Pepper ( Capsicum annuum L. cv. Keystone Resistance Giant 3) seeds were monitored during priming to determine if seed treatments which accelerate the rate of germination could be correlated with specific physiological changes within the seeds. Pepper seeds primed with −0.90 and −1.35 MPa NaCl solutions at 23°C for 18 days did not completely equilibrate with the osmotic potential of the priming solution. Seed respiratory rates indicated that priming extends the lag phase of germination following imbibition. Soluble protein levels increased 115% in primed seeds, and the uptake and incorporation of [14C(U)] labelled amino acids into the acid insoluble fraction increased throughout the priming treatments. Alcohol dehydrogenase (EC 1.1.1.1, anaerobic metabolism), glucose-6-phosphate dehydrogenase (EC 1.1.1.49) and 6-phosphogluconate dehydrogenase (EC 1.1.1.44, pentose phosphate pathway) activities remained stable throughout the priming treatment, but were higher after 6 days. than the water-imbibed controls. Aldolase (EC 4.2.1.1. glycolysis) and isocitrate lyase (EC 4.1.3.1, glyoxylate cycle) activities increased with imbibition and were 61 and 56% (respectively) higher in primed seeds as compared to the water-imbibed controls after 12 days. Treatment with the −0.90 MPa NaCl solution was more effective than the −1.35 MPa solution in improving the rate of germination, yet there were no significant differences between the protein concentrations or enzyme activities of the two priming treatments. However, the incorporation of labelled amino acids into pepper seeds was significantly higher in the −0.90 MPa priming treatment.  相似文献

Samples of three seed lots of each of three cultivars of carrot, celery and onion were primed in polyethylene glycol solution for two weeks at 15 °C. Seedling emergence was recorded in the field for carrot and onion and in the glasshouse for celery. Compared to the untreated control, priming increased the percentage seedling emergence in certain poorly-emerging seed lots of carrot and celery, but had no effect on onion. Mean emergence times were reduced by priming in all seed lots, by 3–5, 5–8 and 3–9 days in carrot, celery and onion respectively. The largest effects occurred in the slowest-emerging seed lots. There were significant interactions between priming and seed lots within cultivars in carrot and celery and between priming and cultivars in celery and onion. Priming generally reduced the spread of emergence times, but the effects were not statistically significant in carrot. Drying back the primed seeds had no effect on percentage emergence in onion, but reduced it (compared to primed seed which had not been dried-back) in certain carrot and celery seed lots. Primed and dried-back seeds emerged later than primed seeds, by up to 1·5, 2·6 and 2·6 days in carrot, celery and onion respectively. The spread of emergence times was generally larger for primed and dried-back seeds than for primed seeds, but the differences were not always statistically significant. Plant fresh weights were recorded 9, 7 and 12 wk after sowing for carrot, celery and onion, respectively. In each species, mean plant weight was inversely related to seedling emergence time; thus plants grown from primed seed were always heavier than the controls, by up to 33%, 182% and 47% in carrot, celery and onion respectively.  相似文献

Plant morphology in the field in Virginia, USA, and growth responses to applied NaCl in a glasshouse were determined for two populations of Opuntia humifusa (Rafinesque) Rafinesque, one from an inland site and the other from the marine strand, which differed in potential exposure to salinity. Cladode dimensions, plant height and rooting depth varied significantly between the populations. Application of NaCl in 50 m M increments up to 400 m M every 3 days for 6 weeks caused the cladodes to decrease up to 30% in thickness, the decrease being about 20% more at 50 to 150 m M NaCl for plants from the inland site than from the marine strand. Inhibition by 150 m M NaCl of the maximum rate of net CO2 uptake and of the total CO2 uptake over a 24-h period was greater for the inland population. Growth, especially of roots, was inhibited by applied NaCl, with a decrease in biomass above 200 m M NaCl for plants from the inland site and not until 400 m M for those from the marine strand. Although the root Na+ level was the same for plants from the two populations, reaching a maximum of about 8 mg (g dry weight)−1 at 200 m M NaCl, the cladode Na+ level was two-fold higher for plants from the marine strand than from the inland site. Thus, exclusion of Na+ from the cladodes is not the reason for the greater NaCl tolerance of O. humifusa from the marine strand, which is a habitat that can experience periodic episodes of high salinity.  相似文献

