The melting temperature and hyperchromia of intestine mucosa tRNA preparations and the effect of inactivating factors on aminoacyl-tRNA-synthetase activity were studied at the stage of hypovitaminosis and under deep A-avitaminosis. It is shown that in the aminoacylation reaction, aminoacyl-tRNA-synthetase is a component more sensitive to vitamin A deficiency.  相似文献

The effect of vitamin A deficiency and vitamin A replacement on spermatogenesis was studied in mice. Breeding pairs of Cpb-N mice were given a vitamin A-deficient diet for at least 4 wk. The born male mice received the same diet and developed signs of vitamin A deficiency at the age of 14-16 wk. At that time, only Sertoli cells and A spermatogonia were present in the seminiferous epithelium. These spermatogonia were topographically arranged as single and paired cells and as clones of 4, 8 and more cells. A few mitoses of single, paired, and clones of 4 A spermatogonia were found, which were randomly distributed over the seminiferous epithelium. When vitamin A-deficient mice were treated with retinol-acetate combined with a normal vitamin A-containing diet, spermatogenesis restarted again synchronously. Only a few successive stages of the cycle of the seminiferous epithelium were present up to at least 43 days after vitamin A replacement. After 20 days, 98.3% of the seminiferous tubules were synchronized, showing pachytene spermatocytes as the most advanced cell type, mostly being in epithelium stages IX-XII. After 35 and 43 days, spermatogenesis was complete in 99.6% of the tubular cross sections, and most tubular cross sections were in stages IV-VII of the cycle of the seminiferous epithelium. The degree of synchronization was comparable or even higher than found in rats. The rate of development of the spermatogenic cells between 8 and 43 days after vitamin A replacement seemed to be similar to that in normal mice. Assuming that the rate of development of the spermatogenic cells is also normal during the first 8 days after vitamin A replacement, it can be deduced that the preleptotene spermatocytes, present after 8 days, were A spermatogonia in the beginning of stage VIII at the moment of vitamin A replacement. These results indicate that the mouse can be used as a model to study epithelial stage-dependent processes in the testis.  相似文献

The cholesterol side-chain cleavage enzyme activity is decreased considerably at the mild stage of vitamin A deficiency in rat testes and ovaries and the decrease in activity becomes more pronounced with progress of deficiency. Supplementation of the deficient rats with retinyl acetate, but not retinoic acid, restores the enzyme activity to normal values. The cholesterol side-chain cleavage enzyme of adrenals is not affected by any of the above treatments.  相似文献

Effects of glucagon and biogenic amines as well as of non-hormonal agents (manganese cations, forskolin, guanine nucleotides) on activity of adenylyl cyclase (AC) was studied in the infusorian Tetrahymena pyriformis, a representative of unicellular eukaryotes. The infusorian cultures at different growth stages were chosen for the study. It has been revealed that the AC basal activity measured in the cultures at the stationary stage is much lower than in those at the exponential stage. The stimulating effects of hormones (glucagon, serotonin, isoproterenol) and of non-hormonal agents were shown to depend markedly on the tetrahymena AC basal activity. These effects are pronounced at the stationary stage of growth (the low basal activity), whereas in cultures at the active division stage (the high basal activity) they either are reduced or change their sign to opposite (inhibition of AC activity. The obtained results not only are another evidence in favor of the existence of the functionally active ACS in lower eukaryotes, but also indicate an important role of this system in regulation of sensitivity of infusoria at different growth stages to external actions.  相似文献

