Spider orb webs are impressive for their apparently uniform geometric patterns. There are, however, consistent, substantial and taxonomically widespread periphery‐to‐hub differences in the distances between both adjacent radii and between sticky spiral lines. Radii in typical orbs were on average about 4–5 times farther apart at the outer edge than the inner edge of the area covered by sticky lines. Distances between sticky spiral loops were on average about two times larger near the outer edge than in more inner portions. This pattern in sticky spiral spacing was absent in the modified orbs of Nephila clavipes, in which distances between radii varied less. Thus, patterns in sticky spiral spacing may be related to inter‐radial spacing; there is, however, probably no single explanation for all of the different patterns of sticky spiral spacing. The patterned differences in radius and sticky spiral spacing have important consequences for understanding orb function, because the lines in a prey's immediate vicinity largely determine whether it will be stopped and then retained, and elementary physics dictates that contact with more lines will tend to increase prey being stopped and retained. Rather than being a unit trap with a single set of prey capture properties, an orb has locally different trapping properties in different sectors. Abandoning the previous typological style of discussion of ‘the’ ability of a given design to stop and retain prey promises to lead to improved understanding of orb web designs. Published 2014. In almost all vertical orb webs the hub is above the geometric centre and consequently, the extent of the capture area is larger below the hub than above. In addition to this vertical web‐extent asymmetry, orb webs show vertical asymmetries in number of spiral loops, mesh widths, and angles between radii. However, it was unknown whether these asymmetries are adaptations to the web‐extent asymmetry or whether they are linked to gravity in a different way than through web‐extent asymmetry. We reviewed known vertical asymmetries of orb webs, and we analysed the asymmetries of webs built by four different Cyclosa species, which show large intra‐ and inter‐specific variation in web‐extent asymmetry. We found all analysed structural asymmetries to be linked both to web‐extent asymmetry and to gravity: Larger web extents below the hub and gravity both led to more sticky‐spiral loops and to smaller angles between radii below the hub, whereas web‐extent asymmetry and gravity had opposing effects on mesh width (mean and peripheral). Independent of web‐extent asymmetry, almost all analysed webs had narrower peripheral meshes and smaller angles between radii below the hub than above. We interpret the narrow peripheral meshes along the web's lower edge as an adaptation to prevent tumbling prey from escaping, and the small angles between radii as an adaptation to prevent the sticky‐spiral lines in these narrow meshes to come into contact with each other. © 2015 The Linnean Society of London, Biological Journal of the Linnean Society, 2015, 114 , 659–672.  相似文献

A field experiment was conducted for five consecutive years to determine upland rice (Oryza sativa L.) and common bean (Phaseolus vulgaris L.) response to eight P sources at three P rates in an Oxisol of Central Brazil. The P sources tested were triple superphosphate (TSP), Arafertil phosphate partially acidulated (APPA), phosphate of Patos partially acidulated (PPPA), phosphate of Araxa concentrated (PAC), phosphate of Catalao (PC), phosphate of Jacupiranga (PJ), phosphate of Patos de Minas (PPM), and phosphate of Abaete (PA). All phosphate rock sources were of Brazilian origin. The P rates used were 87, 174 and 262 kg P ha-1. Yield response to P sources and rates varied from crop to crop. Rice and bean yields were significantly correlated with Bray 1 P, but not Mehlich 1 P. In the first year, TSP and the two partially acidulated phosphate rocks (APPA, PPPA) produced higher grain yields. In the second year and all remaining years of the experiment, the efficiency of phosphate rock sources as measured by grain yield was equivalent to TSP or partially acidulated P sources. The results suggest that these phosphate rock sources could be used in rice-bean rotations on Brazilian Oxisols. Yield losses in the first year could be partially offset by the addition of a small amount of soluble P.  相似文献

