Vasopressin and nonmammalian hormone vasotocin are known to increase the water permeability of mammalian collecting ducts, frog skin and the urinary bladder. Neurohypophysial nonapeptides have also been shown to interfere with the regulation of renal ion transport. The subject of this study was a search for vasopressin and vasotocin analogues with selective effects on renal water, sodium and potassium excretion. During this study, we synthesised the following peptides: 13 vasotocin analogues modified at positions 4 (Thr or Arg), 7 (Gly or Leu) and 8 (d ‐Arg, Lys or Glu); 4 vasopressin analogues modified at positions 4 and 8; and 9 peptides shortened or extended at the C‐terminal or with substitutions for Gly‐NH2. Most of these peptides had mercaptopropionic acid (Mpa) instead of Cys in position 1. The effects of these nonapeptides on renal water, sodium and potassium transport were evaluated in in vivo experiments using Wistar rats. Some nonapeptides possessed antidiuretic, natriuretic and kaliuretic activities ([Mpa1]‐arginine vasotocin, [Mpa1, homoArg8]‐vasotocin, [Mpa1, Thr4]‐arginine vasotocin and [Mpa1, Arg4]‐arginine vasopressin). Substitutions at positions 4 and 8 increased the selectivity of peptide actions. The antidiuretic [d ‐Arg8]‐vasotocin analogues had no effects on sodium excretion. [Mpa1, Arg4]‐arginine vasotocin was antidiuretic and kaliuretic but not natriuretic. [Mpa1, Glu8]‐oxytocin had weak natriuretic activity without any effects on water and potassium transport. In accordance with the data obtained, synthesised vasotocin analogues could be good candidates for pharmaceuticals selectively regulating renal sodium and potassium transport, which is of clinical importance. The effects of systemic infusions of the avian antidiuretic hormone arginine vasotocin on water intake of domestic ducks were investigated under steady conditions of water balance in which angiotensin II was effective as a dipsogen. The study proceeded from the consistent stimulatory effect of arginine vasotocin on angiotensin II-responsive neurons found in the subfornical organ of ducks, suggesting brain-intrinsic vasotocinergic control of these neurons which are also accessible to circulating agents because of the lacking blood-brain barrier. Levels of circulating arginine vasotocin of about 2700 pg·ml-1 which were close to the threshold for activation of subfornical organ neurons in vitro, induced weak but significant drinking responses. Even at this high arginine vasotocin level circulatory effects were absent, thereby excluding their interference with water intake. Arginine vasotocin plasma levels of about 60 pg·ml-1 significantly attenuated the dipsogenic action of angiotensin. While drinking in response to high pharmacological levels of arginine vasotocin is assumed to mimic a stimulatory innervation of angiotensin-responsive subfornical organ neurons by brain-intrinsic vasotocinergic axons, attenuation of angiotensin-induced drinking by high physiological arginine vasotocin levels cannot be explained by its action on central neurons, but may be secondary to body fluid retention caused by the antidiuretic action of arginine vasotocin.Abbreviations ADH antidiuretic hormone - ANGII angiotensin II - AVP arginine vasopressin - AVT arginine vasotocin - BBB blood-brain barrier - HR heart rate - ICV intracerebroventricular - IV intravenous - MAP mean arterial pressure - SFO subfornical organ  相似文献

Administration of 5 ml/100 g body weight of 1% glucose solution to stomach produced the same diuretic kidney response in fasted Wistar rats as administration of the same amount of water. Intragastric administration of arginine vasopressin along with the water load evoked an antidiuretic response. Arginine vasopressin in the same volume of glucose induced no kidney response difference as compared with the hormone action in experiments with water load. 0.1 nmol of arginine vasotocin, having been itroduced into the rat isolated ileum, prevented the effect of glucose on the hormone absoption. 0.1 nmol of arginine vasotocin, having been introduced into the frog isolated ileum along with isotonic glucose solution, increased the hormone absorption; fructose did not affect this process whereas mannitol decreased absorption ofarginine vasotocin. This absorption was also reduced by intraileal introduction of arginine vasotocin with the hypotonic Ringer solution. The findings suggest that glucose in the rat gastrointestinal tract does not affect arginine vasopressin absorption in vivo, whereas in the frog ileum glucose increases arginine vasotocin absorption in vitro.  相似文献

