The light-dependent modulation of ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) activity was studied in two species: Phaseolus vulgaris L., which has high levels of the inhibitor of Rubisco activity, carboxyarabinitol 1-phosphate (CA1P), in the dark, and Chenopodium album L., which has little CA1P. In both species, the ratio of initial to fully-activated Rubisco activity declined by 40–50% within 60 min of a reduction in light from high a photosynthetic photon flux density (PPFD; >700 mol · m–2 · s–1) to a low PPFD (65 ± 15 mol · m–2 · s–1) or to darkness, indicating that decarbamylation of Rubisco is substantially involved in the initial regulatory response of Rubisco to a reduction in PPFD, even in species with potentially extensive CA1P inhibition. Total Rubisco activity was unaffected by PPFD in C. album, and prolonged exposure (2–6 h) to low light or darkness was accompanied by a slow decline in the activity ratio of this species. This indicates that the carbamylation state of Rubisco from C. album gradually declines for hours after the large initial drop in the first 60 min following light reduction. In P. vulgaris, the total activity of Rubisco declined by 10–30% within 1 h after a reduction in PPFD to below 100 mol · m–2 · s–1, indicating CA1P-binding contributes significantly to the reduction of Rubisco capacity during this period, but to a lesser extent than decarbamylation. With continued exposure of P. vulgaris leaves to very low PPFDs (< 30 mol · m–2 · s–1), the total activity of Rubisco declined steadily so that after 6–6.5 h of exposure to very low light or darkness, it was only 10–20% of the high-light value. These results indicate that while decarbamylation is more prominent in the initial regulatory response of Rubisco to a reduction in PPFD in P. vulgaris, binding of CA1P increases over time and after a few hours dominates the regulation of Rubisco activity in darkness and at very low PPFDs.Abbreviations CA1P 2-carboxyarabinitol 1-phosphate - CABP 2-carboxyarabinitol 1,5-bisphosphate - kcat substrate-saturated turnover rate of fully carbamylated enzyme - PPFD photosynthetically active photon flux density (400–700 nm) - Rubisco ribulose-1,5-bisphosphate carboxylase/oxygenase - RuBP ribulose-1,5-bisphosphate  相似文献

Effects of iloprost, which is a stable prostacyclin analogue, on the ischemic myocardium were examined in the open-chest dog heart, in terms of biochemical parameters. Ischemia was initiated by ligating the left anterior descending coronary artery. When the coronary artery was ligated for 3 min, the levels or glycogen, fructose-1,6-diphosphate (FDP), adenosine triphosphate and creatinephosphate decreased, and the levels of glucose-6-phosphate (G6P), fructose-6-phosphate (F6P), lactate, adenosine diphosphate and adenosine monophosphate increased. During ischemia, therefore, energy charge potential was significantly decreased from 0.89±0.01 to 0.82±0.01, and ([G6P]+[F6P])/[FDP] and [lactate]/[pyruvate] ratios were significantly increased from 1.75±0.30 to 29.05±5.70 and 13±3 to 393±112, respectively. Iloprost (0.1, 0.3, or 1 g·kg–1) was injected intravenously 5 min before the onset of ischemia. Iloprost (0.1, 0.3, and 1 g·kg–1) reduced the ischemia-induced decrease in energy charge potential to 94, 74, and 86%, respectively, the increase in ([G6P]+[F6P]/[FDP] to 38, 29, 32%, respectively, and the increase in [lactate]/[pyruvate] to 67, 45, 65%, respectively. These results suggest that iloprost lessens the myocardial metabolic derangements produced by ischemia, and the most potent effect was obtained at the dose of 0.3 g·kg–1.  相似文献