The effect of oral administration of sodium orthovanadate (SOV) and Trigonella foenum graecum seed powder (TSP), a medicinal plant used extensively in Asia, on the mitochondrial metabolism in the alloxan diabetic rats has been investigated. Rats were injected with alloxan monohydrate (20 mg/100 g body wt) or vehicle (Na-acetate buffer), the former were treated with either 2 IU insulin i.p., 0.6 mg/ml SOV ad libitum, 5% TSP ad libitum, and a combination of 0.2% SOV and 5% TSP ad libitum for 21 days. Selected rate-limiting enzymes of the tricarboxylic acid cycle, hydrogen shuttle system, ketone body metabolism, amino acid metabolism and urea cycle were measured in the mitochondrial and cytosolic fractions of liver, kidney and brain tissues of the experimental rats. Majority of the mitochondrial enzymes in the tissues of the diabetic rats had significantly higher activities compared to the control rats. Similarly, the activities of mitochondrial and cytosolic aminotransferases and arginase were significantly higher in liver and kidney tissues of the diabetic rats. The separate administrations of SOV and TSP to diabetic rats were able to restore the activities of these enzymes to control values. The lower dose of SOV (0.2%) administered in combination with TSP to diabetic rats lowered the enzyme activities more significantly than when given in a higher dose (0.6%) separately. This is the first report of the effective combined action of oral SOV and TSP in ameliorating the altered mitochondrial enzyme activities during experimental type-1 diabetes. Our novel combined oral administration of SOV and TSP to diabetic rats thus conclusively proves as a possible method to minimize potential vanadate toxicity without compromising its positive effects in the therapy of experimental type-1 diabetes.  相似文献

Bacterial symbionts that manipulate the reproduction of their host to increase their own transmission are widespread. Most of these bacteria are Wolbachia, but recently a new bacterium, named Cardinium, was discovered that is capable of the same manipulations. In the host species Brevipalpus phoenicis (Acari: Tenuipalpidae) this bacterium induces thelytoky by feminizing unfertilized haploid eggs. The related species B. obovatus and B. californicus are thelytokous too, suggesting that they reproduce in the same remarkable way as B. phoenicis. Here we investigated the mode of thelytokous reproduction in these three species. Isofemale lines were created of all three species and 19 lines were selected based on variation in mitochondrial COI sequences. All B. phoenicis and B. californicus lines (10 and 4 lines, respectively) produced males under laboratory conditions up to 6.7%. In contrast, males were absent from all B. obovatus lines (5 lines). Additional experiments with two B. phoenicis isofemale lines showed that males can be produced by very young females only, while older females produce daughters exclusively. For most lines it was shown that they are indeed feminized by a bacterium as treatment with antibiotics resulted in increased numbers of males up to 13.5%. Amplification and identification of specific gyrB sequences confirmed that those lines were infected with Cardinium. Three out of the five B. obovatus lines did not produce males after treatments with antibiotics, nor did they contain Cardinium or any other bacterium that might induce thelytoky. In these lines thelytoky is probably a genetic property of the mite itself. Despite the different causes of thelytoky, flow cytometry revealed that all 19 lines were haploid. Finally, the taxonomic inferences based on the mitochondrial COI sequences were incongruent with the classical taxonomy based on morphology, suggesting that a taxonomic revision of this group is necessary.  相似文献

Oilseed rape (Brassica napus) lines transformedwith the coat protein (CP) gene of Turnip mosaic virus(TuMV) were used to determine the effectiveness of resistance to TuMV mediatedby CP RNA or coat protein. Lines with one, two, or more copies of transgeneswere produced. T2 and T3 lines containing the CP genewitha functional start codon synthesised coat protein and showed high, but variablelevels of resistance to TuMV (21–96% resistant plants per line). TheT1 and T2 progeny of all lines carrying the CP gene withamutated start codon so that RNA but not protein was expressed, were assusceptible to TuMV as controls. Thus, in these experiments we were able toinduce CP-mediated resistance, but not RNA-mediated resistance.  相似文献

