Effect of d-mannose treatment on different antioxidants, phenolics, protease activity, lipid peroxidation, DNA damage and cell death was investigated in coleoptiles of etiolated wheat seedlings. Modulations in these biochemical parameters were monitored up to 96 h after treatment at 24 h intervals. With accelerating effect on initial signs of cell death, i.e., appearance of long DNA fragmentation and no effect on initiation of terminal stage, i.e., internucleosomal nDNA fragmentation, mannose treatment (1 % = 56 mM) diminished the antioxidant activities in wheat coleoptiles. Mannose treatment decreased the catalase activity at all intervals, while APX and POD activities decreased at 72 h. Peroxidation of lipids increased at 72 h after mannose treatment. Levels of most of antioxidants, i.e., SOD, peroxidases and phenolics were raised during initial time period (24–48 h) of mannose treatment probably as an attempt to counter the stress effect. Protease activity gradually increased and protein content decreased with time in both treated and non-treated coleoptiles. Sharp decrease in CAT, APX and peroxidase activities and increase in lipid peroxidation at 72 h overlaps with apoptotic internucleosomal nDNA fragmentation in this organ. This coincidence points towards the importance of compromised antioxidant defense and involvement of reactive oxygen species in initiation of terminal stage of programmed cell death in wheat coleoptile. In conclusion, accelerating effect on DNA fragmentation and lipid peroxidation along with diminished antioxidant activities at the time of internucleosomal nDNA fragmentation, provide evidence for pro-apoptotic effect of d-mannose in wheat coleoptile.  相似文献

To study the effects of application of salicylic acid (SA) on the antioxidant system and photosystem II (PS II) in wheat seedlings we used two different experiments. The first method was carried out by immersing roots in Hoagland’s nutrient solution containing 0, 0.25, or 2.5 mM SA, and the second method was performed by spraying two-week-old seedlings with the same SA concentrations. After 24 h, chlorophyll fluorescence, thylakoid membrane proteins, antioxidant enzyme activities, and reactive oxygen species were measured. The low concentration of SA caused a significant increase in the antioxidant enzyme activities. However, the treatment with 2.5 mM SA resulted in an increase in the non-photochemical quenching coefficient and a decrease in the antioxidant enzyme activities, the quantum yield of PS II photochemistry, and the photochemical quenching, especially in the first method of application. All these results indicate that the effects of SA on PS II and the antioxidative defense system were dependent on the concentration used and the method of application.  相似文献

The dynamics of changes in total proteolytic activity and activities of various groups of proteases in the coleoptiles of 3- to 12-day-old wheat seedlings grown in light with and without antioxidant BHT (2,6-di-tert-butyl-4-methylphenol) was studied. It was established that the specialized proteases that easily hydrolyze specific synthetic substrates and the enzymes actively hydrolyzing histone H1 dominate in young coleoptiles of 3- to 4-day-old seedlings. Proteases that degrade equally well the majority of the studied substrates are accumulated in the cells of old coleoptiles of 11- to 12-day-old seedlings. Under the effect of BHT, the plants grown in light (in comparison with etiolated seedlings) demonstrated a somewhat changed dynamics of proteolytic activity in young coleoptiles and the disappearance of proteases active toward histone H1. An inhibitory analysis revealed a relative domination of cysteine proteases in young coleoptiles at the initial development stage of seedlings, whereas the fraction of serine proteases markedly increased in old coleoptiles. We presume that the revealed quantitative and qualitative changes in the proteolytic apparatus of the coleoptile cells induced by BHT may be largely responsible for the retardant and geroprotective effect of this antioxidant in plants.  相似文献

