A technique is described to accurately assess the dermal contactdose-mortality response of molluscicides. The technique involvescarbon dioxide anaesthesia of laboratory reared Deroceras reticulatum(Müller) followed by application of test substances insolution to the dorsal surface. The LD50 of the substances testedwere as follows: Sodium pentachlorophenate, 55.9 ng.mg–1,Copper sulphate, 124.1 ng.mg–1, Methiocarb, 314.6 ng.mg–1and Potassium permanganate, 418.9 ng.mg–1 body weight. Externally supplied [3H]myo-inositol and [14C]glucose were incorporatedin cell-wall fractions of segments of etiolated squash hypocotyl.The extent of incorporation of [14C]glucose into cell-wall fractionswas very much greater than that of [3H]myo-inositol. Radioactivityfrom [14C]-glucose was effectively incorporated into hemicelluloseB and cellulose fractions and was incorporated uniformly intohexose, pentose and uronic acid residues, but radioactivityfrom [3H]myo-inositol was incorporated predominantly into uronicacid and pentose residues in the pectin and hemicellulose Bfractions. Exogenously applied ABA significantly suppressed the elongationof segments of squash hypocotyl and the incorporation of radioactivityfrom [l4C]glucose and [3H]myo-inositol into the segments. Furthermore,ABA significantly inhibited the distribution of incorporatedradioactivity from [14C]glucose into the cellulose fraction,but did not affect distribution into the pectic fraction. Bycontrast, ABA only slightly inhibited the distribution of theincorporated radioactivity from [3H]myo-inositol into the pecticfraction. These results suggest that most of the cell-wall polysaccharidesin segments of squash hypocotyl are synthesized via the UDP-sugarpathway, and that ABA significantly inhibits the synthesis ofcellulose but not the synthesis of pectic polysaccharides whenABA suppresses the elongation of the segments. Feeding behavior of the brittle star Ophiura ophiura includesorienting posture, orienting movements, arm ‘walking’,changing the direction of ‘walking’ arm coilingand ingestion. All sequential behavior patterns were releasedor enhanced by single low-molecular-weight compounds. Stimuliwhich released ‘walking’ behavior at high concentrations(10–4 M) in all the test animals are listed in decreasingorder of sensitivity: sarcosine, glycine, urea, L-valine, L-leucine,L-methionine, L-homocysteine, L-norvaline, L-norleucine, L-threonine,L-serine, S-methyl-L-cysteine, L-proline. Threshold values forsingle amino acids were as much as 100 times different in differentindividuals and ranged from 3 x 10–9 to 3 x 10–7M for the most effective stimulus, sarcosine, and from 10–6to 10–4 M for proline. Above 10–5 M, only L-prolineregularly released a second behavior pattern, the arm coilingresponse, which temporarily inhibited the ‘walking’behavior. Behavioral thresholds for the ‘walking’behavior for L(+)-lactate and L-alanine were higher than thosefor the orienting movements. Thyoglycolic acid and ß-alaninereleased tube feet walking, which is not part of the feedingbehavior. Structure—activity comparisons were studied at estimated10–5 M concentrations. Gycline, sarcosine, L-valine, L-norvaline,L-leucine, L-isoleucine, DL-norleucine and DL-homocysteine releasedarm ‘walking’ behavior in more than 75% of all thetests. With the exceptions of S-methyl-and S-ethyl-cysteine,and glycine methylester, derivatives of amino acids were noteffective behavioral stimuli in Ophiura ophiura. L-Isomers ofvaline and leucine regularly stimulated the ‘walking’behavior while their D-isomers were effective in some testsand ineffective in others. Acetylcholine iodide, acetyl-ß-methylcholine chloride and choline phosphate chloride regularly released‘walking’ behavior at concentrations above 10–5M.  相似文献

