For the first time, we studied the karyotype of the flathead sculpin Megalocottus platycephalus taeniopterus (Kner, 1868) from Vostok Bay of the Sea of Japan. The karyotype is stable: 2n = 42 (2 metacentric, 2 submeta-subtelocentric, 30 subtelocentric, and 8 acrocentric chromosomes), NF = 44 + 2. The nucleolar organizers (NOs) were identified using Ag banding in two pairs of chromosomes: in the telomeric parts of the short arm of the medium-size subtelocentric chromosome and the long arm of the large acrocentric chromosome. Variations in the number of nucleolar organizer chromosomes and in the number of NO staining blocks were found. Comparison of the karyotypes of M. p. taeniopterus and previously studied M. p. platycephalus (Pallas, 1814) from the northern Sea of Okhotsk revealed their similarity in the number and morphology of chromosomes and the number of chromosome arms and difference between the subspecies in the number and location of NO, which allows their discrimination.  相似文献

The karyotype of the endemic Okhotsk Sea sculpin Myoxocephalus ochotensis Schmidt (Cottidae) from Odyan Bay was studied. The number and morphology of chromosomes were determined, 2n = 42 (2 metacentric, 20 subtelocentric, and 20 acrocentric chromosomes), NF = 44. Variability of chromosome number was not revealed; no difference between male and female karyotypes was found. The karyotype of the Okhotsk sculpin M. ochotensis was compared with karyotypes of the Far East Steller’s M. stelleri (Tilesius), snow M. brandti (Steindachner), and plain M. jaok (Cuvier) sculpins, and to the European shorthorn sculpin M. scorpius (Linnaeus) from White Sea. The karyotype of the great sculpin, Myoxocephalus polyacanthocephalus (Pallas, 1814) (Pisces: Cottidae) from the Sea of Okhotsk and the Sea of Japan has been studied for the first time. The karyotype is stable; it consists of 40 chromosomes (4 metacentric, 2 submeta-subtelocentric, 20 subtelocentric, and 14 acrocentric chromosomes); the number of chromosomal arms is 44 + 2. Nucleolar organizer regions (NOR) are found in the telomeric region of the arm in one homologue of a pair of small metacentric chromosomes, using the Ag-NOR banding technique. A comparative analysis of the karyotype of M. polyacanthocephalus and the karyotypes of other Myoxocephalus species (M. stelleri, M. brandtii, M. jaok, M. ochotensis, and M. scorpius) has been carried out based on the main karyotype characters, as well as on the number and localization of NORs. The identified differences make it possible to differentiate the studied species, whereas the general traits indicate their taxonomic proximity.  相似文献

Molecular genetic and karyological analyses of antlered sculpin, Enophrys diceraus, from the Sea of Japan and the Sea of Okhotsk were carried out. The karyotype of this species was studied for the first time. On the basis of karyological analysis, it was established that E. diceraus from the Sea of Japan and the Sea of Okhotsk was polymorphic in terms of the number of chromosomes and their morphology (2n = 36, 35, and 37, NF = 40). It was suggested that the karyotype with 35 chromosomes could have been produced as a result of Robertsonian translocation; the karyotype with 37 chromosomes could have been produced by crossing of individuals with different number of chromosomes. On the basis of the molecular genetic analysis of the mitochondrial COI and 16S rRNA genes, considerable differences between E. diceraus from the Sea of Japan and the Sea of Okhotsk corresponding to the level of interspecies genetic variability were established. It is concluded that E. diceraus from the Sea of Japan belongs to another species, most likely, E. namiyei.  相似文献

The karyotype and major ribosomal sites as revealed using silver staining of Anatolian leuciscine cyprinid fish Acanthobrama marmid were studied. The diploid chromosome number was invariably 2n = 50. Karyotype consisted of eight pairs of metacentric, 13 pairs of submetacentric and four pairs of subtelocentric to acrocentric chromosomes. The largest chromosome pair of the complement was subtelo-to acrocentric characteristically, which is a characteristic cytotaxonomic marker for representatives of the cyprinid lineage Leuciscinae. The nucleolar organizer regions (NORs) were detected in the telomeres of two pairs of medium sized submeta-to subtelocentric chromosomes. No heteromorphic sex chromosomes were found. The karyotype pattern of A. marmid is nearly identical to that found in most other representatives of the Eurasian leuciscine cyprinids, while the multiple NOR phenotype appears to be more derived as opposed to a uniform one, ubiquitous in this group.  相似文献

