One- and two-“year”-old seedlings of Pinus silvestris L., from which the buds had been removed, were studied for five weeks during the second and third growth period, respectively. Intact seedlings were used as controls. The seedlings were cultivated under controlled conditions in a climate chamber. The growth of the seedlings was determined and the one-“year”-old needles assayed for changes in net photosynthesis and ribulose bisphosphate carboxylase activity and in the levels of protein, Kjeldahl nitrogen, chlorophyll and starch. In the control the carboxylase activity and the content of protein, Kjeldahl nitrogen and starch in the needles increased in the beginning of the “summer” and decreased during the shoot growth period. The starch content was higher after bud removal (decapitation), since the carbohydrate could not be utilized for the growth of the new shoot. Decapitation did not affect the growth rate of the roots. The content of Kjeldahl nitrogen and total and soluble protein in the needles was higher in the decapitated seedlings during the period of shoot elongation in the control. Total nitrogen, but not protein, reached high levels, indicating accumulation of non-protein compounds. The general course of the chlorophyll pattern was not affected. Higher ribulose bisphosphate carboxylase activity than in the control was observed in the later part of the experimental periods. The higher levels of protein and nitrogen as well as of carboxylase activity after decapitation support the interpretation that soluble protein, including the carboxylase, and possibly other nitrogen compounds in the older needles are used for growth of the shoot. The loss of protein and nitrogen and of carboxylase activity in the control did not seem to be due to mineral deficiency in the substrate. Despite higher levels of carboxylase activity and similar chlorophyll concentrations, light-saturated net photosynthesis was lower after decapitation. The ratio between photosynthesis and photorespiration was not affected.  相似文献

Ribulose bisphosphate carboxylase-oxygenase, RuBP carboxylase (EC 4.1.1.39), was purified from non-hardened and hardened needles of Pinus sylvestris L. Needles were collected from pine seedlings cultivated in nutrient solution in a climate chamber from seedlings grown outdoors, and from a tree in a natural stand. The enzyme was isolated from crude extracts through quantitative precipitation in polyethylene glycol 4000 and MgCl2, followed by sucrose gradient centrifugation in a fixed angle rotor. The purified enzyme seemed homogeneous by the criterion of (sodium dodecylsulphate) polyacrylamide gel electrophoresis. Contamination by nucleic acids was negligible. The RuBP carboxylase protein content of the gradient fractions was estimated as A2801 cm× 0.61 mg ml−1. Carboxylase activities were determined in a radioactive assay at 25°C. The specific activity of RuBP carboxylase isolated from non-hardened needles was approximately 1 μmol CO2 (mg protein)−1 min−1. For enzyme isolated from hardened needles collected during winter the specific activity was somewhat lower due to loss of enzyme activity during the preparation. The described two-step procedure provides a means for quantitation of the RuBP carboxylase protein in pine needles during all seasons.  相似文献

Ribulose bisphosphate carboxylase activity was examined in Pinus silvestris L. during successive seasons. The enzyme activities were studied both in seedlings, kept under controlled conditions in a climate chamber, and in needles from a 15-year-old tree in a natural stand. The enzyme activities were analysed in cell-free extracts prepared with Tween 80 as protective agent. The carboxylase activity fluctuated periodically both in the seedlings and in the natural stand. In the seedlings, the weight-related activity in the older needles increased 50–100% (in the cotyledons c. 200%) in the beginning of the “summer”. It decreased as the new shoot developed. The specific activity increased c. 100%. With chlorophyll as base, the activity usually decreased during “summer”. In the developing current needles the carboxylase activity increased when expressed on a weight or on a protein basis. The decrease in weight-related carboxylase activity in the older needles was preceded by, or simultaneous with, loss of total protein. It is suggested that protein, including the carboxylase, is utilized as nitrogen reserve for the new shoot. During hardening by combined photoperiod and thermoperiod, the carboxylase activity decreased when expressed relative to dry weight and protein. Calculated on a chlorophyll basis, the activity was rather constant. In the natural stand the activity in the one- and two-year-old needles increased during spring and summer and decreased during autumn and winter. Even at severe winter stress substantial carboxylase activity remained in the needles. The activity of the enzyme in vivo is discussed with respect to electron transport and net photosynthesis.  相似文献

