Summary Segregation at one of the loci controlling tiller-base pigmentation was studied to determine the mode of inheritance in tetraploid hybrids between Lolium perenne and L. multiflorum. The results could be explained by tetrasomic inheritance and thus did not support previous reports of a degree of preferential chromosome pairing in this material. However, double reduction and aneuploidy may to some extent have masked any tendency to disomic segregation brought about by preferential pairing. Moreover, there was significant heterogeneity between families in the segregation ratios which may indicate genetically controlled differences in pairing behaviour. The results are related to previous cytological and genetic studies.  相似文献

The effect of B chromosomes on chromosome pairing at meiosis was investigated in the species hybrid Lolium temulentum x L. perenne at both the diploid and tetraploid level. The presence of B chromosomes drastically reduced association of homoeologous chromosomes in both the diploids and tetraploids. This was evident from the high frequency of univalents recorded in PMC's of diploid hybrids with B's and from the predominantly bivalent association of homologous chromosomes in tetraploids of this type. In the absence of B's homoeologous pairing was extensive giving a high frequency of bivalents in the diploids and multivalents as well as bivalents and univalents in the tetraploids.  相似文献

Summary Chromosome association at first meiotic metaphase in tetraploid hybrids between Lolium perenne and L. multiflorum was compared with that in autotetraploid L. perenne. The hybrids were found to have significantly higher levels of bivalent frequency, and lower levels of multivalent and chiasma frequency. A significant increase in multivalent frequency with increasing chiasma formation was found in both groups, but the increase was much less in the hybrids. These differences in chromosome associations between the two groups must therefore reflect differences in chiasma distribution and it is suggested that the results indicate a significant degree of preferential bivalent pairing in the hybrids.  相似文献

Summary Despite an average difference of about 50% in DNA amount, homoeologous chromosomes pair effectively at first metaphase in the diploid interspecific hybrid between Lolium temulentum and Lolium perenne. However, in the presence of accessory B chromosomes and diploidising genes pairing at metaphase I is severely reduced. Reconstruction of serial electron micrographs through pollen mother cell nuclei show that synaptonemal complexes are formed at pachytene between not only homoeologous but also non-homologous chromosome segments resulting in multivalent formation. These associations are largely ineffective in terms of chiasma formation and degenerate at late pachytene. It is highly probable that the pairing determinants exercise their control on chromosome pairing largely by prohibiting the siting of crossovers in homoeologously paired chromosome segments.  相似文献

Comparisons were made between two kinds of tetraploids derived from the hybrid Lolium temulentum x L. perenne. One hybrid behaves like an autotetraploid with multivalents at first metaphase of meiosis in pollen mother cells. The other behaves like an allotetraploid, in which pairing at first metaphase is restricted to bivalents comprised of strictly homologous chromosomes. The diploidisation of the latter form is controlled by determinants located on both the normal, A chromosomes and on supernumary B chromosomes. Reconstruction of synaptonemal complexes and their elements, from serial sections through pollen mother cell nuclei examined under the electron microscope, reveals that at zygotene pairing in both forms results in multivalent formation involving non-homologous as well as homologous chromosomes. The mechanism responsible for the diploidisation is, therefore, not based on a restriction of pairing at early meiosis to homologous chromosomes but on a correction or transformation of the multivalent chromosome associations to bivalents subsequent to zygotene. The transformation is not completed until late pachytene. In the multivalent-forming tetraploid a maximum of four chromosomes are associated at first metaphase. Yet configurations of a higher valency are found at zygotene. There is, therefore, a partial transformation of multivalents even in this autotetraploid form which restricts configurations at metaphase I to homologous and homoeologous chromosomes only. In both hybrids some homologous bivalents are not the product of resolution of multivalents but result from two-by-two pairing from the beginning of zygotene.  相似文献

With a view to eclucidating chromosome relationships between Lolium perenne (Lp), L. multiflorum (Lm) and Festuca pratensis (Fp), chromosome pairing in different diploid (2n=14), auto-allotriploid (2n=3x=21), trispecific (2n=3x=21), amphidiploid (2n=4x=28) and auto-allohexaploid (2n=6x=42) hybrids between them was analysed. At all these levels of ploidy there was very good chiasmate pairing between the chromosomes of the three species and, on the whole, there was little evidence of preferential pairing of the chromosomes of a particular species in the triploid, tetraploid and hexaploid hybrids. A critical test for this also came from the synaptic ability of the chromosomes of the single genome with those of the duplicated genome in the auto-allotriploids which formed predominantly trivalents with 2, 3 or even 4 chiasmata. Moreover, the homology between the Lp and Lm chromosomes seems strong enough to pass the discrimination limits of the B-chromosomes which do not suppress homoeologous pairing in the Lp LmLm triploid and LpLm diploid hybrids. — The triploids having two genomes of a Lolium species and one of F. pratensis had some male and female fertility which suggested genetic compatibility of the parental chromosomes resulting, presumably, in compensation at the gametic level. Also, the occurrence of comparable chiasma frequencies in the auto-allotriploids and trispecific hybrids showed that they were not markedly affected whether two doses of one genome and one of the other or all the three different genomes from the three species were present. From the trend of chromosome pairing in all these hybrids it is concluded that there is little structural differentiation between the chromosomes of the three species, no effective isolation barrier to gene-flow between them, and that they are closely related phylogenetically, having possibly evolved from a common progenitor. Taxonomic revision of the two Lolium species is suggested.  相似文献