Trigonella foenum graecum seed powder (TSP) and Sodium Orthovanadate (SOV) have been shown to demonstrate antidiabetic effects by stabilizing glucose homeostasis and carbohydrate metabolism in experimental type-1 diabetes. However their efficacy in controlling histopathological and biochemical abnormalities in ocular tissues associated with diabetic retinopathy is not known. The purpose of this study was to investigate the comparative efficacy of individual as well as combination therapy of TSP and SOV in 8 weeks diabetic rat lens and retina. Retinas and lenses were taken from control, alloxan-induced diabetic rats and diabetic rats treated separately with insulin, 5%TSP, SOV (0.6 mg/ml) and a combined dose of SOV (0.2 mg/ml) and 5%TSP for 60 days. Control and each experimental group had six rats. Alterations in the activities of enzymes HK (hexokinase), AR (aldose reductase), SDH (sorbitol dehydrogenase), G-6-PD (glucose-6-phosphate dehydrogenase), GPx (glutathione peroxidase), GR (glutathione reductase) and levels of metabolites like sorbitol, fructose, glucose, MDA (malondialdehyde) and GSH (reduced glutathione) were measured in the cytosolic fraction of lenses besides measuring blood glucose levels and glycosylated haemoglobin. Histopathological abnormalities were studied in the lens using photomicrography and retina using transmission electron microscopy. Blood glucose, glycosylated haemoglobin levels and polyol pathway enzymes AR and SDH increased significantly causing accumulation of sorbitol and fructose in the diabetic lens and treatment with SOV and TSP significantly (p < 0.05) decreased these to control levels. Similarly, SOV and TSP treatments modulated the activities of HK, G-6-PD, GPx and GR in the rat lens to control values. Ultrastructure of the diabetic retina revealed disintegration of the inner nuclear layer cells with reduction in rough endoplasmic reticulum and swelling of mitochondria in the bipolar cells; and these histopathological events were effectively restored to control state by SOV and TSP treatments. In this study SOV and TSP effectively controlled ocular histopathological and biochemical abnormalities associated with experimental type-1 diabetes, and a combination regimen of low dose of SOV with TSP demonstrated the most significant effect. In conclusion, the potential of SOV and TSP alone or in low dose combination may be considered as promising approaches for the prevention of diabetic retinopathy and other ocular disorders.  相似文献

The competitiveness of two transgenic oilseed rape (Brassica napus ssp.napus) lines and their fertile transgenic hybrid was tested in field trials in Belgium and Denmark. The lines contained genes for male sterility, restoration of fertility and herbicide resistance. The competitiveness of the three transgenic lines was related to three non-transformed commercially-grown oilseed rape varieties: Drakkar, Topas and Line. As a reference of a more aggressive crucifer, white mustard (Sinapis alba) was also included in the experiment. The experimental design was a complete block design with two locations, monocultures and mixtures with barley (Hordeum vulgare), three plant densities, four harvest times and four blocks. The yield density relationship of the transgenic oilseed rape lines was not different from that of the non-transgenic varieties in either location. The first harvest times showed a vigorous biomass production of white mustard, which in turn produced a significant difference in the competitive ability between oilseed rape and white mustard. Later, this difference decreased, and in Belgium there was no difference at the last harvest time. Variations within populations may blur actual differences between lines and varieties, and it is argued that unless the experimental design covers a range of competitiveness for which it is possible to detect significant differences, test results reporting a lack of difference between transgenic and non-transgenic plants are of little value.  相似文献

Several insect species seem to persist not only in permanent but also in temporary ponds where they face particularly harsh conditions and frequent extinctions. Under such conditions, gene flow may prevent local adaptation to temporary ponds and may promote phenotypic plasticity, or maintain apparent population persistence. The few empirical studies on insects suggest the latter mechanism, but no studies so far quantified gene flow including both pond types. We investigated the effects of pond type and temporal variation on population genetic differentiation and gene flow in the damselfly Lestes viridis in northern Belgium. We report a survey of two allozyme loci (Gpi, Pgm) with polyacrylamide gel electrophoresis in 14 populations from permanent and temporary ponds, and compared these results with similar data from the same permanent populations one year before. The data suggested that neither pond-drying regime, nor temporal variation have a substantial effect on population genetic structuring and did not provide evidence for stable population differentiation in L. viridis in northern Belgium. Gene flow estimates were high within permanent and temporary ponds, and between pond types. Our data are consistent with a source-sink metapopulation system where temporary ponds act as sinks in dry years, and are quickly recolonized after local population extinction. This may create a pattern of apparent population persistence of this species in permanent and temporary ponds without clear local adaptation.  相似文献