Salt inhibition of RuBP carboxylase activity from Aphanothecehalophytica was caused by Cl-, but not by K+ nor Na+. Interstrain differences in bitter taste responses were examinedusing inbred strains of mice. Taste responses were recordedfrom the glossopharyngeal and chorda tympani nerves of SWR/J,LP/J, BDP/J and DBA/2J mice. There were large differences inthe magnitude of responses to sucrose octaacetate (SOA) in boththe glossopharyngeal and chorda tympaninerves of SWR/J miceas compared with the other strains of mice. SOA thresholds ofSWR/J mice were 10–7–6 M, whereas they were– 10–4 M in LP/J mice. On the other hand, no appreciabledifferences were observed in the responses to quinine hydrochlorideand pnenyl-thio-carbamide. The effects of Na application on growth and nitrate reductaseactivity of seven C4 plant species, Zea mays, Echinochloa crus-galli,Panicum miliaceum, Panicum coloratum, Panicum dichotomiflorum,Panicum maximum and Chloris gayana were studied. For three Prorocentrum species, P. minimum (Pavillard) Schiller,P. rhathymum sp. nov., and P. triestinum Schiller, we have foundthat both flagella emerge from a single large pore. This arrangementis contrary to previous observations which suggested they arosefrom separate pores. Prorocentrum triestinum has only one anteriorpore, whereas P. minimum and P. rhathymum sp. nov. have an additionalauxiliary pore. The pores in all three species are formed bya series of small plates. 1-O-trans-p-Coumaroyl-rß-D-glucopyranose (p-coumaroyl-D-glucose)was isolated from slices of sweet potato root which had beenincubated with trans-cinnamic acid. Pre-loaded trans-cinnamicacid efficiently trapped the radioactivity from L-[U-14C]-phenylalanineand reduced its incorporation into chlorogenic acid by 75% ofcontrol values in disks of sweet potato root. In the root diskssupplied with trans-[3-14C]-cinnamic acid, the radioactivitywas transferred first to trans-cinnamoyl- D-glucose, then top-coumaroyl-D-glucose, and subsequently to chlorogenic acidand isochlorogenic acid. These results support our earlier propositionthat p-coumaroyl-D-glucose is involved in the biosynthesis ofchlorogenic acid as an intermediate adjacent in the pathwayto trans-cinnamoyl-D-glucose in sweet potato roots. As reported in a previous article [Kataoka (1988a) Plant CellPhysiol. 29: 1323], growing apices of the xanthophycean coenocyticalga, Vaucheria terrestris, bends away from a unilateral bluelight (BL) source, if they are simultaneously irradiated withstrong background BL in a solution containing 1–4 mM Ca2+.Since the negative bending is a function of the product of theexternal Ca2+ concentration and the fluence rate of backgroundBL, a BL-induced Ca2+-influx at the apex was hypothesized tobe the cause of the phototropic inversion. The present reportprovides strong evidence for this hypothesis. Addition of theCa2+ channel blockers, La3+, verapamil, nifedipine and nitrendipineto media containing 4 mM Ca2+ completely inhibited the phototropicinversion. By contrast, 1 µM A23187 [GenBank] (plus 4 mM Ca2+) notonly enhanced the phototropic inversion under background BL,but also mimicked the background BL; i.e. it caused negativebending under safe red light. Inhibition of phototropic inversionby La3+ is also observed under conditions where the algae areirradiated with unilateral BL for 1 week. A BL-dependent Ca2+influx and a consequential elevation of the cytoplasmic Ca2+-levelin the apical growth region must be involved in the early stepsof phototropic response. Ultrastructural features of spermiogenesis and mature spermatozoaare described for the first time in a cirrate octopod. Comparisonwith other investigated cephalopod species shows that spermatozoaof Opisthoteuthis persephone Berry most closely resemble thoseof the genus Octopus. The shared features include: (1) an elongate,solid acrosome with internal banding and a prominent helicalkeel; (2) a straight, rod-like nucleus; (3) a short midpieceand (4) a post-mitochondrial (‘annular’) skirt.Of these, the acrosomal morphology is most significant taxo-nomically,as the internal banding of the acrosomal vesicle only occursin the Octopoda. Spermatozoa of Opisthoteuthis and Octopus differhowever in the extent of the internal banding of the acrosome(poorly developed in Opisthoteuthis) and substructure of theacrosomal keel (more complex in Opisthoteuthis). Surprisingly,the extensive fibrous plug of Octopus and Vampyroteuthis sperm,is not developed either in Opisthoteuthis or in Eledone. Resultsprovide additional support for the monophyly of the Octopodabut also hint at the possibility that some groups of in-cirratesmay not be as advanced as generally believed. Fleurat-Lessard, P., Roblin, G., Bonmort, J. and Besse, C. 1988.Effects of colchicine, vinblastine, cytochalasin B and phalloidinon the seismonastic movement of Mimosa pudica leaf and on motorcell ultrastructure.