Biochar is an organic amendment used for soil remediation, there are only a few studies documenting the effects of nitrogen on the role of biochar in contaminated soils. A pot experiment was conducted to investigate the impacts of biochar (0%, 1%, and 2.5%, w/w) and nitrogen (0, 100, and 200 mg N kg?1) on plant growth, nutrient and cadmium (Cd) uptake of Cichorium intybus. N, P, Ca, Mg, and Cd concentrations increased with N level in 0% and 1% biochar treatments. In plants treated with 2.5% biochar, 200 mg N kg?1 addition caused significant reductions of N, P, Ca, Mg, and Cd concentrations in comparison to 100 mg N kg?1 treatments. Nitrogen promoted shoot biomass at all biochar treatments, while biochar had no effect on shoot biomass in 0 and 200 mg N kg?1 addition treatments. Nitrogen also significantly increased N, P, K, Ca, Mg, and Cd contents in the 0% and 1.5% biochar addition treatments. Although soil DTPA-extractable Cd concentration showed the lowest values in 1% biochar in combination with 100 and 200 mg N kg?1 addition treatments, lowest shoot Cd concentration, and relatively high shoot biomass occurred in the 2.5% biochar + 200 mg N kg?1 treatment. Based on these results, biochar application at its highest rate (2.5%) in combination with high N supply (200 mg N kg?1) contributed to both crop yield and agricultural product safety. N input alone might increase the risk of human health, and the optimum N dose should be determined during phytostabilization process.  相似文献

Seedlings of Douglas-fir, Pseudotsuga menziesii (Mirb.) Franco, and ponderosa pine, Pinus ponderosa Dougl. ex Laws., were grown in a controlled environment and fertilized with nutrient solutions containing 150 ppm (+N), or 0 ppm nitrogen (−N). These treatments greatly altered seedling growth, and the concentrations of N and carbohydrates in their tissues. Metabolically active tissues, such as roots, incubated with a limited supply of O2 became hypoxic faster and synthesized more ethanol than less active tissues, such as needles. All tissues that were incubated for 4 h in N2 synthesized ethanol. Needles incubated in N2 and light had much lower quantities of ethanol than needles in N2 and dark, suggesting that O2 from photosynthetsis limited internal anoxia. Most tissues from +N seedlings synthesized greater quantities of ethanol in N2 anoxia than tissues from −N seedlings, probably because they were able to produce more enzymes with a greater availability of N. However, this increase in ethanol synthesis between N treatments was most pronounced in the phloem. Ethanol and soluble sugar concentrations were negatively related in needles and positively related in roots of N+ seedlings, but not −N seedlings. Starch concentrations had no effect on the amount of ethanol produced by any tissue. Regardless of N treatments, all tissues from ponderosa pine produced more N2-induced ethanol than Douglas-fir, in part because its tissues contained different concentrations of soluble sugars and N as a consequence of phenological differences between the species. However, ponderosa pine tissues may also maintain greater quantities of anaerobic enzymes, or their isozymes than Douglas-fir. Received: 22 February 1998 / Accepted 23 June 1998  相似文献

Ribulose bisphosphate (RuBP) carboxylase and glycollate oxidase were partially purified from jack pine (Pinus banksiana Lamb.) needles. Preincubation of RuBP carboxylase with HCO3? and Mg2+ markedly stimulated its activity. RuBP carboxylase showed hyperbolic reaction kinetics with respect to HCO3?, Mg2+, and RuBP. Both SO32- and SO42- inhibited RuBP carboxylase, but SO32- was more inhibitory than SO42-. The SO32- inhibition was competitive with respect to HCO3? (whether SO32- was present during activation or was added to the activated enzyme), while the SO42- inhibition was non-competitive with respect to HCO3?. Glycollate oxidase was inhibited more severely by low concentrations of SO32- than by SO42-. Fumigation of jack pine seedlings with 0.34 ppm sulphur dioxide for 24 and 48 hr produced a considerable decline in the activities of these enzymes, but 1 hr of fumigation produced no effect. During the longer exposures the sulphur content of the needles increased considerably, although the needles showed no visible injury. It is suggested that the accumulation of SO32- and SO42- in the needles following sulphur dioxide exposure influenced the enzyme activities.  相似文献

