Soybean [Glycine max (L.) Merrill] seeds and cotyledons weregrown in an in vitro culture system to investigate the relationshipsbetween cell expansion (net water uptake by the seed) and drymatter accumulation. Seeds or cotyledons grown in a completenutrient medium containing 200 mol m–3 sucrose continueddry matter accumulation for up to 16 d after in planta seedsreached physiological maturity (maximum seed dry weight). Seedor cotyledon water content increased throughout the cultureperiod and the water concentration remained above 600 g kg–1fresh weight. These data indicate that the cessation of seeddry matter accumulation is controlled by the physiological environmentof the seed and is not a pre-determined seed characteristic.Adding 600 mol m–3 mannitol to the medium caused a decreasein seed water content and concentration. Seeds in this mediumstopped accumulating dry matter at a water concentration ofapproximately 550 g kg–1. The data suggest that dry matteraccumulation by soybean seeds can continue only as long as thereis a net uptake of water to drive cell expansion. In the absenceof a net water uptake, continued dry matter accumulation causesdesiccation which triggers maturation. Key words: Glycine max (L.) During in vitro growth and maturation of soybean seeds, cessationof embryo growth and dry weight accumulation occurred in thepresence of abundant C and N nutrients. Axis followed by cotyledontissues changed from green to yellow, and post-harvest germinationpotential declined if cultured after yellowing of axis tissues.A tissue specific accumulat;on of methanol occurred during thein vitro culture of immature seeds (i.e. initially 50 to 70mg fresh weight) to maturity in liquid medium. Methanol accumulatedto 3.0 g m–3 or 50 µg seed–1 in the medium,while methanol decreased from 37 to about 3.0 µg g–1fresh weight in cotyledons. By contrast, axis tissues increased20-fold in methanol concentration to 90 µg g–1 during20 d in culture. Ethanol was present only in trace amounts inaxis tissues and medium. Addition of exogenous methanol vapourto in situ grown seeds during precocious maturation decreasedsubsequent seedling vigour and germination with increasing levelsof exposure. Methanol accumulation in axis tissues during thegermination phase was not correlated with high temperature andtissue water content treatments which simulated pre-harvestdeterioration of seeds. However, the accumulation of methanolduring in vitro seed development and maturation in liquid culturemay contribute to reduced post-harvest germination performance. Responses to sucrose and glutamine by soybean embryos grown in vitro   总被引:2,自引:0,他引:2  
Immature soybean (Glycine max [L.] Merr. cv. Ransom) embryos were grown in vitro in the presence of different concentrations of sucrose and glutamine to examine how availability of carbohydrate and nitrogen affects dry matter accumulation and embryo composition. Embryos were transferred to fresh medium every 4 days to maintain sucrose and glutamine concentrations of the culture medium. In all experiments, accumulation of dry matter and protein content increased when the sucrose concentration of the culture medium was increased from 1.5 to 150 mM: however, a relatively greater enhancement of dry matter than of protein accumulation resulted in a lower protein concentration at 150 than at 1.5 mM sucrose. Both content and concentration of protein were increased by the increases in glutamine supply to concentrations exceeding 68% protein at 120 mM glutamine. In combination with 150 mM sucrose, however, oil increased as glutamine supply was increased from 0.6 to 6 mM and then decreased as glutamine supply was increased from 6 lo 120 mM. Varying the concentration of sucrose available during seed development also affected embryo composition. Decreased availability of sucrose during either the early or late portion of the culture period resulted in lower accumulation of dry mailer as well as oil. Protein concentration was actually higher for embryos transferred from 150 to 1.5 nM sucrose than for those remaining in 150 mM throughout the culture period: however, the greater percentage of protein was due lo a decrease in accumulation of dry weight. In addition, embryo composition was affected by altering the availability of glutamine during culture, indicating that variation in the level of nitrogen assimilate delivered during seed development can change embryo composition. Decreasing the glutamine concentration of the medium lowered both protein and oil content. In contrast, increasing the glutamine concentration of the medium from 0.6 to 6 mM 8 days after initiation of culture increased the protein content and concentration of the embryo while oil content was not affected.  相似文献

