The effects of dietary Ergosan on the growth performance and mucosal immunity in rainbow trout skin were investigated. 60 rainbow trout (100-110 g) were randomly assigned to 2 groups in triplicates and fed one of the experimental diet formulated with 5 g kg−1 Ergosan or control diet for 50 days. Results showed that on the 45th day of feeding trial, Ergosan supplementation significantly enhanced the growth performance compared to control group. Various enzyme activities, namely lysozyme, protease, alkaline phosphatase and esterase in treatment group were also enhanced on the 45th and 50th day. Skin mucus in Ergosan-fed fish showed the agglutination of erythrocytes while in control group, no visible agglutination was shown. In addition, skin mucus in treatment group showed strong antibacterial activity against Yersinia ruckeri. In conclusion, the major immune components of rainbow trout mucus that are involved in the non-specific immunity were enhanced by administration of Ergosan in 5 g kg−1.  相似文献

Hatchlings cuttlefish were reared in the laboratory from hatching until 30 days old, fed with live shrimp, frozen shrimp or fish oil-enriched frozen shrimp. Survival of cuttlefish fed with oil-enriched frozen shrimp was better than in animals receiving live shrimp. However, there was no difference with cuttlefish fed with frozen shrimp, even if survival of those receiving oil-enriched frozen shrimp was always higher all along the experiment. Lower survival in animals fed with live shrimp represented the problem of using such food and confirms the necessity to elaborate an artificial food. Utilization of artemia was detrimental to growth and induced low values of instantaneous growth rate (IGR) and conversion rate even after feeding cuttlefish with shrimp. Nevertheless, growth parameters evolutions generally corresponded to those observed by other researchers. The profile noticed at the end of the experiment is typically observed when cuttlefish acquire their adult digestive system. Main differences were observed between groups fed with live shrimp or oil-enriched frozen shrimp. Enrichment did not induce same growth as in cuttlefish receiving live prey. However, at 20 and 25 days after hatching (DAH), in cuttlefish fed with oil-enriched frozen shrimp, ration was lower for the same growth than in other groups. These data showed capacity of juvenile cuttlefish to adjust their digestive enzyme activities according to the diet and the stage of development. Indeed, chymotrypsin was strongly influenced by enrichment, while other enzymes showed difference between live and frozen preys. Trypsin exhibited regulation by diet after 20 DAH. Freezing seemed to delay development as acid phosphatases, characteristic of first stages of cuttlefish, had lower activity in cuttlefish fed with live shrimp at 10 DAH. Moreover, influence of the stage of development was strong as activities between 20 and 30 DAH were different in all groups. This was in relation with evolution of the digestive system. These data illustrated the difficulty to elaborate optimal diet as digestive system evolves.  相似文献

Pacific white shrimp (Litopenaeus vannamei) were injected with Taura syndrome virus (TSV) to assess shrimp immune responses and survival. TSV-infected shrimp suffered high mortality, but mock-infected and untreated shrimp experienced no mortality. Moribund shrimp were a pale, reddish colour and were lethargic and soft-shelled. Their haemolymph was clear red and coagulated poorly. In TSV-infected shrimp, the total haemocyte count (THC), hyalinocyte and granulocyte counts, and total plasma protein decreased significantly to 21%, 24%, 17% and 56% of untreated control values, respectively. Haemocyanin decreased to 67%, and clottable proteins to 80% of control values (P< 0.01). Copper and calcium ions, haemocytic transglutaminase (TGase) activity and plasma growth inhibitory activity against Vibrio harveyi also decreased significantly. Generation of intrahaemocytic superoxide anion, O(-2), in TSV-infected shrimp was significantly greater (P< 0.05) than in both control groups, no matter whether glucan stimulated or unstimulated. But the relative increase of intrahaemocytic O(-2) generation in TSV-infected shrimp response to glucan stimulation was lower in both controls. Plasma phenoloxidase (PO) activity increased significantly in TSV-infected shrimp. The plasma bacterial agglutinin titre against E. coli and V. harveyi, growth inhibition of E. coli and the concentration of magnesium ions in TSV-infected shrimp did not change significantly. In conclusion, ten of thirteen haemolymph parameters changed significantly during the host-TSV interaction. These parameters might be valuable references of shrimp health status.  相似文献