The effects of N application on tree growth and the retranslocationof N, P, and K from young needles to new growth were examinedin young radiata pine (Pinus radiata D. Don) trees. Nitrogen fertilization increased the number and size of needles, rates of shoot production, stem volume growth and tree biomass.Foliar N and P contents (µg per needle) fluctuated ina cyclic fashion with prominent phases of accumulation, retranslocationand replenishment. The patterns of these fluctuations in controland N-fertilized trees were similar, although the fluxes ofN, P and K in and out of needles were increased by N fertilization.Greater translocation (g per tree) of N and K from needles ofN fertilized trees occurred because fertilization increasedthe needle weight and the proportion of N and K retranslocatedfrom individual needles. Nitrogen fertilization increased theretranslocation of P largely as a result of higher needle mass.Trees supplied with more than adequate amounts of P in the soilretranslocated up to 58 per cent of the initial pool of P fromyoung needles. The periods of high retranslocation coincidedwith periods of high concentrations of soil mineral N and withshoot production. Conversely, the periods of rapid replenishmentof N and P into the needles coincided with the time of slowshoot growth and low concentration of soil mineral N. The growthrate of trees, rather than the availability of nutrients inthe soil was the main factor controlling retranslocation. For radiata pine, retranslocation from needles is not a mechanismspecific for coping with low soil fertility. It seems to bea mechanism which enhances the nutrient supply to apical growingpoints, especially during periods of flushing. Scots pine (Pinus sylvestris L.) and Norway spruce (Picea abies Karst.) seedlings were exposed to realistically elevated O3 levels in open‐air experiments over three growing seasons. The total O3 exposure doses were 1.2 × (1991), 1.5 × (1992) and 1.7 × (1993) ambient levels. During the 1992 and 1993 growing seasons pine and spruce seedlings received two different levels of nitrogen supply. Effects on growth, mycorrhiza formation, needle ultrastructure, primary and secondary compounds were studied. Ozone exposure had only slight effects on biomass production, growth height and nutrient content of studied conifers. Higher nitrogen availability improved growth of the seedlings and resulted in higher concentration of nitrogen in needles. In Scots pine O3 exposure did not have effects on quantity of total mycorrhizas and short roots, while higher nitrogen availability decreased quantity of mycorrhizas and short roots. In both tree species O3 exposure induced O3‐related ultrastructural symptoms, e.g. granulation and dark staining of the chloroplast stroma in the needle mesophyll cells, at both nitrogen availability levels. Ozone exposure and nitrogen availability did not have significant effects on starch concentrations in either tree species. Concentrations of some individual terpenes were higher in O3‐exposed needles, while concentrations of individual and total resin acids, total phenolics and catechins were not affected by O3 exposure. Nitrogen availability did not have substantial effects on concentrations of monoterpenes. By contrast, concentrations of some individual and total resin acids were lower in pine needles and higher in spruce needles with higher nitrogen availability, while phenolic concentration in spruce needles decreased at higher nitrogen availability. The results suggest that realistically elevated levels of O3 in the field can have some negative effects on the mesophyll ultrastructure of conifer needles, but carbon allocation to root and shoot growth and secondary metabolites are not affected substantially.  相似文献

The response and adaption mechanisms of seedlings under long-term warming have remained largely unknown. In this study, we investigated the effects of warming for 6 years on growth, and needle carbon, nitrogen, chlorophyll, and carbohydrate levels in a coniferous tree species, Abies faxoniana. Seedlings were grown in even-aged monospecific stands under ambient and warming (ambient +2.2°C) temperature in climate control chambers. Warming caused statistically significant increases in the specific leaf area, leaf area ratio, root biomass, leaf biomass, branch biomass, stem biomass, and total mass of the seedlings, and reduced the root/shoot ratio. Warming also increased total chlorophyll concentrations, specific chlorophyll pigments, and Chlorophyll a/b ratios in both studied needle age classes. In addition, C/N ratios of current-year and 1-year-old needles increased by warming. In contrast, warming decreased the levels of N, sugar, cellulose, and starch in needles, while warming had no effect on the height, stem diameter, needle mass ratio, root mass ratio, and root/needle ratio. We conclude that warming increases branch growth and changes needle chemistry, which enhances the light capture potential of seedlings.  相似文献

