We describe the structure of two contrasting (elevation, topography,climate, vegetation, soil) alpine forest–pasture ecotones located in theCentral Pyrenees (sites Ordesa, O, and Tessó, T). We define ecotonestructure as the spatial distribution of trees of different size classes andgrowth-forms and the relationship between these aspects and the spatialdistribution of understory vegetation and substrate. The studied ecotones aredominated by Pinus uncinata Ram. and have been littleaffected by anthropogenic disturbances (logging, grazing) during this century.One rectangular plot (30 × 140 m) was located within eachsite with its longest side parallel to the slope and encompassing treeline andtimberline. The distribution of size and growth-form classes at site O followeda clear sequence of increasing size downslope from shrubby multistemmedkrummholz individuals to bigger arborescent trees. At site O, regeneration wasconcentrated near the krummholz area and over rocky substrates. This suggeststhat krummholz may modify microenvironment conditions and increase seedlingsurvival. At site T, regeneration was abundant above the treeline where thecover of the dominant understory shrub (Rhododendronferrugineum) decreased. In both ecotones the diversity of plants washigher above the treeline than in the forest and decreased going downslopecoinciding with the increase of P. uncinata cover. Thereduction of plant diversity appeared above the current timberline. At site O,the decrease was steep and spatially heterogeneous what may be due in part tothe edaphic heterogeneity. At site T the change was abrupt though smaller. Therelationships between the plant community and tree regeneration should be takeninto account in future ecological studies of treeline pattern.  相似文献

Enteropeptidase (enterokinase EC 3.4.21.9), catalyzing trypsinogen activation, exhibits unique properties for high efficiency hydrolysis of the polypeptide chain after the N-terminal tetraaspartyl-lysyl sequence. This makes it a convenient tool for the processing of fusion proteins containing this sequence. We found the enteropeptidase-catalysing degradation of some bioactive peptides: cattle hemoglobin beta-chain fragments Hb (2–8) (LTAEEKA) and Hb (1–9) (MLTAEEKAA), human angiotensin II (DRVYIHPF) (AT). Model peptideswith truncated linker WDDRG and WDDKG also were shown to be susceptible to enteropeptidase action. Kinetic parameters ofenteropeptidase hydrolysis for these substrates were determined.K m values for all substrates with truncated linker (10-3 M) are an order of magnitude higher thancorresponding values for typical enteropeptidase artificial peptide or fusion protein substrates with full enteropeptidase linker –DDDDK– (K m 10-4 M). k cat values for AT, Hb (2–8), WDDRG and WDDKG are 30–40 min-1. But one additional amino acid residue at both N- and C-terminus of Hb (2–8) results in a drastic increase of hydrolysis efficiency: k cat value for Hb (1–9) is 1510 min-1. Recent study demonstrates the possibility of undesirable cleavage of target peptides or proteins containing the above-mentioned truncated linker sequences; further, the ability of enteropeptidase to hydrolyse specifically several biologically active peptides in vitro along with its unique natural substrate trypsinogen was demonstrated.  相似文献

Influence of krummholz mat microclimate on needle physiology and survival   总被引:1,自引:0,他引:1  
Summary Microclimate and photosynthesis of krummholz mat growth forms of Picea engelmanii (Parry) and Abies lasiocarpa [Hook.] Nutt. were investigated to determine structural features which may aid survival in alpine environments. The structure of krummholz mats was described in terms of the vertical distribution of leaf area index and leaf area density, which exceeded 50 m-1 (based on total leaf surface area) near the canopy surface and approached zero below 30 cm from the surface in both species. Photosynthetic photon flux density (PPFD, 0.4–0.7 m wavelengths) and wind decreased by an average of 6 and 50-fold, respectively, between 1 m above and 10 cm below mat surfaces in both species. Needle temperatures on a P. engelmannii krummholz mat during July averaged about 2°C above air temperature during the day, with a maximum