Synopsis Embryos in specific stage of the estuarine teleost, Fundulus heteroclitus, were exposed to mercuric chloride (MC) and methylmercuric chloride (MMC) under several distinct treatment conditions. Four-eight cell stage eggs (0-day old) were exposed for 4 days (continuous), 2 days and one day to each mercury compound. One-day old (mid-blastula), 2-day old (mid-neurula) and 5-day old (beating heart) embryos were exposed 4 days to MC and MMC. Mortality for the four days immediately following the initiation of exposure was the embryonic response measured. Under most exposure conditions to the 4–8 cell eggs, progressive and significant reductions in survival were observed at all concentrations above 40 and 30 gHg++l–1 as MC and MMC, respectively. Reducing the duration of exposure to 1 day most significantly increased the survival potential of the 4–8 cell eggs. For all exposure treatments to the 4–8 cell eggs, significant differences in survival, between eggs exposed to MC and MMC, were determined at 40, 60 and 80 gHg++l–1, indicating the presence of compound-dependent response differences. In all cases demonstrating response differences between MC and MMC exposed embryos, survival was significantly lower following exposure to MMC. Survival of embryos was progressively increased when the initiation of continuous exposure (4 days) was delayed 1, 2 and 5 days after fertilization. As a result, compound-dependent response differences were progressively shifted to higher He++ concentrations. For both MC and MMC, survival of 1-day old embryos exposed for 4 days was greater than that of 0-day old eggs exposed for 1 day. Of the embryonic stages examined, it appears that the earlier cleavage stages are the most sensitive to mercury intoxication.  相似文献

Summary Intermoult crayfish (Astacus astacus) were exposed to acid (pH 4), soft water ([Ca++]=100 mol·l–1) in the absence and presence of aluminium (25 mol·l–1) for variable time periods (up to 21 days) in order to assess the consequences for acid-base and electrolyte balance and haemolymph gas transport. Haemolymph osmolality and concentration of major ions decreased drastically and to a similar extent in acid and acid-aluminium water. Muscle tissue ion concentrations were, however, regulated at an almost constant level. A severe metabolic acidosis was gradually developed, attaining a haemolymph metabolic acid load of 6–7 mequiv·l–1 after 12–21 days. The acidosis was partially compensated by ventilatory means, with the postbranchial haemolymph PCO2 decreasing earlier in acidaluminium-exposed than in acid-exposed specimens. Hyperventilation seemed to be a direct acid-base regulatory response, since the rise in postbranchial PCO2 had only minimal influence on haemolymph O2 transport. The Bohr effect of Astacus astacus haemocyanin was low (log P50/GdpH=-0.24), and the mean P50 only increased from 15 to 19 mmHg after 21 days of acid exposure. The decrease in O2 affinity with decreasing pH was accompanied by a decrease in the cooperativity of O2 binding. The haemolymph haemocyanin concentration was not affected by acid and acid-aluminium exposure, but decreased after 21 days due to starvation. Muscle tissue aluminium concentrations were unaffected, whereas gill tissue concentrations increased in acid-aluminium exposed crayfish, most likely due to accumulation of aluminium on the gill surface. Mortality was low, and an internal hypoxia and lactacidosis was not developed in either of the experimental groups. This suggests that the gas transfer qualities of the chitincovered gills of crayfish are much less sensitive to acid and acid-aluminium stress than the gills of teleost fish.Abbreviations Hc haemocyanin - SO2 saturation of Hc with O2 - P 50 oxygen tension of haemolymph at 50% SO2 - n 50 Hills coelficient around 50% SO2  相似文献

Concentrations of cadmium, mercury, lead, arsenic, selenium, copper, zinc, manganese and iron in liver, and cadmium in kidneys, were analysed in 95 carcasses of arctic fox (Alopex lagopus) caught in Svalbard during three winter seasons from 1984 through 1986. The hepatic concentration ranges of cadmium, mercury, lead and arsenic were 0.1–2.4, 0.01–2.2, < 0.5–2.9 and 0.01–1.3 g·g–1 WW, respectively. The range of cadmium concentration in the kidneys was from 0.2 to 13 g·g–1 WW. Cadmium and mercury concentrations were higher in adult animals than in juveniles. The average concentrations of cadmium and lead were similar to recently published levels in polar bear from Svalbard, but the mercury concentrations were lower. Significant geographical differences were observed between trapping areas. Foxes caught north of Isfjorden had lower levels of liver iron and higher levels of all other elements analysed than those caught south of Isfjorden. The recorded concentrations of heavy metals indicate a moderate degree of exposure, which most likely is of natural origin.Gunnar Norheim died January 9, 1991  相似文献