Summary Cytophotometric measurement of the root meristems of seedlings after Feulgen-staining reveals that large differences (up to 58.16%) in nuclear DNA content may occur in the thirty-one cultivated varieties or lines of Helianthus annuus tested. Significant variations (not exceeding 25%) in the amount of DNA, which does not differ between the root and the shoot meristems of a single seedling, are also found to exist within cultivars or lines; even seedlings obtained from seeds collected from different portions of single heads of plants belonging to a selfed line may vary one from the other in this respect. Variations in the number of chromosomes or alterations in the chromosome structure do not account for the differences observed in nuclear DNA content. Karyometric analyses demonstrate that the surface area of squashed interphase nuclei and metaphase chromosomes and the total length of the latter increase with the increase in Feulgen/DNA absorption. DNA thermal denaturation and reassociation kinetics indicate that a frequency variation in repeated DNA sequences goes hand in hand with changes in the size of the genome. These results, supporting the concept that a plant genome is highly flexible, are discussed in relation to other data to be found in the literature on the intraspecific variation in the nuclear DNA content and in relation to the way in which it is produced in H. annuus.  相似文献

A simple tool, TrackBeam, for rapid in situ estimates of potential exposure to the solar beam of objects underneath tree canopies is presented, tested and demonstrated to be useful in insect conservation studies. The tool can be used whenever accurate data on sun exposure is of use e.g., in ecological studies of saproxylic insects. TrackBeam draws upon the principles behind the analysis of hemispherical photographs but its use is much less dependent on the weather at the time of sampling. It may be used for detecting canopy openings in directions corresponding to the solar path. Based on this, periods of time with potential beam exposure and/or the proportion of sky that is not obscured by canopy for objects such as dead wood may be estimated. The results of using TrackBeam compares well between operators and with the corresponding results of analysed hemispherical photographs. Results are presented which show that TrackBeam was successfully used to characterise the habitat light requirements of the saproxylic beetle Melanotus castanipes (Coleoptera: Elateridae).  相似文献

Invasive ants are a significant conservation concern and can have far-reaching effects in ecosystems they invade. We used the experimental control of ant numbers on two pairs of small (<5 ha) offshore islets dominated by either the big-headed ant, Pheidole megacephala or the tropical fire ant, Solenopsis geminata to investigate the influence of these species on seabird hatching success, fledging success and weight. Limited unpublished observations of both ant species attacking nesting seabirds exist, but the frequency of attacks or how they affect seabird growth and survival are unknown. Island-wide treatments with hydramethylnon resulted in the eradication of P. megacephala and the temporary reduction of S. geminata densities. No difference in hatching success, growth, or fledging success of Wedge-tailed Shearwaters (Puffinus pacificus), a common colonial nesting seabird in the Hawaiian Islands was observed on the pair of islets dominated by P. megacephala. On islets dominated by S. geminata, ant control resulted in a temporary increase in fledging success. Injury frequency increased dramatically on the untreated islet (8.3–100%) while remaining the same on the treated islet (27–38%). Severely injured chicks (i.e., chicks that lost >20% of tissue on their feet) weighed significantly less than uninjured chicks and did not fledge. It is unclear if the chicks were being preyed upon or stung in defense of nearby ant colonies. Radical changes in invasive ant populations have been noted, and booming ant populations could cause short-term, but widespread damage to seabird colonies. The negative effects of invasive ants on seabirds may be difficult to detect, and therefore unknown or underestimated throughout the world where the two groups overlap.  相似文献

The UFL-AG-286 cell line, established from embryonic tissue of the lepidopteran insect Anticarsia gemmatalis, has been identified as a good candidate to be used as a cellular substrate in the development of a process for in vitro production of the Anticarsia gemmatalis multicapsid nucleopolyhedrovirus, a baculovirus widely used as bioinsecticide. In order to characterize the technological properties of this cell line and evaluate its feasibility to use it for the large-scale production of Anticarsia gemmatalis multicapsid nucleopolyhedrovirus, UFL-AG-286 cells were adapted to grow as agitated suspension cultures in spinner-flasks. Batch suspension cultures of adapted cells in serum-supplemented TC-100 medium grew with a doubling time of about 29 h and reached a maximum cell density higher than 3.5 × 106 viable cells ml−1. At the end of the growth period glucose was completely depleted from the culture medium, but l-lactate was not produced. Amino acids, with the exception of glutamine, were only negligibly consumed or produced. In contrast to other insect cell lines, UFL-AG-286 cells appeared to be unable to synthesize alanine as a metabolic way to dispose the by-product ammonia. The synchronous infection of suspension cultures with Anticarsia gemmatalis multicapsid nucleopolyhedrovirus in the early to medium exponential growth phase yielded high amounts of both viral progenies per cell and reduced the specific demands of UFL-AG-286 cells for the main nutrients.  相似文献

