The content of chlorophyll, the rate of O2 evolution, the slow phase of fluorescence induction, and photoinduced changes in the intensity of electron paramagnetic resonance (EPR) signal I from the reaction center of photosystem I (P+700) were studied in leaves of Vicia faba L. grown in 10–7–10–3 M aqueous solutions of CdCl2, SnCl2, CuCl2, and MgCl2. At low concentrations of heavy metal (Cd, Sn, and Cu) chlorides, the content of chlorophyll per fresh weight decreased. However, the rate of O2 evolution calculated per chlorophyll basis, O2/(t chlorophyll), increased. High concentrations of heavy metals significantly suppressed plant development and inhibited photosynthetic O2 evolution. In contrast, plant treatment with MgCl2 (10–5–10–3 M) resulted in an increase in the content of chlorophyll and a stimulation of leaf photosynthetic activity. A positive correlation between the F
M/F
T ratio and O2/(t chlorophyll) (r = 0.89, P > 0.999) was found. We observed a negative correlation between the values of P/P
0 of EPR signal I (white/far-red light) and O2/(tchlorophyll) (r = –0.89, P > 0.999). The data obtained indicate nonspecific and nonmonotone changes in the photosynthetic apparatus of plants treated with heavy metals: low concentrations of heavy metals (10–7–10–6 M) stimulated photosynthetic activity, whereas high concentrations (10–4–10–3 M) suppressed it. 相似文献

UV-B辐射作为一种重要的环境信号影响着植物的生长与发育,它能够调控气孔运动和诱导乙烯产生.该试验利用乙烯生物合成抑制剂和乙烯受体抑制剂处理蚕豆叶片表皮条,结合气孔开度分析和乙烯释放量测定,研究乙烯在UV-B辐射调控表皮条气孔运动中的作用.结果发现,将蚕豆叶片表皮条置于0.8 W·m-2的UV-B辐射下1~4 h,乙烯生成和气孔关闭均被显著诱导,且乙烯释放峰先于气孔关闭的起始;乙烯生物合成抑制剂和乙烯受体抑制剂处理均能显著逆转UV-B辐射诱导的气孔关闭;外源乙烯处理也能模拟UV-B辐射的效应诱导可见光下蚕豆表皮条的气孔关闭.可见,乙烯介导了UV-B辐射诱导的蚕豆气孔关闭. 相似文献

细胞外钙调素可能作为多肽第一信使,调节细胞增殖、花粉萌发、特定基因表达等生理过程.气孔能灵敏地对外界刺激作出反应,快速开闭.本文用免疫电镜和免疫荧光显微镜技术证明保卫细胞及其它表皮细胞胞外都存在钙调素.外源纯化钙调素能促进气孔关闭、抑制气孔开放,最适浓度为10-8mol/L;不能透过质膜的大分子钙调素拮抗剂W7-agarose和钙调素抗血清都能抑制气孔关闭、促进开放,说明保卫细胞的内源胞外钙调素确实能促进气孔关闭、抑制开放,而且只能在细胞外起作用.推测在自然情况下,保卫细胞内源胞外钙调素可能作为胞外第一信使和其它信号分子一起调节气孔的开关运动,而且可能在环境刺激与细胞响应之间起重要作用. 相似文献

用激光扫描共聚焦显微技术,初步研究广谱性蛋白激酶抑制剂星型孢菌素(STS)对蚕豆气孔运动的调控效应.结果表明:(1)光下STS对气孔开度无影响但暗中显著促进气孔开放,表明蛋白激酶参与光/暗对气孔运动的调控,光下蛋白激酶活性低而暗中高;(2)与H2O2清除剂抗坏血酸(ASA)和NO清除剂羧基-2-苯-4,4,5,5-四甲基咪唑-1-氧-3-氧化物(cPTIO)一样,STS既降低暗处理和光下外源H2O2、硝普钠(SNP)处理保卫细胞H2O2、NO水平,也促进气孔开放,表明暗中蛋白激酶通过抑制H2O2、NO清除机制提高保卫细胞内源H2O2、NO水平并促进气孔关闭. 相似文献