The effects of differemt S and methionine regimes on growthof developing Vicia faba cotyledons in vitro were studied. Basalmedium (containing adequate S) supplemented with 05 mM methioninemarginally increased d. wt and uncombined amino acid accumulationbut adding 1–5 mM methionine inhibited both growth andprotein accumulation. Sulphur deficiency reduced both d. wtand protein accumulation but incresed accumulation of uncombinedamino acids. Adding 1 mM methionine to the S-deficient mediumrestored growth, normal protein and uncombined amino acid acnunulation.High sulphate medium (7.5 mM ) decreased d. wt, protein anduncombined amino acid accumulation. High sulphate medium or basal medium+methionine (05 mM) changedthe proportions of the seed proteins; legumin increased butvicilin decreased. Sulphur deficiency caused a relative increasein vicilin but a decrease in legumin. The different S and methionineregimes markedly changed the composition of the uncombined aminoacids, especially those derived from aspartic acid but not thecomposition of the protein fraction, except during S deficiency. The data presented indicates a flexibility in the storage proteincomposition of developing cotyledons grown in vitro, with theS and methionine status having a regulatory effect. Soybeans (Glycine max L. Merrill) were grown in the greenhouseand in the field to investigate the effect of variations inthe assimilate supply during the linear phase of seed developmenton the rate and duration of growth of individual seeds. Increasedassimilate supplies, created by partial fruit removal, increasedrates of dry matter accumulation, duration of seed growth, andfinal seed size (weight per seed). Reductions in the supplyof assimilate to the developing seed, created by shading (60per cent) the plants during the linear phase of seed development,lowered seed growth rate but did not affect final seed sizebecause of a longer duration of seed growth. Nitrogen stressduring seed development, created by removing N from the nutrientmedium, did not affect seed growth rate but shortened the durationof seed growth and reduced final seed size. The data indicatethat the growth characteristics of soybean seed are influencedby the supply of assimilate to the seed during the linear phaseof seed development. An in vitro procedure promoting the rapid growth and proteinincrease of soya bean cotyledons has been developed. The amountof protein synthesized varied greatly depending on the nitrogen(N) source provided. Glutamine was the most effective N source,while inorganic forms of N were ineffective. Growth and proteinsynthesis were both more rapid in vitro than in vivo. Underthe best conditions, soya bean cotyledons increased 8-fold bothin dry weight and in protein in 6 days. The formation of the7S and 11S storage proteins in vitro was similar to that invivo. Hence, this in vitro culture method is appropriate forstudying legume seed storage protein synthesis under controlledconditions.  相似文献

Oil and protein are the most valuable components of soybean seed. Evidence indicates that growth and composition of soybean seed are controlled by supplies of carbon and nitrogen provided by the maternal plant to the seed, but it is difficult experimentally to control and quantify the precise amount of carbon and nitrogen provided to the seed by the whole plant. To examine whether oil and protein concentrations are affected by the supply of nitrogen to the seed, immature soybean seeds (Glycine max cv. Williams 82) were grown in vitro in nutrient solutions containing 20, 40, 60 or 80 mM of glutamine. The seeds were incubated in Erlenmeyer flasks for 8 days at 25°C. The rate of dry matter accumulation changed from 7.2 to 8.3 mg seed−1 day−1 as the glutamine concentration increased from 20 to 80 mM but the differences were not significant (P 0.05). Seed protein concentration increased as glutamine concentration increased from 294 mg g−1 at 20 mM glutamine to as high as 445 mg g−1 at 80 mM glutamine. Typical in vivo protein concentration of mature soybean seeds is about 400 mg g−1. Oil and protein concentrations were negatively correlated (r2= 0.44), which indicates that oil and protein synthesis are interrelated. Protein synthesis was favoured over oil synthesis when nitrogen became more abundant. The seeds used in this study clearly demonstrated a capacity to respond to nitrogen availability with changes in seed protein concentration.  相似文献

The effect of exogenous abscisic acid (ABA) on the rate of sucrose uptake by soybean (Glycine max L. Merr.) embryos was evaluated in an in vitro system. In addition, the concentrations of endogenous ABA in seeds of three soybean Plant Introduction (PI) lines, differing in seed size, were commpared to their seed growth rates. ABA (10−7 molar) stimulated in vitro sucrose uptake in soybean (cv `Clay') embryos removed from plants grown in a controlled environment chamber, but not in embryos removed from field-grown plants of the three PI lines. However, the concentration of ABA in seeds of the three field-grown PI lines correlated well with their in situ seed growth rates and in vitro [14C] sucrose uptake rates.

