium in these populations, and we determined their phylogenetic relationships using specific primers for Wolbachia and Cardinium 16S rDNA genes. Third, we also conducted a test using an antibiotic (tetracycline) in an attempt to eliminate the symbionts and evaluate their effects on the reproductive compatibility of their host. Based on the DNA sequences of their COI genes, specimens of the three populations appear to be genetically similar. However, the 16S rDNA gene sequences of their associated endosymbionts differed among the three populations: the Benin and Brazil populations harbour different strains of Wolbachia symbionts, whereas the Ghana population harbours Cardinium symbionts. In response to antibiotic treatment females of each of the three populations became incompatible with untreated males of their own population, similar to that observed in crossings between females from one geographic population and males from another. Compatibility was restored in crosses involving uninfected Brazil females and uninfected Benin males, whereas the reciprocal crosses remained incompatible. Cardinium symbionts seem to be essential for oviposition in the Ghana population. It is concluded that their associated bacterial symbionts are the cause of the post-mating reproductive isolation previously observed among the three geographic populations. This insight is relevant to biological control of coconut mites for which N. paspalivorus is an effective predator, because introducing one geographic strain into the population of another (e.g. in field releases or mass cultures) may cause population growth depression.  相似文献

Crosses between populations or species often display an asymmetry in the fitness of reciprocal F1 hybrids. This pattern, referred to as isolation asymmetry or Darwin''s Corollary to Haldane''s Rule, has been observed in taxa from plants to vertebrates, yet we still know little about which factors determine its magnitude and direction. Here, we show that differences in offspring size predict the direction of isolation asymmetry observed in crosses between populations of a placental fish, Heterandria formosa. In crosses between populations with differences in offspring size, high rates of hybrid inviability occur only when the mother is from a population characterized by small offspring. Crosses between populations that display similarly sized offspring, whether large or small, do not result in high levels of hybrid inviability in either direction. We suggest this asymmetric pattern of reproductive isolation is due to a disruption of parent–offspring coadaptation that emerges from selection for differently sized offspring in different populations.  相似文献

We assessed prezygotic (probability of spawning) and postzygotic (hatching success) reproductive isolation among the three ecologically and morphologically similar species in the Fundulus notatus species complex. We employed a multi-generation breeding experiment to test the hypotheses that karyotypic differences, body size differences, or geographic isolation among populations will increase pre or postzygotic reproductive barriers. Overall, prezygotic barriers were strong and postzygotic barriers weak in crosses of non-hybrid heterospecifics (F1 hybrid crosses) while prezygotic barriers were weaker and postzygotic barriers stronger in crosses involving hybrid individuals (F2 hybrid crosses and backcrosses). Prezygotic barriers among the two smaller species (Fundulus notatus and F. euryzonus) broke down rapidly; first generation hybrids spawned (F2 hybrid crosses and backcrosses) as frequently as parental forms in intraspecific crosses. There was no increase in postzygotic barriers among species with cytogenetic differences. There were increased prezygotic, but not postzygotic, barriers among geographically isolated populations of one species. While pure males and females were just as likely to spawn with hybrids, some types of hybrid females suffered from increased sterility, but not inviability, over hybrid males. Female sterility was only seen in hybrids with a Fundulus euryzonus parent, while other female hybrids produced viable eggs.  相似文献