蚕豆植株经暗饥饿处理40h后,取其下表皮,再用超声波“原位分离”下表皮上的保卫细胞对,然后在无菌、非光合条件下,用外源蔗糖处理蚕豆下表皮上的保卫细胞对,考察其对气孔开放的效应。结果发现,在1d的无菌培养过程中,蔗糖显著促进了气孔的开放。100mmol/L的蔗糖在10mmol/L的MES-NaOH/KOH(pH6.1)缓冲液中,开度分别增加2.0/2.6μm;在1μmol/L的气孔开放促进剂fusicoccin(FC)的存在下,100mmol/L的蔗糖在MES-NaOH/KOH(pH6.1)缓冲液中分别增加开度5.0/5.5μm。不同浓度(5~200mmol/L)的蔗糖处理结果表明气孔开度的增加与蔗糖浓度呈一定的正相关,浓度为100mmol/L的蔗糖处理表现出最大促进作用。同时还初步观察到,蔗糖可维持保卫细胞存活率和叶绿体的完整性。 相似文献

观察了Ca2 + 、Ca2 + 的螯合剂和Ca2 + 通道抑制剂对NO调控的蚕豆气孔运动的影响。结果表明 ,NO的供体 1~ 10 0 μmol/LSNP (sodiumnitroprusside ,硝普纳 )可诱导气孔关闭 ;除去表皮条缓冲液中的Ca2 + 后 ,NO不再影响气孔的运动 ;Ca2 + 的螯合剂EGTA和BAPTA几乎可以完全抑制NO诱导的气孔关闭作用 ;胞内钙通道抑制剂钌红 (rutheniumred)和L型Ca2 + 通道阻断剂硝苯吡啶 (nifedipine)能够减弱SNP诱导气孔运动的关闭趋势 ;加入Ca2 + 通道抑制剂LaCl3 ,则外源NO失去其诱导气孔关闭的作用。说明在NO调控的气孔运动中 ,在NO信号途径的下游可能涉及来自胞内和胞外Ca2 + 的参与 ,并且胞外Ca2 + 更为重要。 相似文献

Activities of noncyclic and alternative pathways of photosynthetic electron transport were studied in intact leaves of broad been (Vicia faba L.) seedlings grown under white light at irradiances of 176, 36, and 18 µmol quanta/(m2 s). Electron flows were followed from light-induced absorbance changes at 830 nm related to redox transformations of P700, the photoactive PSI pigment. The largest absorbance changes at 830 nm, induced by either white or far-red light, were observed in leaves of seedlings grown at irradiance of 176 µmol quanta/(m2 s), which provides evidence for the highest concentration of PSI reaction centers per unit leaf area in these seedlings. When actinic white light of 1800 µmol quanta/(m2 s) was turned on, the P700 oxidation proceeded most rapidly in leaves of seedlings grown at irradiance of 176 µmol quanta/(m2 s). The rates of electron transfer from PSII to PSI were measured from the kinetics of dark P700+ reduction after turning off white light. These rates were similar in leaves of all light treatments studied, and their characteristic reaction times were found to range from 9.2 to 9.5 ms. Four exponentially decaying components were resolved in the kinetics of dark P700+ reduction after leaf exposure to far-red light. A minor but the fastest component of P700+ reduction with a halftime of 30–60 ms was determined by electron transfer from PSII, while the three other slow components were related to the operation of alternative electron transport pathways. Their halftimes and relative magnitudes were almost independent on irradiance during plant cultivation. 以蚕豆为实验材料,利用药理学实验和分光光度法,研究了ABA处理及ABA与H2S合成抑制剂共处理对蚕豆气孔运动的影响,以及体内H2S水平、H2S合成酶L-/D-半胱氨酸脱巯基酶(磷酸吡哆盐依赖性酶)活性变化.结果表明:(1)光下H2S的合成抑制剂羧甲氧基胺半盐酸盐(AOA)、羟氨(NH2OH)、L-/D-半胱氨酸脱巯基酶分解产物C3H3KO3+NH3均明显抑制ABA诱导的蚕豆气孔关闭;(2)外源ABA能够明显提高叶片的H2S水平及L-/D-半胱氨酸脱巯基酶活性;(3)AOA、NH2OH、C3H3KO3和NH3均可以逆转ABA所引起的H2S水平及L-/D-半胱氨酸脱巯基酶活性的升高.研究发现,ABA可通过增强L-/D-半胱氨酸脱巯基酶活性,促进L-/D-半胱氨酸分解生成H2S,进而诱导蚕豆气孔关闭. 相似文献

