SYNOPSIS. Plasmodium lophurae hemozoin (malaria pigment) is a heme-containing protein which is distinctly different from hemoglobin and hematin by immunologic, spectrophotometric, fingerprint, heme-iron, gel filtration, and starch gel electrophoretic analyses. The calculated average molecular weight of P. lophurae hemozoin is ca. 40,000. Hemozoin contains at least 3 antigenic components and shows some indication of cross-reaction with hemoglobin.  相似文献

When larval mosquitoes (Aedes aegypti) infected with the mermithid nematode Romanomermis culicivorax were fed on a diet low in quantity or protein content or both, the number of postparasites which emerged from the hosts decreased and host mortality increased marginally. Parasitic development was prolonged and became asynchronous in nutritionally deprived hosts. Nematodes emerged from insects infected by more than one nematode before the remaining juveniles comprising such infections had completed parasitic development; this resulted in substantial reductions in postparasite nttmbers. Host development was retarded by low protein and/or reduced diets. Postparasites emerging from second and third instars were reduced in size and in the amount of stored nutriment compared to those recovered from hosts fed on a high protein diet ad libitum. A greater proportion of the rnermithids developed into males in hosts fed on reduced diets but not in hosts fed on low protein diets.  相似文献

2011年从辽宁省丹东地区蚊虫样品中分离到6株病毒,采用逆转录-聚合酶链式扩增(RT-PCR)检测方法,结果黄病毒属通用引物和库蚊黄病毒特异性引物均为阳性。6株病毒核苷酸序列经基因库(GenBank)比对后,证实6株病毒为库蚊黄病毒,为我国首次报道。将病毒NS5基因和E基因核苷酸序列与GenBank中10株库蚊黄病毒参考毒株核苷酸序列进行比较,并构建遗传进化树,结果显示本次分离的6株病毒与美国和日本的毒株亲缘关系较近。  相似文献

The increasing global rate of road construction is leading to a parallel increase of areas of degraded soil conditions and steep slopes that need revegetation. Hydroseeding with commercial seeds of fast‐growing grasses and legumes is a common practice in revegetation of motorway slopes. We carried out 3 years of monitoring of vegetation dynamics on hydroseeded and nonhydroseeded motorway slopes (48 slopes) in a maritime Mediterranean zone in Málaga (southern Spain). Our main objectives were to test whether hydroseeding significantly increases species richness and plant cover and whether hydroseeded species act as starters, facilitating the establishment of the vegetation and quickly disappearing once the communities are established. A hydroseeding success index (HSI, ranging from 0 to 1) was used to assess the relative abundance over time of the 14 species from the hydroseeding mixture. Species richness and cover was significantly higher on embankments (50–70 species per embankment, 80–90% cover) than on roadcuts (6–10 species per roadcut, 18–30% cover). Performance of hydroseeded species was poor from the very beginning (HSI, 0.2–0.3). On embankments, either presence or abundance of hydroseeded species did not significantly vary throughout the study. Both hydroseeded and nonhydroseeded communities exhibited a significant decrease in species richness, a significant increase in plant cover, and a highly dynamic species composition over time, with Sorensen index of 0.3–0.5 between years. There were no significant differences in plant cover, species richness, and aboveground biomass between hydroseeded and nonhydroseeded plots on embankments throughout the study. Our results demonstrate that there are situations in which the use of hydroseeding for revegetation is not needed. Further research should focus on understanding the establishment of autochthonous species and identifying environmental conditions under which the addition of commercial seeds may not be needed, or indeed situations where it may be harmful in suppressing autochthonous species.  相似文献

Zika virus(ZIKV) has been isolated from mosquitoes such as Aedes, Mansonia uniformis, and Culex perfuscus; However,the isolation of ZIKV from Anopheles sinensis and Culex tritaeniorhynchus has not yet been reported. In June and July2018, 22,985 mosquitoes and 57,500 midges were collected in Jiangxi Province in southeastern China. Among them, six strains of ZIKV were isolated from mosquitoes: four from An. sinensis and two from Cx. tritaeniorhynchus. Molecular genetic analysis showed that the ZIKV isolated from An. sinensis and Cx. tritaeniorhynchus belonged to genotype 2 in the Asian evolutionary branch of ZIKV. In addition, the ZIKV strains isolated from An. sinensis and Cx. tritaeniorhynchus had amino acid substitutions identical to ZIKV strains prevalent in South America since 2015. This study is the first to isolate ZIKV from mosquito specimens collected in the wild of Jiangxi Province, China; This is also the first time that ZIKV has been isolated from An. sinensis and Cx. tritaeniorhynchus. Given that An. sinensis and Cx. tritaeniorhynchus have a very wide geographical distribution in China and even in eastern and southern Asia, the isolation of several strains of ZIKV from these two mosquitoes poses new challenges for the prevention and control of ZIKV infection in the mainland of China and countries and regions with the same distribution of mosquitoes.  相似文献