In plants of wheat (Triticum aestivum L.) grown in the media with nitrate (NO 3 ? plants), ammonium (NH 4 + plants), and without nitrogen (N-deficient plants), the response to oxidative stress induced by the addition of 300 mM NaCl to the nutrient solution was investigated. Three-day-long salinization induced chlorophyll degradation and accumulation of malondialdehyde (MDA) in the leaves. These signs of oxidative stress were clearly expressed in NO 3 ? and N-deficient plants and weakly manifested in NH 4 + plants. In none of the treatments, salinization induced the accumulation of MDA in the roots. Depending on the conditions of N nutrition, salt stress was accompanied by diverse changes in the activity of antioxidant enzymes in the leaves and roots. Resistance of leaves of NH 4 + plants to oxidative stress correlated with a considerable increase in the activities of ascorbate peroxidase and glutathione reductase. Thus, wheat plants grown on the NH 4 + -containing medium were more resistant to the development of oxidative stress in the leaves than those supplied with nitrate.  相似文献

Rice (Oryza sativa) and wheat (Triticum aestivum) are the most important starch crops in world agriculture. While both germinate with an anatomically similar coleoptile, this tissue defines the early anoxia tolerance of rice and the anoxia intolerance of wheat seedlings. We combined protein and metabolite profiling analysis to compare the differences in response to anoxia between the rice and wheat coleoptiles. Rice coleoptiles responded to anoxia dramatically, not only at the level of protein synthesis but also at the level of altered metabolite pools, while the wheat response to anoxia was slight in comparison. We found significant increases in the abundance of proteins in rice coleoptiles related to protein translation and antioxidant defense and an accumulation of a set of enzymes involved in serine, glycine, and alanine biosynthesis from glyceraldehyde-3-phosphate or pyruvate, which correlates with an observed accumulation of these amino acids in anoxic rice. We show a positive effect on wheat root anoxia tolerance by exogenous addition of these amino acids, indicating that their synthesis could be linked to rice anoxia tolerance. The potential role of amino acid biosynthesis contributing to anoxia tolerance in cells is discussed.  相似文献

Effects of Ca2+ ions on the intensity of lipid peroxidation, activities of guaiacol peroxidase, superoxide dismutase (SOD), and catalase, as well as on heat resistance of winter wheat (Triticum aestivium L.) coleoptiles were examined. A preliminary incubation of coleoptile segments in a 5 mM CaCl2 solution was shown to improve their survival rates after an injuring heat treatment (43.5°C). The effect of Ca2+ was suppressed by the inhibitor of Ca2+ channels (1 mM LaCl3). An incubation of coleoptiles in the presence of 5 mM CaCl2 prior to the stress treatment elevated the content of lipid peroxidation product, malondialdehyde (MDA) and stimulated the activities of guaiacol peroxidase, SOD, and catalase. After the heat exposure of untreated and Ca2+-treated seedlings, differential changes in MDA content and in activities of guaiacol peroxidase, SOD, and catalase were observed. It was established that total proteolytic activity in etiolated wheat seedlings changes in ontogenesis in cycles: peaks of proteolytic activity correspond to the 3rd, 5th, and 8th days of seedling growth, respectively. The maximum of proteolytic activity preceded the maximum of nuclease activity, which may be due to activation of nucleases by proteolytic enzymes. According to inhibitory analysis the cysteine and serine proteases play the main role in apoptosis in wheat coleoptiles. Growing of seedlings in the presence of ethrel stimulated apoptosis in the coleoptile, and it increased (almost 6-fold) the proteolytic activity in its cells. On the other hand, the antioxidant ionol (BHT) suppressed the induction of proteases, particularly at the second stage of coleoptile development, and it slowed down the increase in the nuclease activity after 6th day of the seedling life. It is suggested that phytohormones and antioxidants participate in regulation of apoptosis in the ageing coleoptile, directly or indirectly effecting the proteolytic apparatus in the coleoptile cells.  相似文献

