BACKGROUND AND AIMS: Nuclear DNA amounts of 12 diploid and one tetraploid taxa and 12 natural interspecific hybrids of Cirsium from 102 populations in the Czech Republic, Austria, Slovakia and Hungary were estimated. METHODS: DAPI and PI flow cytometry were used. KEY RESULTS: 2C-values of diploid (2n = 34) species varied from 2.14 pg in C. heterophyllum to 3.60 pg in C. eriophorum (1.68-fold difference); the 2C value for the tetraploid C. vulgare was estimated at 5.54 pg. The DNA contents of hybrids were located between the values of their putative parents, although usually closer to the species with the smaller genome. Biennial species of Cirsium possessed larger nuclear DNA amounts than their perennial relatives. Genome size was negatively correlated with Ellenberg's indicator values for continentality and moisture and with eastern limits of distribution. A negative relationship was also detected between the genome size and the tendency to form natural interspecific hybrids. On the contrary, C-values positively corresponded with the spinyness (degree of spinosity). AT frequency ranged from 48.38 % in C. eriophorum to 51.75 % in C. arvense. Significant intraspecific DNA content variation in DAPI sessions was detected in C. acaule (probably due to the presence of B-chromosomes), and in tetraploid C. vulgare. Only the diploid level was confirmed for the Pannonian C. brachycephalum, generally considered to be tetraploid. In addition, triploidy was discovered for the first time in C. rivulare. CONCLUSIONS: Considerable differences in nuclear DNA content exist among Central European species of Cirsium on the diploid level. Perennial soft spiny Cirsium species of wet habitats and continental distributions generally have smaller genomes. The hybrids of diploid species remain diploid, and their DNA content is smaller than the mean of the parents. Species with smaller genomes produce interspecific hybrids more frequently.  相似文献

The pairing of polytene chromosomes was investigated in Drosophila melanogaster, Drosophila simulans and their hybrids as well as in species of the D. virilis group and in F1 hybrids between the species of this group. The study of frequency and extent of asynapsis revealed non-random distribution along chromosome arms both in interspecific hybrids and pure Drosophila species. It is suggested that definite chromosome regions exhibiting high pairing frequency serve as initiation sites of synapsis in salivary gland chromosomes.  相似文献

Differences in the isozyme patterns of Xenopus laevis and Xenopus mulleri have been utilized to examine the expression of alleles of both species in hybrid animals. Mitochondrial MDH and tetrazolium oxidase phenotypes were examined during the development of non-hybrid embryos of each species and of reciprocal hybrids. Early stages of the hybrids resemble the enzyme phenotype of the maternal parent. Appearance of paternal enzyme takes place just prior to the active feeding tadpole stage for both mitochondrial MDH and oxidase. The maternal effect disappears shortly thereafter in early feeding tadpoles, at which point reciprocal hybrids have identical isozyme patterns. There is no evidence for a predominance of one species over the other. Examination of feeding tadpoles and adult toads indicates that both laevis and mulleri expression is stable. The appearance of paternal mitochondrial MDH does not correspond to the time when other mitochondrial components begin to increase in Xenopus. Multiple bands of MDH in both species and of oxidase in laevis are probably not due to the aggregation of subunits produced by different alleles at the same locus. There is no evidence for the formation of “hybrid” molecules consisting of subunits of both species.  相似文献

Hybrids between the two diploid species Festuca donax and F. drymeja had regular chromosome association, forming 7 bivalents at metaphase I. However, when the two species were crossed with a third species F. scariosa, the hybrids involving F. drymeja showed greater desynapsis than those with F. donax. When the F1 hybrid F. donax × F. drymeja was crossed with F. scariosa, the trispecific progeny could be grouped into three classes according to the degree of desynapsis recorded. Abnormalities associated with the fusion of pollen mother cells, which produced highly polyploid cells, were also observed in the trispecific hybrids. Failure of chromosomal pairing and the occurrence of syncytes is attributed to genotypic interactions.  相似文献

