The ability of the rodent carcinogens 1,4-dioxane (DX) and thiourea (TU) to induce meiotic non-disjunction (ND) was assessed in 3- and 6-day-old Drosophila melanogaster females. The chemicals were administered orally and three 24 h and one 48 h broods were obtained after mating, to sample oocytes treated in increasingly earlier stages of development. The broods represent mainly mature oocytes (brood I), nearly mature oocytes (brood II), early oocytes (brood III) and very early oocytes (brood IV). The toxicity of DX increased with dose (1% (not toxic), 1.5, 2, 3, 3.5%) as well as a reduction in fecundity which was moderate. Induction of ND in mature oocytes was positive with 2, 3 and 3.5% concentrations and was not related to dose. In immature oocytes it was also positive though already at the lowest concentration tested (1%), suggesting a sensitivity higher than that of mature oocytes. TU at 0.10-10%, did not affect viability, but since fecundity was seriously impaired at high doses, ND was not assessed beyond the 1.5% concentration. TU also induced ND in mature and in immature oocytes; neither a threshold nor a dose effect was detected. The response of mature oocytes was lower than that of immature oocytes. TU induced increases of ND in the earliest cells tested in a more consistent fashion than DX. The data clearly show that both chemicals induced ND in mature oocytes and in the three subsets in which immature oocytes were fractionated. Though toxicity may play a significant unspecific role in the induction of chromosome malsegregation by DX and TU, the induction of ND at low doses, moderately toxic to the oocytes, suggests that the interaction with specific targets contributed to the results obtained.  相似文献

The induction of sex chromosomes meiotic nondisjunction (ND) by hydroquinone (HQ) given orally was investigated in Drosophila melanogaster 2-7, 8-22, 24, 48, 72 and 96 h-old females. ND was assessed by a system where exceptional females (XXY) and only 1/4 of the expected regular progeny are viable. Oocytes were treated at different stages of development. 4% HQ tested only in 72 h-old females induced ND in oocytes sampled in brood I (mostly mature oocytes at metaphase I). 6% HQ increased ND in brood I of 8-22 h-old females, while other broods, (including cells treated at early prophase) were also affected in older flies, the highest significance being attained in the 48 h-old series. Newly hatched females (2-7 h-old) were refractory to the treatment, though oocytes sampled in the first three subcultures are comparable to cells showing enhancement of ND in series run with older females. Toxicity of 2, 4 and 6% HQ increased with concentration and females' age: (a) 2% was not toxic; (b) 4% was toxic only to 72 h-old females; (c) 6% was increasingly toxic to females 24, 48 and 72 h-old. The results indicate that age plays a significant role on both chromosomal segregation and toxicity and suggest that in Drosophila HQ is metabolized to its reactive species. The lack of toxic and aneugenic effect in very young females could reflect a more efficient detoxification due to the known high specific activity of glutathione-S-transferase (GST) after eclosion. The decline in GST activity around day 2 of adult life coincides with the high effect of HQ in 48 h-old females.  相似文献

The effect of ethanol on chromosomal segregation was investigated in Drosophila melanogaster females homozygous for a structurally normal X chromosome marked with the recessive mutation yellow (y/y). For chronic treatments the females were kept from eclosion in food supplemented with 10% or 15% (v/v) ethanol, mated 24 or 48 h later to wild-type males and brooded in freshly prepared ethanol food. For the acute treatments 24- or 48-h-old females were exposed for 60 min to a 75% (v/v) ethanol solution by means of soaked tissue paper placed at the bottom of regular culture vials and brooded daily after mating. The results obtained show that: (1) both treatments significantly increased the frequency of X-chromosome nondisjunction; (2) after acute treatment this effect declined in later broods; (3) the yield of malformed flies in the progeny of acutely treated females was significantly higher than control values and also declined in later broods; (4) ovary analysis showed that chronic ethanol treatments caused a cessation of egg production. The induction pattern of nondisjunction and malformed flies is interpreted as giving support to the assumption that these effects may result from a direct action of ethanol. Ethanol toxicity was assessed by exposing females of different ages to a 50% or a 75% (v/v) solution for 60 min and counting the surviving flies 24 h later. The surviving fraction decreased steeply from 1-day-old (100%) to 5-day-old females (1.8%). It is suggested that toxicity may have been due to the action of a metabolite of ethanol, probably acetaldehyde.  相似文献