温室条件下,用0(Control)、8.65kJm-2d-1(TI)及11.2KJm-2d-1(t2)不同剂量的UV-B辐射处理蚕豆幼苗。Ca2 .ATPase及Mg2 -ATPase的活性在辐射处理期间下降。在处理21d,T1和T2微粒体膜的MDA含量明显高于对照,同时IUFA急剧下降,且呈明显的剂量效应。14及21d时,膜磷脂的含量也明显下降。脂氧合酶(Lox)活性在第7及14天与对照相比都显著升高,而21d后迅速下降。结果表明,增强UV-B对微粒体膜的伤害可能是一方面导致正常酶合成与分解之间的平衡失调,另一方面导致了膜脂过氧化作用。 相似文献

水杨酸(salicylic acid,SA)作为植物体内一种内源性的信号分子,具有多种生理功能.实验表明水杨酸以浓度依赖的方式诱导气孔关闭,抑制气孔张开.20U/mL的CAT与SA共同处理时可逆转SA诱导气孔关闭作用的83%~90%.以H2O2荧光探针H2DCFDA结合激光扫描共聚焦显微术直接检测到SA处理可引起保卫细胞内H2O2的产生;在保卫细胞内显微注射CAT可完全阻止SA导致的DCF荧光增强.表明SA诱导的气孔关闭可能与H2O2的产生有关. 相似文献

Different bee flower visitors were observed during the flowering seasons of Broad Bean in Ismailia, eastern Egypt, the Nile Delta and Alexandria, western Egypt. Eight major bee visitors were observed: Andrena ovatula (Kirby, 1802), Andrena sp. (Andrenidae), Chalicodoma siculum (Rossi, 1792) (Megachilidae), Colletes lacunatus Dours, 1872 (Colletidae), Apis mellifera Linnaeus, 1758, Anthophora hispanica (Fabricius, 1787), A. aegyptiaca (Dalla Torre and Friese, 1895) and Xylocopa pubescens (Spinola, 1838) (Apidae). Anthophora hispanica and A. aegyptiaca as well as Chalicodoma siculum had their peak flying period in the middle of the flowering season of the Broad Bean, while Xylocopa pubescens and Andrena sp. had their peak period in the second half of the flowering period. 相似文献

The effect of different light wavelengths on the development of lesions induced by Alternaria tenuissima in broad bean leaves was investigated. Lesion development was completely suppressed in red‐light‐irradiated broad bean leaflets, irrespective of isolate or spore concentration. Pre‐treatment of leaflets with red light for 24 h before inoculation also suppressed lesion development. Alternaria tenuissima failed to produce infection hyphae in red‐light‐irradiated broad bean leaflets. These results indicate that disease suppression in broad bean leaflets is due to light‐induced resistance. Spore germination fluid (SGF) of A. tenuissima allowed non‐pathogenic Alternaria alternata to infect wounded and unwounded broad bean leaflets kept in the dark, results suggesting that SGF induced susceptibility. Red light suppressed susceptibility induced by A. tenuissima SGF; thus, lesion formation and development were suppressed when leaflets inoculated with the spores of A. alternata suspended in A. tenuissima SGF were kept under red light. From these results, we conclude that red light induced resistance in broad bean to leaf spot disease caused by A. tenuissima, and that SGF induced susceptibility of broad bean leaflets to a non‐pathogenic isolate of A. alternata. 相似文献

