In the present study, toluene elimination in the polyurethane (PU) biofilter during long-term (145 day) operation was characterized, and assessed the effects of changing the inlet loading and space velocity (SV). A very high elimination capacity of 3.7 kg·m−3·h−1 was obtained at an inlet loading of 4.0 kg·m–3·h−1 (inlet toluene concentration of 900 ppmv at a SV of 1,040 h−1). Backwashing with irrigation and compressed air allowed maintenance of a pressure drop of < 80 mm H2O·m−1-filter at an SV of 830 h−1 and an elimination efficiency of > 90% during the 145 day of operation. In conclusion, the PU biofilter can overcome the problems of clogging caused by excess biomass growth and of low treatment capacities of conventional biofilters.  相似文献

The biodegradation of toluene was studied in two lab-scale air biofilters operated in parallel, packed respectively with perlite granules (PEG) and polyurethane foam cubes (PUC) and inoculated with the same toluene-degrading fungus. Differences on the material pore size, from micrometres in PEG to millimetres in PUC, were responsible for distinct biomass growth patterns. A compact biofilm was formed around PEG, being the interstitial spaces progressively filled with biomass. Microbial growth concentrated at the core of PUC and the excess of biomass was washed-off, remaining the gas pressure drop comparatively low. Air dispersion in the bed was characterised by tracer studies and modelled as a series of completely stirred tanks (CSTR). The obtained number of CSTR (n) in the PEG packing increased from 33 to 86 along with the applied gas flow (equivalent to empty bed retention times from 48 to 12 s) and with operation time (up to 6 months). In the PUC bed, n varied between 9 and 13, indicating that a stronger and steadier gas dispersion was achieved. Michaelis–Menten half saturation constant (k m) estimates ranged 71–113 mg m−3, depending on the experimental conditions, but such differences were not significant at a 95% confidence interval. The maximum volumetric elimination rate (r m) varied from 23 to 50 g m−3 h−1. Comparison between volumetric and biomass specific biodegradation activities indicated that toluene mass transfer was slower with PEG than with PUC as a consequence of a smaller biofilm surface and to the presence of larger zones of stagnant air.  相似文献

Interactions of toluene and p-xylene in air treatment biofilters packed with an inert filter media were studied. The effect of the inlet load of toluene, p-xylene and mixtures of both compounds on the biodegradation rate was analyzed in three lab-scale biofilters. A maximum elimination capacity (EC) of 26.5 and 40.3 g C m−3 h−1 for an inlet load (IL) of 65.6 and 57.8 g C m−3 h−1 was obtained for p-xylene and toluene biofilters, respectively. Inhibition of p-xylene biodegradation by the presence of toluene took place when the mixture was treated, whereas the presence of p-xylene had an enhancing effect on the toluene removal efficiency. Specific growth rates (μ) from 0.019 to 0.068 h−1 were calculated in the mixed biofilter, where the highest values were similar to mixtures with lower p-xylene levels (ILp-Xyl 8.84 ± 0.29 g C m−3 h−1). Michaelis-Menten and Haldane type models were fitted to experimental EC for p-xylene and toluene biofilters, respectively.  相似文献

Rhinocladiella similis biodegraded volatile organic compounds (VOCs) of different polarity in gas-phase biofilters. Elimination capacities, (EC) of 74 ghexane m−3 h−1, 230 gethanol m−3 h−1, 85 gtoluene m−3 h−1 and 30 gphenol m−3 h−1 were obtained. EC values correlated with the solubility of the VOCs. R. similis grown with n-hexane or ethanol in biofilters packed with Perlite showed that the surface hydrophobicity was higher with n-hexane than ethanol. The hydrophobin-like proteins extracted from the mycelium produced with n-hexane (15 kDa) were different from those in the ethanol biofilter (8.5 kDa and 7 kDa).  相似文献