In Artemia franciscana, enriched for 24 h with emulsified lipids, thefatty acid DHA (22:63) is unstable. In rotifers the level of DHA canbe stabilised by adding algae rich in DHA to the fish tanks. To test thismethod for A. franciscana, enriched A. franciscana was incubated with thealga Isochrysis galbana, which is rich in lipids and DHA, at sixconcentrations ranging from 1 to 20 mg C l–1 at 13–14°C. After lipid enrichment, A. franciscana contained15% lipids of which 79% (116 mg g DW–1) werefatty acids. Of the fatty acids, 17% was DHA (19.8 mg gDW–1). After 72 hours incubation with I. galbana, the level ofDHA decreased to 1.6–3.2 mg g DW–1, which was areduction by 84–92%. This was independent of the algalconcentration. The amount of total fatty acids decreased to 53–73 mg gDW–1, a reduction by 37–54%, independent of thealgal concentration. The rates of loss of the fatty acids, and especiallyDHA, was greater during the first 24 h at the highest algal concentrations(8, 10 and 20 mg C l–1). Enriched A. franciscana shouldtherefore not be incubated with high algal concentrations (>6 mg Cl–1) during first feeding of marine fish larvae.  相似文献

Zinc, lead and mercury accumulation in the amphipod Hyalella azteca increases with increasing exposure to metals. During 10 week chronic toxicity tests, metal accumulated at the highest non-toxic/lowest toxic concentration was 126/136 µg Zn g–1, 7.1/16 µg Pb g–1 and 56/90 µg Hg g–1 dry weight. Concentrations of lead and mercyry in control animals were substantially lower (1.3 µg Pb g–1 and 0.4 µg Hg g–1), but concentrations of zinc in controls (74 µg g–1) were about one half those of the lowest toxic concentration. Copper was completely regulated. Accumulated copper concentrations after 10 weeks exposure to all waterborne copper concentrations resulting in less than 100% mortality were not significantly different from controls (79 µg g–1). Lead and mercury concentrations in wild H. azteca should be useful indicators of potential toxicity. Zinc accumulation may also be a useful indicator of zinc toxicity, but careful comparison with control or reference animals is necessary because of the small differences between toxic and control concentrations. Copper is not accumulated by H. azteca under chronic exposure conditions and body burdens of field animals cannot be used as an indicator of exposure or potential toxic effects. Short term exposures to copper, however, result in elevated copper concentrations in H. azteca, even at concentrations below those causing chronic toxicity. Short term bioaccumulation studies might, therefore, provide a useful indication of potential chronic copper toxicity.  相似文献

Background

Endothelial dysfunction is involved in the pathogenesis of atherosclerosis. Consumption of fish is associated with reduced cardiovascular risk, but there is paucity of data concerning its effect on endothelial function. Furthermore, investigation of the effects of fish consumption on health must take into account the ingestion of contaminants, including transition metals and some metalloids, which may have unfavorable effects on health, including those on the cardiovascular system. We investigated the association between fish consumption, endothelial function (flow mediated dilation of the brachial artery), and serum concentration of some toxic metals in apparently healthy people.

Methods

Twenty-nine high fish consumers (at least 3 portions a week) were compared with 25 low fish consumers (less than 1 portion a week). All participants were free of diabetes, cardiovascular or other systemic diseases. Serum metal (antimonium, arsenic, mercury, lead, cobalt, copper, zinc, selenium, strontium) concentrations were measured in subgroups of 24 high fish consumers and 19 low fish consumers.

Results

Both groups exhibited similar habitual dietary patterns, age and anthropometric characteristics. The high fish consumers had higher flow mediated dilation (9.7±1.8 vs. 7.3±1.9%; P<0.001), but also higher serum concentrations of mercury (5.87±2.69 vs. 1.65±1.10 mcg/L; P<0.001) and arsenic (6.04±3.25 vs. 2.30±1.58 mcg/L; P<0.001). The fasting plasma glucose concentrations were significantly correlated with both mercury (r = 0.39; P = 0.01) and arsenic concentrations (r = 0.55; P<0.001).