The kinetic curves of dark reduction of P700+ (oxidized primary donor of PSI) after far-red light irradiation were studied on broad bean (Vicia faba L.) leaves treated with antimycin A, methyl viologen, or diuron. Four components of P700+ reduction were found in untreated leaves, namely, an ultrafast component with a half-time of 25 ms, and fast (210 ms), middle (790 ms), and slow (6100 ms) components. The fast component disappeared in leaves treated with antimycin A or methyl viologen. At the same time, these substances did not affect other components of P700+ reduction. Treatment of leaves with diuron abolished both the ultrafast and fast components of P700+ reduction. As the length of far-red light exposure was increased, a lag phase appeared in the development of middle component in leaves treated with diuron, antimycin A, or methyl viologen. In thus treated leaves, an exponential pattern of the middle component was displayed with a certain delay after darkening. A conclusion was drawn that the minor ultrafast component of P700+ dark reduction in broad bean leaves was caused by electron donation to PSI from PSII, whereas the fast component of this process was determined by the operation of ferredoxin-dependent electron transport around PSI. 水通道或水通道蛋白是水分运动的主要通道。以RD2 8cDNA和RD2 8抗体为探针证明了蚕豆 (ViciafabaL .)保卫细胞中存在水通道蛋白 ,并以气孔运动为指标 ,结合抗体和抑制剂处理证明水通道蛋白是水分运动的主要通道。研究表明编码质膜水通道蛋白的RD2 8转录体在叶片保卫细胞、叶肉细胞和维管束中高表达 ,尤以保卫细胞中最多 ;荧光免疫染色和Confocal显微镜观察表明 ,RD2 8抗体反应主要位于保卫细胞质膜。进一步采用RD2 8抗体和水通道蛋白抑制剂———HgCl2 (2 5 μmol L) 处理可抑制壳梭孢素 (FC)、光照诱导的气孔开放和原生质体体积膨胀以及ABA诱导的气孔关闭 ,但这种抑制作用可以被水通道抑制剂的逆转剂 β_巯基乙醇 (ME)逆转。表明蚕豆保卫细胞中存在水通道蛋白并参与蚕豆保卫细胞的运动过程。 相似文献

Effects of benzyladenine (BA) and abscisic acid (ABA) applied separately or simultaneously on parameters of gas exchange of Phaseolus vulgaris L. leaves were studied. In the first two experimental sets) 100 M ABA and 10 M BA were applied to plants sufficiently supplied with water. Spraying of leaves with ABA decreased stomatal conductance (g
s) and in consequence transpiration rate (E) and net photosynthetic rate (P
N) already 1 h after application, but 24 h after application the effect almost disappeared. 10 M BA slightly decreased gas exchange parameters, but in simultaneous application with ABA reversed the effect of ABA. Immersion of roots into the same solutions markedly decreased gas exchange parameters and 24 h after ABA application the stomata were completely closed. The effect of ABA was ameliorated by simultaneous BA application, particularly after 1-h treatment. In the third experimental set, plants were pre-treated by immersing roots into water, 1 M BA, or 100 M ABA for 24 h and then the halves of split root system were dipped into different combinations of 1 M BA, 100 M ABA, and water. In plants pre-treated with ABA all gas exchange parameters were small and they did not differ in plants treated with H2O+H2O, H2O+BA, or BA+BA. In plants pre-treated with BA or H2O, markedly lower values of P
N were found when both halves of roots were immersed in ABA. Further, the effects of pre-treatment of plants with water, 1 M BA, 100 M ABA, or ABA+BA on the development of water stress induced by cessation of watering and on the recovery after rehydration were followed. ABA markedly decreased gas exchange parameters at the beginning of the experiment, but in its later phase the effect was compensated by delay in development of water stress. BA also delayed development of water stress and increased P
N in water-stressed leaves. BA reversed the effect of ABA at mild water stress. Positive effects of BA and ABA pre-treatments were observed also after rehydration. 相似文献