A biofiltration system inoculated with the mold Paecilomyces variotii CBS115145 showed a toluene elimination capacity (EC) of around 250 g/m3 of biofilter/h, which was higher than the values usually reported for bacteria. P. variotii assimilated m- and p-cresols but not the o isomer. Initial toluene hydroxylation occurred both on the methyl group and through the p-cresol pathway. These results were corroborated by detecting benzyl alcohol, benzaldehyde, and p-cresol as volatile intermediates. In liquid cultures with toluene as a substrate, the activity of toluene oxygenase (TO) was 5.6 nmol of O2/min/mg of biomass, and that of benzyl alcohol dehydrogenase was 16.2 nmol of NADH/min/mg of protein. Toluene biodegradation determined from the TO activity in the biofilter depended on the biomass distribution and the substrate concentration. The specific enzymatic activity decreased from 6.3 to 1.9 nmol of O2/min/mg of biomass along the reactor. Good agreement was found between the EC calculated from the TO activity and the EC measured on the biofilter. The results were confirmed by short-time biofiltration experiments. Average EC measured in different biofiltration experiments and EC calculated from the TO activity showed a linear relation, suggesting that in the biofilters, EC was limited by biological reaction. As the enzymatic activities of P. variotii were similar to those reported for bacteria, the high performance of the fungal biofilters can possibly be explained by the increased transfer of the hydrophobic compounds, including oxygen, from the gas phase to the mycelia, overcoming the transfer problems associated with the flat bacterial biofilms.  相似文献

Toluene vapour removal in a laboratory-scale biofilter   总被引:4,自引:0,他引:4  
A bench-scale biofilter with a 0.5-m high filter bed, inoculated with a toluene-degrading strain of Acinetobacter sp. NCIMB 9689, was used to study toluene removal from a synthetic waste air stream. Different sets of continuous tests were conducted at influent toluene concentrations ranging over 0.1–4.0 g m−3 and at superficial gas velocities ranging over 17.8–255 m h−1. The maximum volumetric toluene removal rate for the biofilter (242 g m−3 h−1) was obtained at a superficial gas velocity of 127.5 m h−1 (corresponding to a residence time of 28 s) and a toluene inlet concentration of 4.0 g m−3. Under these operating conditions, toluene removal efficiency was only 0.238, which suggested that effective operation required higher residence times. Removal efficiencies higher than 0.9 were achieved at organic loads less than 113.7 g m−3 h−1. A macro-kinetic study, performed using concentration profiles along the bioreactor, revealed this process was limited by diffusion at organic loads less than 100 g m−3 h−1 and by biological reaction beyond this threshold. Pollutant degradation in biotrickling filters for waste air treatment is generally thought to occur only in the biofilm. In two experiments with toluene degrading biotrickling filters, we show that suspended microorganisms in the recycle liquid may substantially contribute to the overall pollutant removal. Two days after reactor start up, the overall toluene elimination capacity reached a maximum of 125 g m−3 h−1, which was twice that found during prolonged operation. High biodegradation activity in the recycle liquid fully accounted for this short-term peak of pollutant elimination. During steady-state operation, the toluene degradation in the recycle liquid was 21% of the overall elimination capacity, although the amount of suspended biomass was only 1% of the amount of immobilized biomass. The results suggest that biotrickling filter performance may be improved by selecting operating conditions allowing for the development of an actively growing suspended culture. The long-term performance and stability of Pseudomonas putida mt-2 cultures, a toluene-sensitive strain harboring the genes responsible for toluene biodegradation in the archetypal plasmid pWW0, was investigated in a chemostat bioreactor functioning under real case operating conditions. The process was operated at a dilution rate of 0.1 h−1 under toluene loading rates of 259 ± 23 and 801 ± 78 g m−3 h−1 (inlet toluene concentrations of 3.5 and 10.9 g m−3, respectively). Despite the deleterious effects of toluene and its degradation intermediates, the phenotype of this sensitive P. putida culture rapidly recovered from a 95% Tol population at day 4 to approx. 100% Tol+ cells from day 13 onward, sustaining elimination capacities of 232 ± 10 g m−3 h−1 at 3.5 g Tol m−3 and 377 ± 13 g m−3 h−1 at 10.9 g Tol m−3, which were comparable to those achieved by highly tolerant strains such as P. putida DOT T1E and P. putida F1 under identical experimental conditions. Only one type of Tol variant, harboring a TOL-like plasmid with a 38.5 kb deletion (containing the upper and meta operons for toluene biodegradation), was identified.  相似文献