Conclusions

Habitual consumption of high amounts of fish is associated with better endothelial function despite higher serum concentrations of mercury and arsenic.  相似文献

A family 19 chitinase (OsChia1c, class I) from rice, Oryza sativa L., and its chitin-binding domain-truncated mutant (OsChia1cCBD, class II) were produced by the Pichia expression system, and the hydrolytic mechanism toward N-acetylglucosamine hexasaccharide [(GlcNAc)6] was investigated by HPLC analysis of the reaction products. The profile of the time-course of (GlcNAc)6 degradation obtained by OsChia1c was identical to that obtained by OsChia1cCBD, indicating that the chitin-binding domain does not significantly participate in oligosaccharide hydrolysis. From the theoretical analysis of the reaction time-course of OsChia1cCBD, the free energy changes of sugar residue binding were estimated to be –0.4, –4.7, +3.4, –0.5, –2.3, and –1.0 kcal/mol for the individual subsites of (–3), (–2), (–1), (+1), (+2), and (+3), respectively. The hexasaccharide substrate appears to bind to the enzyme through interactions at the high-affinity sites, (–2) and (+2), and the sugar residues at both ends more loosely bind to the corresponding subsites, (–3) and (+3). The docking study of (GlcNAc)6 with the modeled structure of OsChia1cCBD supported the subsite structure estimated from the experimental time-course of hexasaccharide degradation. Since the class II chitinase from barley seeds was reported to possess a similar subsite structure from (–3) to (+3) and a similar free energy distribution, substrate-binding mode of plant chitinases of this class would be similar to each other.  相似文献

Muscle, liver, and kidney tissues from 38 polar bears (Ursus maritimus) caught in the Scoresby Sound area, Central East Greenland, were analysed for zinc, cadmium, mercury and selenium. In general, cadmium concentrations were low in muscle, liver and kidney tissue, with geometric means (g.m.) of 0.022 (range: <0.015–0.085), 0.841 (range: 0.092–3.29) and 13.1 (range: 1.04–115) g Cd/g wet weight (ww) respectively. This finding can be explained by low cadmium levels in the blubber of ringed seals. The concentration of mercury in muscle tissue was low (g.m. 0.071; range: 0.039–0.193 g Hg/g ww), whereas concentrations in liver and kidney tissue were relatively high (liver: g.m. 7.87; range: 1.35–24.8 g Hg/g ww, and kidney: g.m. 15.2; range: 1.59–66.6 g Hg/g ww). Mercury and cadmium were positively correlated with age in liver and kidney. Zinc was positively correlated with age in kidney, and selenium was correlated with age in liver. Contrary to other marine mammals, polar bears had higher mercury levels in the kidneys than in the liver. In all three tissues polar bears had significantly lower cadmium levels than ringed seals from the same area. Mercury levels were likewise significantly lower in the muscle tissue of polar bears than in ringed seals, whereas levels in the liver and kidney were significantly higher. The previous geographic trend for cadmium and mercury found in Canadian polar bears could be extended to cover East Greenland as well. Hence cadmium levels were higher in Greenland than in Canada, while the opposite was the case for mercury. Greenland polar bears had higher mercury and cadmium contents in livers and kidneys than polar bears from Svalbard. The mercury levels in muscle and liver tissue from polar bears from East Greenland were twice as high as found in bears from western Alaska, but half the levels found in northern Alaska. Cadmium and zinc were partially correlated in kidney tissue, and this was found for mercury and selenium as well. Cadmium and zinc showed molar ratios close to unity with the highest concentrations occurring in kidney tissue, while the levels of zinc exceeded cadmium in muscle and liver tissue by up to several decades. Mercury and selenium showed molar ratios close to unity in liver and kidneys.  相似文献