Immunostimulants are valuable for control of shrimp diseases and the immunostimulatory effects of some polysaccharide additives for shrimp have been reported. In this study, the Sargassum fusiforme polysaccharide extract (SFPSE) was assessed as a feed additive when supplemented in the diet (0%, 0.5%, 1.0%, and 2.0%) for juvenile shrimp, Fenneropenaeus chinensis, in order to study the effects of SFPSE on vibriosis resistance and immune activity. Shrimp were cultured in the same pond with cages. The body weight, survival, the cumulative mortality after injection with Vibrio harveyi (30 microl V. harveyi suspension at 9.3 x 10(7) CFU ml(-1) per shrimp), the total haemocyte counts (THCs), the protein concentration and the phenoloxidase (PO) activity in supernatant of haemolymph, the lysozyme (LSZ) and superoxide dismutase (SOD) activity in muscle of the shrimp were assayed after 14 days feeding period. The results indicated that shrimp survival under the stress of V. harveyi was affected by the dietary SFPSE. The shrimp treated with 1.0% and 0.5% SFPSE displayed significantly lower cumulative mortalities after being injected with V. harveyi suspension 24 and 30 h later, respectively, compared with that of the control. However, cumulative mortality of 2.0% SFPSE treatment was not significantly different from that of the control. There was no significant difference of cumulative mortality between 0.5% and 1.0% SFPSE treatment groups. The immune activities of the shrimp also were affected by dosage of dietary SFPSE. The THCs of the shrimp rose with increasing SFPSE dosage. The protein concentration and PO activity in supernatant of haemolymph as well as muscular LSZ activity first rose then dropped with increasing SFPSE dosage. The protein concentration in supernatant of haemolymph appeared a maximum of 167.46 mg ml(-1) in 1.0% SFPSE treatment. The PO activity and LSZ activity reached the peaks as 13.20 U and 3.21 U mgprot(-1) in 0.5% SFPSE treatment, respectively. SOD activity of the shrimp was not significantly affected by dietary SFPSE. It is therefore suggested that oral administration of SFPSE at an optimal level of 0.5% and 1.0% for 14 days effectively improved vibriosis resistance and enhanced immune activity of the shrimp in general.  相似文献

We studied the effect of two probiotic Bacillus subtilis strains on the growth performance, digestive enzyme activity, immune gene expression and disease resistance of juvenile white shrimp (Litopenaeus vannamei). A mixture of two probiotic strains, L10 and G1 in equal proportions, was administered at two different doses 10(5) (BM5) and 10(8)?(BM8)?CFU?g(-1) feed to shrimp for eight weeks. In comparison to untreated control group, final weight, weight gain and digestive enzyme activity were significantly greater in shrimp fed BM5 and BM8 diets. Significant differences for specific growth rate (SGR) and survival were recorded in shrimp fed BM8 diet as compared with the control; however, no significant differences were recorded for food conversion ratio (FCR) among all the experimental groups. Eight weeks after the start of the feeding period, shrimp were challenged with Vibrio harveyi. Statistical analysis revealed significant differences in shrimp survival between probiotic and control groups. Cumulative mortality of the control group was 63.3%, whereas cumulative mortality of the shrimp that had been given probiotics was 20.0% with BM8 and 33.3% with BM5. Subsequently, real-time PCR was employed to determine the mRNA levels of prophenoloxidase (proPO), peroxinectin (PE), lipopolysaccharide- and β-1,3-glucan-binding protein (LGBP) and serine protein (SP). The expression of all immune-related genes studied was significantly up-regulated (P?相似文献