As water and nutrient uptake should be related in the response of plants to salinity, the aim of this paper is to establish whether or not aquaporin functionality is related to H+-ATPase activity in root cells of pepper ( Capsicum annuum L.) plants. Thus, H+-ATPase activity was measured in plasma membrane vesicles isolated from roots and aquaporin functionality was measured using a cell pressure probe in intact roots. Salinity was applied as 60 m M NaCl or 60 m M KCl, to determine which ion (Na+, K+ or Cl) is producing the effects. We also investigated whether the effects of both salts were ameliorated by Ca2+. Similar results were obtained for cell hydraulic conductivity, Lpc, and H+-ATPase activity, large reductions in the presence at NaCl or KCl and an ameliorative effect of Ca2+. However, fusicoccin (an activator of H+-ATPase) did not alter osmotic water permeability of protoplasts isolated from roots. Addition of Hg2+ inhibited both ATPase and aquaporins, but ATPase also contains Hg-binding sites. Therefore, the results indicate that H+-ATPase and aquaporin activities may not be related in pepper plants.  相似文献

The sap flow (Jv) and the osmotic pressure-dependent hydraulic conductance (L0) of detached exuding root systems from paprika pepper plants (cv. Albar) were measured. Plants stressed with NaCl (30 m M ) and with six times the macronutrients of the Hoagland nutrient solution (6×HNS) were compared with controls grown in complete Hoagland nutrient solution. Jv of +NaCl and +6×HNS plants decreased markedly, but recovered to values similar to those of controls after removal of the treatments. Hydraulic conductance L0 was always less in NaCl plants than in controls and 6×HNS. A total increase in the ion concentration of the xylem (except Na+ and Cl) was observed with both treatments. In control and 6×HNS plants, HgCl2 treatment (50 μ M ) caused a sharp decline in L0 to values similar to those of NaCl-stressed roots, but were restored by treating with 5 m M dithiothreitol (DTT). However, in NaCl roots only a slight effect of Hg2+ and DTT was observed. In each treatment, there was no difference in the flux of K+ into the xylem after HgCl2 and DTT application. The results suggest that NaCl decreased L0 of the roots by reducing either the activity or abundance of Hg-sensitive water channels. The putative reduction in water-channel function of NaCl-treated plants did not seem to be due to the osmotic effect.  相似文献

Three year data on the effect of water- and mannitol (4%) priming of chickpea seeds (12 h at 25°C) showed higher number and biomass of nodules in the plants from primed seeds than from non-primed seeds. The biomass of nodules increased to 75 DAS but decreased by 90 DAS. Activities of sucrose metabolism enzymes (sucrose synthase (SS) and alkaline invertase) and of nitrogen metabolism (glutamine synthetase (GS), glutamate synthase (GOGAT) and glutamate dehydrogenase (GDH)) in nodules of primed and non-primed crops during development are reported. SS and alkaline invertase activities increased to 70 DAS and then decreased. In primed plants, the higher SS activity in nodules at 60 and 70 DAS might be responsible for providing more energy and carbon skeleton for nitrogen fixation and for ammonium assimilation in primed plants. At 85 DAS, though the SS activity decreased in comparison with the earlier growth stages, it was still higher in nodules of the primed crops than the non-primed crop. Activity of alkaline invertase was maximum at 70 DAS in the nodules of primed and non-primed crops. Priming increased nodule GS activity at 70 and 85 DAS. GOGAT activity was unaffected by priming but GDH activity was greater in nodules from primed crops at 50 DAS. Elevated SS and GS nodule activities in primed chickpeas might be responsible in increasing nodule biomass and metabolic activity thereby increasing seed fill.  相似文献

Cultivated tomato Lycopersicon esculentum (L.) Mill. cv. P-73 and its wild salt-tolerant relative L. pennellii (Correll) D'Arcy accession PE-47 growing on silica sand in a growth chamber were exposed to 0, 70, 140 and 210 m M NaCl nutrient solutions 35 days after sowing. The saline treatments were imposed for 4 days, after which the plants were rinsed with distilled water. Salinity in L. esculentum reduced leaf area and leaf and shoot dry weights. The reductions were more pronounced when sodium chloride was removed from the root medium. Reduction in leaf area and weight in L. pennellii was only observed after the recovery period. In both genotypes salinity induced a progressive reduction in leaf water potential and leaf conductance. During the recovery period leaf water potential (ψ1) and leaf conductance (g1) reached levels similar to those of control plants in wild and cultivated species, respectively. Leaf osmotic potential at full turgor (ψos) decreased in the salt treated plants of both genotypes, whereas the bulk modulus of elasticity was not affected by salinity. Leaf water potential at turgor loss point (ψtlp) and relative water content at turgor loss point (RWCtlp) appeared to be controlled by leaf osmotic potential at full turgor (ψos) and by bulk modulus of elasticity, respectively. At lowest salinity, the wild species carried out the osmotic adjustment based almost exclusively on Cl and Na+, with a marked energy savings. Under highest salinity, this species accommodate the stress through a higher expenditure of energy due to the contribution of organic solutes to the osmotic adjustment. The domesticated species carried out the osmotic adjustment based always on an important contribution of organic solutes.  相似文献

