In the summer of 2003, a microalga strain was isolated from a massive green microalgae bloom in wastewater stabilization ponds at the treatment facility of La Paz, B.C.S., Mexico. Prevailing environmental conditions were air temperatures over 40 degrees C, water temperature of 37 degrees C, and insolation of up to 2400 micromol m2 s(-1) at midday for several hours at the water surface for four months. The microalga was identified as Chlorella sorokiniana Shih. et Krauss, based on sequencing its entire 18S rRNA gene. In a controlled photo-bioreactor, this strain can grow to high population densities in synthetic wastewater at temperatures of 40-42 degrees C and light intensity of 2500 micromol m2 s(-1) for 5h daily and efficiently remove ammonium from the wastewater under these conditions better than under normal lower temperature (28 degrees C) and lower light intensity (60 micromol m2 s(-1)). When co-immobilized with the bacterium Azospirillum brasilense that promotes growth of microalgae, the population of microalga grew faster and removed even more ammonium. Under exposure to extreme growth conditions, the quantity of four photosynthetic pigments increased in the co-immobilized cultures. This strain of microalga has potential as a wastewater treatment agent under extreme conditions of temperature and light intensity.  相似文献

Chlorella vulgaris, a microalga often used in wastewater treatment, was coimmobilized and coincubated either with the plant growth-promoting bacterium Azospirillum brasilense, or with its natural associative bacterium Phyllobacterium myrsinacearum, in alginate beads designed for advanced wastewater treatment. Interactions between the microalga and each of the bacterial species were followed using transmission electron microscopy for 10 days. Initially, most of the small cavities within the beads were colonized by microcolonies of only one microorganism, regardless of the bacterial species cocultured with the microalga. Subsequently, the bacterial and microalgal microcolonies merged to form large, mixed colonies within the cavities. At this stage, the effect of bacterial association with the microalga differed depending on the bacterium present. Though the microalga entered a senescence phase in the presence of P. myrsinacearum, it remained in a growth phase in the presence of A. brasilense. This study suggests that there are commensal interactions between the microalga and the two plant associative bacteria, and that with time the bacterial species determined whether the outcome for the microalga is senescence or continuous multiplication.  相似文献

Growth of and the capacity to take up nitrogen in the freshwater microalgae Chlorella vulgaris were studied while varying the concentrations of ammonium and nitrate, the pH and the source of carbon in a synthetic wastewater growth medium when co-immobilized in alginate beads with the microalgae growth-promoting bacterium Azospirillum brasilense. Analyses of 29 independent experiments showed that co-immobilization of the microalgae with A. brasilense could result in two independent phenomena directly affected by cultivation factors, such as nitrogen species, pH and presence of a carbon source. First, growth of the microalgal population increased without an increase in the capacity of the single cells to take up nitrogen, or second, the capacity of cells to take up nitrogen increased without an increase of the total microalgal population. These phenomena were dependent on the population density of the microalgae, which was in turn affected by cultivation factors. This supports the conclusion that the size of the microalgal population controls the uptake of nitrogen in C. vulgaris cells - the higher the population (regardless the experimental parameters), the less nitrogen each cell takes up.  相似文献

The effect of the microalgae-growth promoting bacterium Azospirillum brasilense on accumulation of total carbohydrates and starch in two species of Chlorella (Chlorella vulgaris and Chlorella sorokiniana), when the bacterium and each microalga were jointly immobilized in alginate beads was studied under autotrophic conditions for 144h in synthetic medium. The interaction of the bacterium with the microalgae enhanced accumulation of total carbohydrate and starch. Cells of Chlorella accumulated the highest amounts of carbohydrate after incubation for 24h. Yet, this did not coincide with the highest affinity and volumetric productivity measured in these cultures. However, after incubation for 72h, mainly in jointly immobilized treatments of both microalgae species, the cultures reached their highest total carbohydrate content (mainly as starch) and also the highest affinity and volumetric productivity. These results demonstrate the potential of A. brasilense to affect carbohydrates and starch accumulation in Chlorella spp. when both microorganisms are co-cultured, which can be an important tool for applications of microalgae.  相似文献