This study was aimed at determining if the increase of body astaxanthin content through dietary supplementation in tiger prawn Penaeus monodon juvenile could enhance its antioxidant defense capability and resistance to ammonia stress. Haemolymph total antioxidant status (TAS) and superoxide dismutase (SOD) were chosen as parameters of shrimp antioxidant capacity. Resistance to chemical stress was evaluated by shrimp survival rate, and haemolymph aspartate aminotransferase (AST) and alanine aminotransferase (ALT). P. monodon 5-day postlarvae were fed diets supplemented with 0 and 71.5 mg kg−1 astaxanthin for 8 weeks. Shrimps were then subjected to 72-h exposure of ammonia at 0.02, 0.2, 2 and 20 mg l−1. The survival rates of the astaxanthin-fed (AX) shrimp were higher than those of the control shrimp under all levels of ammonia except 20 mg l−1, showing that the shrimp's resistance to ammonia stress had been improved by dietary astaxanthin. AX shrimp had higher TAS than control shrimp at ammonia levels higher than 0.02 mg l−1 and lower SOD at all ammonia levels suggested that antioxidation capability had been greatly enhanced. AST in AX shrimp was lower than that in control shrimp under all levels of ammonia stress. ALT in AX shrimp was either lower than or equal to that in control shrimp under various levels of ammonia. Both AST and ALT reflected that shrimp hepatopancreatic function had been improved by dietary astaxanthin. Astaxanthin can become essential for P. monodon when the animal is under ammonia stress.  相似文献

We are reporting results directed to explain the relation between carbohydrates (CHO), protein metabolism, and the energetic balance of Litopenaeus vannamei juveniles. The interaction of dietary CHO and salinity was measured to try to understand the relation between osmotic control and metabolism, both from a biochemical and energetic point of view. Two experiments were done. In the first experiment, shrimp were fed with 0%, 5%, 33%, and 61% CHO and maintained at 15‰ and 40‰ salinity. Glucose, lactate protein, hemocyanin, ammonia concentration, and osmotic pressure were measured in blood. Digestive gland glycogen (DGG) was measured also. In the second experiment, shrimp were fed with 0% and 38% dietary CHO and maintained at 15‰ and 40‰ salinity. From that shrimp, absorbed energy (Abs) was calculated as: Abs=respiration (R)+ammonia excretion (U) and production (P); assimilated energy (As) was calculated as the product of R×P. Osmotic pressure, hemocyanin, protein, lactate, and blood ammonia increased with the reduction in dietary CHO. In contrast, an increase in blood glucose was observed with an increase in dietary CHO. Digestive gland glycogen (DGG) increased following a saturation curve with a DGG maximum at 33% dietary CHO. Blood metabolites of fasting and feeding shrimp showed the same behavior. Energy balance results showed that shrimp maintained in low salinity and fed without CHO waste more energy in U production than for shrimp maintained in high salinity and fed with high CHO levels. Notwithstanding, the production efficiency was higher in shrimp fed without CHO than that observed in shrimp fed with high CHO independent of salinity. A scheme trying to integrate the relation between CHO and protein metabolism and the way in which both are modulated by salinity is presented. From published and present results, there are two factors that apparently control the use of high dietary CHO levels; α-amylase enzyme-dietary CHO level capacity and glycogen saturation in DG. Production of glucose is limited in shrimp because of saturation of α-amylase when shrimp are fed with diets above 33% CHO. This is the first control point of starch metabolism. The digestive gland is saturated with glycogen in shrimp fed with dietary CHO levels >33%. This is apparently the second control point of CHO metabolism that limits growth rate in such conditions. The high metabolic cost related to high CHO diets could explain why shrimp are well adapted to use protein as a source of energy.  相似文献