Growth and reproduction of Akodon molinae (A.m.), A. dolores (A.d.) (Rodentia: Cricetidae), and their hybrids are described. A.d. showed less fertility under animal room conditions, fewer "successful" matings, similar litter sizes, and an altered sex proportion as compared to A.m.; A.d. ovulatory index is supposed to be higher than mean litter size. X-linked lethal gene(s) and chromosome polymorphisms are possible causes of these variations. Interspecific crosses showed a marked seasonal (summer) tendency in parturitions. Intraspecific matings also produced young during winter. F1 hybrids showed a reversed tendency, while F2 matings returned to the parental pattern. A.d. male X A.m. female and F1 crosses presented larger litter sizes than the other pairings. A.m. exhibited sexual differences in weight after 60 days of age. A.d. and F1 hybrids did not show significant differences in weight of both sexes at any age. A.m. males and females were heavier than A.d. individuals except 12 months after birth. F1 hybrids born to A.m. male were heavier at birth than the A.d. female s interspecific offspring, and may be interpreted as a maternal influence of the A.m. females. Hybrids were heavier than their parents at birth and some of them were also heavier at 21 days, and probably is due to pseudoheterosis. The comparison of body measurements between sexes of both species did not give conclusive results. A.m. females were larger than females of A.d. Males of both species showed similar total lengths; A.m. males had larger body lengths and shorter tails. Body measurements were not related to weight variations. However, differences in weight and measurements between sexes appeared simultaneously at 2-6 months. This probably reflected the attainment of maturity.  相似文献

Relative amounts of DNA were determined by Feulgen cytophotometry in 22 diploid species of Ranunculaceae (n=7, 8, 9) representing six genera, and exhibiting large differences in chromosome size, but no marked differences in karyotype pattern. Chemical determination of absolute amounts of DNA for six of these species, allowed conversion of all the photometric data into absolute units of DNA. The mean DNA content per nucleus varied from.13×10–11gm in Aquilegia to 5.25×10–11gm in species of Anemone in the section Homalocarpus. The DNA values obtained appeared to be quantized, and data for the majority of species fitted a non-geometrical series with the observed relative terms: 1—8—12—16—20—24—40. The magnitude of these variations in DNA content, the preservation of the karyotype and the tendency towards a simple numerical progression in DNA values, lead us to prefer an interpretation of the evolution of DNA content in terms of differential polynemy to one postulating changes in size of genetic units in an unchanging number of strands per chromosome.  相似文献

Nucleolar number variation has been investigated in root tip cells by cytologically determining the number of nucleoli per cell in autoploids and alloploids of Hordeum species, their haploids and interspecific hybrids. The nucleolus organisers in autoploid types of H. vulgare or H. bulbosum did not show any alterations irrespective of the ploidy level. The nucleolar number variation in these species results from a definite pattern of fusion and the maximum number of nucleoli per nucleus corresponds to the number of secondary constrictions. Nucleolus formation in alloploids and interspecific hybrids is impaired on some of the NORs, suggesting differential amphiplasty or nucleolar dominance. A comparison of nucleolar formation in the alloploid species (brachyantherum, arizonicum, procerum and parodii), their haploids, and the interspecific hybrids revealed different degrees of variation from the expected mean and maximum numbers of nucleoli. While the deviations in hybrids between alloploids (H. arizonicum and H. brachyantherum or H. procerum and H. brachyantherum) are marginal, nucleolar dominance is more pronounced in hybrids involving H. vulgare or H. bulbosum as one of the parents and is invariably associated with the disappearance of the secondary constriction(s) from the NOR(s) contributed by one of the parents, and the number of nucleoli is appropriately reduced.  相似文献