BTEX catabolism interactions in a toluene-acclimatized biofilter   总被引:1,自引:0,他引:1  
BTEX substrate interactions for a toluene-acclimatized biofilter consortium were investigated. Benzene, ethylbenzene, o-xylene, m-xylene and p-xylene removal efficiencies were determined at a loading rate of 18.07 g m−3 h−1 and retention times of 0.5–3.0 min. This was also repeated for toluene in a 1:1 (m/m) ratio mixture (toluene: benzene, ethylbenzene, or xylene ) with each of the other compounds individually to obtain a final total loading of 18.07 g m−3 h−1. The results obtained were modelled using Michaelis–Menten kinetics and an explicit finite difference scheme to generate v max and K m parameters. The v max/K m ratio (a measure of the catalytic efficiency, or biodegradation capacity, of the reactor) was used to quantify substrate interactions occurring within the biofilter reactor without the need for free-cell suspended and monoculture experimentation. Toluene was found to enhance the catalytic efficiency of the reactor for p-xylene, while catabolism of all the other compounds was inhibited competitively by the presence of toluene. The toluene-acclimatized biofilter was also able to degrade all of the other BTEX compounds, even in the absence of toluene. The catalytic efficiency of the reactor for compounds other than toluene was in the order: ethylbenzene>benzene>o-xylene>m-xylene>p-xylene. The catalytic efficiency for toluene was reduced by the presence of all other tested BTEX compounds, with the greatest inhibitory effect being caused by the presence of benzene, while o-xylene and p-xylene caused the least inhibitory effect. This work illustrated that substrate interactions can be determined directly from biofilter reactor results without the need for free-cell and monoculture experimentation. Increased regulatory constraints on industrial releases of atmospheric volatile organic compounds (VOCs) have resulted in an interest in using biofilters, bioscrubbers and air/liquid membranes for treatment of vapor phase waste streams. In this report, we describe the comparison of the use of two fundamentally different types of membrane module systems that allow the rapid diffusion of vapor phase aromatics and oxygen to an active biofilm for subsequent biodegradation. One system used a commercial membrane module containing microporous polypropylene fibers while the other used a nonporous silicone tubing membrane module for the delivery of substrate (a mixture of benzene, ethylbenzene, toluene, and xylenes [BTEX]) and electron acceptor (O2). Tests of the systems under similar conditions with BTEX in the vapor feed stream showed significant performance advantages for the silicone membrane system. The average surface-area-based BTEX removal rate for the microporous membrane system over 500 h of operation was 7.88 μg h−1 cm−2 while the rate for the silicone membrane system was 23.87 μg h−1 cm−2. The percentages of BTEX removal were also consistently better in the silicone membrane system versus the microporous system. Part of the performance problem associated with the microporous membrane system appeared to be internal water condensation and possible plugging of the pores with biomass over time that could not be resolved with vapor phase backflushing. Two biofilters fed toluene-polluted air were inoculated with new fungal isolates of either Exophiala oligosperma or Paecilomyces variotii, while a third bioreactor was inoculated with a defined consortium composed of both fungi and a co-culture of a Pseudomonas strain and a Bacillus strain. Elimination capacities of 77 g m–3 h–1 and 55 g m–3 h–1 were reached in the fungal biofilters (with removal efficiencies exceeding 99%) in the case of, respectively, E. oligosperma and Paecilomyces variotii when feeding air with a relative humidity (RH) of 85%. The inoculated fungal strains remained the single dominant populations throughout the experiment. Conversely, in the biofilter inoculated with the bacterial–fungal consortium, the bacteria were gradually overgrown by the fungi, reaching a maximum elimination capacity around 77 g m–3 h–1. Determination of carbon dioxide concentrations both in batch assays and in biofiltration studies suggested the near complete mineralization of toluene. The non-linear toluene removal along the height of the biofilters resulted in local elimination capacities of up to 170 g m–3 h–1 and 94 g m–3 h–1 in the reactors inoculated, respectively, with E. oligosperma and P. variotii. Further studies with the most efficient strain, E. oligosperma, showed that the performance was highly dependent on the RH of the air and the pH of the nutrient solution. At a constant 85% RH, the maximum elimination capacity either dropped to 48.7 g m–3 h–1 or increased to 95.6 g m–3 h–1, respectively, when modifying the pH of the nutrient solution from 5.9 to either 4.5 or 7.5. The optimal conditions were 100% RH and pH 7.5, which allowed a maximum elimination capacity of 164.4 g m–3 h–1 under steady-state conditions, with near-complete toluene degradation.  相似文献