overtemperature of greater than 20°C above T air during one sunlit period. At night, needle temperatures averaged 3–4°C below T air.Net photosynthesis in year-old P. engelmannii shoots reached a maximum at 15–20°C during July and August. Surface shoots were light saturated at near 1200 moles m-2s-1 PPFD, and had higher photosynthetic rates than subsurface, predominantly shaded shoots above 800 moles m-2s-1. Shade shoots had higher photosynthetic rates when PPFD was below 600 moles m-2s-1, and at 250 moles m-2s-1 shade shoots maintained about 50% of the net photosynthetic rate of sun shoots at light saturation. Shade shoots appeared capable of benefitting photosynthetically from elevated temperatures within krummholz mats despite relatively low light levels. Especially rapid photosynthesis may occur when canopy needles are illuminated by sunflecks and needle temperatures rise by 10° C or more.Snow cover appears crucial for the survival of needles during winter. Snow accumulated within krummholz needle canopies before the sub-canopy zone of unfoliated branches became filled. The concentrated needle growth in the krummholz canopy captured snow in early autumn without support from ground-level snowpack. Early snow cover in both species prevented cuticle abrasion and resulted in high winter needle water contents and viabilities for subsurface compared to surface needles which became abraded, severely dehydrated, and had high mortality between December and February, especially on windward sides of shoots.Extremely high concentrations of needles within krummholz mat canopies created an aerodynamic structure which elevated needle temperatures to more optimal photosynthetic levels in summer and resulted in more efficient snow accumulation in winter. These factors appear crucial for winter needle survival. Thus, krummholz mats appear to be an important adaptation in growth form which provides survival benefits in both summer and winter.  相似文献

Impacts of petroleum activities on birds in the Amazonian forest of Yasuní National Park and the Huaorani Territory, Ecuador, were evaluated using data from 32 standardized 1-day surveys of twenty one 2-ha sites, spread out over 90 km, located at four different distances (0, 0.5, 1.5, and 4.5 km) into floristically diverse, primary, terra firme forest (240–320 m elev.), except one that was in 6-year-old secondary forest. Width of deforestation for roads was 25 m. The 263 species, identified via sight, sound, and mist-netting (4 nets) between 5:45 and 13:30 on days without heavy or constant rain, were grouped into 14 foraging guilds. Discriminant Function Analysis achieved complete separation of the 10 surveys adjacent to roads from the 22 surveys > 0.3 km into the forest, based on species richness per guild. Terrestrial insectivores contributed by far the most (76%) to this separation, and were the only guild to show a significant difference in the average number of species registered between the 6-edge sites and the 15 interior sites (Mann–Whitney U = 4, P = 0.001). Overwinter and snowmelt processes are thought to be critical to controllersof nitrogen (N) cycling and retention in northern forests. However, therehave been few measurements of basic N cycle processes (e.g.mineralization, nitrification, denitrification) during winter and littleanalysis of the influence of winter climate on growing season N dynamics.In this study, we manipulated snow cover to assess the effects of soilfreezing on in situ rates of N mineralization, nitrification and soilrespiration, denitrification (intact core, C2H2 – based method),microbial biomass C and N content and potential net N mineralization andnitrification in two sugar maple and two yellow birch stands with referenceand snow manipulation treatment plots over a two year period at theHubbard Brook Experimental Forest, New Hampshire, U.S.A. The snowmanipulation treatment, which simulated the late development of snowpackas may occur in a warmer climate, induced mild (temperatures >–5 °C) soil freezing that lasted until snowmelt. The treatmentcaused significant increases in soil nitrate (NO3 )concentrations in sugar maple stands, but did not affect mineralization,nitrification, denitrification or microbial