Bletilla striata (Thunb.) Reichb.f. is a perennial herb with abundant active ingredients. Previous research mainly focused on its tubers, however, the study on flowers, especially the variation of active ingredient contents at different flowering stages, was rarely seen. This study analyzed the total phenols, flavonoids, polysaccharides, anthocyanins, and cyanidin-3-O-glucoside content of B. striata flowers which were in cultivated in Herb Garden of Zhejiang A&F University and collected in May, 2019, in order to investigate the changes in active ingredients and antioxidant capacity among different flowering stages (bud, initial, and full bloom). Changes in radical scavenging capability of DPPH (1,1-Diphenyl-2-picrylhydrazyl radical), ABTS (2,2′-azinobis(3-ethylbenzthiazoline-6-sulphonate)), and hydroxy were analyzed. Significant differences in active ingredient content of flowers were detected among different flowering stages. The total phenolic content increased continuously during the entire flowering stage. The contents of total flavonoid, total polysaccharide, and cyanidin-3-O-glucoside reached peaks at the initial blooming stage and then fell as the flowering process continued. The antioxidant activity in initial stage was the highest than in any other flowering stages. Therefore, we conclude that the initial blooming stage is the best harvesting stage of B. striata flowers. This study provides a robust basis for the harvest and utilization of B. striata flowers in food, medical, and cosmetic industries.  相似文献

The activities of the enzymatic systems involved in the activation and degradation of fatty acids, and in the synthesis of triacylglycerols and phospholipids were studied in vitro using total cellular homogenate and subcellular fractions of eggs of the shrimp Macrobrachium borellii at different developing stages. Egg development was divided into seven stages based on morphological features of the embryo. Palmitoyl-CoA ligase activity increased as the embryo developed and showed its maximum at stage V. An increase in the synthesis of triacylglycerols and diacylglycerols was also observed at this stage. Diacylglycerylethers were synthesized more actively during the first stages of development. The higher specific activity observed in total homogenate than in microsomal fraction suggested that their synthesis was not exclusively microsomal. Phospholipid synthesis was very active all along development, reflecting active membrane biosynthesis. The highest activity of the cytosolic triacylglycerol lipase was observed at stage V. Fatty acid degradation, measured as mitochondrial beta-oxidation activity, did not vary significantly during development. We conclude that both the anabolic and catabolic processes concerning lipid metabolism are very active, with values similar to those described for adult hepatopancreas, revealing the major role of lipids during shrimp embryogenesis energetics, and that the highest activities of lipid synthesis-hydrolysis take place at stage V when embryos are under active organogenesis. The effect of vitamin A deficiency on the functional integrity of the reticuloendothelial system and the phagocytic capacity of circulating polymorphonuclear leukocytes was evaluated in retinoate-cycled vitamin A-deficient rats under conditions such that secondary dietary imbalances were eliminated. Kinetics of blood clearance of 2 X 10(7) Escherichia coli injected intravenously was depressed within 8 days of the withdrawal of retinoic acid; all animals were profoundly affected by Day 12 of deficiency. In vitro, the phagocytic activity of polymorphonuclear leukocytes was similarly affected; by Day 12 of deficiency, phagocytic capacity in all deficient animals was less than 40% of the appropriate control values (P less than 0.01). Animals rendered vitamin A deficient by this procedure also displayed marked susceptibility to endogenous bacterial infection, as judged from the proportion of deficient rats that spontaneously developed bacteremia during the later stages of deficiency. These data together demonstrate unequivocally that reticuloendothelial and polymorphonuclear leukocytic functions are impaired in vitamin A deficiency in the absence of other dietary imbalances.  相似文献

该文选取桂林岩溶石山檵木群落不同恢复阶段(灌木阶段、乔灌阶段和小乔林阶段)作为研究对象,探究凋落物层酶对凋落物分解速率的影响。结果表明:不同恢复阶段凋落物经1 a分解后,凋落物剩余率分别为灌木阶段(59.58%)、乔灌阶段(61.79%)和小乔林阶段(62.02%)。不同恢复阶段凋落物分解速率随演替的进行而减小。3个不同恢复阶段凋落物层多酚氧化酶、脲酶、蔗糖酶活性均在12月份最低,多酚氧化酶活性均在3月份最高,脲酶和蔗糖酶活性均在6月份最高。3个恢复阶段纤维素酶活性变化规律趋势一致,均在6月份酶活性最高,灌木阶段纤维素酶活性在3月份最低,乔灌阶段和小乔林阶段纤维素酶活性均在9月份最低。3个不同恢复阶段的凋落物层酶活性在不同时期均表现为蔗糖酶脲酶纤维素酶多酚氧化酶。不同恢复阶段凋落物层酶活性对凋落物分解速率影响不同。灌木阶段凋落物层蔗糖酶活性与分解速率呈显著正相关(P 0.05),乔灌阶段脲酶活性与分解速率呈显著正相关(P 0.05),小乔林阶段各酶活性与分解速率相关不显著。蔗糖酶、脲酶和多酚氧化酶是影响灌木阶段凋落物分解速率的重要因素,脲酶、纤维素酶和多酚氧化酶是影响乔灌和小乔林阶段分解速率的重要因素。  相似文献