The ability of low doses of X-rays to induce numerical and structural chromosome anomalies in immature oocytes was examined in two experiments. In the first, 10-11-day-old females were given 0.1 or 0.2 Gy of X-rays and sampled at intervals up to 32 weeks later. In the second, 4-5-week-old females were given 0.1 Gy of X-rays and sampled up to 36 weeks post-irradiation. Chromosome anomalies were assessed in metaphase II oocytes. In the first experiment, there was evidence of dose-related increases in both hyperhaploidy (n + 1) and structural chromosome anomalies. In the second experiment, only the frequency of structural chromosome anomalies was found to increase consistently after irradiation. There was no indication that radiation-induced depletion of the oocyte population was associated with an early onset of the maternal age effect on nondisjunction.  相似文献

Various aspects were studied of the brood size and sex allocation strategies, and of size-fitness relationships in Parallorhogas pyralophagus (Marsh), a gregarious ectoparasitoid of Eoreuma loftini Dyar. Brood size was significantly correlated with host size; larger hosts were allocated larger broods. Brood sex ratios were fixed precisely at 1 male per 4 females, and eggs were likely to be deposited in that order; differential mortality did not contribute to this precise sex ratio. The sex allocation strategy of P. pyralophagus is likely to conform to strict, i.e. single foundress, local mate competition. Adoption of this strategy is probably influenced by a limited insemination capacity of males; a smaller proportion of females (0.09 vs. 0.21) remained virgin in broods with precise or higher sex ratios (> or = 0.20 males) relative to broods with lower than precise sex ratios (< 0.20 males). Moreover, all females were inseminated in most broods (60%) with precise or higher sex ratios, whereas this did not occur in broods with lower than precise sex ratios. The hypothesized occurrence of strict local mate competition in P. pyralophagus was supported also by observations that: (i) offspring brood sex ratios were independent of maternal brood sex ratios and number of parental females concurrently allocating offspring to a group of hosts, and; (ii) the rate of superparasitism under no-choice conditions was low (approximately 20%), suggesting that rates of outbreeding in the field are low. Other results suggested that fitness in P. pyralophagus was correlated with adult size; longevity and reproductive capacity both increased with adult size in males and females. However, adult size may be more important for females than for males because the differences in reproductive capacity between the largest and smallest individuals was up to 7.3 times greater in females versus < 2 times in males.  相似文献

Summary The effect of varying X-ray doses (0.05–0.80 Gy) on preovulatory mouse oocytes was studied by measuring nondisjunction during the first meiotic division, as well as structural chromosome anomalies in ovulated oocytes at metaphase stage II. The incidence of nondisjunction (0.1% hyperploid oocytes) found in oocytes from nonirradiated NMRI-Han female mice was in accordance with the results previously obtained with the same strain. Significantly (P<0.05) more hyperploid oocytes (0.9%) were ovulated following irradiation with 0.8 Gy. There was no statistically significant increase of nondisjunction after low doses. Structural chromosome anomalies occurred, however, even after an irradiation dose as low as 0.05 Gy. The dose response for structural chromosome anomalies is altogether different from that of radiation-induced hyperpoidy. We consider that irradiation of mature oocytes might well be less hazardous with regard to its potency for increasing nondisjunction during the first meiotic division when compared with the effect of chemical mutagens.  相似文献