Seven-day-old seedlings of winter wheat (Triticum aestivum L.) in a growth chamber were exposed to ultraviolet-B (UV-B) irradiation for 20 days with daily biologically effective (BE) UV-B irradiation (UV-BBE) at low (4.2 kJ m−2 day−1, LUVB) and high (7.0 kJ m−2 day−1, HUVB) levels. The UV-B irradiated seedlings and the control without UV-B irradiation were then subjected to freezing stress at −6 °C for 6 h and recovered to 20 °C with gradually increased temperature, to investigate the effects of UV-B irradiation on freezing tolerance. During the UV-B exposure, both LUVB and HUVB irradiated seedlings had lower half lethal temperature (LT50) values in comparison with the control, and LUVB more effectively decreased the LT50 values than HUVB. Moreover, foliar concentrations of thiobarbituric acid reactive substances (TBARS) in the UV-B irradiated seedlings were lower than that of control after recovery from freezing stress. Hydrogen peroxide (H2O2) rapidly increased after UV-B exposure, as did activity of superoxide dismutase (SOD). After recovery from freezing stress, activities of catalase (CAT), guaiacol peroxidase (GPX) and glutathione reductase (GR) increased in both LUVB and HUVB leaves, whereas activities of ascorbate peroxidase (APX) and monodehydroascorbate reductase (MDHAR) significantly increased only in the LUVB leaves. Furthermore, the ascorbic acid (AsA) concentration and reduced-to-oxidized ascorbate ratio (AsA/DHA) increased in the LUVB leaves both at the end of UV-B exposure and after recovery from freezing stress. However, the reduced glutathione (GSH) concentration, together with reduced-to-oxidized glutathione ratio (GSH/GSSG) increased in both LUVB and HUVB leaves after recovery from freezing stress. UV-B irradiation increased freezing tolerance in winter wheat seedlings, and this response appears to involve the scavenging enzymes and compounds in the antioxidant defense systems, particularly the ascorbate–glutathione cycle.  相似文献

Reactive carbonyl species methylglyoxal (MG) has always been regarded as a cytotoxic metabolite, but now is emerging to function as signal molecule in plants. However, whether MG can induce salt tolerance is elusive. In this study, treatment of wheat seeds with NaCl reduced seed germination, plant height, root length, fresh weight, and dry weight, indicating the inhibitive effects of NaCl on seed germination and seedling growth. The inhibitive effects of NaCl were alleviated by applying exogenous MG, but aggravated by the MG scavenger N-acetyl-L-cysteine (NAC), suggesting that MG could induce the salt tolerance of wheat. In addition, MG increased glyoxalase I and glyoxalase II activities and decreased endogenous MG content in wheat seedlings under NaCl stress, whereas coapplication of NAC weakened glyoxalase activity and enhanced the endogenous MG level. Also, MG activated superoxide dismutase, catalase, ascorbate peroxidase, and glutathione reductase activities; increased glutathione and ascorbic acid levels; and decreased superoxide radical production and H2O2 and malondialdehyde contents under NaCl stress, while NAC reversed these physiological parameters. Furthermore, MG also induced the accumulation of proline, glycine betaine, and soluble sugar under NaCl stress, whereas this accumulation was weakened by NAC. This work reported for the first time that MG could induce the salt tolerance of wheat, and the acquisition of this salt tolerance was involved in the activation of the glyoxalase system and antioxidant system, as well as the accumulation of osmolytes.  相似文献

We investigated the effects of exogenous sodium nitroprusside (SNP), a nitric oxide (NO) donor, on growth of cucumber (Cucumis sativus L., cv. Jinyou No.1) seedlings and antioxidant enzyme activities in cucumber leaves under waterlogging stress. The growth of cucumber seedlings was significantly inhibited when plants were exposed to waterlogging, whereas shoot spraying with SNP significantly alleviated the inhibition of growth from this type of stress: height, fresh and dry weights of the flooded plants increased obviously. Waterlogging also caused the activation of the antioxidant enzymes (superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), and ascorbate peroxidase (APX)), the reduction of the chlorophyll content, and the accumulation of MDA and protein in leaves. It was found that SNP treatment further potentiated the antioxidant enzyme activities and maintained the chlorophyll and protein content during the entire water-logging period; however, it reduced the MDA content. Thus, NO protects plants from oxidative damage and promotes growth by activation of antioxidant enzymes in leaves in an extent sufficient for the alleviation of membrane injury. However, exogenous NO had no significant effects on cucumber seedlings growth and antioxidant enzyme activities under nonstress conditions.  相似文献