Researchers recognized early that chromosome behavior, as other morphological characters, is under genetic control and gave some cytogenetical examples such as the homoeologous chromosome pairing in wheat. In the intergeneric sexual hybrids between cultivated Brassica species and another crucifer Orychophragmus violaceus, the phenomenon of parental genome separation was found under genetic control during mitosis and meiosis. The cytogenetics of these hybrids was species-specific for Brassica parents. The different chromosome behavior of hybrids with three Brassica diploids (B. rapa, B. nigra and B. oleracea) might contribute to the different cytology of hybrids with three tetraploids (B. napus, B. juncea and B. carinata). The finding that genome-specific retention or loss of chromosomes in hybrids of O. violaceus with B. carinata and synthetic Brassica hexaploids (2n=54, AABBCC) is likely related to nucleolar dominance gives new insight into the molecular mechanisms regarding the cytology in these hybrids. It is proposed that the preferential expressions of genes for centromeric proteins from one parent (such as the well presented centromeric histone H3) are related with chromosome stability in wide hybrids and nucleolar dominance is beneficial to the production of centromere-specific proteins of the rRNAs-donor parent and to the stability of its chromosomes.  相似文献

Summary A number of diploid inbred lines of Lolium perenne were treated with colchicine at the early seedling stage to induce chromosome doubling. In each inbred line the colchicine-treated undoubled diploids were kept as controls, as well as the normal untreated diploids. Comparisons of vegetative growth and development, involving the three treatments within each line, revealed that colchicine treatment of seedlings has long-lasting effects upon plant growth and development independent of chromosome doubling, and that for certain characteristics the effects of chromosome doubling are confounded with other effects of the treatment used to produce tetraploids. This colchicine induced variation in the diploids is transmitted through the seed generations in at least one of the inbred lines. The variation appears to be non-random and also shows a strong genotypic component. In so far as the effects of chromosome doubling could be determined, they showed the usual gigas response but were again strongly influenced by genotype.  相似文献

Homoeologous pairing at metaphase-I was analyzed in wild-type, ph2b, and ph1b hybrids of wheat and a low-pairing type of T. longissimum in order to study the effect of ph mutations on the pairing of T. longissimum chromosomes with wheat chromosomes. Chromosomes of both species, and their arms, were identified by C-banding. The three types of hybrids, with low-, intermediate-, and high-pairing levels, respectively, exhibited a very similar pairing pattern which was characterized by the existence of two types, A-D and B-S1, of preferential pairing. These results confirm that the S1 genome of T. longissimum is closely related to the B genome of wheat. The possible use of ph1b and ph2b mutations in the transfer to wheat of genes from related species is discussed.  相似文献

Summary Primary trisomics (2 n + 1 = 15), double trisomics (2 n + 1 + 1 = 16) and aneuploids with 24 to 30 chromosomes, as well as a diploid and tetraploids, were found in the progeny of a hypertriploid (2 n = 22) plant of perennial ryegrass, Lolium perenne L. Trisomics and double trisomics differed in their mean chromosome association, chiasma number and spike morphology. A few aneuploids and tetraploids had reciprocal translocations. The diploid, primary trisomics and tetraploids were more fertile than the double trisomics and aneuploids. Most trisomics and aneuploids were probably produced through female transmission. One double trisomic had a high univalent number, a low chiasma number and loose chromosome coiling. Both the extra chromosomes carried secondary constrictions. The gene for desynapsis might be located on one of these chromosomes.  相似文献