The biodegradation potential of an innovative enclosed tubular biofilm photobioreactor inoculated with a Chlorella sorokiniana strain and an acclimated activated sludge consortium was evaluated under continuous illumination and increasing pretreated (centrifuged) swine slurry loading rates. This photobioreactor configuration provided simultaneous and efficient carbon, nitrogen, and phosphorous treatment in a single-stage process at sustained nitrogen and phosphorous removals efficiencies ranging from 94% to 100% and 70–90%, respectively. Maximum total organic carbon (TOC), NH4 +, and PO4 3− removal rates of 80 ± 5 g C mr −3 day−1, 89 ± 5 g N mr −3 day−1, and 13 ± 3 g P mr −3 day−1, respectively, were recorded at the highest swine slurry loadings (TOC of 1,247 ± 62 mg L−1, N–NH4 + of 656 ± 37 mg L−1, P–PO4 3+ of 117 ± 19 mg L−1, and 7 days of hydraulic retention time). The unusual substrates diffusional pathways established within the phototrophic biofilm (photosynthetic O2 and TOC/NH4 + diffusing from opposite sides of the biofilm) allowed both the occurrence of a simultaneous denitrification/nitrification process at the highest swine slurry loading rate and the protection of microalgae from any potential inhibitory effect mediated by the combination of high pH and high NH3 concentrations. In addition, this biofilm-based photobioreactor supported efficient biomass retention (>92% of the biomass generated during the pretreated swine slurry biodegradation).  相似文献

Air artificially contaminated with increasing concentrations of benzene was treated in a laboratory scale compost-packed biofilter for 240 days with a removal efficiency of 81–100%. The bacterial community in the packing material (PM) at different heights of the biofilter was analysed every 60 days. Bacterial plate counts and ribosomal intergenic spacer analysis (RISA) of the isolated strains showed that the number of cultivable aerobic heterotrophic bacteria and the species diversity increased with benzene availability. Identification of the isolated species and the main bands in denaturing gradient gel electrophoresis (DGGE) profiles from total compost DNA during the treatment revealed that, at a relatively low volumetric benzene load (1.2≤VBL≤6.4 g m−3 PM h−1), besides low G+C Gram positive bacteria, originally present in the packing compost, bacteroidetes and β- and γ-proteobacteria became detectable in the colonising population. At the VBL value (24.8 g m−3 PM h−1) ensuring the maximum elimination capacity of the biofilter (20.1 g m−3 PM h−1), strains affiliated to the genus Rhodococcus dominated the microflora, followed by β-proteobacteria comprising the genera Bordetella and Neisseria. Under these conditions, more than 35% of the isolated strains were able to grow on benzene as the sole carbon source. Comparison of DGGE and automated RISA profiles of the total community and isolated strains showed that a complex bacterial succession occurred in the reactor in response to the increasing concentrations of the pollutant and that cultivable bacteria played a major role in benzene degradation under the adopted conditions.  相似文献

Composting of leaves and alfalfa (i.e. active compost) was used for thebiofiltration of toluene-contaminated air in a 6-L biofilter (initial bedheight: 180 mm). During the thermophilic phase (45 to 55 °C), toluenebiodegradation rates reached 110 gtoluene.m-3.h-1 at an inlet concentration ofabout 5 g.m-3.h-1 and a gas residence time of 90 seconds. Thehighest rates were obtained late in the thermophilic phase suggesting amicrobial adaptation was occurring. Biodegradation rates decreased rapidly(50% in 48h) in the cooling stage. Under mesophilic conditions, themaximum biodegradation rates that could be obtained by increasing the inlettoluene concentration were near 89 gtoluene.m-3.h-1 which issimilar to that reported in the literature for mature compost biofilters. Novolatile by-product was detected by gas chromatography. Mineralization of14C-toluene and benzene showed that they were completelydegraded into CO2 and H2O under boththermophilic and mesophilic conditions. Bacteria isolated from latemesophilic stage had the capacity to degrade all BTEX compounds but were notable to transform chlorinated compounds. No organisms were isolated whichcould use toluene as their sole source of carbon and energy at 50 °C.Active compost biofiltration should be an excellent process for thetreatment of gaseous BTEX by biofiltration. The biotreatment of complex mixtures of volatile organic compounds (VOCs) such as benzene, toluene, ethylbenzene, and xylene isomers (BTEX) has been investigated by many workers. However, the majority of the work has dealt with the treatment of aqueous or soil phase contamination. The biological treatment of gas and vapor phase sources of VOC wastes has recently received attention with increased usage of biofilters and bioscrubbers. Although these systems are relatively inexpensive, performance problems associated with biomass plugging, gas channeling, and support media acidification have limited their adoption. In this report we describe the development and evaluation of an alternative biotreatment system that allows rapid diffusion of both BTEX and oxygen through a silicone membrane to an active biofilm. The bioreactor system has a rapid liquid recycle, which facilitates nutrient medium mixing over the biofilm and allows for removal of sloughing cell mass. The system removed BTEX at rates up to 30 μg h−1 cm−2 of membrane area. BTEX removal efficiencies ranged from 75% to 99% depending on the BTEX concentration and vapor flowrate. Consequently, the system can be used for continuous removal and destruction of BTEX and other potential target VOCs in vapor phase streams. The composite filler micro-embedded with Pseudomonas putida (P. putida) was prepared and the biodegradation performance of the filler was evaluated in a biofilter. Five phases were set up to evaluate the performance of the biofilter under different toluene inlet loadings and transient shock loadings. In particular, the microbial community structure in the biofilms and fillers was measured by sequence analysis of the 16S rRNA gene. The results show that the biofilter packed with the composite fillers was suitable for the biodegradation of toluene. The biofilter could start up quickly with high removal efficiency (RE), and remain above 90 % RE when the empty bed residence time (EBRT) was 18 s and the inlet loading rates were not higher than 41.4 g/(m3·h). Moreover, the biofilter could tolerate substantial transient shock loadings. The high removal efficiency and elimination capacity contributed to rich bacterial communities for the efficient degradation of toluene. The dominant microbial communities at the phylum level were mainly Firmicutes, Actinobacteria and Proteobacteria. It is noteworthy that the abundance of Bacteroidetes at phylum level and Chungangia and Stenotrophomonas at genus level increased significantly during the re-start period.  相似文献