We inoculated lodgepole pine (Pinus contorta var. latifolia (Dougl.) Engelm.) with Paenibacillus polymyxa P2b-2R, a diazotrophic bacterium previously isolated from internal stem tissue of a naturally regenerating pine seedling to evaluate biological nitrogen fixation and seedling growth promotion by this microorganism. Seedlings generated from pine seed inoculated with strain P2b-2R were grown for up to 13 months in a N-limited soil mix containing 0.7 mM available N labeled as Ca(15NO3)2 to facilitate detection of N2-fixation. Strain P2b-2R developed a persistent endophytic population comprising 102–106?cfu?g?1 plant tissue inside pine roots, stems, and needles during the experiment. At the end of the growth period, P2b-2R had reduced seedling mortality by 14 % and 15N foliar N abundance 79 % and doubled foliar N concentration and seedling biomass compared to controls. Our results suggest that N2-fixation by P. polymyxa enhanced growth of pine seedlings and support the hypothesis that plant-associated diazotrophs capable of endophytic colonization can satisfy a significant proportion of the N required by tree seedlings growing under N-limited conditions.  相似文献

In this study, we surveyed the long term effects of liming and fertilizing in old Scots pine stands on the ectomycorrhiza (ECM) colonization, tree growth and needle nutrient concentration 35 years later. Four mature stands of Scots pine on low productive mineral soil were limed in 1959 and 1964 with total doses of limestone ranging from 3 to 15 Mg ha?1 and fertilized with nitrogen (N) in 1970. Thirty-five years after the first liming treatment, all stands were analysed for tree growth and needle nutrient concentrations and two of the stands were also analysed for ECM colonization. ECM colonization increased significantly with liming from 61.5% in the control plots to 88% in the plot with the highest limestone dose. ECM colonization increased with increasing pH in the humus layer from 62% colonization at pH?=?3.5 to 90% at pH?=?6.5 and decreased with increasing amount of extractable phosphorus (P) in the humus. Liming did not affect the frequencies of different ECM morphotypes or dead short root tips, the fine root biomass or necromass. ECM colonization was uncorrelated with needle nutrient concentrations or tree increment. Liming did not significantly affect tree growth. However, nutrient concentrations of current-year needles were affected by prior liming. Ca concentrations in current-year needles increased from approximately 15 mg g?1 in control treatments to more than 30 mg g?1 in limed plots, whereas concentrations of Mn, Al, Fe, and in two stands, B, decreased due to liming. In conclusion, liming with doses up to 15 Mg ha?1 was detectable in stands 35 years after treatment. The liming significantly increased the ECM colonization of Scots pine fine roots, increased the needle nutrient concentration of Ca and decreased the needle concentrations of Mn, Al, and Fe.  相似文献

We investigated the responses of photon-saturated photosynthesis rate (P sat) and its simultaneous acclimation of anatomy and nitrogen use patterns of current needles of Korean pine (Pinus koraiensis) seedlings grown under factorial combinations of two nitrogen levels and irradiances. Although N supply resulted in a significant increase of N content in needles under both irradiances, the increase of P sat tended to be suppressed only in shade (S). The significant increase of P sat in full sunlight (O) was associated with the increase of ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPCO) and chlorophyll (Chl) contents. In contrast, small increase of Chl content and no increase of RuBPCO content were found in S (90 % cut of full irradiance), which would result in a small increase of P sat. This result suggests that extra N is stocked in needles under shade for the growth in next season. With N supply, a significant decrease of specific leaf area (SLA) was detected only in O. This decrease of SLA was due to the increase of density of needle. Furthermore, the increase of needle density was not due to the increased number and size of mesophyll cells, but the increased density of each mesophyll cell. Therefore, although SLA changed in O, the change did not involve anatomical adaptation to use increased N effectively, at least observable by light microscopy. Hence, even though the SLA would change, N deposition will improve the photosynthetic capacity of Korean pine seedlings, not through the development of needle anatomy but through improvement of the allocation of N in both irradiances.  相似文献