The analysis of 57 synthetic interspecies hybrids revealed that Saccharomyces cerevisiae and Saccharomyces uvarum ( Saccharomyces bayanus var. uvarum) are isolated by a double sterility barrier: by hybrid sterility (hybrid cells cannot produce viable spores) operating in allodiploids and by F1 sterility (F1 cells cannot produce viable spores) operating in allopolyploids. F1-sterility is caused by mating-type heterozygosity. It can be overcome by eliminating chromosome 2 of the S.?uvarum subgenome that carries a MAT locus. The loss of this MAT gene abolishes the repression of mating activity. In cultures of the resulting fertile alloaneuploid F1 segregants, the cells can conjugate with each other like haploids and form zygotes capable of performing meiotic divisions producing viable and fertile F2 spores. To the best of our knowledge, this is the first report on breaking down interspecies hybrid sterility by chromosome loss in eukaryotic organisms. The filial generations are genetically unstable and can undergo additional changes mainly in the S.?uvarum subgenome (directional changes). It is proposed that regaining fertility and subsequent preferential reduction in one of the subgenomes may account for the formation of chimerical ('natural hybrid') genomes found among wine and brewery strains and may also play roles in speciation of hybrid taxa in the Saccharomyces genus.  相似文献

As an initial step in establishing interspecific hybridization to broaden the genetic basis of lentils [Lens culinaris ssp.culinaris (Medikus) Williams], a set of experiments was carried out to produce an efficient in vitro protocol for propagation of lentil and two of its wild relatives (Lens ervoides andLens culinaris ssp.orientalis). The objective of the experiments was to optimize the media (Murashige and Skoog) to regenerate shootsin vitro from nodal segments without a callogenic phase. The number of shoots per explant, the number of nodes per shoot and shoot length showed that species differences, gibberellic acid and benzyladenine levels had the largest effects, with only minor interaction effects. The experiments therefore identified a standard protocol which gave the optimum levels of growth regulators, Murashige and Skoog (MS) salts and sucrose concentrations for maximum plant regeneration from the nodal segment of these species. The medium recommended for optimal shoot regeneration without a callogenic stage contained 2.89 μM GA3 in combination with 1.11 μM BA in MS medium lacking sucrose. The optimal medium for root induction on these shoots had the MS medium supplemented with 5.37 μM NAA. Final successful establishment of regenerated plants was completed by the transfer to a third medium containing half-strength MS salts.  相似文献

Summary Two species of ground crickets, Allonemobius fasciatus (from Corvallis, 44.3 N) and A. socius (from Gainesville, 30.3 N), and their hybrids were compared for nymphal development, wing form, and embryonic diapause. A. fasciatus matured more rapidly at a short photoperiod than at a long photoperiod and also than A. socius which showed no developmental response to photoperiod. The proportion of long-winged forms varied with nymphal photoperiod greatly in A. fasciatus but slightly in A. socius. A. fasciatus laid only diapause eggs, while A. socius responded to a long parental photoperiod and a high incubation temperature by averting diapause. A. fasciatus showed temperature-dependent shift of the diapause stage butA. socius entered diapause only at the early stage. These differences are related to their voltinism and distribution.Reciprocal crosses between the two species produced viable hybrids. In development rate, female hybrids were intermediate between their parents while male hybrids were closer to their mothers, suggesting that the development rate is controlled primarily by the X chromosome. Overdominance occurred in wing form; reciprocal crosses produced either higher or lower proportions of long-winged forms than their parents. Diapause of F1 hybrid embryos was determined only by the female parent and the paternal effect appeared in F2 embryos.  相似文献