HTLV-I causes T-cell leukemia and tropical spastic paraparesis (TSP) in a minority of infected people, whereas the majority remain healthy. The virus differs little in sequence between isolates but has been shown to have a quasispecies structure. Using the Nei and Gojobori algorithm, we have shown that the proportion of nonsynonymous to synonymous changes in HTLV-I proviral tax gene sequences from healthy seropositive subjects (Dn/Ds= 0.9 to 1.3) is significantly higher than those from TSP patients (Dn/Ds= 0.3 to 0.6). Here we show that the distinction between healthy seropositives and TSP patients can only be seen with proviral tax sequences, but not with cDNA, the amino-terminal or carboxy-terminal half of tax, or the rex gene. The Dn/Ds ratio of proviral tax sequences was used to analyze two TSP patients with atypical features and to investigate the influence of cytotoxic T cells (CTL) on the viral quasispecies. We examined the toxicity of a fungicide, sulfur, to the egg parasitoid Anagrus erythroneurae (Hymenoptera: Mymaridae) Trjapitsyn and Chiapini and the vineyard leafhopper pest Erythroneura elegantula Osborn (Homoptera: Cicadellidae) and tested whether or not the use of sulfur in the field affects biological control of E. elegantula. Using field cage bioassays, we demonstrated that sulfur is toxic to adult A. erythroneurae parasitoids, but not toxic to adult E. elegantula leafhoppers. We nonetheless found in a field experiment that sulfur produced no changes in rates of parasitism or E. elegantula egg density, and generated only a very small increase in the density of E. elegantula nymphs. These results suggest that sulfur, although toxic to A. erythroneurae, is not highly disruptive of E. elegantula biological control in vineyards. Our results suggest that simple bioassays of acute toxicity may not accurately predict the impact of agricultural chemicals on biological control.  相似文献

长链非编码RNA(long non-coding RNA,lncRNA)是包括细胞增殖在内的许多细胞过程的重要调节因子。虽然已有研究表明多种lncRNA在造血系统恶性肿瘤的发生发展过程中发挥重要作用,但是缺少一个更全面和无偏倚的方法同时研究多个lncRNA中对白血病细胞系产生功能性影响的lncRNA。在此,我们利用短发夹RNA(short hairpin RNA,shRNA)文库结合高通量测序的方法,筛选对白血病细胞系增殖有影响的lncRNA,确定了74个候选lncRNAs。从中选取lncRNA C20orf204-203作为验证研究对象,发现C20orf204-203在K562和THP-1细胞系中均定位于胞质,敲降C20orf204-203的K562和THP-1细胞系增殖能力降低,早期凋亡细胞增加,BAD基因在mRNA水平上表达量增加,TP53、BCL2蛋白表达量下降,在THP-1细胞系中Caspase 3蛋白表达量减少,激活型Caspase 3蛋白表达量上升,但是二者变化在两种细胞系中不一致。结果表明,在白血病细胞系中敲降lncRNA C20orf204-203会使细胞增殖能力降低。但其在不同细胞系作用途径和机制可能存在差异。这一研究表明了利用shRNA文库结合高通量测序大规模研究lncRNA在白血病细胞系中发挥作用的可行性。  相似文献