Enhancement of salt tolerance in soybean with NaCl pretreatment   总被引:4,自引:0,他引:4  
Acclimation response to salt stress in soybean ( Glycine max [L.] Merr. cv. Lee) was found in this study. Soybean seedlings were exposed to 0, 34 and 68 m M NaCl for 23 days (pretreatment), thereafter plants were exposed to 0, 68 and 137 m M NaCl until maturity (main treatment). There was no effect of pretreatment on growth, but at 137 m M NaCl, Na+ concentration in leaves of the plants pretreated with 34 m M NaCl was lower than that of plants pretreated with 0 and 68 m M NaCl. Furthermore, the survival rate under 137 m M NaCl improved with the 34 m M NaCl pretreatment. Therefore, it is possible that soybean can acclimate to salt stress by its increased survival rate, without showing any improvement in growth. The regulation of Na+ or Cl concentration in leaves could be one of the possible factors involved in salt acclimation of soybean.  相似文献

Water (H15O) translocation from the roots to the top of rice plants ( Oryza saliva L. cv. Nipponbare) was visualized over time by a positron-emitting tracer imaging system (PETIS). H15O flow was activated 8 min after plants were exposed to bright light (1 500 μmol m−2 s−1). When the light was subsequently removed, the flow gradually slowed and completely stopped after 12 min. In plants exposed to low light (500 μmol m−2 s−1), H15O flow was activated more slowly, and a higher translocation rate of H15O was observed in the same low light at the end of the next dark period. NaCl (80 m M ) and methylmercury (1 m M ) directly suppressed absorption of H15O by the roots, while methionine sulfoximine (1 m M ), abscisic acid (10 μ M ) and carbonyl cyanide m -chlorophenylhydrazone (10 m M ) were transported to the leaves and enhanced stomatal closure, reducing H15O translocation.  相似文献

The effects of NaCl on endogenous free levels of the poluamines putrescine, spermi dine and spermine, and the relationships between polyamines, K+ levels and Na+ accumulation were determined in leaves of the cultivated tomato ( Lycopersicon esculentum Mill.) and its wild, salt-tolerant relative L. pennellii (Correll) D' Arcy at different exposure times during a 32-day period. Both stress treatments (100 and 200 m M NaCl) decreased the levels of putrescine and spermidine, although to a different degree for the cultivated and wild tomato species. The spermine levels did not decrease with salinity in L. pennellii over the salinization period, whereas they decreased in L. esculentum , except at the first application of the 100m M NaCl treatment. In both species, the changes induced by salinity in total polyamines and K+ were very similar, with the accumulation of Na+ in the leaf being concomitant with a decrease in both total polyamines and K+. This suggests that the main role of the polyamines in the leaf tissues. In this sense, a direct relationship between total polyamines and K+, and inverse relationship between polyamines and Na+ and between K+ and Na+ were found for both species. In the short term (up to 4 days) a peculiar physiological behavior was found in L. pennellii , as the total polyamine and K+ levels decreased at 100 m M but not at 200 m M NaCl, while after this time the latter plants had values lower than those of the 100 m M NaCl-treated plants at day 11.  相似文献

Aldicarb at 1.4–5.6 kg a.i. ha-l or oxamyl at 1.3–5.2 kg a.i. ha-1 applied over the seed furrows during sowing prevented much of the damage resulting from stem nematode attack in spring-sown onions. Rotting of bulbs in store due to stem nematode infestation was sometimes lessened by these treatments. Neither sowing onion seeds on soil ridges, nor applying aldicarb in two equal doses, one over the seed furrows during sowing, the other over the plants mid-season, controlled the nematode better or increased onion yields more than a single equivalent dose over the seed furrows. Applied as two doses in this way, aldicarb was sometimes more effective against the nematode in summer-sown onions than was an equivalent amount applied over the seed furrows during sowing. Only 2.5 kg or less aldicarb ha-1 can be applied to onions without leaving unacceptable residues (>0.15 μg g-1) in the harvested bulbs.  相似文献