Data are presented on the cytokinin status of seeds and seed components, at different stages of development in Phaseolus coccineus L., as determined with the soybean callus growth bioassay: A change in cytokinin types according to developmental stage occurred: from biologically very active less polar types (zeatin=Z) at early stages to more polar types (zeatin glucoside=Z9G and zeatin riboside=Zr), with relatively low biological activity, at intermediate and late stages of seed development: When cytokinins were analyzed separately in embryos (embryo proper) and suspensors at two embryonic stages: heart-shaped (A) and middle cotyledonary embryos (stage B) respectively, it was found that: i) at stage A, the suspensor showed cytokinin activity at the level of Z, 2iPA (2-isopentenyladenosine) and Zr, whereas more polar cytokinins (Z9G, Zr) were present in the embryo; ii) at stage B, when the embryo seems to become autonomous for cytokinin supply, there was a relative abundance of active cytokinins (Z, 2iPA) in the embryo to which Z9G activity in the suspensor corresponded. Abstract— The activities of three enzymes–cerebroside sulphotransferase, 3'-phospho-adenosine 5'-phosphosulphate synthesizing enzyme and arylsulphatases A and B have been studied in various developmental and recovery stages of experimental allergic encephalomyelitis. The concentrations of cerebroside and sulphatide were also analysed during these stages. It was observed that the sulphatide concentration decreased during the development of the disease, with a concurrent increase in the activity of arylsulphatase and vice versa during the recovery stages. 3'-Phosphoadenosine 5'-phosphosulphate synthesis as well as sulphotransferase activity increased during the pre-acute stage of the disease, reached a maximum at the acute stage and decreased during recovery stages.  相似文献

It was found that calcium exchange disturbances under vitamin E deficiency is due to changes in the metabolism of vitamin D. In vitamin E-deficient rats the serum blood levels of hydroxyvitamin D (25-OHD) showed no significant changes, whereas the concentration of the hormonal form of 1.25-hydroxyvitamin D [1.25(OH)2D], decreased by 40%. In vitro studies showed that the 25-hydroxylase D3 activity in the livers of rats with E-avitaminosis had a tendency to decrease (by 22%), whereas that of 24-hydroxylase dropped drastically (by 52%). The serum blood levels of the parathyroid hormone (PTH) and kidney levels of cAMP under E-avitaminosis were significantly lowered. Preincubation of kidney slices with the adenylate cyclase activator, forskolin, increased the activity of 1-OHase in about the same degree as that in vitamin E-rich rats. The free radical scavenger, BHT, added to kidney slices suppressed the activity of the both enzymes; this finding testifies to the low O2-binding affinity of these monooxygenases. The content of 1.25(OH)2D3 receptors occupied in vivo in the kidneys of vitamin E-deficient rats decreased 2.5-fold; however, the binding of 1.25(OH)2D3-receptor complexes to heterologous DNA was unaffected thereby. The vitamin deficiency in vivo results in the inhibition of vitamin D metabolism in the liver and kidney concomitant with the formation of active metabolites and decreases the concentration of hormone-receptor complexes in target tissues.  相似文献