Across genotypes, the concentration of ABA in seeds peaked at 8.5 micrograms per gram fresh weight, corresponding to the time of most rapid seed growth rate, and declined to 1.2 micrograms per gram at physiological maturity. Seeds of the large-seeded genotype maintained an ABA concentration at least 50% greater than that of the small-seeded genotype throughout the latter half of seed filling. A higher concentration of ABA was found in seed coats and cotyledons than in embryonic axes. Seed coats of the large-seeded genotype always had a higher concentration of ABA than seed coats of the small-seeded line. It is suggested that this higher concentration of ABA in seed coats of the large-seeded genotype stimulates sucrose unloading into the seed coat apoplast and that ABA in cotyledons may enhance sucrose uptake by the cotyledons.

Much of the economic value of soybean (Glycine max) is based on the amount of protein and oil produced in the seeds. To examine the influence of temperature on seed oil and protein concentration, immature soybean seeds (cv. Williams 82) were grown in vitro at temperatures of 17°C, 21°C, 25°C, 29°C and 33°C. Dry growth rate (DGR) was calculated to be maximal at 23.7°C. Oil and protein concentration and seed growth rate did not show statistical difference (P > 0.05) within the temperature range from 21–29°C. Across all temperatures, however, a quadratic regression on oil concentration (R2 = 0.66) showed a minimum at 24.1°C and a quadratic regression on protein concentration (R2 = 0.59) showed a minimum at 24.3°C. Dilution by increased dry matter accumulation in the seed accounted for much of the variation in oil and protein concentration and the two concentrations were equally affected across temperatures. Consequently, oil and protein concentrations were positively related over the tested range of temperature. It was concluded that under these conditions the rate of dry matter accumulation by soybean seeds was critical in influencing seed oil and protein concentrations.  相似文献

The distribution of dry matter and various mineral nutrientsbetween testa and embryo of seeds of Lupinus albus and L. angustifoliusis described It was found that lupin seeds at either end ofa pod contained less dry matter and minerals than seeds in themiddle of the fruit. The transport of dry matter, N, P, K, S,Ca, Mg, Na, Fe, Zn, Mn and Cu from cotyledons of parent seedsof both species to the seedling axis was measured from germinationto the time of cotyledon death. N, P, K and S were retrievedfrom cotyledons with over 90 per cent efficiency, dry matter,Mg, Na, Fe, Zn, Mn and with 59–90 per cent efficiency,and Ca with 26–31 per cent efficiency. There was littlechange in the efficiency of nutrient retrieval from cotyledonswhen seedlings were grown in different culture media. Both speciesshowed a linear relationship between the loss of each elementand dry matter throughout the experiment, and a similar proportioningbetween root and shoot of the amount of a specific nutrientmobilized from cotyledons of parent seeds. Nitrogen (N) supply can limit the yields of soybean [Glycine max (L.) Merr.] in highly productive environments. To explore the physiological mechanisms underlying this limitation, seasonal changes in N dynamics, aboveground dry matter (ADM) accumulation, leaf area index (LAI) and fraction of absorbed radiation (fAPAR) were compared in crops relying only on biological N2 fixation and available soil N (zero-N treatment) versus crops receiving N fertilizer (full-N treatment). Experiments were conducted in seven high-yield environments without water limitation, where crops received optimal management. In the zero-N treatment, biological N2 fixation was not sufficient to meet the N demand of the growing crop from early in the season up to beginning of seed filling. As a result, crop LAI, growth, N accumulation, radiation-use efficiency and fAPAR were consistently higher in the full-N than in the zero-N treatment, leading to improved seed set and yield. Similarly, plants in the full-N treatment had heavier seeds with higher N concentration because of greater N mobilization from vegetative organs to seeds. Future yield gains in high-yield soybean production systems will require an increase in biological N2 fixation, greater supply of N from soil or fertilizer, or alleviation of the trade-off between these two sources of N in order to meet the plant demand.  相似文献