Isopropanol (isopropyl alcohol, 2-propanol, IPA) is a volatile solvent widely used in domestic or industrial environments and reported as innocuous in various test systems. The aim of this work was to search for in vivo genotoxic effects of IPA in Drosophila melanogaster, studying its ability to induce nondisjunction (ND) in females, sex linked recessive lethals (SLRL) in males, and somatic mutation and/or recombination (SMART) in larvae. Treatments were acute (60min) and were administered via inhalation. IPA had low toxicity in adult flies (75% IPA mortality index, MI=12.7% (females) and 2.6% (males)) and larvae (MI=14.3%, 75% IPA). Female fertility was severely affected during the first 24h (brood I, BI) after treatment, but, afterwards, control values were recovered. IPA induced a 50-fold increase of ND (%) in 24h old females, and a six-fold rise in 4-5 d old BI offspring. Nondisjunction frequencies (%) in the offspring of broods II to V (24h in each case) were similar to control values. IPA doses of 25% and 50% (v/v), tested in 24h old females, showed a significant dose-dependent increase of ND(%)in BI only, with control values in subsequent broods. Flies gave normal offspring when kept in regular media for 24h before mating. The eye spot test (SMART) showed a significant increase at 50% IPA (p<0.05, m=2), but the response was not dose-dependent. IPA failed to induce SLRL in any of the spermatogenesis stages tested. These findings suggest that the main effect of IPA is to induce chromosomal malsegregation; IPA must be present at the resumption of M-phase I after fertilization, to exert these effects. The alcohol does not affect DNA directly, but perturbations of the nuclear membrane may be responsible for induction of ND.  相似文献

Female mice of two age groups, 3--4 and 11--14 months old, homozygous for the T(1;13)70H reciprocal mouse translocation were used for cytological observations of bivalents (in primary oocytes) and metaphase II chromosomes (in secondary oocytes). Special attention was given to the behavior of the long (131) and short (113) marker chromosomes. In primary oocytes, univalents were considered "true" or "opposite". The aged females showed an eight-folded increase in "true" univalent frequency for chromosomes 113 over the young ones. A nine-fold rise for nondisjunction with regard to this chromosome was observed. For the other chromosomes, these factors were 2 and 1.7, respectively. The absolute levels of nondisjunction remained low at old age (1.42% for chromosome 113, 1.22% for all other chromosomes). The long marker bivalent 131 was used for chiasma counts. No change in chiasma number with age was observed. It is argued that poorer physiological conditions within the maturing oocytes of older females are the major cause for both the increasing frequencies of "true" and "opposite" univalents and the increased incidence for nondisjunction.  相似文献

The onset of incubation before the end of laying imposes asynchrony at hatching and, therefore, a size hierarchy in the brood. It has been argued that hatching asynchrony might be a strategy to improve reproductive output in terms of quality or quantity of offspring. However, little is known about the mediating effect of hatching asynchrony on offspring quality when brood reduction occurs. Here, we investigate the relationship between phenotypic quality and hatching asynchrony in Common Kestrel Falco tinnunculus nestlings in Spain. Hatching asynchrony did not increase breeding success or nestling quality. Furthermore, hatching asynchrony and brood reduction had different effects on nestlings’ phytohaematogglutinin (PHA)‐mediated immune response and nestling growth. In asynchronous and reduced broods (in which at least one nestling died), nestlings showed a stronger PHA‐mediated immune response and tended to have a smaller body size compared with nestlings raised in synchronous and reduced broods. When brood reduction occurred in broods hatched synchronously, there was no effect on nestling size, but nestlings had a relatively poor PHA‐mediated immune response compared with nestlings raised in asynchronous and reduced broods. We suggest that resources for growth can be directed to immune function only in asynchronously hatched broods, resulting in improved nestling quality, as suggested by their immune response. We also found that males produced a greater PHA‐mediated immune response than females only in brood‐reduced nests without any effect on nestling size or condition, suggesting that females may trade off immune activities and body condition, size or weight. Overall, our results suggest that hatching pattern and brood reduction may mediate resource allocation to different fitness traits. They also highlight that the resolution of immune‐related trade‐offs when brood reduction occurs may differ between male and female nestlings.  相似文献

It is commonly assumed that there exists interbrood competition mediated by in utero growth. This could be manifested by a female reallocating saved resources to future broods. Here, we report results of a manipulation experiment designed to detect such reallocation. Two groups of female mice were allowed each to produce two broods. In the first brood, the test females were mated with phenotypically normal males heterozygous for an insulin-like growth factor 2 (Igf2) null allele, while the control females were mated to a wild-type male. The test sample females invested 20% less into their first brood than did the control sample. In both test and control groups the females were mated with a wild-type male in the second round of mating. Surprisingly, we found that females that invested little into their first brood also invested little (compared with other second broods) into their second brood. This 'priming' effect suggests that the assumptions of classical models of parent-offspring conflict are overly simplistic but cannot disprove the existence of interbrood competition.  相似文献