biomass, and had no significanteffects in yellow birch stands. Annual N mineralization and nitrificationrates varied significantly from year to year. Net mineralization increasedfrom 12.0 g N m–2 y–1 in 1998 to 22 g N m–2 y–1 in 1999 and nitrification increased from 8 g N m–2 y–1 in 1998 to 13 g N m–2 y–1 in 1999.Denitrification rates ranged from 0 to 0.65 g N m–2 y–1. Ourresults suggest that mild soil freezing must increase soil NO3 levels by physical disruption of the soil ecosystem and not by direct stimulation of mineralization and nitrification. Physical disruption canincrease fine root mortality, reduce plant N uptake and reduce competitionfor inorganic N, allowing soil NO3 levels to increase evenwith no increase in net mineralization or nitrification.  相似文献

The biological oceanography of waters adjacent to Australia's North West Cape (21° 49 S, 114° 14 E) was studied during the austral summers of 1997/98 and 1998/99. We measured egg production rate (EPR) by the small paracalanid copepods that dominated the calanoid community. Bottle incubation experiments were conducted at a shallow (20 m) station in the mouth of Exmouth Gulf, and at a shelf-break station (80 m). In 1997/98, we measured EPR by Paracalanus aculeatus, P. indicus, Acrocalanus gracilis and Bestiolina similis, but in 1998/99, we concentrated on P. indicus. Maximal observed EPRs by Paracalanus and Acrocalanus species were 30 eggs female–1 d–1, but B. similis attained only 17 eggs female–1 d–1. Sporadic measurements of EPR by P. aculeatus minor (maximum 4 eggs female–1 d–1) and Parvocalanus crassirostris ( 9 eggs female–1 d–1) were also made. However, maximal EPRs were seldom achieved and were often less than 10 eggs female–1 d–1. There was no difference between EPR of either P. indicus or B. similis in 1997/98 and 1998/99, despite differences in temperature. Trophic resources severely limit copepod egg production in this area. We suggest that variability and skewness of egg production data derived from individual incubations may be used to judge the degree of food limitation of the population and the variability in feeding success between individuals. The dominance of small copepods and the invariance in their EPR suggest that pulses in physical forcing and subsequent primary production will be severely damped by trophodynamic processes before reaching larval fish.  相似文献

Understanding the factors regulating the concentrations of basic cations in soils and surface waters is critical if rates of recovery are to be predicted in response to decreases in acidic deposition. Using a dynamic simulation model (PnET-BGC), we evaluated the extent to which atmospheric deposition of strong acids and associated leaching by strong anions, atmospheric deposition of basic cations through changes in emissions of particulate matter, and historical forest cutting have influenced soil pools of exchangeable basic cations and the acid-base status of stream water at the Hubbard Brook Experimental Forest (HBEF) in New Hampshire. Historical deposition of basic cations was reconstructed from regression relationships with particulate matter emissions. Simulation results indicate that the combination of these factors has resulted in changes in the percent soil base saturation, and stream pH and acid neutralizing capacity (ANC) from pre-industrial estimates of 20%, 6.3 and 45 eq L–1, respectively, to current values of 10%, 5.0 and –5 eq L–1, respectively. These current values fall within the critical thresholds at which forest vegetation and aquatic biotic are at risk from soil and surface water acidification due to acidic deposition. While the deposition of strong acid anions had the largest impact on the acid-base status of soil and stream water, the reduction in deposition of basic cations associated with reductions in particulate emissions was estimated to have contributed about 27% of the depletion in soil Ca2+ exchange pool and 15% of the decreases in stream water concentrations of basic cations. Decline in stream water concentrations of basic cation occurred under both increasing and decreasing exchangeable pools, depending on the process controlling the acid base status of the ecosystem. Model calculations suggest that historical forest cutting has resulted in only slight decreases in soil pools of exchangeable basic cations, and has had a limited effect on stream ANC over the long-term.  相似文献