—J. exp. Bot. 39: 209–221. Colchicine at 1 x 10–3 mol dm–3 does not affectthe seismonastic movement of Mimosa pudica leaves but disruptsmicrotubules in motor cells. Vinblastine at 5 x 10–3 moldm–3 does not affect this movement and partly disruptsmicrotubules. Vinblastine at 1 x 10–4 mol dm–3 alwaysdisrupts microtubules, even after a 12 h reversibility whenthe movement is restored. These drugs, applied at the same respectiveconcentrations, do not alter cytoplasmic and vacuolar fibrils.Cytochalasin B and phalloidin alter the seismonastic movementof Mimosa leaves when applied at concentrations of 1.25 x 10–3and 2.4 x 10–4 mol dm–3 respectively. These drugs,used at the same respective concentrations, also affect themotor cell structure and, in particular, modify the arrangementand the structure of the fibrils but they do not destroy themicrotubules. These data suggest that microtubules are not directly involvedin the seismonastic reaction whereas fibrils, formed by thin(3.0 nm wide) filaments, may be implicated in this reaction. The thermal dependence of two of the reactions catalyzed bythe nitrate reductase from Chlorella vulgaris was determined.The activation energies for NADH:nitrate oxidoreductase (EC1.6.6.1 [EC] ) and NADH:Cytochrome c oxidoreductase (EC 1.6.99.3 [EC] )are 42.1 kJ?mol–1 and 21.5 kJ?mol–1, respectively.Since the thermal dependency of the two enzymes is different,ratios of the activities will vary with temperature. A method is described for the rapid separation of carbonic anhydrase(CA) isozymes by cellulose acetate membrane electrophoresisin which CA activity is detected using the pH-indicating dye,bromcresol purple. This method can detect bovine erythrocyteCA in a 0.3 mm3 sample applied at a concentration of 100 ngcm–3 (total of 30 pg applied) while at higher concentrationsthree isozymes were observed. It was found, using a potentiometrictechnique, that intact cells of Anabaena flos-aquae (Cyanophyceae)and Chlorella ellipsoidea had no detectable activity while C.saccharophila and Chlamydomonas reinhardtii (Chlorophyceae)had external CA activity. CA activity of the extracts suggestedthe presence of internal CA in all species. After electrophoresisit was found that C. saccharophila and C. reinhardtii had twoisozymes while A. flos-aquae and C. ellipsoidea had only a singledetectable band. Spinach had up to five detectable isozymesthat were difficult to resolve. Incubation of spinach extractwith the CA inhibitor ClO4 (500 mol m–3) inhibitedCA activity by 90% using the potentiometric technique, but afterelectrophoresis had no detectable effect. This technique isuseful in identifying isozymes that are substantially differentin electrical charge and in monitoring CA isozyme activity duringenzyme purification. Using permeabilized characean cells in which the ionic conditionsat the cytoplasmic side of the tonoplast are easily controlled,effects of Ca2+ ion on tonoplast potential were examined. Whenthe cell was treated with 1 µM Ca2+, the tonoplast potential(EM became positive in a complicated manner in Chara corallinawhile it simply became negative in Nitella axilliformis. Whenthe cell was treated with 9-antracenecarboxylic acid, a Cl-channelinhibitor, Em became more negative and the response of Em toCa2+ was significantly suppressed. We investigated the role of intracellular Mg2+(Mgi2+) on the ATP regulation ofNa+/Ca2+ exchanger in squid axons and bovineheart. In squid axons and nerve vesicles, the ATP-upregulated exchangerremains activated after removal of cytoplasmic Mg2+, evenin the absence of ATP. Rapid and complete deactivation of theATP-stimulated exchange occurs upon readmission ofMgi2+. At constant ATP concentration, the effectof intracellular Mg2+ concentration([Mg2+]i) on the ATP regulation of exchangeris biphasic: activation at low [Mg2+]i,followed by deactivation as [Mg2+]i isincreased. No correlation was found between the above results and thelevels of phosphatidylinositol 4-phosphate and phosphatidylinositol 4,5-bisphosphate [PtdIns(4,5)P2] measured innerve membrane vesicles. Incorporation ofPtdIns(4,5)P2 into membrane vesicles activates Na+/Ca2+ exchange in mammalian heart but not insquid nerve. Moreover, an exogenous phosphatase prevents MgATPactivation in squid nerves but not in mammalian heart. It is concludedthat 1) Mgi2+ is an essentialcofactor for the deactivation part of ATP regulation of the exchangerand 2) the metabolic pathway of ATP upregulation of theNa+/Ca2+ exchanger is different in mammalianheart and squid nerves.