This study evaluates the feasibility of removing nutrients by the microalgae Chlorella vulgaris, using urban wastewater as culture medium, namely the effluent subjected to secondary biological treatment in a wastewater treatment plant (WWTP). For this, laboratory experiments were performed in batch cultures to study the effect of initial nitrogen and phosphorus concentrations on growth and reduction of nutrient performance of C. vulgaris. The microalga was cultivated in enriched wastewater containing different phosphorus (1.3-143.5 mg x L(-1) P.PO4(3-)), ammonium (5.8-226.8 mg x L(-1) N-NH4+) and nitrate (1.5-198.3 mg x L(-1) N-NO3-) concentrations. The nutrient removal and growth kinetics have been studied: maximum productivity of 0.95 g SS x L(-1) x day(-1), minimum yield factor for cells on substrate (Y) of 11.51 g cells x g nitrogen(-1) and 0.04 g cells x g phosphorus(-1) were observed. The results suggested that C. vulgaris has a high potential to reduce nutrients in secondary WWTP effluents.  相似文献

Three strains of the freshwater microalgae used for wastewater treatment, Chlorella vulgaris and Chlorella sorokiniana co-immobilized separately in alginate beads with the microalgae-growth-promoting bacterium Azospirillum brasilense Cd, resulted in significant changes in microalgal-population size, cell size, cell cytology, pigment, lipid content, and the variety of fatty acids produced in comparison with microalgae immobilized in alginate without the bacterium. Cells of C. vulgaris UTEX 2714 did not change in size, but the population size within the beads significantly increased. On the other hand, C. vulgaris UTEX 395 cells grew 62% larger, but their numbers did not increase. The population of C. sorokiniana UTEX 1602 increased, but not their cell size. The content of pigments chlorophyll a and b, lutein, and violoaxanthin increased in all microalgal species. The lipid content also significantly increased in all three strains, and the number of different fatty acids in the microalgae increased from four to eight. This study indicates that the microalgae-growth-promoting bacterium induced significant changes in the metabolism of the microalgae.  相似文献

Coimmobilization of the freshwater microalga Chlorella vulgaris and the plant-growth-promoting bacterium Azospirillum brasilense in small alginate beads resulted in a significantly increased growth of the microalga. Dry and fresh weight, total number of cells, size of the microalgal clusters (colonies) within the bead, number of microalgal cells per cluster, and the levels of microalgal pigments significantly increased. Light microscopy revealed that both microorganisms colonized the same cavities inside the beads, though the microalgae tended to concentrate in the more aerated periphery while the bacteria colonized the entire bead. The effect of indole-3-acetic acid addition to microalgal culture prior to immobilization of microorganisms in alginate beads partially imitated the effect of A. brasilense. We propose that coimmobilization of microalgae and plant-growth-promoting bacteria is an effective means of increasing microalgal populations within confined environments.  相似文献

Heterotrophic growth of microalgae presents significant economic advantages over the more common autotrophic cultivation. The efficiency of growth and nitrogen, phosphorus, and glucose uptake from synthetic wastewater was compared under heterotrophic, autotrophic, and mixotrophic regimes of Chlorella vulgaris Beij. immobilized in alginate beads, either alone or with the bacterium Azospirillum brasilense. Heterotrophic cultivation of C. vulgaris growing alone was superior to autotrophic cultivation. The added bacteria enhanced growth only under autotrophic and mixotrophic cultivations. Uptake of ammonium by the culture, yield of cells per ammonium unit, and total volumetric productivity of the culture were the highest under heterotrophic conditions when the microalga grew without the bacterium. Uptake of phosphate was higher under autotrophic conditions and similar under the other two regimes. Positive influence of the addition of A. brasilense was found only when light was supplied (autotrophic and mixotrophic), where affinity to phosphate and yield per phosphate unit were the highest under heterotrophic conditions. The pH of the culture was significantly reduced in all regimes where glucose was consumed, similarly in heterotrophic and mixotrophic cultures. It was concluded that the heterotrophic regime, using glucose, is superior to autotrophic and mixotrophic regimes for the uptake of ammonium and phosphate. Addition of A. brasilense positively affects the nutrient uptake only in the two regimes supplied with light.  相似文献