Translocation of NH4+ was studied in relation to the expression of three glutamine synthetase (GS, EC 6.3.1.2) isogenes and total GS activity in roots and leaves of hydroponically grown oilseed rape ( Brassica napus ). The concentration of NH4+ in the stem xylem sap of NO3-fed plants was 0.55–0.70 m M , which was ≈60% higher than that in plants deprived of external nitrogen for 2 days. In NH4+-fed plants, xylem NH4+ concentrations increased linearly both with time of exposure to NH4+ and with increasing external NH4+ concentration. The maximum xylem NH4+ concentration was 8 m M , corresponding to 11% of the nitrogen translocated in the xylem. In the leaf apoplastic solution, the NH4+ concentration increased from 0.03 m M in N-deprived plants to 0.20 m M in N-replete plants. The corresponding values for leaf tissue water were 0.33 and 1.24 m M , respectively. The addition of either NO3 or NH4+ to N-starved plants induced both cytosolic gs isogene expression and GS activity in the roots. In N-replete plants, gs isogene expression and GS activity were repressed, probably due to carbon limitations, thereby protecting the roots against the excessive drainage of photosynthates. Repressed gs isogene expression and GS activity under N-replete conditions caused enhanced NH4+ translocation to the shoots.  相似文献

Leek seed lots of high (91%) and low (82%) viability were primed in aerated polyethylene glycol (PEG) solutions in Bubble-columns and by a non-osmotic priming technique in both 1987 and 1988 and the seeds were then sown in the field. Both methods of priming established a similar seed moisture content during treatment and, in the laboratory, produced seeds with more rapid and uniform germination than untreated seeds, with a greater advantage for Drum compared with Bubble-column priming in PEG. In the field, both priming techniques gave seedling emergence responses similar to those from priming in PEG by the laboratory-scale technique on filter paper. Both large-scale priming methods gave earlier and more uniform emergence than untreated seeds and gave similar or slightly higher levels of seedling emergence, except on one sowing occasion when seeds were stored before sowing followed by sowing into a drying seedbed.  相似文献

The present study examined the response of antioxidant systems to NaCl stress and the relative importance of Na+ and Cl in NaCl-induced antioxidant systems in roots of rice seedlings. NaCl treatment caused an increase in the activities of ascorbate peroxidase (APX) and glutathione reductase (GR) in roots of rice seedlings, but had no effect on the activities of superoxide dismutase (SOD) and catalase (CAT). There were detectable differences in APX and GR isoenzymes between control and NaCl-treated roots. Levels of activity for SOD and CAT isoenzymes did not change in NaCl-stressed roots compared with the control roots. NaCl treatment produced an increase in H2O2, ascorbate (AsA), dehydro-ascorbate (DHA), reduced glutathione (GSH), and oxidized glutathione (GSSG) levels. Treatment with 50 m M Na-gluconate (whose anion is not permeable to membrane) led to a similar Na+ level in roots to that with 100 m M NaCl. It was found that treatment with 50 m M Na-gluconate affected H2O2, AsA, and DHA levels, APX and GR activities, OsAPX and OsGR mRNA induction in the same way as 100 m M NaCl. These observed changes seem to be mediated by Na+ toxicity and not by Cl toxicity. On the other hand, it was found that NaCl, but not Na-gluconate and NaNO3, caused an increase in GSH and GSSG levels, indicating that Cl, rather than Na+, is responsible for the NaCl-increased GSH and GSSG levels in roots of rice seedlings.  相似文献

Plantago species differ in their strategy towards salt stress, a major difference being the uptake and distribution of Na+ ions. A salt-sensitive ( Plantago media L.) and a salt-tolerant ( P. maritima L.) species were compared with respect to Na+/H+ antiport activities at the tonoplast. After exposure of the plants to 50 m M NaCl for 6 days isolated tonoplast vesicles of P. maritima showed Na+/H+ antiport activity with saturation kinetics and a Km of 2.4 m M Na+, NaCl-grown P. media and the control plants of both species showed no antiport activity. Selectivity of the antiport system for Na+ was high and was determined by adding different chloride salts after formation of a Δ pH in the vesicles. Specific tonoplast ATPase activities were similar in the two species and did not alter after exposure to NaCl stress.  相似文献