The karyotype of a subspecies of the golden loach,Sabanejewia aurata balcanica from eastern Slovakia was studied by conventional Giemsa staining, Ag-NOR staining, and C-banding. The diploid chromosome number was 2n = 50. The karyotype comprised 2 pairs of metacentric, 6 pairs of submetacentric and 17 pairs of subtelocentric to acrocentric chromosomes. Both metacentric pairs and 2 large subtelocentric pairs had massive pericentromeric blocks, while all other elements had only weak blocks of heterochromatin. The NORs were localized on the short arms of one middle-sized subtelocentric pair. The karyotype ofS. a. balcanica differs from that ofS. aurata kubanica, suggesting chromosomal polymorphism of this widely distributed, polytypic cobitid species. The polymorphic karyotypes of the golden loach may thus demonstrate transient stages, linking primitive and advanced cobitid karyotypes.  相似文献

The Choeroniscus genus (Glossophaginae, Phyllostomidae) has five monotypic species: C. minor, C. godmani, C. intermedius, C. inca and C. periosus. This paper analyses the karyotype of a female C. minor, collected close to the Guama river (Belém, Para, Brazil). G-, C-banding and NOR-staining were performed. This species has 2n = 20 chromosomes, where there are two bi-armed pairs (numbers 1 and 9) and seven subtelocentric pairs (2, 3, 4, 5, 6, 7 and 8). The probable X chromosome is a submetacentric. The constitutive heterochromatin can be found in the short arm of five subtelocentric pairs (4, 5, 6, 7 and 8) and is centromeric in the bi-armed pairs numbers 1 and 9, and the X chromosome. The heterochromatic bands are heteromorphic in three pairs (1, 2 and 3). Active NOR were observed in the short arms of eight subtelocentric chromosomes, suggesting that at least four pairs are nucleolar organizers. This paper describes for the first time the karyotype of C. minor from the Amazon region.  相似文献

The phylogenetic relationships among five species of sculpins, including Myoxocephalus stelleri, M. brantii, M. jaok, M. ochotensis, and Megalocottus platycephalus, were estimated from the sequence variability of the mtDNA cytochrome b (cytb) and cytochrome oxidase 1 (CO1) genes. Analysis of the topologies of combined phylogenetic trees showed that all of the morphologically described species from different genera represented monophyletic groups with high support of branch robustness. Haplotypes with different karyotypes from different geographical localities with an intragroup nucleotide diversity of 0.42% were combined into the M. stelleri clade. All of the species (except for M. stelleri from the Sea of Japan and the Okhotsk Sea coast of the Hokkaido Island) were characterized by relatively low values of the intragroup variation, along with high values of interspecific variation of mtDNA-encoded markers. Studies of the M. stelleri karyotypes using Ag-banding showed that karyotypes of the individuals from the Sea of Japan and Okhotsk differed not only in the number of chromosomes (2n) but also in the number of active nucleolus organizers (NO) and stained NO blocks, calling into question whether they belong to a single species. The observed discrepancy between the phylogenetic topologies and karyological data is discussed in relation to the informative capacity of mtDNA fragments and the phenomenon of intraspecific chromosomal polymorphism.  相似文献

Resident (lacustrine) chars, inhabiting many lakes of Kamchatka, Chukotka, and continental coast of the Sea of Okhotsk, are of particular interest for understanding the mechanisms of speciation and evolution of Salvelinus chars. Since one of these, the char from Dal'nee Lake (Paratunka River basin, southeastern Kamchatka), is substantially different from lacustrine-riverine and anadromous chars from the Paratunka River basin in several morphological traits, hemoglobin spectrum, ecology, and reproduction, it is considered an isolated species Salvelinus krogiusae. The karyotype of the resident char from Dal'nee Lake was shown to be variable in most individuals, containing 76 to 80 chromosomes in different cells, with NF = 98. This suggests that the variability of the chromosome number in this char form is explained by Robertsonian translocations. In females and males, cells with respectively 2n = 78 and 2n = 77 prevailed. Heteromorphic sex chromosomes were not found in the karyotype of this char species. Nucleolar-organizing regions (NORs) were mostly detected in the telomeric regions of short arms of large submetacentric chromosomes. These features of the Krogius char karyotype demonstrate that this species is isolated from other Kamchatka chars, being similar to Taranetz char S. taranetzi from Chukotka Peninsula, which is in good agreement with morphological data.  相似文献