通过设置绿球藻(Chlorococcum sphacosum GD)的起始接种浓度(25—400 mg/L),研究其对水产养殖废水的处理效果及藻细胞的生长特性。研究结果表明,起始接种浓度为100 mg/L的绿球藻藻液,其生长特性最佳,比生长速率最大,倍增时间最短。随着起始接种浓度的增加,生长速率逐渐降低,倍增时间逐渐增加。在起始接种浓度为100 mg/L的条件下,在5d的培养周期内,绿球藻能够去除水产养殖废水中96.92%的COD、98.08%的氨氮、98.67%的亚硝氮、91.42%的硝氮及98.36%的总磷。低起始接种浓度(尤其是100 mg/L)有利于绿球藻的生长和污染物降解。研究初步探明了微藻起始接种浓度对水产养殖废水处理效果的影响。通过控制微藻接种浓度有望在提高污染物去除率的同时缩短培养周期并提高容积负荷,为今后微藻用于大规模水产养殖废水的处理提供了一定的理论支持。  相似文献

Heterotrophic growth of the microalga Chlorella vulgaris Beij. in synthetic as well as sterilized municipal wastewater of a nonindustrialized city was measured. The city wastewater contained high levels of ammonium and nitrate, medium levels of phosphate, and low levels of nitrite and organic molecules and could not support heterotrophic growth of C. vulgaris. Evaluation of 11 known carbon sources for this microalga that were added to standard synthetic wastewater containing the same levels of nitrogen and phosphorus as the municipal wastewater revealed that the best carbon sources for heterotrophic growth were Na‐acetate and d ‐glucose. These provided the highest growth rates and the largest removal of ammonium. Growth increased with concentration of the supplement to an optimum at 0.12 M Na‐acetate. This carbon source was consumed completely within 10 d of incubation. Higher concentrations inhibited the growth of C. vulgaris. The microalgal populations under heterotrophic growth conditions were one level of magnitude higher than that under autotrophic growth conditions that served as a comparison. No growth occurred in the dark in the absence of a carbon source. Na‐acetate was superior to d ‐glucose. In municipal wastewater, when Na‐acetate or d ‐glucose was added, C. vulgaris significantly enhanced ammonium removal under heterotrophic conditions, and its capacity was equal to ammonium removal under autotrophic growth conditions. This study showed that sterilized wastewater can be treated by C. vulgaris under heterotrophic conditions if supplemented with the appropriate organic carbon source for the microalgae.  相似文献

The ammonia and phosphorus removal efficiencies of the microalgae Chlorella vulgaris and Scenedesmus dimorphus, during biotreatment of secondary effluent from an agroindustrial wastewater of a dairy industry and pig farming, were evaluated. The microalgae were isolated from a wastewater stabilization pond near Santafé de Bogotá, Colombia. Batch cultures were made using both species in 4-1 cylindrical glass bioreactors each containing 2l of culture. Chlorella vulgaris was also cultivated on wastewater in a triangular bioreactor. Three 216-h experimental cycles were run for each microalga and in each bioreactor. In the cylindrical bioreactor, S. dimorphus was more efficient in removing ammonia than C. vulgaris. However, the final efficiency of both microalgae at the end of each cycle was similar. Both microalgae removed phosphorus from the wastewater to the same extent in a cylindrical bioreactor. Using C. vulgaris, the triangular bioreactor was superior for removing ammonia and the cylindrical bioreactor was superior for removing phosphorus. This study shows the potential of using these microalgae to reduce the environmental pollution of heavily contaminated agroindustrial waters currently disposed of untreated into the waterways and streams of tropical Colombia.  相似文献

Genetic transformation is useful for basic research and applied biotechnology. However, genetic transformation of microalgae is usually quite difficult due to the technical limitations of existing methods. We cloned the promoter and terminator of the nitrate reductase gene from the microalga Phaeodactylum tricornutum and used them for optimization of a transformation system of the microalga Chlorella vulgaris. This species has been used for food production and is a promising candidate as a bioreactor for large-scale production of value-added proteins. A construct was made containing the CAT (chloramphenicol acetyltransferase) reporter gene driven by the nitrate reductase promoter. This construct was transferred into the C. vulgaris genome by electroporation. Expression of CAT in transgenic Chlorella conferred resistance to the antibiotic chloramphenicol and enabled growth in selective media. Overall efficiency for the transformation was estimated to be approximately 0.03%, which is relatively high compared with other available Chlorella transformation systems. Expression of CAT was induced in the presence of nitrate and inhibited in the presence of ammonium as a sole nitrogen source. This study presented an inducible recombinant gene expression system, also providing more gene regulation elements with potential for biotechnological applications.  相似文献