A stable microbial consortium that grew on nitrobenzene (NB) as its sole source of carbon, nitrogen and energy and liberated excess nitrogen as ammonia, was immobilized on a perlite-packed trickling air biofilter. On a sustained basis, the biofilter removed 50 g NB m−3 packing h−1 and its operation at pH 8.7 resulted in ammonia stripping, making pH and salinity controls unnecessary. Low maintenance and stable performance during 4 months of continuous operation invite the scale-up of this biofilter for control of NB emissions. The bacterial community structure in a biofilter treating ethanol was investigated using community level physiological profiling. Laboratory scale biofilters of two sizes (5 or 11.5 cm internal diameter with 30 or 67 cm packed height, respectively) were packed with compost and a humidified airstream loaded with ethanol passed through them. Good removal efficiencies (82–100%) and elimination capacities (49–205 g ethanol m − 3 h − 1) were observed in all units. Compost packing media samples were extracted and the community level physiological profiles assayed using Biolog Ecoplates. The community structure was found to be similar over a range of a few centimetres. No differences were observed between sample sizes of 0.5–1 and 6 g, and therefore, the smaller sample size (typical of that used in previous studies) is appropriate for use in the future. Two studies of parallel systems showed that the community structure diverged during the acclimation period (10 days) in one pair, but in another pair, no divergence was observed and a similar shift in community profile was observed in both units between 25 and 40 days of operation. Community level physiological profiling with Biolog Ecoplates is a useful method for detecting differences between and changes within the bacterial communities in biofilters.  相似文献

This study reports the biodegradation of carbon disulfide (CS2) in air biofilters packed with a pelletized mixture of composted manure and sawdust. Experiments were carried out in two lab-scale (1.2 L) biofiltration units. Biofilter B was seeded with activated sludge enriched previously on CS2-degrading biomass under batch conditions, while biofilter A was left as a negative inoculation control. This inoculum was characterized by an acidic pH and sulfate accumulation, and contained Achromobacter xylosoxidans as the main putative CS2 biodegrading bacterium. Biofilter operation start-up was unsuccessfully attempted under xerophilic conditions and significant CS2 elimination was only achieved in biofilter A upon the implementation of an intermittent irrigation regime. Sustained removal efficiencies of 90–100 % at an inlet load of up to 12 g CS2 m?3 h?1 were reached. The CS2 removal in this biofilter was linked to the presence of the chemolithoautotrophic bacterium Thiobacillus thioparus, known among the relatively small number of species with a reported capacity of growing on CS2 as the sole energy source. DGGE molecular profiles confirmed that this microbe had become dominant in biofilter A while it was not detected in samples from biofilter B. Conventional biofilters packed with inexpensive organic materials are suited for the treatment of low-strength CS2 polluted gases (IL <12 g CS2 m?3 h?1), provided that the development of the adequate microorganisms is favored, either upon enrichment or by inoculation. The importance of applying culture-independent techniques for microbial community analysis as a diagnostic tool in the biofiltration of recalcitrant compounds has been highlighted.  相似文献