Effects of dietary vitamin C (l-ascorbyl-2-monophosphate-Mg, C2MP-Mg) on growth, tissue copper (Cu) accumulation, and haemocyte superoxide anion production of juvenile grass shrimp, Penaeus monodon, fed with either adequate or high (8 x adequate) dietary Cu were studied. Three experimental diets were used: basal diet supplemented with adequate levels of both C2MP-Mg (40 mg kg diet(-1)) and Cu (20mg kg diet(-1)) (NC-NCu); basal diet supplemented with adequate C2MP-Mg and high Cu (8 x adequate) (NC-HCu); and basal diet supplemented with high C2MP-Mg (5 x adequate) and high Cu (HC-HCu). These were each fed to triplicate groups of shrimp (mean initial weight: 0.29+/-0.01 g) for 8 weeks. Highest (P< 0.01) weight gain, feed efficiency (FE) and protein efficiency ratio (PER) were observed in shrimp fed NC-NCu diet, intermediate in shrimp fed HC-HCu diet, and lowest in shrimp fed NC-HCu diet. Cu concentrations in hepatopancreas, muscle and haemolymph were highest in shrimp fed NC-HCu diet, followed by shrimp fed HC-HCu diet, and lowest for shrimp fed NC-NCu diet. Survival, total haemocyte count (THC) and intracellular superoxide anion (O-2) production were higher in shrimp fed NC-NCu diet than shrimp fed NC-HCu diet, whereas hepatosomatic index (HSI) was higher in shrimp fed NC-HCu diet than shrimp fed NC-NCu diet. However, all these parameters were similar in shrimp fed NC-NCu diet and shrimp fed HC-HCu diet. These data suggest that increase of dietary vitamin C improved haemocyte respiratory burst response and growth and prevented tissue Cu accumulation in P. monodon fed with high dietary Cu.

Water temperature changes (higher and lower than 24 degrees C) were shown to have a significant effect on dopamine (DA) concentration, haemocyte count and the proPO system in the white shrimp Litopenaeus vannamei. No significant difference in any of the parameters was observed in the control group. DA concentration in haemolymph in the experimental groups increased to a peak value at 0.5 days; meanwhile serine protease (SP) activity and proteinase inhibitor (PI) activity decreased. Total haemocyte count (THC), differential haemocyte count (DHC) and PO activity were lowest at 1 day. All defence parameters became stable after 1-3 days, while the total haemocyte and large granular cell count stabilized after 6 days. After these stabilized, there was no significant difference in DA concentration and PI activity between the control and experimental groups, as was the case for the THC, DHC, PO and SP activities of shrimp held at higher temperatures. However these latter four parameters in the lower temperature groups were distinctly lower than the control group. alpha(2)-Macroglobulin activity in the experimental groups increased to a peak value after 1 day compared with the control and then stabilized after 6 days when the activity levels in higher temperature groups were higher than the control, while the lower temperature groups had no significant difference from the control. It was therefore concluded that water temperature changes modulated the immune system of L. vannamei.  相似文献

The control of crop emptying in foraging honeybees was investigated in individuals trained to collect defined amounts of sugar solutions. Following feeding, they were dissected after fixed periods of time in order to measure crop content and haemolymph sugar titers. Between feeding and dissection, the metabolic rate of every investigated forager was measured using open-flow respirometry, so as to assess the effects of both food quality (concentration, molarity and viscosity of the fed sugar solution) and food quantity on the transport rate through the proventriculus. The sugar transport rate through the proventriculus was observed to be mainly dependent on the metabolic expenditure of the individual. Bee foragers were able to precisely adjust the sugar transport rate to their metabolic rates, but under certain conditions, an excess of sugars was transported through the proventriculus, more than needed to cover the bee’s energetic demands. This excess depended on the nutritive value and quantity of the fed sugar solution, and on the time after feeding. It did not depend on the metabolic rate of the bee, the molarity, or the viscosity of the fed sugar solution. As long as the bees did not exhaust their crop contents, the haemolymph sugar titers were unaffected by this excess amount transported, by the time after feeding, the concentration and the viscosity of the fed sugar solution. For all feeding conditions assayed, the haemolymph trehalose titer remained constant, while the titers of other haemolymph sugars varied. It is suggested that the trehalose concentration in the haemolymph is regulated in honeybees, and that it represents the controlled variable in the feedback loop responsible for the transport rate through the proventriculus.  相似文献