The effect of the bacterium Azospirillum brasilense jointly immobilized with Chlorella vulgaris or C. sorokiniana in alginate beads on total carbohydrates and starch was studied under dark and heterotrophic conditions for 144h in synthetic growth medium supplemented with either d-glucose or Na-acetate as carbon sources. In all treatments, enhanced total carbohydrates and starch content per culture and per cell was obtained after 24h; only jointly immobilized C. vulgaris growing on d-glucose significantly increased total carbohydrates and starch content after 96h. Enhanced accumulation of carbohydrate and starch under jointly immobilized conditions was variable with time of sampling and substrate used. Similar results occurred when the microalgae was immobilized alone. In both microalgae growing on either carbon sources, the bacterium promoted accumulation of carbohydrates and starch; when the microalgae were immobilized alone, they used the carbon sources for cell multiplication. In jointly immobilized conditions with Chlorella spp., affinity to carbon source and volumetric productivity and yield were higher than when Chlorella spp. were immobilized alone; however, the growth rate was higher in microalgae immobilized alone. This study demonstrates that under heterotrophic conditions, A. brasilense promotes the accumulation of carbohydrates in two strains Chlorella spp. under certain time-substrate combinations, producing mainly starch. As such, this bacterium is a biological factor that can change the composition of compounds in microalgae in dark, heterotrophic conditions.  相似文献

In synthetic wastewater, growth and phosphorus absorption by two species of microalgae, Chlorella sorokiniana and Chlorella vulgaris, and in domestic wastewater by C. sorokiniana significantly enhanced after a starvation period of 3 days in saline solution, combined with co-immobilization with the microalgae growth-promoting bacterium (MGPB) Azospirillum brasilense Cd in alginate beads. Starvation of 5 days negatively affected the subsequent growth of C. vulgaris, but not of C. sorokiniana in fresh wastewater. Starvation of immobilized cultures of microalgae separately or microalgae with bacteria, followed by returning the immobilized cultures to the same wastewater did not enhance phosphorus absorption. However, a starvation period followed by subsequent submersion of the cultures in fresh wastewater allowed the continuation of phosphorus absorption. The best phosphorus removal treatment from a batch of synthetic or domestic wastewater was with tandem treatments of wastewater treatment with pre-starved, co-immobilized microalgae and replacement of this culture, after one cycle of phosphorus removal, with a new, similarly starved culture. This combination treatment with two cultures was capable of removing up to 72% of phosphorus from the wastewater. There was a direct correlation between the initial load of phosphorus in the domestic wastewater and the efficiency level of removal, being highest at higher phosphorus loads in co-immobilized cultures. This occurred for both immobilized and co-immobilized cultures. Further, the results showed that negative effects of starving the microalgae were mitigated by the application of the MGPB A. brasilense Cd. This is the first report of this capacity in Azospirillum sp. on a single-cell plant. This study showed that starvation periods, combined with co-immobilization with MGPB, have synergistic effects on absorption of phosphorus from wastewater and merits consideration in designing future biological treatments of wastewater.  相似文献

Four green microalgae (Chlorella sorokiniana, Chlorella vulgaris, Scenedesmus obliquus and Selenastrum capricornutum), a wild Bolivian microalga strain and two cyanobacteria (Anabaena catenula and Microcystis aeruginosa) were compared for tolerance to salicylate, O2 production capacity and ability to support salicylate degradation by a Ralstonia basilensis strain in symbiotic microcosms with the microalgae. Microcystis aeruginosa had the highest tolerance to salicylate at 500 mg l(-1) and 1500 mg l(-1) but only produced 0.7 mg O2 l(-1) h(-1) in the absence of pollutant. Chlorella sorokiniana resisted salicylate at 1500 mg l(-1) with the highest O2 production in the absence of salicylate (26 mg l(-1) h(-1)) closely followed by the Bolivian microalga (23 mg l(-1) h(-1)) and Chlorella vulgaris (21 mg l(-1) h(-1)). Selenastrum capricornutum and Anabaena catenula were completely inhibited by salicylate at 500 mg l(-1). When inoculated with Ralstonia sp. and supplied with salicylate, Chlorella sorokiniana had the highest removal rate (19 mg l(-1) h(-1)), followed by the wild Bolivian strain (18 mg l(-1) h(-1)) and Chlorella vulgaris (14 mg l(-1) h(-1)).  相似文献