Summary The performance of crassulacean acid metabolism (CAM) by dicotyledonous trees of the genusClusia sampled at three sites in the state of Falcon in northern Venezuela is characterized.Clusia leaves have a somewhat succulent appearance. Unlike leaves of many other CAM plants, which are uniformly built up of very large isodiametric cells, there are distinct layers of palisade and spongy mesophyll, with individual cells being smaller. There is no specialized water storage tissue. 13C values indicate thatC. multiflora in the elfin-cloud forest on top of Cerro Santa Ana, at 800 m altitude, performs C3 photosynthesis (13 –27.1). However,C. rosea in the tall cloud forest on Cerro Santa Ana (600m altitude), andC. rosea andC. alata in the dry forest on Serrania San Luis (900 m altitude) perform CAM (13C –14.1 to –19.2). InC. alta andC. rosea there were large day-night changes in the levels of malic and citric acids ranging from 63 to 240 mmol 1–1 for malid acid and from 35 to 112 mmol 1–1 for citric acid. The sum of the changes in malate and citrate levels accounts for the changes of titratable protons measured. With a day-night change of titratable protons of 768 mmol 1–1 in one of the analyses,C. rosea showed the highest value yet encountered in a CAM plant. Oscillations of free sugars (fructose, glucose, sucrose) and of starch were also analysed in the CAM performingClusia species. Carbon skeletons of the precursors involved in nocturnal malate and citrate synthesis largely derive from free sugars and not from polyglucan. Unlike some other CAM plants, there is no clear and quantitative correlation between day-night changes of organic acid levels and cell sap osmolality.Dedicated to Professor Dr. Otto L. Controlled digestion of 150 kD single chain botulinum type E neurotoxin with pepsin atpH 6.0 produced 112, 48, 46, and 16 kD fragments. These were chromatographically purified; their locations in the 1300 amino acid residue long neurotoxin were determined by identifying the amino terminal 10 residues of 112 and 48 kD fragments, 50 residues of 46 kD fragment, and 59 residues of 16 kD fragment. The 48 and 112 kD fragments contain the N-terminal segment of the neurotoxin (i.e., residue no. 1 to 425 and 1 to 990, respectively), the 46 kD fragment corresponds to 407 residues of the C-terminal region, and the 16 kD fragment contains the 140 residues from a segment nearer to the C-terminus. The 48 kD fragment is similar to the 50 kD N-terminal light chain of the 150 kD dichain neurotoxin, which is generated by tryptic cleavage of the 150 kD single chain neurotoxin, and is separated from the 100 kD C-terminal heavy chain by dithiothreitol (DTT) reduction of an intrachain disulfide bond in the presence of 2 M urea (Sathyamoorthy and DasGupta,J. Biol. Chem. 260, 10461, 1985). The pepsin-generated 48 kD fragment, unlike the light chain, was isolated without exposure to DTT and urea. The single chain 112 kD fragment following trypsin digestion yielded 48 and 60 kD fragments that were separable after DTT reduction of the intrachain disulfide which links them. The N-terminal residues of the smaller fragment were identical to that of the single chain 150 kD neurotoxin; the single chain 112 kD fragment is therefore the neurotoxin minus the 50 kD C-terminal half of the heavy chain. The biological activities of the 48 and 112 kD fragments can be demonstrated in permeabilized PC12 cells (Lomnethet al., J. Neurochem. 57, 1413, 1991); they inhibit norepinephrine release.  相似文献