This study examined the ability of jasmonic acid (JA) to enhance drought tolerance in different Brassica species in terms of physiological parameters, antioxidants defense, and glyoxalase system. Ten-day-old seedlings were exposed to drought (15 % polyethylene glycol, PEG-6000) either alone or in combination with 0.5 mM JA. Drought significantly increased lipoxygenase activity and oxidative stress, levels of malondialdehyde and H2O2. Drought reduced seedling biomass, chlorophyll (chl) content, and leaf relative water content (RWC). Drought increased proline, oxidized ascorbate (DHA) and glutathione disulfide (GSSG) levels. Drought affected different species differently: in B. napus, catalase (CAT) and glyoxalase II (Gly II) activities were decreased, while glutathione-S-transferase (GST) and glutathione peroxidase (GPX) activities were increased in drought-stressed compared to unstressed plants; in B. campestris, activities of glutathione reductase (GR), glyoxalase I (Gly I), GST, and GPX were increased, monodehydroascorbate reductase (MDHAR), dehydroascorbate reductase (DHAR), CAT and other enzymes were decreased; in B. juncea, activities of ascorbate peroxidase, GR, GPX, Gly I were increased; Gly II activity was decreased and other enzymes did not change. Spraying drought-stressed seedlings with JA increased GR and Gly I activities in B. napus; increased MDHAR activity in B. campestris; and increased DHAR, GR, GPX, Gly I and Gly II activities in B. juncea. JA improved fresh weight, chl, RWC in all species, dry weight increased only in B. juncea. Brassica juncea had the lowest oxidative stress under drought, indicating its natural drought tolerance capacity. The JA improved drought tolerance of B. juncea to the highest level among studied species.  相似文献

Among the heavy metals (HMs), lead (Pb) is considered as a toxic HM which adversely affects growth and development of crop plants. The present experiment was aimed to investigate the potential role of ascorbic acid (ASC) in the reversal of Pb-inhibited nitrogen and sulfur assimilation enzymes activity and activity of photosynthesis enzyme ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) and growth response in wheat plants. Wheat seedlings were subjected to 0 mM (control) and 0.2 mM and 0.6 mM of ASC with and without 2 mM of Pb. Plants treated with Pb exhibited the following reduced growth characteristics (root length, shoot length, root fresh weight (FW), shoot FW, root dry weight (DW) and shoot DW). A decrease was also observed in the activity of Rubisco and ATP sulfurylase (ATP-S), relative water content (RWC), accumulation of total chlorophyll (Total Chl) and content of nutrients [nitrogen (N), phosphorus (P), potassium (K), calcium (Ca) and magnesium (Mg)] in Pb-treated plants. However, an increase in Chl degradation and in the activity of O-acetylserine(thiol)lyase (OAS-TL) and accumulation of cysteine (Cys), malondialdehyde (MDA) and hydrogen peroxide (H2O2) was observed in plants under Pb stress. On the contrary, exogenous application of ASC mitigated the Pb-toxicity-induced oxidative damage by enhancing the activities of antioxidant enzymes, such as superoxide dismutase, catalase and glutathione reductase. Improved activity of antioxidant enzymes suppressed the formation of MDA and H2O2, which was reflected in the form of improved growth characteristics. Moreover, ASC induced improvement in plants defense systems by reduced Chl degradation and improved the content of essential nutrients (N, P, K, Ca and Mg) and Cys, RWC and the activity of Rubisco, ATP-S, NR and OAS-TL.  相似文献