The present study examined the effect of nutrient supply on acclimation potential and photoinhibitory susceptibility in one-year-old foliage of shade-grown Norway spruce ( Picea abies Karst.) seedlings transferred to high-light, as compared to seedlings grown in shade or in high-light. After the transfer, the photosynthetic response to intercellular CO2 and chlorophyll fluorescence parameters were measured, and the allocation of leaf N within the photosynthetic apparatus was estimated. The effects of light and nutrient supply markedly differed. Light availability positively affected both photosynthetic capacity on an area basis (but not on a mass basis) and leaf N allocation to ribulose-1,5-biphosphate carboxylase/oxygenase (Rubisco), thereby increasing photosynthetic capacity per unit nitrogen. By contrast, nutrient supply in high-light affected photosynthetic capacity both on an area and a mass basis, but did not affect the allocation of N within the photosynthetic apparatus. Fluorescence parameters indicated that shade-grown seedlings transferred to high-light with low-nutrient supply, underwent prolonged photoinhibition. By contrast, transferred seedlings with high-nutrient supply were able to avoid photoinhibition, increasing their photosynthetic capacity on an area basis. On the whole, nutrient shortage was found to prevent the acclimation response and to increase the photoinhibitory susceptibility to changing light conditions in P. abies mature needles.  相似文献

The growth of two provenances of Pinus sylvestris L. were compared with two provenances of Picea abies (L.) Karst. and with Pinus contorta Dougl. when grown in solution cultures with low nutrient concentrations. Nitrogen was added at different exponentially increasing rates, and the other nutrients were added at a rate high enough to ensure free access of them to the seedlings. During an initial period of the culture (a lag phase), when the internal nutrient status was changing from optimum to the level of the treatment, deficiency symptoms appeared. The needles yellowed and the root/shoot ratio increased. The initial phase was followed by a period of exponential growth and steady-state nutrition. The needles turned green again, and the root/shoot ratio stabilized at a level characteristic of the treatment. These patterns were the same as previously reported for other tree species. The relative growth rate during exponential growth was numerically closely equal to the relative nitrogen addition rate. The maximum relative growth rates were about 6 to 7.5% dry weight increase day-1. This is a much lower maximum than for broad-leaved species (about 20 to 30% day-1) under similar growth conditions. The internal nitrogen concentrations of the seedlings and the relative growth rates were stable during the exponential period. Close linear relationships were found between these parameters and the relative addition rate up to maximum growth. During steady state the relative growth rates of the different plant parts were equal. However, there were large differences between genotypes in absolute root growth rate at the same seedling size because of differences in root/shoot ratio. Lodgepole pine had the highest root growth rate, whereas that of Norway spruce, especially the southern provenance, was remarkably low. Yet, Norway spruce had a high ability to utilize available nutrients. In treatments with free nutrient access, growth allocation to the shoot had a high priority in all genotypes, but there was still a marked tendency for luxury uptake of nutrients. Nitrogen productivity (growth rate per unit of nitrogen) was lower than in broadleaved species and highest in lodgepole pine. The relevance of the dynamic factors, i.e. maximum relative growth rate, nutrient uptake rate, nitrogen productivity, growth allocation and root growth rate, are discussed with regard to conifer characteristics and selection value.  相似文献

The relationship between light saturated net photosynthesis (Amax) and nitrogen concentration (N) was studied in needles of both Scots pine seedlings, grown at three relative growth rates (2,6 and 8%) controlled by nutrient addition rate, and Scots pine shoots collected from four sites with different fertility. In the seedlings, Amax was measured on 14 different dates starting at the beginning of the second growing season and ending when growth of the new shoot and the secondary needles had finished. In shoots from the natural stands Amax of the previous-year shoots was measured on 6 dates throughout the growing season. Both in seedlings and shoots, the correlation between Amax and N was poor, when data from all sampling dates were taken together. However, Amax was correlated with N in most instances when the age of the needles was considered and the data were examined either at weekly intervals (seedlings) or separately for each sampling date (shoots). The slope of the Amax vs N relationship varied greatly between sampling dates. In the seedlings the correlation between Amax and N was strongest by the time when the new needles were developing. In the shoots the correlation was significant from mid June until mid August, while no correlation was found in the beginning and at the end of the growing season. Our data indicate that in pine needles the photosynthesis-nitrogen relationship is more complex than in broadleaved species. Contrary to the broadleaved species, where the correlation is independent of sampling time, in this conifer the time of the year affects the correlation and there are phases during the growing season when the correlation is poor or nonexistent.Abbreviations Amax light saturated net photosynthesis - kPa kilopascal - N nitrogen concentration in the needles - PFD photon flux density (400–700 nm) - RGR relative growth rate  相似文献