Aceria guerreronis Keifer (Acari: Eriophyidae) is considered a major pest of the coconut (Cocos nucifera L.), and the use of pesticides is the current method to control it. However, no standard toxicological tests exist to select and assess the efficiency of molecules against the coconut mite. The aim of this study was to develop a methodology that allows for the evaluation of the relative toxicity of acaricides to A. guerreronis through rapid laboratory procedures. We confined A. guerreronis on arenas made out of coconut leaflets and tested two application methods: immersing the leaf fragments in acaricides and spraying acaricides on the leaf fragments under a Potter spray tower. In the latter application method, we sprayed leaf fragments both populated with and devoid of mites. We evaluated the comparative toxicity of two populations (Itamaracá and Petrolina, Pernambuco, Brazil) by spraying on leaflets without mites and submitted the mortality data to probit analysis after 24 h of exposure. No difference was observed in the LC50, regardless of whether the leaflets were immersed or sprayed with acaricide (abamectin, chlorfenapyr or fenpyroximate). The toxicity of chlorfenapyr and fenpyroximate did not differ, irrespective of whether it was applied directly to the leaflet or to the mite; however, the toxicity of abamectin was higher when applied directly to the mite. Chlorpyrifos and abamectin toxicities were lower for the Petrolina population than for the Itamaracá population. Immersing and spraying coconut leaflets can be used to assess the mortality of A. guerreronis under laboratory conditions.  相似文献

Predatory mites identified as Neoseiulus paspalivorus DeLeon (Phytoseiidae) have been considered as agents for classical biological control of the coconut mite, Aceria guerreronis Keifer (Eriophyidae), in Africa and elsewhere. Preliminary identification of geographically distinct populations as belonging to the same species (N. paspalivorus) was based on their morphological similarity. However, laboratory studies recently conducted have shown large differences in feeding behaviors and biological characteristics among individuals collected from three geographic origins: Brazil (South America), Benin and Ghana (West Africa). As morphologically similar specimens do not necessarily belong to the same species, we evaluated under laboratory conditions, reproductive compatibility between the specimens from three geographic locations to ascertain their conspecificity. Morphological measurements were also made to determine whether there is a means of discriminating between them. Inter-population crosses showed complete reproductive isolation between the three geographic populations, but interpopulation discontinuities in morphometric characters were absent. These results indicate that the tested specimens are distinct biological entities despite morphological similarity. Further molecular genetic studies are therefore proposed, including screening for endosymbionts and assessment of genetic differentiation, to determine the cause of reproductive incompatibility and to clarify the taxonomic relationship between those populations.  相似文献

Abstract The population of the citrus red mite, Panonychus citri (McGregor), does not increase on pear from spring to mid-summer but thereafter increases abruptly. To elucidate this phenomenon, we compared the performance of the mites on pear leaves with that on citrus leaves, at different time throughout the pear-growing season. No significant difference was detected between the oviposition rate on pear and that on citrus throughout the season. However, the survival rate of ovipositing females that had fed on pear and the hatch rate of eggs laid by those females were significantly lower than those for females that had fed on citrus, until August. However, no significant difference was observed thereafter. The results showed that the decline of the population of citrus red mite before autumn is due to the high mortality of adult females that had fed on pear leaves and the low hatch rate of the eggs produced by those females.  相似文献

The chalcid, Oomyzus sokolowskii Kurdjumov has been recorded in many parts of the world as a major larval-pupal, gregarious endoparasitoid of the diamondback moth, Plutella xylostella (Linnaeus), a serious pest of brassica vegetable crops worldwide. This study investigated intraspecific variation between two populations of O. sokiolowskii, one from Cape Verde Islands, West Africa and the other from Hangzhou, China. In all crosses and backcrosses between the two geographical populations, the numbers of progeny and sex ratio of progeny were similar to those obtained within each of the populations, demonstrating complete reproductive compatibility between the two populations. The two populations showed similar responses to temperature with respect to development time and survival of immature stages. Observations on the interactions between the two O. sokolowskii populations and Cotesia plutellae (Kurdjumov), another major parasitoid of P. xylostella, showed that neither population could achieve successful parasitism of P. xylostella larvae already parasitized by C. plutellae. However, both O. sokolowskii populations could achieve hyperparasitism by ovipositing into a mid-late stage larva of C. plutellae developing inside the primary host. Contrary to earlier reports, no evidence of intraspecific variations in ability to hyperparasitize between these two populations of O. sokolowskii was found.  相似文献