The aim of our study was to verify, on the basis of statistical analyses of nine quantitative morphological features of pollen grains, the hypothesis that pollen grains of three parental species of Crataegus (C. laevigata, C. monogyna, C. rhipidophylla) differed from the pollen of three spontaneous hybrids of these species (C. × macrocarpa, C. × media, C. × subsphaericea). Contrast analysis revealed that a majority of the pollen features of hybrid species were characterized by significantly higher mean values than those of parental species. Analysis of pollen shape classes indicated that the parental species clearly differed from each other in contrast with hybrids, which were characterized by a similar proportion of pollen in individual pollen shape classes. Statistical analyses showed that the pollen grains of two parental species, C. laevigata and C. monogyna, were most similar to one another. Pollen grains of typical C. rhipidophylla were similar to the pollen of hybrids and the mean values of almost all studied pollen features [P, E, Exp, Exp/P, Le, d, d/E (PAI)] of C. rhipidophylla var. rhipidophylla were intermediate between those of C. monogyna and C. rhipidophylla var. lindmanii. This corroborates Zieliński's conjecture that C. rhipidophylla is probably an old, conserved hybrid between C. monogyna and C. calycina (= C. lindmanii = C. rhipidophylla var. lindmanii). According to the analysis of canonical variables, C. × macrocarpa and C. × media pollen grains were most similar. Biochemical variation at 14 blood loci was reviewed, and specific features compared experimentally in sheep Ovis aries, mouflon Ovis musimon, goat Capra hircus, aoudad Ammotragus lervia and in 2 stillborn aoudad × goat hybrids. Variation at 3 loci was also studied in dall sheep Ovis dalli, bighorn sheep Ovis canadensis and rocky mountain goat Oreamnos americanus. Haemoglobin C production in an anaemic Hb AB mouflon and in mouflon × sheep hybrids was examined. Mouflon differ from domestic sheep in that synthesis of both Hb βAHb βBchains is switched off during Hb C production. The mouflon × sheep hybrids switched off one or both chains depending on whether they had inherited sheep or mouflon Hb β chain genes. In general aoudad showed a closer affinity to goats than to sheep.  相似文献

Natural hybridization commonly produces individuals with intermediate morphological and genetic characteristics, but their response to environmental stress is still uncertain, with some studies showing that transgressive performance would be common. Prosopis chilensis and Prosopis flexuosa are the most important tree species from Arid Chaco, South-America. Both species occupy different ecological niches in terms of water availability. Genetic and morphological studies have demonstrated the existence of interspecific hybrids in contact areas between these species. Hybrids are characterized by clear intermediate morphological characteristics, which have taxonomical value, and genetic structure compared to both parental species. We studied mechanisms implicated in drought stress tolerance in seedlings of P. chilensis, P. flexuosa and their interspecific hybrids trying to elucidate if hybrids have a morpho-physiological, growth and survival intermediate response to drought compared to differential parental responses or if they out-perform both parental species when subjected to drought. Our results suggest that hybridization does not result in individuals with intermediate mechanisms related to drought resistance, but with a unique trait combination leading to high growth when water availability is high (similar to the most vulnerable parental species) and high survival under drought stress (similar to the more resistant parental species). Certain uncoupling between symplastic and apoplastic resistence to drought was observed in hybrids, as well as decreased physiological-wood anatomical plasticity compared to parental species. The long-term consequences in terms of adaptive response to drought of this particular trait combination of hybrids remain still unknown.  相似文献

If sexual selection is to result in speciation, traits involved in mate choice within species need to be capable of producing sexual isolation between species. We investigated the association between mate choice and sexual isolation using interspecific hybrids between two sibling species, Drosophila serrata and Drosophila birchii. A perfuming experiment demonstrated that olfaction was involved in the sexual isolation between the two species. A quantitative genetic analysis using 30 populations of hybrids between the two species indicated that mating success in hybrid individuals was predominately determined by cuticular hydrocarbons; the average genetic correlation between mating success and cuticular hydrocarbon profile was 0.84, and in some instances exceeded 0.95. Multivariate analysis of the cuticular hydrocarbons of the two species revealed that there were three independent blends of cuticular hydrocarbons that separated three levels of organization: species, sex, and sex within species. The hydrocarbons used by hybrids in mate choice included those that separated the two species, demonstrating that species-specific characters may be used in mate choice within populations. The interspecific reciprocal cross had major effect on which cuticular hydrocarbons were associated with mating success, indicating that the expression of the cuticular hydrocarbons was strongly sex linked.  相似文献

Background and Aims

Floral traits, such as floral volatiles, can contribute to pre-zygotic reproductive isolation by promoting species-specific pollinator foraging. When hybrid zones form, floral traits could also influence post-zygotic isolation. This study examined floral volatiles in parental species and natural hybrids in order to explore potential scent mediation of pre-zygotic and post-zygotic isolation.