Uniparental maternal brood care often coincides with multiple paternity and single maternity of broods, possibly reflecting benefits of polyandry and costs of uniparental care. Genetic data from the maternally mouthbrooding cichlid fish Simochromis pleurospilus revealed the opposite pattern--low polyandry and allomaternal care. More than 70% of the investigated females had mated with a single male, and 14% of the females had unrelated fry in their broods. Broods with foreign fry were in the late stage of brood care, in which females guard free-swimming fry and recall the broods into their mouths for protection. With one exception, fostering females were related to their adopted fry at the level of first cousins (R(QG) > 0.12), but relatedness between fosters and adopted fry was not significantly higher than between fosters and fry tended by other females. Relatedness among breeders extended to the level of first-order relatives. Mean relatedness among contemporaneously breeding dams (R(QG) = 0.08) was significantly higher than among dams breeding in different seasons (R(QG) = -0.04), which suggests a temporal or spatial concentration of mouthbrooding relatives. Indeed, females sometimes brood in small groups. This behaviour may reduce brood predation but will increase the risk of brood mixing, which is possibly mitigated by low costs of brood care and indirect benefits accrued by relatedness among the breeders in the group. Remarkably, the apparent inbreeding potential did not give rise to bet-hedging polyandry or active avoidance of relatives, as half of the mated individuals were related at R(QG) > 0.13 and polyandry did not coincide with high within-pair relatedness.  相似文献

There is little experimental evidence testing whether currentbrood size and past brood mortality influence mate desertion.In the cichlid Aequidens coeruleopunctatus both parents initiallydefend offspring. In a field study, all experimental broods,irrespective of initial brood size (222.9 ± 60.4, mean± SD), were manipulated to a size of 100 fry. Neitherthe duration nor investment of females in parental care differed between control and brood reduced pairs, even though care seemedcostly. On average, females lost 5.1 ± 4.8% of initialweight while guarding a brood until independence. In contrast,males with experimentally reduced broods guarded fry for significantlyfewer days before deserting their mate than did males fromcontrol pairs with natural-sized broods (20.5 ± 7.5 vs. 14.2 ± 6.2 days). In at least 20% of cases (n = 9/45),the deserting male immediately mated with another female. Maleswith experimentally reduced broods also spent less time guardingfry before deserting and attacked fewer brood predators thandid males with control broods. For broods manipulated to have100 fry, there was a significant negative relationship betweenthe days until male desertion and the proportion of the initialbrood removed. This indicates that male assessment of the futuresuccess of the current brood (hence its reproductive value)is based on past mortality and/or that there is variation amongmales in the expected size of future broods. Both current broodsize and brood size relative to initial brood size are thereforepredictors of male, but not female, parental behavior and matedesertion. Female care may be unaffected by brood reductiondue to limited breeding opportunities and partial compensationfor reduced male care.  相似文献

To test whether nest abandonment is associated with parental health state, reproductive parameters and parental condition indices were examined in relation to brood desertion in great tits. Before desertion, pairs that abandoned their broods in the second half of the nestling period had significantly higher nestling mortality as well as lower average weight of nestlings and entire broods. Independently of brood size, female great tits that deserted their broods on average weighed 1 g (>5%) more than non-deserters. Comparison of metabolic profiles revealed that deserting females were in better nutritional condition (inclined to fat deposition) than non-deserters, which showed symptoms of postresorptive catabolic state, as indicated by a lower level of plasma triglycerides, very low density lipoproteins, and a higher level of free fatty acids and β-hydroxy-butyrate. These results suggest that desertion can be regarded as a reproductive restraint and that non-deserting females invested at least some of their maintenance resources on brood rearing. We found no evidence that desertion or non-desertion was associated with age- or disease-related differences in residual reproductive values. The possible interaction between X-ray- and transposon-induced chromosome damage was monitored in the P-M system of hybrid dysgenesis in Drosophila melanogaster. One- to two-day-old F1 dysgenic males originating from a cross between M strain females and P strain males were irradiated with 5.5 Gy (550 rad) or used as controls to monitor X-Y translocations and transmission ratio distortion. Two 3-day sperm broods were sampled for the former and two 4-day broods for the latter to detect damage induced in the most radiosensitive cells. F1 nondysgenic males derived from M female to M male crosses (controls) were treated identically. X-Y chromosome translocations induced by P element mobility alone declined sharply with a decrease in temperature (18 versus 21 degrees C) and they were significantly reduced with aging of hybrid males from brood 2, 4-8 days of age, to brood 3, 7-11 days of age. No significant increase in translocations was observed when X irradiation and P-M dysgenesis were combined, showing no interaction between damages induced by the two mutator systems. In contrast, interaction was observed in transmission ratio distortion which was significantly increased by X irradiation of hybrid males derived from both reciprocal M X P and P x M crosses. The preferential elimination of P element-bearing autosomes occurred when either spermatocytes or spermatids were irradiated. An aging effect was also observed, resulting in less distortion in 9- to 14-day-old dysgenic males compared to 5- to 10-day-old hybrids.  相似文献