The present study investigated the immunomodulatory activity of Ergosan, an algal extract containing alginic acid, and Macrogard, a yeast extract containing beta-glucans, on innate and specific immunity in sea bass (Dicentrarchus labrax). Four cycles of experimental feeding using normal fish feed formulation (control group) supplemented with Ergosan (0.5%) or Macrogard (0.1%) were performed at 60-day intervals (15 days of treatment+45 days of suspension). Serum complement, lysozyme, total proteins and heat shock protein (HSP) concentrations were measured at 15, 30 and 45 days from the end of the first 15-day feeding cycle (short term) and 45 days after the end of each feeding cycle over a 35-week period (long term). The percentage of B- and T-lymphocytes in peripheral blood leucocytes and gut were measured over long-term trial. Significant elevation (P < 0.05) in serum complement activity occurred in sea bass fed with alginic acid and glucans, at 15 days from the end of first cycle of treatment. Significant elevation (P < 0.05) in serum lysozyme, gill and liver HSP concentration were observed in the same experimental groups at 30 days from the end of treatment, whereas a significant increase (P < 0.05) of complement activity was only observed in fish that received an Ergosan diet. At 45 days from the end of treatment, complement, lysozyme and HSP concentration did not differ among groups. Over the long-term period, no significant differences were observed in innate and specific immune parameters, survival, growth performances and conversion index in treated and control fish. A dramatic decrease of both innate and acquired immune parameters was observed during the winter season in all groups, followed by a partial recovery when water temperature increased. Reduction in complement and lysozyme activities was significatively correlated (p < 0.01) to water temperature variation. The results suggested the potential of alginic acid and beta-glucans to activate some innate immune responses in sea bass, and particularly under conditions of immunodepression related to environmental stress.  相似文献

An 8-week feeding trial was conducted to investigate the effects of dietary mannan oligosaccharide (MOS) on growth performance, gut morphology, and NH(3) stress tolerance of Pacific white shrimp Litopenaeus vannamei. Juvenile Pacific white shrimp (1080 individuals with initial weight of 2.52?±?0.01?g) were fed either control diet without MOS or one of five dietary MOS (1.0, 2.0, 4.0, 6.0 and 8.0?g?kg(-1)) diets. After the 8-week feeding trial, growth parameters, immune parameters, intestinal microvilli length and resistance against NH(3) stress were assessed. Weight gain (WG) and specific growth rate (SGR) were significantly higher (P??0.05) in all experimental groups. Compared with control group, TEM analysis revealed that 2.0, 4.0, 6.0 and 8.0?g?kg(-1) MOS supplementation could significantly increase (P?相似文献

The mannose-specific snowdrop lectin (Galanthus nivalis agglutinin: GNA), when fed to insects, binds to the gut epithelium and passes into the haemolymph. The ability of GNA to act as a carrier protein to deliver an insecticidal spider venom neurotoxin (Segestria florentina toxin 1: SFI1) to the haemolymph of lepidopteran larvae was investigated. Constructs encoding SFI1 and an SFI1/GNA fusion protein were expressed in Pichia pastoris. The insecticidal activity of purified recombinant proteins on injection was found to be comparable to published values for SfI1 purified from spider venom [Toxicon 40 (2002) 125]. Whereas neither GNA nor SFI1 alone showed acute toxicity when fed to larvae of tomato moth (Lacanobia oleracea), feeding SFI1/GNA fusion at 2.5% of dietary proteins was insecticidal to first stadium larvae, causing 100% mortality after 6 days. The protein also showed a significant, dose dependent, toxicity towards fourth and fifth stadium larvae, with growth reduced by up to approximately 90% over a 4-day assay period compared to controls. Delivery of intact SFI1/GNA to the haemolymph in these insects was shown by western blotting; haemolymph samples from fusion-fed larvae contained a GNA-immunoreactive protein of the same molecular weight as the SFI1/GNA fusion. SFI1/GNA and similar fusion proteins offer a novel and effective approach for delivering haemolymph active toxins by oral administration, which could be used in crop protection by expression in transgenic plants.  相似文献