Indoleacetic acid at 0.017 millimolar inhibited the formation of three peroxidase isoenzymes in both soluble and wall-bound enzyme fractions of wheat coleoptile (Triticum vulgare) tissue. Hydroxyproline at 1 millimolar prevented the indoleacetic acid-induced inhibition. Indoleacetic acid oxidase activity in the soluble fraction was decreased by indoleacetic acid and was restored by hydroxyproline. Most of the indoleacetic acid oxidase activity was located in the electrophoretic zones occupied by two of the peroxidase isoenzymes influenced by indoleacetic acid and hydroxyproline. At least part of the effect of hydroxyproline on auxin-induced elongation of coleoptile tissue may be through control of auxin levels by indoleacetic acid oxidase.  相似文献

高寒山区植物根抗氧化酶系统的季节变化与抗冷冻关系   总被引:5,自引:0,他引:5  
在高寒山区(海拔2900m)和选取4种多年生草本植物,即无芒雀麦(Bromus inermis)、草地早熟禾(Poa sphyondylodes)、花誉麦(Bromus sinensis)和垂重申披碱草(Elymus nutans),测定了秋末、冬初、冬季、春季气温变化过程中其根中丙二醛(MDA)含量和抗氧酶活力(过氧化氢酶(CAT)、过氧化物酶(POD)、超氧物歧化酶(SOD))和抗坏血酸氧化酶(APX)变化,分析了抗氧酶系统在根抗冷适应中的作用,结果表明,随秋末降温植物根中MDA含量增加,尔后下降,在冬季和翌年春季保持相对稳定。从9月初到10月下旬,4种植物根中SOD、CAT、POD活力平均增加170%、130%和56%。在冬季下降,但仍远高于9月,在春季气温上升过程中酶活力上升。根能在组织结冰状况下生存与其具备完善的保护酶系统,能及时清除氧自由基抑制膜脂过氧化维持膜完整性有关,据降温过程中MDA含量和抗氧酶活力变化,可将根冷适应分为两个阶段,即第1阶段平均气温在0℃以上,抗氧酶活力增强,MDA增加阶段,第2阶段平均气温降至0℃以下,最低气温降到-15℃以下,抗氧酶活力下降,MDA无明显变化阶段。  相似文献

The effects of salicylic acid (SA) and salinity on the activity of apoplastic antioxidant enzymes were studied in the leaves of two wheat (Triticum aestivam L.) cultivars: salt-tolerant (Gerek-79) and salt-sensitive (Bezostaya). The leaves of 10-d-old seedlings grown at nutrient solution with 0 (control), 250 or 500 mM NaCl were sprayed with 0.01 or 0.1 mM SA. Then, the activities of catalase (CAT), peroxidase (POX) and superoxide dismutase (SOD) were determined in the fresh leaves obtained from 15-d-old seedlings. The NaCl applications increased CAT and SOD activities in both cultivars, compared to those of untreated control plants. In addition, the NaCl increased POX activity in the salt-tolerant while decreased in the salt-sensitive cultivar. In control plants of the both cultivars, 0.1 mM SA increased CAT activity, while 0.01 mM SA slightly decreased it. SA treatments also stimulated SOD and POX activity in the salt-tolerant cultivar but significantly decreased POX activity and had no effect on SOD activity in the saltsensitive cultivar. Under salinity, the SA treatments significantly inhibited CAT activity, whereas increased POX activity. The increases in POX activity caused by SA were more pronounced in the salt-tolerant than in the salt-sensitive cultivar. SOD activity was increased by 0.01 mM SA in the salt-tolerant while increased by 0.1 mM SA treatment in the salt-sensitive cultivar.  相似文献

The effect of pretreatment with 0.05 mM salicylic acid (SA) on the activity of superoxide dismutase (SOD) and peroxidase in the roots of four-day-old seedlings of wheat (Triticum aestivum L.) was studied under conditions of salination. The level of the stress-induced accumulation of active oxygen species and, therefore, activities of SOD and peroxidase in seedlings pretreated with SA were significantly lower than in untreated seedlings, which indicates that these enzymes contribute to the protective effect of SA on plants under conditions of salination.  相似文献