The dynamics of the microbial food sources for Aedes triseriatus larvae in microcosms were found to be strongly influenced by larval presence. The total abundance of bacteria in water samples generally increased in response to larvae, including populations of cultivable, facultatively anaerobic bacteria. Additionally, a portion of the community shifted from Pseudomonaceae to Enterobacteriaceae. Bacterial abundance on leaf material was significantly reduced in the presence of actively feeding larvae. Principle-component analysis of whole community fatty acid methyl ester (FAME) profiles showed that larvae changed the microbial community structure in both the water column and the leaf material. Cyclopropyl FAMEs, typically associated with bacteria, were reduced in microcosms containing larvae; however, other bacterial fatty acids showed no consistent response. Long-chain polyunsaturated fatty acids characteristic of microeukaryotes (protozoans and meiofauna) declined in abundance when larvae were present, indicating that larval feeding reduced the densities of these microorganisms. However, presumed fungal lipid markers either increased or were unchanged in response to larvae. Larval presence also affected microbial nitrogen metabolism through modification of the physiochemical conditions or by grazing on populations of bacteria involved in nitrification-denitrification. Stemflow primarily influenced inorganic ion and organic compound concentrations in the microcosms and had less-pronounced effects on microbial community parameters than did larval presence. Stemflow treatments diluted concentrations of all inorganic ions (chloride, sulfate, and ammonium) and organic compounds (total dissolved organic carbon, soluble carbohydrates, and total protein) measured, with the exceptions of nitrite and nitrate. Stemflow addition did not measurably affect larval biomass in the microcosms but did enhance development rates and early emergence patterns of adults.  相似文献

ABSTRACT. Cell-free schizonts of Plasmodium knowlesi , a simian malaria parasite, possess significant isocitrate dehydrogenase (IDH) activity, about 90% of which is contributed by the NADP-specific enzyme that is localized in the cytosolic fraction. The enzyme has been partially purified by affinity chromatography using Blue sepharose CL-6B. Although unstable in nature, it is stabilized by citrate and glycerol. Kinetic studies with dl -isocitrate and NADP yielded hyperbolic curves with Michaelis constants of 0.210 and 0.038 mM, respectively. Manganous or magnesium ions are essential for activity. The enzyme is thermosensitive, shows maximum activity at pH 8.0, and has a molecular mass of about 48.5 kDa. It is strongly inhibited by thiolblocking agents but protected against them by thiol-providing agents. Cupric and argentic ions also have a marked inhibitory effect on its activity. The enzyme is significantly inhibited by chloroquine and oxytetracycline in vitro, but to a lesser degree by tetracycline.  相似文献

ABSTRACT Understanding the population structure of Plasmodium parasites is essential for malaria intervention. A survey of parasites in vectors and host infections was conducted in an area of intense mortality due to malaria in a captive penguin ( Spheniscus demersus ) colony, using a novel method for identification of Plasmodium species by amplification of ribosomal sequences in DNA or RNA. Three phylogenetically distinct groups of avian Plasmodium were detected in mosquitoes ( Culex ) collected at the study site (Baltimore Zoo, Baltimore, MD) during a period of high transmission. One of the three clades of Plasmodium was found to be prevalent in penguins monitored through the malaria transmission season and consistent with morphological identification as Plasmodium relictum. This parasite sequence was directly associated with the death of a penguin. Thus, a complete transmission cycle is defined at this site. Phylogenetic comparison of ribosomal sequences to an authenticated reference strain of Plasmodium relictum indicates that this is not the parasite causing death in the penguins, suggesting that different parasites may be morphologically indistinguishable.  相似文献