Male albino rats received vasopressin, vasotocin, pressinoic acid or placebo and were tested on an aversively motivated brightness discrimination task. Treatment with both vasopressin and vasotocin had no effect on acquisition but facilitated the reversal of the discrimination. Pressinoic acid had an inconsistent effect. The results are interpreted to show that the C terminal of the peptides vasopressin and vasotocin influence potency of these peptides. Furthermore, the results are interpreted as showing that both vasotocin and vasopressin influence selective attention during aversively motivated tasks.  相似文献

In experiments on non-anesthetized Wistar white rats there was studied reaction of kidney to an intramuscular injection of arginine vasotocin or arginine vasopressin at doses from 0.001 to 0.05 µg/100 g body mass on the background of a water load. Water (5 ml/100 g body mass) was administered through a catheter into stomach to suppress secretion of endogenous antidiuretic hormone (ADH). In experiments with water administration, diuresis increased due to a decrease of osmotic permeability of renal tubules and to excretion of osmotically free water, with the constant clearance of sodium ions. Injection of 0.05 µg arginine vasopressin led to a marked decrease of diuresis due to a rise of reabsorption of osmotically free water without elevation of excretion of osmotically active substances. Injection of the same dose of arginine vasotocin resulted in no increase of diuresis; however, reabsorption of osmotically free water and excretion of osmotically active substances including sodium ions were more pronounced. Hence, both vasotocin and vasopressin increased osmotic permeability of the tubular epithelium, but vasotocin, unlike vasopressin, promoted reduction of reabsorption of sodium ions and their loss with urine. A suggestion is made that one of the reasons for replacement in mammals of the molecular ADH forms (vasotocin by vasopressin) was the absence of the pronounced natriuretic effect in arginine vasopressin. This was of crucial significance to preserve sodium ions in the organism, to maintain water–salt balance in animals adapted to the terrestrial life, and to provide not only osmo-, but also volumoregulation.  相似文献

The present study was designed to determine whether the responses of the avian kidney to circulating arginine vasotocin (AVT), under different osmotic conditions, involve an interaction with prostaglandins (PGs). The renal effects of intravenously infused AVT at a dose of 0.1 ng.kg–1.min–1 for 45 min were compared in Pekin ducks given maintenance infusions of either 200 mosmol NaCl or glucose at 1 ml.min–1, with and without PG inhibition by indomethacin. Birds infused with glucose responded with similar degrees of AVT-induced antidiuresis with and without indomethacin, however the urinary excretion of sodium was significantly reduced when PG production was prevented. In ducks receiving saline without indomethacin, the antidiuretic response to AVT was markedly less than that in the glucose-infused birds, however, indomethacin treatment increased the degree of antidiuresis to a level similar to that in the glucose-infused ducks. The results indicate that PGs have important renotropic actions in birds and in particular modulate the antidiuretic effect of AVT in salt- and volume-loaded animals.Abbreviations ANP atrial natriuretic peptide - AVT arginine vasotocin - PGs prostaglandinsCommunicated by I.D. The effect of dilution of the interstitial fluids on the responsiveness of the toad urinary bladder to antidiuretic hormones has been examined in vivo and in vitro. Toads were given periodic injections with vasopressin while in water so that their plasma osmolality fell below 190 mosmoles/kg H2O. The hydraulic conductivity of bladders which had been removed from the animal and fixed with 1% glutaraldehyde was 10-fold less in overhydrated toads than in normally hydrated controls. A similar inhibitory phenomenon was observed in in vitro studies, when the tonicity of Ringer's fluid in which the bladders were suspended was lowered from its isotonic value. Mannitol, but not urea, could be effectively substituted for one-half of the NaCl content of Ringer's fluid. In other experiments it has been shown that the responsiveness of the bladder to vasotocin is depressed during bulk water movement across the tissue. This "flux inhibition" was found to depend upon the velocity and the duration of water flow from mucosa to the serosa. It is suggested that the responsiveness of the toad bladder to antidiuretic hormones diminishes as the effective osmotic pressure of the interstitial fluids declines.  相似文献