Semper's larvae were obtained from <300 out of 1800 plankton tows taken in the world's oceans (1964–1993). Zoanthellae (larvae of Sphenopidae) occurred at 217 stations and zoanthinae (larvae of Zoanthidae) at 86, the two larval types showing distributions clearly delimited by a minimum sea temperature (22 °C for zoanthellae, 18 °C for zoanthinae; a statistically significant difference, P<0.001). Length of formalin-fixed zoanthellae was 2–8.6 mm and of zoanthinae 1.5–5.9 mm. Endodermal zooxanthellae were present in 9/24 zoanthinae but in no zoanthellae (of 19). Three larvae contained an endo-commensal/parasitic amphipod. Septa were externally visible in larger zoanthinae and were counted in transverse sections of other larvae, a majority of which (both kinds) had 12 septa, the normal maximum. The pattern was brachycnemic in 40/43 larvae and anomalous (but non-macrocnemic) in three. If macrocnemic genera reproduce by Semper's larvae, they should have been represented in such a large sample. The distribution of adult Epizoanthus was examined: many species are deep sea (recorded down to 5000 m) but shallow-water species are relatively plentiful in, for example, the Adriatic and North Seas. No Semper's larva has ever been recorded from either. Some Parazoanthus species also occur in shallow water, especially associated with western Atlantic reef sponges. If they produce Semper's larvae, these have never been found. It is probable that macrocnemic zoanthids settle from planulae that do not develop into recognizable zoanthellae or zoanthinae.  相似文献

The nitrogen source utilization by Fagus crenata distributed on soils with different forms of inorganic nitrogen in a cool-temperate deciduous forest in central Japan was determined by measuring foliar 15N. Two soil habitat types along a slope were delineated based on nitrogen transformation patterns, i.e., soils with high net nitrification rates and with no or low net nitrification, respectively. Despite differences in soil types, the study species, F. crenata, was distributed along the entire slope. The foliar 15N value of F. crenata from the lower slope area was significantly lower than that from the upper slope. Given the finding of a previous study that the 15N of NO3 was lower than that of NH4+, our results indicate that reliance on NO3 as a nitrogen source was greater in the lower slope area than in the upper slope area. Differences in the values of foliar 15N were about 1, which is far less than the 10 15N value of soil inorganic N reported in the previous study. This discrepancy might suggest that the study species utilized NO3 even in the upper site where net nitrification had not been detected. Measurements of nitrate reductase activity, an index of NO3 uptake, also supported this interpretation. Nitrate reduction occurred in leaves and roots at both the lower and the upper sites. Thus, the study species may be able to use NO3 even in soils with no net nitrification, a factor that could allow the distribution of F. crenata along the entire length of the slope.  相似文献

This paper presents the first Holocene continuous record from the southern Bolivian Altiplano. In this area, the climate is now characterized by weak summer monsoon rains. The record is located north of Salar de Uyuni in a non-glacial valley (Rio Baja). Between 11600 and 2210 cal year BP, the rivers accumulated fine deposits, while under the present climatic conditions, the fine particles are carried downstream by strong water floods. These deposits contain a rich diatom flora showing that the valley floor was occupied by paleowetlands. Water input needed to be more or less continuous to explain that the paleowetlands survived over a long period of time. We show that diatoms can be used to reconstruct the relative variations in the water level and the salinity throughout time, despite of the spatial complexity of this type of environment. During the Holocene, the water level was low except during some periods, dated 11600–9800, 6330–5300, and 3110–2210 cal year BP. Saline and freshwater microhabitats were simultaneously present in the valley floor as indicated by a mixed diatom flora evidenced throughout the record. We propose a paleoclimatic scenario based on the assumption that the NE wet atmospheric flow of the monsoon was replaced by the westerlies of the southern hemisphere at the latitude of the study site.  相似文献