Procedures for the transfer of genes for drought resistance from Festuca glaucescens (2n=4x=28) into Lolium multiflorum (2n=2x=14) are described. Following the initial hybridisation of a synthetic autotetraploid of L. multiflorum (2n=4x=28) with F. glaucescens, the F1 hybrid was backcrossed twice onto diploid L. multiflorum (2n=2x=14) to produce a diploid Lolium genotype with a single F. glaucescens introgression located distally on the nucleolar organiser region arm of chromosome 3. The transmission of F. glaucescens-derived amplified fragment length polymorphisms and a sequence-tagged-site (STS) marker was monitored throughout the breeding programme. Those genotypes of a mapping population of backcross 3 that survived combined severe drought and heat stress all contained the F. glaucescens-derived markers. The STS marker provided a prototype for a PCR-based system for high-throughput screening during cultivar development for the presence of the F. glaucescens-derived genes for drought resistance. The frequency of intergeneric recombination between L. multiflorum and F. glaucescens is described. During the initial stages of the breeding programme, preferential intraspecific chromosome pairing between Lolium homologues and Festuca homoeologues dominated with low frequencies of intergeneric chromosome associations. However, these increased in the backcross 1 due to the absence of opportunities for intraspecific chromosome pairing between homoeologous Festuca chromosomes following the loss of half of the Festuca chromosomes. Once transferred to Lolium, F. glaucescens sequences recombined with Lolium at high frequencies, thereby enabling the loss of potentially deleterious gene combinations that might reduce the forage quality of Lolium.  相似文献

The anticipated complexity of multifunctional grasslands with environment‐friendly and sustainable management practices demands better understanding of traits, their interactions, and their genetic control. Intergeneric hybrids between closely related Lolium and Festuca species are being used to broaden the gene pool and provide the plant breeder with options to combine complementary traits aimed at high quality but more robust grass varieties for the future. New techniques in introgression mapping provide opportunities for precision breeding whereby desirable gene combinations transferred from one species into another are selected preferentially, with the exclusion of deleterious alien genes. The close homology between genomes of Lolium and Festuca species allows high levels of chromosome pairing and recombination. Using genomic in situ hybridisation (GISH) on Lolium/Festuca hybrids and their derivatives, recombination between Lolium and Festuca chromosomes is observed at any point along the chromosome. The system provides unlimited access to any combination of Lolium and Festuca DNA sequence. Moreover, genes transferred between homoeologous chromosome sites are expected to function normally at their new locations. Alien chromosome segments may be reduced in size by further recombination events thereby reducing linkage drag. Molecular markers such as AFLPs, SSRs, SNPs, or RFLPs are being targeted to genes of interest to allow their selection through different generations in plant breeding programmes. Relatively simple PCR‐based marker systems are used for specific traits as breeders' toolkits in plant breeding programmes.  相似文献

Colchicine induced tetraploids (4x=28) from diploidSecale cereale heterozygous for a translocation showed a strong tendency of non-preferential pairing for the interchanged chromosomes. The normal chromosomes associated in configurations up to quadrivalents, and the translocation complex formed multivalents up to octavalents. Most of the interchanged chromosome associations were characterized by their heteromorphic nature. The percentage of the chromosomes in the interchange complex forming multivalent associations was far higher than that of the remaining twenty chromosomes. Abnormalities were observed at anaphase I and II in the pollen mother cells. The tetraploids appeared to be completely sterile. It is suggested that the high frequency of multivalent formation may be explained on the basis that the interchange might have involved a region of localized chiasmata. The absence of polyploidy in the genusSecale as against its widespread occurrence in the related grass genera may be accounted for, in part, on the basis of non-preferential pairing.  相似文献

Morphometric karyotype characters were studied in 25Angelica spp. (Umbelliferae, Apioideae) and in one species of the related genusTommasinia. For three species the chromosome numbers are new. In our study the majority of the species investigated are diploids with 2n = 22, some are tetraploids with 2n = 44 (for these tetraploids also diploid cytotypes are reported in the literature). Among the diploid species,A. miqueliana has a distinct karyotype consisting of submetacentric and acrocentric chromosomes only, the remaining diploids with 2n = 22 as well as tetraploids with 2n = 44 have rather symmetrical karyotypes, consisting of metacentric and submetacentric chromosomes. The very different chromosome number 2n = 28 has been found inA. gmelinii. Its karyotype includes two distinct groups of chromosomes: 8 pairs of rather large metacentrics and submetacentrics and 6 pairs of very short and asymmetrical chromosomes. Chromosome numbers and structures appear to be useful in the taxonomy of some intrageneric taxa inAngelica.  相似文献

Plants were regenerated from cultured excised leaf segments ofTurnera ulmifolia (2n = 6x = 30). Cytological studies have demonstrated that chromosome doubling occurred in 100% of the regenerated plants. Probably it was produced by endomitosis, induced by excess of auxins in relation to cytokinins. High bivalent and low quadrivalent frequency, and univalents to octovalents were observed in metaphase I; lagging chromosomes were also found. Probably, the presence of multivalents may be due to the pairing among homoeologous chromosomes because the mother plant is a segmental allohexaploid. The high bivalent frequency may have been caused by preferential pairing of identical chromosomes against the homologous.  相似文献