Anaerobic digestion of microalgae is hampered by its complex cell wall. Against this background, cyanobacteria cell walls render this biomass as an ideal substrate for overcoming this drawback. The aim of the present study was to compare the growth of two cyanobacteria (Aphanizomenon ovalisporum and Anabaena planctonica) and a microalga (Chlorella vulgaris) in urban wastewater when varying the temperature (22, 27 and 32 °C). Cyanobacterial optimal growth for both strains was attained at 22 °C, while C. vulgaris did not show remarkable differences among temperatures. For all the microorganisms, ammonium removal was higher than phosphate. Biomass collected was subjected to anaerobic digestion. Methane yield of C. vulgaris was 184.8 mL CH4 g COD in?1 while with A. ovalisporum and A. planctonica the methane production was 1.2- and 1.4-fold higher. This study showed that cyanobacteria growth rates could be comparable to microalgae while presenting the additional benefit of an increased anaerobic digestibility.  相似文献

微藻是可广泛用于健康食品及水产养殖的饵料,同时,微藻细胞内积累的油脂可作为可再生生物燃料,因此微藻的生长和代谢受到广泛关注。温度和pH对微藻的生物量积累有很大影响,考察不同温度和pH条件下微藻细胞的生长有助于寻找最佳的条件进行微藻的培养。自絮凝小球藻JSC-7(Chlorella vulgaris JSC-7)可实现自沉降采收,有利于降低微藻生产成本,优化其生长条件对更好地利用该微藻具有重要意义。考察了温度(22∽40℃)及pH(6.0∽10.0)对其细胞生长、叶绿素含量和油脂产量的影响。在所选取的温度及pH范围内,JSC-7细胞均可生长,显示该藻种可以适应广泛的温度和pH条件。适合细胞生长的温度依次为31℃〉28℃〉35℃〉25℃,pH依次为7.0〉8.0〉6.0。pH 8.0时生物量和油脂的积累量最多,说明该藻株在弱碱条件下更适合生长和产油。当温度为31℃、pH为7.0时,可获得最高的生长量(OD690=0.941)、叶绿素含量(19 mg/L)及油脂产量(39.07%/克干重)。  相似文献

The feasibility of using a microalga Chlorella vulgaris YSW-04 was investigated for removal of nutrients from piggery wastewater effluent. The consequent lipid production by the microalga was also identified and quantitatively determined. The wastewater effluent was diluted to different concentrations ranging from 20 to 80 % of the original using either synthetic media or distilled water. The dilution effect on both lipid production and nutrient removal was evaluated, and growth rate of C. vulgaris was also monitored. Dilution of the wastewater effluent improved microalgal growth, lipid productivity, and nutrient removal. The growth rate of C. vulgaris was increased with decreased concentration of piggery wastewater in the culture media regardless of the diluent type. Lipid production was relatively higher when using synthetic media than using distilled water for dilution of wastewater. The composition of fatty acids accumulated in microalgal biomass was dependent upon both dilution ratio and diluent type. The microalga grown on a 20 % concentration of wastewater effluent diluted with distilled water was more promising for generating high-efficient biodiesel compared to the other culture conditions. The highest removal of inorganic nutrients was also achieved at the same dilution condition. Our results revealed the optimal pretreatment condition for the biodegradation of piggery wastewater with microalgae for subsequent production of high-efficient biodiesel.  相似文献

Microalgae cultures are receiving attention because of increasing biotechnological and biomedical production of active biomolecules. We evaluated various fertilizer-based culture media to scale up production of the marine microalga Phaeodactylum tricornutum for production of exocellular polysaccharides (EPS), soluble proteins, and cellular superoxide dismutase (SOD). The standard source of sodium nitrate was the same as that used in the synthetic f/2 culture medium and ammonium nitrate, urea, ammonium sulfate, and calcium nitrate as alternative sources of nitrogen. The maximum production of EPS was achieved in microalgae cells grown in the culture media containing 63 and 23% nitrogen from ammonium sulfate, and also in microalgae cells grown in the culture media containing 3% nitrogen from ammonium nitrate. The maximum production of cellular SOD was achieved in microalgae cells grown in the culture media containing 35 and 26% nitrogen from ammonium sulfate, and in the culture media containing 17% nitrogen from urea. The results suggest that it is possible to use a source of nitrogen, other than sodium nitrate, to scale up growth of P. tricornutum for production of EPS and SOD at reduced costs.  相似文献