This study was conducted to evaluate the influence of brassinosteroid (24-epibrassinolide, EBL) seed priming and optimal nitrogen (N) supply in improving salt tolerance in soybean. The experimental treatments were (a) control (nutrient solution without N and without EBL priming), (b) nutrient solution without N and EBL seed priming, (c) N supplemented nutrient solution without EBL priming and (d) EBL seed priming + N supplemented nutrient solution under optimal (0 mM NaCl) and salt stress (0 mM NaCl) conditions. Salt stress caused significant reduction in growth and biomass accumulation of soybean. However, EBL seed priming and application of N improved the soybean performance under optimal and salt stress conditions. In this regard, treatments receiving both EBL and N were more effective. EBL priming and N, alone and in combination, triggered the accumulation of osmolytes including proline, glycine betaine and sugars resulting in better photo-protection through maintenance of tissue water content. Antioxidant activity and osmolyte accumulation significantly increased due to combined treatment of N and EBL under normal as well as salt stress conditions. In conclusion, salt stress caused reduction in growth and biomass soybean due to oxidative damage and osmotic stresses. However, soybean performance was improved by seed priming with EBL. Supplementation of N further improved the effectiveness of EBL treatment in improving salt tolerance in soybean.  相似文献

Water deficits during seed filling decrease seed size in soybean (Glycine max L.). This may result from a reduction in the supply of assimilates from the maternal plant and/or an inhibition of seed metabolism. To determine whether maternal or zygotic factors limited seed growth, we examined the effects of a plant water deficit on the supply of sucrose to and its utilization by developing embryos. Plants were grown in the greenhouse, and water deficits were imposed by withholding water for a period of 6 days during linear seed fill. When water was withheld, leaf water potential decreased rapidly, inhibiting canopy photosynthesis completely within 3 days. However, seed dry weight (nodes 7-11) continued to increase at or near the control rate. The level of total extractable carbohydrates in leaf, stem, and pericarp tissue decreased by 70, 50, and 45%, respectively, indicating that reserves were mobilized to support seed growth. Cotyledon sucrose content decreased from about 60 milligrams per gram dry weight to 30 milligrams per gram dry weight. Similarly, the concentration of sucrose in the interfacial apoplast of the cotyledons decreased from approximately 100 millimolar to 50 millimolar. However, the rate of sucrose accumulation by excised embryos, measured in a short-term in vitro assay, increased in response to the water deficit. These results indicate that both source and sink activity in soybean are altered by water deficits to maintain the flux of assimilates to the developing embryos. This may explain why seed growth is maintained, albeit for a shorter duration, when soybean is exposed to water deficits during the seed filling period.  相似文献