Freshwater salmonids exposed to low environmental pH typically suffer a net loss of ions, primarily Na+ and Cl, across the gills, resulting in reduced plasma and tissue ion concentrations. However, in recent experiments in our laboratory, juvenile rainbow trout, Oncorhynchus mykiss, fed a ration of 1% body weight d–1 or greater showed no ionoregulatory disturbance during chronic, sublethal acidification. This raised the possibility that these fish had acclimated to low pH in that they would be better able to withstand further, more severe acidification than fish that had no prior experience of acid conditions: previous studies had concluded that such acclimation does not occur. This hypothesis was tested by measuring unidirectional ion fluxes during a 24h acute acid challenge (pH 4.2) in juvenile rainbow trout that had previously been exposed to either ambient pH 6.2 (naive fish) or sublethal low pH 5.2 (acid pre-exposed fish) for 90 days, and fed a ration of either 1.0 or 0.25% d–1 (wet basis). No mortalities were observed during the acute acid challenge in the fish fed the higher ration and no differences between the two groups in the response of Na+ fluxes were observed. Sodium influx in both groups was significantly inhibited throughout the challenge and Na+ net flux was significantly stimulated over the first 6h. Prior to the acute acid challenge, the fish fed the lower ration that had previously been exposed to pH 5.2 had significantly lower plasma ion concentrations than those fish previously exposed to pH 6.2. Both groups suffered mortalities; those of the naive fish (22% by 24h) being markedly lower than those of the acid pre-exposed fish (68% by 24h). However, there were no significant differences in either Na+ or Cl fluxes between the two groups of fish during the acid challenge: both showed significant inhibition of ion influxes and significantly greater net ion losses, resulting in reduced plasma ion concentrations. These results indicate that rainbow trout are unable to acclimate to environmental acidification irrespective of the availability of dietary salts.  相似文献

The 98 amino acid (a. a.) N-terminus of the 126 a. a. atrial natriuretic factor (ANF) prohormone contains three peptides consisting of a. a. 1–30 (proANF 1–30), a. a. 31–67 (proANF 31–67) and a. a. 79–98 (proANF 79–98) with blood pressure lowering, sodium and/or potassium excreting properties similar to atrial natriuretic factor (a. a. 99–126, C-terminus of prohormone). ProANF 1–30 and proANF 31–67 have separate and distinct receptors from ANF in both vasculature and in the kidney to help mediate the above effects. At the cellular level proANFs 1–30, 31–67, and 79–98 as well as ANF's effects are mediated by enhancement of the guanylate cyclase (EC 4.6.1.2) — cyclic GMP system in vasculature and in the kidney. These peptides from the N-terminus of the ANF prohormone circulate normally in man and in all animal species tested. The object of the present investigation was to determine if these peptides have the ability to enhance either guanylate cyclase and/or adenylate cyclase in a variety of other tissues in addition to kidney and vasculature. ProANF 1–30, proANF 31–67, proANF 79–98, and ANF all increased rat lung, liver, heart and testes, but not spleen, particulate guanylate cyclase 2- to 3-fold at their 100 nM concentrations. Dose response curves revealed that maximal stimulation of particulate guanylate cyclase activity by these newly discovered peptides was at their 1 M concentrations, with no further increase in activity above their 1 M concentrations. Half-maximal (EC50) enhancement of particulate guanylate cyclase occurred at 0.15 ± 0.01, 0.3 ± 0.02, 0.5 ± 0.03, and 0.9 ± 0.03 nM for proANF 1–30, proANF 31–67, proANF 79–98 and ANF, respectively. ProANFs 1–30, 31–67, 79–98, and 99–126 (i.e., ANF) each increased cyclic GMP but not cyclic AMP levels in tissue slices of liver, lung, small intestine, heart, and testes. None of these peptides enhanced either adenylate cyclase or the soluble 100,000 G form of guanylate cyclase. The ability of these N-terminal peptides to enhance particulate guanylate cyclase activity in a wide variety of tissues suggests that they may have effects in a much wider variety of tissues than presently thought.  相似文献