First and second meiotic metaphases (MI and MII, respectively) from female mice of Robertsonian translocation (Rb) stock, trisomic for chromosome 16 (Ts16) or 19 (Ts19), were studied. The mature trisomic oocytes were derived from explanted fetal ovaries that had been cultured and then transplanted so as to mature heterotopically. Multivalent configurations involving the Rb chromosomes and the additional trisomic acrocentric were analysed. Pentavalent configurations occurred in 74.5% of 98 Ts16 MI and 44.2% of 249 Ts19 MI oocytes; quadrivalents (with a univalent acrocentric) were found in 9.2% of Ts16 MI and 10.8% of Ts19 MI oocytes. In 1% of Ts16 MI and 4% of Ts19 MI oocytes, there were two Rb bivalents and a univalent trisomic acrocentric. Rb trivalents and Rb bivalents occurred together in 14.3% of Ts16 MI and 39.4% of Ts19 MI oocytes. Chiasma frequencies were similar in trisomic and chromosomally balanced MI. Chiasma position, distribution, and localization were nearly identical, whether they were found in Rb multivalents or acrocentric bivalents, but one control group (from chromosomally balanced Ts19 littermates) had significantly more terminal chiasmata. Within the triple homologous region of 8% of Rb pentavalents, two chiasmata were observed in the same relative position in the two sister chromatids of one of the three homologs, suggesting a lapse in chiasma position interference. Assortment at MI anaphase was influenced by secondary nondisjunction of the Rb. The ratio of balanced to unbalanced MII oocytes was 1:4 in both trisomies.  相似文献

In Drosophila melanogaster suppression of oviposition results in an aging of both mature and immature oocytes. When oviposition was suppressed for four days, the incidence of X-chromosomal nondisjunction (XXY exceptions) in mature oocytes was more than doubled, whereas in immature oocytes the nondisjunction frequency was not increased. It is shown that this special susceptibility of mature oocytes to aging-induced nondisjunction has to be considered in experiments on the induction of nondisjunction by chemical agents.  相似文献

In many bird species, females undergo a marked decline in body condition during the first days of the nestling period. This decline may be because brooding young chicks reduces the time available for foraging. Alternatively, it might be viewed as an adaptive way to reduce flight costs when the food demand of the brood is highest. To test these hypotheses we modified the brooding commitment of House Sparrows Passer domesticus by manipulating brood size to see if changes in time spent brooding affects adult body condition. During the nestling period, females provided on average three times as much brooding as males. Reduced broods received 14% more brooding than large broods and time spent brooding declined with brood size and chick age according to an exponential decay function. Male body condition was unaffected by brood size and remained stable throughout the reproductive period. Body condition of females with enlarged broods decreased gradually during the nestling period, whereas that of females tending reduced broods dropped abruptly and significantly upon hatching. This resulted in females with reduced broods having lower body condition during the first half of the nestling period than those with enlarged broods. The sharp drop in body condition of females with reduced broods coincided with the period that brooding was most intensive. Indeed, female body condition at the end of the nestling period was negatively correlated with the proportion of time they spent brooding during the first half of the nestling period. Thus, the probable lower homeothermic capacities of reduced broods implies a higher brooding commitment for female House Sparrows that, in turn, may reduce their opportunity to forage and consequently also their body condition.  相似文献