During 7 d of precocious maturation of soybean seed (Glycinemax), the starch content declined and soluble sugar levels increasedin patterns similar to natural seed dehydration and maturation.Total seed protein content and total seed dry weight increasedwhereas oil content remained relatively unchanged. Overall,the proportions of the constituents in precociously maturedseeds were comparable to naturally mature seeds. Precociouslymatured soybean seeds showed much the same germination and seedlinggrowth frequency patterns as naturally matured seeds. The effect of a substituted pyridazinone, 4-chloro-5-(dimethylamino)-2-phenyl-3(2H)-pyridazinone (San 9785), on the reduction of linolenate content was examined in the cotyledons of the soybean cv. Century and a low linolenate mutant of soybeans, C1640, cultured in vitro. No apparent changes in dry matter and total fatty acid accumulation were observed in the cotyledons developing in the presence of San 9785. However, a significant reduction of linolenate content with a corresponding increase in linoleate resulted from growth of the cotyledons in culture medium containing San 9785. San 9785 had a greater effect on decreasing the linolenate content in cotyledons excised in early developmental phases than later stages from both the wild type and mutant soybeans. This result supports other observations that the biosynthesis oflinolenate relative to the other major fatty acids of soybean cotyledons declines in later developmental stages. The compound becomes progressively less effective in reducing the content of linolenate during seed development in the mutant than in the wild type. Both San 9785 and the mutation result in changes in phosphatidylcholine molecular species which indicates the presence of a number oflinolenate desaturation systems in developing soybean cotyledons. Nitrogen (N) has been suggested as a determinant of seed production especially in species with high seed N content. Assuming that seed yield was determined as the balance between N demand and supply for seed production, we studied the effect of N fertilization after flowering on soybean (Glycine max L. Merr.) yield. Seed N concentration was nearly constant irrespective of N fertilization, indicating that seed production was proportional to the amount of N available for seed growth. N demand for seed production was analyzed as the product of seed number, the rate of N filling in individual seeds, and the length of the reproductive period. N fertilization increased seed number and the reproductive period, but did not influence the N filling rate. Seed number was positively correlated with dry mass productivity after flowering. Three N sources were distinguished: mineral N uptake, symbiotic N2 fixation and N remobilization from vegetative body. N fertilization increased N uptake and N remobilization, but lowered N2 fixation. We concluded that N availability in the reproductive period determined seed yield directly through increasing N supply for seed growth and indirectly through increasing seed N demand with enhanced plant dry mass productivity.  相似文献

The accumulation is described of N, P, K, S, Ca, Mg, Na, Fe,Zn, Mn and Cu in the developing pericarp and seed of two speciesof seagrass. Both species showed essentially the same patterns,which resemble those of herbaceous terrestrial plants. The general pattern of decrease of the 'critical' plant N concentration(i.e. minimum concentration required for maximum growth rate)during growth has been described for several C3 non grain-legumespecies, and this can be used as a reference curve for diagnosisof N nutrition in these species. The present study was undertakento investigate changes of N concentration during growth of agrain legume, in different conditions of N nutrition. Whitelupin (Lupinus albus L.) was grown for six crop seasons in fieldtrials in which inoculation with Rhizobium lupini, nitrogenfertilizer rate, cultivar and plant density were density weremanipulated. The yield and dry matter production of noninoculatedplants were lower than, or at the best similar to, those ofinoculated plants, whatever the level of N supply. From anthesisto the beginning of seed filling, the N concentration of shootsof inoculated plants was found to be remarkably stable betweenyears, N fertilization regimes, cultivars, and for individualplants within a plot. Nitrogen concentration only varied withplant density. By contrast, the N concentration of noninoculatedplants was highly variable and generally lower than that ofinoculated plants, whatever the level of N supply. The highand stable N concentration of inoculated plants did not appearto be necessary for maximum growth rate but seemed to be requiredfor maximum production of seed dry matter and N. Soybean seeds [Glycine max (L.) Merr.] synthesize de novo andaccumulate several non-storage, soluble polypeptides duringnatural and precocious seed maturation. These polypeptides havepreviously been coined ‘maturation polypeptides’.The objective of this study was to determine the fate of maturationpolypeptides in naturally and precociously matured soybean seedsduring rehydration, germination, and seedling growth. Developingsoybean seeds harvested 35 d after flowering (mid-development)were precociously matured through controlled dehydration, whereasnaturally matured soybean seeds were harvested directly fromthe plant. Seeds were rehydrated with water for various timesbetween 5 and 120 h. Total soluble proteins and proteins radio-labelledin vivo were extracted from the cotyledons and embryonic axesof precociously and naturally matured and rehydrated seed tissuesand analyzed by one-dimensional PAGE and fluorography. The resultsindicated that three of the maturation polypeptides (21, 31and 128 kDa) that had accumulated in the maturing seeds (maturationpolypeptides) continued to be synthesized during early stagesof seed rehydration and germination (5–30 h after imbibition).However, the progression from seed germination into seedlinggrowth (between 30 and 72 h after imbibition) was marked bythe cessation of synthesis of the maturation polypeptides followedby the hydrolysis of storage polypeptides that had been synthesizedand accumulated during seed development. This implied a drasticredirection in seed metabolism for the precociously maturedseeds as these seeds, if not matured early, would have continuedto synthesize storage protein reserves.