The characteristics and habitat of the Patagonian toothfish (Dissostichus eleginoides) are typical of fish that accumulate high concentrations of mercury. In this study, mercury determinations were made on samples of muscle tissue from Macquarie Island toothfish and the Southern Ocean deepwater warty squid (Moroteuthis ingens). The analysis of mercury in the biological tissues was made by cold vapour-atomic absorption spectrometry following acid digestion. Performance of the analytical procedure was assessed by analysis of certified reference material (DORM-2, dogfish muscle). Mercury concentrations of 16 Macquarie Island toothfish ranged from 0.12 mg kg–1 (550 g, 381 mm TL) to 0.59 mg kg–1 (6,100 g, 823 mm TL), with a mean concentration of 0.33±0.12 mg kg–1. A significant correlation was found between mercury and either toothfish weight or total length. The fish analysed were juveniles, which suggests that larger individuals would have higher mercury concentrations well exceeding food standard code limits for mercury in fish (typically 0.5 mg kg–1). Warty squid, also from around Macquarie Island, had a low mean mercury concentration of 0.086 mg kg–1 in mantle tissue; no significant correlation existed between mercury concentration and either squid mantle length or total weight. It is postulated that the squid have a mechanism, possibly involving the digestive gland, that prevents bioaccumulation of mercury in the mantle, and presumably other body tissues.  相似文献

Background

In 2003 mean cord blood mercury concentrations in pregnant Bermudian women exceeded levels associated with adverse health outcomes in children. The principal mercury source was local fish species. Public health messages were developed suggesting pregnant women reduce consumption of fish species with higher mercury concentrations (e.g. swordfish), substituting species containing lower mercury concentrations, and elevated omega-3 fatty acids (e.g. anchovies). Recent evidence indicates mercury concentrations in Bermuda’s pregnant women have fallen five- fold.

Objectives

Assess whether changes in women’s fish eating patterns during pregnancy are consistent with the public health messaging. Determine who is making changes to their diet during pregnancy and why.

Methods

Mixed methods study with a cross-sectional survey of 121 pregnant women, including 13 opened-ended interviews. Health system, social vulnerability, public health messaging, and socio-demographic variables were characterized and related to changes in fish consumption during pregnancy. Qualitative data were coded according to nutritional advice messages, comprehension of communication strategies, and sources of information.

Results

95% of women surveyed encountered recommendations about fish consumption during pregnancy. 75% reported modifying fish eating behaviors because of recommendations. Principal sources of information about fish consumption in pregnancy were health care providers and the Internet. 71% of women reported reducing consumption of large fish species with greater mercury levels, but 60% reported reduced consumption of smaller, low mercury fish. No participant mentioned hearing about the benefits of fish consumption. More frequent exposure to public health messages during pregnancy was associated with lower reported consumption. Bermudian born women were less likely to reduce consumption of large fish species during pregnancy.

Conclusions

In Bermuda, public health messages advocating reduced consumption of larger, higher mercury-containing fish species appear effective, but masked the nutritional value message of small fish species, with low mercury concentration. Adjustment is needed to better balance the risk communication.  相似文献

A laboratory experiment was performed to study the responsiveness of selected condition measures to starvation in herring, Clupea harengus, larvae in relation to temperature and ontogeny. The larvae at two intervals of development, i.e. stage 1 larvae with initial exogenous feeding and stage 2 larvae with established feeding prior to notochord flexion, were reared at three temperatures (5, 8 and 11°C) and subjected to sub-lethal durations of starvation. Temporal changes in standard length, dry weight, DNA concentration (% of dry weight), RNA concentration (% of dry weight), Fulton's condition factor (CF) and RNA/DNA were assessed and compared with fed controls. Starvation led to decreases in dry weight, CF, RNA concentration and RNA/DNA, while it led to an increase in DNA concentration. Higher responsiveness to starvation was observed at higher temperatures, and the magnitude of the changes was higher in stage 2 larvae. The shortest latency in starvation response was found with respect to RNA/DNA which was length independent in the size range studied. RNA/DNA was also significantly related to average DNA growth rate, and the model for DNA growth rate was, SGRDNA=4.49 RNA/DNA + 7.14 T – 0.42 T2 – 37.5 ; n=32, r2=0.85, p < 0.001). While the model seemed to adequately represent the average temperature dependent DNA growth, a relatively low classification success made it unsuitable for depicting individually starving larvae. Critical levels in DNA concentration can be used (2.2% for stage 1 larvae, 2.9% for stage 2 larvae) to differentiate starving larvae (after 3 – 5 days) from feeding larvae. RNA/DNA was the most sensitive and suitable condition index studied in detecting early starvation of herring larvae.  相似文献