The significance of perfection of osmoregulation in vertebrates for an increase of stability of the cell volume is substantiated. The functional significance of different types of osmoregulation is shown and the role of the isoosmotic regulation in sturgeons at their sea period of life is substantiated. A new hypothesis is put forward on the origin of the antidiuretic effect of vasotocin as a contreffect to autacoids. Presented are the data on the practical application of the results of study of evolution of the water–salt balance for space medicine, treatment of ischemic insult, nocturnal enuresis in children, prevention of nephrotoxic effect of antitumor drugs. The effects of dehydration and hemorrhage on plasma ionic, osmotic, and antidiuretic hormone (arginine vasotocin) concentrations and of hemorrhage on salt gland secretion and glomerular filtration rate were evaluated in glaucous-winged gulls, Larus glaucescens. Dehydration for 24 h did not affect plasma ionic, osmotic or arginine vasotocin concentrations; 72 h dehydration significantly elevated plasma osmolality, plasma sodium and chloride concentrations, and plasma arginine vasotocin concentration, but did not affect plasma potassium concentration. Constant infusion of 0.8 mol·l-1 NaCl increased plasma arginine vasotocin concentration and produced salt gland secretion in seven gulls; four secreted well, while three secreted less well. Removal of 20% blood volume during saline infusion immediately reduced (P<0.001) salt gland secretion rate in all gulls. After bleeding, good secretors maintained glomerular filtration rate and urine flow rate; the poorer secretors increased glomerular filtration rate and became diuretic. Blood replacement returned salt gland secretion rate to the prebleeding level (P<0.05) without affecting salt gland secretions sodium concentration in gulls which secreted well, but did not restimulate salt gland secretion in gulls which secreted poorly. Reinfusion of blood had no effect on glomerular filtration rate. Desacetyl alpha-MSH predominates over alpha-MSH during development, but whether it is biologically active and has a physiological role is unclear. We compared the effects of 0.3 microg.g(-1).day(-1) desacetyl alpha-MSH with that of 0.3 microg.g(-1).day(-1) alpha-MSH on postnatal body growth by administering the peptides subcutaneously daily for postnatal days 0-14 and also used a two-dimensional gel electrophoresis gel-based proteomic approach to analyze protein changes in hypothalami, the relay center for body weight and growth regulation, after 14 days of treatment. We found that the growth rate between days 1 and 10 was significantly decreased by desacetyl alpha-MSH but not by alpha-MSH, but by day 14, a time reported for development of a mature pattern of hypothalamic innervation, both peptides had significantly increased neonatal growth compared with PBS-treated control rats. Desacetyl alpha-MSH significantly increased spleen weight, but alpha-MSH had no effect. alpha-MSH significantly decreased kidney weight, but desacetyl alpha-MSH had no effect. Both desacetyl alpha-MSH and alpha-MSH significantly decreased brain weight. By 14 days, both peptides significantly changed expression of a number of hypothalamic proteins, specifically metabolic enzymes, cytoskeleton, signaling, and stress response proteins. We show that peripherally administered desacetyl alpha-MSH is biologically active and induces responses that can differ from those for alpha-MSH. In conclusion, desacetyl alpha-MSH appears to be an important regulator of neonatal rat growth.  相似文献

We studied the effect of alpha-melanotropin hormone (alpha-MSH) on experimental autoimmune oophoritis (EAO), an inflammatory process induced in female rats. During proestrus, serum levels of LH and progesterone in rats with EAO were higher than those of control rats. However, administration of alpha-MSH to these rats decreased the levels of LH. Similarly, in the following diestrus, rats with EAO had high levels of LH but treatment with alpha-MSH decreased the levels to diestrus 2 control values. Treatment with alpha-MSH also reduced the LH levels of control rats in diestrus 2 compared to untreated controls. However, alpha-MSH treatment had no effect on progesterone levels of either control or rats with EAO. Thus, although alpha-MSH induced notable changes in levels of LH, this decrease was unable to block the illness.  相似文献