Methods

Floral bouquets were analysed for the sister species Ipomopsis aggregata and I. tenuituba and their natural hybrids at two contact sites differing in both hybridization rate and temporal foraging pattern of hawkmoth pollinators. Floral volatiles were quantified in diurnal and nocturnal scent samples using gas chromatography–mass spectrometry.

Key Results

The bouquets of parental species and hybrids showed qualitative overlap. All flowers emitted similar sets of monoterpenoid, sesquiterpenoid, aliphatic and benzenoid compounds, but separated into groups defined by multivariate analysis of quantitative emissions. The parental species differed most strikingly in the nitrogenous compound indole, which was found almost exclusively in nocturnal bouquets of I. tenuituba. Natural hybrid bouquets were highly variable, and showed emission rates of several compounds that appeared transgressive. However, indole emission rates were intermediate in the hybrids compared with rates in the parents. Volatile bouquets at the contact site with lower hybridization did not show greater species specificity in overall scent emission, but I. tenuituba presented a stronger indole signal during peak hawkmoth activity at that site.

Conclusions

The two species of Ipomopsis differed in patterns of floral bouquets, with indole emitted in nocturnal I. tenuituba, but not in I. aggregata. Natural hybrid bouquets were not consistently intermediate between the parents, although hybrids were intermediate in indole emission. The indole signal could potentially serve as a hawkmoth attractant that mediates reproductive isolation both before and after hybrid formation.  相似文献

Giemsa C-banding patterns in two species of mosquitoes, Aedes aegypti and Aedes mascarensis, their hybrids and backcross progeny revealed differences in the sex chromosome pair. In A. aegypti, the female determining or the m chromosome in both males and females shows a conspicuous band in the centromere region and another band in one arm. The male determining or the M chromosome is devoid of any bands. Progeny of crosses involving A. aegypti females and A. mascarensis males showed interesting albeit unexpected results. The intercalary band was suppressed in both sons and daughters. When such F1 sons were backcrossed to A. aegypti females, a proportion of males developed into intersexes. These intersex progeny differed from the normal males in terms of their banding pattern. In the reciprocal cross (A. mascarensis female × A. aegypti male), the F1 and the backcross progeny yielded the expected C-banding patterns. The implications of the reversible expression of the intercalary band on the A. aegypti m chromosome and its relevance to genetic regulation are discussed.  相似文献

Despite the commonality and study of hybridization in plants, there are few studies between invasive and noninvasive species that examine the genetic variability and gene flow of cytoplasmic DNA. We describe the phylogeographical structure of chloroplast DNA (cpDNA) variation within and among several interspecific populations of the putative native, Carpobrotus chilensis and the introduced, Carpobrotus edulis (Aizoaceae). These species co-occur throughout much of coastal California and form several 'geographical hybrid populations'. Two hundred and thirty-seven individuals were analysed for variation in an approximate 7.0 kb region of the chloroplast genome using PCR-RFLP (polymerase chain reaction - restriction fragment length polymorphism) data. Phylogenetic analyses and cpDNA population differentiation were conducted for all morphotypes. Historic geographical dispersion and the coefficient of ancestry of the haplotypes were determined using nested clade analyses. Two haplotypic groupings (I and II) were represented in C. chilensis and C. edulis, respectively. The variation in cpDNA data is in agreement with the previously reported allozyme and morphological data; this supports relatively limited variation and high population differentiation among C. chilensis and hybrids and more wide-ranging variation in C. edulis and C. edulis populations backcrossed with C. chilensis. C. chilensis disproportionately contributes to the creation of hybrids with the direction of gene flow from C. chilensis into C. edulis. The cpDNA data support C. chilensis as the maternal contributor to the hybrid populations.  相似文献