In a sample of 108 underground voles from 23 Greek localities, the species Microtus felteni, M. guentheri, M. rossiaemeridionalis and M. subterraneus were identified, based on external body morphology and karyotype. Moreover, the implemented C-banding staining technique revealed the heterochromatin distribution in the chromosomes of the above species. All M. guentheri (2n=54, FN=54) and M. rossiaemeridionalis (2n=54, FN=56) specimens displayed the typical karyotypes of these species, respectively. The M. subterraneus specimens belonged to the chromosomal race with 2n=52, FN=60 of this species, apart from a single individual that demonstrated a medium-sized, subtelocentric autosome in heterozygous condition (2n=52, FN=61). Furthermore, M. felteni individuals, trapped again after many years, were karyologically studied (2n=54, FN=56) and the C-banding pattern for this species is hereby presented for the first time. Finally, the study of meiotic preparations in M. guentheri and M. rossiaemeridionalis males verified the asynaptic behaviour of their sex chromosomes. The karyotype of the four studied Microtus species does not seem to have diversified much from the putative ancestral arvicoline karyotype (2n=56, FN=56). On the other hand, the heterochromatin accumulation in the sex chromosomes, particularly prominent in M. felteni and M. rossiaemeridionalis, represents this common tendency in several Microtus species.  相似文献

A karyometric analysis of the chromosome set of the marine turbellariansMonocelis fusca, M. lineata andParotoplana macrostyla has been carried out. The karyotype of the twoMonocelis species investigated (2n=6) is formed by three pairs of small and similarly sized chromosomes: InM. fusca, chromosome 1 is metacentric, chromosome 2 acrocentric and chromosome 3 is subtelocentric.M. lineata also presents one pair of metacentric chromosomes (chromosome 2), while chromosomes 1 and 3 are submetacentric.P. macrostyla (2n=12) reveals two pairs of large metacentric and four pairs of small chromosomes, three of which are metacentric, whereas the last is subtelocentric.  相似文献

The diploid chromosome number of Gymnothorax unicolor (Delaroche, 1809) is 2n=42, the karyotype comprising six pairs of meta-submetacentric and fifteen pairs of acrocentric chromosomes. C-positive chromatin is present in the centromeres of all chromosomes as well as in the paracentromeric regions of some chromosomes. A nucleolar organizer region was identified on the long arm of chromosome 9, near the centromere. This region is also positive to C-banding.Cytotaxonomical relationships are evidenced between the described karyotype and that of the related species Muraena helena.  相似文献

The karyotypes of four species of Dyscophinae and eight species of Cophylinae were analyzed. The chromosome number was 2n=26 in all cases. Between the two subfamilies a difference in the form of the karyotype was observed; the chromosomes show a gradual decrease in length in the Dyscophinae, whereas in the Cophylinae the karyotype demonstrates a clear discontinuity of size between pairs 5 and 6.Chromosomal polymorphism was found in Plethodontohyla tuberata, the chromosomes of pair 4 were subtelocentric in the homozygous specimens, whereas this pair showed a subtelocentric and a submetacentric chromosome of equal length in the heterozygous one, suggesting a pericentric inversion. Although in the Cophylinae the chromosome number is constant, the number of chromosome arms is variable. Pericentric inversions seem to play an important role in the chromosomal evolution of the Cophylinae.  相似文献

A new karyotype for blind mole rats was recorded in Tunceli province in Eastern Turkey. The karyotype contained 44 chromosomes, including 13 biarmed pairs, 7 acrocentric pairs, and one heteromorphic pair with a submetacentric and an acrocentric homologue in the autosomal complement (FNa=69). The X chromosome was submetacentric and the Y chromosome medium-sized subtelocentric (FN=73). Distinct dark centromeric C-bands were observed on most of the biarmed and three pairs of the acrocentric autosomes. The NORs were detected on short arms of three subtelocentric pairs and one acrocentric pair of autosomes. The diploid number of chromosomes and the karyotype characteristics observed are obviously unique among hitherto studied populations of blind mole rats and the complement can be evaluated as a new chromosome race of Nannospalax xanthodon. The distribution ranges of individual chromosome races of the species recorded in Eastern Anatolia are revised and possible interracial hybridization is discussed in respect of the finding of a new race.  相似文献

Morphometric and karyotypic studies were made on two species of ricefishes collected from Yunnan, southwestern China.Oryzias latipes from Yunnan had the same morphological and karyological characteristics asO. latipes collected from eastern China. The Yunnan populations had 2n, 46 chromosomes consisting of 3 metacentric, 8 submetacentric, 2 subtelocentric, and 10 acrocentric pairs, the arm number (NF) being 68 (2n=46, NF=68, 3M+8SM+2ST+10A). The karyotype was characterized by having a “large” metacentric pair and nucleolus organizer regions (NORs) on the short arms of a submetacentric pair.Oryzias minutilius from Yunnan had the same morphological characteristics asO. minuiillus from Thailand and Burma, although the karyotype was different from that collected from Thailand. The Yunnan population had 2n, 42 chromosomes consisting of 21 acrocentric pairs, NF being 42 (2n=42, NF=42, 21A). The karyotype was characterized by having NORs at the telomeric regions of an acrocentric pair.Oryzias latipes occurs widely on the eastern Yunnan Plateau where the climate is temperate or subtropical, whereasO. minutilius is found in Xishiangbanna, the southern low mountain areas of Yunnan, where the climate is tropical.  相似文献