The effects of 9-tetrahydrocannabinol (9-tetrahydrocannabinol-THC) administration on the central serotoninergic system were evaluated by biochemical assays of tissue levels of indoleamines; a measure of the serotonin (5-HT) innervation was obtained by using [3H]paroxetine as a maker of 5-HT uptake sites. Two different 9-THC treatments were chosen, i.e: acute and chronic perinatal maternal exposure. Following acute treatment (5mg/kg), the 5-HT content increased in dorsal hippocampus (+35%), Substantia nigra (+61%) and neostriatum (+62%) but remained unchanged in cingulate cortex, Raphe nuclei, Locus coeruleus and anterior hypothalamus. Endogenous 5-hydroxyindole-3-acetic acid (5-HIAA) decreased in anterior hypothalamus (–23%) and Raphe nuclei (–21%). Following maternal exposure to 9-THC (5 mg/kg per day; from gestational day 13 to postnatal day 7), levels of 5-HT were increased in the neostriatum (+22%) but decreased in anterior hypothalamus (–25%), Raphe nuclei (–29%) and Locus coeruleus (–20%) of the litters. Tissue 5-HIAA was increased in anterior hypothalamus (+23%) and Substantia nigra (+48%). There were no changes in 5-HT uptake site density, determined by [3H]paroxetine binding, except for an increase (+50%) in the cingulate cortex of perinatal-treated rats when compared to acutely-treated animals. The present results show that acute and maternal exposure to 9-THC produced different effects on the central 5-HT system of the offspring, with a clear regional especifity, but with no changes in the densities of 5-HT uptake sites.Abbreviations Hypo anterior hypothalamus - Cin cingulate cortex - dHipp dorsal hippocampus - 5-HIAA 5-hydroxyindole-3-acetic acid - HPLC high-performance liquid chromatography - 5-HT serotonin - 5-HTP 5 hydroxy-1-tryptophan - LC Locus coeruleus - rNS rostral neostriatum - MRN midbrain Raphe nuclei region - SN Substantia nigra - 9-THC 9-Tetrahydrocannabinol  相似文献

Macroalgal blooms have increased globally in recent decades as a result of increased nutrient enrichment and eutrophication of coastal waters. In Lee County, Florida, this problem reached a critical stage in 2003/2004 when massive rhodophyte blooms washed ashore, making beaches unsuitable for recreation and requiring an expensive removal program. To better understand the ecology of these blooms, water quality and macroalgae sampling was conducted in August 2004, prior to hurricane Charley, and again in late October following several months of large freshwater discharges from the Caloosahatchee River. During both samplings, water and macroalgae were collected along a gradient extending from the Caloosahatchee River to natural and artificial reefs up to 26 km from shore.Dissolved nutrient concentrations were generally high throughout the study area, with significantly higher concentrations in the Caloosahatchee River. Mean dissolved inorganic nitrogen concentrations in the Caloosahatchee River increased from Ortona Lock (<18 μM) to Franklin Lock (23–28 μM) downstream during both samplings, indicating significant enrichment within the basin. On coastal reefs, mean ammonium concentrations increased six-fold (≤0.20–1.31 μM) and soluble reactive phosphorus increased three-fold (0.30–0.92 μM) from August to October, respectively. Mean reef macroalgae C:N ratios were low and similar in August (13.9) and October (13.5), and C:P and N:P ratios were also low but decreased significantly from August to October (386–242 and 27.4–17.5, respectively). Macroalgal δ15N values increased from Ortona Lock (+8 to 9‰) to Franklin Lock (+12 to 15‰) during both samplings, were within the sewage nitrogen range, and decreased with increasing distance from shore to +3.0‰ at the most offshore reef. Macroalgae (Gracilaria, Hypnea, Botryocladia, Eucheuma, Sargassum) collected in July 2004 from Lee County beaches had mean δ15N values >+6.0‰, similar to values for macroalgae on inshore reefs and within the sewage nitrogen range. However, mean δ15N values of reef macroalgae decreased from August (+5.84‰) to October (+3.89‰) as Caloosahatchee River discharges increased, suggesting relatively larger contributions from nitrogen sources with low δ15N values (<+3‰), such as rainfall and agricultural fertilizers, in the wet season. Improved management of freshwater releases from Lake Okeechobee, combined with nutrient removal from sewage effluent within the Caloosahatchee River drainage basin, could help mitigate future macroalgal blooms in Lee County's coastal waters.  相似文献