This paper studies the relative importance of endogenous ABA and ion toxicity in the leaf growth inhibition caused by NaCl in salt-adapted and unadapted bush beans. Adaptation to salt-stress was achieved by germination of seeds in 75 m M NaCl, while unadapted plants were germinated in tap water. The adaptation process caused a transitory increase in leaf ABA, Na+ and Cl concentrations, while leaf expansion was inhibited. However, when grown for 8 or 13 days in 75 m M NaCl-containing nutrient solution, primary and first trifoliolate leaves of salt-adapted plants had greater areas than those of unadapted plants. Concentrations of ABA, Na+ and Cl in these leaves were lower in adapted plants, and a strong negative correlation between leaf expansion growth and either leaf Na+, Cl or ABA concentrations could be established. However, in the second trifoliolate leaves only the ABA, but not the Na+ or Cl, concentrations were significantly correlated with leaf expansion. Our results suggest that salt-induced inhibition of leaf expansion growth in bush beans is mediated by ABA rather than Na+ or Cl toxicity. Moreover, the increase of ABA, induced by the salt-pretreatment, seems to play an important role in limiting the accumulation of Na+ and Cl in the leaves, leading to adaptation of bush beans to salt-stress.  相似文献

Peanuts ( Arachis hypogaea L. cv. Shulamit) grown with NO3 and saline water in hydroponics responded positively to addition of nitrogen (N) in their vegetative growth, but not in desert dune sand. In order to clarify these conflicting results, peanut plants were grown in a greenhouse pot experiment with fine calcareous sand. The nutrient solution contained 0 or 50 m M NaCl and 2 or 6 m M N in the form of Ca(NO3)2, NH4NO3 or (NH4)2SO4. Three replicates were harvested after 48 days (beginning of reproductive stage) and three after 109 days (pod filling). In addition, gynophores were treated with 0, 50, 100, 150 or 200 m M NaCl outside the growth pot to check their sensitivity to salt. Shoot dry weight became greater with increasing NH4+/NO3 ratio. Increasing the N concentration from 2 to 6 m M did not change shoot dry weight of the NH4NO3 or NH4+-fed plants, but caused a reduction in shoot dry weight of NO3-fed plants. Shoot dry weight was not affected by increasing the NaCl concentration to 50 m M . Salt caused an increase in the number of gynophores per plant and a reduction of the mean pod weight. A NaCl concentration of 100 m M and above reduced gynophore vitality. It is concluded that the salt sensitivity of peanut plants resides mainly in the sensitivity of the reproductive organs.  相似文献

Seedlings of spring wheat ( Triticum aestivum L. cv. Svenno) were cultivated at 20°C in continuous light or darkness with the roots in nutrient solutions for six days. The plants were starved for K+ during different periods of time to produce plants with various K+ status. In one cultivation light-grown plants were pretreated in darkness, and vice versa, before the uptake experiment. In all experiments, roots were put in a complete nutrient medium containing 2.0 m M K+ radiolabelled with 86Rb. The uptake time was varied (5, 60 or 120 min).
The K+ concentration in the roots, [K+]root, increased during the course of the uptake experiments, especially in light and at initially low [K+]root, At the same time K+ (86Rb) influx in the roots decreased. The simoidal relationship obtained between K+ (86Rb) influx and [K+]root was affected by these changes, and Hill plots gave various Hill coefficients, nH, depending on the duration of the uptake experiments. nH from three apparently straight line segments of the same plot, in different [K+]root - intervals, indicated a falling degree of interaction between the binding sites as [K+]root increased. For the dark-grown plants negative cooperativity could not be demonstrated.  相似文献

Carrot and leek seed was osmotically primed in polyethylene glycol solution (273 g/kg water and 342 g/kg water respectively) for 10, 14 or 17 days before accelerated ageing for 0, 24, 48, 72 or 96 h. Priming reduced the germination time compared with non-primed seed. Accelerated ageing increased germination and emergence times and decreased percentage germination and emergence to a greater extent for the primed seeds than for non-primed seeds in both species. Primed and dried but non-aged seed from both species was stored at 10°C for 12 months. There was no loss of viability and improvements in germination time due to priming were maintained throughout the storage period for all the priming treatments in leek, and for the 10 and 14 day priming treatments in carrot. Carrot seed primed for 17 days lost some viability after 12 months storage compared with non-stored seed.  相似文献