目的探讨肠道菌群和粪便炎性标志物在炎症性肠病(IBD)活动度评估中的临床价值。方法共纳入120例IBD患者为研究组,其中溃疡性结肠炎(UC)患者68例,克罗恩病(CD)患者52例。选择30例经结肠镜检查正常的健康体检者为对照组。采集全部研究对象的新鲜粪便标本进行粪便细菌培养及炎性标志物检测,比较不同疾病活动度IBD患者的肠道菌群及粪便钙卫蛋白(FC)、乳铁蛋白(LF)、基质金属蛋白酶-9(MMP-9)、髓过氧化酶(MPO)水平的变化。结果与对照组比,UC和CD患者肠道中肠杆菌、肠球菌、拟杆菌、消化球菌及酵母菌数量均明显增加(P0.05),双歧杆菌、乳杆菌及真杆菌数量明显减少(P0.05)。UC患者梭菌数量较对照组增加(P0.05),CD患者梭菌数量较对照组减少(P0.05)。UC、CD活动期患者肠杆菌、肠球菌、拟杆菌、消化球菌及酵母菌数量明显多于缓解期患者(P0.05),双歧杆菌、乳杆菌及真杆菌数量明显少于缓解期患者(P0.05),且重度活动期患者肠道菌群改变较轻、中度活动期改变更明显(P0.05)。UC活动期患者梭菌数量明显多于缓解期(P0.05),CD活动期患者梭菌数量明显少于缓解期(P0.05)。UC和CD患者粪便中FC、LF、MMP-9及MPO水平均显著高于对照组(P0.05)。UC、CD活动期患者FC、LF、MMP-9及MPO水平显著高于缓解期患者(P0.05),且重度活动期患者高于轻、中度活动期患者(P0.05)。结论肠道菌群变化和粪便中FC、LF、MMP-9及MPO水平可作为IBD患者疾病活动性评估的辅助指标。  相似文献

选用200羽14日龄健康AA肉鸡,以电子自旋共振(ESR)捕集法和生物化学法对肉仔鸡血液和组织器官的不同自由基进行直接或间接测定,探讨VE和Se对肉鸡不同自由基代谢的作用及其动态变化.结果表明:组织一氧化氮(NO)自由基水平随日粮VE含量升高而降低,二者呈负相关关系,日粮高水平Se有诱导产生NO自由基的倾向;高VE和Se日粮显著提高血清和肝脏中SOD和GSH-Px的活性,但随处理时间的延长,组织SOD活性逐渐降低,而GSH-Px活性逐渐升高,说明日粮VE和/或Se不足均会诱导机体产生O.2-、H2O2自由基,且O2.-自由基会持续大量产生,而H2O2自由基仅在缺乏初期大量产生,而后趋于缓和;低VE和/或低Se均显著提高组织MDA含量,且低Se比低VE更为显著.VE和Se对肉鸡NO、O.2-和H2O2自由基代谢的作用存在协同效应.  相似文献

DNA-dependent RNA polymerase has been purified from Bacillus subtilis at various stages of vegetative cell growth. Polymerase isolated from cultures approaching the end of the logarithmic growth phase was associated with a 60,000-dalton polypeptide and was only 10–20% as active as polymerase isolated from rapidly growing cells. Appearance of this new polypeptide and the change in template activity occur prior to stage 0 of sporulation.  相似文献

Summary The activity of RN-ase at zero zinc level was found to be 2, 173-1 and 2 times higher in all the varieties of rice (Jaya and HM484) and Maize (Ganga 5 and Vijay) than that of 5 ppm zinc at 15/20 days, 30/40 days and 45/60 days growth stages respectively, but the difference in the zinc content of leaves between two treatments was not found to be significant except that of 30/40 days growth stage. Similarly the potash content in both the treatments did not vary, but the dry matter production of whole plant and protein content of leaves except 15/20 days growth stage varied significantly. The data suggest thirty ppm P released per 100 g protein per hour (RN-ase activity) could be taken as threshold value above which hidden deficiency at an early growth stage is detected. Also an average concentration of 27 and 25 ppm zinc in rice and maize leaves respectively could be taken as critical limit for zinc deficiency at their 15/20 days of growth respectively. On the basis of RN-ase activity the hidden hunger of zinc in crop can be predicted at an early growth stage (15/20 days) where the zinc content in plant tissue fail to reveal the true picture of zinc deficiency.  相似文献