Eukaryotic parasites of the genus Plasmodium cause malaria by invading and developing within host erythrocytes. Here, we demonstrate that PfShelph2, a gene product of Plasmodium falciparum that belongs to the Shewanella-like phosphatase (Shelph) subfamily, selectively hydrolyzes phosphotyrosine, as shown for other previously studied Shelph family members. In the extracellular merozoite stage, PfShelph2 localizes to vesicles that appear to be distinct from those of rhoptry, dense granule, or microneme organelles. During invasion, PfShelph2 is released from these vesicles and exported to the host erythrocyte. In vitro, PfShelph2 shows tyrosine phosphatase activity against the host erythrocyte protein Band 3, which is the most abundant tyrosine-phosphorylated species of the erythrocyte. During P. falciparum invasion, Band 3 undergoes dynamic and rapid clearance from the invasion junction within 1 to 2 s of parasite attachment to the erythrocyte. Release of Pfshelph2 occurs after clearance of Band 3 from the parasite-host cell interface and when the parasite is nearly or completely enclosed in the nascent vacuole. We propose a model in which the phosphatase modifies Band 3 in time to restore its interaction with the cytoskeleton and thus reestablishes the erythrocyte cytoskeletal network at the end of the invasion process.  相似文献

Antibodies to Toxoplasma gondii were determined in serum samples from 616 equids (454 horses, 80 mules and 82 donkeys) in a cross-sectional study of 420 herds in Andalusia (Southern Spain), the region with the highest number of equids in Spain. Antibodies to T. gondii were found in 10.8% horses, 15.0% mules and 25.6% donkeys by using the modified agglutination test (MAT) at a cut-off of 1:25. Herd seroprevalence for horses, mules and donkeys was 14.7% (48/327), 23.9% (11/46) and 34.0% (16/47), respectively, and 75 herds (17.8%) had at least one seropositive animal. Significant differences in T. gondii seroprevalence were observed among species, with donkeys having the highest seroprevalence and horses the lowest (P=0.04). Seroprevalence was significantly higher in herds with presence of domestic ruminants. This study is the first report of the presence of T. gondii antibodies in equine species in Spain and the first reporting T. gondii infection in donkeys in Europe. The presence of antibodies is indication of contact with the parasite and therefore, consumption of equine meat could be a potential source of human infection in Spain.  相似文献

Ultrastructural investigations of P. falciparum cultivated in vitro in human erythrocytes revealed new features of the feeding mechanism of the parasite. Mature trophozoites and schizonts take up a portion of the host cytosol by endocytosis which is restricted to cytostomes and which involves the invagination of both parasitophorous and parasite membranes. The resulting endocytic vesicles, surrounded by two concentric membranes, migrate towards the central food vacuole membrane. The external membrane of the endocytic vesicles apposes that of the food vacuole, leading to the internalization of vesicles bounded by a single membrane into the vacuolar space where they are rapidly degraded. We conclude from this sequence of events that endocytic vesicles fuse with the food vacuole. Treatment of infected cells with therapeutic concentrations of chloroquine inhibited the last step of the feeding process, i.e. vacuolar degradation. This was manifested by the accumulation within the vacuolar space of intact vesicles bounded by single membranes. The implications of these findings for the antimalarial activity of chloroquine are discussed.  相似文献

Namibia supports a highly diverse avifauna of 644 species, including over 90 species endemic to the southern African subregion and 13 species endemic to the country. Patterns of species diversity in relation to protected areas and habitat types were analysed using data from the Southern African Bird Atlas Project. A modified Shannon index appropriate for atlas data was used to derive an index of diversity for all species, wetland, terrestrial, threatened, and regional endemic species. Species richness for Namibian endemics was mapped. Overall species diversity is highest in the northeast of Namibia where wetland and riparian habitats coincide. Both wetland and terrestrial species show highest diversity in this area. The greatest diversity of southern African endemics falls within the Savanna-Karoo systems. Several key areas are identified for red data species, including the Caprivi Strip, Kunene and Orange Rivers, coastal wetlands and ephemeral river mouths and pans. This highlights the pressures operating on wetland and riparian habitats in arid environments. Concentrations of Namibian endemics are found in the northwestern (Kaoko) escarpment of the country. Although much of the area of high diversity of wetland, terrestrial and red data species falls within protected areas, national and regional endemics are poorly represented within national parks.  相似文献