Synopsis Arsenic persists in Chautauqua Lake, New York waters 13 years after cessation of herbicide (sodium arsenite) application and continues to cycle within the lake. Arsenic concentrations in lake water ranged from 22.4–114.81 g l–1, = 49.0 ag l–1. Well water samples generally contained less than 10 g l–1 arsenic. Arsenic concentrations in lake water exceeded U.S. Public Health Service recommended maximum concentrations (10 g l–1) and many samples exceeded the maximum permissible limit (50 g l–1). Fish accumulated arsenic from water but did not magnify it. Fish to water arsenic ratios ranged from 0.4–41.6. Black crappie (Pomoxis nigromaculatus) contained the highest arsenic concentrations (0.14–2.04 g g–1 ), X = 0.7 g g–1) while perch (Perca flavescens), muskellunge (Esox masquinongy) and largemouth bass (Micropterus salmoides) contained the lowest concentrations (0.02–0.13 g g–1). Arsenic concentrations in fish do not appear to pose a health hazard for human consumers.  相似文献

A survey carried out in Lake Tana in 2015 found that Hg levels in some fish species exceeded internationally accepted safe levels for fish consumption. The current study assesses human exposure to Hg through fish consumption around the Lake Tana. Of particular interest was that a dietary intake of fishes is currently a health risk for Bihar Dar residents and anglers. Hair samples were collected from three different groups: anglers, college students and teachers, and daily laborers. A questionary includes gender, age, weight, activity. Frequency of fish consumption and origin of the eaten fish were completed by each participant. Mercury concentrations in hair were significantly higher (P value <0.05) for anglers (mean?±?standard deviation 0.120?±?0.199 μg/g) than college students (mean?±?standard deviation 0.018?±?0.039 μg/g) or daily workers (mean?±?standard deviation 16?±?9.5 ng/g). Anglers consumed fish more often than daily workers and college group. Moreover, there was also a strong correlation (P value <0.05) between the logarithms of total mercury and age associated with mercury concentration in scalp hair. Mercury concentrations in the hair of men were on average twice the value of the women. Also, users of skin lightening soap on a daily basis had 2.5 times greater mercury in scalp hair than non-users. Despite the different sources of mercury exposure mentioned above, the mercury concentrations of the scalp hair of participants of this study were below levels deemed to pose a threat to health.  相似文献

Biofilms were allowed to develop on wooden slides of the River Red Gum (Eucalyptus camaldulensis Dehnh., Myrtaceae) submerged in two billabongs of south-eastern Australia. The slides were placed in the photic zone and the aphotic zone, and the biofilms sampled after eight week's growth over the summer of 1989–1990 and winter of 1990. Bacterial numbers, estimated with epifluorescence microscopy, ranged from 4–78 × 106 cells cm–2. Bacteria were more abundant in the photic zone than the aphotic zone, and more abundant in summer than winter. Fewer than 0.5% of the bacteria could be cultivated on nutrient agar plates. Concentrations of phospholipids ranged from 8–79 ng cm–2, which corresponded to bacterial abundances of 2–17 × 106 cells cm–2. Fifty five phospholipid fatty acids (PLFA) were identified, of which 16:0 (13–29% of total PFLA) was the most common. Other abundant PFLA included 16:17c (6–28%), 18:26 (3–16%), 18:33 (4–12%), 18:19c (3–5%), 18:l7c (5–11%) and 18:0 (2–8%). Minor PLFA included 14:0, i and a 15:0, 15:0, 16:l5c, 16:113c, 18:36, 18:43, 20:46 and 20:53. The PLFA profiles of the biofilms were quite different from those of the sediments and plankton. There was a clear distinction between the PLFA profiles of summer and winter biofilms, but less evidence for unequivocal site or light-regime effects.  相似文献