Mucosal cells of the small intestine obtained from rats deprived of vitamin D or given excessive amounts of the vitamin accumulated significantly more calcium than did cells from control animals. 2. Mucosal cells from vitamin D-deficient rats released less calcium than did cells from normal or hypervitaminotic D animals. 3. Studies in vivo showed that the transfer of (45)Ca from the intestine to the blood was delayed in vitamin D deficiency, but was accelerated in hypervitaminosis D. 4. The findings support the thesis that vitamin D is involved in the release of calcium rather than in its uptake by mucosal cells. 5. Further evidence is presented suggesting that uptake of calcium by intestinal mucosal cells at 0 degrees is primarily passive, whereas at 38 degrees uptake and release are effected by an active process that depends on energy derived from glycolytic activity.  相似文献

Summary Gibberellins and auxins were extracted from embryos and suspensors of Phaseolus coccineus L. at two stages of development: A) heart-shaped embryo and B) cotyledonary embryo with suspensor in the initial stage of degeneration. The time interval between the two stages was 5–6 days.In both embryos and suspensors, gibberellin (GA)-like activity was found in three fractions: F-1 (ethyl acetate fraction at pH 8.0), F-2 (free GAs) and F-3 (bound GAs). At stage A, the total GA activity in the suspensor was about 30 times greater than in the embryo and the bound GAs contributed by about 90% to the total GA content. A dramatic decrease in level of bound GA-like substances was found in suspensors at stage B, when the level of total GAs in the embryo had increased to 10 times that at stage A. This might suggest a transport of GAs from the suspensor to the embryo. In both embryo and suspensor, qualitative changes in GAs with shift in activity of the fractions tested occurred at the two developmental stages.The methanolic extracts of stage A suspensors showed two inhibitors, one much more active than the other, and two large peaks of growth promoting activity at Rf 0.4–0.7; in stage A embryos, the general activity of the extracts was lower and the promoting effect was spread over Rf 0.3–0.9.The present results seem to support the view that the suspensor plays a role in embryogenesis by acting as a site of synthesis of growth regulators needed by the embryo.Abbreviations F-1 ethyl acetate fraction at pH 8.0 - F-2 free gibberellins - F-3 bound gibberellins - GA gibberellic acid - Stage A heart-shaped embryo - stage B cotyledonary embryo with suspensor in the initial stage of degeneration  相似文献

研究了FACE条件下(CO2浓度增加200μmol·mol^-1)水稻、小麦不同生育期0~10cm土层土壤脲酶、磷酸酶、芳基硫酸酯酶、脱氢酶活性的变化.结果表明,FACE条件下,土壤脲酶活性在冬小麦生育前期低于对照,在孕穗期高于对照;在水稻生育前期高于对照,在成熟期低于对照.磷酸单酯酶活性在冬小麦生育期高于对照;在水稻分蘖期高于对照,在生育后期(拔节期、抽穗期和成熟期)低于对照.芳基硫酸酯酶活性在小麦越冬期和孕穗期低于对照,在分蘖期和成熟期高于对照;在水稻生育期间均高于对照.脱氢酶活性在小麦和水稻的生育前期低于对照,在后期高于对照.  相似文献

The effects of phosphorus deficiency on carbohydrate fractions of Mentha arvensis L. var. piperascens were studied. Mint plants were grown in sand cultures under full nutrition and phosphorus deficiency conditions. Component organs, viz., leaf, stem and root, were sampled at four different stages of the growth cycle and analysed for various sugar fractions. In leaf and root phosphorus deficiency brought about a definite increase in all sugar fractions whereas in stem a reduction was noticed. The stem appeared to be the principal storage organ throughout the growth. Maximum sugar concentration was recorded at the age of 70 days, which represents the full maturity stage, and coincided with maximum essential oil accumulation.  相似文献