Summary A set of species-specific repetitive DNA sequences was isolated from Lolium multiflorum and Festuca arundinacea. The degree of their species specificity as well as possible homologies among them were determined by dot-blot hybridization analysis. In order to understand the genomic organization of representative Lolium and Festuca-specific repetitive DNA sequences, we performed Southern blot hybridization and in situ hybridization to metaphase chromosomes.Southern blot hybridization analysis of eight different repetitive DNA sequences of L. multiflorum and one of F. arundinacea indicated either tandem and clustered arrangements of partially dispersed localization in their respective genomes. Some of these sequences, e.g. LMB3, showed a similar genomic organization in F. arundinacea and F. pratensis, but a slightly different organization and degree of redundancy in L. multiflorum. Clones sequences varied in size between 100 bp and 1.2 kb. Estimated copy number in the corresponding haploid genomes varied between 300 and 2×104. Sequence analysis of the highly species-specific sequences from plasmids pLMH2 and pLMB4 (L. multiflorum specific) and from pFAH1 (F. arundinacea specific) revealed some internal repeats without higher order. No homologies between the sequences or to other repetitive sequences were observed. In situ hybridization with these latter sequences to metaphase chromosomes from L. multiflorum, F. arundinacea and from symmetric sexual Festulolium hybrid revealed their relatively even distribution in the corresponding genomes. Crosses ofHordelymus europaeus (2n = 4x = 28) with four genera in theTriticeae were attempted. Adult hybrids were obtained in combinations withHordeum bogdanii (2x),Hordeum depressum (4x), andSecale cereale (2x). The meiotic pairing was very low in the hybrids withH. bogdanii andSecale cereale (0.12 and 0.30 chiasmata/cell, respectively), whereas high pairing (9.90 chiasmata/cell) was found in hybrids withH. depressum due to autosyndetic pairing ofH. depressum chromosomes. The chromosome complement ofHordelymus europaeus comprised 16 metacentrics, 8 submetacentrics, and 4 SAT-chromosomes. The Giemsa C-banding patterns of the chromosomes were characterized by small to minute bands at no preferential positions. It is hypothesized thatHordelymus europaeus may genomically be closest related toTaeniatherum andPsathyrostachys spp.  相似文献

Genomic in situ hybridisation (GISH) was used to reveal chromosome pairing in two partly fertile, triploid (2n = 3x = 21) hybrids obtained by crossing the diploid (2n = 2x = 14) Festuca pratensis Huds. (designated FpFp), used as a female parent, with the autotetraploid (2n = 4x = 28) Lolium multiflorum Lam. (designated LmLmLmLm), used as a male parent. The pattern of chromosome pairing calculated on the basis of the mean values of chromosome configurations identified in all 100 PMCs analysed, was: 0.71I Lm + 2.24I Fp + 2.18II Lm/Lm + 0.54II Lm/Fp + 4.18III Lm/Lm/Fp. A relatively high number of Lm/Lm bivalents and Fp univalents, and a low number of Lm/Fp bivalents and Lm univalents indicated that the pairing was preferential between L. multiflorum chromosomes. Other observations regarding chromosome pairing within the Lm/Lm/Fp trivalents also confirmed this preferential pairing in the analysed triploids, as the Fp chromosome was not randomly located in the chain- and frying-pan-shaped trivalents. The similarities and differences in chromosome pairing at metaphase I and the level of preferential pairing between Lolium chromosomes in the different triploid Lolium-Festuca hybrids are discussed.  相似文献

Six diploid populations of Lolium rigidum were used in crosses with a standard homozygous line of Lolium temulentum in order to analyse the variation in the effect of B chromosomes on homoeologous pairing in the interspecific hybrid. — Large differences in chiasma frequency were found, both between the progeny of the different populations and, in one case, between the progeny of different plants from the same population. That this variation was due to differences in B chromosome genotype was ruled out by the use of appropriate 1B Lolium rigidum parents. It is concluded therefore that the effect of B's on homoeologous pairing in this interspecific hybrid can be modified quite substantially by the A chromosome genotype of the parents.  相似文献

Summary A number of inbred lines of Lolium perenne have been developed as far as the fifth inbred generation and are being used for basic studies on the cytogenetics of the species. An analysis on variation in the pattern of chiasma frequency and distribution, including the parent plants and all generations down to the S5, reveals that the effect of inbreeding is to reduce chiasma frequency and to increase both the cell and bivalent variances. Evidence is presented for a genetic basis of polygenic control of chiasma formation and distribution, and a model is suggested for control over the three related components of variation which involves a two-stage level of regulation.  相似文献