Fifteen or 30 days of anaerobic growth conditions significantly reduced shoot and root nitrogen, potassium, phosphorus, iron and manganese concentrations in seedlings of pond pine ( Pinus serotina Michx.), sand pine [ P. clausa (Engelm.) Sarg.] and drought-hardy and wet-site loblolly pine ( P. taeda L.) grown in a culture system using non-circulating, continuously flowing solution. Calcium and shoot magnesium levels were least affected by anaerobic growth conditions – largely reflecting the passive nature of their uptake. Shoot and root nutrient content (mg nutrient pot-1) followed similar trends, with wet-site loblolly and pond pine seedlings least affected by anaerobic solution culture. Shoot biomass of wet-site loblolly and pond pine seedlings was not affected by anaerobiosis, suggesting an increase in shoot nutrient utilization efficiency. Root biomass was significantly reduced by 15 or 30 days of anaerobiosis, with sand pine exhibiting the largest reduction in root dry weight (57%).
These results suggest that anaerobiosis interferes with net nutrient acquisition, even under the high nutrient conditions provided by solution culture. Sand pine suffered the largest reductions in shoot and root biomass and nutrient concentrations, showing earlier symptoms of waterlogging injury and nutrient stress than drought-hardy loblolly pine seedlings. Whether net nutrient acquisition decreased because of the reduction in root surface area available for absorption and/or reduced uptake efficiency cannot be ascertained from these data.  相似文献

The effects of aluminium chloride (AICI3) treatments (50 and 150 mg/l) on 3-year-old Scots pine (Pinus sylvestris L.) seedlings were studied in a sand culture during 2 growing periods in an open field experiment. Even by the end of the first growing period, a decline was observed in the concentrations of Ca, Mg and P within the needles, and of Ca and Mg in the roots. After the second growing period, increased N and K concentrations were observed in the needles of Al-treated seedlings. Both the needles and roots of Al-treated seedlings showed, after the second growing period, a decline in growth and increased concentrations of AI as the amount of AICI3 in the nutrient solution increased. Al-induced changes in needle structure were found to be symptomatic of a nutrient imbalance, particularly of Mg and P. Al-stress did not result in any observable changes in root anatomy or in the number of mycorrhizas. Scots pine proved to be rather resistant to Al-stress, indicating that direct Al-injuries are not likely in the field, though Al-stress may be a contributing factor in the formation of nutrient imbalances.  相似文献

The content of shikimic acid in the vegetative parts of a number of plants growing in the Altai Territory has been studied. The greatest content of shikimic acid (about 1.5%) was found in the coniferous needles of Scots pine. Shikimic acid accumulated in buckwheat cells cultivated in vitro on glyphosate-containing medium. The best results were obtained with buckwheat explants cultivated in vitro on Murashige and Skoog nutrient medium in the presence of 0.5 × 10?3 mg/L of glyphosate.  相似文献

Fumigations of 4–7 week old seelings of Pinus silvestris , with ca 400 μg SO2 m-3 for 4–5 days resulted in a decrease both in net photosynthetic rate and in the levels of ribulose bisphosphate carboxylase activity of the primary needles. The effect on net photosynthesis was reversible. No visible damages were observed on the needles. Lower concentrations, ca 200 μg SO2 m-3, for ca 6 days inhibited net photosynthesis reversibly. The effect on net photosynthesis was not the result of stomata closure or of increased respiration. A dose-response relation for the inhibition of net photosynthesis and ribulose bisphosphate carboxylase is suggested. A similar relation was observed for the chlorophyll content of the needles. The reversible inhibition of photosynthesis was evident only at high quantum flux density, and the quantum yield of photosynthesis at rate limiting quantum flux was not affected. This was also suggested from an experiment with Scots pine in the field.  相似文献