In order to study the mechanisms of nondisjunction at meiosis I in oocytes gonadotropin-stimulated Djungarian hamsters were treated at two stages [4.5 and 6 h post human chorionic gonadotropin (HCG)] during the preovulatory period with 1000 mg/kg Carbendazim (MBC). The compound, known to bind fast but reversibly to mammalian tubulin, was chosen to investigate whether the stage at which spindle function is inhibited affects the pattern of nondisjunction. Ovulated oocytes were cytologically prepared and scored for hyperhaploidy, diploidy and presegregation. Application at an early spindle phase, 4.5 h post HCG, to females stimulated with a low gonadotropin dose [3 IU pregnant mares serum (PMS); 2 IU HCG] caused a high frequency of nondisjunction (40.6%) with a more or less nonspecific pattern of malsegregated bivalents. Treatment at a late stage of spindle function (6 h post HCG) resulted in a less frequent (22.5%) but highly preferential malsegregation of those A-D group bivalents thought earlier to be late segregators. On the other hand, oocytes from females primed with a high (10 IU PMS and HCG) gonadotropin dose, a treatment assumed to delay meiosis by approximately 1.5 h, responded to MBC treatment at the late stage (6 h) with a nonspecific pattern and a high frequency (71.2%) of nondisjunction. The latter result is comparable to that in which MBC was given at the early stage (4.5 h) and after a low gonadotropin dose. The high nondisjunction response additionally indicates that spindles in hypergonadotropic stimulated oocytes are more susceptible and/or that the concentration of the inhibitor is higher in such oocytes. Only few oocytes with presegregation (3.1%; 0.0%; 1.7%) and few diploid oocytes (3.3%; 1.5%; 3.2%) with complete inhibition of meiosis I were observed. We conclude, that in Djungarian hamsters (1) the segregation of bivalents at meiosis I is asynchronous with the large A-D bivalents segregating last, (2) the phase in which spindle function is inhibited determines the pattern of nondisjunction, and (3) the resumption of meiosis I — from dictyotene to metaphase II — does not follow a rigidly timed programme but depends on the conditions of follicular maturation.  相似文献

We tested the hypothesis that the weight lost by female Great and Blue Tits Parus major and P. caeruleus while raising their first brood influences their ability to start a second brood. The evening weight of female parents was recorded when the nestlings were 5 and 13 days old, in different years and habitats. Several predictions were tested: (1) both species lose weight while raising nestlings and Great Tit females which start a second brood lose less weight than females which do not; (2) differences in the average weight lost between years and areas correlate with differences in the proportion of second broods; (3) the relative weight loss in Blue Tits, which only rarely undertake second broods, is higher than in Great Tits in which second broods are more common. Other factors also are related to the probability of undertaking a second brood: more second broods are undertaken by more successful females, adult females and females that lay earlier.
The comparison of Great and Blue Tits suggests that the two species use different reproductive strategies.  相似文献

The effect of maternal age on the incidence of chromosomal abnormalities was investigated on a large sample of 3,042 in vitro unfertilized human oocytes II obtained from 792 women aged 19-46 years and participating in an in vitro fertilization program for various indications. The chromosomal analysis combined a gradual fixation of oocytes and an adapted R-banding technique. A total of 1,397 interpretable karyotypes were obtained. Various types of numerical aberration were observed, involving conventional chromosome nondisjunction (3.5%), single-chromatid nondisjunction (5.9%), complex (0.8%) or extreme aneuploidy (0.5%), diploidy (5.4%), and set of single chromatids (3.8%). No significant difference was found in the mean age of women according to the various types of chromosomal abnormalities. A positive relationship was found between maternal age and the global rate of aneuploidy, in agreement with the findings of epidemiological studies. The incidence of both whole-chromosome nondisjunction and precocious chromatid separation were correlated to maternal aging but the most significant correlation was found between maternal aging and single-chromatid nondisjunction. The rate of diploidy was also correlated to a slight extent to maternal aging, whereas no correlation was found between maternal age and the rate of single-chromatid sets. These data reveal that single-chromatid malsegregation is an essential factor in the age-dependent occurrence of nondisjunction in human oocytes.