Retusa obtusa held individually in the laboratory carried largeoocytes in the gonad but did not spawn in December-late January.In late Jan-early Feb, however, the first few individuals producedbatches of 8–17 eggs and, by early February, most individualswere producing means of 2–4 eggs d–1 and maximain late Feb of <7.9 d–1. Spawning declined to low levelsin few individuals in late March. By early April, spawning hadvirtually ceased prior to the drastic loss of adults in thepopulation by May. Larger R. obtusa generally produced more eggs than smaller onesand the few specimens shorter than 3.8 mm produced markedlyfewer than those of 3.8–5.0 mm shell length. Twenty of the 34 R. obtusa held in the main breeding period(4 sets started 29 Jan-3 Mar) spawned in 3 or more successiveweeks whereas only 6 of them failed to spawn in 2 consecutiveweeks, so indicating that several successive batches of eggsare commonly produced. Yet in virtually all individuals (exceptat the start of breeding) more eggs were produced in the firstweek of captivity than subsequently. These first-week valuesimply total production by individuals in the spawning seasonof some 112-305 eggs comprising about 2.8-7.6 mg dry weightor, by the Forth estuary population, of 26880-73200 eggs m–2comprising 0.67-1.83 g dry weight m–2. The growth rates of four saline-lake diatom taxa were measuredunder varying conditions of salinity (5, 8 and 11), brine type(sulfate- versus bicarbonate-dominated) and nitrogen form (NH4+versus NO3), using a full factorial design. With NO3as the nitrogen source, Cyclotella quillensis, Cymbella pusillaand Anomoeoneis costata exhibited lower growth rates in thesulfate versus bicarbonate media. The strain of Chaetoceroselmorei used in these experiments, isolated from a sulfate-dominatedlake, was unable to grow on NO3 alone. In the NH4+ treatments,neither salinity nor brine type affected the growth rates ofC.quillensis or C.elmorei. When supplied with NH4+, C.pusillaand A.costata had higher growth rates in the bicarbonate versussulfate media, although for C.pusilla the difference on NH4+was not as great as on NO3. The impact of brine typeon NO3 use is consistent with the theory that sulfateinhibits molybdate uptake, as molybdenum is required for NO3use but not NH4+. Cymbella pusilla was the only taxon affectedby changes in salinity. The four taxa used in these experimentsare frequently found in saline lakes and saline-lake sediments,hence they are used in paleoclimate reconstructions; the resultspresented here provide additional information that may enhancethese diatom-based reconstructions.  相似文献

Post-embryonic development time, egg production rate and adultbody size of calanoid copepod Eurytemora affinis from Lake Ohnuma,Japan, were determined under six temperature-food conditions(103,5 x 103, 104 and 5 x 104 cells ml–1 at 15°C,5 x 104 cells ml–1 at 10and20°C) in the laboratory.The measured parameters varied with both temperature and foodconcentration. Development time from hatching to adult femalewas 9.2, 11.4 and 22.8 days at 20, 15 and 10°C respectively,at the highest food concentration. The males developed to adultat one to two days earlier than the females. An effect of foodshortage on development time occurred at the lowest food concentration.This development time was 24.8 days even at 15°C, beingtwice as long as that at the highest food concentration. Prosomelength of these food-limited females was approximately 75% ofwell-fed ones, which reduced by only 10% with increasing temperaturefrom 10 to 20°C. Clutch size (C, eggs clutch–1) ofwell-fed individuals depended on prosome length of the adultfemale (L, mm), and was expressed as an equation: C = 65.2 L383. Clutch size of individuals reared at less than 104 cellsml–1, however, mostly laid below the estimated curve,especially at the lowest food concentration being only 10% ofthat at the highest food concentration. These results suggestthat food availability is a more important factor affectingpopulation growth of E.affinis in Lake Ohnuma than variationof temperature.  相似文献

Cellular membrane fractions, including endoplasmic reticum (ER),Golgi-enriched membrane, plasma membrane and tonoplasts, wereisolated from Vigna radiata seedlings. Each of these membranefractions was associated with specific ATPases which were highlydependent on Mg2+. ATPases of ER, Golgi-enriched membrane andplasma membrane were sensitive to vanadate but the tonoplastATPase was not. ATPases were mostly dependent on Cl1, but aslight stimulation by K+ was observed in the case of ATPasesof Golgi-enriched membrane and plasma membrane. KNO3 inhibitedtonoplast ATPase but stimulated the other ATPases. ER ATPasecan be distinguished from other ATPases by the following characteristics:specific inhibition by KNO2 and Triton X-100, stimulation bylow concentrations of diethylstilbestrol and 4,4'-diisothiocyanostilbene-2,2'-disulfonicacid, and high sensitivity to heat. The ATPases showed typicalMichaelis-Menten kinetics and had Km values of 0.5 to 0.6 ITIMMg2+-ATP for ER, Golgienriched-membrane and tonoplast ATPases,and 2.27 msi Mg2+-ATP for plasma membrane ATPase. ATPases ofGolgi-enriched membranes and plasma membranes had similar properties,but they were still distinguishable by the differences in theirKm values and their responses to Triton X-100.