Botulinum neurotoxin (NT) serotype A is a 150-kDa dichain protein. Posttranslational nicking of the single-chain NT (residues Pro 1–Leu 1295) by the protease(s) endogenous to Clostridium botulinum excises 10 residues, leaving Pro 1–Lys 437 and Ala 448–Leu 1295 in the 50-kDa light (L) and 100-kDa heavy (H) chains, respectively, connected by a Cys 429–Cys 453 disulfide and noncovalent bonds [Krieglstein et al. (1994), J. Protein Chem. 13, 49–57]. The L chain is a metalloprotease, while the amino- and carboxy-terminal halves of the H chain have channel-forming and receptor-binding activities, respectively [Montecucco and Schiavo (1995), Q. Rev. Biophys. 28, 423–472]. Endoproteinase Glu-C and -chymotrypsin were used for controlled digestion at pH 7.4 of the 150-kDa dichain NT and the isolated 100-kDa H chain (i.e., freed from the L chain) in order to map the cleavage sites and isolate the proteolytic fragments. The dichain NT appeared more resistant to cleavage by endoproteinase Glu-C than the isolated H chain. In contrast, the NT with its disulfide(s) reduced showed rapid digestion of both chains, including a cleavage between Glu 251 and Met 252 (resulting in 30- and 20-kDa fragments of the L chain) which was not noted unless the NT was reduced. Interestingly, an adjacent bond, Tyr 249–Tyr 250, was noted earlier [DasGupta and Foley (1989), Biochimie 71, 1193–1200] to undergo self-cleavage following reductive separation of the L chain from the H chain. The site Tyr–Tyr–Glu–Met (residues 249–252) appears to become exposed following reduction of Cys 429–Cys 453 disulfide. Identification of Glu 669–Ile 670 and Tyr 683–Ile 684 as protease-susceptible sites demonstrated for the first time that at least two peptide bonds in the segment of the H chain (residues 659–684), part of which (residues 659–681) is thought to interact with the endosomal membranes and forms channels [Oblatt-Montal et al., (1995), Protein Sci. 4, 1490–1497], are exposed on the surface of the NT. Two of the fragments of the H chain we generated and purified by chromatography are suitable for structure–function studies; the 85- and 45-kDa fragments beginning at residue Leu 544 and Ser 884, respectively (both extend presumably to Leu 1295) contain the channel-forming segment and receptor-binding segments, respectively. In determining partial amino acid sequences of 10 fragments, a total of 149 amino acids in the 1275-residue NT were chemically identified.  相似文献

Flash-induced optical kinetics at room temperature of cytochrome (Cyt) c 551 and an Fe-S center (CFA/CFB) bound to a purified reaction center (RC) complex from the green sulfur photosynthetic bacterium Chlorobium tepidum were studied. At 551 nm, the flash-induced absorbance change decayed with a t 1/2 of several hundred ms, and the decay was accelerated by 1-methoxy-5-methylphenazinium methyl sulfate (mPMS). In the blue region, the absorbance change was composed of mPMS-dependent (Cyt) and mPMS-independent component (CFA/CFB) which decayed with a t 1/2 of 400–650 ms. Decay of the latter was effectively accelerated by benzyl viologen (Em –360 mV) and methyl viologen (–440 mV), and less effectively by triquat (–540 mV). The difference spectrum of Cyt c had negative peaks at 551, 520 and 420 nm, with a positive rise at 440 to 500 nm. The giant, 3.6-MDa hexagonal bilayer hemoglobin (Hb) of Lumbricus terrestris consist of twelve 213-kDa globin subassemblies, each comprised of three disulfide-bonded trimers and three monomer globin chains, tethered to a central scaffolding of 36–42 linkers L1–L4 (24–32 kDa). It is known to contain 50–80 Ca and 2–4 Cu and Zn; the latter are thought to be responsible for the superoxide dismutase activity of the Hb. Total reflection X-ray fluorescence spectrometry was used to determine the Ca, Cu, and Zn contents of the Hb dissociated at pH 2.2, the globin dodecamer subassembly, and linker subunits L2 and L4. Although the dissociated Hb retained 20 Ca2+ and all the Cu and Zn, the globin subassembly had 0.4 to 3 Ca2+, depending on the method of isolation, and only traces of Cu and Zn. The linkers L2 and L4, isolated by reversed-phase high-pressure liquid chromatography at pH 2.2, had 1 Ca per mole and very little Cu and Zn. Electrospray ionization mass spectrometry of linker L3 at pH 2.2 and at neutral pH demonstrated avid binding of 1 Ca2+ and additional weaker binding of 7 Ca2+ in the presence of added Ca2+. Based on these and previous results which document the heterogeneous nature of the Ca2+-binding sites in Lumbricus Hb, we propose three classes of Ca2+-binding sites with affinities increasing in