The role of endogenous pyrogens induced by gram-positive bacterial pyrogens is not known. Intravenous alpha-MSH (2.5 micrograms) significantly reduced only the first phase of the biphasic thermal response to IV S. aureus cell walls (5 x 10(7)). Intracerebroventricular alpha-MSH (200 ng) had no effect on the fever response. The fall in serum iron concentration was significantly attenuated by the IV alpha-MSH but was not affected by the ICV alpha-MSH. Intravenous alpha-MSH had no effect on fever or the serum iron response caused by muramyl dipeptide (MDP). We conclude that the first phase of the thermal response to S. aureus cell walls is mediated by an endogenous pyrogen (EP) and the second phase of the response by a mechanism not involving EP, but possibly a muramyl peptide.  相似文献

alpha-MSH reduces fever in rabbits when administered IV, ICV, or by gavage; however, the applicability of this finding to higher species, specifically to primates, has not been determined. In this study, we chose the squirrel monkey as an appropriate primate model since it responds reliably to peripheral administration of bacterial endotoxins that cause fever in man. From pilot studies, doses of S. typhosa endotoxin necessary to produce maximum fever and doses of alpha-MSH which did not cause hypothermia were determined for each animal. In the main experiments endotoxin was given via an indwelling catheter in the saphenous vein, followed by alpha-MSH injections when the rectal temperature increased 0.3 degrees C. alpha-MSH (100-400 micrograms) reduced the area under the fever curve an average of 50.0%, but had no effect on afebrile temperature. Molar equivalent amounts of the antipyretic drug acetaminophen had little effect on fever. These findings support the idea, based on research on rabbits, that alpha-MSH has a role in central modulation of fever.  相似文献

Injections of arginine vasotocin into bullfrogs had no effect on plasma glucose after 30 min. but caused hyperplycemia after 120 min. Plasma levels of growth hormone were elevated earlier (30 min.) and returned to control levels after 120 min. Plasma free fatty acids were unaltered following vasotocin administration. It is suggested that the hyperglycemic effect of vasotocin may be mediated via growth hormone.  相似文献

The effects of intraperitoneal (i.p.) administration of kainic acid (KA) and alpha-melanocyte-stimulating hormone (alpha-MSH) alone or in combination, on core temperature of freely moving rats were examined. KA or saline was administered once (10 mg/kg) and alpha-MSH or saline was given repeatedly i.e. 10 min before and 10, 30 and 60 min after the administration of saline or KA. Two doses of alpha-MSH were used: 0.5 and 2.5 mg/kg. KA alone produced a biphasic effect on core temperature, i.e. an initial short-lasting hypothermia followed by hyperthermia that lasted about 6 h. The higher dose of alpha-MSH had a potentiating effect on KA-induced hypothermia, while the lower dose of alpha-MSH increased the hyperthermia produced by KA. alpha-MSH administered alone produced a late (3 h), dose-dependent increase in core temperature. It is conceivable that repeated administration of alpha-MSH in the doses used in our study may cause a cumulative effect in raising body temperature for a limited period of time.The previously described interactions between KA and alpha-MSH, respectively, with dopaminergic and serotoninergic systems may account for the effects on core temperature in rats observed in our study.  相似文献

The effects of arginine vasotocin (AVT) on the estrogen-induced surge of LH and FSH were examined in ovariectomized adult rats. Two and one-half weeks after ovariectomy, animals that were treated with a single subcutaneous (s.c.) dose of 5 μg of estradiol benzoate (EB) exhibited a surge of LH and FSH at 1700 and 1900 hours, respectively, two days after the administration of the EB. AVT, antidiuretic hormone (ADH) and oxytocin (OT) were administered s.c. in 1 μg dose every 4 hours beginning at 1500 hours on day 1 after EB treatment and then every 2 hours beginning at 1200 hours on day 2 after EB treatment. AVT completely prevented the LH surge at 1700 hours but was without effect on the FSH surge at 1900 hours on the day 2 after steroid treatment. Neither ADH nor OT had any significant effect on the afternoon surge of these hormones. It is postulated that AVT may interfere with the mechanisms mediating the estrogen-induced afternoon surge of LH in ovariectomized rats.  相似文献