Oral administration of polysaccharide gel (PG) in shrimp diets revealed immunostimulating potential and disease resistance in Penaeus monodon (black tiger shrimp). PG from the fruit-rind of Durio zibethinus has been characterized to be a pectic polysaccharide with immunomodulating and antibacterial activities. PG inhibited growth of the shrimp bacterial pathogen, Vibrio harveyi 1526, by agar diffusion and broth microdilution tests. Clear inhibition zones on agar plates were observed at the lowest PG concentration of 3.1 mg/ml, where minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) values for PG were 6.3 and 12.5 mg/ml, respectively. Each group of juvenile shrimps, initial mean body weight 0.29 ± 0.04 g, was housed in a closed-recirculating treated water system and was fed with PG-supplemented diets containing 1, 2 and 3% PG or shrimp basal diet in the control group for 8 and 12 weeks. PG-supplemented diets did not contribute to the overall growth of black tiger shrimp. The immune response was evaluated by analysis of prophenoloxidase activity and total hemocyte count in the shrimp fed PG-supplemented diets for 12 weeks. Prophenoloxidase activity in shrimp fed the 1, 2 and 3% PG-supplemented diet and total hemocyte count in shrimp fed the 1 and 2% PG-supplemented diet were higher (P < 0.05) than those of the control group. The percent survival was higher in groups fed the 1–3% PG-supplemented diets in challenge tests with either white spot syndrome virus (WSSV) or the bacterium V. harveyi 1526 than that of the control group. Relative percent survival (RPS) values in groups fed the 2% PG-supplemented diet showed the highest RPS value for disease resistance of 100% (at Day 6) and 36% (at Day 4) in treated shrimp against viral and bacterial infection, respectively. Mortality of PG-supplemented diets in treated shrimps against WSSV infection was also found to be much lower (P < 0.05) than that of the control group.  相似文献

Penaeus japonicus (15.7 ± 1.4 g) were exposed individually in 30 ppt seawater to 0.01 (control), 5, 10, 20 and 50 mg/l nitrite-N for 24 hr. Haemolymph ammonia, urea, nitrite and whole shrimp ammonia-N excretion and nitrite-N uptake were then determined. Ammonia excretion of P. japonicus increased with increased ambient nitrite, and with a concomitant decrease of haemolymph ammonia, as occurring increased concentrations of nitrite. Concentrations of nitrite-N and urea-N in the haemolymph of shrimp increased with increased ambient nitrite-N. However, no urea-N excretion was observed for shrimp exposed to any nitrite treatments.  相似文献

Haemolymph ammonia-N of Penaeus japonicus (15.6 ± 2.17 g) increased with increased ambient ammonia-N, as they were exposed individually in 30 ppt seawater to 0.003 (control), 0.367, 0.731, 1.439 and 3.665 mmol/1 ammonia-N after 24 hr. Ammonia-N excretion was inhibited and net ammonia-N uptake occurred, as shrimps were exposed to 0.367 mmol/1 ammonia-N or greater. Haemocyanin and protein levels in the haemolymph of shrimp decreased, whereas free amino acid levels increased with increased ambient ammonia-N in the range of 0.003–1.439 mmol/1 ammonia-N. Exposure of shrimp to ambient ammonia-N at 1.439 mmol/1 caused accumulation of haemolymph ammonia and urea, and caused catabolism of haemocyanin and protein to free amino acids. Urea, taurine and glutamine are the major organic constituents in the haemolymph of shrimp under ammonia stress.  相似文献