the following order: (i) a large number of sites (>100) with affinities lower than EDTA associated with linker L3 and dodecamer subassembly, (ii) 30 sites with affinities higher than EDTA occurring within the cysteine-rich domains of linker L3 and dodecamer subassembly, and (iii) 25 very high affinity sites associated with the linker subunits L1, L2, and L4. It is likely that the low-affinity type (i) sites are the ones involved in the effects of 1–100 mM Group IIA cations on Lumbricus Hb structure and function, namely increased stability of its quaternary structure and increased affinity and cooperativity of its oxygen binding.  相似文献

Summary Eimeria cyclopei n.sp. is described from the silky anteater, Cyclopes didactylus, from Pará State, north Brazil. Undifferentiated oocysts, passed in the faeces, complete sporulation in seven days at 26 to 28°C. Oocysts are ellipsoidal to sub-spherical, with a mean size of 28.1 × 23.6 m: the wall is 1.5 to 2.0 m thick, apparently with an outer thin, colourless membrane and two inner, thicker, striated and yellowish layers. There is no micropyle, oocyst residuum or polar body. The mean measurements of sporocysts are 19.0 × 9.0 m, and they are slightly asymmetrical, elongate pear-shape, with a plug-shaped Steida body projecting beyond the end of the sporocyst. Sporozoites are as long as or longer than the sporocysts: The sporocyst residuum is scattered between sporozoites in younger specimens and becomes condensed into rounded mass in older ones. The endogenous stages occur in the epithelial cells of the ileum, on the lumenal side of the host-cell nucleus. Uninucleate meront, microgamont and macrogamont precursors are recognizable morphologically. Mature meronts are 20.0 × 15.7 m some produce 12 to 20 merozoites which are 8.7 × 2.0 m, and others 10 to 26 merozoites which are 11.4 × 2.0 to 15.0 × 3.0 m. Mature microgamonts which are 27.5 × 24.1 m, produce from 150 to 170 microgametes of 7.1 × 1.0 m: microgametes have two flagella of unequal length. Mature macrogamonts are 28.4 × 24.5 m Eimeria choloepi n.sp. is recorded from the two-toed sloth, Choloepus didactylus, from the same area of Brazil. Undifferentiated oocysts, passed in the faeces, complete sporulation in 23 days at 26 to 28°C. Oocysts with a mean size of 23.0 × 20.3 m, have a wall 2.0 to 2.5 m thick which is composed of two thick, yellowish and striated outer layers and a delicate, colourless inner one. There is no micropyle, oocyst residuum or polar granule. Mature sporocysts with a mean size of 11.3 × 7.1 m, are ellipsoidal to egg-shaped and have a poorly developed Steida body. The sporocyst residuum is composed of a small number of large globules: The sporozoites are longer than the sporocyst and strongly recurved. The endogenous stages occur in epithelial cells of the ileum, on the lumenal side of the host-cell nucleus. Dimorphic meronts produce 8 to 18 merozoites which are either 13.0 × 2.0 m or 13.0 × 3.0 m. Summary The relationship between lipid composition and phase transition was investigated by differential scanning calorimetry for intact and membrane phospholipid extracts of wild-type (w/t) and thecel (Tw 40) mutant ofNeurospora crassa. Thecel (Tw 40) mutant (grown on minimal, sucrose medium supplemented with Tween 40 at 34 °C) had approximately twice the saturated fatty acid content ofw/t organisms grown at 22 °C. The gel-liquid crystal phase transitions of ergosterol-free extracts derived fromw/t andcel (Tw 40) occur at –31 and –11 °C, respectively. The heats of transition (H) of these extracts were 1 and 13 cal/g, respectively. The addition of ergosterol (the predominant sterol inNeurospora) to the phospholipid extracts decreased the observed heats of transition, but did not alter the transition temperature. IntactNeurospora, whetherw/t orcel (Tw 40) did not manifest similar gel-liquid crystal phase transitions in the differential scanning calorimeter. However, an endothermic peak at approximately 30 °C was observed in intact cells and extracted phospholipids of bothw/t andcel (Tw 40) organisms. Summary The dielectric breakdown in the membranes of cells ofValonia utricularis was investigated using intracellular electrodes and 500-sec current pulses. Electrical