To determine how nutritional indices for insects fed leaves are affected by the experimental conditions and the physiology of the plant material, we used larvae of the buckmoth, Hemileuca lucina Hy. Ed. (Saturniidae) and their hostplant Spiraea latifolia Ait. Bork (Rosaceae). Under experimental conditions identical to those used to determine larval nutritional indices, we found that the age of leaves (new versus mature) significantly affected their metabolism and water loss, but simulated herbivory did not directly affect leaf metabolism. Over a 6-day test, nitrogen concentration showed an initial increase followed by a gradual decline, and was higher in new leaves compared to mature leaves. New leaves increased in protein concentration and then gradually returned to the initial level, whereas mature leaves changed little over the 6-day test. These changes in percent nitrogen and protein may largely reflect the disproportional changes in non-nitrogenous materials. Solitary and grouped larvae had similar growth rates on new leaves, but they differed on mature leaves. Deliberate manipulation of larvae during the course of an experiment significantly reduced relative growth rates by increasing duration of the stadium rather than by decreasing biomass gained. Two methods of estimating larval gut contents at mid-stadium were compared: weight of frass produced and weight of digestive tract and contents. After the end of the 4-day test period used to determine nutritional indices, the digestive tracts with food accounted for 10.8% of the larval dry weight. Larval frass produced in 24 h after the end of the test period comprised 9.3% of the larval dry weight. Correction factors for plant metabolism changed nutritional indices by 1 to 8%, while those for larval gut contents altered indices by 2 to 15%.  相似文献

Tailed bacteriophages specific for Gram‐negative bacteria encounter lipopolysaccharide (LPS) during the first infection steps. Yet, it is not well understood how biochemistry of these initial interactions relates to subsequent events that orchestrate phage adsorption and tail rearrangements to initiate cell entry. For many phages, long O‐antigen chains found on the LPS of smooth bacterial strains serve as essential receptor recognized by their tailspike proteins (TSP). Many TSP are depolymerases and O‐antigen cleavage was described as necessary step for subsequent orientation towards a secondary receptor. However, O‐antigen specific host attachment must not always come along with O‐antigen degradation. In this issue of Molecular Microbiology Prokhorov et al. report that coliphage G7C carries a TSP that deacetylates O‐antigen but does not degrade it, whereas rough strains or strains lacking O‐antigen acetylation remain unaffected. Bacteriophage G7C specifically functionalizes its tail by attaching the deacetylase TSP directly to a second TSP that is nonfunctional on the host's O‐antigen. This challenges the view that bacteriophages use their TSP only to clear their way to a secondary receptor. Rather, O‐antigen specific phages may employ enzymatically active TSP as a tool for irreversible LPS membrane binding to initiate subsequent infection steps.  相似文献

Insect cell lines in culture are used for a variety of studies. In this laboratory imaginal disc cell lines have been established from primary cultures from third instar larvae, and used for a number of experiments. The effect of ageing on the morphology and physiology of Drosophila cell lines has received very little attention, although problems of genotypic or phenotypic changes in cell lines with age are recognized in other areas of animal cell culture. We tested our cell line CI8+ for any difference in growth, morphology and response to 20-hydroxyecdysone (20HE) at different ages (passage numbers). The cells were found to multiply faster, adhere less firmly to the substrate and to lose the tendency to aggregate at higher passages. The response to 20HE in terms of cell numbers and induction of β-galactosidase was similar at all passage numbers but morphological changes in hormone-treated cells were less obvious in the higher passages. Cell lines are likely to vary in the extent of ageing effects but workers are advised to be aware of the possibilities. We suggest the effects of age on cell lines should be established, and passage numbers noted in experimental reports.  相似文献

Summary With the idea to develop a selection system for asymmetric somatic hybrids between oilseed rape (Brassica napus) and black mustard (B. nigra), the marker gene hygromycin resistance was introduced in this last species by protoplast transformation with the disarmed Agrobacterium tumefaciens strain C58 pGV 3850 HPT. The B. nigra lines used for transformation had been previously selected for resistance to two important rape pathogens (Phoma lingam, Plasmodiophora brassicae). Asymmetric somatic hybrids were obtained through fusion of X-ray irradiated (mitotically inactivated) B. nigra protoplasts from transformed lines as donor with intact protoplasts of B. napus, using the hygromycin resistance as selection marker for fusion products. The somatic hybrids hitherto obtained expressed both hygromycin phosphotransferase and nopaline synthase genes. Previous experience with other plant species had demonstrated that besides the T-DNA, other genes of the donor genome can be co-transferred. In this way, the produced hybrids constitute a valuable material for studying the possibility to transfer agronomically relevant characters — in our case, diseases resistances — through asymmetric protoplast fusion.  相似文献