A study was conducted on the chromosomes of a Rhamdia hilarii (Pisces, Pimelodidae) population. The results suggest that the basic chromosome number is 2n=58, with numerical variation up to a limit of 2n=63, due to the presence of supernumerary chromosomes which seem to be mitotically stable. These chromosomes are metacentrics and can be different in size. The C-banding pattern, showing heterochromatin especially in both telomeric regions, permits their identification in the karyotype. The NORs are located on secondary terminal constrictions on the short arm of a pair of subtelocentric chromosomes. However, there may be heteromorphism in the size of the secondary constrictions and, consequently, in the size of the NORs.  相似文献

Almost all species of the Curimatidae family have a stable karyotype, with a diploid number of 54 metacentric (M) and submetacentric (SM) chromosomes, and one sole nucleolus organizer pair. This family has considerable specific diversity in Argentinean fluvial basins; however, no cytogenetic data are available. Eight species from the Paraná River (Argentina): Cyphocharax voga, C. spilotus, C. platanus, Steindachnerina brevipinna, S. conspersa, Curimatella dorsalis, Psectrogaster curviventris, and Potamorhina squamoralevis were analyzed cytogenetically. Chromosome preparations were obtained from direct samples and through cell culture, and they were processed for conventional, C- and nucleolar organizer region-banding. Six of the species exhibited the standard family karyotype, with 2n = 54 M-SM and fundamental number of chromosomes (FN) = 108, as well as variations in the chromosome formula, and in heterochromatic and nucleolar organizer regions. Though nucleolar organizer regions were located on only one chromosome pair, they varied in both carrier chromosomes and pairs involved. On the other hand, C. platanus showed a complement of 2n = 58 M-SM and subtelocentric with FN = 116, and P. squamoralevis presented 2n = 102, with some M-SM and a large number of acrocentric chromosomes. Even though the karyotype macrostructure appears to be conserved, the speciation process within the family has been accompanied by micro-structural rearrangements, as evidenced by pattern diversity in the heterochromatin and nucleolar organizer regions. Some changes in chromosome macrostructure have also occurred in this group, primarily in C. platanus and P. squamoralevis, in which there have been centric dissociations and inversions.  相似文献

Karyotype, location of the nucleolar organiser region (NOR) and heterochromatin presence and composition were studied in the Antarctic scallop Adamussium colbecki Smith, 1902. The karyotype exhibits 2n = 38 chromosomes with 11 pairs of metacentrics, 5 of submetacentrics, one subtelocentric and two telocentrics. Ag–NOR, CMA3, DA/MM and NOR–FISH evidenced paracentromeric NORs on the short arm of 2nd pair chromosomes. Digestion with three restriction endonucleases followed by sequential staining with Giemsa, CMA3 and DAPI evidenced on all chromosomes centromeric heterochromatin positive for both DAPI and CMA3. In situ hybridisation analysis showed the presence of an AT-rich satellite DNA in the centromeric heterochromatin of several chromosomes. A mosaicism was detected in the germinal cell lines of one specimen, as in six of the 20 plates examined the set had 37 chromosomes with a missing pair of telocentrics and an unpaired metacentric. Comparison of the chromosome sets of all the pectinids studied to date and comparison with a phyletic tree obtained from molecular mitochondrial genes studies yielded good agreement between karyotype morphology and taxonomic classification.  相似文献

Within a total of 50 analyzed specimens a male individual of Trichomycterus davisi has been recorded with 81 chromosomes including 60 metacentric, 18 submetacentric and three subtelocentric chromosomes. When compared with diploid individuals (2n = 54) and the morphological standard of chromosomes, this male is a triploid with 3n = 81 chromosomes. Since staining with silver nitrate indicates three active nucleolar organizer regions (NORs), the three NOR-bearing chromosomes in this individual are genetically active. Analysis of the synaptonemal complex (SC) by electronic microscopy shows that there is an incomplete pairing of the third set of chromosomes in the triploid individual.  相似文献

We determined the karyotypes of nine species of Lessingianthus, eight of which are here analyzed for the first time. The results include the first chromosome count for L. plantaginoides, which is tetraploid with 2n = 64. All species showed a high proportion of metacentric chromosomes combined with a lower proportion of submetacentric pairs. Only L. coriaceus had a subtelocentric chromosome pair. B chromosomes were observed in L. coriaceus and L. varroniifolius, which were subtelocentric. Differences among the karyotypes of the studied species were small, suggesting that karyotype diversity in the genus evolved by small changes in the structure of chromosomes. Karyotype features appear useful to distinguish Lessingianthus from the closely related genera Chrysolaena and Lepidaploa.  相似文献