breakdown, which occurs when the membrane potential reaches a well-defined critical value, is not associated with global damage to the cell or its membranes (the membrane reseals in <5 sec). It was thus possible to investigate the effect of temperature on dielectric breakdown in single cells. It was found that the critical potential for breakdown was strongly dependent on temperature, decreasing from 1000 mV at 4°C to 640 mV at 30°C. The decrease in the breakdown potential with increasing temperature and the very short rise-time of the breakdown current (1 sec) suggests that the Wien field dissociation does not play a major role in the breakdown process. It is shown that the nonlinearI–V characteristics observed at different temperatures can be accurately accounted for with no adjustable parameters, by considerations of the mechanical compression of the membrane due to stresses induced by the electric field. Electrical breakdown on this scheme results from an electromechanical instability in the membrane. On this basis the present results indicate that the elastic modulus of the region of the membrane where breakdown occurs, decreases by a factor of 2 with increasing temperature from 4 to 30°C. On the assumption of a thickness of 4.0 nm and a dielectric constant of 5, the elastic modulus is estimated to have a value of 5×106 Nm–2 at 20°C.  相似文献

Summary The three-dimensional organization of the motor endplates of the slow fibers of the rectus abdominis muscle in the Japanese meadow frog (Rana nigromaculata nigromaculata Hallowell) is visualized by use of a field-emission scanning electron microscope after removal of connective tissue components by HCl hydrolysis. Clusters of shallow oval depressions 1–3 m in diameter are seen in the postsynaptic membrane at intervals of about 150 m. On the surface of these depressions, a few low bulges of postsynaptic membrane are irregularly arranged. Terminal boutons, 1–3 m in diameter, occur along the length of nerve branches and terminals and fit into the shallow oval depressions of the postsynaptic membrane. The Schwann cells covering the terminal branches exhibit a simpler organization than those in twitch fibers.  相似文献

Grassland to forest conversions currently affect some of the world's most productive regions and have the potential to modify many soil properties. We used afforestation of native temperate humid grassland in the Pampas with eucalypts as an experimental system to 1) isolate forest and grassland imprints on soil acidity and base cation cycling and 2) evaluate the mechanisms of soil acidification. We characterized soil changes with afforestation using ten paired stands of native grasslands and Eucalyptus plantations (10–100 years of age). Compared to grasslands, afforested stands had lower soil pH (4.6 vs.5.6, p < 0.0001) and 40% lower exchangeable Ca (p < 0.001) in the top 20 cm of the soil. At three afforested stands where we further characterized soil changes to one meter depth, soil became increasingly acidic from 5 to 35 cm depth but more alkaline below 60 cm compared to adjacent grasslands, with few differences observed between 35 and 60 cm. These changes corresponded with gains of exchangeable acidity and Na in intermediate and deeper soil layers. Inferred ecosystem cation balances (biomass + forest floor + first meter of mineral soil) revealed substantial vertical redistributions of Ca and Mn and a tripling of Na pools within the mineral soil after afforestation. Soil exchangeable acidity increased 0.5–1.2 kmolc.Ha–1.yr–1 across afforested stands, although no aboveground acidic inputs were detected in wet + dry deposition, throughfall and forest floor leachates. Our results suggest that cation cycling and redistribution by trees, rather than cation leaching by organic acids or enhanced carbonic acid production in the soil, is the dominant mechanism of acidification in this system. The magnitude of soil changes that we observed within half a century of tree establishment in the Pampas emphasizes the rapid influence of vegetation on soil formation and suggests that massive afforestation of grasslands for carbon sequestration could have important consequences for soil fertility and base cation cycles.  相似文献