Summary Extracts of the carp urophysis elicit a marked decrease in urine flow in the anaesthetized hydrated rat. Reproducible dose-dependent responses are obtained within the range of 2 to 16 g of acetonedried carp urophysis per 100 gBW of the rat. The carp urophysial antidiuretic substance is peptidic, and is different from the neurohypophysial peptides. The bulk of antidiuretic activity is located in the electrondense granules in the carp urophysis. The antidiuretic substance, probably urotensin I, is found generally in teleost urophyses. We investigated dose-dependent effects of alpha-melanocyte-stimulating hormone (alpha-MSH) on habituation in the Texas toad, Bufo speciosus. Additionally, we determined changes in plasma and brain levels of alpha-MSH following peripheral administration of the peptide or following exposure to an ether stressor. The ability of alpha-MSH to facilitate acquisition of habituation was dose dependent. Plasma alpha-MSH concentrations were elevated within 5 min of dorsal lymph sac injection and remained elevated up to 600% over controls after 30 min. Administration of 50 microgram alpha-MSH had no effect on plasma corticosterone levels. Radiolabeled alpha-MSH was detected in cerebrospinal fluid microdialysates within minutes of peripheral injection. Concentrations of alpha-MSH in the telencephalon and preoptic area were significantly lowered after ether exposure, whereas levels in the optic tectum, thalamus/hypothalamus, brainstem, and plasma were unchanged. We conclude that alpha-MSH administered peripherally facilitates habituation in a dose-dependent fashion. Our results confirm that the effects of alpha-MSH are independent of corticosterone secretion. The peptide is cleared rapidly into the bloodstream and enters the cerebrospinal fluid after dorsal lymph sac injection. Neuronal alpha-MSH may help toads gather information about their environment when exposed to certain stressors.  相似文献

Arginine vasotocin, 0.02--1 nM, increases osmotic water permeability of frog urinary bladder, arginine vasotocin after a simultaneous addition to the mucosal and serosal Ringer solutions rises the water permeability to a lesser degree than on the hormone addition only to the serosal solution. 1 nM remestyp, an agonist of V1-receptors, from the apical membrane decreases the hydroosmotic effect of arginine vasotocin added to the serosal Ringer solution. When added to the mucosal solution, combination of the same concentrations of arginine vasotocin and SR 49059, an antagonist of V--receptors, or desmopressin, agonist of V2-receptor alone, increases the effect of the same concentration of arginine vasotocin added to the serosal solution. 1 nM arginine vasotocin at the luminal membrane increases secretion into the Ringer solution of prostaglandin E, and prostaglandin E1 but not of prostaglandin F2 alpha. The data obtained indicate the presence of the arginine vasotocin receptors responsible for the hydroosmotic effect only in the basolateral membranes, while arginine prostaglandin E, participation is shown in modulation of the arginine vasotocin effect.  相似文献

Volemia and osmolality homeostasis is ensured in vertebrates through neuroendocrine reflexes, involving an afferent neural branch from baro- and osmo-receptors to hypothalamus and an efferent endocrine branch from secretory neurons to target hydroosmotic cells equipped with receptors and effectors. Whereas the osmoregulatory system in the tadpole comprises three organs, namely gut, kidney and gills, as in freshwater fishes, the adult displays a quaternary strategy with gut, kidney, urinary bladder and skin. In particular, the cutaneous permeability entails a great evaporative water loss when the animal is in the open air, loss that must be compensated by water reabsorption through the nephron and the urinary bladder and mainly by water uptake through the skin. Adaptation occurred at the level of these organs by regulation of their permeability through neurohypophysial hormones. Aside from vasotocin, active on the three organs, all anuran Amphibia possess hydrin 2 (vasotocinyl-Gly), a peptide resulting from a down-regulation of provasotocin processing. Exceptionally Xenopus laevis, a permanent aquatic toad, has hydrin 1 (vasotocinyl-Gly-Lys-Arg) instead of hydrin 2. Hydrins are somewhat more active than vasotocin on water permeation of skin and bladder but are devoid of antidiuretic activity. Adaptive evolution has created, along with the vasotocin-nephron system, preserved in all terrestrial non-mammalian tetrapods, additional functions such as the hydrin-skin and hydrin-bladder rehydration mechanisms. Specific hydrin receptors might exist in the skin and the bladder, different from those of vasotocin in the kidney. It is assumed that the water channel recruitment mechanism, found for vasopressin acting on the collecting duct principal cells in mammals, is also involved when vasotocin and hydrins stimulate their hydroosmotic target cells and that hormone-regulated aquaporin 2-like proteins could be identified in the three osmoregulatory organs of amphibians.  相似文献