In some systems, the identity of a prey species' dominant predator(s) may not be constant over time. In cases in which a prey species exhibits different responses to various predator species, such changes in predator identity may have population-wide consequences. Our goals were to determine (1) whether mortality of and refuge use by the grass shrimp, Palaemonetes pugio, were predator-specific, and (2) how effects of prey size and habitat interacted with predator type. Striped bass (Morone saxatilis) exerted twice as much predation pressure as mummichog (Fundulus heteroclitus), although not equally as great on large (female) and small (male) shrimp. Mummichog, which fed preferentially on large shrimp, forced a partitioning of habitat between the two shrimp size classes. In contrast, large and small shrimp occupied similar habitats when subjected to striped bass, which fed on both size classes equally. Refuge use of grass shrimp depended on predator type. In the presence of mummichog, which occupied shallower depths in the water column than striped bass, shrimp stayed deep and close to structural habitat. Striped bass, which were deeper, caused shrimp to move high in the water column away from structural habitat. When both predators were present, shrimp distribution was similar to that when only striped bass were present, striped bass predation rate was enhanced, and overall mortality was higher than with either predator alone. Results suggest that at times when mummichogs are the dominant predators, large (female) shrimp experience higher predation than small (male) shrimp and are physically separated from their potential mates. When striped bass are more abundant, male and female shrimp may share a similar, shallow, less structure-oriented distribution and be subjected to higher mortality. When both predators are present, mortality rates may be higher still. This predator-, size-, and habitat-specificity of grass shrimp behavior suggests significant population and distribution consequences of fluctuating predator guilds and fluctuating cover of structural habitats in the field.  相似文献

Two hundred and two strains of lactic acid bacteria (LAB) isolated from digestive tracts of cultivated and wild adult shrimp, including Litopenaeus vannamei, Metapenaeus brevicornis and Penaeus merguiensis were selected based on their antibacterial activity against Vibrio harveyi. LAB strain of MRO3.12 exhibiting highest reduction of V. harveyi was identified as Lactobacillus plantarum MRO3.12 based on the nucleotide sequence of its 16S rDNA, which showed 99% (780/786 bp) homology to L. plantarum strain L5 (GenBank accession number DQ 239698.1). Co-cultivation of V. harveyi and L. plantarum MRO3.12 showed complete reduction of V. harveyi at 24 h under aerobic and anaerobic conditions, whereas L. plantarum increased from 5.29 to 9.47 log CFU ml−1. After 6-week feeding trial with L. plantarum supplemented diet, white shrimp (L. vannamei) exhibited significant differences (p < 0.05) in relative growth rate (% RGR), feed conversion ratio (FCR) and survival compared to the control group fed with non-supplemented diet. LAB-fed group showed 98.89% survival, whereas only 68.89% survival was observed in the control group. LAB from the digestive tract of probiotic-fed shrimp showed higher level of 5.0 ± 0.14 log CFU/g than the non-supplemented ones (3.34 ± 0.21 log CFU/g). However, total bacterial and non-fermenting vibrios counts decreased in shrimps fed on L. plantarum. Ten days after infection with V. harveyi (5.3-5.5 log CFU ml−1), significant survival (p < 0.05) of 77% was observed in LAB supplemented shrimp, while only 67% survival was observed in the control.  相似文献

A monoclonal antibody specific to yellow head virus (YHV) was produced from a mouse immunized with gill extracts prepared from laboratory-reared Penaeus monodon dually infected with YHV and white spot syndrome virus (WSSV). One clone designated V3-2B specifically bound to native and SDS-treated viral specific antigens. Immunocytochemical studies of infected gills revealed viral specific immunoreactivities in the cytoplasm of gill tissue and in haemocytes. No antibody binding was observed in gills from non-infected shrimp. In addition, immunocytochemical examination of tissues from shrimp experimentally infected with YHV gave a positive reaction, while tissues from uninfected control shrimp or shrimp experimentally infected with WSSV did not. Western blot analysis indicated that the antibody reacted with a protein of approximately 135 kD that was present only in shrimp infected with YHV. In dot-blot indirect immunoperoxidase assays, the antibody was able to detect viral associated antigen in diluted haemolymph up to 1:50 dilution and in an ammonium sulfate precipitate of haemolymph up to 1:1000 dilution. The results suggested that this antibody might be useful for development of effective diagnostic techniques for both heavy and mild YHV infections in shrimp.  相似文献
